Growing pigs with ileum and ileocecal re-entrant canulae were given 5 different rations (pig fattening feed 1 and 2, rations with dried skim milk, wheat gluten + lysine and wheat + wheat gluten + lysine). The crude protein and amino acid excretion was quantitatively determined with ileum chyme and feces. The calculation of the amino acid balances (apparent and true digestibility) at the end of the small intestine, at the end of the wholedigestive tract and the isolated colon showed characteristic differences for the individual amino acids. On ileum level the amino acids with relatively high endogenous quotas, threonine, tryptophane, alanine, aspartic acid and glycine showed a distinctly lower, the amino acids arginine, methionine, phenylalanine, glutamic acid, proline and tyrosine a distinctly higher digestibilityhan crude protein. In comparison of the values on the feces level, including the true digestibility, these differences are largely balanced out. At the end of the small intestine the absorption of the amino acids is in the main completed. The disappearance rate of amino acids from the colon shows large differences with reference to the different rations as well as the individual amino acids. They were particularly high for various rations as far as proline, tryptophane, glycine and cystine are concerned. If wheat gluten + lysine rations were given, a net synthesis of methionine in the colon could be proved.  相似文献

Re-entrance cannulae were applied by way of operations at the end of the small intestine of a larger number of growing pigs. After a post-operative phase these animals were fed with various rations (N-free diet, fattening feed for pigs, rations with dried skim milk resp. wheat gluten + lysine and wheat + wheat gluten + lysine). During the main experiment periods the ileum digesta (24 hour-periods of collecting) and the feces were quantitatively registered. The protein and amino acid content ascertained in the feed rations, the digesta and feces samples are reported, comparatively evaluated and discussed. The amount of amino acids (in mg per kg intake of dry matter) contained in the ileum digesta after N-free feeding were statistically calculated with regard to differences occuring between the animals and days. For most amino acids a significant decrease in the ileum digesta could be detected during the course of the N-free feeding period. Moreover, the amount of amino acids in the ileum digesta was compared with the metabolic fecal amino acids, from which conclusions could be drawn concerning the disappearance rate of the individual amino acids in large intestine.  相似文献

The amino acid composition in the chyme and faecel protein after feeding pigs with different diets is described. It is to be stated that chyme protein cannot necessarily be considered homogenous. Faecal protein shows less considerable variations in its composition and deviates distinctly from the composition of the faecal bacteria. The quota of bacterial crude protein was calculated according to the content of 2,6 diamino pimelic acid. In the chyme protein (end of the ileum) it amounted to approximately 30% and in the faecal protein to approximately 50%.  相似文献

Three female pigs of about 50 kg liveweight were provided with re-entrant canulas in the upper and lower sections of the small intestine and were catheterized to collect urine and blood (vena jugularis). After feeding once the experimental diet containing 15N-labelled, fat extracted dry curd as protein component, it was possible to determine, on two days, the chyme passed through the canulas and to separate aliquote portions thereof for analysis. The cellulose contained in the food was used as indicator for the passage rate of the chyme in the small intestine. The passage rates in the small intestine calculated from the amounts of cellulose and chyme were found to agree fairly well. The passage rates in the upper part of the small intestine were characterized by strongly marked, short-time peaks, right after feed intake, and by periods of relatively even chyme flow some eight hours after feeding. At the end of the small intestine almost constant chyme passage rates were observed over the whole experimental period.  相似文献

Studies of the apparent and true ileal digestibility of crude protein and the apparent and true amino acid (AA) absorption were carried out with 7 female pigs in the live weight range between 22 and 34 kg after ileorectostomy. Protein sources of varied quality with regard to the method of treatment (toasted) and untoasted soybean oilmeal) and to the AA composition (vital gluten or vital gluten + L-lysine X HCl) were used. Distinct differences regarding the apparent ileal digestibility of crude protein and the apparent absorption of amino acids were discovered between the protein sources, the cause of which is to be seen in the different amounts of endogenous N. The true ileal digestibility of crude protein and amino acid absorption were partly distinctly higher than the values of the apparent ileal digestibility of crude protein and AA absorption. On the whole one can say that the results of the investigation gained by means of ileorectostomy can be compared to the values gained from cannulated animals. Thus the experimental method used is suitable for studying the absorption process to the end of the small intestine of pigs.  相似文献

