将经过体外成熟培养(IVM),体外受精(IVF)后获得的牛早期胚胎,按其所处卵裂阶段的不同,划分为2细胞、3~4细胞和5细胞以上3个试验组,在体外条件十继续培养到发育囊胚和孵化囊胚阶段,以观察在胚胎早期,受精卵裂发育的快慢与其后胚胎所获得的继续发育能力的关系。结果显示,牛早期胚胎体囊胚发育率的高低,与其在体外培养(IVC)前卵裂发育的快慢成正比。在IVF后42~46h处于2细胞、3~4细胞和5细胞  相似文献

比较了体外受精后不同时间收集的牛囊胚期胚胎冷冻后的存活率。试验一收集受精后第6,7,8和9天出现的胚胎,分3步加入0.75mol/L甘油和0.5mol/L丙二醇,将胚胎装入0.25mL细管后,放入已降温至5℃的冷冻机内,立即以2℃/min的速率降温至-7℃,停留5min后植冰。植冰后5min,以0.3℃/min的速率降温至-25℃,取出胚胎,并立即投入液氮中保存。解冻时,先让细管在空气中暴露10s  相似文献

牛体外受精胚胎一步脱防冻剂冷冻方法的研究   总被引:7,自引:1,他引:7  
为了研究适合于牛体外受精胚胎的一步除防冻剂的冷冻方法,特进行两个试验。试验1分别用1.5M甘油+0.25M蔗糖、1.5M乙二醇和1.5M丙二醇作防冻剂冷冻体外受精后第7天,已发育至囊胚阶段的牛胚胎。使胚胎降温至-7℃后,植冰,然后以0.3℃/分的速率降温至-30℃,立即将胚胎投入液氮中冷冻保存。在37℃水中解冻胚胎后,使其直接在含15%胎犊血清(FCS)的磷酸缓冲液(PBS)中脱去防冻剂。经体外培养72h后,3组胚胎的孵化率分别为77.27%(102/132)、73.24%(104/142)、47.90%(57/119),第1、2组的孵化率极显著高于第3组(P<0.01),说明用1.5M丙二醇溶液冷冻的胚胎,在解冻后不宜直接在PBS中脱防冻剂。试验2比较用不同浓度(1.0M01.5M,2.0M)的乙二醇和不同投液氮温度(-25℃,-30℃,-35℃)冷冻胚胎的效果。结果表明乙二醇浓度和投液氮温度对胚胎孵化率均无显著影响(P>0.05),各组胚胎的孵化率变动于74.44%-85.48%之间。  相似文献

比较了3种不同浓度的甘油(1.4,1.0,0.7mol/L)和投液氮温度(-25℃、-30℃和-35℃)对牛体外受精胚胎冷冻效果的影响。结果发现,最终投液氮温度为-25℃时,胚胎冻后存活率以1.4mol/L甘油组为最高,同1.0mol/L甘油组相比,差异显著(78.5%vs 64.5%,P〈0.05);与0.7mol/L甘油组相比,差异极显著(78.5%vs53.5%,P〈0.01)。以1.0mol/L的甘油冷冻,-30℃投液氮的效果优于-25℃的,它们之间冻后胚胎的孵化率差异显著(77.1%vs 64.5%,P〈0.05)。用0.7mol/L甘油冷冻,投液氮温度以-30℃的为最好,冻后胚胎的孵化率(73.3%)极显著(P〈0.01)高于-25℃的(53.5%)。用1.4mol/L甘油冷冻,以-25℃投液氮为最好,冻后胚胎的孵化率(78.5%)显著(P〈0.05)高于-35℃的(65.7%)。结果表明,不同的甘油浓度和投液氮温度对牛体外受精胚胎的冷冻效果有显著的影响。  相似文献

我们对牛胚胎快速冷冻技术进行了研究。1992年8月一11月间在木兰县,共移植受体母牛99头,有46头受体母牛妊娠,妊娠率为46.5%,有29头受体牛产犊,产犊率为29.3%。通过研究得知,牛胚胎快速冷冻技术可以应用于生产。  相似文献

