Nine adult horses were fed alfalfa hay cubes containing approximately 10% Senecio vulgaris until all horses had consumed approximately the same amount of toxic components of S vulgaris, pyrrolizidine alkaloids (PA). The amount of PA consumed was determined by the amount that induced clinical signs of PA toxicosis in 3 horses. The 6 other horses were given similar amounts per kilogram of body weight. An initial decrease of feed intake was observed when horses' diets were changed from alfalfa cubes to alfalfa/Senecio cubes, and feed intake was decreased further over 89 to 98 days. From 50 to 159 days, body weight decreased in all horses. Liver disease was induced in all 9 horses after they ate an average of 233 +/- 9.2 mg of PA/kg of body weight. Eight horses died or were euthanatized. Treatment with branched chain amino acids had no effect on mortality, but appeared to reduce neurologic problems. Clinical signs of PA-induced liver disease included ataxia, head pressing, and decreased feed intake. Other clinical signs of toxicosis were observed individual horses, but did not develop in most horses. Megalocytic hepatopathy developed. Liver abnormalities proceeded as PA was consumed and were severe in 8 of 9 horses before clinical signs of toxicosis appeared. Sulfobromophthalein sodium clearance did not decrease until PA-induced liver disease was advanced. Bile acid (BA) concentrations increased to greater than or equal to 50 mumol/L, in the 8 horses that died. One horse had hepatopathy and increased BA concentration, but survived. Thermoregulation is an energy-expensive process, which can be mitigated by blanketing horses in cold climates, potentially preventing weight loss or leading to decreased feed intake. The objective of this study was to evaluate feed intake, body weight (BW), and body condition scores (BCSs) in blanketed and nonblanketed horses. In October 2019, 16 mature adult horses were blocked by breed and BCS and randomly assigned to a blanketed (n = 8) or nonblanketed (n = 8) treatment; blankets were placed. Data were collected in December 2019 and January 2020 in River Falls, Wisconsin. During the study, horses were housed in dry lots and fed grass-legume mixed round bales. At the start of each trial period, BW and BCSs of horses were taken, hay cores were taken for nutritive analysis, and hay bales were weighed. Hay waste was collected daily, and when hay could no longer be consumed ad libitum, the remaining hay was removed from both pens (orts) and new bales were fed. Hay waste and orts were dried and daily dry matter intake was estimated as the difference between hay provided minus hay waste and orts during the trial period divided by the total BW of the pen. The average bale weight, forage nutritive value, BW, and BCS did not differ across treatment groups (P ≥ .05). However, the daily dry matter intake differed (P ≤ .05) at 2.31% BW for blanketed horses and 2.51% BW for nonblanketed horses. These results suggest blanketed horses conserve energy leading to decreased feed intake.  相似文献

We performed intake, digestibility and ingesta passage studies in 11 Indian rhinoceroses (Rhinoceros unicornis) from four zoological institutions, using total faecal collection for the quantification of faecal output. The regularly fed zoo ration of roughage and concentrates (ration RC) and a roughage-only ration (ration R) were used; the roughage source differed between the facilities and comprised grass hay, grass silage, straw and lucerne hay. Dry matter intake ranged between 0.8 and 1.3% of body weight on ration RC and 0.5-1.2% on ration R. Digestibility coefficients achieved were similar to those reported for horses on diets of comparable composition. Endogenous losses as determined by linear regression analysis were within the range reported for horses. Measurements of faecal volatile fatty acids, faecal lactate and faecal pH also showed similarity to similar measurements in horses. The mean retention times of fluids (Co-EDTA) and particles (Cr-mordanted fibre <2 mm) in the whole gastrointestinal tract averaged 42 and 61 h, respectively, and were the longest ever recorded in a monogastric ungulate with this marker system. The results suggest that the horse is a useful model animal for designing diets for Indian rhinoceroses. Why digestive parameters are similar between these species in spite of enormous differences in body weight and retention times remains to be answered.  相似文献

