Effect of testing environment on adrenal cortical responses to an injection of ACTH in clinically normal dogs was examined in three locations, presumably of increasing order of stress elicitation: in a home; veterinary hospital (VH), 4 hours in a cage; and VH, overnight in a cage. Basal cortisol (hydrocortisone) values for plasma were significantly lower (P less than 0.001) for the home group (1.8 microgram/dl) when compared with values for the VH, 4-hour cage (3.8 microgram/dl) or the VH, overnight cage (3.9 microgram/dl) groups. However, significant differences (P greater than 0.05) were not observed 2 hours after ACTH admininstration for the home group (13.7 microgram/dl); VH, 4-hour cage group (14.8 microgram/dl); or VH, overnight cage group (16.0 microgram/dl). Responses of individual dogs were consistent (P less than 0.005). The testing environment did not markedly affect results of adrenal cortical function tests for dogs when ACTH stimulation was utilized. The response of dogs to ACTH, as monitored by immunologic assay techniques (competitive protein-binding assay or radioimmunoassay), was consistent and was useful as a diagnostic aid for adrenal malfunction.  相似文献

OBJECTIVE: To evaluate adrenal sex hormone concentrations in neutered dogs with hypercortisolemia. DESIGN: Case series. ANIMALS: 11 neutered dogs with hypercortisolemia. PROCEDURE: Serum samples obtained before and 1 hour after administration of ACTH were evaluated for concentrations of cortisol, progesterone, testosterone, dehydroepiandrosterone sulfate or androstenedione or both, and 17-hydroxyprogesterone. RESULTS: For all dogs, concentrations of 1 or more adrenal sex hormones were substantially greater than reference range values before or after administration of ACTH. Testosterone concentration was not greater than reference range values in any of the dogs. CONCLUSIONS AND CLINICAL RELEVANCE: Results emphasize the importance of ruling out hypercortisolemia before measuring adrenal sex hormone concentrations as a means of diagnosing adrenal hyperplasia syndrome (alopecia X) in dogs.  相似文献

Previously, GH response to GHRH challenge at weaning has been shown to be indicative of ADG during a standard postweaning growth performance test in Angus cattle. In this study, we tested the hypothesis that GH response to GHRH before weaning would predict postweaning ADG. Bulls with the highest and lowest GH responses to GHRH over a 3-yr period, relative to their contemporaries, were used as sires, to allow for examination of the persistence of GH response to GHRH through selection. The selected calves in this study were sired by one of four Angus bulls chosen based on their GH response to GHRH (high response, n = 2; low response, n = 2). Forty-nine Angus calves (bulls, n = 24; heifers, n = 25) were challenged with GHRH at approximately 60, 105, and 150 d of age and at weaning (219 d; SD = 25). Blood samples were taken immediately prior to and 10 min following an i.v. clearance dose of 4.5 microg of GHRH/100 kg BW and, 2 h later, immediately prior to and 10 min following a challenge dose of either 1.5 or 4.5 microg of GHRH/100 kg BW. Two hours later, the procedure was repeated, with each calf receiving the other challenge dose. Body weight was measured every 28 d and ADG was calculated over a 140-d growth performance test (heifers and bulls maintained separately). Data were log-transformed for statistical analyses. In the selected bulls and heifers, response of GH to 1.5 microg of GHRH/100 kg BW at 60 and 105 d of age was positively related (P < 0.05) to postweaning ADG. Response to 4.5 microg of GHRH/100 kg BW at 105 d of age and at weaning was positively related (P < 0.01) to postweaning ADG. Inclusion of sire in the analysis improved the relationship between GH response and ADG for calves of sires with high GH responses from R2 = 0.18 (P = 0.01) to R2 = 0.33 (P = 0.02). When the GH response to GHRH of the unselected calves at weaning was added to the data from the selected animals and analyzed, the GH response of the bulls was related to postweaning ADG (R2 = 0.09; P = 0.04). In conclusion, GH response to GHRH as early as 60 d of age is indicative of postweaning ADG in beef cattle. In addition, the relationship between GH response to GHRH and postweaning ADG is improved with selection for greater GH response to GHRH.  相似文献

