Fusarium culmorum is a phytopathogenic, toxigenic fungus causing seedling diseases, foot rot and head blight of cereals. For estimating competition effects in mixtures of two single-spore isolates, two winter rye single crosses were tested with either four isolates individually or four 1 : 1 mixtures of the same isolates in six field environments. Two isolates (FC46, FC64) were highly aggressive deoxynivalenol (DON) and 3-acetyl DON-producers, the other two (FC30, FC71) were medium aggressive nivalenol-producers. Rye heads were inoculated during flowering with conidia of pairs of isolates expressing similar (FC46 + FC64, FC30 + FC71) or contrary (FC46 + FC71, FC30 + FC64) levels of aggressiveness and similar or different concentrations and chemotypes of mycotoxins, respectively. Head blight rating and yield components relative to the non-inoculated plots were recorded as aggressiveness traits. Additionally, mycotoxin concentrations were measured in the rye grain. Random pathogen samples were re-isolated from heads at the onset of symptom development and analysed by molecular markers (RAPD–PCR) in one environment. Aggressiveness of the isolate mixtures was significantly lower than that of the isolates applied individually on both rye genotypes. Similarly, mycotoxin concentrations were significantly lower in the mixtures in seven out of eleven comparisons. Among the re-isolates, the component genotypes of a mixture significantly deviated from the inoculated 1 : 1 ratio when a particular isolate (FC46) was present in the mixture. This isolate displayed a superior competitive ability irrespective of the aggressiveness or mycotoxin profile of the mixing partner illustrating that pathogenic fitness is caused by additional factors that have not, as yet, been identified.  相似文献

Vegetative compatibility among 17 isolates ofVerticillium dahliae obtained from watermelon, originating from eight regions of Greece, was investigated using complementation tests between nitrate-nonutilizing(nit) mutants. Among 529 chlorate-resistant sectors obtained, only 107 werenit mutants. These mutants were paired with tester strains (from Greece and other countries) of previously described vegetative compatibility groups (VCGs), and also were paired in many combinations among themselves. All isolates were self-compatible. Sixteen isolates were found to belong to VCG2. Only isolate V75 could not be assigned to a VCG, because the threenit mutants obtained from it showed negative reactions with the tester strains of four VCGs and with complementary mutants from other isolates. Based on this sample, we conclude that the population ofV. dahliae from watermelon in Greece is homogeneous in respect to VCG.  相似文献

The Fusarium species predominantly found associated with Fusarium head blight (FHB) in wheat and other small-grain cereals all over Europe are F. graminearum, F. avenaceum and F. culmorum. Among the less frequently encountered species are several others which are less pathogenic or opportunistic, but also toxigenic. These include F. poae, F. cerealis F. equiseti F. sporotrichioides F. tricinctum and, to a lesser extent, F. acuminatum F. subglutinans F. solani F. oxysporum F. verticillioides F. semitectum and F. proliferatum. The species profile of FHB is due to several factors, primarily climatic conditions, particularly rain and the temperature at flowering stage, but also agronomic factors, such as soil cultivation, nitrogen fertilization, fungicides, crop rotation, and host genotype. The most frequently encountered Fusarium mycotoxins in FHB in Europe has proved to be deoxynivalenol and zearalenone produced by F. graminearum and F. culmorum with the former more common in southern (warmer) and the latter in northern (colder) European areas. Nivalenol was usually found associated with deoxynivalenol and its derivatives (mono-acetyldeoxynivalenols), together with fusarenone-X, formed by F. graminearum F. cerealis F. culmorum and, in northern areas, by F. poae. Moreover, from central to northern European countries, moniliformin has been consistently reported, as a consequence of the widespread distribution of F. avenaceum whereas the occurrence of T-2 toxin derivatives, such as T-2 toxin and HT-2 toxin, and diacetoxyscirpenol have been recorded in conjunction with sporadic epidemics of F. sporotrichioides and F. poae. Finally, beauvericin and various enniatins have recently been found in Finnish wheat colonized by F.avenaceum and F. poae.  相似文献

