Existence of diazinon, an organophosphorous pesticide, in river waters of Iran near rice paddy fields has been reported by some authors. The present research aimed to determine the acute toxicity and evaluate the effect of sub-lethal concentrations of diazinon on some biochemical parameters of rainbow trout, Oncorhynchus mykiss after 7, 14 and 28 days. No significant differences were observed in the plasma levels of creatinine among the treatment groups at different sampling intervals. Acetylcholinesterase activity and the levels of total protein, albumin as well as globulin in plasma were significantly reduced at both concentrations tested (p < 0.05). Lactate dehydrogenase activity was only decreased on 7th day in 0.1 mg/L diazinon treatment (p < 0.05). Creatine kinase activity was significantly lower in 0.1 mg/L diazinon group at 14th and 28th sampling periods, whereas its activity significantly increased in fishes exposed to 0.2 mf/L diazinon only on 7th day (p < 0.05). Aspartate aminotransferase, alanine aminotransferase activities and glucose levels in diazinon treated groups were significantly higher than the controlled group at experimental periods (p < 0.05). In conclusion, long-term exposure to diazinon at sub-lethal concentrations induced biochemical alterations in rainbow trout, and offers a simply tool to evaluate toxicity-derived alterations.  相似文献

The toxicity of fenvalerate to the prawn Penaeus monodon was evaluated using biomarkers of stress. In a preliminary bioassay test, P. monodon was exposed to a series of fenvalerate concentrations, which showed 4, 6.5 and 8.5 μg L−1 to be sublethal, median lethal and lethal, respectively. Sublethal effect of fenvalerate was further evaluated in hepatopancreas, muscle and gills of prawns with reference to oxidative stress biomarkers. Significant induction of lipid peroxidation and glutathione-S-transferase activity was found in hepatopancreas, muscle and gills of prawns exposed to fenvalerate when compared to control (P < 0.001, P < 0.05 and P < 0.05). On the contrary, the activities of Superoxide dismutase, catalase, glutathione peroxidase, vitamin C, vitamin E and glutathione were found to be reduced in the experimental group of prawns when compared to control. The results suggest that the animals were under oxidative stress when exposed to sublethal concentration of fenvalerate.  相似文献

In the present study, 40 male Wistar albino rats were used and divided into 4 groups. The first group served as the control group; the second group was administered Saw palmetto extract at the dose of 20 mg/kg/bw; the third group was administered flumethrin at the dose of 15 mg/kg/bw; and the fourth group was administered a combination of 20 mg/kg/bw Saw palmetto extract and 15 mg/kg/bw flumethrin, for 21 days, orally. After the trial period, blood and tissue (liver, kidney and brain) samples were taken from the rats. Saw palmetto extract did not cause significant alterations in plasma and tissue malondialdehyde (MDA) levels, serum and tissue nitric oxide (NO) levels, erythrocyte and tissue superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH-Px) activities when compared to the controls (p > 0.05). Flumethrin led to increased plasma and tissue MDA levels, serum and tissue NO levels, tissue GSH-Px activities and decreased erythrocyte and tissue SOD and CAT activities, and erythrocyte GSH-Px activity, compared to the controls (p < 0.05). The flumethrin and Saw palmetto extract combination increased erythrocyte SOD activity and decreased brain GSH-Px activity as compared to flumethrin (p < 0.05). In conclusion, it was determined that Saw palmetto extract did not cause any negative effect on the prooxidant-antioxidant balance. While flumethrin stimulated lipid peroxidation; Saw palmetto extract at the dose of 20 mg/kg/bw did not exhibit enough antioxidant effect in rats.  相似文献

