OBJECTIVE: To determine whether stress associated with transportation or feed withdrawal increased fecal shedding of Salmonella Typhimurium among pigs experimentally infected with the organism. ANIMALS: 86 healthy pigs. PROCEDURE: Pigs were challenge exposed with Salmonella Typhimurium at 4 weeks old and reared conventionally. When pigs reached market weight, they were assigned to groups and subjected to various combinations of transportation and feed withdrawal. Ileocecal contents were collected after slaughter and tested for Salmonella Typhimurium. RESULTS: Salmonella Typhimurium was not detected in feces collected from pigs just prior to slaughter. When feed was withheld for 24 hours prior to slaughter, the proportion of transported pigs with Salmonella Typhimurium in ileocecal contents at the time of slaughter was not significantly different from the proportion of nontransported pigs. However, when feed was not withheld prior to slaughter, the proportion of transported pigs with Salmonella Typhimurium in ileocecal contents at the time of slaughter was significantly higher than the proportion of nontransported pigs. CONCLUSIONS AND CLINICAL RELEVANCE: When carrier pigs remained on feed, transportation stress increased the proportion positive for Salmonella sp. On the basis of results reported here, it is suggested that producers withhold feed from pigs for 24 hours prior to transportation to a slaughter plant.  相似文献

The aim of this study was to evaluate the impact of the pre-slaughter process on Salmonella caecal contamination of pigs at slaughter. An observational study was carried out in 2001 on 101 conventional farrow-to-finish pig farms. On each farm, one batch of contemporary pigs was followed from the end of the fattening period until slaughter. The Salmonella bacteriological status of the batches was assessed by environmental samples of faecal material. The serological Salmonella status was obtained on 30 individually identified market-age pigs using an indirect ELISA test. At the slaughterhouse, 25 g of caecal contents were taken from 10 of the identified pigs. Faecal and caecal material were analysed according to a classical bacteriological method. A questionnaire was designed to obtain information about the type of feeding during the fattening period (dry versus wet), the duration of fasting on the farm before leaving for the slaughterhouse, the duration of transport between the farm and the slaughterhouse, the holding time in lairage at the slaughterhouse and loading and unloading conditions on the farm and at the slaughterhouse. To assess the relationships between these factors and the Salmonella caecal status of the pigs and the batches, two logistic models were fitted at the individual and at the batch level, respectively. The first analysis was performed using a random effects logistic regression model. The second analysis was based on a cumulative logit model with a positive caecal rate classified into three classes as the outcome variable. The results showed that the Salmonella status of market-age pigs assessed on the farm either by serological or bacteriological examinations and the time spent in lairage before slaughtering played a crucial role on caecal contamination. In the light of these results, actions should be considered both on the farm and at the slaughterhouse to decrease the risk of Salmonella contamination of the caecal contents.  相似文献

OBJECTIVE: To determine whether withdrawing feed from pigs prior to slaughter had any effects on prevalence or severity of gastric ulcers. DESIGN: Split-plot design. ANIMALS: 873 pigs. PROCEDURES: At the finishing barn, pigs were assigned to 30 pens. Feed withdrawal times (0, 12, or 24 hours) were assigned to pens at random, and pigs in each pen were marketed in 3 groups over a period of 4 weeks. The first marketing group consisted of the 10 heaviest pigs in each pen, the second consisted of the next 10 heaviest, and the third consisted of all remaining pigs. Feed was withheld from all pigs in each pen prior to removal of each marketing group. Thus, feed was withheld once, twice, or 3 times for pigs in the first, second, and third marketing groups, respectively. RESULTS: Feed withdrawal time was not significantly associated with ulcer score at the time of slaughter. Ulcer scores and prevalence of chronic damage were higher in the third marketing group, regardless of feed withdrawal time. Prevalence of severe damage, prevalence of chronic damage, and prevalence of esophageal constriction increased as carcass weight decreased. No pigs died of gastric ulceration. CONCLUSION AND CLINICAL RELEVANCE: Results suggest that withdrawal of feed from pigs prior to slaughter does not increase damage to the stomach and that repeated feed withdrawal does not result in fatal gastric ulceration.  相似文献

