Human immunoglobulin G (IgG) concentrates are immune-modulating,
anti-inflammatory plasma-derived products. Clinical studies in recent years have suggested
that IgG attenuates neuropathic pain. In this study, effects of sulphonated IgG on the
development and maintenance of a mechanical allodynia-like response were examined in mice
with neuropathic pain induced by a partial sciatic nerve ligation (PSL). When sulphonated
IgG (400 or 1,000 mg/kg/day, i.p.) was administered for 5 days, from 1 day before surgery
to post-operative day (POD) 3, the development of a mechanical allodynia-like response was
attenuated. On the other hand, sulphonated IgG had little effect on the maintenance of a
mechanical allodynia-like response when administered for 5 days, from POD 11 to POD 15, at
which time a mechanical allodynia-like response had already been developed. To explore the
mechanism of sulphonated IgG, the mRNA expression of inflammatory cytokines was evaluated
in the injured sciatic nerve. Sulphonated IgG (1,000 mg/kg/day, i.p.) that was
administered for 3 days, from 1 day before surgery to POD 1, significantly attenuated the
up-regulation of tumor necrosis factor-α and monocyte chemotactic protein-1 mRNAs on POD
1. These results suggest that prophylactic treatment with sulphonated IgG attenuates the
development of mechanical allodynia-like response by inhibition of inflammatory cytokine

Dourine, caused by infection with Trypanosoma equiperdum, is one of the trypanosomiasis in equids. The clinical course of dourine is long-term, ranging from 1–2 months to several years. Since the pathogenesis of dourine has not yet been elucidated, experimental studies using mouse infection models are needed. Although mice are not susceptible to most T. equiperdum strains, some strains can infect mice. Even in such strains, infected mice develop rapidly transient parasitemia and die within 2–8 days. Therefore, mice experimentally infected with these T. equiperdum strains are not suitable for mouse infection models to analysis the pathogenesis of dourine. A sequential method of isolating parasites from dourine-affected horses and adapting them to in vitro cultures using soft agarose media was recently developed. Various T. equiperdum strains adapted to in vitro conditions have been established using this technique. We used one of these strains, the T. equiperdum IVM-t2 strain. In the present study, T. equiperdum IVM-t2 strain inoculated mice developed periodic parasitemia during the experimental period of 60 days. Histopathologically, vaginitis and dermatitis were observed. These findings were comparable to those of dourine-affected horses. Therefore, mice infected with T. equiperdum IVM-t2 strain may be a valuable tool for pathological, immunological, and parasitological in vivo research, and will contribute to investigations on the mechanisms underlying the disease process and the host-parasite relationship. 相似文献

The aims of this pilot study were to evaluate the feasibility and efficacy of high-dose hypofractionated volumetric modulated arc radiotherapy (VMAT) applied to whole pelvic region radiotherapy (WPRT) with multilevel simultaneous integrated boost (MLSIB) combined with piroxicam and chemotherapy for the treatment of canine transitional cell carcinoma (TCC) of the lower urinary tract with muscle invasion TCC. Twelve dogs were enrolled, according to stage, in two groups: group 1, TCC confined to the urinary tract; group 2, TCC with metastasis. The planning target volume dose was tailored from 36 to 42 Gy in 6 fractions. All dogs were prescribed piroxicam and radiosensitizing carboplatin, and six received chemotherapy after radiotherapy. Serial follow-ups with computed tomography and magnetic resonance imaging were performed. Disease control and toxicity effects were evaluated according to the Response Evaluation Criteria in Solid Tumors and Veterinary Radiation Therapy Oncology Group criteria. The treatment was well tolerated, and no high-grade side effects were reported. The median overall survival times for groups 1 and 2 were 1,230 and 150 days, respectively. A considerable percentage of patients in group1 (50%) were still alive at the time of writing this paper, and a longer follow-up could enable a more accurate survival analysis. This preliminary analysis shows that VMAT applied to the WPRT with MLSIB is an effective and safe option for dogs with lower urinary TCC, although the presence of metastases worsens the prognosis. 相似文献

Excessive stress response causes disability in social life. There are many diseases caused by stress, such as gastrointestinal motility disorders, depression, eating disorders, and cardiovascular diseases. Transient receptor potential (TRP) channels underlie non-selective cation currents and are downstream effectors of G protein-coupled receptors. Ca2+ influx is important for smooth muscle contraction, which is responsible for gastrointestinal motility. Little is known about the possible involvement of TRP channels in the gastrointestinal motility disorders due to stress. The purpose of this study was to measure the changes in gastrointestinal motility caused by stress and to elucidate the mechanism of these changes. The stress model used the water immersion restraint stress. Gastrointestinal motility, especially the ileum, was recorded responses to electric field stimulation (EFS) by isometric transducer. EFS-induced contraction was significantly reduced in the ileum of stressed mouse. Even under the conditions treated with atropine, EFS-induced contraction was significantly reduced in the ileum of stressed mouse. In addition, carbachol-induced, neurokinin A-induced, and substance P-induced contractions were all significantly reduced in the ileum of stressed mouse. Furthermore, the expression of TRPC3 was decreased in the ileum of stressed mouse. These results suggest that the gastrointestinal motility disorders due to stress is associated with specific non-selective cation channel. 相似文献

