Thyroid status is compromised in a variety of acute and chronic infections and toxin-mediated disease states. Conversion of thyroxine (T4) into the metabolically active hormone, triiodothyronine (T3), is catalyzed by 5'-deiodinase (5'D). Our objective was to determine the effect of endotoxin (LPS) challenge with and without L-arginine (Arg) infusion on hepatic activity of 5'D and plasma concentrations of T4 and T3. In a 2 x 2 factorial, beef heifers (275-310 kg b.wt.) were fed low (8% CP; 6.5 kg/d) or high (14% CP; 7.2 kg/d) isocaloric protein diets (1.96 Mcal/kg DM) for 10 d before LPS challenge. L-Arginine in saline (0.5 g/kg b.wt.) or saline alone was infused i.v. throughout an 8 hr period starting 2 hr before bolus LPS injection (Escherichia coli, 055: B5; 0.2 microg/kg; i.v.). Blood samples were collected at -2, 0, 3, 6, 12, and 24 hr relative to LPS injection. Liver samples were obtained 20 hr before, and then 6 and 24 hr after LPS challenge using a biopsy needle. Plasma T4 and T3 concentrations were not affected by dietary CP or Arg. Compared with levels at 0 hr, LPS challenge decreased plasma T4 (P < 0.01) and T3 (P < 0.001), respectively, 8.4% and 28.9% at 6 hr and 19.7% and 31.3% at 24 hr. Consistent with these changes, the T3:T4 ratio was lower than that at 0 hr (P < 0.001) 22.0% at 6 hr and 13.5% at 24 hr. Hepatic 5'D activities 20 hr before LPS injection were 2.80 +/- 0.11 nmol I- x hr(-1) x mg protein(-1) and decreased 24 hr after LPS, respectively, 45.4% (P < 0.01) and 17.6% (P < 0.05) in saline- and Arg-infused heifers. The results indicate that mild LPS challenge in cattle inhibits hepatic generation of T3 and decreases plasma concentrations of thyroid hormones. Photoperiod manipulation, specifically a long-day photoperiod (LDPP), increases milk production in lactating cattle. We have previously reported that the galactopoietic effect of LDPP is associated with an increase in circulating IGF-I, which seems to occur independently of changes in concentrations of GH, IGFBP-2, and IGFBP-3. This study tested the hypothesis that LDPP increases the expression of GH receptor (GHR) 1A messenger RNA (mRNA) in the liver. Two groups of Holstein steer calves (98 +/- 4 d old) were maintained indoors and exposed to LDPP (16-h light: 8-h dark; n = 6) or short-day photoperiod (SDPP; 8-h light: 16-h dark; n = 6) for 60 d. Calves were individually fed a grain- and alfalfa-based diet. Jugular blood samples were collected weekly and via cannula at 15-min intervals for a 4-h period on d 1, 26, and 55 of the study to monitor pulsatile hormone secretion. Serum was harvested and assayed for IGF-I, prolactin (PRL), and GH using RIA. Liver biopsies were obtained at 3-wk intervals to quantify changes in hepatic IGF-I and GHR 1A mRNA using real-time PCR. Steer BW increased during the study but did not differ between treatments. No differences in ADG or total DMI were observed. Relative to SDPP, calves on LDPP had higher (P < 0.05) serum IGF-I concentrations. Concentrations of PRL increased (P < 0.01) in calves exposed to LDPP compared with calves exposed to SDPP. Differences (P < 0.05) in pulsatile GH secretion were also detected. Hepatic IGF-I and GHR 1A mRNA were positively correlated with circulating IGF-I concentrations, and although both increased with time, they were not affected by photoperiod treatment. These results confirm that LDPP increases circulating concentrations of IGF-I, but this occurs independently of changes in IGF-I synthesis and GHR 1A mRNA expression in the liver. Over a 2-year period involving a total of 168 Angus cows and their calves, 2 studies were conducted to evaluate the effect of once-daily suckling on cow reproduction, calf growth, and gastrointestinal parasitism. Experiment I compared 2 restricted suckling (RS) groups with 1 normal suckling (NS) group. Each group consisted of 24 cows with their calves (24 cows-calves/group = 24 cows and 24 calves/group). In the RS groups, suckling by the calves was restricted to once a day for 45 days, and then the calves were allowed to suckle normally