Hendra has been recognized in Australia as a new zoonotic disease of horses since 1994/5 and subsequent work has shown that the viral agent is endemic in certain species of fruit bat. The Hendra virus is the type species of a new genus within the sub-family Paramyxovirinae, which also contains another newly identified zoonotic bat virus, namely Nipah. It is assumed that contact with bats has led to the Hendra virus being transferred to horses on each of the three separate incidents that have been reported in the last five years. No evidence has been found for widespread subclinical infection of horses.Infected horses can develop a severe and often fatal respiratory disease characterized by dyspnoea, vascular endothelial damage and pulmonary oedema. Nervous signs may also occur. Fatal respiratory disease has been seen in cats and guinea pigs following experimentally induced infections. Transmission of the virus from horses to other horses or man seems to have taken place, but very close contact was required. Three human cases have been recognized, all in association with equine cases. There have been two human fatalities, one due to respiratory failure and the other from a delayed-onset encephalitis. A number of diagnostic methods have been developed, but great care must be taken in obtaining samples from suspected cases.  相似文献

Leptospirosis is a worldwide zoonosis which can affect many species. Control programs need accurate diagnosis to be successful, and currently, diagnosis relies on serology. It presents three main issues: the sampling, the antigen panel, and the cutoff point. Herein, we propose a systematic review on leptospirosis among dogs, pigs, and horses in Latin America in order to improve the understanding of the seroepidemiology of leptospirosis in these species in the region as well as the temporal development of the research on this topic and, consequently, improve the chances of success on control programs. Internet databases were consulted over 2015. Inclusion criteria included serosurvey using MAT; a relevant number of animals; the presence in the antigen panel of at least one representative of serogroups Icterohaemorrhagiae and Canicola for dogs, Icterohaemorrhagiae, Australis, and Pomona for pigs, and Icterohaemorrhagiae and Australis for horses; and a cutoff point of ≥100. Overall, 240 papers were studied, of which 87 referred to dogs, 66 to pigs, 39 to horses, and 48 to more than one of the studied species. In relation to those that met all the inclusion criteria, it was 45 (66.2%) in dogs, 23 (41.8%) in pigs, and 23 (63.9%) in horses. Leptospirosis is widespread in Latin America. Predominant serogroups are Canicola to dogs and Icterohaemorrhagiae to pigs and horses. Therefore, research on animal leptospirosis should be encouraged in Latin America, in order to reach a greater standardization in studies and then achieve better results on control programs.  相似文献

Alfalfa, teff, oat, and wheat hays were fed to eight mature horses to determine voluntary intake of each hay type. After a 2-week adaptation period during which each horse was offered all four hay types each day, a 4-week study was conducted in which two horses were offered each type of hay each week, usually at the rate of 2.2% of initial body weight. Refused feed was collected and weighed the last 5 days of each week to calculate voluntary intake. Horses consumed significantly more alfalfa hay than the other hays. Wheat and teff hays were consumed in significantly lower amounts than alfalfa, but significantly more than oat hay. Across the 5 days during which voluntary intake was measured each week, hay consumption increased each day, reaching levels significantly greater than day 1 by day 4, when measured as a percentage of body weight, and day 5 when measured as a percentage of feed offered. Only alfalfa hay, at the amount consumed, met 100% of nutrient requirements for digestible energy, crude protein, lysine, calcium, and phosphorus for horses in maintenance condition. Teff hay met all nutrient requirements except digestible energy. Neither wheat nor oat hays met 100% of any of these nutrient requirements. Alfalfa and teff hays averaged about 12% nonstructural carbohydrate content (NSC), whereas wheat and oats hays contained about 30% NSC. The lower NSC content of the alfalfa and teff hays makes these more suitable for horses with problems related to carbohydrate metabolism or for refeeding starving horses.  相似文献

