Summary Solanum L. sect. Petota Dumort., the potato and its wild relatives, contains 232 species, according to the latest taxonomic interpretation of Hawkes (1990). Section Petota is distributed in the Americas from the southwestern United States to southern Chile. This economically important group has attracted the attention of numerous taxonomists with various taxonomic philosophies. There are 531 validly published basionyms in the group, plus 67 nomina nuda or nomina dubia, and subsequent transfers to other ranks raise the number of names to 664. The taxonomy of sect. Petota has been dominated in recent years by J. G. Hawkes & J. P. Hjerting (co-workers) and C. M. Ochoa. The near simultaneous and independent publication of three recent books by these workers, treating the Bolivian species of sect. Petota, provides a novel opportunity to compare their taxonomic philosophies. Additionally, a recent independent treatment of all of the South American species by L. E. Gorbatenko provides a new interpretation of affiliations of species to series that is compared to the recent treatment of Hawkes and earlier treatments of S. M. Bukasov and D. S. Correll. These treatments differ in the placement of species into series, species boundaries, rank of infraspecific taxa, and hypotheses of hybridization. Our analysis illustrates the wide differences of taxonomic interpretation possible when independent workers treat the same material. The comparison provides insights into unresolved taxonomic questions in sect. Petota and indicates the need for a practical taxonomic resolution that will benefit plant breeders and other researchers on wild potatoes. Reasons for the discrepancies are discussed and suggestions for future research are provided.  相似文献

Isozyme variation was studied in 87 plants from 32 cultivated and wild accessions of banana passion fruit (P. tripartita var. mollissima, P. tripartita var. tripartita, P. tarminiana, and P. mixta), rosy passion fruit (P. cumbalensis), tin-tin (P. pinnatistipula), gulián (P. ampullacea), P. antioquensis, P. bracteosa, and P. manicata, from the Andes of Venezuela, Colombia, and Ecuador. Six polymorphic enzyme systems (IDH, PGDH, PGM, DIA, PRX, and ACP) revealed 31 zymotypes characterized by the presence or absence of 31 electromorphs. Cluster analysis separated clearly the accessions of P. tarminiana, P. tripartita, P. mixta, and P. cumbalensis from the less typical species of subgenus Tacsonia, which is consistent with morphological evidence. P. mixta showed the highest intraspecific variation and the closest affinity with P. tripartita. The accessions of these two species formed two clusters, one dominated by Colombian genotypes and the other dominated by Ecuadorian genotypes. One of the P. tripartita var. mollissima accessions clustered close to P. tarminiana accessions. The affinity between these three species is particularly interesting for conservation and use of banana passion fruit genetic resources. All the other species formed monospecific clusters.  相似文献

