Soil samples were collected from Panchamarhi dry deciduous forest in Satpuda Biosphere Reserve, India to determine the effect of hill slopes and altitude on the population size of methanotrophic bacteria. Population size, in range of 4×105-3.6×107 g−1 dry soil, was negatively correlated with altitude and increased exponentially (r2=0.97, P<0.001) at steep slope (60°) while logarithmically (r2=0.97, P<0.001) at low slope (45°). Soil organic C, total N, and soil moisture increased while C/N ratio and temperature decreased down the hill slope. The results indicated that nutritional status of the soil across the slopes determines the methanotrophic bacterial population size.  相似文献

Organic nitrogen (DON) was extracted from two improved pasture soils, one of which had been re-colonized by acid heath vegetation, and a blanket peat. Although the quantities extracted in H2O, 10 mM CaCl2, 500 mM K2SO4 and 50 mM Na2HPO4 were not consistent, mean extractable DON as a proportion of total N was greater in the two grazed pastures (0.4%) than in the peat (0.2%). Averaged over the four extractants, free α-amino N was greater in the peat and least in the improved pasture soil and accounted for 26% of DON in the peat and less than 5% in the mineral soil. Amino N increased after 6 M HCl hydrolysis, and this combined N contributed 56% to DON in extracts of the mineral soil compared with only 36% in the peat This variation in the relative contributions of free and combined amino N to DON indicated qualitative differences in the composition of DON between the three soils.  相似文献

Precipitation, throughfall, stemflow and soil water content were measured, and interception, transpiration, evaporation, runoff, deep percolation and soil water recharge were estimated in the natural Liaotung Oak (Quercus liaotungensis) and regrown Black Locust (Robinia pseudoacacia) forestlands in the hill and gully region of the China Loess Plateau. Four stands (south- and north-facing slopes) of two forests were studied between May 27, 2006 and October 31, 2007. Hydrological fluxes were calculated using a coupled water and heat flow model called CoupModel. Throughfall, stemflow and soil water content were used to calibrate the model. The simulations indicated that, interception, vegetation transpiration and soil water evaporation were the main components of water consumption in the 4 stands, accounting for about 90% of the precipitation. The simulated interception and vegetation transpiration in the south-facing slope (154 and 327 mm in regrown forestland and 173 and 338 mm in natural forestland) were lower than those in the north-facing slope (219 and 344 mm in regrown forestland and 203 and 342 mm in natural forestland). Soil water evaporation in the south-facing slope (416 mm in regrown forestland and 373 mm in natural forestland) was larger than that in the north-facing slope (325 mm in regrown forestland and 330 mm in natural forestland) in the same vegetation stands. This was mainly due to greater vegetation density in the north-facing slope than in the south-facing slope. For the regrown forestlands, the simulated soil water recharge was larger under north-facing slope stands (90 mm) than under south-facing slope stands (76 mm), and the natural forestland in the north-facing slope had the largest soil water recharge (104 mm). The results indicated that vegetation species and slope aspects significantly influenced the water balance budget in the soil–vegetation–atmosphere system. The water budget differences among the 4 stands indicate that care is required for properly selecting regrown tree-species. Soil and water conservation measures must be applied scientifically when converting farmland to forest in the Loess Plateau of China, especially on the south-facing slopes.  相似文献

Agricultural peat soils in the Sacramento-San Joaquin Delta, California have been identified as an important source of dissolved organic carbon (DOC) and trihalomethane precursors in waters exported for drinking. The objectives of this study were to examine the primary sources of DOC from soil profiles (surface vs. subsurface), factors (temperature, soil water content and wet-dry cycles) controlling DOC production, and the relationship between C mineralization and DOC concentration in cultivated peat soils. Surface and subsurface peat soils were incubated for 60 d under a range of temperature (10, 20, and 30 °C) and soil water contents (0.3-10.0 g-water g-soil−1). Both CO2-C and DOC were monitored during the incubation period. Results showed that significant amount of DOC was produced only in the surface soil under constantly flooded conditions or flooding/non-flooding cycles. The DOC production was independent of temperature and soil water content under non-flooded condition, although CO2 evolution was highly correlated with these parameters. Aromatic carbon and hydrophobic acid contents in surface DOC were increased with wetter incubation treatments. In addition, positive linear correlations (r2=0.87) between CO2-C mineralization rate and DOC concentration were observed in the surface soil, but negative linear correlations (r2=0.70) were observed in the subsurface soil. Results imply that mineralization of soil organic carbon by microbes prevailed in the subsurface soil. A conceptual model using a kinetic approach is proposed to describe the relationships between CO2-C mineralization rate and DOC concentration in these soils.  相似文献