throughout the remainder of the experiment. In the NS group, the calves suckled normally throughout the experiment. Experiment II compared 2 groups of RS cow-calves (24 cow-calves/group) with 2 groups of 24 NS cow-calves/group. In experiment I, there were no differences (P less than 0.05) in pregnancy rate or in the average number of nematode eggs/g of feces (epg) between the cows from the 2 treatment groups. The RS calves consistently had higher epg counts than did the NS calves during the 45 days of RS (169 vs 62 epg, P less than 0.05) and from the time of their return to pasture until weaning (266 vs 186 EPG, P = 0.27). The RS calves also had a higher average coccidia oocyst count during the RS period only. Although Se is essential for antioxidant and thyroid hormone function, factors influencing its requirement are not well understood. A survey and two experiments were conducted to determine the influence of cattle breed and age on selenoprotein activity and the effect of maternal Se supplementation on cow and calf selenoprotein activity and neonatal thyroid hormone production. In our survey, four cowherds of different ages representing three breeds were bled to determine the influence of breed and age on erythrocyte glutathione peroxidase activity (RBC GPX-1). All females were nonlactating, pregnant, and consumed total mixed diets (Holstein) or grazed pasture (Angus and Hereford). In our survey of beef breeds, yearlings had greater average RBC GPX-1 activity than mature cows. In Exp. 1, neonatal Holstein heifers (n = 8) were bled daily from 0 to 6 d of age to determine thyroid hormone profile. An injection of Se and vitamin E (BO-SE) was given after the initial bleeding. Thyroxine (T4) and triiodothyronine (T3) concentrations were greatest on d 0 and decreased (P < 0.05) continuously until d 5 postpartum (156.13 to 65.88 and 6.69 to 1.95 nmol/L, d 0 to 5 for T4 and T3, respectively). Reverse T3 concentrations were 3.1 nmol/L on d 0 and decreased (P < 0.05) to 0.52 nmol/ L by d 5. In Exp. 2, multiparous Hereford cows were drenched weekly with either a placebo containing 10 mL of double-deionized H2O (n = 14) or 20 mg of Se as sodium selenite (n = 13). After 2 mo of treatment, Se-drenched cows had greater (P < 0.01) plasma concentrations than control cows (84.92 vs. 67.08 ng/mL), and at parturition, they had plasma Se concentrations twofold greater than (P < 0.05) control cows (95.51 vs. 47.14 ng Se/mL). After 4 mo, cows receiving Se had greater (P < 0.05) RBC GPX-1 activity than controls; this trend continued until parturition. Colostrum Se concentration was twofold greater (P < 0.05) in Se-drenched cows than control cows (169.97 vs. 87.00 ng/mL). Calves born to cows drenched with Se had greater (P < 0.05) plasma Se concentration, RBC GPX-1, and plasma glutathione peroxidase activity on d 0 compared with calves born to control cows. By d 7, no differences in plasma glutathione peroxidase activity in calves were observed. Maternal Se supplementation did not influence calf thyroid hormone concentrations. Selenium provided by salt and forages is not adequate for cattle in Se-deficient states. 相似文献

Previously, GH response to GHRH challenge at weaning has been shown to be indicative of ADG during a standard postweaning growth performance test in Angus cattle. In this study, we tested the hypothesis that GH response to GHRH before weaning would predict postweaning ADG. Bulls with the highest and lowest GH responses to GHRH over a 3-yr period, relative to their contemporaries, were used as sires, to allow for examination of the persistence of GH response to GHRH through selection. The selected calves in this study were sired by one of four Angus bulls chosen based on their GH response to GHRH (high response, n = 2; low response, n = 2). Forty-nine Angus calves (bulls, n = 24; heifers, n = 25) were challenged with GHRH at approximately 60, 105, and 150 d of age and at weaning (219 d; SD = 25). Blood samples were taken immediately prior to and 10 min following an i.v. clearance dose of 4.5 microg of GHRH/100 kg BW and, 