Plasma fibrinogen concentration was measured in beef calves at various points within the system presently used to assemble, market and transport calves from one production point to another in order to determine the effect of the stresses encountered. A short haul of 160 km immediately after weaning did not significantly elevate fibrinogen concentration above the pretransit values. Yearling steers transported 400 km and confined in unfamiliar surroundings for 15 h did have an elevated (P less than 0.01) concentration of fibrinogen, but this increase was not significantly different from that of steers which were confined but not transported, thus confinement may be a significant portion of the stress associated with transit. The change in plasma fibrinogen concentration during assembly and transit was dependent upon the farm from which the calves originated. The magnitude of the change in fibrinogen concentration as a result of assembly and transit varied between the years studied. In one year pretransit assembly for ten days resulted in a higher fibrinogen concentration before and after transit than assembly for four days, but no difference was noted between the two groups in the second year. Bovine plasma fibrinogen concentration does increase in response to the stresses associated with assembly and transit. The stress of fasting and housing in unfamiliar surroundings also increase bovine plasma fibrinogen concentration and are present in the assembly and transit system. These two stresses may account for a majority of the stress associated with marketing and transit. The response of beef calves to the marketing and transit system varied between years.  相似文献

Four-month-old beef calves were implanted with 36 mg zeranol, and their growth compared with untreated calves. Treated steer and heifer calves gained 10 kg more bodyweight to weaning at 7.5 months of age. The trial was conducted at 2 research centres and calves at both centres showed significant responses for the first 45 days (P less than 0.001) after treatment but during the following 30 days only calves at one centre showed a continued response (P less than 0.05). No differences in growth rate occurred between treated and untreated calves over the final 30 days before weaning at either centre. The backfat measurement of untreated animals increased more than that of the implanted animals (P less than 0.05) at one centre but not at the other.  相似文献

Albendazole (methyl 5-propylthio-1 H-benzimidazol-2-yl carbamate) was used as an anthelmintic in a 3-yr study involving 578 beef cows and 438 nursing calves. Infection levels for nematodes, coccidia, and tapeworm were relatively low throughout the 3-yr period. Eggs per gram of feces in cows and calves were lower (P less than .01) 2 wk posttreatment but were not different 5 mo later, when calves were weaned. Cow weight gain, rate and time of conception, and adjusted calf weaning weights were not affected significantly by deworming of either cows or calves. Level of nematode infection measured as eggs per gram of feces was higher (P less than .01) in younger cows than in mature cows. Although deworming with Albendazole lowered (P less than .01) nematode infection levels, no responses were observed in cow or calf performance.  相似文献

The objective herein was to estimate heterosis and breed effects in purebred and crossbred Romosinuano, Brahman, and Angus calves on acute phase protein response to weaning and transportation. Calves (n = 1,032) were weaned in September of 2002, 2003, and 2004 at approximately 7 mo of age. Approximately 28 d after weaning, steer calves (n = 482) were transported 1,800 km (20 h) to Oklahoma. Concentrations of 3 acute phase proteins (ceruloplasmin, fibrinogen, and haptoglobin) were measured in blood samples. Calves (steers and heifers) were sampled at weaning, and 24 and 72 h postweaning. For separate analyses, steers sent to Oklahoma were sampled before shipment, upon arrival, and 24 and 72 h after arrival. Combinations of the following fixed effects were investigated: sire breed, dam breed, sampling time, birth location, calf sex (weaning only), year, cow age, and interactions. Effects of special interest were sire breed x dam breed as an indication of breed group of calf, and the interaction of sire and dam breeds with sampling time. Weaning age and BW were investigated as linear and quadratic covariates. Sire of calf within sire breed was a random term. The correlation structure of repeated measures was determined by comparison of information criterion values for different structures within each analysis. In general, plasma acute phase protein concentrations in weaned calves increased with sampling time. Concentrations in the transported steers increased through sampling at 24 h after arrival, and were lower at 72 h. Significant estimates of heterosis were detected for Brahman-Angus haptoglobin concentrations at weaning (0.38 +/- 0.14 mg/dL x 100; 44%), and for Romosinuano-Angus fibrinogen concentrations at weaning (11.4 +/- 5.5 mg/dL; 10%) and in transported steers (22.5 +/- 8.4 mg/dL; 20%). The direct effect of Romosinuano was to increase (P <0.004) ceruloplasmin concentrations of weaned calves (4.1 +/- 0.9 mg/dL) and of transported steers (3.9 +/- 1.3 mg/dL). The direct effect of Angus was to lower ceruloplasmin concentrations in weaned calves (-3.9 +/- 1.2; P = 0.001). Significant maternal effects were detected at weaning for ceruloplasmin concentrations in Romosinuano (-1.4 +/- 0.5 mg/ dL) and Angus (1.6 +/- 0.7 mg/dL) and fibrinogen concentrations in Brahman calves (-17.7 +/- 8.8 mg/dL). These data imply that acute phase protein concentrations in response to weaning and transportation are impacted by cattle breed.  相似文献

