Over ten-year routine inspection results on organochlorine pesticide (OCP) residue were summarized, OCPs residues, including BHC isomers (α, β, γ, and δ-BHC), DDT analogs (p,p′-DDD, p,p′-DDE, o,p′-DDT, and p,p′-DDT), and pentachloronitrobenzene (PCNB) and its metabolites (pentachloroaniline and methyl pentachlorophenyl sulfide (MPCPS)), in 1,665 samples for 37 types of Chinese herbal medicine (CHM) using the QuEChERS method coupled with the GC-ECD. Based on the maximal residue levels for OCPs set by Asian pharmacopeias, PCNB contamination in Ginseng radix as well as the total DDT and PCNB contamination in Panacis quinquefolii radix are of concern. OCP residues in different parts of Panax ginseng were also compared. The total BHC residue in leaf and fibrous root, as well as the total DDT and PCNB residue in all parts, exceeded MRL of 0.1 mg/kg. Overall, this study provided meaningful results about OCP residue in CHM for pharmaceutical industries and consumers.  相似文献

A total of 146 samples of different kinds of cheeses produced in Spain were analysed in order to ascertain the specific contamination pattern. The organochlorine compounds studied were those most commonly investigated in previous surveys: α-HCH, β-HCH, γ-HCH (lindane), γ-HCH, chlordane, aldrin, dieldrin, endrin, heptachlor, heptachlor epoxide, and the isomers and metabolites of DDT. α-HCH, β-HCH, γ-HCH, chlordane, p,p′, DDT, and p,p′-DDE were found in more than 76% of samples; p,p′-DDE and γ-HCH were the most frequently detected, with frequencies of 100 and 97.9% respectively. γ-HCH, aldrin, dieldrin, heptachlor, heptachlor epoxide, o,p′-DDT, p,p′-DDD and o,p′-DDD were observed at lower frequencies. No residues of endrin were detected in any sample. Insecticides exceeding the maximum residue limits (MRLs) were chlordane, β-HCH, α-HCH and γ-HCH, with 42, 20, eight and six samples respectively. Mean residues of organochlorines found were as follows (μ kg?1 butterfat): α-HCH = 46.3; β-HCH = 46.5; γ-HCH = 54.2; δ-HCH = 16.9; aldrin = 16.7; dieldrin = 9.7; heptachlor = 15.9; heptachlor epoxide = 14.8; chlordane = 50.2; o,p′-DDT = 5.1; p,p′-DDT = 12.4; o,p′-DDT = 19.6; p,p′-DDD = 46.7; o,p′-DDE = 6.9; p,p′-DDE = 40.7 (.DDT = 55.0). Estimated dietary intakes (EDIs) from cheese consumption were compared to acceptable daily intakes (ADIs) for the pesticides where residues exceeded the MRL. EDIs calculated were in all cases below ADIs, and, therefore, based on the ADIs, there is no health risk involved in the consumption of cheese from Spain arising from organochlorine residues.  相似文献

Raf-1 is the best characterized member of the mammalian raf family. The raf-1 kinase has many important physiological functions but its regulation is complex. 1,1-Dichloro-2,2 bis(p-chlorophenyl) ethylene (p,p-DDE), a major and important metabolite of organochlorine pesticide DDT, is a widespread environmental pollutant. Previous studies have reported that p,p′-DDE influences the MAPK pathway, but little details are known regarding how the raf-1 activity is influenced and regulated by p,p′-DDE in those events. And here, we used 20 male rats that received different doses of p,p′-DDE (0, 20, 60, 100 mg/kg b.wt) every other day by intraperitoneal injection for 10 days. Results showed that the raf-1 mRNA and protein expressions showed significant down-regulation. The thyroxine (T4) level and the protein kinase C alpha (PKCα) mRNA expression declined, but the raf kinase inhibitory protein (RKIP) up regulated. In addition, the p,p′-DDE-induced oxidative stress resulted in declines of glutathione peroxidase (GSH-PX) and superoxide dismutase (SOD) activities. The elevations of ERK mRNA level and phosphorylated ERK protein expression were also observed. Taken together, these results suggest that the raf-1 is dually down regulated by p,p′-DDE via two pathways. One pathway is that the p,p′-DDE exposure elevates RKIP level by decreasing T4 and PKCα level, and the increased RKIP eventually down regulates the raf-1. The other is that the p,p′-DDE exposure activates the ERK by oxidative stress, and the activated ERK down regulates the raf-1 through the negative feedback phosphorylation.  相似文献