2 h later, immediately prior to and 10 min following a challenge dose of either 1.5 or 4.5 microg of GHRH/100 kg BW. Two hours later, the procedure was repeated, with each calf receiving the other challenge dose. Body weight was measured every 28 d and ADG was calculated over a 140-d growth performance test (heifers and bulls maintained separately). Data were log-transformed for statistical analyses. In the selected bulls and heifers, response of GH to 1.5 microg of GHRH/100 kg BW at 60 and 105 d of age was positively related (P < 0.05) to postweaning ADG. Response to 4.5 microg of GHRH/100 kg BW at 105 d of age and at weaning was positively related (P < 0.01) to postweaning ADG. Inclusion of sire in the analysis improved the relationship between GH response and ADG for calves of sires with high GH responses from R2 = 0.18 (P = 0.01) to R2 = 0.33 (P = 0.02). When the GH response to GHRH of the unselected calves at weaning was added to the data from the selected animals and analyzed, the GH response of the bulls was related to postweaning ADG (R2 = 0.09; P = 0.04). In conclusion, GH response to GHRH as early as 60 d of age is indicative of postweaning ADG in beef cattle. The effect of triglycerides (Tg) on GRF-mediated GH secretion was examined in 2 groups of twelve ten-day old male calves. Twelve calves were intravenously infused with a lipid-heparin solution (5 mg Tg and 0.3 IU heparin/kg body wt/min for 90 min). The twelve control calves received in the same way, the same volume of saline. Thirty minutes after the start of infusion, GRF 1–29 (human amide, 0.16 μg/kg body wt) was intravenously injected in six animals of each group. Mean plasma GH levels reached peak concentrations in the 2 groups 5 min after GRF injection. However the area under the GH response curve, when lipid-heparin was given, was significantly diminished compared to the response when saline was given. In the same time, lipid-heparin treatment increased plasma SRIF concentration. These data suggest that an increase in plasma Tg concentration, induced by lipid-heparin infusion, inhibits GRF-mediated GH secretion, possibly through stimulation of SRIF secretion. 相似文献

This study examined the influence of bovine growth hormone (bGH) on liver and kidney thyroxine-5'-monodeiodinase activity (TMA) in growing beef cattle. In a preliminary trial (trial 1), tissue samples were obtained at slaughter from two placebo-injected and two bGH-injected (29.2 IU/day for 14 days before slaughter) Hereford heifers (398 kg avg slaughter wt), with one heifer on each treatment fed at either 1.0 or 2.0 times maintenance energy requirement. In a second trial, tissue and plasma samples were obtained from six placebo-injected and six bGH-injected (29.2 IU/day for 19 days) Hereford steers (331 kg avg slaughter wt) fed at 1.8 times maintenance energy requirement. In a third trial, liver tissue and plasma samples were obtained from 14 Angus X Hereford steers (270 kg avg wt) fed either an 8% protein, 1.96 Mcal-metabolizable energy/kg diet (8 steers) or a 14% protein, 2.67 Mcal-metabolizable energy/kg diet (6 steers) in association with acute administration of bGH. Half the steers in each group were given two injections per steer of either placebo or 37.8 IU bGH at 24-hr intervals and slaughtered 24 hr after the second injection. Units of tissue TMA in all trials were measured at slaughter; one unit defined as 1 ng T3 generated/hr/mg protein during incubation with T4 (1.3 fM). In trial 1, TMA in liver and kidney was 2.86 and 1.21 times greater, respectively, for bGH than for placebo treatments. In trial 2, units of TMA in liver homogenates were higher (P less than .05) for bGH (4.14) than for placebo (2.83) treatments and higher (P less than .02) in liver microsomal preparations for bGH (32.7) than for placebo (27.3) treatments. Units of TMA in kidney homogenates were also higher (P less than .10) for bGH (1.48) than for placebo (.87) treatments and higher (P less than .02) in kidney microsomal preparations for bGH (23.0) than for placebo (16.4) treatments. Following acute