Normal chickens given infectious avian encephalomyelitis virus (IAEV) orally at 1, 7 or 14 days of age developed infectious avian encephalomyelitis (IAE), whereas those dosed with the virus at 21, 28 and 35 days of age did not. Chickens in all of these age groups that had been treated with cyclophosphamide or testosterone developed clinical IAE. Intraperitoneal inoculation of IAEV immunoglobulin at the time of dosing with the virus, or 48 hours later, protected normal and immunosuppressed chickens against the onset of clinical IAE. The titre of IAEV serum neutralising antibody was found to be lower in normal chickens given the virus at 1, 7 and 14 days of age in older chickens. These results suggest a significant role for the humoral immune system in the pathogenesis of IAE and in the development of immunity to the disease.  相似文献

An association was demonstrated between the development of clinical infectious avian encephalomyelitis (IAE), the persistence and titre of infectious avian encephalomyelitis virus (IAEV) in the brain of the chicken, the duration of detectable viraemia and the age of the chicken at the time of infection with the virus. The older the chicken at the time of infection the milder the disease, the lower the virus titre in the brain and the shorter the period of viraemia. IAEV serum neutralising antibody was produced earlier after infection in older chickens, and its detection was associated with decreasing virus titres in the brain and the cessation of detectable viraemia. Treatment of chickens with testosterone in ova, to inhibit the development of antibody synthesis, prevented the onset of age-associated resistance and testosterone treated birds were as susceptible to clinical IAE as baby chickens. The results suggested that the ability to produce IAEV serum neutralising antibody was an important component of age-associated resistance to IAE.  相似文献

The age of the chicken at the time of infection with infectious avian encephalomyelitis virus (IAEV) and the route of the administration of the virus had a marked effect on the development of the clinical disease. Chickens given the virus by intramuscular, intraperitoneal and oral routes exhibited a decreased susceptibility to development of clinical disease with increasing age. Irrespective of age, chickens were consistently susceptible to intracerebral inoculation of IAEV. Chickens infected with the virus at 1-, 7- and 14-days of age developed lower neutralising antibody titres to IAEV than chickens infected at 21 and 28 days. A relationship between the ability to produce specific neutralising antibody to IAEV and susceptibility to the development of clinical disease is discussed.  相似文献

The B1 strain of Newcastle disease virus (NDV-B1), which is nonpathogenic for newly hatched chickens, killed embryos when it was used to inoculate chicken eggs at embryonation day 18. Treatment of NDV-B1 with an alkylating agent, ethylmethane sulfonate (EMS) markedly reduced the pathogenicity of the virus for 18-day-old chicken embryos. Eggs inoculated with the modified virus (NDV-B1-EMS) hatched, and the virus was isolated from lungs and spleen of 1-day-old chickens. The hatched chickens developed antibody to NDV and were protected against challenge exposure (at 4 weeks of age) with a highly virulent GB-Texas strain of NDV. Presence of maternal antibody to NDV in embryonating eggs did not influence the protective ability of NDV-B1-EMS, which also induced protective immunity when administered to 4-week-old chickens. The 50% protective dose of NDV-B1-EMS in maternal antibody-negative and -positive embryos was calculated to be 10.77 and 17.70 embryo 50% lethal doses, respectively. Results of the study indicated that NDV-B1-EMS may be used as an embryo vaccine to protect chickens against Newcastle disease.  相似文献

The level of maternal antibody to infectious bursal disease (IBD) virus in the circulation of one-day-old layer strain chickens was found to be on average, 45% of the antibody titre in their respective dam, while the minimum ELISA titre which protected against a challenge of 1000 CID50 of virus was 400. Maternal antibody was found to disappear from the circulation of these crossbred chickens with a half-life of 6.7 days. From these data it is possible to estimate the ELISA titre necessary in vaccinated hens to provide the desired duration of passive protection; protection being assessed by an ELISA which measures IBD viral antigen in the bursa of Fabricius following challenge.  相似文献

应用电镜技术、血清学检查、生物学试验等方法,从未注射过疫苗的某自然发病鸡群分离出一株新城疫病毒。该分离株对10日龄鸡胚的平均致死时间为116小时,静脉接种致病指数为0.06,其毒力介于新城疫病毒的LaSota株与V4株之间。免疫原性试验结果表明,该分离株具有免疫后无不良反应、免疫后6天开始产生抗体、产生的抗体效价高、对新城疫强毒的攻击能100%保护等特点,是一株良好的新城疫候选疫苗株  相似文献