Rotenone, an insecticide, causes toxicity through inhibition of mitochondrial electron transport chain at complex I and oxidative injury to the tissues. The aim of the present study was to determine in vivo effects of rotenone on myocardium and cardio-protective effects of caffeic acid phenethyl ester (CAPE), an antioxidant agent, against rotenone toxicity in rats. The rats were divided into three groups: untreated control, rotenone (2.5 mg/kg/day for 60 days, i.p.) and rotenone + CAPE groups. CAPE was administrated i.p. 10 μmol/kg/day for 62 days started two days before first dose rotenone injection. The malondialdehyde, nitric oxide levels and xanthine oxidase activity of rotenone group was significantly higher than control and rotenone + CAPE groups (p < 0.05). However, catalase activity in the rotenone group was decreased in comparison with the other groups (p < 0.05). The superoxide dismutase activity of rotenone group was insignificantly decreased compared to the others. In conclusion, rotenone caused lipid peroxidation in myocardial tissue and CAPE treatment prevented this rotenone-induced lipid peroxidation in rats. CAPE might be a cardio-protective agent against myocardial toxicities.  相似文献

In this study, the effects of sublethal exposures to the anticholinesterase insecticides azinphos methyl (AzMe) and carbaryl on the detoxifying responses of juvenile rainbow trout Oncorhynchus mykiss were investigated. Juvenile specimen were exposed to sublethal concentrations of AzMe (2.5 and 5 μg/L) and carbaryl (1 and 3 mg/L) for 24, 48 and 96 h. Carboxylesterase (CbE), catalase (CAT) and glutathione S-transferase (GST) activities as well as reduced glutathione (GSH) and cytochrome P450-1A (CYP1A) levels were monitored in liver and/or kidney. In all exposed groups liver CbE was significantly inhibited. Liver and kidney GSH level was reduced after sublethal exposure to both compounds. Carbaryl induced CAT activity during the first 48 h of exposure, followed by a significant decrease, whereas AzMe continuously decreased CAT activity. GST activity and CYP1A were transiently induced at 24 h by carbaryl exposure (3 mg/L) but sublethal exposure to AzMe did not affect GST activity or CYP1A. Our results show that the O. mykiss detoxifying system are a target for carbaryl and AzMe action, probably affecting redox balance. Although the responses showed similar trends in both organs, they were more important in liver than in kidney. The early inhibitory effect in CAT activity and GSH content produced by AzMe may be associated with a high degree of oxidative stress. Early induction of CYP1A, GST and CAT by carbaryl followed by enzyme inhibition suggests a milder or delayed oxidative stress, revealing differences between both pesticides metabolization. CbE inhibition is a good biomarker for AzMe and carbaryl exposure.  相似文献

Angiotensin-converting enzyme inhibitors (ACEi) were shown to ameliorate endothelial dysfunction in various human diseases and some of these inhibitors have been reported to enhance antioxidant defenses. The objective of the present study was to shown the abilities of enalapril and lisinopril as two nonthiol ACEi on mitochondrial toxicity due to paraquat. In this study, mitochondrial isolation from rat liver was divided into six groups. Group 1 was considered as control, group 2 received paraquat (5 mM), group 3 received enalapril (0.25 mM), group 4 received lisinopril (0.01 mM), group 5 received paraquat (5 mM) + enalapril (0.25 mM), and group 6 received paraquat(5 mM) + lisinopril (0.01 mM). Viability, lipid peroxidation, catalase activity, GSH (reduced glutathione) and GSSG (oxidized glutathione) concentrations were also determined. Simultaneous treatment of mitochondria with enalapril (0.25 mM) + paraquat (5 mM) and lisinopril (0.0.01 mM) + paraquat (5 mM) did not significantly ameliorate the mitochondrial toxicity induced by paraquat (5 mM) alone (p > 0.05). However, the nonthiol ACEi, enalapril showed to partially improve target of lipid peroxidation due to paraquat. In conclusion, nonthiol ACEi treatment did not improve the increased oxidative stress and the decreased antioxidant mechanisms.  相似文献