injection of bGH (trial 3), units of TMA in liver homogenates were higher (P less than .01) for bGH (2.5) than for placebo (1.8) treatments. Plasma T4 (70.0 vs 73.5 ng/ml) and T3 (1.08 vs 1.18 ng/ml) concentrations appeared slightly higher in bGH-treated steers (trial 2) and increased by 20 and .5 ng/ml, respectively, (P less than .01) within 6 to 12 h after acute injection of bGH (trial 3). Human pancreatic growth hormone releasing factors (hpGRF) (1–40) and (1–44) were administered iv in sheep, pigs and calves to determine their effectiveness in stimulating GH release in these species. Both peptides produced a rapid increase in plasma GH concentration in all three species at dose levels ranging from .0065 to .65 nmol/kg. Moreover, there was no difference in the GH-secretory response observed between hpGRF(1–44)NH 2 and (1–40)OH in sheep. Sheep also responded to hpGRF(1–40)NH 2 and (1–40)OH as well as [his 1]- and [tyr 1]-hpGRF(1–40)NH 2 in a similar manner. Rat hypothalamic GRF was less effective than [his 1]-hpGRF(1–40), while the response to bGRF was not significantly different from hpGRF(1–40) in stimulating GH secretion in sheep. Although all three species responded to hpGRF, the elevation in plasma GH levels above baseline were greater after hpGRF injection in sheep than in pigs or calves. Subcutaneous injection of hpGRF in sheep was an effective mode of administration of the peptide, although the effect was not as long-lasting as that after iv injections and higher doses were required to stimulate GH secretion. 相似文献

We investigated the effect of cellooligosaccharide (CE) or a combination of dextran and Lactobacillus casei ssp. casei strain JCM1134 T (synbiotic; SB) feeding on growth performance, fecal condition and hormone concentrations in Holstein calves. Fifty‐two female Holstein calves were randomly assigned to three treatment groups: CE feeding group ( n = 16), SB feeding group ( n = 18), and control group ( n = 18). Body weight at 90 days of age, as well as daily body weight gain (DG) and feed efficiency after weaning to 90 days of age were greater ( P < 0.05) in the CE feeding group than in the control group. The total fecal score tended to be lower ( P < 0.1) in the SB feeding group than in the control group. Plasma insulin concentration was higher ( P < 0.05) in the CE feeding group than in the control group at 90 days of age. Our results indicate that CE feeding improved DG and feed efficiency in calves. On the other hand, there was less effect on growth performance and fecal Escherichia coli counts in calves fed SB. 相似文献

To determine the effect of chronic treatment with human growth hormone-releasing factor (1-29)NH2 (GRF) and(or) thyrotropin-releasing factor (TRF), 20 calves averaging 70.2 kg BW were divided into four groups (n = 5) according to a 2 X 2 factorial design. For 86 d, calves in each group received twice daily s.c. injections of either .9% NaCl, GRF (5 micrograms/kg BW), TRF (1 microgram/kg BW) or GRF (5 micrograms/kg BW) plus TRF (1 microgram/kg BW). On d 87, all calves received a s.c. injection of GRF (5 micrograms/kg BW) plus TRF (1 microgram/kg BW). Blood samples were collected every 20 min for 18 h on d 1, 29, 57 and 85, and for 8 h on d 87. Hormone responses were measured as area under the hormone concentration curve over time. GRF and TRF acted in synergy (P less than .10) on GH release throughout the treatment period. Growth hormone responsiveness to GRF and(or) TRF decreased (P less than .01) with days of treatment, but this decrease was due to aging rather than to chronic treatment, because GH response to GRF plus TRF was similar (P greater than .10) between control and treated calves on d 87. TRF increased prolactin (Prl) concentration until the end of the treatment period (P less than .01). The response of thyroid-stimulating hormone (TSH) to TRF disappeared (P greater than .10) after 1 mo of treatment, whereas the thyroxine (T4) response decreased (P less than .01) throughout the treatment period. The use of the biochemical and serological parameters pepsinogen, gastrin