鸡痘母源抗体对重组鸡痘疫苗免疫效果的影响   总被引:2,自引:1,他引:1  
为了检测抗鸡痘病毒母源抗体对喉气管炎重组鸡痘疫苗的影响,孵化一批来自禽痘病毒高免母鸡的雏鸡,采用ELISA方法检测鸡痘疫苗免疫鸡后代的血清抗体。检测结果表明,雏鸡自孵出2d开始,血清鸡痘病毒抗体水平就开始缓慢下降,到15日龄时下降至临界值,已有部分鸡开始出现抗体阴性反应;到21日龄时,全部被检血清抗体水平均转为阴性。分别于不同日龄对试验雏鸡免疫接种重组鸡痘疫苗,结果只有当鸡体内的鸡痘病毒母源抗体全部为阴性(21日龄)后免疫时才能产生可靠的保护作用,保护率达到80%以上。这说明鸡痘病毒母源抗体对重组鸡痘疫苗的效果有一定的影响,因此重组疫苗合理的首免时间应选择在3周龄以后。  相似文献

疫苗的接触传播是疫苗免疫接种需要考虑的重要因素,为了检测重组鸡痘病毒载体疫苗水平传播的能力,对隔离条件下饲养的SPF鸡用重组鸡痘病毒基因工程疫苗接种,同时设立非免疫对照鸡,饲养期间特意延长清粪时间以增加感染的机会,1个月之后攻击传染性喉气管炎WG株强毒和鸡痘102株强毒,疫苗免疫鸡全部获得保护,而非免疫鸡则全部发病.在试验动物饲养场的自然条件下,将免疫鸡和试验对照两组鸡饲养在同一个鸡舍内,让疫苗毒的传播更接近自然条件.在每个月的攻毒试验中,对照鸡都没有获得对鸡痘和传染性喉气管炎强毒的保护.在疫苗免疫期间进行连续5个月的跟踪检测,同居未免疫鸡没有检测到抗传染性喉气管炎病毒gB抗体.这些实验结果表明抗鸡传染性喉气管炎重组鸡痘病毒基因工程疫苗不能通过接触传播.  相似文献

The immunogenicity of three Newcastle disease virus (NDV) strains--V4, Hitchner B1 and La Sota, was assessed in chickens that had varying levels of maternal antibody to the virus. Chickens were immunised at different ages by the eye drop and aerosol methods. The immune response of chickens to similar doses of the strains was affected by the presence of maternal antibody. Both the level of maternal antibody and the strain of virus used affected the result. Strain La Sota was least affected as it took higher levels of maternal antibody to depress response to it than were required to have a similar effect on Hitchner B1 and V4. Strain V4 was the strain most affected by circulating maternal antibody. The results demonstrated that strain V4 was less immunogenic than Hitchner B1 and La Sota when used in chickens with maternal antibody to NDV.  相似文献

Objectives To assess the efficacy of the vaccination procedure and the effect of the transfer of maternal antibodies to progeny chickens on reovirus pathogenicity.
Design To vaccinate chickens and challenge progeny chickens with high doses of homologous and heterologous viruses.
Procedure High doses of reovirus strains RAM-1, 1091 and 724 were used to induce tenosynovitis lesions. High doses were produced by concentration of viruses grown in cell culture. Then similar doses of viruses were used to challenge immunised chickens progeny.
Result Vaccination of breeding hens with the RAM-1 strain of avian reovirus, which resulted in the passive transfer of neutralising antibody to progeny chickens, completely prevented the development of tenosynovitis in 80% of progeny chickens infected with the homologous virus. Even though multiple injection of hens resulted in broadening of the normal type-specificity of the neutralising antibody response against heterologous serotypes of avian reovirus, only marginal protection against strains of two heterologous serotypes of avian reovirus was obtained.
Conclusions A model for assessing the efficacy of vaccination against avian reovirus strains on clinical signs such as tenosynovitis was developed that overcome the normal low virulence of Australian strains of avian reovirus. Breeding hens can be immunised with Australian strain of avian reovirus with passive transfer of antibody via the yolk to the progeny chickens. Although the neutralising antibody response to three injections of inactivated virus decreased the specificity of the neutralising antibody response against antigenically heterologous strains of avian reovirus, the protective immunity appeared to retain type-specificity.  相似文献

The immunosuppressive effect of infectious bursal disease virus (IBDV) on vaccination against Newcastle disease (ND) was compared among 2-, 3-, and 4-week-old chickens inoculated with the highly virulent IBDV field isolate 90-11 and the reference serotype 1 strain GBF-1. In all age groups, isolate 90-11 severely suppressed antibody response to ND vaccination and protective vaccinal immunity against ND. In contrast, chickens inoculated with strain GBF-1 and vaccinated with ND vaccine were well protected from the ND virus challenge. The mitogenic response to phytohemagglutinin of splenic lymphocytes from chickens inoculated with isolate 90-11 or strain GBF-1 was significantly lower than that of uninoculated controls. There was no difference between the two inoculated groups in responsiveness, although lymphocyte depletion in the thymus was more severe in chickens inoculated with isolate 90-11 than in chickens inoculated with strain GBF-1.  相似文献

