AbstractThis study evaluated the accuracy of an in vitro 2-step method combined with an in vitro gas production technique (2+IVGPT) for the estimation of dietary fiber (DF) digestibility coefficient and fermentability of feedstuffs in the large intestine of growing pigs. Digestibility coefficients of corn, wheat, soybean meal, and wheat bran, estimated by the in vitro methods, including the 2-step, 3-step, 2+IVGPT, and in vivo were compared. The fermentation characteristics of feedstuffs were also determined through the 2+IVGPT. The results showed that the in vivo DF digestibility coefficient of wheat bran was 0.39, which was similar to the result estimated by the 2+IVGPT (0.40). In regard to the fermentation characteristics, the lowest maximum rate of gas production (Rmax) was obtained for wheat bran (11.0 ml h?1). To conclude, in vivo total tract digestibility of DF in the fibrous feedstuff (e.g. wheat bran) could be accurately estimated by the 2+IVGPT. 相似文献

The effect of phytase and xylanase supplementation of a wheat-based pig diet on the ileal and total tract apparent digestibility of dietary components and minerals were studied in eight growing pigs fitted with a PVTC cannula in a randomized block design experiment. The diets (A and B) were similar in major ingredient composition and in nutrient content. In diet A, part of the limestone was replaced with di-calcium phosphate to increase the content of available phosphorus (P). Diet B was fed without or with supplementation with phytase (500 FTU/kg; diet BP), xylanase (4000 XU/kg; diet BX) and phytase + xylanase (500 FTU and 4000 XU/kg; diet BPX). There were no differences (P > 0.05) between diets in the ileal or total tract digestibility of organic matter (OM), NDF and crude protein (CP). The ileal and total tract digestibility for P and Ca differed (P < 0.05) between diets, while there were no treatment effects for Zn. The ileal and total tract digestibility for P and Ca was higher (P < 0.05) on diets BP and BPX than on the other diets. In conclusion, phytase improved the utilization of dietary P and Ca in a wheat-based diet, while xylanase had no additional benefits in terms of OM and CP digestibility or mineral utilization. Two experiments were conducted to investigate the effects of 1,25(OH)2D3 to stimulate Na+-dependent phosphate uptake in Caco-2 cells, and the effects of dietary vitamin D supplementation to vitamin D-deficient nursery pigs on Na+-dependent nutrient uptake and mRNA expression of NaPi-IIb cotransporter and calbindin D9k in the jejunum. In Exp. 1, 250,000 Caco-2 cells were seeded on Costar 12 mm Snapwell inserts with a 0.40 µm polycarbonate filter and a seeding density of 0.25 × 106 and studied at 15 d postconfluence. Cells were treated with 10 nM of either 1,25(OH)2D3 or vehicle for 48 h and then mounted in modified Ussing chambers for transepithelial measurements. In Exp. 2, pigs (n = 32) were removed from sows at 3 d of age, placed on a vitamin D-deficient milk replacer diet and housed in a room devoid of sunlight and UV light in the range of 280 to 300 nm. On day 28, serum 25(OH)D3 concentrations were measured to verify low vitamin D status. Pigs (BW 10.10 ± 0.38 kg) were then individually housed day 28 postweaning and allotted to 1 of 2 dietary treatments. Dietary treatments consisted of corn-soybean-based diets with vitamin D supplementations of 0 or 1,500 IU/kg diet for 12 d. Blood samples were taken from the brachiocephalic vein on the initial (day 0) and final day (day 10, 11, or 12) of the study for analysis of serum 25(OH)D3, P, and Ca. Pigs were euthanized and jejunal segments were harvested and used in modified Ussing chambers and for RNA isolation and subsequent quantitative RT-PCR analysis. In Exp. 1, treating Caco-2 cells with 10 nM 1,25(OH)2D3 resulted in a 52% increase (P < 0.005) in Na+-dependent phosphate uptake compared with cells treated with a vehicle. In Exp. 2, Na+-dependent phosphate and glucose transport did not differ (P > 0.10) among treatment groups. Additionally, NaPi-IIb and calbindin D9k mRNA expression were not different (P > 0.10) between treatment groups. No differences (P > 0.10) were detected in final serum P or 25(OH)D3 concentrations between treatments. However, serum Ca linearly increased (P < 0.05) as the concentration of supplemental vitamin D increased in the diet. Overall, while 1,25(OH)2D3 stimulated Na+-dependent phosphate uptake in Caco-2 cells, supplementing diets with 1,500 IU/kg vitamin D3 from cholecalciferol did not increase jejunal Na+-dependent phosphate uptake or NaPi-IIb mRNA expression over that of pigs fed diets with no supplemental cholecalciferol. 相似文献

The effect of soya bean meal (SBM) substitution in the diet with white lupine (WL) seeds on performances and some parameters of the digestive tract of pigs were studied. The digestibility experiment was performed on 30 pigs using difference method combined with marker method, where one group was offered basal diet and in diet of experimental groups 25% of the basal diet was substituted with SBM or WL meal. The growth experiment was conducted on 48 pigs of approximately 20 kg body weight. The animals from the control group were fed with diet containing SBM, whereas in the experimental groups 50% or 100% of SBM was substituted with WL seeds. At the end of the experiment, eight pigs from each group were euthanized, and digesta and tissues were sampled. The apparent total tract digestibility of crude protein and dry matter was higher (p < 0.05), but crude fibre and ether extract were lower (p < 0.05) in the WL seeds than in SBM. Substituting 50% of SBM in the control diet with WL seeds did not affect significantly pig performance. Total substitution of SBM with WL seeds impacted negatively pigs growth and feed intake (p < 0.05), but it did not affect the length of villi and crypt depth. It also significantly increased ileum digesta viscosity and ammonia concentration in the ileum and caecum digesta. In the ileal digesta of animals offered WL seeds, an increased (p < 0.05) total bacterial and Enterobacteriaceae numbers were found, whereas reduced number of yeast and mould was found in the caecal digesta. The substitution of 50% SBM with WL seeds in the growing pig diet did not affect pigs' results, but the total substitution significantly decreased the performance of pigs and affected intestinal ecology. 相似文献