In a field study with fattening pigs, effects of feed particle size at the dietary presence of organic acids on Salmonella prevalence were measured. On two farms (f1/f2), each holding ~800 pigs, diets based on finely ground (control) or coarsely ground ingredients (experiment) were fed as crumbs. On f1 both control and experimental grower and finisher diets contained identical concentrations of formic and propionic acid (0.4% and 0.2% respectively). On f2 only finisher diet of the experimental group contained 1.2% potassium diformate. At the start of the fattening period no statistical differences were measured between Salmonella prevalence in animals fed control and experimental diets on both farms. At slaughter Salmonella prevalence in caecal contents was lower (p < 0.05) on f1 in animals fed the experimental diet. Furthermore, the number of seronegative meat juice samples taken from these animals [optical density (OD) <10] was higher (p < 0.001); seropositive as well as distinct seropositive samples (OD ≥20 and ≥40 respectively) were less frequent (p < 0.01) compared to samples from animals fed the control diet. Feeding the experimental diet on f2 resulted in a lower Salmonella detection rate in faeces before slaughter (p < 0.01). Salmonella prevalence was lower in caecal content at slaughter for pigs fed the experimental diet compared to those fed control diet (p < 0.0001). The number of distinct seropositive meat juice samples (OD ≥40) was lower (p < 0.01) for pigs fed the experimental than for those fed the control diet. In comparison to pigs in the control group, starch concentrations in the caecal content from pigs in the experimental groups on both farms were higher (p < 0.05) and the pH values lower (p < 0.05). Propionate (p < 0.0001) and butyrate (p < 0.01) concentrations were higher in the caecal content taken from pigs in the experimental group on f2.  相似文献

The objective of this study was to evaluate the influence of different feed withdrawal and lairage times prior to slaughter on the gastrointestinal tract (GIT) weight and on the fermentation pattern and numbers of Enterobacteriaceae in the cecum of pigs. A total of 72 finishing pigs (6 pens, 12 animals each) were included in the study and were sent to the slaughterhouse on three consecutive days. On each day pigs from two pens were deprived of feed for either 2 or 12 h before leaving the farm. Pigs from each of the two pens were divided in sub-groups (4 pigs each) on the moment of loading onto the truck and followed the same distribution at the slaughterhouse. Once there, each sub-group corresponding to the same feed withdrawal time was held in different holding pens for 0, 5 or 10 h before slaughter. The weight of the gastro-intestinal tract (GIT) was determined and cecal content was collected to evaluate pH, short chain fatty acids (SCFA) and ammonia (NH3) concentrations as well as lactobacilli and Enterobacteriaceae numbers. Total GIT weight decreased as feed withdrawal (P < 0.0001) and lairage time (P < 0.001) increased within a range between 6.69 and 4.59 kg. Cecal pH increased with feed withdrawal (P < 0.0001) and lairage time (P = 0.0001) (ranging from 5.79 to 6.79) in parallel to a decrease in the total SCFA concentration (P < 0.005) (from 211 to 100 mM). The percentages of acetic and propionic acid were not modified by the experimental treatments but significant decreases were registered in the percentage of butyric acid with the increased times of both withdrawal (P < 0.0001) and lairage (P < 0.005). The percentage of valeric and branched SCFA and also NH3 concentration showed an increase with withdrawal (P < 0.005, P < 0.0001 and P < 0.005 respectively) and lairage (P < 0.005, P < 0.0001 and P < 0.001 respectively). Lactobacilli numbers decreased as lairage time increased (from 9.97 to 8.63 log 16S rRNA gene copies/g FM; P < 0.05) and Enterobacteriaceae numbers increased with both, feed withdrawal (P < 0.05) and lairage (P < 0.05) (ranging from 8.53 to 9.95 log 16S rRNA gene copies/g FM).The previous to slaughter increase in feed withdrawal and lairage times decreased the GIT weight but involved changes in the gut microbial ecosystem caused through changes in the fermentation pattern that lead to the increase of Enterobacteriaceae numbers. This increase could represent a higher risk of carcass contamination by the enteropathogens of this group such as Salmonella. However this potential risk should be further investigated.  相似文献

