The levels of activity of sorbitol dehydrogenase (SDH), alanine transaminase (ALT), aspartate transaminase (AST), lactate dehydrogenase (LDH) and its isoenzymes, α-hydroxybutyric dehydrogenase (α-HBDH) and creatine kinase (CK) were determined in various tissues of six clinically normal cats. The plasma half times (T1/2) of several hepatic enzymes were also measured in three clinically normal cats.  相似文献

The activities of serum creatine kinase (CK), lactate dehydrogenase (LD) and LD isoenzymes were studied in 14 Prestice black pied pigs from a herd affected with congenital tremor. Mean CK activity was 19.57 +/- 3.56 mu kat.l-1 for 6 adult pigs, and it was 21.03 +/- 1.33 mu kat.l-1 and 20.42 +/- 1.23 mu kat.l-1 for the affected (n = 5) and control (n = 3) piglets, respectively. No significant differences were demonstrable between the groups in CK activity. Total serum LD and LD-4 as well as LD-5 isoenzyme activities were higher in sows. Piglets affected with congenital tremor showed an increase in total LD enzyme and LD-5 isoenzyme activity. It is concluded that no relationship exists between congenital tremor and serum CK activity in piglets. At the same time, there is a positive relationship between congenital tremor and significantly (P < 0.01) elevated LD enzyme and LD-5 isoenzyme activity. The results allow us to suggest that total lactate dehydrogenase and LD-5 isoenzyme activities could be used as biological markers of congenital tremor in piglets.  相似文献

Changes in hepatic function tests to induced toxicity in the bovine liver   总被引:1,自引:0,他引:1  
Graded hepatic damage was induced in mature lactating dairy cows to measure the sensitivity of several hepatic diagnostic tests. In a preliminary study, cows were dosed with .05, .10 and .20 ml/kg body weight of carbon tetrachloride. Extreme changes occurred in hepatic tests by 24 h post-dosing, and all died by 35 h with massive diffuse centrilobular necrosis of hepatic cord cells. Dosing was decreased to induce non-fatal hepatic changes. Cows in Groups 1, 2, 3 and 4 were orally dosed with .002, .004, .006 or .01 ml/kg body weight of carbon tetrachloride, respectively. Serum enzymes of hepatic origin, bilirubin, plus bromosulfophthalein dye clearance were assayed before dosing and up to d 14 post-dosing. Liver biopsies were performed 24 h post-dosing for histological evaluation and cytochrome P-450 content. Hepatic concentrations of cytochrome P-450 were decreased in all the dosed cows. Serum activities of sorbitol dehydrogenase and gamma-glutamyl transferase were elevated in cows of Groups 3 and 4 and glutamic-oxaloacetic transaminase in cows of Group 4 by 24 h. Serum alkaline phosphatase, glutamic-pyruvate transaminase, lactate dehydrogenase, bilirubin, urobilinogen and bromosulfophthalein dye clearance were not significantly different. Mild to moderate diffuse centrolobular necrosis was observed in livers of cow of Groups 3 and 4, but no pathological changes were seen in Groups 1 and 2.  相似文献

Changes in serum enzyme levels, liver histology and liver function tests have been correlated to determine the usefulness of these tests in assessing liver status. The effects of carbon tetrachloride administration on these parameters has been determined in a group of 20 sheep. Normal levels, elevated levels after injury and the effect of elapsed time after injury are reported for serum glutamic dehydrogenase, sorbitol dehydrogenase, glutamic-oxaloacetic transaminase, glutamic-pyruvic transaminase, lactate dehydrogenase, fructose-1-phosphate adlolase, alkaline phosphatase, cholesterol and proteins. Variation in the time of elevation of enzyme activities may be useful in determining the elapsed time between acute injury and serum sampling. In comparison to sheep fed an adequate diet, a diet with a restricted protein intake was associated with increased severity of histological lesions and decreased liver function.  相似文献

After racing 722 m, 16 Greyhounds were evaluated to determine changes in hematologic, biochemical, blood-gas, and acid-base values following exercise. Values were determined before racing (T0), immediately after racing (T1), and 3 hours after racing (T2). Significant changes detected immediately after racing included increased heart rate, respiratory rate, and rectal temperature. Significant changes in hematologic values included increases in PCV, total plasma protein, hemoglobin, RBC, WBC, neutrophils, and lymphocytes. Change was not detected in values for monocytes, eosinophils, and neutrophil/lymphocyte ratio. Other increases included those for plasma concentrations of sodium, chloride, calcium, lactic acid, aspartate transaminase, alanine transaminase, alkaline phosphatase, creatine kinase, lactate dehydrogenase, and glucose. Concentrations of potassium and urea did not change. Measurement of blood-gas and acid-base status revealed significant increases in PaO2 and base deficit, whereas PaCO2, pH, and bicarbonate decreased. Three hours after exercise, all vital signs and blood-gas and acid-base values, except for PaCO2, which was still slightly low, had returned to baseline (T0) values. Most biochemical values had also returned to baseline, although sodium, chloride, aspartate transaminase, and creatine kinase were still high, and urea was low. Many hematologic values were still different from baseline values, with high values for WBC, neutrophils and neutrophil/lymphocyte ratio, and low values for PCV, total plasma protein, hemoglobin, RBC, and lymphocytes.  相似文献