Six species of the genus Prunus occur inAndalusia. Matters regarding their ecology, ethnobotanical aspectsand conservation are discussed, as well as knowledge about thegermplasm of those species with greater economic importance and theirdegree of conservation, namely: P.avium, P.mahaleb and P.insititia. Various conservation measures areproposed for wild germplasm and for the germplasm of local varieties:a) conservation efforts should target the best populations,except in the case of P.avium and P.insititia, where every individual should beaddressed, including both wild and local cultivars; b) theDehesa del Camarate (Sierra Nevada) is proposed as an areafor in situ germplasm conservation; c) theawareness of the importance of Prunus germplasmshould be increased among forest workers, environmental managers andthe public; d) some silvicultural techniques are proposed, suchas mixed Prunus patches, hedge treatment,Z-tree selection at the early stages and the protection ofroot suckers; e) improved, selected P.avium and P.mahaleb germplasm should be used at highlyproductive sites since these two species can be cultivated as highvalue timber trees; f) some Prunus speciescan also provide profitable NTFPs in the region. Details regardingdifferent collection areas are covered. Finally, some topics arerecommended for scientific research. A wild tetraploid oat Avena maroccana Gdgr. was collected from the 11 populations in the periphery of Rommani and Casablanca geographic groups of Morocco. Genetic diversity of the species was investigated using six allozyme systems. Allelic frequencies were scored representing eight polymorphic and five monomorphic loci. Coefficient of gene differentiation (Gst) was 0.3019, which indicated great genetic differentiation. The number of alleles per locus was 2.6154, the percentage of polymorphic loci was 61.54, and the expected heterozygosity was 0.2462 in all populations. Genetic diversity in A. maroccana was high in comparison to self-pollinated species. In total, nine heterozygotes resulting from outcrossing were found in the progeny from M1, M3, M4, M22 and M26. The population of M7 had peculiar alleles Pgd–2SS and Pgd-1SS in high frequency. M9 had the lowest level of diversity out of the 11 populations. Geographic and genetic distances between all the populations were not significantly correlated with each other (r = 0.0996). Cluster analysis showed that two groups, (M1, M22, M2 and M4) and (M3, M23, M8, M5 and M26) were apparently differentiated. Two populations of the Casablanca group, M7 and M9 were independent from each other, and were separated distinctly from the other populations. Genetic diversity of the Rommani and Casablanca groups was almost the same in all the parameters. This was due to the similar man-made habitat such as roadside or rich fertile soil and brown clay soils. The population size of A. maroccana was small and restricted to the narrow central Morocco with great genetic differentiation so that genetic diversity may be reflected from the results of genetic drift and outcrossing heterozygote segregation.  相似文献

Oryza latifolia is the most abundant and widespread wild relative of rice in the lowlands of Costa Rica. It also has the most diverse morphology. However, little is known about the genetic diversity and mating system of this allotetraploid species. Genetic analyses of nine populations from different life zones from the Pacific and Atlantic slopes were performed using six isozyme loci. Differences in genetic variability were observed among populations, of which Palo Verde and Santa Rosa were the most diverse. Most Atlantic populations clustered together, and a similar result was obtained with the Pacific populations. High levels of interpopulation diversity were observed while most populations were monomorphic for at least one genotype, suggesting little genetic flow within populations. Even in polymorphic populations no variation was observed within progeny groups, combined with Hardy-Weinberg disequilibria in most populations, commonly observed in autogamous species or in species with clonal reproduction. However, the high frequency of heterozygous-like patterns may suggest that the reproductive system of O. latifolia might be more complex. Future research may explain the genetic patterns as well as the reproductive biology of this species. The knowledge of the genetic diversity and mating system of O. latifolia could contribute to the implementation of genetic resource conservation strategies and gene flow analyses, as well as of breeding programs for rice improvement.  相似文献

Isozyme studies of genetic diversity and evolution in Hippophae   总被引:4,自引:0,他引:4  
Summary To provide information on the genetic variation, differentiation and evolution in Hippophae seed samples of 25 populations from China, Finland and Russia were electrophoretically analyzed. Of six loci investigated, four were good genetic markers for identifying species and subspecies. The percentages of polymorphic loci per population were 40.3% and 62.5% at 0.95 and 0.99 polymorphic criteria respectively. The mean number of alleles per locus per population was 2.1. Total genetic diversity in the material was 0.4614. Genetic diversity partitioning showed that there was a large amount of diversity residing within geographical populations (0.1354), between subspecies within species (0.1046) and between species (0.2566) but not between geographical populations (0.0114). There were nearly twice as many negative fixation indices as positive ones in Hippophae populations. The phylogenetic tree agreed very well with botanic classifications of the species and subspecies and their geographical distributions, and quantitatively presented the genetic relationships of 25 populations. A detailed view of the evolutionary stages in Hippophae showed clearly a general decline of similarity as evolutionary divergence continued, which further explained the evolution process in Hippophae.  相似文献