The endogenous nitrogen in the duodenum and the distal segment of the small intestine of 3 fistulated pigs fed a 15-N-labelled feed protein was determined using the isotope dilution technique. Over 24 hours, the endogenous N secretion was found at least 12.5 g at the end of the duodenum. For the upper segment of the small intestine, a statistically secured correlation between endogenous nitrogen and chyme amount was found to exist. It was possible to determine the true and apparent absorption of the nitrogen for both individual segments of and the whole digestive tract.  相似文献

An experiment was conducted to compare apparent total tract protein digestibilities and apparent ileal digestibilities of protein and amino acids in growing pigs and adult pregnant and lactating sows. Twelve growing pigs and 12 sows were used and surgically fitted with simple T-cannulas at the distal ileum. Six experimental diets based on corn, barley, wheat, soybean meal, canola meal, or meat and bone meal were formulated, and each diet was fed to growing pigs, gestating sows, and lactating sows for 7 d. Chromium oxide was included in all diets as an indigestible marker (.25%) for calculating nutrient digestibilities. Fecal material was collected on d 5 of each feeding period by grab sampling, and ileal samples were collected for 12 h/d during the last 2 d of each feeding period. Apparent fecal protein digestibilities for all feed ingredients were higher (P < .05) in gestating and lactating sows compared to growing pigs, but no differences between the two groups of sows were observed (P > .05). At the distal ileum, no differences (P > .05) in protein digestibilities were detected between sows and growing pigs regardless of feed ingredient. For all feed ingredients tested, lactating sows had apparent ileal digestibilities of most amino acids that were two to six percentage units higher than those obtained in growing pigs, but not all of the differences were significant. Gestating sows had digestibilities of most amino acids that were intermediate between those of growing pigs and lactating sows. The combined results from the six feed ingredients showed that lactating sows had higher (P < .05) digestibilities of all indispensable amino acids except arginine, and gestating sows had higher (P < .05) digestibilities of five of the indispensable amino acids than did growing pigs. The results of this experiment indicate that apparent fecal protein and apparent ileal amino acid digestibilities obtained in growing pigs are not always representative of digestibilities in either gestating or lactating sows.  相似文献

Ileo-rectal anastomosis (IRA), which is frequently used to measure prececal digestibility in pigs, could induce some disturbances of the normal absorptive function. Our aim was to investigate the effects of different IRA surgical procedures on the main histologic characteristics of the small intestine in pigs. The 4 different IRA procedures compared to intact pigs (INT) were the following: either end to end (EE) or end to side (ES) with or without preservation of the ileocecal valve (EEV, EE, ESV, ES respectively). At 147 d after surgery, samples of the wall of the duodenum, jejunum and ileum were taken under anesthesia and histometric examinations were performed on HE- and PAS-colored sections to estimate changes mainly of mucosa and muscle layers. The values recorded for villus length, crypt depth, and whole thickness of the mucosa suggested that the EE procedures disturb the small intestine less than the ES models. A new parameter, called epithelial quotient and calculated as [(villus length/crypt depth)/mitotic index], was proposed to improve the comparisons. According to this quotient, EE procedures did not significantly affect the mucosa of the whole small intestine. An increased density of goblet cells was recorded in all operated pigs along the small intestine, but mainly in the ileum after EE-IRA. The lymphatic follicle area was reduced. These findings, which were in agreement with a reduced mitotic index in the ileum of EE-pigs, indicated a decreased effect of noxious factors on the small intestinal mucosa in IRA-pigs, especially after the EE-IRA procedure. Some atrophic or hypertrophic effects on the muscle layers were related to the absence or preservation of the ileo-cecal valve. Finally it was concluded that i) there was no major disturbance after IRA, and ii) the end to end procedure was most beneficial for the structural integrity of the small intestine.  相似文献