Pancreatic lipase immunoreactivity (Spec fPL) is currently considered to be the
most accurate blood test for the diagnosis of feline pancreatitis. In this study, we
measured lipase activity in cats using a newer catalytic lipase assay of dry-chemistry
system (FDC-v-LIP) to determine the reference range and compared the results with those
for Spec fPL. Based on the results of healthy cats, the reference range of FDC-v-LIP was
determined to be less than 30 U/l. FDC-v-lip did not show a strong
correlation with Spec fPL in cats with various diseases, which resulted in the low
sensitivity and positive predictive value. However, the relatively high (>90%)
specificity and negative predictive value indicated that FDC-v-LIP could be a useful
patient-side screening test for the exclusion of feline pancreatitis. 相似文献

The antibody response in horses inoculated with 2 doses of a live equine herpesvirus type 1 vaccine with different vaccination intervals (1 to 3 months) was evaluated with regard to the persistence of virus-neutralizing (VN) antibodies. The durations for which the geometric mean VN titers were maintained significantly higher than those before the first vaccination (P<0.05) were up to 5 months in horses that received the vaccination with a 1-month interval (n=17) and 7 months for those that received it with a 2-month (n=17) or 3-month interval (n=14 or 17). The vaccination program with the 2-month interval was the most effective in maintaining VN antibodies for a long duration with the smallest gap of antibody decline between the first and second vaccinations. 相似文献

The experiment was conducted to investigate the effects of trace amounts of antibiotic on growth performance, diarrhea, systemic immunity, and intestinal health of weaned pigs experimentally infected with an enterotoxigenic Escherichia coli. Weaned pigs (n = 34, 6.88 ± 1.03 kg body weight [BW]) were individually housed in disease containment rooms and randomly allotted to one of the three dietary treatments: nursery basal diet (CON) and two additional diets supplemented with 0.5 or 50 mg/kg carbadox to the nursery basal diet (TRA or REC), respectively. The experiment lasted 18 d with 7 d before and 11 d after the first E. coli inoculation. The E. coli F18 inoculum was orally provided to all pigs with a dose of 1010 colony-forming unit (CFU)/3 mL for three consecutive days. Fecal and blood samples were collected on day 0 before inoculation and days 2, 5, 8, and 11 postinoculation (PI) to test the percentage of β-hemolytic coliforms in total coliforms and complete blood cell count, respectively. Sixteen pigs were euthanized on day 5 PI, whereas the remaining pigs were euthanized at the end of the experiment to collect the jejunal and ileal mucosa and mesenteric lymph node for gene expression and bacterial translocation, respectively. Pigs in REC had greater (P < 0.05) final BW and lower (P < 0.05) overall frequency of diarrhea compared with pigs in the CON and TRA groups. Pigs in TRA had the lowest (P < 0.05) average daily gain and feed efficiency from day 0 to 5 PI, highest (P < 0.05) percentage of β-hemolytic coliforms in fecal samples on days 2 and 5 PI, and greatest (P < 0.05) bacterial colonies in mesenteric lymph nodes on day 11 PI compared with pigs in the CON and REC groups. Pigs in TRA had the greatest (P < 0.05) neutrophils on day 5 PI and higher (P < 0.05) white blood cell counts and lymphocytes than other groups on day 11 PI. Pigs in TRA had the greatest (P < 0.05) serum C-reactive protein on days 2 and 5 PI and serum tumor necrosis factor-α on day 5 PI, compared with pigs in the CON and REC groups. Pigs fed REC had increased (P < 0.05) mRNA expression of zona occludens-1 (ZO-1) and occludin (OCDN) and reduced (P < 0.05) interleukin-1 beta (IL1B), interleukin-6 (IL6), and tumor necrosis factor-alpha (TNFA) in ileal mucosa on day 5 PI, compared with the CON, whereas TRA upregulated (P < 0.05) mRNA expression of IL1B, IL6, and cyclooxygenase-2 (COX2) in the ileal mucosa on day 11 PI, compared with the REC. In conclusion, trace amounts of antibiotic may exacerbate the detrimental effects of E. coli infection on pig performance by increasing diarrhea and systemic inflammation of weanling pigs. 相似文献

The common marmoset (Callithrix
jacchus) is now widely used in various
research fields, including toxicology. However,
information about the background pathology of this
species is scarce. Here, we report a case of
rhabdomyosarcoma that spontaneously occurred in a
common marmoset. A 44-month-old male common
marmoset was euthanized due to bilateral hind limb
paralysis. At necropsy, a 2×2×5-cm intramuscular
mass was observed in the lower right back.
Histologically, the mass was mainly composed of
interlacing bundles of spindle-shaped tumor cells.
Immunohistochemically, the tumor cells were
positive for myogenin, desmin, vimentin and
alpha-smooth muscle actin. Ultrastructurally, the
tumor cells contained bundles of myofilaments with
Z-band-like structures. Thus, the tumor was
diagnosed as a rhabdomyosarcoma. To our knowledge,
this is the first report of spontaneous
rhabdomyosarcoma that was definitely diagnosed in