In the present study, we investigated the possible protective role of vitamin E, selenium (Se) and vitamin E plus Se in fenthion-induced organophosphates (OP) toxicity in rats. Serum concentrations of ascorbic acid, retinol, β-carotene, ceruloplasmin, nitrite and nitrate as well as levels of malondialdehyde (MDA) and reduced glutathion (GSH) in whole blood and in some tissues such as brain, heart, jejunum, kidney, liver, lung, muscle and pancreas were measured in sham, control, vitamin E, Se and vitamin E + Se groups. Compare to the sham group, the MDA (p < 0.001) and GSH (p < 0.01) levels in whole blood and some in tissues were significantly higher in the control animals. Ceruloplasmin levels of the control (p < 0.05), vitamin E (p < 0.05) and vitamin E + Se (p < 0.01), groups were higher than the sham group. Ascorbic acid, retinol, β-carotene as well as nitrite and nitrate levels in the control group were significantly lower than sham, vitamin E, Se and vitamin E + Se groups. We concluded that fenthion toxicity-induced lipid peroxidation and generation of free radicals in whole blood and tissues. Additionally, the antioxidants we tested did show a significant protective effect against OP-induced tissue and blood injury at the biochemical level.  相似文献

Experiment was carried out to determine the median lethal concentration (LC50) of fenvalerate to Labeo rohita fingerlings. After determining the LC50 value of fenvalerate, a sub-lethal concentration (1/3rd of LC50) of fenvalerate was exposed for 15 days. Significant alterations in SOD (P < 0.05) activity of liver and gill was observed due to fenvalerate. Catalase activity in gills of fishes was also affected significantly (P < 0.05). WBC, NBT and Hct values were reduced significantly in fenvalerate exposed fishes as compared to control group, whereas blood glucose level showed higher values in fenvalerate exposed group. Serum total protein and albumin were also reduced significantly as a result of fenvalerate exposure. Significant increase in the serum GOT, serum GPT, creatinine, triglyceride and serum ACP was noticed after 15 days of fenvalerate exposure. Results indicated that short term exposure of fenvalerate can induce biochemical and haematological alterations causing stress to L. rohita fingerlings.  相似文献

Paraquat is a highly toxic herbicide that is used in most countries without restriction. The cytotoxic action of paraquat is mediated by reactive radicals that are products of its metabolic reduction in cells. It has already been hypothesized that some angiotensin-converting enzyme inhibitors (e.g., captopril and enalapril) could show antioxidant and radical scavenging activity through their structural thiol groups, increasing antioxidant enzymes production or nitric oxide synthesis. In this study the hepatoprotective effect of captopril and enalapril against paraquat induced oxidative stress cytotoxicity was evaluated in isolated rat hepatocyte. Subtoxic concentrations of captopril (0.2 mM) and enalapril (0.2 mM) significantly (p < 0.05) protected the hepatocytes against paraquat (2 mM) induced oxidative stress cytotoxicity markers including: cell lysis, reactive oxygen species (ROS) generation, lipid peroxidation, glutathione depletion, mitochondrial membrane potential decrease, lysosomal membrane oxidative damage and cellular proteolysis. Moreover, we showed that non-thiol enalapril acts as well as thiol containing captopril at inhibiting oxidative stress cytotoxicity markers. Finally, our results support the hypothesis that it is the increase in nitric oxide synthesis and not the presence of the thiol group that accounts for the antioxidant activity of ACE inhibitors.  相似文献

The main propose of the present study was to determine the effects of diazinon on the activity and gene expression of glutamate dehydrogenase (GDH) as the key enzyme of Langerhans islet for secretion of insulin. Diazinon was administered intraperitoneally at doses of 15, 30, and 60 mg/kg. Langerhans islets were isolated from the pancreas of rats by a standard collagenase digestion, separation by centrifugation, and hand-picking technique. The activity and gene expression of the mitochondrial GDH was determined in the islets homogenates. Glutamate, C-peptide, and insulin were determined in plasma.Diazinon at all tested doses (15, 30, and 60 mg/kg) significantly (p < 0.01) decreased plasma insulin after 1 h while the values did not differ from control when examined after 18 h. Diazinon at all tested doses (15, 30, and 60 mg/kg) significantly (p < 0.01) increased concentration of C-peptide both 1 and 18 h post-administration. Diazinon at all tested doses (15, 30, and 60 mg/kg) significantly (p < 0.05) increased production of glutamate while the values did not differ from control when tested after 18 h. Administration of diazinon at doses of 30 and 60 mg/kg significantly (p < 0.001) increased activity of GDH after 1 h while all doses of diazinon increased GDH activity when measured after 18 h. Diazinon at dose of 60 mg/kg significantly (p < 0.01) decreased expression of GDH gene 18 h post-administration.It is concluded that GDH is a component of diazinon-induced changes in release of improper insulin.  相似文献