The adrenal cortex is believed to be implicated in the high incidence of abortion in the Angora goat. Stimulation testing with adrenocorticotrophic hormone (ACTH) was used to assess the adrenal cortical function in 5 Angora does from herds with a history of abortion and 5 non-Angora does. An acute test involving a single intramuscular (i.m.) injection of 0.25 mg of synthetic ACTH was given during anoestrus, at mid-oestrus, on day 90 and on day 120 of gestation. Blood samples were collected from the jugular vein at 30 min intervals for 1 h before and 5 h after injection. Cortisol concentrations rose within 30 min and returned to baseline values within 3.5 h. Cortisol production was lower (p<0.01) in the pregnant state compared to the non-pregnant state in both groups. Production of cortisol was consistently lower (p<0.05) in the Angora does compared to the non-Angora does during anoestrus and pregnancy and marginally so at mid-oestrus. A chronic stimulation test involving once daily injections of 0.5 mg of a depot form of ACTH i.m. for 7 days commencing on day 90 of pregnancy was also conducted. Cortisol concentrations rose to reach a peak on the third day of treatment in both groups. The values then declined in the Angora does despite continued ACTH treatment, while those for the non-Angora does exhibited a second peak. During and following this treatment, two non-Angora does delivered live kids (day 95, day 120). Out of 7 Angora pregnancies, one Angora doe aborted two dead fetuses at day 116. No significant difference in the cortisol response in the acute test was detected between the animals that aborted and their respective cohorts, but the two non-Angora does that aborted had significantly lower cortisol concentrations during depot ACTH administration. Progesterone and oestradiol levels did not differ between Angora and non-Angora animals during pregnancy or on the test days. The results suggest that the steroidogenic response of the adrenal cortex to ACTH stimulation is significantly less in Angora does with a history of abortion than it is in non-Angora does and support the view that the Angora goat would make a more limited adrenal cortical response to a stressful occurrence during pregnancy.  相似文献

The objective of this experiment was to investigate the effects of injectable trace minerals on humoral responses of calves receiving a viral vaccination. Beef steer calves (n = 99; average BW = 316 ± 4.2 kg), seronegative for bovine herpesvirus-1 (BHV-1) and bovine viral diarrhea virus, genotypes 1 and 2 (BVDV-1 and BVDV-2), were sourced from 2 locations. All calves, except 15 non-vaccinated (sentinel) calves, received a single dose of a multivalent modified live vaccine (Titanium 5; AgriLabs, St. Joseph, MO) containing BHV-1, BVDV-1, BVDV-2, bovine parainfluenza virus type 3, and bovine respiratory syncytial virus. Among the vaccinated calves, 2 treatments were concurrently and randomly applied on the basis of initial serum Se status and BW, including 1) injectable trace mineral supplement (ITM; n = 42; 7 mL subcutaneous.; MultiMin, Fort Collins, CO) containing 15, 40, 10, and 5 mg/mL of Cu, Zn, Mn (all as disodium EDTA salts), and Se (as Na selenite) or 2) saline-injected control (Control; n = 42). As a measure of humoral immunity, neutralizing antibody titers were measured on d 0, 14, 30, 60, and 90, relative to vaccine administration. All calves were seronegative for each of the 3 viruses on d 0, and sentinel calves remained seronegative throughout the study. Serum mineral concentrations were evaluated on d 0 and 14. No differences (P ≥ 0.30) in serum Cu, Zn, Mn, or Se were observed between treatments on d 0. Control steers experienced a decrease (P < 0.001) in serum Zn and Se, and ITM steers had an increase (P = 0.007) in serum Cu on d 14 relative to initial d 0 values. On d 14, serum Zn and Se concentrations were greater (P < 0.01) in ITM compared with Control steers. Vaccinated calves experienced marked increases in neutralizing antibody titers by d 30 following vaccine administration. Calves receiving ITM at the time of vaccination experienced greater (P ≤ 0.003) neutralizing antibody titers to BHV-1 on d 14, 30, and 60 compared with Control. These results demonstrate that the injectable trace mineral formulation evaluated in this study, administered concurrently to viral vaccination, does not impair humoral immune responses in beef calves. Further, concurrent administration of ITM and BHV-1 vaccine may enhance the production of neutralizing antibodies to BHV-1 in previously na?ve beef calves.  相似文献