The allozyme variation of 15 Tunisian wild populations of Mentha pulegium L. threatened by human activities (clearing, hard-grazing, ploughing, traditional uses) was surveyed by the analysis of 14 isozyme loci using horizontal gel starch electrophoresis. The species exhibited a high level of genetic variation within populations (the mean Ap = 2.20, P = 72%, Ho = 0.349 and He = 0.229), which indicates a predominately outcrossing mating system and the recruitment of new genotypes via dispersal seeds. The genetic structure analysis of the populations using F statistics indicates no inbreeding, and showed an excess of heterozygosity for few loci. The moderate differentiation of populations (FST = 0.110) and the low rate of gene flow between them (Nm = 2.02) might been caused by recent isolation of the populations through biotope disturbances. The value of Nei's genetic identity varied from 0.839 to 0.999 reflecting a relatively low genetic divergence between populations. Cluster analysis using UPGMA method and Nei's genetic identity values, showed that populations geographically close didn't always cluster together. However, populations within the same bioclimatic stage generally subclustered together indicating that differentiation between bioclimatic regions occurred.  相似文献

Amplified fragment length polymorphism (AFLP) analysis was used to evaluate the genetic relationships and diversities of Chinese vegetable mustards. Fourteen pairs of primers generated a total of 366 scorable fragments among 16 accessions of Brassica juncea studied, of which 296 bands were polymorphic with an average of 21.1% polymorphic bands per primer combination. Genetic similarities were obtained using Nei and Li similarity coefficients, and a dendrogram of the 16 accessions was made by UPGMA clustering method. The Nei and Li Similarity coefficient value ranged from 0.63 to 0.88. This result indicated that the 16 accessions of B. juncea possessed high level genetic variations. The cluster analysis showed that the vegetable mustards could be grouped into two main groups and some minor rami, which was partially in accordance with the traditional classification that based on different edible organs of vegetable mustards. The incongruity between morphological and molecular classification might be attributed to the high selection pressure during domestication of Chinese vegetable mustards, producing some accessions with similar genetic backgrounds evolving into abundant morphological variations. The great diversification among Chinese vegetable mustards not only provides an excellent object for molecular evolution research of B. juncea but also is of great value for widening the genetic basis of breeding programs and breeding materials selection. Besides, our study also indicates that AFLP are informative and can provide significant insights for genetic diversity research in B. juncea.  相似文献

Random amplified polymorphic DNA (RAPD) and isozyme electrophoresis (IE) techniques were used to estimate the level of genetic diversity in a sample of cacao germplasm existing at the International Cocoa Genebank, Trinidad. Twenty-six cocoa populations represented by 459 cocoa genotypes were analysed using IE and 22 populations represented by 353 cocoa genotypes were analysed using RAPD. Despite few differences in the classification of the populations, both techniques revealed three major groups: the indigenous trees, the cultivated Trinitario and the cultivated trees from Ecuador. Two-thirds of the partitioned diversity were found within populations and one-third between the populations, with both techniques.  相似文献

Diversity in morphological characters of 206 accessions of 14 wild Vigna species from India was assessed. Of these, 12 species belonged to Asian Vigna in the subgenus Ceratotropis and two were V. vexillata and V. pilosa belonging to subgenus Plectotropis and Dolichovigna, respectively. Data on 71 morphological traits, both qualitative and quantitative, were recorded. Data on 45 qualitative and quantitative traits exhibiting higher variation were subjected to multivariate analysis for establishing species relationships and assessing the pattern of intraspecific variation. Of the three easily distinguishable groups in the subgenus Ceratotropis, all the species in mungo-radiata group, except V. khandalensis, viz. V. radiata var. sublobata, V. radiata var. setulosa, V. mungo var. silvestris and V. hainiana showed greater homology in vegetative morphology and growth habit. The species, however, differed in other plant, flower, pod and seed characteristics. Within species variation was higher in V. mungo var. silvestris populations and three distinct clusters could be identified in multivariate analysis. V. umbellata showed more similarity to V. dalzelliana than V. bourneae and V. minima in the angularis-umbellata (azuki bean) group. Within species variations was higher in V. umbellata than other species in the group. In the aconitifolia-trilobata (mothbean), V. trilobata populations, were more diverse than V. aconitifolia. The cultigens of the conspecific wild species were more robust in growth, with large vegetative parts and often of erect growth with three- to five-fold increase in seed size and seed weight, except V. aconitifolia, which has still retained the wild type morphology to a greater extent. More intensive collection, characterisation and conservation of species diversity and intraspecific variations, particularly of the close wild relatives of Asian Vigna with valuable characters such as resistance to biotic/abiotic stresses, more number of pod bearing clusters per plant etc. assumes great priority in crop improvement programmes.  相似文献