Recovery study was performed at regular intervals to establish the time course of 50% and 100% recovery in neurotransmitter enzyme (acetylcholinesterase, AChE, EC 3.1.1.7) and locomotor behaviour response of mosquito fish, Gambusia affinis exposed to lethal concentration (20.49 mg L−1) of an organophosphorous pesticide, monocrotophos (MCP) for 96 h. In vitro AChE activity studies indicated that MCP could cause 50% inhibition (I50) at 10.2 × 10−5 M. A positive correlation was observed between brain AChE activity and swimming speed during the recovery study. Also, the recovery response of the antioxidant enzymes superoxide dismutase (SOD, EC 1.15.1.1), catalase (CAT, EC 1.11.1.6) and glutathione reductase (GR, EC 1.6.4.2) as well as lipid peroxidation (LPO) as biomarkers of oxidative stress were assessed in viscera of G. affinis. The results showed that the MCP besides its inhibitory effect on target enzyme AChE activity and induction in antioxidant enzyme activities as a characteristic of oxidative stress, which can be used as biomarkers in the pesticide contaminated aquatic streams.  相似文献

Rainbow trout (Oncorhynchus mykiss; 116.88 ± 21.69 g) were exposed to sublethal concentrations (25 μg/L) of carbosulfan for 60 days to test if the long term exposure of fish to carbosulfan affects red blood cells acetylcholinesterase (AChE), δ-aminolevulinic acid dehydratase (ALA-D) and paraoxonase (PON) enzyme activity and induces genotoxic and/or mutagenic effects. The exposure resulted in inhibition of AChE and ALA-D activity of rainbow trout when compared to control fish. The activity of PON was not affected by carbosulfan. Interestingly, carbosulfan was found to induce DNA damage in red blood cells (comet assay) and proved to be mutagenic as revealed by the Ames test. Results indicate that blood AChE and ALA-D of rainbow trout may be a sensitive biomarker for assessment of carbosulfan contaminated water bodies. Furthermore, because the Ames test and comet assay were proven successful to detect the genotoxicity of carbosulfan, we proposed that nonlethal techniques such as blood collection from caudal vein of fish should be used to determine potential toxic effects of other pesticides to surrounding environment.  相似文献

Deltamethrin and thiachloprid are an α-cyano class pyrethroid and neonicotinoid insecticide, respectively. Recently, a pesticide combining deltamethrin and thiacloprid has also been released. In the present study, the acute and subacute toxic effects of deltamethrin, thiachlopride, and a combination of these insecticides, on the lymphoid organs (spleen, thymus and bone marrow), polymorphonuclear leukocytes (PMNs) and plasma of rats, were determined to better understand mammalian antioxidant-oxidant and inflammatory system responses. For this purpose, rats were treated orally with different doses of thiacloprid (single acute dose of 112.5 mg/kg); subacute dose of 22.5 mg/kg/day for 30 days; deltamethrin (single acute dose of 15 mg/kg); subacute dose (3 mg/kg/day for 30 days), or a combination of these pesticides. Results were compared with those from a comparable dosing regimen with the known immunosuppressive drug cyclophosphamide. Pesticide treatments caused significant changes in the levels of liver and kidney injury markers. Antioxidant enzyme (catalase and glutathione peroxidase), glutathione and plasma antioxidant levels decreased but lipid peroxidation increased in all lymphoid organs and the plasma. Glutathione-S-transferase and especially DT-diaphorase activity, decreased after thiacloprid treatment. Myeloperoxidase activity, carbonyl content, lipid peroxidation and total nitrite levels increased in PMNs and plasma. When evaluated as a whole, the oxidative and inflammatory stresses seen in the pesticide combination groups were not much more pronounced than in the groups treated with a single pesticide. In terms of the evaluated biochemical parameters, the pesticides showed similar effects to cyclophosphamide.  相似文献