Soil respiration was measured with the enclosed chamber method in an ungrazed Leymus chinensis steppe during the growing seasons of 2001 and 2002. Soil respiration rate (RS) was significantly influenced by air temperature (T) at the diurnal scale, and could be described by Van't Hoff's equation (RS = R10 exp(β(T − 10))). At the seasonal scale, the normalized soil respiration rate at 10 °C (R10) was mainly controlled by soil water content (R2 = 0.717, P < 0.001), while the sensitivity of soil respiration to temperature (Q10) was partially affected by absolute growth rate (R2 = 0.482, P = 0.004). Thus, soil respiration could be described as RS = (20.015W − 84.085) (0.103AGR + 1.786)(T−10)/10 during the growing seasons, integrating soil water content (W) and absolute growth rate (AGR) into the temperature-dependent soil respiration equation. It was validated by the observed soil respiration rates in this study (R2 = 0.890, P < 0.001) and observations from near-field experiment (R2 = 0.687, P = 0.011). It implied that accurately evaluating annual soil respiration should include the effects of plant biomass production and other abiotic factors besides air temperature.  相似文献

To increase wetland acreage and biodiversity, Delaware agencies constructed >220 depressional wetlands. During construction, agencies included amendments thought to increase biodiversity. Because the efficacy of amendments is unknown, we investigated their effects on macroinvertebrate and vegetative communities. We selected 20 standardized wetlands (five contained coarse woody debris (CWD) and microtopography amendments (land surface ridges and furrows), five had neither, five had CWD only, and five had microtopography only). Additionally, 12 wetlands had received organic matter amendments (i.e., straw). Insect richness (P = 0.010; r2 = 0.16), insect biomass (P = 0.023; r2 = 0.13), intolerant insect biomass (P = 0.033, r2 = 0.03), Ephemeroptera biomass (P = 0.027; r2 = 0.12), and Odonata biomass (P = 0.046; r2 = 0.10) increased with CWD volume. Obligate plant percent cover increased with microtopographic variation (P = 0.029; r2 = 0.120). Although organic matter amendments did not increase percent soil organic matter (t13.7 = −1.16, P = 0.264), total (P = 0.027; r2 = 0.12), native (P = 0.036; r2 = 0.11), and facultative (P = 0.001; r2 = 0.24) plant richness increased with percent soil organic matter. To enhance biodiversity, constructed wetlands should contain CWD, but additional research is needed to understand the benefits of microtopography and organic matter amendments.  相似文献

Soil water erosion and shallow landslides depend on aggregate stability and soil shear strength. We investigated the effect of vegetation on both soil aggregate stability and shear strength (through direct shear tests) in former croplands converted to vegetated erosion protection areas within the context of China's sloping land conversion programme. Four treatments were analysed in plots comprised of (i) 4 year old crop trees, Vernicia fordii, where understory vegetation was removed; (ii) V. fordii and the dominant understory species Artemisia codonocephala; (iii) only A. codonocephala and (iv) no vegetation. Soil samples were taken at depths of 0–5 cm and 45–50 cm. Root length density (RLD) in five diameter classes was measured, soil organic carbon (SOC), hot water extractable carbon (HWEC), texture and Fe and Al oxides were also measured. We found that mean weight diameter after slow wetting (MWDSW) in the A-horizon, was significantly greater (0.94–1.01 mm) when A. codonocephala was present compared to plots without A. codonocephala (0.57–0.59 mm). SOC and RLD in the smallest diameter class (< 0.5 mm), were the variables which best explained variability in MWDSW. A significant positive linear relationship existed between MWDSW and soil cohesion but not with internal angle of friction. As herbaceous vegetation was more efficient than trees in improving aggregate stability, this result suggests that the mechanisms involved include modifications of the cohesive forces between soil particles adjacent to plant roots and located in the enriched in SOC rhizosphere, thus also affecting shear strength of the corresponding soil volume. Thus, vegetation stabilised soil under different hierarchical levels of aggregate organisation, i.e. intra- and inter-aggregate. Our results have implications for the efficacy of techniques used in land conversion programs dedicated to control of soil erosion and shallow landslides. We suggest that mixtures of different plant functional types would improve soil conservation on slopes, by reducing both surface water erosion and shallow substrate mass movement. Planting trees for cropping or logging, and removing understory vegetation is most likely detrimental to soil conservation.  相似文献