Gibberella zeae (anamorph Fusarium graminearum ) causes head blight of cereals and contaminates grains with mycotoxins such as deoxynivalenol (DON). To determine the correlations among aggressiveness traits, fungal colonization and DON production, 50 progeny from a segregating population of G. zeae were inoculated onto a susceptible winter wheat cultivar in three field environments (year–location combinations). Aggressiveness traits were measured as head-blight rating and plot yield relative to noninoculated plots. Fungal colonization, measured as Fusarium exoantigen (ExAg) content, and DON production were analysed with two ELISA formats. Disease severity was moderate to high based on head-blight rating and relative plot yield. Fusarium ExAg content and DON production ranged from 0·26–1·41 units and from 4·18–43·70 mg kg−1, respectively. Significant ( P  = 0·01) genotypic variation was found for all traits. Heritability for Fusarium ExAg content was rather low because of high progeny–environment interaction and error. DON/ Fusarium ExAg ratio did not vary significantly ( P  > 0·1) among progeny. Correlation between DON production and Fusarium ExAg content across environments was high ( r  = 0·8, P  = 0·01), but no covariation existed between aggressiveness traits and DON/ Fusarium ExAg content ratio.  相似文献

Details of our long-term research programme concerning the epidemiology of Fusarium spp. and mycotoxin production are summarized. Evaluation of the occurrence of Fusarium spp., mainly on winter wheat (Triticum aestivum), was carried out by investigating Fusarium infection and mycotoxin contamination. Two to 15% of grains were infested during 1995–1998 at three climatologically differing localities of the Rhineland, Germany. Disease progress was accelerated by rainfall during the flowering season. The species most frequently isolated were Fusarium avenaceum, F. poae, F. culmorum and F. graminearum. The mean deoxynivalenol (DON) content varied from 19gkg–1 (1995) to 310gkg–1 (1998) and was not always correlated with disease severity. Organic farming systems showed lower rates of infection with ear blight and lower mycotoxin contamination than conventional farming systems.  相似文献

Summary An infection of bread wheat by fusarium head blight contaminates the crop with mycotoxins, particularly deoxynivalenol (DON) and nivalenol (NIV). The toxicity and natural occurrence of these mycotoxins in wheat are reviewed. Based on 8 years data of fusarium head blight epidemics of wheat in the Netherlands, DON contamination of the grain was estimated. Fusarium head blight ratings averaged an infection of 1.7% of all spikelets; estimates for DON contamination averaged 0.9 mg kg–1. Taking a guideline level for DON in uncleaned bread wheat of 2 mg kg–1, in 1979 and 1982 a wheat crop was produced with estimated DON concentrations above the limit of tolerance. Human and animal exposure to mycotoxins in the Netherlands appears to be small but chronic. The information presented in this paper illustrates the need for an annual evaluation of the crop for fusarium head blight incidence and mycotoxin content, and the necessity of fusarium head blight resistant wheat cultivars.Samenvatting Aaraantasting van tarwe doorFusarium culmorum enFusarium graminearum leidt tot vorming van mycotoxinen in het graan, waarvan deoxynivalenol (DON) en nivalenol (NIV) de belangrijkste toxinen zijn. In dit artikel wordt een overzicht gegeven van de toxicologische aspecten, en het voorkomen van deze toxinen in tarwe. Informatie over DON en NIV in tarwe in West-Europa is schaars. Gebaseerd op gegevens vanFusarium epidemieën in de jaren 1979–1986 wordt een schatting gegeven van de concentratie DON in Nederlandse tarwe. Rekening houdend met de herkomst en verwerking van tarwe, blijken zowel in dierlijk als menselijk voedsel lage concentraties DON chronisch voor te komen. Op basis van een maximaal toelaatbare dagelijkse dosis DON van 3 g kg–1 lichaamsgewicht is de schatting van de dagelijkse opname van DON in het jaar volgend op de oogst van 1982 net op de grens. An in vitro detached leaf assay, involving the inoculation of detached leaves with Microdochium nivale, was further developed and used to compare with whole plant resistance ratings to Fusarium head blight (FHB) of 22 commercial cultivars and published information on 21 wheat genotypes, identified as potential sources for FHB resistance. An incubation temperature of 10 °C and isolates of M. nivale var. majus of intermediate pathogenicity were found to be the most suitable for the differential expression of several components of partial disease resistance (PDR), namely incubation period, latent period and lesion length, in wheat genotypes used in the detached leaf assay. There were highly significant differences (P < 0.001) for each component of PDR within commercial cultivars and CIMMYT genotypes. Positive correlations were found between incubation period and latent period (r = 0.606; P < 0.001 and r = 0.498; P < 0.001, respectively, for commercial cultivars and CIMMYT genotypes), inverse correlations between incubation period and lesion length (r = -0.466; P < 0.01 and r = –0.685; P < 0.001, respectively) and latent period and lesion length (r = –0.825; P < 0.001 and r = –0.848; P < 0.001, respectively). Spearman rank correlations between individual PDR components and UK 2003 recommended list ratings were significant for incubation period (rs = 0.53; P < 0.05) and latent period (rs = 0.70; P < 0.01) but not for lesion length (r s = –0.26). Commercial cultivars identified with high resistances across all three PDR components in the detached leaf assay also had high whole plant FHB resistance ratings, with the exception of cv. Tanker which is more susceptible than the results of the detached leaf assay suggested, indicating an additional susceptibility factor could be present. Agreement between resistances found in the detached leaf assay and resistance to FHB suggests resistances detected in detached leaves are under the same genetic control as much of the resistances expressed in the wheat head of the commercial cultivars evaluated. In contrast, high resistances in each of the PDR components were associated with higher susceptibility across 19 CIMMYT genotypes previously evaluated as potential breeding sources of FHB resistance (incubation period: r = 0.52; P < 0.01, latent period: r = 0.53; P < 0.01, lesion length: r = –0.49; P < 0.01). In particular, the CIMMYT genotypes E2 and E12 together with Summai #3, known to have high levels of whole plant FHB resistance, showed low levels of resistance in each PDR component in the detached leaf assay. Such whole plant resistances, which are highly effective and not detected by the detached leaf assay, do not appear to be present in Irish and UK commercial cultivars. The most resistant Irish and UK commercial cultivars were comparable to the genotype Frontana and the most resistant CIMMYT germplasm evaluated in the leaf assay.  相似文献