The activity of the mutant CYPBM3 “21B3”, which is able to use hydrogen peroxide as the final electron acceptor, was evaluated against two major environmental pollutants; organochlorine and organophosphorus pesticides. This evolved CYP from Bacillus megaterium is able to transform a variety of structurally different pesticides. The catalytic parameters for two organochlorine; dichlorophen (kcat = 9.2 min−1, KM = 64.1 μM) and linuron (kcat = 226.5 min−1, KM = 468.2 μM), and two organophosphorus compounds; parathion (kcat = 10.9 min−1, KM = 59.3 μM) and chlorpyrifos (kcat = 9.2 min−1, KM = 226.5 μM) were determined giving catalytic efficiencies between 0.143 and 1.107 min−1 μM−1. CYPBM3 “21B3” has the ability to both activate and detoxify organophosphorus pesticides, as demonstrated by the chemical nature of the reaction products. The capacity to transform structurally diverse compounds together with the great stability, easy production and relatively inexpensive cofactors needed, makes CYPBM3 “21B3” an enzyme with a potential use on the environmental field.  相似文献

Here we investigated the in vitro and in vivo effects of the pesticides, deltamethrin, diazinon, propoxur and cypermethrin, on the activity of rainbow trout (rt) gill carbonic anhydrase (CA). The enzyme was purified from rainbow trout gills using Sepharose 4B-aniline-sulfanilamide affinity chromatography method. The overall purification was approx. 214-fold. SDS-polyacrylamide gel electrophoresis showed a single band corresponding to a molecular weight of approx. 29 kDa. The four pesticides dose-dependently inhibited in vitro CA activity. IC50 values for deltamethrin, diazinon, propoxur and cypermethrin were 0.137, 0.267, 0.420 and 0.460 μM, respectively. In vitro results showed that pesticides inhibit rtCA activity with rank order of deltamethrin > diazinon > propoxur > cypermethrin. Besides, in vivo studies of deltamethrin were performed on CA activity of rainbow trout gill. rtCA was significantly inhibited at three concentrations (0.25, 1.0 and 2.5 μg/L) at 24 and 48 h.  相似文献

The effects of p,p′-DDT and four of its analogs on electrical activity in the central nervous system of the cockroach, Periplaneta americana (L.), were investigated. Cockroaches were injected intraabdominally with an organochlorine compound at LD50 96-hr doses (except for p,p′-DDE). Extracellular recordings were made from the central nervous system at 1 hr, 24 hr, or 3 weeks postinjection. p,p′-DDT, methoxychlor, and p,p′-DDD induced behavioral changes (tremors, jitters, hyperexcitability) and repetitive firing in the central nervous system prior to 1 hr postinjection. By 24 hr postinjection, most behavioral signs of poisoning had disappeared, though repetitive firing could still be readily elicited in the central nervous system. Cockroaches injected with o,p′-DDT, however, usually required about 48 hr before overt signs of poisoning became apparent. Cockroaches treated with p,p′-DDT or o,p′-DDT behaved normally at 3 weeks postinjection but still displayed a significant occurrence of repetitive firing in the central nervous system. A mechanism is proposed to explain how a cockroach might recover behaviorally from a sublethal dose of an organochlorine compound but still display repetitive firing in its central nervous system. A direct “cause and effect” relationship between repetitive firing in the central nervous system and mortality (and external signs of poisoning) is therefore questioned.  相似文献