Three female pigs provided with re-entrant canulas at the beginning and the end of the small intestine and catheterized for blood and urine collection were once fed a 15N-labelled mixed diet. Until 48 hours after feed intake chyme from the canulas and urine and blood were collected at varying intervals. All 15N-frequencies measured in the duodenal chyme were found to be below the labelling of the feed protein. Until 11 hours after intake of the labelled feed, the residue obtained by centrifugation of the intestinal contents revealed the highest 15N-labelling. Only slight differences were found in the frequencies of the other fractions (proteins, peptides, free amino acids). The almost constant labelling of all fractions in that period is indicative of a dilution with endogenous nitrogen being constant in comparison with food N. The 15N-frequencies of the chyme fractions observed at the end of the small intestine allowed to conclude that the breakdown of the food proteins is more complete than the disintegration of the endogenous proteins.  相似文献

One hundred and twenty pigs were used to evaluate the effects of different dietary lysine levels on the growth performance, apparent nutrient digestibility, and abundance of cationic amino acid transporter messenger RNA (mRNA) in the small intestine of finishing pigs. Pigs received a low lysine diet (LL, 0.60% lysine), moderate lysine diet (ML, 0.80% lysine) or a high lysine diet (HL, 1.00% lysine) for 28 days. A digestion test was carried out during the third week. Although the apparent nutrient digestibility in pigs fed experimental diets were different (P < 0.05) and the highest when pigs were fed ML diet, diets did not change the growth performance. In the duodenum, mRNA abundance of PepT-1, as detected by real-time RT-PCR, was reduced in the LL diet (P < 0.05). A greater abundance of b0,+AT and PepT-1 mRNA was associated with the ML diet (P < 0.05) in the jejunum and ileum, respectively. In the ileum, the HL diet had a lower abundance of CAT-1 mRNA compared with other diets. These results showed that the finishing pigs would gain better nutrient digestibility when the dietary lysine content was 0.80%, and dietary lysine levels influenced the expression of cationic amino acid transporter mRNA in the small intestine of finishing pigs.  相似文献

A replicated trial of 3 X 3 Latin square design was conducted with growing pigs (about 29 kg initially) to determine the effects of different levels of feed intake on nutrient digestibilities determined near the end of the small intestine and over the total digestive tract. Pigs were fiftted with simple T-cannula. Feeding levels were ad libitum (AL) and limit-fed (4.5 or 3% of body weight/d). A 16% sorghum-soybean diet was used. Limit-fed pigs were fed at 12-h intervals and water was limited to 2 liters/kg of diet; AL pigs received water ad libitum. Consumption by pigs fed ad libitum averaged 6% of body weight/d. Neither feeding method nor level greatly affected nutrient digestibility measured at the end of the small intestine, but values tended to decrease as feeding level decreased. Comparisons of nutrient digestibility between AL and limit-fed pigs ranged from 2.9 to .7 percentage units, with only N (P less than .10) and methionine (P less than .05) reaching significance. Differences between 4.5 and 3% were slightly larger, ranging from 4.3 to -.2 percentage units, with differences for dry matter, N, gross energy and several amino acids reaching significance (P less than .10 or P less than .05). Measured over the total tract, observed differences among feeding levels were again small, but the trend was reversed, with digestibilities increasing as feeding level decreased. Two experiments were carried out using a balanced feed mixture composed of wheat, corn, soyabean meals, casein, and crystalline amino acids (AA) and supplemented with 0, 4, or 8% of apple pectin. The daily dietary allowance provided pigs with the same level of nutrients, apart from the pectin. One experiment was conducted on pigs (within 25 to 40 kg BW) with the post valve T-caecum (PVTC) cannula to determine AA digestibility and ileal digesta viscosity. In the second experiment, the N-balance was measured twice on male pigs at about 20 and 28 kg BW using 6 animals per diet and feed intake and body weight were measured twice during the 28 d. After 40 d of feeding the diets, the pigs were sacrificed and the small intestines were weighed and sampled. The added pectin decreased standardized ileal AA digestibility (on average up to 5% dig. units) and increased digesta viscosity from about 1 to 88 mPa s. The higher pectin level did not affect motility of the duodenum and mid-jejunum, measured as responses to electrical field stimulation and to acetylcholine. However, the response of the duodenum to electrical field stimulation was after feeding the diet with 4% pectin increased compared with other diets. Pectin supplementation did not alter the weight and the length of the small intestine, but induced changes in the intestinal morphology. Muscle layer width increased significantly in the duodenum, the mid-jejunum, and the ileum while villi length increased in the duodenum and ileum. The addition of pectin decreased N retention and increased the F/G ratio but did not affect significantly the ADG of the pigs.  相似文献

