Maize was grown for 36 days in solution culture with roots either under axenic conditions or in the presence of rhizosphere organisms. In other experiments with sterile roots the plants were grown with different concentrations of potassium. At the end of the experiments sugars, organic acids and amino acids in the nutrient solutions were determined. Under axenic conditions the exudates consisted of up to 65% sugars, up to 33% organic acids and only up to 2% amino acids. The same substances were detected in non-sterile nutrient solutions. In the presence of microorganisms fructose, arabinose and the predominating glucose decreased to almost one half, while sucrose was not affected. The amounts of organic acids were not changed by microbial growth. The main amino acid, glutamic acid, was nearly doubled by the presence of microorganisms, whereas other amino acids remained unchanged. The lower O2 content of the nutrient solutions of non-sterile roots suggested microbial decomposition of monosaccharides. In another experiment with roots grown under axenic conditions and with different K treatments low K supply significantly increased the total amounts of sugars, organic acids and amino acids exuded g?1 root dry matter. As in the previous experiment glucose, fumaric and oxalacetic acid as well as glutamic and aspartic acid dominated in the respective fractions. Again sugars and organic acids represented the major quantity of exudates, while amino acids amounted to less than 2%. In an additional experiment with another cultivar, with nitrate as N source and a 5-day longer growth period, somewhat different results were obtained. In the exudates sugars were found in lower amounts, probably due to a higher growth rate. Under these conditions organic acids were the prevailing root exudates. Unlike sugars and amino acids, their total quantity was not affected by K nutrition, but the proportion of malic acid increased with increasing K supply, while oxalacetic acid dominated at low K nutrition. Similarly the total amount of organic acids within the root was independent of K nutrition. However malic acid content increased with increasing K application, while the likewise dominating citric and oxalacetic acid decreased.  相似文献

Root exudates play a major role in the mobilization of sparingly soluble nutrients in the rhizosphere. Since the amount and composition of major metabolites in root exudates from one plant species have not yet been systematically compared under different nutrient deficiencies, relations between exudation patterns and the type of nutrient being deficient remain poorly understood. Comparing root exudates from axenically grown maize plants exposed to N, K, P, or Fe deficiency showed a higher release of glutamate, glucose, ribitol, and citrate from Fe‐deficient plants, while P deficiency stimulated the release of γ‐aminobutyric acid and carbohydrates. Potassium‐starved plants released less sugars, in particular glycerol, ribitol, fructose, and maltose, while under N deficiency lower amounts of amino acids were found in root exudates. Principal‐component analysis revealed a clear separation in the variation of the root‐exudate composition between Fe or P deficiency versus N or K deficiency in the first principal component, which explained 46% of the variation in the data. In addition, a negative correlation was found between the amounts of sugars, organic and amino acids released under deficiency of a certain nutrient and the diffusion coefficient of the respective nutrient in soils. We thus hypothesize that the release of dominant root exudates such as sugars, amino acids, and organic acids by roots may reflect an ancient strategy to cope with limiting nutrient supply.  相似文献

Plant root systems mediate ecological processes in the rhizosphere through the exudation of organic compounds. Although exudate composition is thought to depend strongly on plant nutrient status, little is known about the influence of multi‐nutrient stresses. In this study, we examined responses to short‐term (3 d) nutrient limitation in Helianthus annuus (common sunflower), and root exudates were collected for 2, 4, or 6 h with the trap‐solution method. Root exudates, analyzed by means of gas chromatography‐mass spectrometry, consisted of over 60 sugars, sugar alcohols, amino acids, organic acids, and phosphates, with sugars and organic acids generally detected in the highest quantities. Twenty‐five of the detected metabolites, including half of the organic acids, sugars, and sugar alcohols, differed in relative abundance among the three sampling intervals, exhibiting higher abundance in sampling intervals greater than 2 h. Similarly, 24 of the detected metabolites, including half of the amino acids, phosphates, and sugar alcohols, were affected by nutrient supply, with 20 exhibiting higher abundance in the high‐nutrient treatment. Fumaric acid, quinic acid, and glucose were detected at significantly higher levels in the low‐nutrient treatment, potentially representing an adaptive response to nutrient limitation in sunflower. However, as sampling interval exerted a strong influence on the apparent effects of nutrient supply, future studies should consider the potential impacts of sampling‐interval length in comparative analyses of genotypes or treatments.  相似文献