Pyrrolizidine alkaloids belong to a class of phytotoxins which are present in more than 6000 plant species. The disease seneciosis in farm animals represents the severe poisoning by pyrrolizidine alkaloids from plants of the genus Senecio. This form of poisoning has been known since the end of the 19th century in Germany, the USA, Canada and New Zealand, and is mainly caused by Senecio jacobaea and related Senecio spp. in farm animals, including poultry. Animal poisoning by pyrrolizidine alkaloids is of worldwide importance. In Germany poisoning of horses and cattle by Senecio jacobaea, which was earlier named Schweinsberg disease, is of renewed relevance for veterinary medicine. The disease occurs almost entirely as a consequence of chronic poisoning and in general ends fatally. The ultimate cause is the formation of toxic metabolites of pyrrolizidine alkaloids in the liver, and their covalent binding to nucleic acids and proteins leading to liver cirrhosis. Because many pyrrolizidine alkaloids possess mutagenic, and a few also carcinogenic properties, European and international authorities are concerned about possible residue levels in food of animal origin. The review addresses in its first part several aspects, being the occurrence, the chemistry, and the toxicology of pyrrolizidine alkaloids as well as animal intoxications by poisonous plants. In the second part (46) clinical characteristics of animal seneciosis, the therapeutic interventions, the significant species differences and a critical assessment of so-called nontoxic amounts of Senecio plants in animal fodder with reference to cumulative lethal toxin doses are presented.  相似文献

The monoclonal antibody AM-3K, which was developed using human pulmonary macrophages as the immunogen, immunocytochemically labels most human macrophages except for blood monocytes and dendritic cell populations. AM-3K also shows cross-reactivity in some animal species. To evaluate the usefulness of AM-3K, the present study investigated the detailed distribution of AM-3K-immunopositive macrophages in normal and diseased tissues of dogs, cats, horses, cattle, pigs, and rabbits. Zamboni's solution-fixed, paraffin-embedded sections were the most available for the immunocytochemistry with AM-3K. In all animal species examined, AM-3K labeled most macrophages in splenic red pulp, lymph node sinuses and thymus, and tissue macrophages in the interstitium of various organs and sites such as the kidneys, lungs, heart, pancreas, intestines, and skin. Alveolar macrophages and perivascular microglial cells were also immunoreactive for AM-3K. Interestingly, Kupffer cells of dogs, cats, and horses were labeled for AM-3K, but those of cattle, pigs, and rabbits were not. Furthermore, in tumor tissues and inflammatory lesions such as liver fibrosis and encephalomalacia that were obtained from dogs, infiltrating macrophages were stained with AM-3K, but not all infiltrating macrophages reacted to AM-3K. In addition, only 30-50% of pulmonary and peritoneal macrophages obtained from cats and dogs were reactive for AM-3K. AM-3K did not react with blood monocytes, dendritic cell populations, and osteoclasts. These observations indicate that AM-3K specifically labels most exudate and tissue macrophages in the animal species examined. However, the expression of antigens recognized by AM-3K on macrophages may be dependent on differential maturation stages or different functions evoked by some conditions. AM-3K immunoreaction products were seen on the cytoplasmic membrane of macrophages by immunoelectron microscopy. AM-3K would be useful for detection of macrophage populations in the animal species examined here.  相似文献

In central Brazil, there are two well-defined climatic periods, the rain and the dry season, the latter being marked by massive forage shortages in pastures. In this context, forage conservation such as making hay and silage for herbivores is imperative during the dry season. Grass hay is most commonly used for horses, but sorghum silage is often used for ruminants. The objective was to compare the nutrient digestibility, fecal production, and dry matter intake (DMI) of sorghum silage (Sorghum bicolor l. Moench CV. BRS 655) to that of vaquero hay (Cynodon CV. 90160 CD × CV. Mirage). Twelve Mangalarga Marchador fillies (8 months of age) were divided into two groups: S (sorghum silage, n = 6) and F (vaquero hay, n = 6). The ration was formulated to supply 3.0% body weight in dry matter (DM) per day, with a forage:concentrate ratio of 50:50 on a DM basis. The concentrate used was a corn-based ration formulated to meet or exceed requirements for growth when fed at 1.5% body weight DM to young horses. The animals were housed in individual stalls (3 × 3m) for a period of 25 days, with 21 days for adaptation, followed by 4 days of total collection of feces. During the collection period, an external marker of digestibility, Purified and Enriched Lignin (LIPE), was added to the concentrate as an external marker to determine the apparent digestibility coefficients of DM, crude protein, gross energy, neutral detergent fiber, acid detergent fiber, hemicelluloses, and cellulose. The forage DMI was calculated by subtracting the quantity supplied at each meal minus the amount of forage that was not eaten by the next meal (orts). The measures of digestibility of the sorghum silage were not different (P > .05) from that of vaquero hay. However, DMI of the sorghum silage was lower (P < .05) than the vaquero hay and may be of concern if fed for prolonged periods of time and not supplemented with carefully balanced concentrates. Purified and Enriched Lignin can be used as a marker for evaluation of digestibility of nutrients in weanling horses.  相似文献