In this study we examined the effects of glucose-C on the activities of fungi and bacteria determined by the method of substrate-induced respiration (SIR) in combination with the selective inhibition technique, the immobilized-S and the arylsulphatase (ARS) activity in two calcareous arable and fallow soils. The amounts of glucose-C were added at six doses: 0, 125, 250, 500, 750 and 1000 mg kg− 1 soil to the soils and then incubated for one week with a Na235SO4 solution (518.9 kBq kg− 1 dry soil and 20 mg S kg− 1 dry soil) prior to analysis. At the highest dose of 1000 mg kg− 1 soil, fungal activity increased by 59.1% (of the dose 0) versus 45.5% for bacterial activity in the arable soil, while in the fallow soil the increases were more marked and corresponded to 69.9% and 71.1%, respectively. Largest increase in immobilized-S was observed in the arable soil (300.7%) compared with the fallow soil (153.1%). In contrast, the ARS activity increased by 16.4% in the arable soil versus 32.1% in the fallow soil. These results indicate that glucose proportionately affected more the intensities of immobilized-S than those of ARS. Strong positive correlation coefficients were found between fungal activities and immobilized-S in the arable soil (r = 0.96, P < 0.01) and in the fallow soil (r = 0.98, P < 0.001). However, non-significant correlations were observed between fungal activities and ARS in both studied soils. As to bacterial activities, positive significant correlation coefficients were found with immobilized-S in the arable soil (r = 0.95, P < 0.01) and in the fallow soil (r = 0.90, P < 0.05) as well as with ARS activities in the arable soil (r = 0.83, P < 0.05) and in the fallow soil (r = 0.97, P < 0.01). Overall, we also found positive and significant correlation coefficients of immobilized-S with ARS activities in the arable soil (r = 0.86, P < 0.05) and in the fallow soil (r = 0.83, P < 0.05). Accordingly, the results showed a presence of extracellular arylsulphatase activity of 38.7 mg p-nitrophenol kg− 1 soil h− 1 in the arable soil and of 63.5 mg p-nitrophenol kg− 1 soil h− 1 in the fallow soil. It was concluded that fallowing maintained larger activities of fungi, bacteria and arylsulphatase compared with the arable soil.  相似文献

Soil microarthropods are considered to be among the strongest determinants of plant litter decomposition in warm, humid sites. Little is known, however, about the regulation of plant litter decomposition dynamics along an elevation gradient in such sites. Our study examined the contributions of soil microarthropods to leaf litter decomposition of a single substrate (Castanopsis carlesii) along an elevation gradient across four types of zonal vegetations in southeastern China: evergreen broadleaf forest (EVB): coniferous forest (COF): dwarf forest (DWF): and alpine meadow (ALM) during April 2007 to March 2008. Leaf litter decomposition of C. carlesii was significantly accelerated by the presence of fauna in all four sites. After 360 days in the field, mass loss rates with the full decomposer assemblage and in the reduction of microarthropods were 62.9% and 41.2% in EVB, 48.1% and 30.6% in COF, 36.4% and 27.8% in DWF, 30.3% and 23.7% in ALM, respectively. The percentage of total decomposition due to the presence of soil fauna was 37% in EVB, 25% in COF, 12% in DWF, and 8% in ALM, thus showing strong systematic variation along the elevational gradient. The mass losses in control plots at the four sites were significantly correlated with the abundances of total Acari, Collembolans, and Mesostigmata mites. Although the proportion of Oribatid mites at EVB was not the highest among the four sites, there were elevated proportions of Mesostigmatid and Prostigmata mites, many of which were microbe-consuming species and induce an indirect influence on litter decomposition. Moreover, Shannon Index (F = 2.455, p = 0.093) and Group Number (F = 5.830, p = 0.005) both decreased along the elevation gradients. Mass losses were also found to be distinctively related to H′ (r2 = 0.984, p = 0.016), and GN (r2 = 0.952, p = 0.048) across the four sites. Our results suggest that the faunal contribution to plant litter decomposition varies markedly across environmental gradients that differ in litter faunal diversity.  相似文献