The genetic diversity of 38 cultivated populations of Sesamum indicum L. from four different regions of Turkey was estimated at the DNA level with the random amplified polymorphic DNA (RAPD) technique. Sixty-one bands were obtained for all populations 78% of which were polymorphic. Analysis of molecular variance (AMOVA) was used to investigate the genetic diversity of the populations which yielded highly significant differences among populations within regions (91.9% of the total genetic diversity). According to AMOVA and Shannon's index that were performed separately for each region, the highest value of genetic variation was observed among Northwest region populations (CV = 7.7; H0 = 0.304) and lowest in the Southeast regions' populations (CV = 2.6; H0 = 0.068). Nei and Li's similarity index was calculated and phylogenetic tree was established using the neighbor-joining algorithm. This phenetic analysis grouped 35 of 38 accessions in six groups leaving three highly diverse accessions outside. Wagner phylogenetic method was used to assess the phylogenetic relationships among the populations. In the majority-rule consensus tree, only 7 of the 32 forks showed above 60% occurrence. Using Principal Coordinate Analysis (PCO) of the RAPD data set, the groups were clearly separated along the first three axis. These results indicate that RAPD technique is useful for sesame systematics, and should be valuable for the maintenance of germplasm banks and the efficient choice of parents in breeding programs.  相似文献

Information on genetic diversity and genetic relationships among genotypes of Brassica carinata is currently limited. The objectives of this study were to evaluate patterns and levels of genetic diversity in B. carinata based on amplified fragment length polymorphisms (AFLP) as compared with Brassica juncea and Brassica nigra, and to evaluate agronomic and seed quality data for plants grown in the field in western Canada. A total of 296 AFLP bands were generated from four primer pair combinations and scored for presence/absence in 66, 20 and 7 accessions of B. carinata, B. juncea and B. nigra, respectively. B. carinata was less genetically diverse than the other two species. Differences in diversity were evident in the proportion of polymorphic loci within each species: 23, 35 and 50% for B. carinata, B. nigra and B. juncea, respectively. Pair-wise similarity measures based on the Jaccard coefficient were highest among accessions of B. carinata and showed the narrowest range: 0.911 (0.810–0.981) compared to B. nigra: 0.569 (0.438–0.660) and B. juncea: 0.715 (0.345–0.951). AFLP-based genetic distance information can be used by plant breeders to select diverse genotypes. AFLPs are also useful for fingerprinting cultivars and two primer pair combinations were sufficient to uniquely identify all the accessions of B. carinata. More variation among accessions was identified in the agronomic trial than had previously been described in studies of B. carinata in western Canada, but the data were too limited to draw conclusions regarding specific accessions. Overall, the findings were in agreement with other published work describing the favourable agronomic potential of this species.  相似文献

RAPD analysis was conducted in 22 cultivars of P. nigrum(black pepper) from South India and one accession each of P. longum and P. colubrinum. Twenty-four primers generated 372 RAPD markers of which 367 were polymorphic. Jaccard's similarity between pairs of accessions ranged between 0.11 and 0.66 with a mean of 0.38. Among P. nigrum cultivars, the similarity ranged between 0.20 and 0.66 and the mean was 0.42. The existence of wide genetic diversity as revealed in the present study is supported by earlier reports of extensive inter- and intrapopulation morphological variability in pepper cultivars from South India. UPGMA dendrogram and PCO plot revealed P. colubrinum to be most distant of the three species. Genetic proximity among P. nigrum cultivars could be related to their phenotypic similarities or geographical distribution. Greater divergence was observed among landraces than among advanced cultivars. Landraces grown in southern parts of coastal India and those grown in more northern parts were grouped in separate clusters of the dendrogram.  相似文献

