14头中国实验用小型猪肌肉注射复合麻醉剂(XFM)0.15mL/kg,并在注药前及注药后5、10、30、45、60、80、100、120min进行收缩压(SBP)、舒张压(DBP)、平均动脉压(MAP)和心率(HR)的监测并同步采取前腔静脉血样,采用比色法和放免法测定血清中一氧化氮(NO)、血浆中内皮素(ET)、6-酮-前列腺素F1α(6-keto-PGF1α)和血栓素B2(TXB2)的含量。结果显示,血压和HR主要是在注药后5~10min及80min时出现明显的变化(P<0.01或P<0.05)。ET和TXB2与SBP、DBP和MAP呈相关或显著相关性(P<0.05或P<0.01);而6-keto-PGF1α与SBP、DBP和MAP呈负相关或显著负相关(P<0.05或P<0.01)。NO血压和HR的变化无相关性(P>0.05)。结果表明,ET、TXB2和6-keto-PGF1α共同参与了XFM引起的小型猪血流动力学变化过程,是引起小型猪血压发生变化的主要原因之一。  相似文献

为研究鹿复合麻醉剂作用下血浆中一氧化氮(NO)、内皮素(ET)和降钙素基因相关肽(CGRP)含量变化与血流动力学变化的相关性,探讨鹿复合麻醉剂引起鹿血流动力学变化的部分机理。试验选取6只健康成年梅花鹿,肌肉注射鹿复合麻醉剂0.04 m L/kg体重,注药前及注药后15、30、45、60、75、90、120 min进行收缩压、舒张压、平均动脉压及心率的监测,并同步采集颈静脉血样测定NO、ET和CGRP含量。结果表明,麻醉期鹿血浆ET变化与收缩压(SBP)、舒张压(DBP)、平均动脉压(MAP)和心率(HR)变化趋势大致相似,存在着一定相关性(P0.01);CGRP变化与上述指标变化趋势相反,存在一定负相关性(P0.01);NO变化不显著(P0.05)。结果提示,鹿复合麻醉剂作用下引起ET和CGRP含量的变化参与了鹿血流动力学的调节。  相似文献

实验一选择9只健康、6-10月龄雄性白山羊,经粪便学检查和Dot-ELISA检测,确认无肝片吸虫感染,随机分成感染组(n=5)和对照组(n=4),试验组每只一次口服接种150个肝片吸虫囊蚴,每周定时从颈静脉采集感染前(0周)和感染后23周山羊血液一次,分离血浆,实验二将60只大鼠随机分成感染组(n=30)和对照组(n=30),感染组大鼠一次口服25个囊蚴,对照组不感染,于感染前(0周)和感染后(1,3,5,7,9周)采集血样,分别测定试验组和对照组羊和鼠花生四烯酸代谢物6-酮-前列腺素F1α(6-keto-PGF1α)和血栓素B2(TXB2)的变化。结果表明肝片吸虫感染后,山羊血浆中TXB2水平变化,在1-3周与对照组相比无显著差异,从第5周开始到实验结束,试验组血浆中TXB2水平均显著或极显著地高于对照组,大鼠血栓素水平在感染后前5周逐步下降,以后急剧上升,至实验结束时显著高于对照组;而整个实验期间,两种动物血浆中6-keto-PGF1α水平变化不显著,提示肝片吸虫感染后,机体发生多种功能的改变,花生四烯酸代谢物参与了肝片吸虫病的发展过程。  相似文献