Differences in alpha and beta bat diversity among montane rainforest and five shaded coffee plantations under different management regimes, as well as some environmental factors and vegetation parameters influencing bat richness, were evaluated for the first time in southeastern Chiapas, Mexico. In each site, bats were captured every 2 months from March 2004 to July 2005, with six mist-nets, during two nights, using the capture-recapture method. We captured 2970 individuals of 43 bat species. Montane rainforest had the greatest alpha diversity (H′ = 2.681; n = 37), whereas alpha diversity was similar among coffee plantations (H′ = 2.229-2.364; n = 23-26). The number of frugivorous and nectarivorous species was similar among the sites; the greatest exchange in species composition (beta diversity) occurred for insectivorous bats, which reduce their number in coffee plantations as pesticides are incorporated. Bat richness species was significantly related to the number of vegetation strata, height, and cover of trees. We suggest that coffee plantations could act as corridors, facilitating connection among different elements of the landscape in the Sierra Madre de Chiapas for some frugivorous and nectarivorous bats.  相似文献

Inventories of peat volume and carbon storage often include general values for peat depth, but more spatially explicit and accurate estimates are required if carbon management strategies are to be developed at scales appropriate for the management. This article presents a methodology for estimating peat depth for large blanket peat areas using field sampling and GIS modelling to map peat depth on Dartmoor in south‐west England. The study area was divided into carbon unit areas (CUAs) based on soil and vegetation. Approximately 1000 peat depth measurements were taken, each consisting of a mean (n = 5) from depths within a 32 m2 area. Sampling points were stratified according to CUA area and proportional extent of slope and elevation classes. Regression analyses were used to determine the relationships between slope, elevation and peat depth within each CUA. The strongest relationship was for blanket peat (r2 = 0.53), with weaker ones for areas where peat was shallow and depth was less variable. A digital elevation model was used in a GIS to model peat depths for the whole of Dartmoor. Results were tested against a data set of 200 peat depths on a 250 m grid covering 1325 ha. We conclude that peat depth can be modelled using easily available topographic data combined with well‐designed field sampling over larger spatial scales. The approach can result in accurate mapping of peat depth and carbon storage for blanket peatlands in the United Kingdom and perhaps also elsewhere.  相似文献

The concepts of “genoform” and “phenoform” distinguish the genetically-defined soil series and the variation of soil properties resulted from different land uses and management practices. With the repeated field measurements over time, we attempted to understand the difference of soil hydraulic properties among different land uses for a given soil series, and their temporal dynamics. Four soil series (Glenelg, Hagerstown, Joanna, and Morrison) in Pennsylvania with contrasting textures, structures, and parent materials were investigated. Within each soil series, four common land uses (woodland, cropland, pasture, and urban) were examined. At each site of soil series–land use combination, field-saturated and near-saturated hydraulic conductivities, K(ψ), were measured at the soil surface using standard tension infiltrometers at water supply potentials (ψ) of − 0.12, − .06, − 0.03, − 0.02, − 0.01, and 0 m. Surface infiltration measurements were repeated at each site in May and October from 2004 to 2006. The analysis of variance indicated that the measurement time (May or October) had the greatest impact on all measured hydraulic conductivities (p < 0.001), followed by the land use (< 0.05 for Kψ = 0 and Kψ = − 0.06) and soil series (p < 0.06 for Kψ = − 0.01 to Kψ = − 0.03). The interactions between the time and land use and between the soil series and land use were statistically significant for Kψ = 0 and Kψ = − 0.01. When separated by the measurement time, land use showed greater impacts in October than in May, while soil series had greater impacts in May than in October. Among the four land uses, woodland showed less obvious temporal change compared to the other three land uses because of less human-induced impacts and more consistent ground cover. Other three land uses generally showed a higher hydraulic conductivity in May than in October due to the drier initial soil moisture condition and related management practices in the spring that gave rise to more significant macropore flow. The results suggested that the initial soil moisture is an important variable that drives the temporal variation of the surface soil hydraulic properties.  相似文献