Background:Gut microbiota plays a key role in health,immunity,digestion,and production in layers.Factors such as environment,diet,diseases,stress,and flock management significantly affect gut microbiota;however,it is not known how potential stressors such as intramuscular injections or feed withdrawal alter the composition of gut microbiota that result in increased the shedding level of foodborne pathogens.In the current study,the effects of intramuscular corticosterone injection and feed withdrawal were evaluated to understand their role in Salmonella Typhimurium shedding and changes in the composition of gut microbiota in layers.Results:Salmonella shedding was observed for 8 weeks post-infection.There was a significant increase in Salmonella Typhimurium count after intramuscular injection and feed withdrawal.The Salmonella infected and the negative control groups showed significant differences in the abundance of different genera in gut microbiota at week 1 and up to week 7 post infection.The infected group showed a significant reduction in alpha diversity of gut microbiota.Firmicutes reduced significantly(P<0.05)after intramuscular injection,while the feed withdrawal groups did not cause any significant changes in Proteobacteria-Firmicutes ratio.Furthermore,intramuscular injection resulted in a significant change in alpha diversity of gut microbiota.Conclusions:Exposure of chicks to relatively low dose of Salmonella Typhimurium can lead to persistent shedding in pullets.The Salmonella Typhimurium infection disrupted the gut microbiota composition immediately after infection.The potential stress of intramuscular injection and feed withdrawal significantly increased the Salmonella Typhimurium count in faeces.The intramuscular injection also resulted in a significant alteration of the Proteobacteria-Firmicutes ratio,which could increase the risk of dysbiosis.  相似文献

This study was carried out to evaluate the effect of feed withdrawal and handling intensity on blood acid-base responses and muscle glycolytic potential in slaughter-weight pigs. Sixty crossbred pigs (BW = 107.7 +/- 0.56 kg; 44 barrows and 16 gilts) were used in a randomized complete block design with a 2 x 2 factorial arrangement of treatments: 1) feed withdrawal (0 vs. 24 h), and 2) handling intensity (low vs. high). The high-intensity handling treatment consisted of moving the pigs through a passage (12.2 m long x 0.91 m wide) for eight laps using an electric goad two times per lap. Pigs in the low-intensity handling treatment were moved at their own pace through the passage for eight laps using a livestock panel and paddle. Biopsy samples were collected from the LM at the beginning of feed withdrawal, at the end of the handling procedure, and 4 h after handling. Blood samples were collected 2 h before and immediately after the handling procedure. There were no interactions between feed withdrawal and handling intensity for any of the variables measured. Feed withdrawal decreased (P < 0.05) baseline and posthandling body temperature (38.85 vs. 38.65 degrees C; SEM = 0.060 and 39.70 vs. 39.37 degrees C; SEM = 0.04, respectively) and blood glucose, lowered (P < 0.05) baseline partial pressure of oxygen and partial pressure of carbon dioxide, and increased (P < 0.01) baseline and posthandling plasma free fatty acid concentrations. High-intensity handling produced higher (P < 0.01) posthandling lactate and glucose, and lower (P < 0.01) posthandling blood pH (7.33 vs. 7.18 +/- 0.02, respectively), bicarbonate, base excess, and total carbon dioxide than low-intensity handling. Longissimus muscle glycolytic potential of fasted pigs was lower (P < 0.01) than in fed pigs at the end of the handling procedure (177.2 vs. 137.0 micromol/g of wet tissue; SEM = 10.08, respectively). There was no effect of handling intensity on longissimus muscle glycolytic potential. Feed withdrawal did not attenuate the blood acid-base changes caused by handling; however, the combination of feed withdrawal and handling decreased muscle glycolytic potential.  相似文献

In this study, we investigated the effect of adding organic acids to the drinking water of finishing pigs 2 weeks prior to slaughter on the shedding and prevalence rate of Salmonella at slaughter. Approximately 600 animals from four Belgian pig herds infected with Salmonella were included. At two herds, the study was conducted twice. Before the start of the study, overshoes were taken at the different herds. Two weeks prior to the expected slaughter date, the pigs were randomly divided into two groups (treatment and control group) each containing on average 50 animals within each herd. The treatment group received from this day onwards acidified drinking water (pH = 3.6-4.0), the control group received non-treated water (pH = 7.8-8.5). All other housing, feeding and management factors were identical in both groups. At the slaughterhouse, 10 pigs of each group (20 pigs for each group of study group 6) were randomly selected and sampled (blood, contents of ileum and rectum, mesenteric lymph nodes and carcass swabs). All samples were immediately transported to the laboratory and submitted to Salmonella isolation. Salmonella was isolated out of 11.9% (66/554) of the samples taken at the slaughterhouse, with the highest frequency found in the content of the ileum (18.7%), followed by 17.8% in the lymph nodes, 7.2% in the content of the rectum and 3.6% in the carcass swabs. The results did not reveal a significant difference between the treatment and control groups for the different slaughterhouse samples. The study documented that the investigated control strategy namely, the strategic application of organic acids during the last 2 weeks prior to slaughter was insufficient to decrease Salmonella shedding and contamination shortly before and during slaughter.  相似文献