选取经传染性法氏囊病病毒(IBDV)弱毒苗免疫和未免疫的AA 鸡120 羽,研究了感染IBDV 强毒后血浆肿瘤坏死因子-α(TNF-α)和花生四烯酸代谢物水平的动态变化及相互关系。结果表明,攻毒后第1 天,未免疫攻毒鸡(A 组)血浆TNF-α即表现较大的变化,至第3 天,A 组和免疫攻毒鸡(B组)TNF-α显著高于未免疫未攻毒鸡(C组),至第5 天3 个组虽均下降,但A 组仍高于或明显高于B、C组,以后3 组间无明显差异;攻毒后的3~5 d,A 组血浆前列腺素E(PGE2 )上升或明显上升,高于或明显高于B、C组,B组虽也见上升但与C组无明显差异,5 d 后3 组均下降,同期各组相比无明显差异;血栓素B2 (TXB2 )除在攻毒后第3 天C组明显高于A、B组以及攻毒前水平外,其他时间各组间无显著差异;A、B组的6-酮-前列腺素F1α(6-keto-PGF1α)在攻毒后头5 d 内升高,但组间无差异。分析3 组TNF-α与几种花生四烯酸代谢物水平间的相关性发现,仅A 组的TNF-α分别与PGE2 、TXB2 和6-keto-PGF1α呈显著正相关。  相似文献

为探讨山豆根多糖(SSP)对体外培养的鸡脾脏淋巴细胞信号分子的影响,采用终浓度为50、100、200、400μg/mL的山豆根多糖分4、8、12、24 h 4个时间段刺激培养鸡脾脏淋巴细胞,测定培养上清或细胞内环磷酸腺苷(cAMP)、环磷酸鸟苷(cGMP)、6-酮-前列腺素F1α(6-keto-PGF1α)、血栓素B2(TXB2)和一氧化氮(NO)信号转导分子水平。结果表明,山豆根多糖在终浓度为100μg/mL时与鸡脾脏淋巴细胞共同培养8 h后能显著升高cAMP(P0.01)、6-keto-PGF1α(P0.05)、TXB2(P0.01)水平,4种浓度的山豆根多糖对NO分子水平随时间的变化有不同程度的升高(P0.05),影响cAMP/cGMP和6-keto-PGF1α/TXB2信号体系。提示山豆根多糖可通过影响免疫细胞内的信号转导而影响机体的免疫功能。  相似文献

本研究旨在探讨围产期窒息时内皮素( ET)和一氧化氮 ( NO)的变化与急性胃黏膜损伤的关系及 NO的前体 L-精氨酸 ( L-Arg)和NO合成酶抑制剂 L-硝基 -精氨酸甲酯 ( L-NAME)在胃黏膜损伤中的作用。结果显示窒息组血浆 ET和胃壁 NO水平显著高于正常对照组 ( p<0 .0 1 ) ,胃黏膜损伤明显 ;L -Arg组血浆ET水平较窒息组显著下降 ( p <0 .0 1 ) ,血浆和胃壁 NO含量则显著升高 ( p <0 .0 1 ) ,胃黏膜损伤减轻 ;L-NAME组血浆 ET水平和 L I显著高于正常对照组和 L -Arg组 ( p <0 .0 1 ) ,血浆和胃壁 NO含量较 L-Arg组显著下降 ( p <0 .0 1 ) ,表明围产期窒息时 NO和 ET的变化与胃黏膜损伤的程度密切相关。NO在围产期窒息时有保护胃黏膜的作用 ,而 ET则作用相反。L -Arg的保护作用和 L -NAME的损伤作用均是通过 NO的变化来实现的。  相似文献

用结扎颈动脉和缺氧的方法建立一侧缺氧缺血性脑损伤(HIE)动物模型,观察脑活素对HIE大鼠脑组织损伤的治疗作用。结果发现,缺氧缺血后新生大鼠脑组织血栓素B2(TXB2)和6酮--前列腺素F1α(6-keto-PGF1α)含量、细胞凋亡数及对侧脑组织含水量均显著升高,表明新生大鼠缺氧缺血性脑损伤与脑血流改变、脑细胞凋亡及脑组织含水量的变化密切相关;缺氧缺血大鼠应用脑活素治疗后,其脑组织TXB2和6-keto-PGF1α含量、细胞凋亡数及脑组织含水量均下降,表明脑活素通过改善脑血流、降低脑细胞凋亡及脑水肿程度来减轻缺氧缺血造成的损伤,对新生大鼠缺氧缺血性脑损伤发挥有效的治疗作用。  相似文献

