Five types of presumed ciliate sensory receptors were detected in the forebody papillae of the adult fish trematode, Crepidostomum metoecus (Braun, 1900). The cilia are short and submerged in a tegumental pit. The apical bulb part of all types of receptors observed is supported by a dense collar and connected to the tegument basal plasma membrane by a circular septate junction. In sensory receptors types I and III no rootlet is present; the bulbs of sensory receptors types III and IV contain an electron-dense formation.  相似文献

Two pairs of hitherto unknown multiciliate sensory endings have been demonstrated in free-swimming cercariae of Echinostoma revolutum using transmission and scanning electron microscopy. This type of receptor consists of a bundle of 36--40 short, conically projecting cilia ensheathed by a collar formed by a circular evagination of tegument in shape of a cup. The cilia projecting from the centre of a widened nerve fibre, filled with electron-lucid vesicles, possess a well developed basal body and do not contain a ciliary rootlet. In addition to uniciliate sensory endings, three types of multiciliate sensory endings have been demonstrated on the anterior end of Echinostoma revolutum redia using transmission and scanning electron microscopy. This is the first finding of multiciliate sensory endings in the parthenogenetic generation of trematodes. In the first type, 12 cilia are situated in a pit communicating with the tegument surface. In the second and third types, there are two and four short cilia, respectively. They protrude from the nerve bulb above the surface of the tegument. The significance of multiciliate sensory endings in rediae and their similarity to multiciliate sensory endings in miracidia and cercariae is discussed.  相似文献

Studies involving comparisons of taxa that vary in their degree of relatedness may allow the distinction of functional and phylogenetic components in cercarial sensory systems. In this study, cercariae of allocreadiids Bunodera Railliet, 1896 and Crepidostomum Braun, 1900, lecithodendriid Allassogonoporus Olivier, 1938 and opecoelid Allopodocotyle Pritchard, 1966 were compared as regards ultrastructure and chaetotaxy of sensory receptors as well as neuromorphology. Cercariae were treated with acetylthiocholine iodide and silver nitrate and some were processed for scanning and transmission electron microscopy. The types of cercarial sensory receptors differed in the presence of a tegumentary sheath, a dome-like base and a tegumentary collar, number of cilia (0, 1, 2 or more), cilium length (short, moderately long or long) and tegumentary collar length (low to moderately low, high or very high). Chaetotaxic patterns were consistent at the family level in all taxa studied. Irregular cholinergic nerve networks were identified. The present study indicates that the major categories of cercarial sensory receptors are nonciliated (including sheathed and subtegumentary types) and ciliated (including uncollared and collared types) receptors. It also allows the distinction of functional and phylogenetic components in the sensory systems of the cercariae studied. Functional components were reflected in the numbers of sensory receptors associated with each nerve region and in the ultrastructure and site-specificity of receptor types. Phylogenetic components included taxon-specific chaetotaxic patterns and receptor types.  相似文献

E. revolutum redia digestive and excretory system ultrastructure, as well as body wall tegument were studied in TEM. The body tegument protrudes in short microvilli and contains uni- and multiciliate sensory endings. The anterior part of the digestive system (pharynx, short oesophagus) is lined with body tegument, the caecum being covered with a flat epithelium with sporadic microvilli. Oesophageal gland cells are filled with minute electron-dense granules. The excretory capillaries joined to large flame cells are composed of spirally wrapped cells, in the cytoplasm of which there are clusters of microtubules. The special features of the digestive system of echinostome rediae, as well as their significance in the antagonistic relationship to the partenitae of other families of trematodes and their pathogenic effect on the intermediate host, are discussed.  相似文献

The digestive tract of a fully formed free-swimming cercaria of Echinostoma revolutum (Froelich, 1802) Dietz, 1909 was studied by electron microscopy. The oral sucker and pharynx are covered by a tegument. The oesophagus and the caeca are not luminized. They are packed with giant epithelial cells with electron-dense granules of irregular shape, rough endoplasmic reticulum and its cisterns, and Golgi apparatus. The tegument of the oral sucker cavity contains numerous uniciliate receptors.  相似文献