Male and female rats were orally administered chlorpyrifos at a dose of 6.75 mg kg−1 body weight for 28 consecutive days. An additional chlorpyrifos group received zinc (227 mg l−1) in drinking water throughout the experimental duration. Two groups more served as controls; one received water only and the other received zinc in drinking water. Administration of chlorpyrifos resulted in a significant increase in lipid peroxidation (LPO) level and significant decrease in the activities of superoxide dismutase (SOD), glutathione-s-transferase (GST), catalase (CAT) and acetylcholinesterase (AChE) in erythrocytes of male and female rats. In contrast, zinc-chlorpyrifos treatment showed insignificant differences (p ? 0.05-0.01), compared to control results, regarding LPO, SOD, GST and CAT. In case of AChE, supplementation of zinc showed little alteration in the activity of this enzyme in the rats treated with chlorpyrifos. It can deduce that chlorpyrifos induced oxidative stress and lipid peroxidation in erythrocytes of male and female rats. The overall results reveal the pronounced ameliorating effect of zinc in chlorpyrifos-intoxicated rats and variation in the response of male and female animals regarding alteration in the level of some biochemical parameters and LPO.  相似文献

Five experimental diets with various combinations of probiotics, namely T1 & T6 (basal feed (BF) without probiotics), T2 & T7 (BF + Bacillus subtilis + Lactococcus lactis), T3 & T8 (BF + L. lactis + Saccharomyces cerevisiae), T4 & T9 (BF + B. subtilis + S. cerevisiae) and T5 & T10 (BF + B. subtilis + L. lactis + S. cerevisiae) were fed to Labeo rohita fingerlings for 30 days. Treatment groups T1, T2, T3, T4 and T5 were exposed to Fenvalerate, at a concentration of 1.79 μg L−1. The SOD and CAT activity was significantly affected (P < 0.01) in fenvalerate treated groups. However, the supplementation of the three-probiotic mixture at equal concentration showed markedly reduced activity. Similarly, RBC, Hb, NBT, total protein and albumin values were reduced significantly (P < 0.01) in the fenvalerate exposed fish as compared to the probiotic supplemented fish. Fenvalerate exposure also showed increased serum ALP, ACP and Bilirubin values (P < 0.01) in comparison to the non-exposed fish. Histological observations of the gills, kidney and liver showed tissue degeneration after fenvalerate exposure, which however showed marked recovery on the three-probiotic mixture supplementation. Therefore, these results indicate that a mixture of multi-species probiotic supplementation in equal concentration acts beneficially in mitigating the stressful effects of fenvalerate.  相似文献

Amitraz is a pesticide targeting the octopaminergic receptors. In a previous study, octopamine, a biogenic amine, was found to induce a biphasic effect on the honeybee heart, inhibition at low concentrations and excitation at high concentrations. Furthermore, the honeybee heart was found to be far more sensitive to octopamine compared to other insect hearts. The objective of the present study was to investigate the effects of amitraz on the electrical and mechanical properties of the honeybee heart ex vivo and on the heart rate in vivo. In ex vivo conditions, amitraz at 10−12 M caused a significant inhibition in the mechanical (p < 0.05, n = 4) and electrical properties (p < 0.05, n = 4). Higher concentrations such as 10−9 and 10−6 M induced a biphasic effect, with total inhibition for 7.86 ± 1.26 min (n = 7), followed by strong excitation of spontaneously-generated contractions (n = 7). The initial elimination of heart activity was caused by strong hyperpolarization, while the subsequent excitation was caused by a depolarization in the membrane potential of pacemaker cells at 10−9 M (n = 8). In the in vivo experiments, abdominal injection or oral application of 0.20 ng of amitraz per bee induced a persistent increase of 134.28 ± 4.07% (p < 0.05, n = 4) in the frequency of the cardiac action potentials. The above responses clearly show that the heart of the honeybee is extremely vulnerable to amitraz, which is nevertheless still used inside beehives, ostensibly to “protect” the honeybees against their main parasite, Varroa destructor.  相似文献