探讨单纯隆朋麻醉和隆朋麻醉下手术对犬内皮素(ET)和降钙素基因相关肽(CGRP)分泌的影响。方法:选取健康犬12只,将其随机均分为单纯隆朋麻醉组和隆朋麻醉手术组。两组均于麻醉前、0.5 h、2 h、8 h、24 h、48 h、72 h和120 h采集静脉血,并迅速离心取血浆,用放射性免疫法检测ET和CGRP的浓度。结果:麻醉组,ET出现上升趋势而CGRP呈现下降趋势。ET到2 h时恢复至麻前水平,CGRP到8 h时恢复至麻前水平。手术组,手术开始后ET和CGRP都呈现上升趋势,两者都在24 h时恢复至正常水平。结论:隆朋单纯麻醉时对ET和CGRP有轻微的影响,但隆朋麻醉手术时ET和CGRP都出现上升趋势,机体的应激反应较强烈。  相似文献

为探究伊犁马对补喂不同水平α-酮异己酸的吸收及其某些相关代谢,采取单因素多水平试验设计方法,选取20匹年龄为1岁,体重(295.25±25.15)kg的伊犁马,分成4组,每组5匹。试验Ⅰ、Ⅱ、Ⅲ组分别补喂6、12、18 gα-酮异己酸,进行为期25 d的试验。在试验第25天采集血液样品。结果表明:(1)随着α-酮异己酸补喂水平的增加,血浆中α-酮异己酸浓度的变化呈先上升后下降的趋势。与对照组相比,补喂α-酮异己酸1 h后各试验组血浆α-酮异己酸浓度分别提高了11.2%、7.86%和21.51%,且均达到了峰值(P>0.05);(2)随着α-酮异己酸补喂水平的增加,血浆中β-羟基-β-甲基丁酸浓度的变化呈先下降后上升再下降的趋势。与对照组相比,各试验组补喂前0 h血浆β-羟基-β-甲基丁酸浓度呈下降趋势,补喂后1、2、4 h血浆β-羟基-β-甲基丁酸浓度均有所升高(P>0.05);(3)与对照组相比,各试验组血浆α-酮异己酸和β-羟基-β-甲基丁酸浓度均差异不显著(P>0.05);试验Ⅱ组和试验Ⅲ组血浆亮氨酸浓度分别提高了22.40%和22.24%(P>0.05);试验Ⅰ组和试验Ⅲ组血浆异亮氨酸浓度极显著降低(P<0.01);血浆缬氨酸浓度显著降低(P<0.05);试验Ⅲ组血浆赖氨酸、甲硫氨酸、精氨酸和谷氨酰胺浓度均显著降低(P<0.05);各试验组血浆5-羟色胺浓度极显著降低(P<0.01);试验Ⅲ组血浆三甲基组氨酸浓度降低了18.74%(P>0.05)。补喂α-酮异己酸提高了伊犁马血浆α-酮异己酸浓度,且各试验组均在补喂后1 h达到峰值;提高了血浆β-羟基-β-甲基丁酸和亮氨酸的浓度。通过补喂不同水平的α-酮异己酸,改变了伊犁马对某些氨基酸的吸收利用。  相似文献