In an attempt to find the biorational pesticides, we synthesized 12 pyridinyl derivatives of podophyllotoxin (PPT) and 4′-demthylepipodophylltoxin (4′-DMEP) in this study. Their structures and the α/β substitution at C-4 were confirmed by 1H NMR, IR, MS spectral analyses and elemental analysis. The insecticidal activities were tested against fifth-instar larvae of Pieris rapae and the third-instar larvae of Cullex pipiens pallens at concentrations of 250 and 10 μg ml−1. Four derivatives of PPT, 4.1, 4.2, 4.3 and 4.5, showed higher insecticidal activities against P. rapae than PPT, while three derivatives of PPT, 4.4, 4.5 and 4.6, displayed higher mosquito larvicidal activity than PPT, with LC50 values of 1.66, 3.96 and 1.54 mg l−1, respectively. Interestingly, we also found that the pyridine ring derivatives of PPT showed delayed insecticidal activity, which is different from traditional neurotic insecticides. The results suggest that 4′-OCH3 in the PPT derivatives is essential to keep the insecticidal activity and the insecticidal activities of pyridine ring derivatives of PPT are higher than that of the derivatives of 4′-DMEP, supporting PPT has the potential to be a lead structure of semi-synthetic insecticides.  相似文献

Four groups of Indian buffaloes were fed daily with 25 mg of p,p′-DDT p,p′-TDE p,p′-DDE or o,p′-DDT for 100 days. Milk was analysed for organochlorine residues during this period and also for 100 days after pesticide administration had been discontinued. For the period showing ‘plateau level’ residues, 17.2% of p,p′-DDE, 17% of p,p′-TDE, 14% of p,p′-DDT as p,p′-DDT (3.5%); p,p′-TDE (10.5%); 3.2% of p,p′-DDT as o,p′-DDT (1.3%) and o,p'-TDE (1.9%) of their administered amounts were excreted in the milk. Since these compounds were excreted at different rates, the residue levels in the milk expected from a given feed would depend on their concentration and proportional distribution in the feed. The maximum tolerable content of DDT analogues in feed was derived to be 0.1 mg kg?1 (dry weight basis) by using the maximum accumulation coefficient and incorporation of the necessary safety margin. It is concluded that Indian buffaloes fed with rations contaminated with a total of DDT analogues below this limit will yield milk of acceptable quality. Following the termination of feeding with contaminated rations, the elimination of p,p′-DDE in the milk took the longest time and that of o,p′-DDT the shortest. These results suggest that the time required for the initial high residue concentration to decline to less than the legal limit would be determined by the relative amounts of DDE, TDE and DDT in the milk, after elimination of the potent source of contamination.  相似文献

The effects of paraoxon and atrazine on the spontaneously beating auricle, isolated from the heart of Sparus aurata, were assessed. Paraoxon, 5 μM, eliminated the atria contraction within 28.4 ± 2.8 min, an effect which was fully reversed by 15 μM atropine, an antagonist of muscarinic cholinergic receptors. The IC50 was estimated to be 3.2 ± 1.5 μΜ. Atrazine, 50 and 100 μM, induced a 22.5 ± 3.2 and 32.9 ± 2.3% increase in the force of auricle contraction, caused by excitation of sympathetic synaptic terminals releasing adrenaline. This effect was reversed by 50 μM propranolol, a blocker of β-adrenoreceptors. The results have shown that both sympathetic and parasympathetic nerve terminals are activated by atrazine. Also, the auricle contraction is mainly under sympathetic control, while the frequency is dominated by cholinergic system. Finally, the detailed parameters of the auricle contraction estimated during exposure to specific pesticides, force, frequency, time-response curves and electromechanical coupling can be further used to assess and compare the toxic effects of other compounds, anticholinesterases for example, on the heart of the fish.  相似文献

Brucea javanica (L.) Merr. is a medicine plant distributed widely throughout Asia where its bitter fruits have been used traditionally in medicine for treating various ailments and controlling some pests. In recent years, concerns over the potential impact of synthetic pesticides on human health and environment have now become more pressing to develop environmentally friendly pesticides. In this paper, brusatol, a quassinoid, was isolated from the fruit of B. javanica, and identified using X-ray crystallographic analysis. Results showed that brusatol has potent contact toxicity (LD50, 2.91 μg/larva, 72 h) and anfieedant activity (AFC50, 17.4 mg/L, 48 h) against the third-instar larvae of Spodoptera exigua. Brusatol demonstrated cytotoxic effects to the tested insect cell lines, IOZCAS-Spex-II and Sf21, in a time- and dose-dependent manner. After brusatol treatment, apoptotic cell death with the DNA fragmentation, activation of caspase-3 and release of cytochrome c was preliminarily observed in both IOZCAS-Spex-II and Sf21. These results indicated the existence of apoptotic death with the mitochondrial-dependent pathway induced by brusatol in Sf21 and IOZCAS-Spex-II cell lines. Our studies will provide important knowledge to understand mechanisms of action of brusatol and to develop brusatol and its derivatives as insecticides.  相似文献