对胚胎因素影响奶牛体外受精胚胎培养效果进行了研究。结果发现:在胚胎因素中,胚胎类型(冻胚和鲜胚)与胚胎发育阶段对胚胎培养效果没有显著影响(P〉0.05);冷冻胚胎解冻后停留时间对胚胎培养效果影响显著(P〈0.01);冷冻胚胎解冻后胚胎等级对胚胎培养效果影响显著(P〈0.05),解冻后A级胚胎存活率(80%,128/160)、囊胚孵化率(58.75%,94/160)显著高于B级胚胎存活率(59.38%,95/160)和囊胚孵化率(37.5%,60/160)(P〈0.05);解冻水浴温度(20℃、30℃)对培养效果有极显著影响(P〈0.01)。  相似文献

将采自肉联厂屠宰车间的牛卵巢中的卵母细胞置于TCM-199+10%ECS(0d)+FSH(1万IU/L)+LH(5mg/L)中培养成熟,用含0.3%透明质酸酶的消化液将卵母细胞周围的卵丘细胞去掉,并以7.5mg/L的CB液处理后,用微吸管吸去透明带内的第一极体及极体下面的部分卵母细胞质,然后将经体外受精并发育至8~16细胞期胚胎的卵裂球注入去核卵母细胞的卵周隙中,再将移核胚置于电融合槽内进行融合(电场强度为1200V/cm,持续时间为80μs,1次脉冲刺激)。将融合的胚胎移入生长有单层贴壁颗粒细胞的发育培养液(TCM-199+10%牛血清)中培养,观察移核胚的融合率及发育率,部分去核卵母细胞经染色后观察去核率。结果表明:(1)移核胚的融合率为86.9%(53/61);(2)发育至2~8细胞期的胚胎占培养的移核胚胎总数的21.3%(10/47);(3)卵母细胞的去核率为68.2%(45/66)。  相似文献

对牛卵泡卵母细胞体外受精(IVF)168h的致密桑椹胚、囊胚用常规快速冷冻法、预冷和不预冷的超快速冷冻法进行了冷冻保存试验。结果表明:IVF囊胚采用含10%甘油的常规快速冷冻法、含2.1mol/L甘油和0.25mol/L蔗糖预冷5min的一步冷冻法及含25%甘油和25%乙二醇预冷5min的玻璃化冷冻法等3种方法进行冷冻保存,解冻后的继续发育率(68.0%,59.0%,65.7%)均无显著差异(P>0.05),可用快速、简便、预冷的一步冷冻法或玻璃化冷冻法替代常规快速冷冻法;IVF致密桑椹胚可用一步冷冻法(2.1mol/L甘油+0.25mol/L蔗糖)和玻璃化冷冻法(25%甘油+25%乙二醇或25%甘油+25%1,2-丙二醇)进行预冷的超快速冷冻保存;冻前预冷(5min)能显著提高IVF囊胚的冻后形态正常率和继续发育率(P<0.05);IVF囊胚冷冻—解冻后的继续发育率高于IVF致密桑椹胚。  相似文献

Gustafsson H., U. Jaakma and M. Shamsuddin: Viability of fresh and frozen-thawed biopsied bovine embryos. Acta vet. scand. 1994,35,217-222.– Bovine embryos were biopsied using a simplified splitting technique and frozen-thawed according to a standard method with glycerol as cryoprotectant. The viability of fresh and frozen-thawed biopsied and intact embryos were evaluated after in vitro culture, by means of fluorescence test or following transfer to recipients. The survival rates after in vitro culture of fresh intact and biopsied embryos and of frozen-thawed intact and zona free embryos were not significantly different (70%, 60%, 68% and 52%, respectively), but significantly reduced for bipsied frozen-thawed embryos (16%) (p≤0.05). The pregnancy results after transfer of biopsied frozen-thawed embryos were also significantly lower (8%) compared to fresh biopsied embryos (39%) (p≤0.05). Both intact and biopsied embryos fluoresced after incubation with diacetylfluorescin but with higher intensity for the intact embryos. It is suggested that the reduced survivability for the frozen-thawed biopsied embryos might be caused by combined effects of the loss of the zona pellucida and the reduction of cells as a result of the simplified biopsy technique. It is concluded that improved biopsy and/or freezing techniques must be used if biopsied embryos have to be frozen.  相似文献