Coastal Bermuda grass hay was fed as the sole source of nutrients to determine comparative voluntary intake and nutrient digestibility of hay in yearling and mature horses. Yearling horses consumed dry matter at a higher (P<.10) percentage of their body weight (2.5%) than mature horses (2.0%). digestion of hemicellulose was higher (P<.05) in mature horses, but digestion of dry matter, gross energy, neutral detergent fiber and crude protein was similar between the 2 classes of horses.  相似文献

Horses fed beyond their nutritional requirement and that are physically inactive will develop obesity, which is often accompanied by insulin resistance and heightened risk of laminitis. The use of pharmacologic agents in combination with nutritional restriction may promote weight loss in obese horses unable to exercise because of laminitic pain. This study shows that reducing feed intake of brome grass hay to 75% of ad libitum intake in obese pony mares reduces body weight without induced exercise. Additional supplementation of ractopamine hydrochloride for 6 weeks resulted in a tendency for increased weight loss. Subsequent modulation of obesity-associated hormones, leptin and insulin, as a result of caloric restriction was observed.  相似文献

BACKGROUND: Fourteen horses at a boarding stable in Virginia were diagnosed with hepatic disease and locally grown hay was implicated as the cause. HYPOTHESIS: Panicum dichotomiflorum, the predominant grass species in the hay, is hepatotoxic to horses. ANIMALS: Naturally occurring cases were adult horses of various breeds. Two healthy adult horses and 2 healthy adult sheep were used in feeding trials. METHODS: Blood and liver specimens collected from affected animals during the outbreak were analyzed. Some of the affected animals were treated supportively; the main intervention was hay withdrawal. Feeding trials were not blinded and no treatments were provided. Blood and liver specimens were collected and analyzed throughout the trials. RESULTS: Five affected animals were euthanized, whereas the others recovered. One research horse was euthanized for postmortem examination, and the other research animals recovered after hay withdrawal. All affected animals had evidence of hepatic disease with abnormally high aspartate aminotransferase (AST), sorbitol dehydrogenase (SDH), gamma glutamyl transferase (GGT), and alkaline phosphatase (ALP) activity. Evaluation of liver biopsy specimens disclosed mild lymphocytic and histiocytic inflammation, mild vacuolar change (hydropic degeneration), prominently clumped chromatin, and necrosis of individual hepatocytes. CONCLUSIONS AND CLINICAL IMPORTANCE: Severe hepatotoxicosis developed rapidly after Panicum hay exposure. Patchy hepatocyte necrosis was observed, implicating apoptosis as the mechanism of hepatotoxicosis. Absence of fibrosis in the research animals indicates that immediate withdrawal of Panicum hay should allow all but severely affected animals to recover from acute exposure.  相似文献

To complete a studyaimed at investigating the pattern of the basal activities of liver xenobioticmetabolizing enzymes in major and minor species intended for meat production, microsomal carboxylesterases and some conjugating enzyme activities were determined and compared in liver preparations from horses, cattle, pigs, rabbits and broiler chicks, using the rat as a reference species. Horses and broiler chicks exhibited a lower microsomal carboxylesterase activity towards indophenyl or p-nitrophenyl acetate than that measured in cattle or pig subfractions. Among food-producing species, the rate of glucuronidation of either 1-naphthol or p-nitrophenol was in the order pigs ∼rabbits > horses >> cattle > broiler chicks. The widest variations were observed in the acetylation capacity towards p-aminobenzoic acid or isoniazid, which in rabbits was 3-fold to 11-fold greater than that displayed by any other examined species; low but measurable activities were found in equine and bovine cytosols. The activity of cytosolic glutathione S-transferase (GST) accepting the general substrate 1-chloro-2,4-dinitrobenzene was significantly higher in rabbits, horses and pigs than in rat, broiler chicks and cattle. Finally, an uneven pattern of activity towards the other tested GST substrates – 3,4-dichloronitrobenzene, ethacrinic acid or 1,2-epoxybutane – was observed, possibly reflecting the species-related expression of different GST classes; in this respect, the conjugative capacity displayed by horses was higher than or comparable to that found in the other food-producing species.  相似文献