The experiment was carried out on 3 pigs of 34 kg body weight equipped with re-entrant cannulas both in the duodenum and terminal ileum. Furthermore, catheters were inserted into the external jugular vein and into the carotid artery. During a preliminary period digesta from both cannulas were collected and stored. This digesta was than reintroduced during a 12 hours period of intravenous infusion of 14C-leucine while outflowing digesta from the proximal parts of the cannulas were collected, stored and analyzed for leucine content, 14C-radioactivity and specific radioactivity of leucine. This paper reports the net secretion of 14C-activity and of 14C-leucine by the small intestine and the recovery of both in the content of the digestive tract at the end of the infusion. It was concluded that endogenous leucine enters the lumen of the small intestine through the intestinal wall mainly in a protein bound form. Free leucine is secreted, however, only in small amounts. Metabolites of leucine were also secreted into or formed in the lumen of the small intestine. A proportion of endogenous leucine was reabsorbed during the passage of digesta along the small intestine.  相似文献

Differently structured isolated proteins with the same amino acid composition were tested with growing albino rats in an N-balance and absorption experiment. The following items were tested: --isolated horse bean protein --hydrolysed isolated horse bean protein with an amino acid supplement to make up for losses due to hydrolysis --horse bean protein-casein (1:1) fibres --isolated horse bean protein with an amino acid supplement to achieve the same amino acid concentrations as in the horse bean protein-casein fibres. The lowest digestibility and utilisation values were ascertained for the hydrolysed protein. Endogenous N-excretion calculated according to the isotope dilution method was higher than for intact proteins, the transport of the chyme in the digestive tract and the true absorption of N in the small intestine had significantly diminished. In comparison with horse bean protein supplemented with amino acids, the horse bean protein-casein fibres showed decreased protein utilisation and increased endogenous N-excretion. Independent of the protein fed and of the time after feeding a relatively constant endogenous N-quota of approximately 70% could be ascertained in the chyme in the small intestine. In contrast to this, the relation between exogenous and endogenous N determined from the contents of the large intestine proved to be dependent on the intermediary utilisation of the protein fed.  相似文献

An experiment was conducted to investigate the effects of BW, feed intake, and the physiological condition of the animal on the loss and amino acid composition of endogenous protein in swine. Ten growing barrows and five multiparous sows were equipped with a T-cannula in the distal ileum for digesta collection. A protein-free diet was fed to all animals. The barrows were given free access to the experimental diet. The sows also were allowed to consume the diet on an ad libitum basis, and digesta were collected during lactation and in the following gestation period. In addition, digesta from the gravid sows were collected after restricting the sows to 2 kg of feed per day. For each animal group, the endogenous losses of protein and amino acids were calculated in relation to DMI, and the amino acid composition of endogenous protein was calculated. The total endogenous gut protein loss at the distal ileum of growing pigs, lactating sows, and gestating sows, given free access to feed, was 12.4, 9.4, and 11.2 g/kg DMI, respectively. These values were not different (P > .10). However, when gestating sows were fed only 2 kg/d, 17.8 g of endogenous protein was lost per kilogram of DMI, which was higher (P < .05) than for any of the other groups. This difference was mainly caused by higher (P < .05) losses of glycine, proline, and serine. There were no differences (P > .05) in amino acid composition of endogenous protein between growing pigs, lactating sows, and gestating sows given free access to feed, but restricted-fed gestating sows had an amino acid composition of endogenous protein that was significantly different from that of the other groups. The results from the experiment showed that age, BW, and the physiological condition of the animal have little or no effect on the amount of endogenous protein and amino acids lost at the distal ileum of hogs if calculated in relation to DMI. Likewise, the amino acid composition was not affected by the BW or physiological condition of the animal. However, DMI had a significant effect on endogenous protein losses in sows as well as on amino acid composition of endogenous protein.  相似文献