When planning optimal conservation strategies for wild and cultivated types of a plant species, a number of influencing biological and environmental factors should be considered from the outset. In the present study Brassica rapa was used to illustrate this: to develop Scandinavian conservation strategies for wild and cultivated B. rapa, DNA-marker analysis was performed on 15 cultivated and 17 wild accessions of B. rapa plus 8 accessions of the cross compatible B. napus. The B. rapa cultivars were bred in Sweden and Finland in 1944–1997 and the wild B. rapa material was collected from Denmark, Sweden and United Kingdom. The B. napus accessions were bred within the last 20 years in the Scandinavian countries. Results were based on scoring of 131 polymorphic ISSR markers in the total plant material. A Bayesian Markov chain Monte Carlo (MCMC) approach implemented in NewHybrids demonstrated a clear distinction of B. rapa and B. napus individuals except for three individuals that seemed to be backcrosses. The backcrossed hybrids descended from two Swedish populations, one wild and one escaped. The overall pattern of genetic variation and structure in B. rapa showed that cultivated and wild B. rapa accessions formed two almost separated clusters. Geographical origin and breeding history of cultivars were reflected in these genetic relationships. In addition, wild populations from Denmark and Sweden seemed to be closely related, except for a Swedish population, which seemingly was an escaped cultivar. The study point to that many processes, e.g. spontaneous introgression, naturalisation, breeding and agricultural practise affected the genetic structure of wild and cultivated B. rapa populations.  相似文献

Genetic diversity throughout the rapeseed (Brassica napus ssp. napus) primary gene pool was examined by obtaining detailed molecular genetic information at simple sequence repeat (SSR) loci for a broad range of winter and spring oilseed, fodder and leaf rape gene bank accessions. The plant material investigated was selected from a preliminary B. napus core collection developed from European gene bank material, and was intended to cover as broadly as possible the diversity present in the species, excluding swedes (B. napus ssp. napobrassica (L.) Hanelt). A set of 96 genotypes was characterised using publicly available mapped SSR markers spread over the B. napus genome. Allelic information from 30 SSR primer combinations amplifying 220 alleles at 51 polymorphic loci provided unique genetic fingerprints for all genotypes. UPGMA clustering enabled identification of four general groups with increasing genetic diversity as follows (1) spring oilseed and fodder; (2) winter oilseed; (3) winter fodder; (4) vegetable genotypes. The most extreme allelic variation was observed in a spring kale from the United Kingdom and a Japanese spring vegetable genotype, and two winter rape accessions from Korea and Japan, respectively. Unexpectedly the next most distinct genotypes were two old winter oilseed varieties from Germany and Ukraine, respectively. A number of other accessions were also found to be genetically distinct from the other material of the same type. The molecular genetic information gained enables the identification of untapped genetic variability for rapeseed breeding and is potentially interesting with respect to increasing heterosis in oilseed rape hybrids.  相似文献

Gliadin composition has been analysed in 403 accessions of spelt wheat (Triticum aestivum ssp. spelta); 61 different patterns were found for the -gliadins, 44 for the -gliadins, 19 for the -gliadins and 15 for the -gliadins. A subset of 333 accessions belonging to fifty populations from Asturias, North of Spain, showed high levels of genetic variation (A = 3.89, P = 0.88, Ne = 3.35 and He = 0.553), indicating that 82.5% of the genetic variation was within populations, and only 18.5% among populations. Thirty-five of these populations presented more of five accessions, in this new subset the values of genetic variation were higher that those of fifty populations (A = 4.49, P = 0.91, Ne = 3.80 and He = 0.595). The genetic variation within populations was 59.7% of the total, and 40.3% among populations, which could be associated to fixation effects of some alleles by genetic drift.  相似文献