N2-fixation by free-living (diazotrophic) microorganism is a key process affecting ecosystem functioning in soils. Understanding drivers affecting diazotrophic community assemblages and activities may lead to management practices to increase primary production and/or environmental sustainability. We used PCR-DGGE to determine the fundamental relationships between diazotrophic community structure and in a wide range of soils across southern Australia. In addition qPCR, RT-qPCR and N2-fixation (acetylene reduction) were used to investigate factors influencing gene abundance, expression and processes in similar soils with different agricultural inputs. Across 22 soils, the structural composition of the nifH community was significantly influenced by site (ANOSIM R = 0.876; P = 0.001). The effects of management practices were evident, and often larger than between-soil differences, but were only present at some sites. Differences in nifH communities between sites correlated to particulate organic carbon (POC; measured by mid-infrared spectroscopy) content of the soils (BIO-ENV test; ρ = 0.502; P = 0.001), but not other factors including total soil C. In 3 soils from the Murrumbidgee irrigation region of NSW, intensification of the farming systems was associated with increasing N2-fixation (P < 0.05), except where rice was cultivated. N2-fixation correlated either with nifH abundance or gene expression in soils, but not both. Our data shows that soil C is closely linked to diazotrophic ecology. Principally, the amount of C entering the soil system is directly related to the abundance and N2-fixation activity of free-living bacteria. However, we also show that C in the POC pool has associative links to the genetic diversity of the soil diazotroph community. Given the importance of diversity and abundance of functional organisms in supporting ecosystem processes, we suggest that soil C inputs should be considered for both qualitative and quantitative properties when considering impacts on diazotrophic bacterial ecology.  相似文献

At the global scale peatlands are an important soil organic carbon (SOC) pool. They sequester, store and emit carbon dioxide and methane and have a large carbon content per unit area. In Ireland, peatlands cover between 17% and 20% of the land area and contain a significant, but poorly quantified amount of SOC. Peatlands may function as a persistent sink for atmospheric CO2. In Ireland the detailed information that is required to calculate the peatland SOC pool, such as peat depth, area and carbon density, is inconsistent in quality and coverage. The objective of this research was to develop an improved method for estimating the depth of blanket peat from elevation, slope and disturbance data to allow more accurate estimations of the SOC pool for blanket peatlands. The model was formulated to predict peat depth at a resolution of 100 ha (1 km2). The model correctly captured the trend and accounted for 58 to 63% of the observed variation in peat depth in the Wicklow Mountains on the east coast of Ireland. Given that the surface of a blanket peatland masks unknown undulations at the mineral/peat interface this was a successful outcome. Using the peat depth model, it was estimated that blanket peatland in the Wicklow Mountains contained 2.30 Mt of carbon. This compares to the previously published values ranging from 0.45 Mt C to 2.18 Mt C.  相似文献