A recombinant fowlpox virus (rFPV) coexpressing the Newcastle disease virus (NDV) fusion and hemagglutinin-neuraminidase genes and infectious laryngothracheitis virus (ILTV) glycoprotein B gene was constructed. This virus was then evaluated for its ability to protect specific-pathogen-free (SPF) chickens against clinical symptoms and death after challenge by virulent NDV and ILTV. SPF chickens were grouped and vaccinated with the rFPV and commercial NDV (La Sota) and ILTV attenuated live vaccine (Nobilis ILT), respectively. After challenge with NDV 10 days postvaccination, 70% of chickens vaccinated with rFPV were protected from death, whereas 100% of the commercial NDV-vaccinated chickens were protected from death. In contrast, 100% of the unvaccinated chickens died after challenge. After challenge with ILTV, both the rFPV and commercial ILTV-vaccinated chickens were completely protected from death and 70% of chickens were protected from respiratory signs. In comparison, 100% of the unvaccinated chickens developed severe respiratory disease and 10% of chickens died. The protective efficacy was also measured by the antibody responses and isolation of challenge viruses. Results showed that this rFPV could be a potential vaccine for preventing NDV and ILTV by a single immunization.  相似文献

Lachrymal fluid specific IgG and IgA were detected by ELISA in chickens with specific maternal antibodies and in chickens free of antibodies (SPF), after vaccination at 1 day of age with the H-120 vaccine strain of infectious bronchitis virus. Samples were obtained at 3 day intervals and until Day 37 of age. Optical densities were in all instances low but significant differences could be detected within and between the experimental groups. Both class-specific immunoglobulins showed a similar kinetic pattern. Nevertheless, the SPF group increased its IgA level on Day 13 while chickens with maternal immunity increased their level on Day 16. The antibody levels of both IgG and IgA were also different, being higher in the SPF group. In both chicken groups, higher levels of IgA than IgG were detected.  相似文献

利用表达 H 5亚型禽流感病毒 (AIV)血凝素基因的重组鸡痘病毒 (r FPV- HA)以不同剂量免疫 1日龄 SPF鸡、有或无母源抗体 (FPV、AIV H5)的商品鸡 ,并于免疫后 2 1d利用同亚型 AIV通过肌肉注射进行致死性攻击 ,通过检测免疫后 HI抗体应答、比较攻毒后发病率和死亡率评价免疫剂量和母源抗体对 r FPV- HA免疫效力的影响。结果发现 ,免疫后 2 1d,15 %~ 2 0 %的 SPF鸡和无母源抗体商品鸡可检出 HI抗体 ,而含母源抗体商品鸡检测不到 HI抗体。利用H5亚型 AIV致死性攻击后 ,10 3~ 10 6 PFU的 r FPV- HA可保护 95 %~ 10 0 %的 SPF鸡和无母源抗体商品鸡抵御强毒攻击 ,使之免于发病和死亡 ;而不同剂量 r FPV- HA接种的含母源抗体商品鸡有 80 %~ 90 %发病和死亡。结果表明 ,在较宽的免疫剂量范围内 ,r FPV- HA对 SPF鸡和无母源抗体商品鸡可提供良好的保护 ,显示出一定的应用前景 ;母源抗体影响 r FPV- HA诱导的免疫应答 ,且提高免疫剂量亦不能克服其干扰作用 ,这提示在实际应用中需优化免疫程序 ,避免母源抗体干扰。  相似文献

Laboratory and field tests were conducted to evaluate the immunogenicity of a live-virus vaccine against infectious laryngotracheitis (ILT) of chickens that was prepared using tissue-culture-modified strain C7. Eighty-three percent or more of chickens vaccinated by the ocular (OC) or intranasal (IN) route with 10(4.75) TCID50 of the attenuated strain C7 at 50 days of age were protected against challenge with a virulent strain of ILT virus without showing any clinical signs for 4 weeks post-vaccination (PV). When vaccine was administered by aerosol, however, only 65% of vaccinated chickens were protected against challenge. Fifty-seven percent of chickens vaccinated at 70 days of age maintained immunity for 6 months PV. Immune response in younger chickens was inferior to that in older ones. In the field trials, clinical observation revealed no adverse reactions caused by the vaccination, and 60% or more of broilers and 80% or more of layers vaccinated by the OC route were protected against challenge at 4 weeks PV. These results confirmed the safety and efficacy of the vaccine.  相似文献