During the cooking process of Agave tequilana Weber var. azul to produce tequila, besides the hydrolysis of inulin to generate fermentable sugars, many volatiles, mainly Maillard compounds, are produced, most of which may have a significant impact on the overall flavor of tequila. Exudates (agave juice) from a tequila company were collected periodically, and color, Brix, fructose concentration, and reducing sugars were determined as inulin breakdown took place. Maillard compounds were obtained by extraction with CH(2)Cl(2), and the extracts were analyzed by GC-MS. Increments in color, Brix, and reducing sugars were observed as a function of time, but a decrease in fructose concentration was found. Many Maillard compounds were identified in the exudates, including furans, pyrans, aldehydes, and nitrogen and sulfur compounds. The most abundant Maillard compounds were methyl-2-furoate, 2,3-dihydroxy-3,5-dihydro-6-methyl-4(H)-pyran-4-one, and 5-(hydroxymethyl)furfural. In addition, a series of short- and long-chain fatty acids was also found. A large number of the volatiles in A. tequilana Weber var. azul were also detected in tequila extracts, and most of these have been reported as a powerful odorants, responsible for the unique tequila flavor.  相似文献

蚕豆枯萎病是土传病害,其发生与蚕豆根系分泌物有密切关系。本文以3个枯萎病不同抗性蚕豆品种——‘89-147’(高抗)、‘8363’(中抗)和‘云豆324’(感病)为材料,通过水培试验收集根系分泌物,测定根系分泌物对镰刀菌孢子萌发和菌丝生长的影响,分析对枯萎病表现出不同抗性的蚕豆品种根系分泌物中糖、氨基酸和有机酸的含量,分离鉴定了根系分泌物中氨基酸和有机酸的组分。结果表明,抗病品种的根系分泌物抑制了尖孢镰刀菌的孢子萌发和菌丝生长,在加入5 mL中抗品种根系分泌物时,显著促进尖孢镰刀菌孢子萌发,但对菌丝生长无显著影响;而在加入1 mL感病品种根系分泌物时,就能显著促进尖孢镰刀菌孢子萌发和菌丝生长。不同抗性蚕豆品种根系分泌物中氨基酸总量和总糖含量随抗性的降低而升高,有机酸分泌总量则随蚕豆品种对枯萎病的抗性增加而升高。感病品种和中抗品种中检出15种氨基酸,而高抗品种中检出14种,组氨酸只存在于中抗品种中,脯氨酸仅在感病品种中检出,3个蚕豆品种根系分泌物中均未检出精氨酸。蚕豆根系分泌物中天门冬氨酸、谷氨酸、苯丙氨酸、酪氨酸和亮氨酸含量高,可能会促进枯萎病的发生,而蛋氨酸、赖氨酸和丝氨酸含量高可能抑制枯萎病发生。酒石酸仅在抗病品种中存在,根系分泌物中有机酸种类丰富,有助于提高蚕豆对枯萎病的抗性。蚕豆对枯萎病的抗性不同,根分泌物对镰刀菌孢子萌发和菌丝生长的影响也不同,而这种抗病性差异与蚕豆根系分泌物中糖、氨基酸、有机酸的含量和组分密切相关。  相似文献