A natural compound plumbagin (5-hydroxy-2-methyl-1,4-naphthoquinone) was isolated from the leaves of Plumbago auriculata and found to inhibit the enzyme, 8-amino-7-oxononanoate synthase (AONS, also known as 7-keto-8-aminopelargonate synthase, KAPAS) an IC50 of 2.1 μM in vitro. Biotin supplement significantly rescued the plant injury caused by the plumbagin treatment, and this result confirmed the target site, AONS. Foliar application of 1000 ∼ 2000 μg/mL plumbagin in a greenhouse condition showed lethal activity against eight species of weeds, containing three grass species of Sorghum bicolor, Echinochloa crus-galli, Digitaria sanguinalis and five broad leaf species of Solanum nigrum, Aeschynomene indica, Abutilon avicennae, Xanthium strumarium, Calystegia japonica. Field trial of foliar application with plumbagin 2000 μg/mL have successfully controlled 10 ∼ 15 leaf-stages and 2 ∼ 3 m vine lengths of Sicyos angulatus at the natural habitats around riparian zone in the Nam-Han River in Korea. Visual symptom of desiccation might be induced by the physiological cellular leakage which was significantly dose dependent on the plumbagin treatment regardless of light.  相似文献

Amounts of DDT and its breakdown products were determined in soil in an apple orchard in Herefordshire. Samples were taken for a number of years (1972–79) after use of the insecticide in the orchard had ceased in 1969. The results were compared with those obtained in an investigation of the same orchard in 1968. From 1968 to 1979, soil residues of pp′-DDT, p′--DDT and pp′--TDE decreased gradually whereas those of pp′--DDE increased, and there were linear relationships between log (concentration) and time. The calculated time for 50% decrease in concentration (Dt50) was 11.7 years for pp′--DDT, 3.3 years for pp′--TDE and 7.1 years for op′--DDT; the time for doubling the concentration for pp′--DDE was 9.1 years. Regression analysis on the two major components (pp′--DDT+pp′--DDE) indicated that the total amount (2.7 mg kg?1) was not decreasing with time. It was concluded that during a post-spray era, the breakdown of pp′--DDT to pp′--DDE was a significant feature of the persistence of DDT, and that, in contrast to the findings of other workers who sampled when DDT was being used, there were no losses by volatilisation. There was an exponential decrease in the amount of DDT residues with increasing soil depth and approximately 90% was found in the top 10 cm of the undisturbed soil profile.  相似文献

Agricultural chemicals can induce genetic alterations on aquatic organisms that have been associated with effects on growth, reproduction and population dynamics. The evaluation of DNA damage in fish using the comet assay (CA) frequently involves the utilization of erythrocytes. However, epithelial gill cells (EGC) can be more sensitive, as they are constantly dividing and in direct contact with potentially stressing compounds from the aquatic environment. The aim of the present study was to evaluate (1) the sensitivity and suitability of epithelial gill cells of Prochilodus lineatus in response to different genotoxic agents through the application of the CA, (2) the induction of DNA damage in this cell population after in vivo exposure to cypermethrin. Baseline value of the CA damage index (DI) for EGC of juvenile P. lineatus was 144.68 ± 5.69. Damage increased in a dose-dependent manner after in vitro exposure of EGC to methyl methanesulfonate (MMS) and H2O2, two known genotoxic agents. In vivo exposure of fish to cypermethrin induced a significant increase in DNA DI of EGC at 0.150 μg/l (DI: 239.62 ± 6.21) and 0.300 μg/l (270.63 ± 2.09) compared to control (150.25 ± 4.38) but no effect was observed at 0.075 μg/l (168.50 ± 10.77). This study shows that EGC of this species are sensitive for the application of the CA, demonstrating DNA damage in response to alkylation (MMS), oxidative damage (H2O2), and to the insecticide cypermethryn. These data, together with our previous study on DNA damage induction on erythrocytes of this species, provides useful information for future work involving biomonitoring in regions where P. lineatus is naturally exposed to pesticides and other genotoxic agents.  相似文献