A study on slaughter pigs from Saskatchewan detected Salmonella organisms in 12.5% and 5.2% of cecal content and ileocaecal lymph node samples, respectively. Cecal content prevalence was associated with larger farms and longer lairage periods. Antimicrobial resistance was detected in 41.5% of the isolates. Porcine carcasses contaminated with Salmonella typhimurium pose significant public health problems. Prolonged faecal shedding of Salmonella in pigs contributes to the contamination level of carcasses. Although the mechanism of prolonged faecal shedding is not yet clarified, the CS54 Island, and more specifically the shdA gene encoding a fibronectin binding autotransporter protein, was identified as an important locus for intestinal colonization and persistence of Salmonella typhimurium in mice. The aim of this study was to assess the contribution of ShdA in faecal shedding of Salmonella typhimurium in pigs. Pigs were orally inoculated with a Salmonella typhimurium wild type field strain or its isogenic shdA mutant strain. For the first few days after inoculation, the shdA mutant strain was excreted more, the diarrhoea was more pronounced and higher numbers of internal organs were infected. No effect on long-term shedding was found. In a porcine ileal loop model, the wild type strain and shdA mutant strain did not show any differences in the induction of neutrophil influx into the intestinal wall and lumen. In conclusion, we have shown that a Salmonella typhimurium deletion mutant in shdA is more virulent during the first days after inoculation and is not significantly impaired in persistence or prolonged shedding in pigs.  相似文献

In Denmark, a serological Salmonella surveillance programme in finishing pig herds has been in place since 1995. The programme was founded on data from experimental studies, which demonstrated a strong association between Salmonella serology and the prevalence of these bacteria. The current study was carried out in three Danish abattoirs to evaluate the correlation under field conditions. A total of 160 Danish finishing pig herds were included. Seven out of these were examined twice, yielding a total of 167 observations. The herds were selected according to their herd serology based on data from the national surveillance. From each herd, samples were taken from 10 finishers at slaughter. The prevalence of Salmonella bacteria was measured at four sites: (1) caecal-content; (2) carcass surface; (3) pharynx; and (4) caecal lymph nodes. A logistic regression model was constructed for each sampling site. Abattoir, sanitary slaughter and herd seroprevalence were used as explanatory variables. The results demonstrated that there was a strong association between herd serology and the prevalence of Salmonella bacteria measured at three of the sampling sites: caecal-content, pharynx, and carcass surface. For these sites, the odds for being culture-positive for Salmonella varied from 1.3 to 1.5 for each increase of 10% in herd serology (P < 0.0001). Keeping pigs in lairage provides a buffer to supply the slaughter line and allows the pigs to recover from the stress of transport. It has implications for the pigs' welfare by influencing their behaviour and ease of handling. In terms of pork quality, optimal lairage times for most pigs in the UK are probably one to three hours. Shorter lairage times are associated with more pale, soft, exudative (PSE) meat and longer times are associated with more dark, firm, dry (DFD) meat, more skin blemishes caused by fighting, and a lower carcase yield. However, when pigs have not been previously stressed by handling procedures, resting them in lairage may have no beneficial effects on the pigs' welfare or meat quality. At very high environmental temperatures there is evidence that a holding period of less than one hour may help to reduce the number of PSE carcases. At temperatures above 10 degrees C showering the pigs with water is beneficial. Fasting times need to be controlled to minimise the potential loss of carcase yield while reducing the volume of gut contents to reduce the risk of carcase contamination. Lairages can act as reservoirs of infection by pathogenic bacteria and there is evidence that longer holding times increase the risk of cross-contamination.  相似文献