Most research in the past using genetically modified crops (GM crops) has focused on the ecological safety of foreign gene (i.e., the gene flow), gene products (for example, Bt (Bacillus thuringiensis) protein), and the safety of transgenic food for humans. In this study, changes in both the species and amounts of low-molecular-weight components in cotton (Gossypium hirsutum L.) root exudates after foreign Bt gene overexpression were investigated under different nutritional conditions. Transgenic cotton containing Bt (Bt-cotton), supplemented with all the mineral nutrients, secreted more organic acids than the wild-type cotton (WT). When nitrogen was removed from the full-nutrient solution, the amount of organic acids secretion of Bt-cotton was lesser than that of WT. The roots of the transgenic cotton secreted lesser amounts of amino acids and soluble sugars than the WT roots in the full-nutrient solution. Deficiencies of P and K caused a large increase in the total amino acid and soluble sugar secretions of both Bt-cotton and WT, with larger increases observed in Bt-cotton. Because transferring the foreign Bt gene into cotton can result in alterations in the components of the root exudates, with the effect varying depending on the nutritional status, the cultivation of genetically modified crops, such as Bt-cotton, in soil environments should be more carefully assessed, and the possible effects as a result of the alterations in the root exudate components should be considered.  相似文献

采用砂培和室内模拟方法,研究了两个抗病性不同的大豆品种水溶性根系分泌物中氨基酸组分随作物生长的变化;同时检测了培养基中添加大豆根系分泌物和纯品氨基酸对大豆根腐病菌菌落生长的影响。结果表明,添加大豆苗期和花荚期根分泌物均显著促进尖镰孢菌菌丝生长,添加成熟期根分泌物显著促进腐皮镰孢菌菌丝生长。易感根腐病大豆品种合丰25号花荚期以后根分泌物中氨基酸种类多于抗根腐病大豆品种绥农10号。感病大豆品种根系分泌的氨基酸总量随生育时期增加,在鼓粒期达到最高;抗病大豆品种根系分泌的氨基酸总量在花荚期最高。感病大豆品种根系分泌的主要氨基酸为精氨酸,抗病大豆品种根系分泌的氨基酸主要为天冬氨酸。氨基酸纯品培养中,添加精氨酸和酪氨酸处理的尖镰孢菌菌落直径显著高于不加氨基酸的对照菌落直径;添加丝氨酸和天冬氨酸的处理菌落直径则显著低于对照处理。同时,添加天冬氨酸的培养基上腐皮镰孢菌菌落直径显著低于不加氨基酸的对照。可见,不同大豆品种根系分泌物中氨基酸组分对病原菌生长起着一定的作用,其表现的作用受根际氨基酸种类和氨基酸浓度影响较大,对于不同病原菌的作用存在差异。  相似文献

应用水培的方法采集油菜根系分泌物;采用高效液相色谱测定出油菜根系分泌物中所含的16种氨基酸;使用烧杯法和平板趋化法进行趋化性试验,分别测定枯草芽孢杆菌Tu-100对16种氨基酸、油菜根系和油菜菌核的趋化性。结果表明Tu-100仅对组氨酸、脯氨酸、精氨酸、天门冬氨酸和谷氨酸等5种氨基酸有趋化性,其中组氨酸的趋化性最强;Tu-100对油菜根系和油菜菌核也有趋化性,对油菜根系的趋化性要大于油菜菌核。  相似文献

Five chick pea (Cicer arietinum L.) varieties when inoculated with an effective Rhizobium strain showed significant variations in nodule number. The root exudates of the plant varieties were analysed for sugars, TCA cycle intermediates and amino acids. Xylose, citrate, alanine and phenylalanine were detected in exudates of one or two varieties, in addition to compounds common to all varieties such as glucose, glucosamine, ribose, serine, homoserine and glutamic acid. However, the concentration of all these compounds in the exudates collectively was inadequate to support Rhizobium growth under aseptic conditions. Several of the exuded compounds were chemotactic at very low concentrations. The rhizobia showed taxes toward growing roots of all the varieties in agar plates. Competition experiments showed that the rhizobia have a multiple-chemotactic system like that of Escherichia coli. It appears that root exudates accumulate Rhizobium along the growing roots through chemotaxis. The minor differences in exudate composition among varieties is not a factor responsible for variation in nodule number.  相似文献