Trypanorhynch cestodes from Indonesian coastal waters (East Indian Ocean)   总被引:4,自引:0,他引:4  
Results of a study on trypanorhynch cestodes of fishes from Indonesian coastal waters are presented. A new species, Dasyrhynchus thomasi sp. n., is described, and five species are recorded which all represent new locality records: Tentacularia coryphaenae Bosc, 1797; Nybelinia africana Dollfus, 1960; Nybelinia scoliodoni (Vijayalakshmi, Vijayalakshmi et Gangadharam, 1996); Sphyriocephalus dollfusi Bussieras et Aldrin, 1968; and Otobothrium penetrans Linton, 1907. Their known ranges of distribution are extended to the East-Indian Ocean. Scanning and transmission electron microscopy was used to clarify details of the tentacular armature and surface morphology of T. coryphaenae, D. thomasi and O. penetrans. In T. coryphaenae, hook-like microtriches along the bothridial tegument are embedded in the distal cytoplasm, sometimes showing a split base. The solid tentacular hooks are embedded into a fibrillar, highly ordered tentacular wall. D. thomasi is distinguished by its characteristically shaped bothridia and a triple chainette with winged hooks on the external surface of the tentacle. Tufts of microtriches with ciliated sensory receptors are regularly arranged on the bothridial surface of O. penetrans. They show similarities to sensory receptors reported from other trypanorhynch cestodes. Otobothrium pephrikos Dollfus, 1969 is considered a junior synonym for O. penetrans, and the variability of the scolex within trypanorhynch cestodes is emphasised.  相似文献

Six types of gland cells occurring in the body of developing Echinostoma revolutum cercaria are described at ultrastructural level. Three types of the gland cells, lateral, ventral and dorsal, release their secretion into the tegument of cercaria during its development in the media. Another three types, paraoesophageal, penetration and proper cytogenic gland cells are preserved in the body of the free-swimming cercaria. The paraoesophageal gland cells open on the tegument surface. Secretory granules of all types of gland cells and stratification of the body tegument arising by gradual release of the contents of the above mentioned three types of gland cells are characterized in detail.  相似文献

Scanning electron microscopy examinations of trematode specimens belonging to Crepidostomum farionis (O.F. Muller, 1784) and C. metoecus Braun, 1900, collected from brown trout, Salmo trutta fario L., in the Czech Republic, made it possible to study their surface morphology including details not described previously. The tegument of both species bears numerous characteristic papillae around the oral sucker (in C. metoecus also around the ventral sucker) and the ventral and dorsal surfaces of the forebody, exhibiting a high degree of variability in numbers and arrangement, with tegumental bosses forming lateral fields on the forebody and minute sensory receptors with submerged cilia scattered on the surface of the dorsal part of the oral sucker. In addition to marked differences in the size, shape and position of the oral muscular lobes, both species distinctly differ in the number of genital pores: two separate pores in C. farionis and a single pore in C. metoecus.  相似文献

The authors have found that pinocytosis occurs in the tegument of C. bovis from the fourth week after infection. Electron-lucid bladders surrounded by plasma membrane were encountered in the distal cytoplasm. The pinocytosis occurred in form of micropinocytotic bladders only in the bladder tegument but not in the scolex. The bladders appeared first in the superficial part of the distal cytoplasm. During the following periods of development of the larva they were dispersed in the whole distal cytoplasm and were found even in the processes of subtegumental cells and inside these cells near the heterolysosomes.  相似文献