Interactions between Barley yellow dwarf virus (BYDV) and Fusarium species causing Fusarium head blight (FHB) in winter wheat cvs Agent (susceptible to FHB) and Petrus (moderately resistant to FHB) were studied over three years (2001–2003) in outdoor pot experiments. FHB developed more rapidly in cv. Agent than in cv. Petrus. The spread of FHB was greater in BYDV-infected plants than in BYDV-free plants. Thousand grain weight (TGW) was reduced more in Fusarium-infected heads of cv. Agent than in cv. Petrus. A highly significant negative correlation was found between disease index and TGW in cv. Agent (r = −0.916), while in cv. Petrus the correlation was less significant (r = −0.765). Virus infection reduced TGW in cv. Petrus more than in cv. Agent. In plants with both infections, TGW reductions in cv. Petrus corresponded to those of BYDV infection, and in cv. Agent TGW was more diminished than in BYDV infection. Effects of different treatments determined over three years on ergosterol contents in grain were generally similar to effects on disease indices. Grain weight per ear and ear weight of the different treatments of both cultivars largely corresponded with the TGW results. Deoxynivalenol (DON) content in grain of cv. Agent infected with Fusarium spp. was 11–25 times higher compared to the corresponding treatments in cv. Petrus. The DON content in grain of plants of the two cultivars infected with both pathogens was higher than that of plants infected only with Fusarium over the three years.  相似文献

Inoculation experiments with 14 genotypes of oats (10 cultivars and 4 lines) were performed during 1996, 1997 and 1998 in Sitaniec, South-Eastern Poland. Panicles of oats were inoculated with a conidial suspension of Fusarium avenaceum, which caused a reduction in yield by 33% and in 1000 kernel weight (TKW) by 21%. During the period between inoculation and harvest, F. avenaceum was able to accumulate moniliformin (MON) in kernels at an average level of 0.13mgkg–1 (gg–1). The highest reduction of yield components caused by the F. avenaceum inoculation was found for cv. Santor, followed by lines CHD 1171, STH 2795 and cvs: Kwant and Farys, while cvs Slawko, Dukat, Borys and Komes exhibited the highest resistance to the disease in terms of TKW and yield reductions after inoculation.  相似文献

为研究小麦UDP-葡萄糖基转移酶7(UDP-glycosyltransferase 7,TaUGT7)的抗赤霉病功能,利用DNAMAN 6.0软件对Ta UGT7及其同源蛋白进行序列比对,应用实时荧光定量PCR(quantitative real-time PCR,qRT-PCR)技术分析经赤霉菌Fusarium graminearum和脱氧雪腐镰刀菌烯醇(deoxynivalenol,DON)处理后的苏麦3号小穗中TaUGT7基因的表达特征,利用基因枪在洋葱表皮细胞瞬时表达TaUGT7-eGFP进行亚细胞定位,采用农杆菌介导法在小麦品种Fielder中过量表达TaUGT7基因并进行赤霉病抗性鉴定。结果表明,TaUGT7在氨基酸序列上与已知赤霉病抗性相关UGT相似性较低;TaUGT7在赤霉菌接种24 h后开始被诱导表达,在DON处理2 h后逐步被诱导表达;Ta UGT7蛋白亚细胞定位于细胞膜和细胞核中;qRT-PCR检测发现,TaUGT7在8株独立的过表达转基因株系中均有不同程度的上调表达;与野生型对照相比,过表达株系TaUGT7-395和TaUGT7-457中的平均病小穗率显著下降。...  相似文献