Porcine circovirus type 2 (PCV2) is considered as the causative agent of postweaning multisystemic wasting syndrome (PMWS) in domestic pigs, where the virus is ubiquitous as evidenced by serological surveys. We present the results of the first nationwide sero-survey on the presence of PCV2 antibodies in European wild boars, and report the first PMWS case in a wild boar from Spain. Sera from 656 hunter harvested wild boars from 45 different geographical sites and 22 additional imported animals were analysed by means of an immunoperoxidase monolayer assay (IPMA). We also examined the tissues from 55 healthy and one diseased wild boars for the presence of PCV2 nucleic acid and PMWS lesions by in situ hybridisation and histopathology, respectively. Additionally, abundance estimates of wild boars and field interviews were carried out on 30 sampling sites. The prevalence of medium to high PCV2 serological titres among the examined wild boars was 47.89 +/- 1.9%. Seropositive wild boars appeared in all but one of the geographical regions analysed. Seroprevalence and titre of PCV2 antibodies were closely related to the management of the wild boar populations. Wild boars from intensively managed, farm-like populations had higher prevalence than wild boars living in more natural situations. The effect of wild boar abundance and management on PCV2 antibody prevalence was further evidenced by the high correlation existing between the relative abundance estimates of animals and the percentage of wild boars with medium to high levels of PCV2 antibodies. PCV2 nucleic acid was detected in the tissues of three wild boars. One of these was diagnosed as PMWS. The results, in addition to information on piglet mortalities, suggest a potential role of PMWS in piglet mortality in intensively managed wild boar populations.  相似文献

为了研究杂交野猪的生长性能、繁育水平与养殖环境、运动量、饲料配制的关系,根据野猪生活习性,对杂交野猪放养区内生活环境进行模拟(包括野猪巢穴、生态环境、植被),使杂交野猪能够有更多的运动范围和获取野外食物的机会。结果表明,放养模式下的杂交野猪产仔率(P<0.05)、仔猪存活率(P<0.01)均高于圈养模式杂交野猪,但增重速度显著(P<0.05)低于圈养模式下饲养杂交野猪。由试验结果可以得出,圈养模式下杂交育肥野猪增重速度显著高于放养杂交野猪,饲养管理难度较低,适合集约化大规模饲养。  相似文献

The aim of the present study was to determine the effects of dietary supplements of vitamins on vitamin status, libido, and semen characteristics in young boars under normal and intensive semen collection. Sixty Landrace, Yorkshire, and Duroc boars were allocated randomly from 6 to 10 mo of age to one of the following diets: 1) basal diet (industry level) for minerals and vitamins (Control, n = 15); 2) basal diet supplemented with vitamin C (ASC, n = 15); 3) basal diet supplemented with fat-soluble vitamins (FSV, n = 15); and 4) basal diet supplemented with water-soluble vitamins (WSV, n = 15). After puberty (approximately 12 mo of age), semen was collected at a regular frequency (three times every 2 wk) for 5 wk. Thereafter, all boars were intensively collected (daily during 2 wk). A recovery period (semen collection three times every 2 wk) followed and lasted for 10 wk. Sperm quality (percentage of motile cells and percentage of morphologically normal cells) and quantity (sperm concentration, semen volume, and total sperm number) were recorded as well as direct and hormone related measurements of boar libido. Blood and seminal plasma samples were taken to monitor vitamin status. High concentrations of B6 (P < 0.05) and folic acid (P < 0.05) were observed in the blood plasma of WSV boars, whereas greater concentrations of vitamin E (P < 0.01) were obtained in FSV boars. In the seminal plasma, folic acid concentrations tended to be greater in WSV boars (P < 0.08). During the intensive collection period, there was a tendency (P < 0.06) for semen production to be greater in WSV boars, the effect being less pronounced (P < 0.10) in FSV boars. During the recovery period, the percentage of motile sperm cells was greater in WSV boars (P < 0.03) and, to a lesser extent, in FSV boars (P < 0.10) compared with Control boars. Sperm morphology and libido were not affected by treatments. These results indicate that the transfer of vitamins from blood to seminal plasma is limited and the dietary supplements of water-soluble and fat-soluble vitamins may increase semen production during intensive semen collection.  相似文献

