Two enzyme-linked immunosorbent assays (ELISA), one based on a mouse anti-Trypanosoma brucei group-specific monoclonal antibody and the other on rabbit anti-Trypanosoma evansi polyclonal antibodies, have been evaluated for their ability to detect circulating trypanosome antigens in camel sera as a means for the diagnosis of T. evansi infections. All 91 sera from a negative control camel herd from Kenya gave negative antigen-ELISA results in the monoclonal antibody-based ELISA and only 2 of them (2.2%) gave false positive results in the polyclonal antibody-based ELISA. In subsequent analyses of sera from infected camels (as determined by mouse inoculation), the monoclonal antibody-based ELISA detected antigens in 90 (83.3%) out of the 108 sera tested. This percentage was lower for the polyclonal antibody-based ELISA which was able to detect antigens in 67 (60.9%) out of the 110 sera tested. The two tests detected probably different antigens and when the results were combined, 99 out of 107 (92.5%) sera were shown to be ELISA positive. In a survey involving 316 camels from the Gao and Nara areas, in Mali, a high proportion of animals tested were antigen positive (43.5 and 42.9%, respectively for the mono- and polyclonal antibody-based ELISA) compared to only 22 (7.0%) diagnosed by the parasite detection techniques. Thus, these immunoassays were at least six times more sensitive than the haematocrit centrifugation technique. As a large proportion of cases may be antigen positive but parasite negative, these two of "surra" immunoassays should be used in routine diagnosis in addition to the parasite detection techniques in the dromedary camel.  相似文献

The monoclonal antibody ES78 was used in a sandwich immunosorbent assay (Sandwich ELISA) for the detection of antigens in sera and faeces in the course of Fasciola hepatica infection in 10 experimentally infected sheep. All infected sheep had circulating antigens in the first week post-infection (WPI). Antigenemia was detectable until WPI 3 in four infected sheep, WPI 4 in five infected sheep and in only one sheep by WPI 5. The detection of coproantigens (Fa(g)) was possible in five infected sheep at WPI-4, in four sheep at WPI-5 and in one sheep only at WPI-6. This technique was compared to an indirect ELISA for the detection of antibodies using excretory secretory antigens of F. hepatica. A significant correlation was found between Fa(g) and egg output and also with adult worm numbers. Our method demonstrated that the diagnosis of active fasciolosis in sheep is possible during all periods of infection.  相似文献

Circulating antibody against Fasciola hepatica antigens was determined by enzyme-linked immunosorbent assay (ELISA) and immunoelectrophoresis in alpacas naturally exposed to F. hepatica. Serological assay parameters were established by using sera from eight infected animals and seven controls with no record of this parasitic infection. Excretory--secretory (ES-) products, Fas1- and Fas2-ELISA were used to survey 307 alpacas from a F. hepatica endemic area in the Peruvian Andes. Seroprevalence of F. hepatica infection varied from 56.7, 64.8 and 66.8% measured by Fas1-, Fas2- and ES-ELISA, respectively. The sensitivity for ES-ELISA was 95%, corresponding Fas1- and Fas2-ELISA sensitivity values were 90 and 95%. In this population, 7% of animals were positive for F. hepatica eggs in faeces, other parasites detected were Trichuris sp. (40%), Nematodirus sp. (34.6%), Lamanema sp. (12.8%) and Eimeria sp. (11.8%). The results show that F. hepatica infected animals elicit circulating antibodies against ES, Fas1 and Fas2. Fas2-ELISA may be proposed as a sensitive assay for the immunodiagnosis of fasciolosis in alpacas.  相似文献

False-positive reactions were not observed with K99-negative strains and with several viral antigens. Only bovine coronavirus caused slight positive reactions which could be eliminated by a blocking test. As compared with the conventional procedure for the detection of the K99 antigen, ELISA seemed to be at least as sensitive and had the advantage that samples could be stored at -20° C before testing. In addition many samples could be handled at the same time and the results became available quickly. By carrying out the assay as a blocking test, specific antibody against K99 in serum or colostrum could be detected and titrated.  相似文献

The sensitivity of an indirect enzyme-linked immunosorbent assay (ELISA) for bovine IgG serum antibody to Pasteurella haemolytica was compared with that of an indirect hemagglutination (IHA) test. Pasteurella haemolytica serotypes were grown in a chemically defined cell culture medium, and soluble antigens released into the growth medium were used in the ELISA and IHA test. An ELISA with serotype-1 antigen consistently detected antibody in sera that were positive by IHA test (correlation, 99%). Sera reacting with serotype-1 ELISA antigens also reacted with ELISA antigens prepared from other serotypes. Although ELISA titers averaged 5 log2 units higher than IHA titers, plots of titers determined by the 2 methods were approximately linear. Titer increases detected in paired serum samples by either test were similar. The ELISA was more sensitive than was the IHA in detecting colostral IgG antibody in serum of newborn calves. The ELISA uses a simple, stable antigen preparation and detects antibody to P haemolytica serotypes that commonly infect cattle.  相似文献