Chronic inflammatory liver disease regardless of aetiology leads to failing regeneration and fibrosis, ending in cirrhosis. Both in man and in animals this worldwide health problem has no definitive cure. Chronic liver injury causes hepatic stellate cells to proliferate and differentiate into matrix-producing cells. New therapeutic options will be developed upon detailed understanding of the molecular mechanisms driving liver fibrosis. This may lead to new anti-fibrotic therapies which need to be tested in suitable models before application in the veterinary and human clinic. On the other side, to restore the failing regenerative capacity of the diseased liver cells, adult progenitor cells are of interest, as an alternative to whole organ transplantation. In order to find the most suitable large animal model it is important to recognise that the typical histopathological reaction pattern of the liver can differ between mammalian species. It is therefore imperative that specialists in veterinary internal medicine and pathology, being familiar with the diseases and pathologies of the liver in different animal species, are teaming-up in finding the best models for veterinary and human liver diseases. Several large animal models have been mentioned, like pigs, sheep, and dogs. Based on the observations that man and dog share the same hepatopathies and have identical clinical, pathological and pathogenetic reaction patterns during the development of liver disease, the dog seems to be a properly suited species to test new therapeutic strategies for pets and their best friends.  相似文献

Toxicity and metabolism of pyrrolizidine alkaloids   总被引:7,自引:0,他引:7  
Pyrrolizidine alkaloids (PA) are found mainly in plants of three families: boraginaceae, Compositae and Leguminosae. In North America, PA poisoning of livestock is caused primarily by consumption of Senecio and Crotalaria spp. The PA of Senecio spp. cause irreversible hepatic damage; toxicity signs are a consequence of impaired liver function. Crotalaria intoxication leads to pulmonary damage as a primary effect; hepatic effects are less prominent. Large species differences exist in susceptibility to PA toxicosis. Small herbivores such as sheep, goats, rabbits, guinea pigs and other herbivorous laboratory animals are highly resistant to PA toxicity, associated with a low rate of hepatic production of reactive metabolites (pyrroles) and(or) a high rate of activity of detoxifying enzymes. Diester PA common to Heliotropium and Echium spp. are metabolized in the ovine rumen to 1-methyl metabolites, whereas the macrocyclic ester PA of Senecio spp. are not. Exposure to PA results in high concentrations of liver Cu, reduced liver Zn, and abnormal Fe metabolism with hematopoiesis markedly impaired. Pyrrolizidine alkaloid toxicity alters vitamin A metabolism in rats, depressing plasma and liver levels of vitamin A. Synthetic antioxidants in the diet confer protective activity in laboratory animals (e.g., rats, mice) against PA toxicoses. The PA and their metabolites are secreted in the milk of lactating animals, but this probably does not represent a significant human health hazard.  相似文献

Intravenous glucose tolerance testing (0.5 g/kg of body weight) was done on 2 groups of healthy horses maintained with hay (group 1, n = 5) and with hay plus grain supplementation (group 2, n = 5) and on a group of horses with clinically diagnosed pituitary adenoma (group 3, n = 10). Healthy horses showed an immediate increase of plasma glucose concentration after the IV glucose injection, with return of values to base line in 1 hour. Group 3 horses showed resting hyperglycemia and a delayed return of glucose values to base line (3 hours). Group 3 horses showed resting hyperinsulinemia and a feeble (nonsignificant) response to the glycemic stimulus, with gradual decrease of insulin values to base line. In addition to the apparently reduced tissue sensitivity to insulin in group 3 horses, as evidenced by hyperglycemia, hyperinsulinemia, and protracted glucose and insulin curves, the initial decrease in the insulin/glucose ratio indicates that there was secretory deficiency in response to acute IV glucose loading.  相似文献