In Europe, on-farm biosecurity measures, involving a strict all-in/all-out batch-management system and decontamination of the rearing rooms between consecutive batches, are recommended to control Salmonella infection in growing pigs. However, implementation of these measures is often relaxed under common farming conditions. Therefore, this study was conducted to assess the relative contributions of batch-management system and room decontamination efficacy on Salmonella seroprevalence for different growing rates and subsequent slaughter ages of pigs. Because the impact of these factors cannot be easily evaluated by an observational approach in commercial farms, a stochastic simulation model representing the population dynamics, herd management, and Salmonella infection within a farrow-to-finish pig herd was used. Realistic levels were set for each factor under study (3 for batch-management system and slaughter age; 4 for room decontamination) to generate 54 simulation scenarios. Salmonella shedding prevalence in groups of slaughter pigs was then compared. A sensitivity analysis was performed to rank the impacts of the 3 factors on output. Batch-management system had little effect. In contrast, room decontamination efficacy had the greatest impact on Salmonella prevalence in pigs at slaughter. A drop in decontamination efficacy from 100 to 50%, with a strict all-in/all-out batch-management system and for all slaughter ages tested, noticeably increased (P<0.001) the prevalence and almost doubled it for the reference slaughter age. Our results suggest that the control of Salmonella in pig herds should primarily focus on room decontamination efficacy. Provided that a good level of room decontamination is ensured, some flexibility in batch management, in terms of pig mixing, would be acceptable to limit the number of underweight pigs delivered to the slaughterhouse.  相似文献

Pigs from three farms were deprived of food for up to one hour, 12 hours or 18 hours before being sent for slaughter. In lairage, the animals' behaviour was monitored, and at slaughter a blood sample was collected and analysed for cortisol, lactate and creatine phosphokinase, potential indicators of stress and physical activity. The carcases were assessed for skin damage as an index of fighting, and rigor in the hind leg as an indicator of stress and/or fatigue. Measurements were also made of cold carcase weight, backfat thickness and liver glycogen concentration. General activity was very high on entry to the lairage pen. Drinking and mounting occurred almost immediately. Fighting developed after an exploratory period, and could last up to 60 minutes. There were large differences in the behaviour of pigs from the three farms. Pigs from farm A fought frequently but showed little mounting activity, whereas pigs from farm C were involved in mounting but little fighting. The period of food deprivation had no effect on average skin damage or rigor score, but the frequency of carcases with the highest scores was different The pigs deprived of food for up to an hour had the lowest incidence of severe skin damage and high rigor scores. Boars had a higher incidence of severe skin damage but a lower incidence of carcases with a high rigor score than gilts. Liver glycogen was almost completely depleted in the pigs deprived of food for 12 and 18 hours and was lower in the pigs deprived for up to an hour than in animals fed immediately before slaughter. The period of food deprivation had no effect on the levels of cortisol, creatine phosphokinase or lactate in the blood.  相似文献

试验采用单因子设计,在保证营养水平相同的情况下,共设5种日粮,其中包括对照组(基础日粮)、鱼粉组(基础日粮+2.5%鱼粉)、酵母组(基础日粮+2.5%酵母)、酵母去酶组(基础日粮+2.5%酵母并扣除酶制剂)和酵母去药组(基础日粮+2.5%酵母并扣除抗生素)。选用28日龄断奶的DLY三元杂交仔猪60头,试验期为28d。试验主要考察平均日采食量(ADFI)、平均日增重(ADG)和料肉比(F/G)。结果表明,从整个试验期来看,添加酵母组和鱼粉组都能提高仔猪的生产性能,尤以酵母组的提高幅度最大,酵母组的ADG、ADFI分别较对照组提高14.9%(P<0.05)、4.2%,料肉比降低了9.5%(P<0.05);ADG、ADFI分别较鱼粉组提高10.6%、5.4%,料肉比降低4.7%(P<0.05)。通过比较对照组、鱼粉组、酵母去酶组、酵母去药组这4组全期生产性能指标,发现对照组虽然采食量较高,但ADG和料肉比仍是最差,其中料肉比明显高于酵母去酶组和鱼粉组(P<0.05);酵母去酶组较对照组ADG提高6.4%,料肉比降低11.9%(P<0.05);酵母去药组较对照组ADG提高1.7%,料肉比降低2.0%,差异不显著(P>0.05)。试验结果说明,酵母代替鱼粉的效果是肯定的,同时具有替代酶制剂的潜力,另外,酵母饲料作为一种绿色的饲料来源,在一定条件下代替抗生素也是可行的。  相似文献