p, p'-DDT, HEOD, endrin, heptachlor epoxide, hexachlorobenzene and lindane were fed in combination to day-old broiler chickens at levels of 0.05, 0.15 and 0.30 part/million in the feed, for a period of about 7 weeks. Birds were killed at regular intervals and samples of abdominal fat and other tissues examined for residues of the chlorinated pesticides by gas-liquid chromatography. The lipid content of each sample was determined and the residue levels calculated for the whole tissue and also for the fat contained in the sample. At the end of the 7 weeks (when in practice broilers are slaughtered) residues were highest for heptachlor epoxide, followed by hexachlorobenzene, HEOD, DDT (total), endrin, and lindane. Heptachlor epoxide residue levels in fat were 13 times the levels in the feed, for lindane the ratio was 3.3. Fat residues of each pesticide reached a plateau level relatively quickly, and these levels were proportional to the fortification rates in the feed. HEOD and p, p'-DDT alone were fed to separate groups of birds. No differences in residue build-up of these compounds were found between these groups and the groups that had received these compounds in combination with the other pesticides. The total amounts of each pesticide in the tissues were reduced when the meat was fried, probably by loss of fat during preparation and frying. For p, p'-DDT and heptachlor epoxide also smaller residue were observed in the remaining fat after frying.  相似文献

It is known that organophosphate pesticides and their metabolites are generally eliminated through urine and are likely to affect nephrons. Despite the widespread application of Dichlorvos only limited studies appear to have been done on its toxicity to kidney. Intraperitoneal administration of 400 μg/kg Dichlorvos in mice exhibited maximum reduction in total protein concentration of kidney after exposure to 120 h (52-fold decrease, t-test, P < 0.001). Variations in renal clearance and percent reabsorption of acid phosphatase, alkaline phosphatase and nonspecific esterase enzymes were significant at P < 0.01 and at P < 0.001 (2-way ANOVA), respectively. Maximum significant increase in renal contents of Na+ and Ca++ was induced by 200 μg/kg dose after 120 and 240 h exposure (P < 0.001), while significantly highest retention of K+ and Cl ions was caused by 400 μg/kg dose after 24 and 72 h exposure (P < 0.01). Histopathological changes in glomeruli, PCTs, DCTs and CTs along with altered renal tubular function and renal clearance of enzymes and various ions indicate the development of acute renal disturbances under the influence of Dichlorvos.  相似文献

The present paper deals with the investigations of optimal conditions for the myeloperoxidase (MPO) mediated oxidation of chlorpyrifos, azinphos-methyl and phorate, organophosphorous pesticides (OPs) containing phosphorothionate group, from thio- to oxo-forms, in the presence of hydrogen peroxide. The aim of the work was to apply this oxidation method in the AChE based bioanalytical tests for OPs determination. The maximum concentration of oxo-forms for all tested pesticides was achieved after 10 min incubation of OPs in 50 mM phosphate buffer (pH 6.0) with 100 nM MPO in the presence of 50 μM H2O2. Optimal temperature for obtaining maximal concentration of oxo-forms was 37 °C. Only the parent compounds and their oxo-forms were identified chromatographically in the OPs samples after their exposure to MPO. Moreover, no hydrolysis products were detected in the time interval of 1 h after the MPO catalyzed reaction was stopped by catalase. The efficiency of OPs transformation from thio- to oxo-forms was measured using acethylcholinesterase (AChE) test, by comparison of percent of AChE inhibition before and after exposure to the oxidized sample.  相似文献