Experiments were conducted to compare clostridial antibody response of beef heifers that do and do not develop injection-site lesions, evaluate long-term antibody response of a single- and multiple-dose toxoid, and evaluate the ability of a clostridial toxoid to elicit an active antibody response in newborn calves. In Exp. 1, 37 weaned heifers were vaccinated (d 0) with a clostridial vaccine (Alpha-7, 2 mL, s.c.). Serum samples were collected on d 0, 28, 56, 84, and 112 to determine clostridial antibody titers. On d 28, heifers were visually inspected and palpated for injection-site lesions. The percentage of heifers that developed lesions was 64.9%. Lesioned heifers had elevated antibody titers for Clostridium chauvoei (CC) on d 28 (P < 0.08) and 84 (P < 0.07) compared with non-lesioned heifers. Clostridium sordellii (CS) and perfringens type D (CPD) antibody titers were greater in lesioned heifers than in non-lesioned heifers on d 28 and 56. In Exp. 2, long-term antibody response of Alpha-7 (A7) and Ultrabac 7 (UB7) was investigated in stocker heifers. The A7 heifers (n = 15) received one 2-mL vaccination (d 0), and the UB7 heifers (n = 15) received a 5-mL vaccination on d 0 and 28. Blood samples were collected on d 0, 28, 56, 84, 112, 140, and 180. Clostridium chauvoei, CPD, and Cl. novyi (CN) antibody titers from the A7 heifers were greater than those from the UB7 heifers on d 28. Due to the second UB7 injection, CC, CS, CN, and Cl. perfringens type C (CPC) antibody titers were greater in UB7 heifers than in A7 heifers on d 56. By d 112, titers were not different, and by d 140 all antibody titers were below detectable levels. In Exp. 3, 58 pregnant, mature, crossbred cows were vaccinated with A7 before calving. At birth, calves were carefully observed to ensure consumption of colostrum. Calves were blocked according to parturition date, and calves in each block were randomly allocated to receive A7 (s.c. at 3 +/- 3 d of age) or remain unvaccinated controls. Calves were bled at the time of vaccination (d 0) and on d 28, 56, 84, and 112. Antibody titers for CC, CPC, and CPD were elevated on d 0 and decreased throughout the experimental period (P < 0.01), but no titer differences (P > 0.10) were detected between treatment groups on any of the days sampled. These data indicated that antibody titers against clostridial diseases are enhanced when injection-site lesions develop. One injection of Alpha-7 seemed to provide the same length of protection as two injections of Ultrabac 7.  相似文献

Influence of supplemental Se on humoral immune response was measured in 60 weaned beef calves with marginal blood Se status. Calves were fed a Se-deficient diet consisting of corn silage, corn grain, and soybean meal. Blood Se concentrations, primary and secondary humoral immune responses to hen egg lysozyme inoculation, and weight gain were determined in a 70-day trial. Calves fed 20 mg of Se/kg of mineral mixture ad libitum had lower antibody responses (P less than 0.02), compared with calves fed 20 mg of Se/kg of mineral mixture and given 0.1 mg of Se and 0.22 IU of vitamin E/kg of body weight, IM, or with calves fed 80, 120, 160, or 200 mg of Se/kg of mineral mixture. Calves fed 80, 120, 160, or 200 mg of Se/kg of mineral mixture had higher (P less than 0.001) blood Se concentrations on day 70, compared with calves fed 20 mg of Se/kg of mineral mixture and given 0.1 mg of Se and 0.22 IU of vitamin E/kg of body weight, IM. Selenium supplementation had no effect on weight gain.  相似文献