This study assesses the influence of saccharides in the rhizodeposition on the phosphate solubilizing ability of rhizosphere bacteria. Water‐soluble rhizodeposits were analyzed of 14C‐labeled pea plants (Pisum sativum, cv. ‘Grapis’) which were grown at two different levels of P‐nutrition. The sugars produced were fed in vitro either as single compounds or as synthetic mixtures to three bacterial strains and the ability of the bacteria to mobilize Ca3(PO4)2 was measured. The relative glucose proportion of pea exudates decreased under P deficiency while the content of galactose, ribose, xylose and fucose increased. In vitro feeding of single sugars and sugar mixtures showed that the ability of Pseudomonas fluorescens (PsIA12) to dissolve tertiary calcium phosphate was lower with pentoses and the mixed sugars of the P‐deficient plants than with glucose. On the other hand, the shifted sugar pattern observed under P deficiency increased the phosphate mobilization ability of Pantoea agglomerans (D5/23) and Azospirillum sp. (CC 322) considerably. This observation can only partly be explained by the acidification of the nutrient medium. Bacteria also produced different carboxylic anions depending on sugar supply. In addition to low‐molecular mono‐, di‐, and tricarboxylic acids which are known as P‐solubilizing substances, sugar acids also played an important role in cultures D 5/23 and CC 322.  相似文献

Root exudates comprising soluble low‐molecular‐weight organic compounds (LMWOCs) play a crucial role in the rhizosphere processes. Therefore, accurate determination of the composition and quantity of these compounds is of importance. A continuous trapping system (CTS) with XAD‐4 macroporous resin is widely used for collecting root exudates in hydroponics, and ion exchange resins are used in the separation of root exudates into neutral, acid and basic fractions. Here, we studied the adsorption equilibrium isotherms and dynamic adsorption properties of 27 standard LMWOCs on XAD‐4 resin. The adsorption isotherms of most LMWOCs matched the Langmuir isotherm equations. Furthermore, we investigated the adsorption efficiency of the CTS for LMWOCs and the recovery ratios during fractionation by ion exchange resins. The adsorption capacities of the CTS for LMWOCs which are non‐polar or having large non‐polar moieties were higher. The recovery ratios of most LMWOCs were larger than 80% during fractionation. The overall recovery ratios of LMWOCs were ca. 10, 20, and 30% for sugars, organic acids, and amino acids, respectively. Using this collection method, we present the composition and quantity of root exudates of cucumber (Cumumis sativus L.) at four different growth stages. The major components of root exudates were similar to previous works, whereas the quantities were different. The various collection methods may be the main reason for these differences. Therefore, it is necessary to study the overall recovery ratios when the compositions of root exudates via different collection methods are compared.  相似文献

Hokkaikintoki, Himetebou, and Kurodane-kinugasa) of common bean ( Phaseolus vulgaris L.) were fractionated based on several physicochemical properties, and the nutrient substances in the fractions effective for Rhizobium proliferation were determined. Regardless of the common bean cultivars, the low molecular weight and high polarity fraction showed the most beneficial effect on Rhizobium proliferation. After further separation of the low molecular weight and high polarity fraction, the promoting effect was detected in the non-ionic and the cationic plus ampho-teric subtractions. Although the amount of sugars or amino acids exuded from a seed was, respectively, similar in all the cultivars except for the sugars in Kurodanekinugasa, the content of these compounds per seed-weight was inversely proportional to the seed size. Ten sugars and twenty-one amino acids were, respectively, identified in the non-ionic and cat-ionic plus amphoteric subtractions, and there were some differences in the composition of these substances between common bean cultivars.  相似文献

Microbial population both in diversity as well as numbers in soil is influenced by the amount and type of various compounds entering soil through plant litter, root exudates and management factors like mineral and organic fertilizers under various climatic conditions. This in turn affects crop production and sustainability of soil health. Therefore, we tested total and number of various groups of microflora and also substrate use by Biolog to assess the metabolic capabilities of various groups of microbes from soils of long term fertilization experiment, Halle (Germany) having different mineral and organic fertilizer treatments in unplanted and planted soils with two crops viz., Secale cereale and Medicago sativa . High number of total microorganisms, nitrogen fixers and phosphate solubilizers were observed in the rhizosphere of Medicago sativa and Secale cereale compared to bulk soils. Microbial counts were specially higher in treatments of NPK+FYM (farmyard manure) and NPK in Secale. Medicago had more P solubilizers in yearly addition of phosphatic fertilizers compared to three yearly treatment. During substrate utilization test (Biolog) maximum utilization was found for sugars followed by amino acids in Secale whereas in Medicago sugars were followed by carboxylic acids.  相似文献