在流行病学调查中分离到1株病毒,经鉴定为伪狂犬病弱毒株,定名为F971株。分离病毒经克隆纯化后测得其毒价为10^7.59TCID50/ml,通过细胞中和试验表明分离病毒能也有效地被猪伪狂犬病毒闽A株阳性血清中和。病毒在电镜下可以清楚地观察到囊膜及外周纤突。分离株对3日龄乳鼠有一定的致病力,但对家兔、3日龄乳猪及妊娠母猪都有很高的安全性。用不同的剂量10^0、10^-1、10^-2肌肉注射3日龄乳猪后14天用10^5.7TCID50伪狂犬病强毒攻击,所有试验仔猪均得到保护。用分离株免疫母猪,其后代可获高滴度的母源抗体,15日龄的仔猪能抵御10^5.7TCID50强毒的攻击。用ELISA普查试剂盒测定免疫猪抗体,结果均为阳性,而用g^1-ELISA试剂盒测定抗体时,结果均为阴性。证明分离株具有缺损g^1糖蛋白的特性。综合上述特性,确定F971为1株g^1糖蛋白缺损的猪伪狂犬病弱毒株。  相似文献

An international effort (sponsored by the Australian Centre for International Agricultural Research) is being made to develop oral vaccines that will protect village chickens against Newcastle disease. The vaccines being used are derivatives of the avirulent Australian V4 strain that have been selected for enhanced heat resistance. The present study, undertaken in Sri Lanka, used local processed (parboiled) rice as a vehicle for the vaccine. Chickens receiving two doses of vaccine on cooked, parboiled rice were completely protected against contact challenge with the virulent SL 88/1 Sri Lankan strain of Newcastle disease virus Chickens kept in contact with these vaccinated chickens were similarly protected. Lower levels of protection were achieved with vaccine given on uncooked parboiled rice. V4 vaccine administered intranasally also gave complete protection. Serums from vaccinated chickens that survived challenge were tested for haemagglutination-inhibition antibodies, using both vaccine virus and challenge virus as antigens. One-week-old chickens were vaccinated with live or formalin-killed temperature-sensitive (TS) Mycoplasma gallisepticum (MG) either intranasally (IN) or subcutaneously (SQ). Live TS MG protected chickens against S6 strain challenge directly into the air sacs, regardless of route of vaccination. Killed MG, however, protected chickens only when administered SQ. Antibody to MG was detected in sera and in the tracheal and air-sac washings of only the chickens given live vaccine IN. The antibody present in tracheal and air-sac washings may be one of the mechanisms that play a role in resistance to MG challenge.  相似文献

The efficacy of four different commercial live vaccines (vaccines A, B, C, and D) against the infectious laryngotracheitis virus (ILTV) was assessed in specific-pathogen-free (SPF) chickens. SPF chickens were vaccinated intraocularly at 6 wk old with ILTV live vaccines and were challenged intratracheally with the N91B01 strain of virulent Korean ILTV 2 wk after vaccination. The immunity against ILTV live vaccines was assessed by the incidence of latent infection by the challenge virus in the chickens' tracheas and trigeminal ganglia, the reisolation rate of the challenge virus, and the clinical signs in the chickens challenged with the N91B01 strain of ILTV. The latent infection in chickens was assessed by nested polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Our data showed that the clinical signs and challenge virus isolation were negative in all chickens receiving four difference commercial ILTV live vaccines. The viral DNA of the vaccine strain, but not that of the challenge virus, was detected in chickens vaccinated with vaccine A by nested PCR-RFLP. The viral DNAs of both the vaccine and challenge strains were detected from chickens vaccinated with vaccines B, C, and D. This study showed that only vaccine A can protect chickens from latent infection with the field virulent ILTV. We speculate that the efficacy of infectious laryngotracheitis live vaccines to protect chickens from latent infection with virulent ILTVs can be assessed by nested PCR-RFLP analysis.  相似文献

蛋用型海兰褐母鸡在用网状内皮增生病病毒(REV)细胞适应毒REV-C99(p30)免疫接种后,均在2周内产生REV特异性抗体。来自免疫种鸡的雏鸡在7日龄内100%(10/10)REV母源抗体阳性。分别在有母源抗体和没有母源抗体的1日龄鸡接种与疫苗株同源的低传代毒REV-C99(p3)或异源的REV中国野毒株HA9901(p5),以此比较母源抗体对同源和异源REV病毒感染造成的免疫抑制的预防作用。结果表明,REV母源抗体能同等有效地预防同源和异源REV感染造成的对H5和H9禽流感灭活疫苗HI抗体反应的抑制作用。  相似文献