An antibody-capture enzyme-linked immunosorbent assay (ELISA) for detection of antibody to turkey coronavirus (TCV) utilizing infectious bronchitis virus (IBV) antigen was developed. Anti-TCV hyperimmune turkey serum and normal turkey serum were used as positive or negative control serum for optimization of the ELISA system. Goat anti-turkey immunoglobulin G (light plus heavy chains) conjugated with horseradish peroxidase was used as detector antibody. The performance of the ELISA system was evaluated with 45 normal turkey sera and 325 turkey sera from the field and the cutoff point was determined. Serum samples of turkeys experimentally infected with TCV collected sequentially from 1 to 63 days postinfection were applied to the established antibody-capture ELISA using IBV antigens. The optimum conditions for differentiation between anti-TCV hyperimmune serum and normal turkey serum were serum dilution at 1:40 and conjugate dilution at 1:1600. Of the 325 sera from the field, 175 were positive for TCV by immunofluorescent antibody (IFA) assay. The sensitivity and specificity of the ELISA relative to IFA test were 93.1% and 96.7%, respectively, based on the results of serum samples from the field turkey flocks using the optimum cutoff point of 0.18 as determined by the logistic regression method. The ELISA values of all 45 normal turkey sera were completely separated from that of IFA-positive sera. The ELISA results of serum samples collected from turkeys experimentally infected with TCV were comparable to that of the IFA assay. Reactivity of anti-rotavirus, anti-reovirus, anti-adenovirus, or anti-enterovirus antibodies with the IBV antigens coated in the commercially available ELISA plates coated with IBV antigens could be utilized for detection of antibodies to TCV in antibody-capture ELISA.  相似文献

The sensitivity and specificity of the enzyme-linked immunosorbent assay (ELISA) for the diagnosis of Taenia solium cysticercosis was evaluated in experimentally and naturally infected pigs, using T. solium larval scoleces and its fractionated 1st and 2nd peaks on Sephadex G-200 as antigens. First peak antigen gave maximum sensitivity and highest antibody titres. The overall sensitivity of this test was found to be 91.5, 95.8 and 70.8% with scolex, 1st and 2nd peak antigens, respectively. False positive reactions occurred in 9.09% of uninfected pigs with scolex and 1st peak antigens and cross-reactions occurred in 25% of Taenia hydatigena-infected animals using scolex and 2nd peak antigens. No cross-reaction was observed using 1st peak antigen. The specificity of the test was 92.3, 96.2 and 92.3% with scolex, 1st and 2nd peak antigens, respectively.  相似文献

Recombinant 26 kDa capsid (CA) proteins of bovine lentiviruses, bovine immunodeficiency virus (BIV) and Jembrana disease virus (JDV), were expressed in Escherichia coli and utilised as antigens for an enzyme-linked immunosorbent assay (ELISA) and a western immunoblot (WIB) procedure for the detection of antibody in dairy cattle in Western Australia. A total of 690 serum samples, 30 from each of 23 farms, were tested by ELISA with a JDV CA protein antigen, and antibody was detected in 3.8% (p<0.05) of the sera. Nine sera from each farm were also tested by WIB with JDV CA protein antigens and antibody was detected in 15.9% of these samples. All ELISA-positive results were also WIB-positive, and all sera antibody-positive by WIB with JDV CA protein antigens were also antibody-positive by the WIB using recombinant BIV CA antigens. This study showed that recombinant protein antigens can be used for serological tests to detect bovine lentivirus infection in Australia.  相似文献

Calves infested with Psoroptes ovis (Hering, 1838) for the first time (naive) or previously infested calves were examined for serum antibody activity to P. ovis (obtained from rabbits) antigens by enzyme-linked immunosorbent assay (ELISA) to determine the temporal appearance of specific serum antibody activity. The development of the serum antibody activity to P. ovis antigens was then correlated with the development of lesions (% scab) and with changes in the number of P. ovis. The serum antibody activity [ELISA OD414 value (EODV) greater than 0.290] to P. ovis antigens in naive calves, in most cases, is first detected at the same time or slightly after the detection of mites and mite-caused lesions; and the development of specific serum antibody activity paralleled the increase in the P. ovis population and the percentage scab until 7 weeks post-infestation (PI). In previously infested calves, if serum antibody activity to P. ovis antigens was not already present from the previous infestation, specific serum antibody activity was detected at the same time and developed in a similar manner as in the naive calves. The serum antibody activity to P. ovis antigens could be detected after calves were relieved of their P. ovis burdens by pesticide treatment or after the P. ovis population began to decline when the calves were allowed to groom themselves. Serum antibody activity to P. ovis antigens was not detected in any of the control calves during the test period.  相似文献