Thirteen crossbred boars were immunized at 1 mo of age against either testosterone-3-oxime-equine serum albumin (treated boars) or equine serum albumin (control boars) to test the hypothesis that active immunization against testosterone stimulates testicular growth and development in the prepubertal boar. All boars were injected with the appropriate antigen at 2, 3, 4, 5 and 6 mo of age and were slaughtered at 14 mo of age. Active immunization against testosterone resulted in an increase (P less than .05) in tritiated-testosterone binding by plasma within 60 d after the primary immunization; the degree of binding decreased by 6 mo but remained elevated (P less than .05) relative to controls through 12 mo of age. There was no effect of treatment on body weights through 12 mo of age. Concentrations of testosterone in plasma were higher (P less than .05) in testosterone-immunized boars than in controls; this increase was likely due to antibody binding rather than increased testosterone secretion because (1) concentrations of androgen in testicular parenchyma at slaughter were not altered by treatment and (2) plasma concentrations of estrogens were generally not affected by treatment. Concentrations of luteinizing hormone (LH) and follicle stimulating hormone (FSH) were markedly suppressed in testosterone-immunized boars during the time when concentrations of these gonadotropins were high in control boars (greater than 3 mo of age). In spite of suppression of average LH and FSH concentrations, testicular weights, daily sperm production rates and seminal characteristics were similar for the two groups of boars at slaughter. In the present study, we used morphological and genetic analyzes to distinguish bones of domestic boars from those of wild boars. We analyzed 65 Sus bones (cranium, mandible and teeth) stored in three research institutes in Vietnam and in a village in Vietnam. Based on comparison of bucco‐lingual measurements of mandibular parts, the 58 specimens were morphologically classified into two size groups: a large bone group and a small bone group. Analysis of 572‐bp mitochondrial DNA (mtDNA) sequences indicated that the large bones had genetic links to wild boar lineage including Ryukyu, Taiwan and Korean wild boars, and that the small bone group was closely related to East Asian domestic pigs. The phylogenetic analysis and parsimonious networks constructed among mtDNA haplotypes belonging to Ryukyu wild boar lineage showed that the Ryukyu wild boar is closely related to the Vietnamese wild boars, and uniquely miniaturized on their islands after the Ryukyu archipelago became isolated from the Asian continent.  相似文献

The prevalence of Lawsonia intracellularis between wild boar (Sus scrofa) and fallow deer (Dama dama) bred in one game reserve was investigated using the nested PCR method. In the study, 88 clinically healthy wild boars of different age categories and two fallow deer bagged in the game reserve were examined. Lawsonia intracellularis was demonstrated in the mucous membrane of the intestine of eight (9.1%) wild boars and one fallow deer. Of the nine wild boar whose tissues of corresponding lymph nodes were examined in addition to the mucous membrane of the ileum, one tested positive for the microorganism. A relationship between the occurrence of L. intracellularis and age of wild boar was demonstrated. Because wild boar and fallow deer are bred together in one game reserve, the possibility of inter-species transfer of L. intracellularis should be borne in mind.  相似文献

Twelve male neonatal calves (39 to 50 kg) were allotted to 3 groups of 4 calves each. All calves were anesthetized with halothane, and then Escherichia coli endotoxin was given intravenously (3 times) and intraperitoneally (3 times) during a 6-hour period. Group-1 calves were untreated, group-2 calves were pretreated with a low dose of flunixin meglumine (1.1 mg/kg of body weight), and group-3 calves were pretreated with a high dose of flunixin meglumine (4.4 mg/kg). In calves of group 1, the mean systemic arterial blood pressure (MABP) and cardiac output (CO) decreased, but pulmonary arterial pressure increased after the initial intravenous and intraperitoneal injections of endotoxin. In calves of this group, these changes were accompanied by increased plasma thromboxane B2 (TxB2) concentration. During this period, increased plasma TxB2 concentration or hemodynamic changes were not detected in calves of groups 2 and 3. Only calves of group 1 had altered hemodynamics early in the experiment; however, after 6 hours, calves of all 3 groups had similarly decreased CO and MABP. In calves of the untreated group, plasma 6-keto-prostaglandin (PG)F1 alpha concentration increased steadily from the beginning of the experiment until 3 hours later. The CO and MABP were low at the time when serum 6-keto-PGF1 alpha concentration was high; however, these 2 measurements also were low in treated calves who did not have correspondingly high plasma 6-keto-PGF1 alpha concentration. Serum samples from 1,011 wild boars hunted in 2012 were collected for serological surveillance for 4 subtypes (pandemic A (H1N1) 2009 and classical H1N1, H1N2 and H3N2) of swine influenza virus (SIV). Samples from 12 of the boars were identified as positive for SIV (pandemic A (H1N1) 2009, n=9; classical H1N1, n=2; and H1N2, n=1) by a hemagglutination inhibition test (HI test) and a nucleoprotein (NP)-based enzyme-linked immunosorbent assay (NP-ELISA). Although the overall seroprevalence of SIV in the Korean wild boar population was somewhat low compared with that in China and the U.S.A., the apparent prevalence of pandemic H1N1 was notable. Therefore, continuous monitoring of the wild boar population is needed as it may be a major reservoir for pandemic H1N1, facilitating its spread to humans and domestic pigs.  相似文献