为了观察脱防冻剂方法对牛体外受精冷冻胚胎在体内、外发育率的影响,应用常规冷冻法在0.75mol/L甘油+0.5mol/L丙二醇溶液中冷冻受精后7、8d的囊胚,解冻后在0.25、0.5mol/L蔗糖液中二步或在0.25mol/L蔗糖液中三步脱防冻剂的胚胎孵化率(分别为68.6%、62.2%、68.7%)与用PBS/FCS六步脱防冻剂的差异不显著(70.4%,P>0.05),但在0.5mol/L蔗糖液中三步脱防冻剂后,孵化率显著降低(47.6%,P<0.01)。用0.25mol/L或0.5mol/L蔗糖或海藻糖预先使胚胎脱水后冷冻,解冻后可使胚胎直接在PBS中一步脱掉防冻剂,胚胎孵化率(45.0%~49.5%)与在0.25mol/L蔗糖液中二步脱防冻剂的相似(51.5%,P>0.05)。移植试验表明,在0.25mol/L蔗糖液中二步脱防冻剂获得的妊娠率与六步脱防冻剂相似,与在PBS中一步脱防冻剂的亦无明显差异,但妊娠率均偏低(21.4%~37.5%)。  相似文献

选用不同品种母牛作为试验动物,试验在排除胚胎质量和胚胎移植人员技术水平对胚胎移植影响的前提下,分别对不同品种、不同管理方式、不同营养水平、不同外源激素对牛体外受精冷冻胚胎移植妊娠率的影响进行研究,同时通过测定内源激素水平来验证黄体触诊法在实际胚胎移植工作中的可行性。结果为:①不同品种受体母牛对体外受精冷冻胚胎移植妊娠率没有显著影响;②不同管理方式、营养水平以及注射亚硒酸钠维生素E对体外受精冷冻胚胎移植妊娠率没有显著差异,但都有所提高;③注射外源激素对体外受精冷冻胚胎移植妊娠率没有显著影响。  相似文献

探讨了人工合成培养液CR1aa和小鼠胎仔成纤维细胞对牛体外受精早期胚胎体外发育的影响。结果表明,牛体外受精卵在CR1aa液中的卵裂率达76.2%,8细胞胚的比率达44.8%。小鼠胎仔成纤维细胞能够显著促进牛体外受精的早期囊胚以上胚胎的发育。牛体外受精后第5、6天的早期胚胎分别与小鼠胎仔成纤维细胞共培养,在受精后第7天发育至囊胚以上的比率分别达19.8%和24.6%;受精后第8天,孵化的囊胚比例分别达5.2%和7.5%。实验表明,受精后第5、6天的牛体外受精早期胚胎与小鼠胎仔成纤维细胞共培养,可显著提高扩张囊胚和孵化囊胚数量。小鼠成纤维细胞对胚胎发育的支持作用取决于胚胎发育阶段  相似文献

Important differences exist between in vitro fertilized (IVF) and nuclear transfer (NT) bovine embryos. Studies have shown that although in vitro development is comparable, post-implantation survival is greatly reduced in NT embryos. In this study, we compare serum and bovine serum albumin (BSA) supplementation during oocyte maturation and embryo culture of IVF and NT embryos. In experiment 1, oocytes and embryos were randomly distributed into different treatment groups consisting of synthetic oviductal fluid (SOF) medium supplemented with either serum, fatty acid-free BSA (FAF) or fraction V BSA during maturation and/or culture to assess IVF embryo development. In experiment 2, oocytes were matured in SOF + serum or SOF + FAF and reconstructed embryos were cultured in SOF + FAF to assess NT embryo development. Among the IVF treatment groups, a greater number of blastocysts were observed in the steer serum (SER) group (IVM and IVC in SOF + serum) on day 6; however, no significant differences were seen in blastocyst development from day 8 onwards. Hatching frequencies on days 8 and 9 were significantly greater in groups with serum, with the exception of FAF (IVM and IVC in SOF + FAF) on day 9. For the NT treatment groups, the presence of serum during IVM resulted in a higher proportion of MII oocytes and increased blastocyst development and hatching rates were compared with supplementation of FAF. These results indicate that both serum and FAF provide comparable embryo development for IVF but not for NT bovine embryos.  相似文献