and antibodies against Cooperia, Ostertagia and Dictyocaulus in an enzyme-linked immunosorbent assay was evaluated in diagnosing gastrointestinal parasitism and lungworm disease in seven oxfendazole pulse-release bolus (OPRB)-treated and seven non-treated first-season grazing calves. During the grazing season, no difference in growth rate was observed and the control animals showed no clinical symptoms. The Dictyocaulus antibody levels in the controls increased from turn-out, while in the treated animals the rise was postponed until August. The Cooperia antibody titres for both groups were comparable. At the end of the grazing season, the control group showed significantly higher pepsinogen and gastrin values, and Ostertagia antibody titres. It was concluded that these parameters are a useful tool in confirming subclinical parasitism and that preventive treatment with the OPRB in a low to moderately infected environment, together with a high plane of nutrition, provided little benefit to the farmer. 相似文献

The purpose of the present study was to clarify the hypothalamic action of leptin on the secretion of luteinizing hormone (LH) and growth hormone (GH) in cattle. Intracerebroventricular (the third ventricle) injections of leptin were given to fully fed castrated Holstein calves. Blood samples were collected at 10‐min intervals for 60 min after injection and 20‐min intervals for 60 min before injection and for 60–180 min after injection through an indwelling catheter in the external jugular vein. Plasma LH and GH levels were examined by homologous radioimmunoassay. The administration of 10 µg of leptin stimulated a significant ( P < 0.05) release of GH but not LH. Average GH levels began to rise after 30 min and were significantly increased at 40, 50 and 60 min after the injection, compared with the respective control values ( P < 0.05). The present result suggests that leptin may act partly on the hypothalamus to stimulate the release of GH in castrated calves. 相似文献

A study was conducted to determine the effects of BST on the growth rate, hormone profiles and carcass composition of preweaned Holstein bull calves. Injection of BST resulted in increased average daily gains and feed intakes (P less than .05) compared to that of control animals. This increased rate of gain coupled with increased intakes resulted in identical feed conversion efficiencies between treatments. Fractional weights (organ weight/body weight) of selected organs and tissues were unaffected by BST treatment. Carcass fat percentage was reduced in BST-treated calves (28.1 +/- 1.18% and 32.8 +/- 1.84% for BST and controls groups respectively, P less than .05) while no differences were seen for carcass DM, CP, ash or gross energy content. Plasma BST levels were higher (P less than .0001) in BST treated calves while plasma IGF-1 levels were not different between treatments. Plasma IGF-1 concentrations correlated (P less than .0001, R = .71) with average daily gains. The results of this trial suggest that growth responses are inducible with BST in young calves (7 days to 3 months). 相似文献

Blood samples were taken from 19 double-muscled (DM) and 20 normal (N) bull calves at the ages of 1.5, 2, 2.5, 3, 3.5, 4, 5.5, 6.5 and 9 mo to compare the plasma concentrations of growth hormone and insulin in DM with those in N bull calves and to relate these to differences in growth rate between the two breed groups. Double-muscled bull calves were lighter (P less than .0001) than N calves at all ages and had lower (P less than .001) preweaning and postweaning rates of gain. Double-muscled bull calves had lower (P less than .01) mean growth hormone concentration than N calves. Mean growth hormone concentration was correlated positively with body weight and preweaning rate of gain. The effect of age on growth hormone concentration was linear (P less than .05); however, mean growth hormone concentration fluctuated between ages 1.5 to 4.5 mo but stabilized after 5.5 mo of age in both breed groups. Mean insulin concentration was lower (P less than .01) in DM than in N bull calves. The effect of age on insulin concentration was both linear and quadratic (P less than .0001). Mean insulin concentration generally was constant in both breed groups, at around .75 ng/ml, from 1.5 to 6.5 mo of age but rose sharply to around 1.67 ng/ml after weaning when the bulls were put on a high-energy diet. 相似文献