Deltamethrin, a synthetic pyrethroid pesticide contaminating aquatic ecosystems as a pollutant, was investigated in the present study for toxic effects on embryos and larvae of common carp, Cyprinus carpio as a model. The control and five test experiments were repeated five times. The water temperature in the experimental units was kept at 24 ± 1 °C. The number of dead embryos significantly increased in response to deltamethrin concentrations 0.005, 0.05, 0.5, 5, 25, and 50 μg L−1 (p<0.05 for each cases). Dose-response decreases in hatching success were recorded as 75.2, 64.6, 47.4, 26.0, 14.4, and 9.0%, respectively. The lowest concentration of deltamethrin (0.005 μg L−1) produced a significantly decrease in number of dead larvae compared to control group (p<0.05). With increasing deltamethrin concentrations, the larvae exposed duration 1-48 h significantly increased the number of dead larvae (p<0.05 for each cases). The 48 h LC50 values (with 95% confidence limits) of deltamethrin for common carp embryos and larvae were estimated as 0.213 (0.103-0.404) and 0.074 (0.011-0.260) μg L−1, respectively. The results provide evidence that deltamethrin pollution may have an adverse effect on the reproduction and development of carp, which should be considered when this chemical is used in agricultural areas near aquatic ecosystems.  相似文献

In this study, the effects of malachite green on selected immunological parameters, oxidative stress and antioxidant status biomarkers in blood, liver, kidney, spleen and gill of rainbow trout, Oncorhynchus mykiss, was examined. During 5 days the malachite green was applied at concentrations of 1/15,000 and 1/150,000 for 30 s and 60 min, respectively. Immunological parameters (nitroblue tetrazolium (NBT) activity, total plasma protein (TP), total immunoglobulin (TI)) and biochemical parameters (lipid peroxidation (MDA), catalase (CAT) activity, reduced glutathione (GSH) levels) were evaluated after exposed to malachite green. It has been observed that NBT activity (p < 0.05, p < 0.001), total protein (p < 0.01, p < 0.001) and total immunoglobulin (p < 0.05, p < 0.001) levels were decreased compared with control group. In the rainbow tout exposed to malachite green duration 5 days significantly increased lipid peroxidation, which might be associated to decreased levels of reduced glutathione and catalase activity in the whole tissues of O. mykiss (p < 0.05, p < 0.01, p < 0.001 for each cases).  相似文献

Anti-vitamin K drugs are widely used as anticoagulant in human thromboembolic diseases. Similar compounds have also been used as rodenticides to control rodent population since 1950s. Massive use of first generation anticoagulants, especially warfarin, has lead to the development of genetic resistances in rodents. Similar resistances have been reported in human. In both cases, polymorphisms in VKORC1 (Vitamin K epoxide reductase subunit 1), the subunit 1 of the VKOR (Vitamin K epoxide reductase) complex, were involved. In rats (Rattus norvegicus), the Y139F mutation confers a high degree of resistance to warfarin. Little is known about the in vitro consequences of Y139F mutation on inhibitory effect of different anticoagulants available. A warfarin-susceptible and a warfarin-resistant Y139F strain of wild rats (Rattus norvegicus) are maintained in enclosures of the Lyon College of Veterinary Medicine (France). Using liver microsomes from susceptible or resistant rats, we studied inhibition parameters by warfarin (Ki = 0.72 ± 0.1 μM; 29 ± 4.1 μM), chlorophacinone (Ki = 0.08 ± 0.01 μM; 1.6 ± 0.1 μM), diphacinone (Ki = 0.07 ± 0.01 μM; 5.0 ± 0.8 μM), coumachlor (Ki = 0.12 ± 0.02 μM; 1.9 ± 0.2 μM), coumatetralyl (Ki = 0.13 ± 0.02 μM; 3.1 ± 0.4 μM), difenacoum (Ki = 0.07 ± 0.01 μM; 0.26 ± 0.02 μM), bromadiolone (Ki = 0.13 ± 0.02 μM; 0.91 ± 0.07 μM), and brodifacoum (Ki = 0.04 ± 0.01 μM; 0.09 ± 0.01 μM) on VKOR activity. Analysis of the results leads us to highlight different anticoagulant structural elements, which influence inhibition parameters in both susceptible and Y139F resistant rats.  相似文献