BackgroundAfrican swine fever (ASF) is a hemorrhagic fever occurring in wild boars (Sus scrofa) and domestic pigs. The epidemic situation of ASF in South Korean wild boars has increased the risk of ASF in domestic pig farms. Although basic reproduction number (R0) can be applied for control policies, it is challenging to estimate the R0 for ASF in wild boars due to surveillance bias, lack of wild boar population data, and the effect of ASF-positive wild boar carcass on disease dynamics.ObjectivesThis study was undertaken to estimate the R0 of ASF in wild boars in South Korea, and subsequently analyze the spatiotemporal heterogeneity.MethodsWe detected the local transmission clusters using the spatiotemporal clustering algorithm, which was modified to incorporate the effect of ASF-positive wild boar carcass. With the assumption of exponential growth, R0 was estimated for each cluster. The temporal change of the estimates and its association with the habitat suitability of wild boar were analyzed.ResultsTotally, 22 local transmission clusters were detected, showing seasonal patterns occurring in winter and spring. Mean value of R0 of each cluster was 1.54. The estimates showed a temporal increasing trend and positive association with habitat suitability of wild boar.ConclusionsThe disease dynamics among wild boars seems to have worsened over time. Thus, in areas with a high elevation and suitable for wild boars, practical methods need to be contrived to ratify the control policies for wild boars.  相似文献

本试验旨在研究骨桥蛋白(osteopontin,OPN)在长白公猪生殖细胞中的表达和定位,并探究其与公猪繁殖性能的相关性。试验采集了长白公猪精液和不同阶段(3日龄、3月龄、6月龄和12月龄)的睾丸组织,通过蛋白印迹的方法检测OPN蛋白在精液和不同月龄睾丸中的表达,通过免疫组化的方法对OPN蛋白在公猪睾丸细胞中进行定位;同时,根据配种胎次≥ 20胎,3次配种公猪为同一头的标准,筛选并采集17头长白种公猪精液,统计相对应的1 388头母猪的生产成绩,计算得到公猪繁殖性能指标(包括窝产总仔数、窝产活仔数、分娩率和繁殖力)。低温离心精液分离得到精子和精浆,丙酮法提取精浆蛋白,Lysis buffer方法提取精子蛋白,最后运用BCA和ELISA的方法检测精子和精浆中OPN蛋白的含量,分析OPN蛋白与公猪繁殖性能的相关性。蛋白印迹结果显示,OPN在精子、精浆和各月龄阶段的长白公猪睾丸中均以两种形式表达(67.4和33.7 ku),且67.4 ku的形式在3月龄公猪睾丸中表达量最高;免疫组化的结果显示,OPN在长白公猪的初级精母细胞、次级精母细胞和精子细胞中表达,在精母细胞、支持细胞和间质细胞中无表达;BCA和ELISA结果显示,精子中的OPN蛋白含量是精浆中的7倍(P<0.05),精液中的OPN蛋白与公猪窝产活仔数显著正相关(P<0.05)。本试验结果表明,OPN在各阶段的长白公猪睾丸中都有表达,且在精子和精浆中也有表达,这可能与公猪的繁殖性能有关,从而为后期OPN蛋白在公猪受精力的研究奠定基础。  相似文献