The normal activities of lactate dehydrogenase and creatine kinase isoenzymes in the tissues and serum of clinically normal five-and-a-half-month-old lambs are presented. The percentage distribution of the lactate dehydrogenase isoenzymes in serum, liver, heart, lung and skeletal muscle were in agreement with previous studies but the distribution in ovine abomasum, small intestine, large intestine and red blood cells has not been previously described. Up to five creatine kinase isoenzymes were detected in ovine tissues and four in serum. One creatine kinase isoenzyme present in serum and tissues was thought to represent a mitochondrial isoenzyme which is absent from the normal serum of other species. Estimations of both lactate dehydrogenase and creatine kinase isoenzymes allowed all the tissues examined to be distinguished and were therefore more likely to allow tissue-specific isoenzyme patterns to be detected in serum than the estimation of the isoenzymes of either enzyme alone.  相似文献

Serum lactate dehydrogenase (LDH) isoenzyme activity was analyzed in cetaceans. Animals that were treated by i.m. injection and others that received azole therapy had distinctly different LDH isoenzyme profiles. A third distinctive pattern was occasionally observed in clinically normal animals with elevations in total transaminase and LDH activity levels. DH isoenzyme activity patterns were not affected by mild or moderate hemolysis, refrigeration after 24 hr, or freezing for 24 hr with subsequent thawing. However, severe hemolysis produced artifactual changes similar to those observed in individuals that received injections but of a lesser magnitude. DH isoenzyme activity patterns may provide useful corroboration of other clinical findings when diagnostic modalities are limited, especially to differentiate nonspecific enzyme elevation from nonpathologic elevations in serum enzyme concentrations due to i.m. injections or azole therapy.  相似文献

Serum enzyme activities of sorbitol dehydrogenase, glutamate dehydrogenase, gamma glutamyltransferase, alkaline phosphatase, aspartic aminotransferase, and creatine kinase, were measured in five clinically normal mixed-breed goats. Tissue activities of these enzymes were also measured in two goats. These basal serum values were then used to determine the response to treatment with carbon tetrachloride (CCl4). The basal value for serum and hepatic tissue sorbitol dehydrogenase were appreciably greater for goats than previously reported for sheep and cattle. The change in the above serum enzymes after CCl4 treatment resembled that in sheep, but the amount of sorbitol dehydrogenase increase was less than that in sheep. This study established basal tissue and serum enzyme activity values and demonstrated the efficacy of the use of changes in serum S.D.H. and G.D.H. activity as indicators of acute hepatopathy in goats.  相似文献

Tissue and plasma enzyme activities in juvenile green iguanas   总被引:1,自引:0,他引:1  
OBJECTIVE: To determine activities of intracellular enzymes in 8 major organs in juvenile green iguanas and to compare tissue and plasma activities. ANIMALS: 6 green iguanas < 1 year old. PROCEDURE: Lysates of liver, kidney, epaxial muscle, heart, lung, spleen, small intestine, and pancreas were analyzed for alkaline phosphatase (ALP), lactate dehydrogenase (LDH), aspartate transaminase (AST), alanine transaminase (ALT), gamma-glutamyltransferase (GGT), creatine kinase (CK), glutamate dehydrogenase (GMD), and amylase (AMS) activities. RESULTS: In general, low tissue enzyme activity coincided with low plasma activity. The CK activity was high in epaxial muscle and the heart and low in all other tissues tested. The AMS activity was found exclusively in the pancreas. Moderate LDH and AST activities were found in all tissues. Low ALT and ALP activities were found in a variety of tissues. Plasma and tissue activities of GGT and GMD were low or undetectable. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggest that plasma CK activity may be muscle specific in iguanas, but high values may not always indicate overt muscle disease. The AMS activity may be specific for the pancreas, but the wide range of plasma activity would likely limit its diagnostic usefulness. Activities of AST and LDH may reflect tissue damage or inflammation, but probably do not reflect damage to specific tissues or organs.  相似文献