Sixteen Bonsmara calves (4 males, 12 females) between 10 and 18 months of age were blocked according to age and sex and randomly assigned to 2 groups. They were offered licks containing bone meal and salt (50:50 ratio) (control) and bone meal and ammonium sulphate (NH4SO4) at 1.25, 2.5, 5, 10, 15, and 18% (treatment) to evaluate the effects of dietary anions on bone phosphate (P) concentration. Bone P concentration was significantly (P < 0.05) higher in the NH4SO4 group compared with the control group, indicating that NH4SO4 was able to increase the P content of bone at each of the 6 concentrations used in the lick relative to the control animals, thereby improving the P status of the animals. Ammonium sulphate at 15% and 18 % in the lick also significantly (P < 0.05) increased bone P compared with the lower concentrations of NH4SO4. Bone calcium (Ca) fluctuated as a result of the acidogenic lick. There was absorption of Ca when P was being resorbed and resorption of Ca when P was being absorbed into and out of bone. Bone Ca:P ratio ranged from 3.2 to 6.4 among the control group and 1.6 to 4.3 among the treatment group. Animals receiving the acidogenic lick had a higher percentage ash compared to the control group for most of the experimental period. Bone magnesium (Mg) fluctuated in response to the acidogenic lick, and it was difficult to show a relationship between bone Mg and Ca or P. The overall mean cortical bone thickness was significantly (P < 0.05) greater in treatment (1.60 mm) compared with control (1.43 mm) calves and this was also true at sampling periods 2, 4, 5 and 6. Bone thickness followed bone P and not bone Ca. Results from this research indicate that the addition of ammonium sulphate to a lick had a beneficial effect in improving the P status by increasing bone P and improving the mineral status of bone by increasing the thickness of cortical bone and percentage ash.  相似文献

A retrospective study was performed on 46 abomasotomies in young beef calves. Clinical abomasal disease in the herds under investigation had an average incidence of 1.0% for the year of the study. The range in incidence among herds was 0.2-5.7%. In 80% (37/46) of abomasal surgeries, abomasal ulcers were found. In 76% (35/46) of the cases, hair was present in the abomasum. These findings indicate that there could be a link between abomasal ulcers and the ingestion of hair by young beef calves.

Housing appeared to be a significant factor in relation to the relative risk of abomasal disease. Calves housed in pens or on stubble fields were nearly three times as likely to receive surgery for abomasal disease than were those housed on pasture.

The results also indicated that mineral supplements may be helpful in reducing the incidence of abomasal disease.

The influence of the suckling stimulus and ovarian secretions on LH response to naloxone was studied in 16 postpartum anestrous beef cows that were assigned randomly to one of four groups (n = 4/group): intact suckled (IS), intact nonsuckled (IN), ovariectomized suckled (OS) or ovariectomized nonsuckled (ON). Ovariectomy (OS + ON) and calf removal (IN + ON) were performed on d 2, 3 or 4 after parturition. Jugular venous blood was collected at 15-min intervals for 4 h before and 4 h after administration of naloxone (1 mg/kg BW, i.v.) on d 14 and d 28 after parturition. Gonadotropin-releasing hormone (5 micrograms, i.v.) was given 3 h after naloxone. Both IN and OS increased (P less than .05) mean pretreatment LH above IS values (mean +/- SE, ng/ml; IS 1.6 +/- .1 vs IN 2.5 +/- .3 and OS 2.7 +/- .4; P less than .01), whereas ON increased (P less than .01) LH (3.7 +/- .3 ng/ml) even further. Mean LH increased (P less than .05) after naloxone administration in all treatment groups. However, magnitude of this response was variable and dependent on ovarian status. Amplitude of the naloxone-induced LH response was greater (P less than .05) for ovariectomized (5.9 +/- 1.1 ng/ml) than for intact groups (2.7 +/- .5 ng/ml). Gonadotropin-releasing hormone increased mean LH concentrations in all groups. We suggest that ovarian secretions and the suckling stimulus contribute to endogenous opioid inhibition of LH during the postpartum interval.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献