Glasshouse studies were undertaken to determine if fungicides used for the control of Fusarium head blight (FHB) result in elevated concentrations of the trichothecene mycotoxin, deoxynivalenol (DON) in harvested wheat grain. Metconazole and azoxystrobin, at double, full, half or quarter the manufacturer's recommended dose rate, were applied to ears of wheat (cv. Cadenza), artificially inoculated with conidia of either Fusarium culmorum or F. graminearum. Metconazole demonstrated high activity against both pathogens, reducing significantly the severity of FHB and the DON concentrations at each of the four dose rates tested when compared to untreated controls. Applications of azoxystrobin significantly reduced FHB and DON compared to unsprayed controls. However, their effectiveness was significantly less than that of metconazole and no dose rate response was observed. Quantification of the amount of trichothecene-producing Fusarium present in harvested grain was determined using a competitive PCR assay based on primers derived from the trichodiene synthase gene (Tri5). Simple linear regression analyses revealed strong relationships between the amount of trichothecene-producing Fusarium present in grain and the DON concentrations (r 2=0.72–0.97). It is concluded that fungicides, applied for the control of FHB, affect DON concentrations indirectly by influencing the amount of trichothecene-producing Fusarium species present in wheat grain. There was no evidence that fungicide applications directly increase the concentration of DON in grain.  相似文献

Several Fusarium species occurring worldwide on maize as causal agents of ear rot, are capable of producing mycotoxins in infected kernels, some of which have a notable impact on human and animal health. The main groups of Fusarium toxins commonly found are: trichothecenes, zearalenones, fumonisins, and moniliformin. In addition, beauvericin and fusaproliferin have been found in Fusarium-infected maize ears. Zearalenone and deoxynivalenol are commonly found in maize red ear rot, which is essentially caused by species of the Discolour section, particularly F. graminearum. Moreover, nivalenol and fusarenone-X were often found associated with the occasional occurrence of F. cerealis, and diacetoxyscirpenol and T-2 toxin with the occurrence of F. poae and F. sporotrichioides, respectively. In addition, the occurrence of F. avenaceum and F. subglutinans usually led to the accumulation of moniliformin. In maize pink ear rot, which is mainly caused by F. verticillioides, there is increasing evidence of the wide occurrence of fumonisin B1. This carcinogenic toxin is usually found in association with moniliformin, beauvericin, and fusaproliferin, both in central Europe due to the co-occurrence of F. subglutinans, and in southern Europe where the spread of F. verticillioides is reinforced by the widespread presence of F. proliferatum capable of producing fumonisin B1, moniliformin, beauvericin, and fusaproliferin.  相似文献

为明确镰刀菌毒素在我国不同小麦产区的污染与分布,利用超高效液相色谱-串联质谱仪对全国214个小麦品种籽粒中的7种镰刀菌毒素含量进行了测定和比较分析。结果表明:7种镰刀菌毒素的平均回收率在95.02%~116.52%之间,相对标准差为6.32%~21.50%。所有测试小麦样品均被镰刀菌毒素污染,7种毒素中以雪腐镰刀菌烯醇(nivalenol,NIV)含量最高,即使在NIV含量最低的长江中下游冬麦区,平均含量也高达182.56μg/kg,虽然目前并没有关于NIV的相关限量标准,但也应该予以足够重视;脱氧雪腐镰刀菌烯醇(deoxynivalenol,DON)含量较高,在8.07~337.23μg/kg之间,低于国家限量标准1 000μg/kg;T-2含量在4.05~78.05μg/kg之间,低于现有国际限量标准100μg/kg;玉米赤霉烯酮(zearalmone,ZEA)含量在5.30~12.50μg/kg之间,低于国家限量标准60μg/kg;其它3种毒素含量较低,均低于17.73μg/kg。不同麦区间毒素比较发现,DON和3-乙酰脱氧雪腐镰刀菌烯醇的含量在长江中下游冬麦区最高,分别为43.09μg/kg和6.39μg/kg,且显著高于其它麦区;来自长江中下游冬麦组和黄淮冬麦区南片水地组的小麦品种籽粒中毒素T-2和HT-2的含量显著高于其它麦区;其余毒素在不同麦区间均无显著差异。  相似文献