A monoclonal antibody against a plasma membrane antigen of Trypanosoma rhodesiense was used for the detection of T brucei group-specific circulating antigen in 24 adult local dogs experimentally infected with T brucei brucei strain 8/18. Ten of the dogs were splenectomised and the remainder non-splenectomised (intact). Five dogs each from the splenectomised and intact groups were inoculated intravenously with try-panosomes. The infected dogs developed trypanosomiasis between days 4 and 8 after infection. The circulating antigens were detected as early as six days after infection and remained high until two weeks after treatment, when the circulating antigen declined. The detection of the antigens showed the existence of infection unlike the antibody test. The treatment of the infected dogs with diminazene aceturate (Berenil; Hoechst) at a dose of 7-0 mg/kg on day 21 after infection cleared all the parasites but elevated the circulating antigen levels. The antigen capture enzyme-linked immunosorbent assay is a useful diagnostic tool for complementing parasitological diagnosis, for detecting infection in the field and for ascertaining the efficacy of trypanocidal drugs.  相似文献

An enzyme-linked immunosorbent assay (ELISA) was developed for detection of specific immunoglobulin G (IgG) against canine distemper virus (CDV) antigens. Sucrose gradient separation of viral and cellular proteins was required to produce coating antigens for the ELISA. The specificity of the ELISA was demonstrated by blocking CDV-positive canine sera with CDV-specific antisera produced in goats and rabbits and adsorption of positive sera with CDV antigens. A comparison of the ELISA with the serum-neutralization technique for the detection of CDV antibodies was conducted. Anti-CDV IgG was detected in conventional dogs as early as 6 days after inoculation with a commercial vaccine to CDV. Paired sera from the immunized dogs were evaluated by both techniques and a statistically (P less than 0.01) significant agreement between the ELISA and the serum-neutralization technique was shown (r = 0.6121, n = 75).  相似文献

Several diagnostic methods including immunofluorescence, enzyme-linked immunosorbent assay, polymerase chain reaction and immunohistochemistry have been developed for the detection of porcine epidemic diarrhea virus (PEDV). An immunohistochemical method using a new recombinant antibody produced by a phage antibody system (PAS16) kit was investigated and compared with that using a monoclonal antibody for PEDV detection in PEDV-infected piglets. In both the immunohistochemical methods, PEDV antigens were detected in the cytoplasm of villous enterocytes and in the macrophages infiltrated in the lamina propria at 18 to 110 hr post inoculation. The positive signals with the recombinant PAS16 antibody were similar to those with the monoclonal antibody. This result suggests that the recombinant PAS16 antibody can be applicable for the rapid immunohistochemical diagnosis of PEDV infection.  相似文献

An improved enzyme-linked immunosorbent assay (ELISA) for detection of heterophile Hanganutziu-Deicher (HD) antibodies and antigens, which are frequently detected in sera and/or cancerous tissues from patients with various cancers was developed using biotinylated chicken anti-GM3(NeuGc) antibody and avidin-horseradish peroxidase conjugate. The N-glycolylneuraminyllactosyl-ceramide, GM3(NeuGc) ganglioside was purified from horse erythrocyte membranes. The ELISA procedure required 300 ng GM3(NeuGc) antigen to coat plastic microtiter plates and 190 ng biotinylated antibody per well to give optimum product formation. The technique could detect 6 ng antigen in tissue homogenate as compared to 0.6 ng of the pure compound by inhibition. Chicken anti-GM3(NeuGc) antibody quantitatively inhibited the biotinylated antibody, however, this procedure was not suitable to quantify lower affinity HD antibody in patient sera. Immunostaining specific for HD antigen-positive cells, in tissue sections was by 4 micrograms/ml biotinylated antibody and 200 dilution of Avidin-biotinylated peroxidase complex reagent using pig intestine and lymph node as positive tissues and chicken intestine and lung as negative tissues.  相似文献