A glasshouse experiment was conducted using a root-bag technique to study the root exudates, rhizosphere Zn fractions, and Zn concentrations and accumulations of two ryegrass cultivars (Lolium perenne L. cvs. Airs and Tede) at different soil Zn levels (0, 2, 4, 8, and 16 mmol kg-1 soil). Results indicated that plant growth of the two cultivars was not adversely affected at soil Zn level≤8 mmol kg-1. Plants accumulated more Zn as soil Zn levels increased, and Zn concentrations of shoots were about 540 /μg g-1 in Aris and 583.9μg g-1 in Tede in response to 16 mmol Zn kg-1 soil. Zn ratios of shoots to roots across the soil Zn levels were higher in Tede than in Airs, corresponding with higher rhizosphere available Zn fractions (exchangeable, bound to manganese oxides, and bound to organic matter) in Airs than in Tede. Low-molecular-weight (LMW) organic acids (oxalic, tartaric, malic, and succinic acids) and amino acids (proline, threonine, glutamic acid, and aspartic acid, etc.) were detected in root exudates, and the concentrations of LMW organic acids and amino acids increased with addition of 4 mmol Zn kg-1 soil compared with zero Zn addition. Higher rhizosphere concentrations of oxalic acid, glutamic acid, alanine, phenylalanine, leucine, and proline in Tede than in Airs likely resulted in increased Zn uptake from the soil by Tede than by Airs. The results suggested that genotypic differences in Zn accumulations were mainly because of different root exudates and rhizosphere Zn fractions.  相似文献

A solution culture experiment was conducted to investigate the effects of collection time and interfering ions on separation and determination of low-molecular-weight organic acids in root exudates of soybean using the method for directly collecting root exudates.The suitable cooection time of root exudates and the interferiung ions affecting organic acid determination were determined.The method for removing the interfering ions was established and analyzed.The release amount of root exudates increased with the increase of collection time from 0 to 120min but decreased with increasing of collection time from 120to 240min.The maximum exuding amounts of organic acids were observed in root exudates at the collection time of 120min.There was a significant difference of organic acid components between the treatments of collection time of 120min and 240min,Citric acid was founnd only in the treatment of 120min collection time,NO3^- was the main interfering ion in organic acid determination and had the same retention time as oxalic acid.Anion exchange resin(SAX)properly treated by HPLC(high performance liquid chromatography)solvent could remove NO3^- anion in sample solution of root exudates,thus enhancing the recoveries of organic acids in root exudates.There was no significant effect of the chemicals added into sample solution such as H3PO4,SAX and KNO3 on the retention time of organic acids.  相似文献

Effects of seed and root exudates obtained from common bean on the proliferation of Rhizobium sp. (Phaseolus) were examined in a combination of three plant cultivars with three Rhizobium strains. In the first experiment, seed or root exudate was mixed with an Andosol soil extract, and bacterial proliferation in the mixture was traced. Seed exudate was prepared from hydroponic solution used in seed imbibition for 24 h, and a series of root exudates was prepared from a hydroponic solution collected every 24 h from the initiation of rooting up to 96 h after rooting. Regardless of the common bean cultivars and Rhizobium strains used, Rhizobium population markedly increased of the 24 h of culture in the mixture containing seed exudates, whereas a negligible increase was detected in the mixture with root exudates. The mixture containing root exudates collected within a period of 72–96 h after initial rooting (96–120 h after seed imbibition) exerted an inhibitory effect on Rhizobium proliferation. The seed exudates contained large amounts of sugars, amino acids, nitrogen, phosphorus, potassium, and magnesium compared to any root exudates. In the second experiment, Rhizobium was inoculated directly to common bean seeds sowed in a vermiculite bed which was sterilized and moistened with a plant nutrient solution. Compared with the control (without seed), a remarkable increase in the number of bacterial cells was observed in all the combinations of plant cultivars and Rhizobium strains 24 h after sowing. These results reveal that seed exudates of common bean have a substantial potential to promote Rhizobium proliferation, and that root exudates in a particular period of culture contain some inhibitory factors.  相似文献

