ObjectiveTo study the effect of mate (Ilex paraguariensis) on serum lipids and antioxidant status in normocholesterolaemic and hypercholesterolaemic rats.MethodsTriglycerides (TG), total, LDL- and HDL-cholesterol levels, total antioxidant capacity (FRAP and ABTS assays), malondialdehyde (MDA) and protein carbonyls were analysed in serum, and MDA, glutathione and antioxidant enzyme activity in livers of rats drinking water or mate fed normal or cholesterol–cholic supplemented diets.ResultsABTS, glutathione and antioxidant enzymes were not affected by any treatment. In normocholesterolaemic animals, mate had no effect on serum lipids or antioxidant status, yet it increased serum carbonyls and liver MDA concentrations. In hypercholesterolaemic rats, mate consumption had no effect on HDL-cholesterol or protein carbonyls, yet it showed a marked hypolipidaemic action, decreasing TG, total and LDL-cholesterol, and serum MDA levels that had been increased after consuming the high-cholesterol diet.ConclusionPotential beneficial effect of mate on markers of cardiovascular risk seems to be restricted to hyperlipaemic animals.  相似文献

The effect of schisandrin B (Sch B) on long-term ethanol-induced oxidative stress in various rat tissues was investigated. Long-term ethanol treatment increased reactive oxygen metabolites (ROM) level in plasma. The ethanol-induced oxidative stress was assessed by mitochondrial glutathione and α-tocopherol levels, antioxidant enzyme activities, malondialdehyde (mtMDA) production and heat shock protein (Hsp) 25/70 levels. Liver was most susceptible to oxidative stress with a significant increase in mtMDA production. Long-term Sch B treatment enhanced mitochondrial antioxidant status in a tissue non-specific manner. Sch B co-treatment ameliorated the alterations in plasma ROM levels, mtMDA production and Hsp 25/70 expression in rat tissues.  相似文献

The protective effects of Ginkgoselect Phytosome((R)) (GBP) on Rifampicin (RMP) induced hepatotoxicity and the probable mechanism(s) involved in this protection were investigated in rats. Liver damage was induced in Wistar rats by administering rifampicin (500 mg/kg, p.o.) daily for 30 days. Simultaneously, GBP at 25 mg/kg and 50 mg/kg, and the reference drug silymarin (100 mg/kg) were administered orally for 30 days/daily to RMP treated rats. Levels of marker enzymes (SGOT, SGPT and SALP), albaumin (Alb) and total proteins (TP) were assessed in serum. The effects of GBP on lipid peroxidation (LPO), reduced glutathione (GSH), superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPX) and glutathione reductase (GR) were assayed in liver homogenates to evaluate antioxidant activity. GBP (25 and 50 mg/kg) and silymarin elicited a significant hepatoprotective activity by lowering the levels of serum marker enzymes and lipid peroxidation and elevated the levels of GSH, SOD, CAT, GPX, GR, Alb and TP in a dose dependant manner. The present findings suggest that the hepatoprotective effect of GBP in RMP induced oxidative damage may be related to its antioxidant and free radical scavenging activity.  相似文献

We evaluated the hypoglycemic and antioxidant effects of the total alkaloids of leaves and twigs of Catharanthus roseus Linn.(CTA) in streptozotocin(STZ)-induced diabetic rats. The hypoglycemic effect was measured by blood glucose and plasma insulin level. Oxidative stress was measured in heart, liver and kidney by levels of antioxidant markers, free radical scavengers and lipid peroxides i.e. superoxide dismutase(SOD), catalase(CAT), glutathione(GSH) and thiobarbituric acid reactive substances(TBARS). Biochemical parameters, i.e. aspartate aminotransferase(AST), alanine aminotransferase(ALT), alkaline phosphate(ALP) were observed in diabetic control and treated rats. Oral administration of CTA for30 days was followed by a significant(P \ 0.05) decrease in fasting blood glucose and increase in insulin level as compared with untreated diabetic rats. Also it significantly(P \ 0.05) reduced ALT, AST and ALP. The treatment also resulted in significant(P \ 0.05) reductions in GSH,SOD, CAT, and decrease in TBARS in the heart, liver and kidney of diabetic rats. The results suggest that CTA can effectively normalize the impaired antioxidant status in STZ-induced diabetes in a dose-dependent manner.CTA exerted rapid protective effects against lipid peroxidation by scavenging of free radicals and reducing the risk of diabetic complications.  相似文献

The antioxidant activity of flavonoids from banana (Musa paradisiaca) was studied in rats fed normal as well as high fat diets. Concentrations of peroxidation products namely malondialdehyde, hydroperoxides and conjugated diens were significantly decreased whereas the activities of catalase and superoxide dismutase were enhanced significantly. Gongronema latifolium is a rainforest plant, which has been traditionally used in the South Eastern part of Nigeria for the management of diabetes. The effects of oral administration of aqueous and ethanolic G. latifolium leaf extracts for 2 weeks on streptozotocin-induced diabetic rats were investigated. Both extracts were shown to significantly increase the activity of superoxide dismutase and the level of reduced glutathione. The aqueous extract further increased the activity of glutathione reductase while the ethanolic extract caused a significant increase in the activity of glutathione peroxidase and glucose-6-phosphate dehydrogenase and a significant decrease in lipid peroxidation. These results suggest that the extracts from G. latifolium leaves could exert their antidiabetic activities through their antioxidant properties.  相似文献