A series of studies examined the binding characteristics and ontogeny of hepatic growth hormone binding sites in dairy bulls on d 2, 30, 180, and 365 of age. Binding of iodinated recombinant bovine growth hormone ([125I]rbGH) to liver membrane receptors was membrane protein-dependent. Receptors were considered growth hormone-specific, because physiological concentrations of bovine prolactin (bPRL) failed to displace [125I]rbGH from bovine hepatocyte membranes. Only 50% of [125I]rbGH was bound reversibly to hepatic microsomes. Addition of dithiothreitol (DTT) to the receptor-assay buffer increased the binding of [125I]rbGH to hepatic membranes in a time-dependent manner. Moderate concentrations of Ca++ and Mg++ in the receptor-assay buffer had no detectable effects on binding of [125I]rbGH to hepatic microsomes. In growing dairy bulls, specific binding of [125I]rbGH per milligram of membrane protein increased from 1.9 +/- 1.8% at d 2 to 14.1 +/- 1.8% at d 180 and then declined to 5.2 +/- 1.6% at d 365. Likewise, concentration of insulin-like growth factor (IGF)-I in serum was low during the 1st mo of age (d 2, 13.3 +/- 8.8 ng/ml; d 30, 9.7 +/- 8.8 ng/ml), but it became maximal at d 180 (151.0 +/- 8.8 ng/ml). Circulating concentrations of IGF-II increased linearly during the 1st yr of growth. Serum concentrations of GH, triiodothyronine, and thyroxine declined from 39.9 +/- 6.5, 2.7 +/- .2, and 75.4 +/- 4.6 ng/ml at d 2 to 16.5 +/- 6.5, 1.3 +/- .2, and 53.4 +/- 4.6 ng/ml at d 30, respectively, and remained low through 1 yr of age. Insulin concentration in serum did not change significantly with development. Results indicated that increasing concentrations of specific bGH receptors in the bovine liver may play a key role in regulating postnatal growth in cattle. 相似文献

Sub-clinical parasitism in spring-born single suckled beef calves was investigated from the middle of their first grazing season until weaning or housing later the same year. The study was conducted on four beef suckler herds in southern England over a 3-year period and involved a total of 334 spring-born beef suckler calves and their dams. The animals were grazed extensively on pastures naturally infected with nematode larvae. At the start of each period of observation, faecal samples were taken from calves and cows and subjected to routine worm egg counts; calves were re-sampled at the end of the grazing season.In July in each year and at each location the calves were ranked by initial weight within sex, paired according to rank and randomly allocated to either an untreated control group or a group in which the calves were each treated with an ivermectin sustained-release (SR) bolus. The calves in both trial groups, and their dams, were grazed together until weaning or housing. The calves were weighed at the initial allocation and at the end of the study. The adult cows were not treated with any anthelmintic during the study.The faecal nematode egg counts (FECs) conducted in July showed that the suckler cows were excreting worm eggs at low concentrations: range 0-100 eggs per gram (epg), with one individual count of 500epg, 88% of the cows sampled had counts of <50epg. Similarly, the counts from the calf samples were fairly low in July: range 0-250epg, 73% of the calves sampled had counts of <50epg. By the end of the grazing season, the faecal samples from the untreated control calves showed higher values: range 0-650epg, with only 58% having an epg of <50.The average rate of daily liveweight gain in the untreated heifer calves was 0.79kg per day, the corresponding figure for the heifer calves treated with the ivermectin SR bolus in mid-summer was 0.88kg per day; the difference of 90g per day was significantly different (P=0.0118). The average rate of daily liveweight gain in the untreated bull calves was 0.91kg per day, the corresponding figure for the bull calves treated with the ivermectin SR bolus in mid-summer was 1.01kg per day; the difference was significantly different (P=0.0169). 相似文献