An enzyme linked immunosorbent assay (ELISA) for the detection of Chlamydia psittaci in vaginal swabs of aborted ewes and goats has been developed using microtiter plates coated with sheep anti-Chlamydia immunoglobulin G. This technique was compared to the direct isolation of the agent by plaque assay on McCoy cells. Among 89 specimens from animals in infected flocks, 58 were positive by both methods, seven were only positive by ELISA, and nine others were only positive by direct isolation (plaque assay). None of the 75 specimens from animals in healthy flocks gave a positive response in ELISA or the plaque assay. Unlike direct isolation in cell culture, the ELISA technique permitted the detection of Chlamydia even in the absence of special care in sampling and conservation of specimens.  相似文献

Toxoplasma gondii is widely distributed in humans and other animals including domestic poultry throughout the world, but the data on prevalence of T. gondii in free-ranged (FR) chickens in People's Republic of China (PRC) are limited. In the present study, the seroprevalence of T. gondii infection in FR chickens was investigated in 13 provinces/municipalities of China during the period from January to June 2010. A total of 1173 serum samples were collected and assayed for T. gondii circulating antigens (TCA) and antibodies (TCAb) using enzyme linked immunosorbent assay (ELISA) technique. Out of this number, 199 samples were TCA positive (16.97%), 226 samples were TCAb positive (19.27%), 69 samples were positive for both TCA and TCAb (5.88%), and the total seropositive rate was found in 356 of 1173 (30.36%). The results of the present survey indicated that infection with T. gondii in FR chickens is widely spread in China.  相似文献

Blood samples were collected from 121 individuals of three species of wild-caught nonhuman primates from Kenya, including African green monkeys (Cercopithecus aethiops), Syke's monkeys (C. mitis), and olive baboons (Papio cynocephalus anubis), and were examined for circulating Trypanosoma brucei and for T. brucei antigen and anti-trypanosome antibody. Indirect antibody enzyme-linked immunosorbent assay detected titers of anti-T. brucei antibodies in 13 of the primates sampled, and field-oriented latex agglutination test detected invariant T. brucei antigens in 10 (8.3%) of the primates. However, no trypanosomes were visible in blood smears, on wet blood films, or by buffy coat technique, nor were they demonstrable in a subset of C. aethiops individuals that were studied using mouse subinoculation.  相似文献

Bulk and individual milk samples from 117 herds located in Brittany (west France) were used to assess: (i) the performance characteristics of an indirect enzyme‐linked immunosorbent assay (ELISA) applied to individual milk for the detection of antibodies to bovine viral diarrhoea virus (BVDV); and (ii) the relationship between the bulk milk result obtained from this test and the within‐herd prevalence of antibody‐positive lactating cows. This ELISA test was based on a monoclonal antibody directed against non‐structural protein NS2‐3 of pestiviruses. At the individual level, based on 1113 matched milk/serum samples, the sensitivity and specificity of this test applied to milk, compared with the virus neutralization test on serum, were 95.0 and 97.7%, respectively. At the herd level, the relationship between the optical density percentage (OD%) of bulk milk and the within‐herd prevalence of antibody‐positive lactating cows was assessed using the receiver operating characteristics (ROC) analysis. Classes of OD% of bulk milk were determined so that they were associated with minimum intraclass and maximum between‐class variances of within‐herd prevalence of antibody‐positive cows. The ROC analysis resulted in two classes of bulk milk results corresponding to different expected levels of within‐herd prevalence. Herds with an OD% of bulk milk <75% and ≥75% had a mean observed prevalence of antibody‐positive cows of 8.9 and 60.6%, respectively. Herds with a bulk milk result <75% were expected to be BVDV free, whereas large variations in prevalence of antibody‐positive cows existed in the herds with OD% ≥75%. The test described in this study is suitable to identify herds likely to have a low prevalence of BVDV antibody‐positive cows.  相似文献

为了探讨用抗独特型抗体替代病毒或病毒亚单位检测H9亚型禽流感血清抗体的可行性,本试验分别用全病毒抗原和抗独特型抗体(Ab2)作检测原,通过琼脂免疫扩散试验(AGP)和间接酶联免疫吸附试验(ELISA)检测了鸡的血清样品。在AGP试验中,分别用Ab2和禽流感病毒(AIV)对10份血清样品检测的符合率为70%。在间接ELISA试验中,Ab2和AIV抗原对10份血清样品检测的符合率为90%。用Ab2间接ELISA试验检测出的血清阳性率(8/10)高于血凝抑制试验(6/10)和AGP试验(5/10)。结果表明,在一定的情况下Ab2可以用来代替具有污染环境风险的病毒抗原来检测抗病毒抗体。  相似文献