Ogasawara Islands are important ecosystems sustaining many indigenous spices. To clarify the indigenous soil environments of Ogasawara Islands, we studied the chemistry of the soils. Many surface soils were low in bio-available P (0 to 0.55 g P2O5 kg−1, average: 0.04 g P2O5 kg−1 as Bray II P, n = 22), but several soils were found to contain extremely large amounts of bio-available P (1.36 to 6.98 g P2O5 kg−1, average: 2.93 g P2O5 kg−1, n = 5). From soil profile analyses, the authors concluded that the extremely large amount of bio-available P could not be explained by the effects of parent materials with high P contents nor the effect of fertilizations by human activity, but the effects of natural seabird activities in the past could be the cause. The soil profiles with large amounts of bio-available P indicate deep migration of soil materials from A horizons, which could be a result of intensive mixing of upper horizons by seabird activities. The intensive mixing was supported by the low mechanical impedance of the horizons for the P-accumulating soils (8.17 ± 2.54 kg cm−2, n = 8) than those for the non-P-accumulating soils (17.46 ± 3.52 kg cm−2, n = 36). It is likely that in the past seabirds, such as shearwaters, made burrows in the soils for nesting and propagating and inadvertently transported a large amount of P from the sea to the soils, resulting in the extremely large amounts of bio-available P in the present soils.  相似文献

Forest soils contain the largest carbon stock of all terrestrial biomes and are probably the most important source of carbon dioxide (CO2) to atmosphere. Soil CO2 fluxes from 54 to 72-year-old monospecific stands in Rwanda were quantified from March 2006 to December 2007. The influences of soil temperature, soil water content, soil carbon (C) and nitrogen (N) stocks, soil pH, and stand characteristics on soil CO2 flux were investigated. The mean annual soil CO2 flux was highest under Eucalyptus saligna (3.92 μmol m−2 s−1) and lowest under Entandrophragma excelsum (3.13 μmol m−2 s−1). The seasonal variation in soil CO2 flux from all stands followed the same trend and was highest in rainy seasons and lowest in dry seasons. Soil CO2 flux was mainly correlated to soil water content (R2 = 0.36-0.77), stand age (R2 = 0.45), soil C stock (R2 = 0.33), basal area (R2 = 0.21), and soil temperature (R2 = 0.06-0.17). The results contribute to the understanding of factors that influence soil CO2 flux in monocultural plantations grown under the same microclimatic and soil conditions. The results can be used to construct models that predict soil CO2 emissions in the tropics.  相似文献

Assessment of diversity and understanding factors underlying species distribution are fundamental themes in ecology. However, the diversity of native arbuscular mycorrhizal fungi (AMF) species in African tropical agro-ecosystems remains weakly known. This research was carried out to assess the morphological diversity of indigenous AMF species associated with rhizosphere of cowpea (Vigna unguiculata (L.) Walp.) in different agro-ecological zones (AEZ) of Benin and to examine the effects of soil chemical properties, climatic factors and agricultural practices on this diversity. Results showed that, in Benin, cowpea was grown by farmers in very exhausted soils, where available phosphorus and potassium were deficient. The indigenous AMF spore density was on average 202 spores per 100 g dry soil and there was no difference in the density among the agro-ecological zones (P = 0.56). Fifteen AMF morphospecies belonging to eight genera (Gigaspora, Scutellospora, Racocetra, Acaulospora, Funneliformis, Rhizophagus, Glomus and Claroideoglomus) were detected. The computed species richness estimators indicated that a limited number of additional undetected morphospecies are probably present in cowpea fields. According to analysis of similarity (ANOSIM), the arbuscular mycorrhizas community composition of the Sudanian zone and Guinean zone were not significantly different (r = −0.01; P = 0.517). The diversity of AMF morphospecies in the fields was weak with prevalence of Glomeraceae (92%). Furthermore, AMF diversity and evenness indices were negatively correlated with annual rainfall (P < 0.01) and with available phosphorus (P < 0.05). However, no significant correlation was observed between AMF diversity indices and soil organic carbon. Ultimately, this study tends to confirm that soil management practices (tillage, soil disturbance) have negative effect on AMF diversity.  相似文献