The antiulcerogenic effects of the ethanol extract of Picrorrhiza kurroa rhizomes and roots (PK) on HCl/ethanol-induced ulceration in rats with respect to acid/pepsin, lipid peroxidation, antioxidant status, proteins and glycoproteins in the gastric mucosa have been investigated. Turnera ulmifolia is a plant popularly known in Brazil and South America as chanana. Some species of Turnera are widely used in folk medicine for different types of inflammatory diseases. In this study, the preventive intestinal antiinflammatory activity of a lyophilized infusion obtained from the aerial parts of T. ulmifolia was tested in the trinitrobenzenesulphonic acid (TNBS) model of rat colitis. The results obtained revealed that pretreatment to colitic rats with the extract, at 250 and 500 mg/kg, significantly attenuated the colonic damage induced by TNBS. This beneficial effect was associated with an improvement in the colonic oxidative status, since the infusion prevented the glutathione depletion that occurred as a consequence of the colonic inflammation. On the other hand, this antioxidant activity was confirmed in in vitro studies. In conclusion, the preventive effect exerted by the lyophilized infusion of T. ulmifolia in the TNBS model of rat colitis is probably related to its antioxidant properties, due to its flavonoids content.  相似文献

Schisandrin B (Sch B) and schisandrin C (Sch C), but not schisandrin A and dimethyl diphenyl bicarboxylate, protected rat skin tissue against solar irradiation-induced oxidative injury, as evidenced by a reversal of solar irradiation-induced changes in cellular reduced glutathione and α-tocopherol levels, as well as antioxidant enzyme activities and malondialdehyde production. The cytochrome P-450-mediated metabolism of Sch B or Sch C caused ROS production in rat skin microsomes. Taken together, Sch B or Sch C, by virtue of its pro-oxidant action and the subsequent eliciting of a glutathione antioxidant response, may prevent photo-aging of skin.  相似文献

The cytoprotective effect of piperine on benzo[a]pyrene (B[a]P) induced experimental lung cancer was investigated in male Swiss albino mice. Oral administration of piperine (100 mg/kg body wt.) effectively suppressed lung cancer initiated with B[a]P as revealed by the decrease in the extent of lipid peroxidation with concomitant increase in the activities of enzymatic antioxidants (superoxide dismutase, catalase and glutathione peroxidase) and non-enzymatic antioxidant (reduced glutathione, vitamin E and vitamin C) levels when compared to lung cancer bearing animals. Our data suggest that piperine may extend its chemopreventive effect by modulating lipid peroxidation and augmenting antioxidant defense system.  相似文献

本研究报道沙棘(胡颓子科)对雄性白化病白鼠由铬诱导氧化胁迫的抗氧化活性。氧化胁迫通过对小白鼠进行强制性喂养30 d,以浓度为30 m g/kg的铬相对体重比例的重铬酸钾盐实现。铬促使体重下降,而却明显增加了器官与体重比例。铬处理明显减少了谷胱甘肽的降低,增加了丙二醛和肌氨酸磷酸激酶的水平;而且它还加强了血清中谷氨酸草酰乙酸转移酶和谷氨酸丙酮酸转移酶的浓度。用不同剂量的沙棘叶子提取物(乙醇提取)对保护铬元素诱导的氧化胁迫进行了评估,结果表明叶子提取物在浓度为100到250 m g/kg铬与体重比的情况下可以明显保护动物避免由铬所诱导的氧化伤害。  相似文献

The antioxidative properties of bulb, leaf and stalk of Allium ursinum were investigated. Activities of antioxidant enzymes (superoxide dismutase, catalase, peroxidase, glutathione peroxidase), quantities of malonyldialdehyde, superoxide and hydroxyl radicals and reduced glutathione and also the contents of total flavonoids, chlorophylls a and b and carotenoids were determined. The extracts from all plant organs exhibited antioxidant activity, the highest having been observed in the leaves. Furthermore, ESR signal of PBN-OH radical adducts in the presence of leaves phosphate buffer (pH 7) extract was reduced for 87.61%.  相似文献

通过测定黄藤熟笋、生笋(鲜切笋)、熟笋干和生笋干的水和4种不同浓度乙醇提取物中的自由基清除能力、总还原能力、总多酚含量和总黄酮含量,研究了煮熟、干燥加工方法对黄藤笋提取物抗氧化活性的影响。结果表明,煮熟处理能保持或提高提取物的抗氧化活性,干燥处理则相反。乙醇提取物比水提取物具有更高的抗氧化活性,以50%~75%的乙醇提取溶剂效果最好。熟笋在所有溶剂中的自由基清除能力、总还原能力、总多酚含量和总黄酮含量均表现出最高水平,表明煮熟是黄藤笋适宜的加工利用方式。  相似文献

Ethanol mediated free radical generation plays an important role in the pathogenesis of liver injuries and alcoholic liver diseases. In the present study two ellagitannins namely geraniin and amariin isolated from Phyllanthus amarus were examined for their ability to protect mouse liver slices against ethanol induced toxicity and possible mechanism of its protection. Oxidative stress markers such as, lipid peroxidation, protein carbonyl formation, amount of 8-hydroxy-2-deoxyguanosine and antioxidant enzymes levels were measured using specific biochemical assays. Poly (ADP-ribose) polymerase (PARP), Bax and Bcl2 were checked to assess the induction of apoptosis using western blots. The results showed that geraniin and amariin protected mouse liver slices against ethanol induced cytotoxicity. Both compounds inhibited oxidation of lipid, protein and formation of 8-hydroxy-2-deoxyguanosine, all of which were found to be elevated on exposure to ethanol. These compounds restored the antioxidant enzymes altered on ethanol exposure. Compounds also inhibited the cleavage of PARP and bax and restored Bcl2, induced on exposure to ethanol. In summary, both ellagitannins effectively protected mouse liver slices against ethanol induced cytotoxicity and apoptosis by reducing oxidative damage to biological molecules and modulating Bax/Bcl-2 ratio respectively, thus minimizing liver injury.  相似文献