Genetic subdivision of Fusarium asiaticum was investigated using a collection of 478 isolates originating from the Kyushu area and Aichi Prefecture, Japan and Zhejiang Province in China. Trichothecene-type determination by a multiplex PCR-test indicated that all isolates were either of a nivalenol (NIV) or a 3-acetyl deoxynivalenol (3ADON) type. The 15-acetyl deoxynivalenol (15ADON) type was not detected in this collection. Based on a Bayesian model-based clustering method using allele data obtained with 11 variable number of tandem repeats (VNTR) markers, we detected three genetic clusters. The majority of isolates in the clusters were NIV isolates from both Japan and China, Japanese 3ADON and Chinese 3ADON isolates, respectively. High levels of fixation indices and low levels of effective number of migrants were observed between the genetic clusters. Data was re-analyzed by classifying the isolates into six groups according to trichothecene type and geographic location. Population analyses of these re-classified groups indicated that the genetic subdivisions of F. asiaticum were correlated with both trichothecene type and geographic differences. Different sets of wheat genotypes were tested under field conditions by spraying inocula of isolates of seven Fusarium spp. and Microdochium nivale (formerly F. nivale) in the period 1998–2002. The severity of Fusarium head blight (FHB), Fusarium-damaged kernels (FDK), the yield reduction and the deoxynivalenol (DON) contamination were also measured to describe the nature of the resistance. The degrees of FHB severity of genotypes to F. graminearum, F. culmorum, F. avenaceum, F. sporotrichioides, F. poae, F.␣verticillioides, F. sambucinum and M. nivale were very similar, indicating that the resistance to F.␣graminearum was similar to that for other Fusarium spp. listed. This is an important message to breeders as the resistance relates not only to any particular isolate of F. graminearum, but similarly to isolates of other Fusarium spp. This holds true for all the parameters measured. The DON contamination refers only to DON-producers F. graminearum and F. culmorum. Highly significant correlations were found between FHB, FDK, yield loss and DON contamination. Resistance components such as resistance to kernel infection, resistance to DON and tolerance were identified in the more susceptible genotypes. As compared with western European genotypes which produced up to 700 mg kg−1 DON, the Hungarian genotypes produced only 100 mg kg−1 at a similar FDK level. This research demonstrates the importance of measuring both FDK and DON in the breeding and selection of resistant germplasm and cultivars.  相似文献

In a field experiment between 2004 and 2006, 14 winter wheat varieties were inoculated with either a mixture of three isolates of F. poae or a mixture of three isolates of F. avenaceum. In a subsequent climate chamber experiment, the wheat variety Apogee was inoculated with individual single conidium isolates derived from the original poly conidium isolates used in the field. Disease symptoms on wheat heads were visually assessed, and the yield as well as the fungal incidence on harvested grains (field only) was determined. Furthermore, grains were analysed using LC-MS/MS to determine the content of Fusarium mycotoxins. In samples from field and climate chamber experiments, 60 to 4,860 μg kg−1 nivalenol and 2,400 to 17,000 μg kg−1 moniliformin were detected in grains infected with F. poae and F. avenaceum, respectively. Overall, isolate mixtures and individual isolates of F. avenaceum proved to be more pathogenic than those of F. poae, leading to a higher disease level, yield reductions up to 25%, and greater toxin contamination. For F. poae, all variables except for yield were strongly influenced by variety (field) and by isolate (climate chamber). For F. avenaceum, variety had a strong effect on all variables, but isolate effects on visual disease were not reflected in toxin production. Correlations between visual symptoms, fungal incidence, and toxin accumulation in grains are discussed.  相似文献

Severe rot of leaves, peduncles and flowers caused by Gibberella zeae (anamorph: Fusarium graminearum) was found on potted plants of hyacinth (Hyacinthus orientalis), a liliaceous ornamental, in greenhouses in Kagawa Prefecture, Japan, in January 2001. This disease was named “Fusarium rot of hyacinth” as a new disease because only the anamorph, F. graminearum, was identified on the diseased host plant. The authors contributed equally to this work. The fungal isolate and its nucleotide sequence data obtained in this study were deposited in the Genebank, National Institute of Agrobiological Sciences and the DDBJ/EMBL/GenBank databases under the accession numbers MAFF239499 and AB366161, respectively.  相似文献