Abstract

An experiment was conducted to investigate changes in the lumen and wall of the intestinal tract of weaned pigs caused by fructo-oligosaccharides (FOS) and consequences for growth. Twelve male pigs weaned at 24 days were individually housed and fed either control, 0.25% FOS, or 3% FOS diets for 21 days after weaning. The pH of chyme in the caecum and proximal colon were lower (p<0.001) in 3% FOS compared to other treatments. Mean molar proportions (%) of total SCFA values along the large intestine were significantly affected by FOS diet. Acetic acid was significantly higher in control compared with other treatments. Butyric acid and isobutyric acid were significantly increased in 0.25 and 3% FOS compared to the control. Feed intake, empty body weight gain, disaccharidase activities and villous height in the small intestine were not significantly affected by FOS. It was concluded that dietary FOS stimulates fermentation in the large intestine, but does not significantly affect enzyme activity, ammonia content, villous height and growth.  相似文献

The crude protein digestibility and the amino acid absorption of six female pigs (average live weight 61 kg) with duodenal and ileocecal re-entrant cannulae, which were fed with a wheat + wheat gluten + lysine ration and an N-free ration, were determined in various segments of the intestines. Comparative experiments concerning the N-metabolism with pigs without and with cannulae showed that the animals renormalised their metabolism 14 days after narcosis and fistulation of the intestines. The extents of secretion and absorption of the various amino acids vary as it is shown from the values of the apparent and true digestibility resp. rate of passage through various segments of the intestines. While for methionine and glutamic acid absorption exceeds endogenous secretion already in the duodenum, the amino acids with a high endogenous quota (glycine, alanine, threonine, tryptophan) are, even at the terminal ileum, not as well absorbed as the others. Methionine is obviously synthesised on a large scale by the colon flora and excreted in feces. The fractionation of the duodenal and ileum chyme after feeding wheat + wheat gluten + lysine as well as N-free mixture, into the fractions "solid particles", "peptides-free amino acids" and "proteines precipitable by trichlor-acetic acid" supplies information on the degree of protein degradation in various segments of the intestines.  相似文献

A dynamic model of protein digestion in the small intestine of pigs   总被引:5,自引:0,他引:5  
A dynamic mathematical model of the digestion of proteins in the small intestine of pigs was developed. The model integrates current knowledge on the transit of digesta along the small intestine, endogenous secretions, digestion of proteins, and absorption of amino acids into a mechanistic representation of digestion. The main characteristics of the model are the following: the small intestine is divided into several segments of variable length but with equal digesta retention time; the rate of transfer of digesta between segments is based on the progression of myoelectric migration complexes; pancreatic and biliary secretions are poured into the first segment, whereas intestinal secretions enter all intestinal segments; protein hydrolysis is described by first-order equations; and an intestinal absorption capacity is used to estimate absorption of hydrolyzed protein. Simulation results are consistent with observed data, although more information is needed to represent reality more closely. The sensitivity analysis shows that parameters for protein hydrolysis largely determine protein digestibility. The absorption capacity of the small intestine limits the absorption of amino acids at the beginning of a meal and modulates the appearance of amino nitrogen in the portal vein. It also shows that amino acid absorption can be limiting to protein digestibility when large amounts of protein are eaten in a single daily meal. The model is useful in evaluating the dynamics of protein digestion and absorption of feedstuffs. The model can be used in evaluating protein digestion of different feedstuffs and feeding strategies.  相似文献