Eight- and nine-week-old Hungarian Landrace pigs were tested with halothane as described by Laky et al. (1985). Immediately after the test blood samples were taken for determination of the activity of serum creatine kinase (CK), creatine kinase MB (CK-MB) isoenzyme, aldolase (ALD), lactate dehydrogenase (LDH) and alpha-hydroxybutyrate dehydrogenase (alpha-HBDH). Elevated creatine kinase, creatine kinase MB isoenzyme and aldolase activities indicating enhanced susceptibility to stressors were found in 92% of the halothane reacted and 16% of the halothane non-reacted animals. In these individuals the activities of lactate dehydrogenase and alpha-hydroxybutyrate dehydrogenase were also high. Data of the literature show a close relationship between enhanced susceptibility to stressors and halothane reaction in pigs. It was suggested, therefore, that determination of the activity of appropriate serum enzymes might be used for detecting this enhanced susceptibility.  相似文献

Otic medications containing triamcinolone or dexamethasone were administered twice daily for 21 days to 2 groups of 4 healthy dogs each. Serum alkaline phosphatase, gamma-glutamyltransferase, alanine transaminase, and aspartate transaminase activities, and serum bile acid and lipoprotein-X concentrations were assayed for 35 days. Increased serum activities for gamma-glutamyltransferase, alkaline phosphatase, and alanine transaminase were detected by day 7 and peaked at day 21. Increases were greater in dogs given the dexamethasone-containing medication. Enzyme activity returned to baseline by day 35. Serum aspartate transaminase activity and bile acid and lipoprotein-X concentrations did not increase.  相似文献

Serum and CSF lactate dehydrogenase (LDH) activity and LDH isoenzyme profile, as well as total protein, were measured in samples obtained from 26 healthy Beagles. The LDH activity in serum was approximately 10 times that in the CSF. The CSF LDH isoenzyme profile was a mirror image of the serum LDH isoenzyme profile. Age was negatively correlated (ie, as age increased, the activity decreased) with LDH3 in CSF and LDH5 in serum and was positively correlated with LDH1 in serum. Seemingly, simultaneous measurement of serum and CSF LDH activity and LDH isoenzyme profile can aid in establishing status of the blood-brain barrier, whether brain damage exists and whether other organ systems are involved.  相似文献

The effects of a diet containing a high proportion of rapeseed meal on the activity of certain plasma enzymes were studied in laying birds. The enzymes studied were alkaline phosphatase (AP), aspartate transaminase (AST), L-gamma-glutamyl-transferase (gamma-GT), isocitrate dehydrogenase (ICDH), leucine arylamidase (LAP), malate dehydrogenase (MDH) and sorbitol dehydrogenase (SDH). No notable differences were observed between the plasma AP, LAP or SDH activities of the birds given the rapeseed meal and the birds receiving a soyabean meal control diet throughout the experiment. However, the plasma AST and gamma-GT activities of the treated birds showed slight elevations while their plasma ICDH and MDH activities showed more marked elevations, which are indicative of liver damage, in response to the diet. Macroscopic observations of the livers of the birds at the end of the experiment were in fairly good accord with the elevation in plasma ICDH and MDH activities noted for the individual birds.  相似文献

Ten male Holstein-Friesian calves naturally infected by Mycobacterium paratuberculosis were experimentally re-infected orally at an average of 17 days. Monthly measurements were conduced of the following activities, in the period between post infection days 160 and 400: total protein (TPR), albumin (ALB), cholesterol (CHOL), triglycerides (TRIG), Zn and Cu concentrations as well as sorbitol dehydrogenase, lactate dehydrogenase (LDH), alpha-hydroxybutyrate dehydrogenase (alpha-HBDH), gamma-glutamyltransferase, aspartate aminotransferase (AST), alanine aminotransferase (ALT), creatine kinase (CK), alkaline phosphatase and fructose-1,6-diphosphate aldolase (ALD). TPR, ALB, TRIG, and CHOL were reduced by day 400, in conjunction with disorders of digestion and absorption. Increased activities of CK, ALD, LDH, alpha-HBDH, AST and ALT primarily indicated damage to skeletal muscle and/or liver. Serum CK and ALD activities as well as TRIG and TPR concentrations may serve as aids to specific diagnosis of paratuberculosis, particularly in the advanced stage of the disease.  相似文献