ABSTRACT Spiroplasma citri, a helical, wall-less prokaryote in the class Molli-cutes, is transmitted by the beet leafhopper, Circulifer tenellus. Invasion of leafhopper tissues and cytopathological effects by S. citri were investigated by transmission electron microscopy. All eight cell types of the principle salivary glands, as well as the adjacent muscle cells and the cells of the accessory salivary glands, were colonized by the spiroplas-mas. In both midgut epithelia and salivary gland cells, spiroplasmas usually occurred in membrane-bound cytoplasmic vesicles that often were located near the cell periphery. In several salivary gland cells, spiroplas-mas were also observed within membranous pockets apparently formed by invagination of the plasmalemma beneath intact basal lamina. These observations are consistent with spiroplasma entry into the insect cells by receptor-mediated endocytosis. Cytopathological effects of spiroplasma infection in salivary cells included loss of membrane and basal lamina integrity, presence in some cells of irregular inclusion-like structures containing dense matrices of filamentous material that labeled with anti S. citri antibodies, and apparent disorganization of the endoplasmic reticulum. Compared to the tightly aligned fiber bundles in healthy muscle cells, bundles in spiroplasma-containing muscle cells appeared fragmented and loosely arranged. Such symptoms could contribute to the reduction in longevity and fecundity that has been previously reported for S. citri-infected C. tenellus.  相似文献

The ultrastructure of the tail fins and muscles of Echinostoma revolutum cercaria is described. The longitudinal muscles consist of a primitive type of transversely striated muscle fibres with a well-developed sarcoplasmic reticulum in the Z-band. The fins are formed by a longitudinal tegument fold containing the same components as the remaining part of the tail tegument. Isolates of Rhizoctonia solani AG2-2 obtained from turf with symptoms of large-patch disease of warm-season turfgrasses were compared with known AG2-2 isolates belonging to cultural types IIIB and IV. Some isolates that were previously identified as type IV have been separated here and named LP isolates. Comparisons among isolates were based on cultural morphology, hyphal growth rate, pathogenicity and restriction fragment length polymorphism (RFLP) analysis in the nuclear encoded ribosomal DNA (rDNA) genes. The cultural characteristics of LP isolates varied from those of types IIIB and IV. LP isolates did not show distinct sclerotial formation and zonation, and the colour of their mycelia and pigment deposition was dark brown. LP isolates had slower hyphal growth rates than types IIIB and IV, with an optimum temperature of 25°C compared with 28°C for types IIIB and IV. LP isolates were less virulent on radish but highly virulent on zoysia grass when compared with isolates of types IIIB and IV. Genomic DNA was digested separately with Eco RI, Ban III, Xba I and Sal I, and probed with cloned rDNA from Alternaria alternata in Southern hybridizations. LP isolates had one RFLP pattern, while both IIIB and IV possessed four different patterns each. Cluster analysis of RFLPs showed that R. solani AG2-2 is divided into three genetic subgroups, consisting of the IIIB, IV and LP isolates, respectively. The polymerase chain reaction (PCR) amplified rDNA internally transcribed spacer (ITS) regions of the IIIB, IV and LP isolates had the same length but produced different restriction patterns when digested with Msp I and Taq I. These results indicate that there are three cultural types in R. solani AG2-2, namely IIIB, IV and LP.  相似文献

The cercariae of Eurytrema pancreaticum (Janson, 1889) possess four types of gland cells - proper cystogenic, penetration, ventral and dorsal gland cells. The secretion of ventral and dorsal gland cells is released into the tegument. The proper cystogenic gland cells are the largest and their contents serve for the formation of the cyst wall of metacercariae in the second intermediate host. The secretion of proper cystogenic gland cells contains besides neutral mucosubstances also acid mucosubstances with both carboxyl- and sulphogroups digestible with beta-glucuronidase. The secretion of penetration gland cells contains neutral mucosubstances and proteins with tyrosine, tryptophan and SS groups. The ventral gland cells contain mostly acid mucosubstances with sulphogroups, which are digested with beta-glucuronidase, and proteins with tyrosine, tryptophan and SH groups. The rudimentary dorsal gland cells contain a small amount of acid mucosubstances. The whole tegument of cercariae and the two main collecting canals of the excretory system exhibit a high alkaline phosphatase activity. The nerve ring and the main nerve truncs contain proteins with SH groups and hydrophilic lipids and exhibit a cholinesterase activity. The suckers contain a larger amount of glycogen.  相似文献