The present study compared the efficiency of transgenic (TG) cloned embryo production by somatic cell nuclear transfer (SCNT) with fetal-derived fibroblast cells (FFCs) which were transfected with pEGFP-N1 to in vitro-fertilized (IVF), parthenogenetic and SCNT counterparts by evaluating the rates of cleavage and blastocyst formation, apoptosis rate at different developmental stages, cell number, ploidy and gene expression in blastocysts. In SCNT and TG embryos, the rates of cleavage and blastocyst formation were significantly lower (p < 0.05) than those of IVF controls, but it did not differ between SCNT and TG embryos. In IVF control, 86.7% embryos displayed diploid chromosomal complements and the rates were significantly (p < 0.05) higher than those of SCNT and TG embryos. Most TG embryos (79%) with FFCs expressed the gene by both PCR and under fluorescence microscopy. The expression of apoptosis by TUNEL was first detected at six to eight cell stages in all embryos of IVF, SCNT and TG groups, but the expression rate at each developmental stages was significantly higher (p < 0.05) in SCNT and TG embryos than in IVF counterparts. The expression rate in inner cell mass (ICM) of TG embryos was significantly higher (p < 0.05) than in SCNT and IVF embryos. These results indicate that the high occurrence of apoptosis observed in SCNT and TG embryos compared with IVF counterparts might influence the developmental competence. Moreover, the SCNT embryos derived using non-transfected donor cells exhibited a lower apoptosis expression in ICM cells than in TG embryos derived using pEGP-N1-transfected donor cells suggesting a possible role of negative gene effect in TG embryos.  相似文献

为探讨褪黑素(MLT)对牛卵母细胞成熟、精子获能和胚胎发育的抗氧化效果。在体外培养液中添加10-9 M MLT,以评估牛卵母细胞成熟率及胚胎发育效果;同时利用目测法、低渗膨胀法、JC-1和考马斯亮兰染色法评估不同浓度MLT(0 M、10-3 M、10-4 M和10-5M)添加在牛精子获能液经上游法处理45min后对精子质量的影响。结果表明:与对照组相比,添加10-9 M MLT既可显著提高牛卵母细胞的成熟率(62.5%vs 77.8%)(P0.05),又可明显刺激受精后胚胎体外发育的卵裂率(53.2%vs 67.6%)(P0.05)和囊胚率(20.3%vs35.4%)。在精子获能液中添加10-4 M MLT上游处理45 min后,其精子活力(84.6%vs71.6%)、质膜完整率(88.8%vs 75.2%)、线粒体高膜电位百分率(83.0%vs 70.2%)和顶体完整率(87.0%vs 74.4%)均显著高于对照组(P0.05)。这些结果有助于优化IVF技术体系,利于体外胚胎生产效率提高。  相似文献

选用40头泌乳中期(169.8±8)DIM的中国荷斯坦奶牛随机分为4组,采用完全随机试验设计,研究日粮亚油酸和亚麻酸水平对乳脂CLA合成的影响。试验共设4个处理,分别为高亚油酸组(LA)、高亚麻酸组(LEA)、高亚油酸和亚麻酸组合组(HLALEA)和低亚油酸和亚麻酸组(LLALEA)。通过9周的试验结果表明,不同处理对干物质采食量、泌乳净能校正产奶量、乳蛋白及乳脂肪的含量和日产量均没有显著影响。增加日粮中LA或LEA的含量,能够显著降低乳脂C12∶0、C14∶0和C16∶0的含量(P<0.05),显著增加乳中硬脂酸(C18∶0,P<0.05)和18碳长链不饱和脂肪酸的含量(P<0.01),但对20碳以上不饱和脂肪酸的影响小(P>0.05)。随着日粮中亚油酸含量的增加,LLALEA、HLEA、HLALEA和HLA组乳脂TVA和c9t11CLA的含量呈线性增加。HLEA组、HLALEA和HLA组乳脂TVA的含量分别比LLALEA组增加1.56、3.05和4.71 g/100 g,而c9t11CLA分别比LLALEA组提高2.5、2.8和3.7倍。因此表明,日粮亚油酸对乳脂c9t11CLA合成的贡献效果高于亚麻酸,而亚油酸调控乳脂CLA合成机制主要以提供乳腺SCD合成c9t11CLA所需的底物(TVA)为主。  相似文献