The association between hepatic lipidosis (HL) and disease in 59 anorectic, ketotic, lactating Holstein heifers and cows was investigated. Severe HL, as determined by histologic evaluation of liver tissue, was present in 46 animals; only half of these animals required intensive treatment for ketosis, and only half had serum biochemical evidence of liver disease, as determined by the presence of a test value of 2-fold or greater than the upper limit of the reference range for at least 2 of the 4 serum tests: gamma-glutamyl transferase, aspartate aminotransferase, and sorbitol dehydrogenase activities and bile acid concentrations. Most cattle with biochemical evidence of liver disease and severe HL had been lactating for 14 or more days. Cows that required intensive treatment inconsistently had serum biochemical evidence of liver disease.
Although cattle with severe HL had significantly higher serum bilirubin concentrations and aspartate aminotransferase and sorbitol dehydrogenase activities than cattle with less severe lipidosis, the specificity of abnormally high serum sorbitol dehydrogenase activity or bilirubin concentration for severe lipidosis was only 8%. Abnormally high serum aspartate aminotransferase activity was 83% sensitive and 62% specific for severe lipidosis. Serum glucose and total carbon dioxide concentrations were significantly lower in cattle with severe lipidosis than in those with mild or moderate lipidosis, and low serum glucose or total carbon dioxide concentrations were rare in cattle without severe lipidosis. From these data, we conclude that the use of a single biochemical or histopathologic criterion to define severity of disease or degree of liver compromise in anorectic, ketotic cows results in the misidentification of many animals.  相似文献

Nineteen purebred Beagles of various ages (4, 5, 13, and 47 weeks) were inoculated with North American Trypanosoma cruzi isolates obtained from an opossum (Tc-O), an armadillo (Tc-A), or a dog (Tc-D). Dogs were grouped on the basis of clinical outcome of infection. During the acute stage of disease, dogs of group 1 (n = 7 inoculated with Tc-O or Tc-A) died or were euthanatized because of the severity of disease. Dogs of group 2 (n = 5 inoculated with Tc-O or Tc-A) developed acute disease, but survived to develop chronic disease. Dogs of group 3 (n = 7 Tc-D-inoculated dogs) developed neither acute nor chronic disease. Dogs of group 4 (n = 4--2 dogs 13 weeks old and 2 dogs 47 weeks old) served as noninoculated controls. Clinical signs associated with severe acute myocarditis developed in dogs of groups 1 and 2 between postinoculation day (PID) 15 and 28. Generalized lymphadenopathy and lymphocytosis were observed in all dogs of groups 1, 2, and 3 between PID 14 and 17. Serum alanine transaminase and aspartate transaminase activities and urea nitrogen concentration were high, and glucose concentration was low prior to death of dogs in group 1. Serum activities of isoenzymes of creatine kinase were significantly (P less than 0.05) high in only 1 dog (group 1), whereas serum lactate dehydrogenase isoenzyme activities were not significantly high in any dog.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Plasma bile acid concentration was determined in normal dogs,sheep, calves and ponies for three days before and six days after liver damage, induced by carbon tetrachloride. In all species, a significant increase in plasma bile acid concentration was associated with a concomitant significant increase in plasma sorbitol dehydrogenase and transferase activity. Plasma bilirubin also significantly increased in all animals except the dogs. Results suggested that plasma bile acid levels could be used to test liver function in domestic animals.  相似文献

Total lactate dehydrogenase and the absolute and percentage levels of its isoenzymes were measured in lung lesions and macroscopically normal areas of lung from lambs with chronic proliferative exudative pneumonia and acute pasteurella pneumonia. Lung lesions had a higher total enzyme activity which was associated mainly with increases in the activity of the LDH4 and LDH5 isoenzymes, particularly in chronic pneumonia, and gave lung lesions a considerable potential for altering the serum isoenzyme distribution. Thus, the nature of any changes in the serum isoenzyme distribution will depend on whether the isoenzymes are released from abnormal or normal areas of lung. This appears to be the first report on lactate dehydrogenase isoenzymes in ovine pneumonia.  相似文献

To differentiate the origin of high total lactate dehydrogenase (LD) activity in canine sera, a spectrophotometric method based on the preferential inhibition of cardiac LD isoenzymes by pyruvate was performed. Comparison with the electrophoretic separation of LD isoenzyme activities and determination of the hydroxybutyrate dehydrogenase-to-LD ratio indicated that the method proposed gave a better discrimination between cardiac and hepatic LD activities than did the other tests.  相似文献

Twenty sheep were dosed with either Pachystigma pygmaeum or Fadogia homblei belonging to the Rubiaceae. The experimentally-induced cardiotoxicoses were monitored by various clinical pathological parameters and heart function tests. Elevated AST (aspartate transaminase) activity in the serum proved to be a more reliable indicator of cardiac damage in gousiekte than either LD (lactate dehydrogenase) or CK (creatine kinase). Persistent increases of AST activity were recorded from c. 14 days after commencement of dosing, and this activity sometimes peaked as late as 30 days after the dosing had ceased. Tachycardia and diminished heart function were registered only terminally. Lesions of gousiekte were present in all the sheep that were exposed to the plants. In a field outbreak of P. pygmaeum, where 60 out of 90 sheep died, 14 out of the 15 animals examined had increased AST levels compared with none of the 15 controls. These results indicated that increased enzyme levels can be of use to identify affected animals during latency in a natural outbreak of gousiekte.  相似文献