Endosulfan is an organochlorine pesticide widely used in agriculture and hence finds its way into natural water bodies, thus affecting aquatic life. The purpose of this study was to determine LC50 of endosulfan (99%; α:β ratio of 7:3) in Tilapia, Oreochromis mossambicus and study its effect on anti-oxidative enzymes (superoxide dismutase, glutathione-S-transferase and catalase), immuno-hematological profile (RBC, WBC, Hb, serum protein, albumin-A, globulin-G, A/G ratio, phygocytic activity as indicated by nitroblue tetrazolium reduction, serum cortisol and serum lipid peroxidation) and neurotransmitter acetylcholine esterase enzyme activity. The LC50 value at 96 h and 95% confidence limit for tilapia (46.78 g) was estimated as 3.6 μg/L. Activities of anti-oxidative enzymes, immuno-hematological profile, blood glucose and neurotransmitter activity was significantly influenced (P < 0.01) in dose dependent manner. This was reflected in the behavior of fish that was altered from normal during acute toxicity.  相似文献

Diazinon is commonly used for pest control in the agricultural fields surrounding freshwater reservoirs. So this study was conducted to determine the acute toxicity of this organophosphorous pesticide, contaminating aquatic ecosystems as a pollutant, and its effects on behavior, and some hematological parameters of fingerling European catfish, Silurus glanis. Diazinon was applied at concentrations of 1, 2, 4, 8, 16, 32, and 64 mg L−1. The water temperature in the experimental units was kept at 16 ± 1 °C. The number of dead fishes significantly increased in response to diazinon concentrations 2-64 mg L−1 (p < 0.05). With increasing diazinon concentrations, the fishes exposed duration 1 to 96 h significantly increased the number of dead fishes (p < 0.05 for each cases). The 1, 24, 48, 72, and 96 h LC50 values (with 95% confidence limits) of diazinon for fingerling European catfish were estimated as 14.597 (12.985-16.340), 12.487 (11.079-14.471), 8.932 (7.907-10.348), 6.326 (no data because of p > 0.05), and 4.142 (no data because of p > 0.05) mg L−1, respectively. Compared to the control specimens, fish after an acute exposure to diazinon was significantly lower erythrocyte, leukocyte, hemoglobin, hematocrit, MCV, MCH, and MCHC values (p < 0.05). In addition, it was also showed a significantly negative correlation between these hematological parameters and exposure times of diazinon (p < 0.01).  相似文献

Rice (Oryza sativa), a relatively tolerant species, and early watergrass (Echinochloa oryzoides; EWG), a relatively susceptible species, were exposed to 14C-labeled clomazone to determine accumulation, biotransformation, and mass balance. On a total mass basis, rice absorbed more clomazone than EWG (p < 0.05), but on a nmol/g basis, there was no significant difference between the two species (p > 0.05). Rice contained more extractable 14C residues (7.7 ± 0.5 vs. 4.8 ± 0.5 nmol in rice vs. EWG, respectively; p < 0.5), but the concentration in EWG was significantly higher (4.2 ± 0.5 vs. 1.8 ± 0.1 nmol/g in EWG vs. rice, respectively; p < 0.01). More metabolized residue was measured in EWG compared to rice (84.1% vs. 67.9%; p < 0.01). Both species produced hydroxylated forms, β-d-glucoside conjugates, and several other unidentified polar metabolites, but EWG generally produced higher metabolite concentrations. The concentration of the suspected active metabolite, 5-ketoclomazone, was significantly higher in EWG vs. rice (21 ± 2 vs. 5.7 ± 0.5 pmol/g, respectively; p < 0.01). Differences in sensitivity to clomazone between rice and EWG appear to be due to differential metabolism, but in this case the more susceptible EWG qualitatively and quantitatively metabolized more clomazone than the more tolerant rice. This is consistent with the action of a metabolically activated herbicide. This metabolic difference could be exploited to develop herbicide safeners for use with clomazone.  相似文献