Two barley cultivars (‘Sahara’ = Zn-efficient and ‘Clipper’ = Zn-inefficient) were grown at different soil Zn fertilization (0, 0.2, 0.8, 1.6 and 3.2 mg Zn kg?1 soil). Root exudates were collected 16 and 28 days after sowing. At Zn = 0, shoot dry matter was decreased in both genotypes, but more distinctly in ‘Clipper’. At 0.2 mg Zn kg?1, the ‘Sahara’ shoot concentrations of Zn was 130% higher and shoot Zn content 44% greater compared with ‘Clipper’. Low-molecular-weight organic acid anions (=carboxylates) (malate, maleate, fumarate and cis-aconitate) and amino acids (alanine, valine, proline, aspartic acid and glutamic acid) were detected in root exudates, with the highest concentration at Zn = 0.2 mg kg?1 soil. Higher concentrations of organic acid anions as well as amino acids were noted in the rhizosphere of ‘Sahara’ than ‘Clipper’. The genotypic differences in Zn acquisition from soil may be linked to differential carboxylate and amino acid composition of root exudates.  相似文献

Studies performed on model systems using pyrolysis-GC-MS analysis and (13)C-labeled sugars and amino acids in addition to ascorbic acid have indicated that certain amino acids such as serine and cysteine can degrade and produce acetaldehyde and glycolaldehyde that can undergo aldol condensation to produce furan after cyclization and dehydration steps. Other amino acids such as aspartic acid, threonine, and alpha-alanine can degrade and produce only acetaldehyde and thus need sugars as a source of glycolaldehyde to generate furan. On the other hand, monosaccharides are also known to undergo degradation to produce both acetaldehyde and glycolaldehyde; however, (13)C-labeling studies have revealed that hexoses in general will mainly degrade into the following aldotetrose derivatives to produce the parent furan-aldotetrose itself, incorporating the C3-C4-C5-C6 carbon chain of glucose (70%); 2-deoxy-3-ketoaldotetrose; incorporating the C1-C2-C3-C4 carbon chain of glucose (15%); and 2-deoxyaldotetrose, incorporating the C2-C3-C4-C5 carbon chain of glucose (15%). Furthermore, it was also proposed that under nonoxidative conditions of pyrolysis, ascorbic acid can generate the 2-deoxyaldotetrose moiety, a direct precursor of the parent furan. In addition, 4-hydroxy-2-butenal-a known decomposition product of lipid peroxidation-was proposed as a precursor of furan originating from polyunsaturated fatty acids. Among the model systems studied, ascorbic acid had the highest potential to produce furan, followed by glycolaldehyde/alanine > erythrose > ribose/serine > sucrose/serine > fructose/serine > glucose/cysteine.  相似文献

The quality of Tombul (Round) hazelnut, grown in the Giresun province of Turkey, was determined by measuring proximate composition, minerals, vitamins, dietary fiber, amino acids, and taste active components (free amino acids, sugars, and organic acids). Fat was the predominant component in Tombul hazelnut (approximately 61%). The major minerals were potassium, phosphorus, calcium, magnesium, and selenium. Hazelnut was also found to serve as an excellent source of vitamin E (24 mg/100 g) and a good source of water soluble (B complex) vitamins and dietary fiber. The major amino acids were glutamic acid, arginine, and aspartic acid. The three nonessential amino acids and the essential amino acids contributed 44.9 and 30.9% to the total amino acids present, respectively, while lysine and tryptophan were the limiting amino acids in Tombul hazelnut. Twenty-one free amino acids, six sugars, and six organic acids were positively identified; among these, arginine, sucrose, and malic acid predominated, respectively. These taste active components may play a significant role in the taste and flavor characteristics of hazelnut.