Electron microscopical studies of the cyst of 1-month-old H. diminuta larvae revealed considerable thickening of the superficial syncytium of tegument and formation of a large number of cavities and canals. The cytoplasm of the tegument is filled with microtubules produced by cytons of parenchymal layer which are analogs of tonofibrils. These peculiarities, together with numerous processes of the external fibrous layer, are regarded as adaptative to the changes of the hydrostatic pressure of the hemocoel of the intermediate host. The superficial syncytium is covered with microvilli and keeps polycellular cytoplasmatic bonds with cytons, which ensures energetic and plastic requirements for the stabilization of the hypertrophied syncytium and its physiological regeneration. Accumulations of fibres identical with those of the connective tissue were found in the cytons of the tegument. The cyst parenchyma consists mostly of cells with widened canals of granular endoplasmic reticulum associated with the microfibrils and production of fibrous filaments. The internal fibrous layer is produced by typical fibroblasts. The tegument of the cercomer is thinner and is considered to be less differentiated.  相似文献

The forebody and foregut of Crepidostomum metoecus Braun, 1900 are invested with a tegument bearing regularly arranged surface tubercles comparable with the aspidogastrean surface structures. The tegument of the ventrolateral lobes and of the prepharynx is penetrated by ducts of eccrine gland cells. The frontal and prepharyngeal gland cells, localised in the parenchyma, discharge electron-dense granules. Their ducts are lined by peripheral microtubules and fixed to the tegument plasmalemma by a septate junction. The functional roles of these glands are discussed.  相似文献

Selectivity to insects over mammals is one of the important characteristics for a chemical to become a useful insecticide. Fipronil was found to block cockroach GABA receptors more potently than rat GABAA receptors. Furthermore, glutamate-activated chloride channels (GluCls), which are present in cockroaches but not in mammals, were very sensitive to the blocking action of fipronil. The IC50s of fipronil block were 30 nM in cockroach GABA receptors and 1600 nM in rat GABAA receptors. Moreover, GluCls of cockroach neurons had low IC50s for fipronil. Two types of glutamate-induced chloride current were observed: desensitizing and non-desensitizing, with fipronil IC50s of 800 and 10 nM, respectively. We have developed methods to separately record these two types of GluCls. The non-desensitizing and desensitizing currents were selectively inhibited by trypsin and polyvinylpyrrolidone, respectively. In conclusion, in addition to GABA receptors, GluCls play a crucial role in selectivity of fipronil to insects over mammals. GluCls form the basis for development of selective and safe insecticides.  相似文献

The external morphology of two bucephalid digenean parasites of Conger conger (Linnaeus) (Congridae, Anguilliformes) caught northwest of the Iberian Peninsula, Prosorhynchus crucibulum (Rudolphi, 1819) Odhner, 1905 and P. aculeatus Odhner, 1905, were studied using scanning electron microscopy (SEM). SEM techniques elucidated new external morphological details, mainly relating to the tegument and protruding organs, such as, in P. crucibulum, a papilla-like structure associated with the pharynx and, in P. aculeatus, the cirrus. The tegument bears scale-like spines, which in both species are arranged quincuncially. The spines of P. crucibulum are wider than long and cover the major part of the body and rhynchus. However, no spines were found in either the central apical depression of the rhynchus or in the middle of the ventral indentation. Also, spines were rarely seen on the tegument around mouth, around the genital aperture or close to the excretory pore. P. aculeatus has spines of a different shape, as wide as they are long and with a rounded margin. They cover the whole body and almost the entire rhynchus, but none were found in the middle of the rhynchus or on its neck region.  相似文献