The aim of this study was to evaluate the immune responses to intranasal and
intrapulmonary vaccinations with the attenuated Mycoplasma hyopneumoniae
(Mhp) 168 strain in the local respiratory tract in pigs. Twenty-four
pigs were randomly divided into 4 groups: an intranasal immunization group, an
intrapulmonary immunization group, an intramuscular immunization group and a control
group. The levels of local respiratory tract cellular and humoral immune responses were
investigated. The levels of interleukin (IL)-6 in the early stage of immunization
(P<0.01), local specific secretory IgA (sIgA) in nasal swab samples
(P<0.01); and IgA- and IgG-secreting cells in the nasal mucosa and
trachea were higher after intranasal vaccination (P<0.01) than in the
control group. Interestingly, intrapulmonary immunization induced much stronger immune
responses than intranasal immunization. Intrapulmonary immunization also significantly
increased the secretion of IL-6 and local specific sIgA and the numbers of IgA- and
IgG-secreting cells. The levels of IL-10 and interferon-γ in the nasal swab samples and
the numbers of CD4+ and CD8+ T lymphocytes in the lung and hilar
lymph nodes were significantly increased by intrapulmonary immunization compared with
those in the control group (P<0.01). These data suggest that
intrapulmonary immunization with attenuated Mhp is effective in evoking
local cellular and humoral immune responses in the respiratory tract. Intrapulmonary
immunization with Mhp may be a promising route for defense against

Eight of 9 juvenile raccoons at a rehabilitation center died without obvious prior clinical signs. Gross changes were unremarkable except for mildly distended intestines. Microscopically, crypt loss, distension, necrosis, and regeneration with intranuclear viral inclusions were observed in the small intestine, with marked lymphoid depletion and necrosis in Peyer patches and mesenteric lymph nodes. Immunohistochemistry with a canine parvovirus antibody showed intensive signals of parvoviral antigens in the crypts and lymphoid germinal centers. Metagenomic sequencing allowed assembly of a complete parvoviral genome with >99% identity to canine parvovirus 2a, as well as Salmonella enterica subsp. enterica. Also, S. enterica subsp. enterica serovar Thompson with multiple antimicrobial resistance was isolated from the intestinal contents. Concurrent infection with parvovirus and Salmonella should be included as a differential diagnosis in raccoons with sudden death. 相似文献

To assess factors for canine skin extensibility, our study investigated associations between the dogs’ skin extension index and the following factors, gender, age, neuter status, weight, coat color and six coat color related gene polymorphisms. Swab samples were collected from 69 toy poodles to extract DNA. The skin extension indices of the lower back and the neck were measured using the following formula: vertical height of the skin fold divided by body length multiplied by 100. The dogs’ age, weight, gender, neuter status and coat color were also recorded, as well as polymorphisms of the following six selected coat color related genes, Melanocortin 1 receptor, Tyrosinase-related protein 1, Melanophilin, Canine β-defensin-1, Major Facilitator Superfamily Domain Containing 12 and Agouti-signaling protein (ASIP). Univariable analysis showed there was a meaningful association between the lower back skin extension index and both gender and age (P<0.001 and P=0.048, respectively). Also, there was a possible association between the lower back skin extension index and ASIP Single nucleotide polymorphism (SNP) (R96C) (P=0.078). Linear model analysis showed there was a significant association between the lower back skin extension index and gender (P<0.001), and there was a tendency of the association between the lower back skin extension index and ASIP SNP (R96C) (P=0.098). In addition, there was an association between gender and age for the skin extension index. (P=0.048). Therefore, these results suggest that a greater risk of skin extensibility in toy poodle could be related to being female and the ASIP SNP (R96C), because these factors were associated with higher lower back skin extension index. 相似文献

A 4-year-2-month-old female Japanese domestic cat was diagnosed with lymphangiosarcoma
through tissue biopsy of an amputated leg. Two months later, the cat was euthanized, and
postmortem findings revealed edema, and bruising at the caudal region of the trunk,
pulmonary hemorrhage, pulmonary nodules and mediastinal lymphadenopathy. Microscopically,
neoplastic tissues were observed in the dermis and subcutis of the trunk, lung,
mediastinal lymph nodes, diaphragm, omentum and mesentery. The tumor cells were spindle to
polygonal-shaped with nuclear pleomorphism aligning along pre-existing collagen bundles
and forming irregular vascular channels in which the erythrocytes were rarely observed.
These cells were immunopositive for vimentin, von Willebrand factor and CD31. Based on the
histopathological and immunohistochemical features, the neoplasia was diagnosed as
lymphangiosarcoma with systemic metastases. 相似文献