Understanding the sensitivity of soil respiration to temperature change and its impacting factors is an important base for accurately evaluating the response of terrestrial carbon balance to future climatic change, and thus has received much recent attention. In this study, we synthesized 161 field measurement data from 52 published papers to quantify temperature sensitivity of soil respiration in different Chinese ecosystems and its relationship with climate factors, such as temperature and precipitation. The results show that the observed Q10 value (the factor by which respiration rates increase for a 10 °C increase in temperature) is strongly dependent on the soil temperature measurement depth. Generally, Q10 significantly increased with the depth (0 cm, 5 cm, and 10 cm) of soil temperature measuring point. Different ecosystem types also exhibit different Q10 values. In response to soil temperature at the depth of 5 cm, alpine meadow and tundra has the largest Q10 value with magnitude of 3.05 ± 1.06, while the Q10 value of evergreen broadleaf forests is approximately half that amount (Q10 = 1.81 ± 0.43). Spatial correlation analysis also shows that the Q10 value of forest ecosystems is significantly and negatively correlated with mean annual temperature (R = −0.51, P < 0.001) and mean annual precipitation (R = −0.5, P < 0.001). This result not only implies that the temperature sensitivity of soil respiration will decline under continued global warming, but also suggests that such acclimation of soil respiration to warming should be taken into account in forecasting future terrestrial carbon cycle and its feedback to climate system.  相似文献

Long-term diversity-disturbance responses of soil bacterial communities to copper were determined from field-soils (Spalding; South Australia) exposed to Cu in doses ranging from 0 through to 4012 mg Cu kg−1 soil. Nearly 6 years after application of Cu, the structure of the total bacterial community showed change over the Cu gradient (PCR-DGGE profiling). 16S rRNA clone libraries, generated from unexposed and exposed (1003 mg Cu added kg−1 soil) treatments, had significantly different taxa composition. In particular, Acidobacteria were abundant in unexposed soil but were nearly absent from the Cu-exposed sample (P<0.05), which was dominated by Firmicute bacteria (P<0.05). Analysis of community profiles of Acidobacteria, Bacillus, Pseudomonas and Sphingomonas showed significant changes in structural composition with increasing soil Cu. The diversity (Simpsons index) of the Acidobacteria community was more sensitive to increasing concentrations of CaCl-extractable soil Cu (CuExt) than other groups, with decline in diversity occurring at 0.13 CuExt mg kg−1 soil. In contrast, diversity in the Bacillus community increased until 10.4 CuExt mg kg−1 soil, showing that this group was 2 orders of magnitude more resistant to Cu than Acidobacteria. Sphingomonas was the most resistant to Cu; however, this group along with Pseudomonas represented only a small percentage of total soil bacteria. Changes in bacterial community structure, but not diversity, were concomitant with a decrease in catabolic function (BioLog). Reduction in function followed a dose-response pattern with CuExt levels (R2=0.86). The EC50 for functional loss was 0.21 CuExt mg kg−1 soil, which coincided with loss of Acidobacteria diversity. The microbial responses were confirmed as being due to Cu and not shifts in soil pH (from use of CuSO4) as parallel Zn-based field plots (ZnSO4) were dissimilar. Changes in the diversity of most bacterial groups with soil Cu followed a unimodal response - i.e. diversity initially increased with Cu addition until a critical value was reached, whereupon it sharply decreased. These responses are indicative of the intermediate-disturbance-hypothesis, a macroecological theory that has not been widely tested in environmental microbial ecosystems.  相似文献

Two Finnish agricultural soils (peat soil and loamy sand) were exposed to four freeze-thaw cycles (FTC), with a temperature change from −17.3±0.4 °C to +4.1±0.4 °C. Control cores from both soils were kept at constant temperature (+6.6±2.0 °C) without FTCs. Soil N2O and CO2 emissions were monitored during soil thawing, and the effects of FTCs on soil microbes were studied. N2O emissions were extremely low in peat soil, possibly due to low soil water content. Loamy sand had high N2O emission, with the highest emission after the second FTC. Soil freeze-thaw increased anaerobic respiration in both soil types during the first 3-4 FTCs, and this increase was higher in the peat soil. The microbial community structure and biomass analysed with lipid biomarkers (phospholipid fatty acids, 3- and 2- hydroxy fatty acids) were not affected by freezing-thawing cycles, nor was soil microbial biomass carbon (MIB-C). Molecular analysis of the microbial community structure with temperature gradient gel electrophoresis (TGGE) also showed no changes due the FTCs. These results show that freezing and thawing of boreal soils does not have a strong effect on microbial biomass or community structure.  相似文献