Objective —To evaluate the seasonal influence, signalment, type of hay consumed, clinical examination findings, and outcome after surgery for horses with ileal impaction. Study Design —A retrospective study. Results —Between 1988 and 1993, 28 horses had surgical correction of ileal impaction. There was a significantly higher rate of ileal impaction during the Fall (September-November, P= .0041). Mean duration of clinical signs of abdominal pain before referral was 15 hours. Transrectal palpation was used to localize the impaction in 11 horses. The ileal impaction was reduced by extraluminal massage aided by admixing of intestinal fluid oral to the impaction or injection of fluids intraluminally and then movement of the ingesta into the cecum alone in 24 horses. A total of 26 horses recovered from surgery; 24 horses were discharged from the hospital and eventually returned to previous use. Two horses had fatal postoperative complications: jejunocec-ostomy dehiscence and development of extensive small intestine adhesions after manual reduction of the impaction. One horse initially treated by manual reduction required jejunocecostomy twice for management of recurrent ileal impaction. Follow-up information was obtained for 21 horses, of which 20 were alive 1 year or longer after surgery. A total of 27 of 28 horses were fed Coastal Bermudagrass hay as the primary type of hay consumed. A total of 9 horses continued to be fed Coastal Bermudagrass hay as the only roughage source, whereas 6 horses were fed Coastal Bermudagrass with at least 50% other hay, and in 6 horses, Coastal Bermudagrass hay was entirely eliminated from the diet. Conclusion and Clinical Relevance—Ileal impactions can be successfully reduced by celiotomy and extraluminal massage and injection techniques to soften the ingesta for passage into the cecum without enterotomy or bypass techniques in most horses. Changes in weather and feeding practices in the Fall may account for an increased risk of ileal impaction in horses in the southeastern United States at that time of year.  相似文献

Concentrations of dioctyl sodium sulfosuccinate (DSS) ranging from three to five times the recommended dosage produced severe diarrhea, rapid dehydration and death in seven horses and 66 guinea pigs when administered experimentally per os. Clinicopathological findings indicated hemoconcentration in both horses and guinea pigs. There was a leucocytosis in the guinea pigs given the highest dosages. In all cases the principal finding at necropsy was extreme fluid distention of the intestinal tract. There was histopathological evidence of epithelial denudation and vascular stasis. The LD50 in the guinea pig was approximately 0.65 g DSS/kg body weight.  相似文献

Despite being largely studied as models of equine nutrition, few published data exist on actual nutritional requirements of Miniature horses. Small equines are particularly prone to obesity, insulin resistance, and the metabolic disorders for which these are risk factors. Improper feed rationing, overfeeding in particular, puts horses at risk for these metabolic problems. Feed manufacturers, however, generally supply feeding recommendations for horses weighing ≥360 kg, whereas the average Miniature horse weighs around 100 kg. Lack of nutritional guidelines for small equines makes it difficult for owners to properly estimate the amount of hay and grain to feed their Miniature horse. The purpose of this study was to determine whether feeding recommendations provided by feed manufacturers for average-sized horses could be linearly extrapolated to Miniature horses. This study surveyed nutritional management of 12 mature Miniature horses that represented the ideal body condition of the breed. Each horse’s hay, grain, and pasture intake was recorded and the average daily digestible energy (DE) intake was calculated from those data. The daily DE intake of the surveyed horses was compared with the extrapolated recommendations published by several different sources. When linearly extrapolated to fit the body weight of a Miniature horse, recommendations significantly (P < .002) misestimated the amount of daily DE that the surveyed horses were consuming. The majority of recommendations overestimated the DE consumed by the Miniature horses per day. However, one source that was designed specifically for Miniature horses, severely (P < .001) underestimated their required DE.  相似文献