This investigation was conducted to assess changes in the genetic structure of two varieties of two species of annual Brassica. Seeds of B. napus cv. Topas and B. rapa Broccoletto were sent to nine research institutes in different geographical areas of Europe for regeneration. The multiplied material was sent back after one year of regeneration and analysed electrophoretically. The original populations of each species and their multiplied samples were stained for 12 different enzymes, of which 4 were found to be polymorphic (DIA, SKD, GPI and PER). It was possible to detect considerable differences in isozyme patterns in B. napus and allelic frequencies in B. rapa, both within and between populations. When the original population was compared with the regenerated samples, the Chi-square homogeneity test for all pairwise comparisons revealed distinctness with a 99% probability for B. napus and 95% probability for B. rapa with one or more of the enzyme systems examined. Furthermore, the average of gene diversity analysis (Nei, 1973) revealed that some regenerated populations have less while others have increased genetic variation compared with the original population. These observations indicated that the frequencies were non-random and considerable shifts in genetic diversity have occurred during multiplication. In addition, different regeneration procedures have caused the fixation of certain alleles.  相似文献

Lolium and Festuca are two important genera of cool-season forage and turf grasses worldwide. Lolium temulentum L. (darnel ryegrass) has been proposed as a model species for genomics studies of cool-season forage and turf grasses. A study with 41 darnel ryegrass, three tall fescue (Festuca arundinacea Schreb.), two tetraploid fescue (F. glaucescens), and two meadow fescue (F. pratensis) genotypes was initiated to (i) identify a set of microsatellite (simple sequence repeats) markers useful for L. temulentum L., and (ii) to utilize such markers for assessing the genetic variability of L. temulentum accessions collected from different geographical regions of the world. A total of 40 tall fescue (TF) EST-SSRs and 60 Festuca–Lolium (F × L) genomic SSRs were screened on a subset of eight genotypes. The selected 30 tall fescue EST-SSRs and 32 F × L genomic SSRs were used for further analysis of genotypes. The TF-EST- and the F × L genomic-SSRs identified 10.3 and 9.3 alleles per marker, respectively with an average polymorphic information content (PIC) value of 0.66. The phenogram based on 319 EST-SSR and 296 genomic SSR fragments, grouped L. temulentum accessions into three major clusters except for accession ABY-BA 8892.78. Lolium temulentum accession ABY-BA 8892.78 did not cluster with any other accession. The Festuca clusters were distantly related with darnel ryegrass clusters with a similarity coefficient of 0.26. The selected set of tall fescue EST- and F × L genomic SSRs were useful in assessing L. temulentum genetic diversity and could benefit the genetic improvement of members of the Festuca–Lolium complex.  相似文献

Brahmi (Bacopa monnieri) is atraditional Indian medicinal plant known for its natural nootropicaction of saponins present in large amount in its shoots. Acollection of 24 B.monnieri accessions from differentagro-climatic zones of India and an introduction from Malaysiamaintained in the field genebank at CIMAP was analysed for RAPDvariation. Among the 40 random primers tested, 29 primers generatedone or more polymorphic bands. The number of polymorphic bandsgenerated was primer dependent, ranging from 2 to maximum of 8.Similarity matrices were generated from the RAPD data on the basis ofNei's estimates of similarity indices and dendrograms wereconstructed based on UPGMA clustering. All the accessions were foundto be in the range of 0.8–1.0 of similarity, which isindicative of a narrow genetic base among the various accessions witha medium level of polymorphism. It was possible to differentiateindividual accessions, showing differences in morphological andgrowth properties at DNA level. The observed low levels of geneticvariation were attributed to interplay of sexual and vegetative modesof reproduction and similarity of local environments in habitats ofB. monnieri.  相似文献