In addition to a basic ration (barley, dried green fodder, fishmeal) pigs (castrated males, 100 ... 130 kg live weight) received a supplement of raw potato starch, raw ensiled potatoes, cooked potato starch or steamed ensiled potatoes; nutrient and amino acid passage and digestibility (absorption) were measured in the small intestine, the upper large intestine and the total digestive tract with the help of simple cannulae at the distal ileum and the middle of the hind gut and the estimation of the HCl insoluble ash as an indicator. In addition, the amounts of bacteria in faeces were determined and the influence of raw and cooked potato starch on N retention was ascertained. When raw potato products were fed, distinctly higher amounts of dry matter, organic matter and starch pass the ileum, which get into the colon undigested, i.e. the digestibility of these substances is low at the end of the ileum (raw potato starch 24%, starch of raw ensiled potatoes 36%). Up to the middle of the colon there is intensive bacterial starch degradation so that in this place the difference between raw and thermically treated potatoes is only small. No matter what the previous treatment of the potatoes was, there is hardly any starch in faeces, i.e. it was nearly 100% digested up to the end of the digestive tract. The passage and digestibility resp. of ash and crude cellulose from the basic ration is not influenced by the supplement of raw or thermically treated potato products in the individual sections of the digestive tract. At the end of the ileum there are no differences between the apparent digestibility of the crude protein and absorption of amino acids of supplemented raw or cooked potato starch or steamed ensiled potatoes; after the use of raw ensiled potatoes they are, however, significantly more reduced than the comparison of analyses of faeces shows. In the large intestine the high amount of potato starch stimulates the metabolism and the reproduction of faecal bacteria as is shown by an increase by 20-30% in bacteria N excretion in faeces. Apparent crude protein digestibility and amino acid absorption are diminished by that. The increased N excretion in faeces after the supplement of raw potato starch to the basic ration is-in comparison with the supplement of heated starch-compensated by a commensurately lower N excretion in urine so that N retention practically remains uninfluenced by the supplement of variously treated potato starch.(ABSTRACT TRUNCATED AT 400 WORDS)  相似文献

Eight Angus x Gelbvieh heifers (445 +/- 74.5 kg) fitted with ruminal and duodenal cannulas were used in a 4 x 4 Latin square double double-crossover designed experiment to assess the effect of restricted forage intake on site and extent of digestion and flow of essential AA amino acids to the small intestine. Heifers were fed chopped (2.54 cm) bromegrass hay (9.2% CP, 64% NDF on an OM basis) at one of four percentages of maintenance (30, 60, 90, and 120%). Experimental periods were 21 d in length, with 17 d of adaptation followed by 4 d of intensive sample collection, after which maintenance requirements and subsequent level of intake were adjusted for BW change. True ruminal OM, NDF, and N digestion (g/d) decreased linearly (P < 0.001) with decreasing forage intake. When expressed as a percentage of OM intake, true ruminal OM and N digestibility were not affected (P = 0.23 to 0.87), whereas ruminal NDF digestibility tended to increase (P = 0.09) as forage intake decreased. Total and microbial essential amino acid flow to the duodenum decreased linearly (P = 0.001) from 496.1 to 132.1 g/d and 329.1 to 96.0 g/d, as intake decreased from 120 to 30% of maintenance intake, respectively. Although the profile of individual essential amino acids in duodenal digesta (P = 0.001 to 0.07) and isolated ruminal microbes differed (P = 0.001 to 0.09) across treatment, the greatest difference noted for total and microbial essential amino acid profile was only 0.3 percentage units. Because total and microbial flow of essential amino acids to the small intestine decreased as OM intake decreased, but true ruminal degradability of individual essential amino acids (P = 0.17 to 0.99) and digesta essential amino acid profile were comparable across treatments, total essential amino acid supply to the small intestine was predicted using OM intake as the independent variable. The resulting simple linear regression equation was: total essential amino acid flow = (0.055 x OM intake) + 1.546 (r2 = 0.91). The model developed in this experiment accounted for more of the variation in the data set than the current beef cattle NRC model, which under-predicted total flow of essential amino acids to the duodenum. The prediction equation developed herein can be used to estimate the supply of essential amino acids reaching the small intestine when formulating supplements to compensate for potential amino acid deficiencies resulting from restricted forage intake.  相似文献