The effect of strategic treatments with ivermectin in first-season calves exposed to trichostrongyle nematodes on naturally contaminated pasture was studied. Twenty first season heifer calves were divided into 2 groups, according to live weight, and on 22nd May each group was turned out onto a 1 hectare pasture. Group A (Plot A) was treated with ivermectin at weeks 3, 8 and 13 after turn out, while group B (Plot B) served as an untreated control group. The study showed that control calves exhibited increase in trichostrongyle egg counts in August, while treated calves were excreting low numbers of trichostrongyle eggs. Pasture larval counts on Plot B (control animals) were low during the first part of the grazing season, followed by a steep rise towards the end of July. In contrast, the numbers of infective larvae recovered from Plot A remained low throughout the season. Both groups showed comparable weight gains from May up to the middle of July. However, from then on, Group B (controls) had lower weight gains than ivermectin treated Group A. From the end of July onwards, most untreated calves (Group B) showed clinical signs of parasitic gastroenteritis. It can be concluded that the strategical ivermectin treatments were successful, and faecal egg counts, pepsinogen levels and herbage larval counts clearly demonstrated that this was accomplished through suppression of pasture contamination with nematode eggs and subsequent reduction of pasture infectivity. 相似文献

Unthrifty calves occurred sporadically in Japanese Black (beef cattle) in an area in northeastern Japan. The states of unthrifty development, pedigree, clinico-biochemistry and the secretory function of bovine growth hormone (bGH) in pituitary were investigated. The total cholesterol concentration and CK, AST and LDH activities in the serum showed higher values than those of control calves. Basal bGH concentrations in the serum and bGH secretory reactivity in the insulin tolerance test (ITT) were showed to be significantly lower than those of the control calves. Furthermore, sperm donated from a specific bull had been used for these unthrifty calves. This study suggested that the present occurrence of unthrifty calves represented ateliosis possibly caused by congenital hypopituitarism which decreased of bGH secretory function. 相似文献

A study was initiated to determine whether development of a functional ruminant digestive system was associated with alterations in plasma growth hormone (GH) concentration. Holstein bull calves were fed milk or milk with grain until studied at the age of 1 month (n = 12). Calves placed on pasture with some grain supplementation were studied at the age of 3 months (n = 6) to determine plasma GH concentration in an animal with fully developed ruminant metabolism. Blood samples were taken at 10-minute intervals for 5 hours, followed by administration of bovine GH-releasing factor (0.075 micrograms/kg of body weight) and subsequent blood sample collection for 1 hour. On the following day, a blood sample was collected via jugular cannula, clonidine (10 micrograms/kg) was administered, and blood samples were subsequently obtained. Data indicated that milk-fed calves had higher mean plasma GH concentration than did either milk/grain-fed or older calves. The difference in mean plasma GH concentration was related to higher secretory pulse amplitude. Pituitary responses to bovine GH-releasing factor did not differ among the 3 groups, but response to clonidine were greater in milk-fed calves than in calves of the other groups. These data indicate that the change from a nonruminant to a ruminant-type gastrointestinal tract, perhaps attributable to subsequent changes in metabolism, may induce changes in hypothalmic function to decrease GH concentration. 相似文献