为深入了解家兔消化道中CD4+T细胞的组织学定位以及在消化道不同解剖学部位的数量变化,本试验运用免疫组织化学方法,探讨了家兔胃(胃底腺区、幽门腺区)和小肠及大肠不同解剖学部位中的CD4+T细胞定位特点和数量变化规律。结果表明,在胃肠中,CD4+T细胞主要分布于固有层。胃底腺区阳性细胞数目要明显比幽门腺区少。小肠、大肠各段的阳性细胞分布数量未见明显差异,由十二指肠到回肠表现为小幅度递增,由盲肠到直肠表现为小幅度递减。胃肠中CD4+T细胞在各部位的数量随年龄增长表现出先增后减的变化趋势,老年组和幼年组CD4+T细胞数量在大肠中未见明显差异。结果提示,CD4+T细胞在青年家兔胃肠中的分布最为丰富。 相似文献

Hanwoo (Korean native) steers (274.8 ± 4.6 kg) with ruminal and duodenal cannulae were used in a 4 × 4 Latin square design experiment to examine the effects of dietary treatments on starch disappearance in the gastrointestinal tract. Dietary treatments consisted of concentrate that were based on ground corn with soybean meal (C‐SBM), ground corn with corn gluten meal (C‐CGM), ground barley with soybean meal (B‐SBM) and ground barley with corn gluten meal (B‐CGM). Although the intakes of starch and protein for steers fed experimental diets were different, it did not change ruminal pH and total volatile fatty acid concentrations. Average duodenal CP flow and quantity of CP apparently digested post‐ruminally was higher (P = 0.001) for CGM‐based diets than SBM‐based diets. There were increases in quantity (P < 0.001) and percentage (P < 0.001) of corn starch digested post‐ruminally compared to barley starch. Synchronized diets showed higher percentages (P = 0.03) of starch apparently digested post‐ruminally than asynchronization. Hanwoo steers fed a corn‐based diet with a large quantity of starch reaching the duodenum and fed C‐CGM supplying great amounts of protein to the small intestine may have contributed to increased post‐ruminal starch digestion. 相似文献

The effect of source of fiber on ileal and fecal amino acid digestibility and bacterial nitrogen excretion in growing pigs. 总被引:4,自引:0,他引:4
Studies were carried out with six barrows (initial BW 50 kg) to determine the effect of two fiber sources on the apparent ileal and fecal N and amino acid digestibilities and bacterial N excretion in feces. The pigs, fitted with a simple T-cannula at the distal ileum, were fed three cornstarch-based diets formulated to contain 16% CP from soybean meal according to a repeated 3 x 3 Latin square design. Diet 1 did not contain additional fiber. Diets 2 and 3 contained 10% powdered cellulose and 10% barley straw (ground through a 2-mm mesh screen), included at the expense of cornstarch, respectively. The pigs were fed 900 g at each meal twice daily, 0800 and 2000. Feces were collected for 48 h after a 10-d adaptation period. Thereafter, ileal digesta were collected for 24 h. Chromic oxide was used as a digestibility marker. With the exception of leucine, the inclusion of fiber did not affect (P greater than .05) the ileal digestibilities of the indispensable amino acids. The values for the average of the ileal digestibilities of the indispensable amino acids were 85.3, 82.9, and 83.2% for diets 1, 2, and 3, respectively. On the other hand, the inclusion of fiber decreased (P less than .05) the fecal digestibilities of all the indispensable amino acids. The values for the average of the fecal digestibilities of the indispensable amino acids were 91.7, 87.4, and 89.1% for diets 1, 2, and 3, respectively. These decreases resulted from an increase (P less than .05) in the excretion of bacterial N, which was measured using 2,6-diaminopimelic acid as a marker. In conclusion, as measured with the ileal analysis method, the inclusion of fiber had little effect on the digestible amino acid supply. 相似文献

Changes in the activity of enzymes involved in oxidative metabolism of glutamine, and in protein content, in the epithelial tissue along the gastrointestinal (GI) tract of growing pigs exposed to nivalenol (NIV) in the diet were investigated. The epithelial tissue was taken from the stomach, small intestine and colon of three groups of animals fed diets without NIV (control), with inclusion of 2.5 mg NIV/kg diet (low dose) and with inclusion of 5.0 mg NIV/kg diet (high dose). The activities of glutaminase, glutamate dehydrogenase, oxoglutarate dehydrogenase, isocitrate dehydrogenase and alanine aminotransferase were determined. In the control pigs the activities of oxoglutarate dehydrogenase and alanine aminotransferase were higher (P < 0.05) in the epithelium of the small intestine as compared with the stomach and colon, while there were no differences in the activities of glutaminase, glutamate dehydrogenase and isocitrate dehydrogenase. With increasing inclusion of NIV in the diet the activity of oxoglutarate dehydrogenase decreased (P < 0.05) in the epithelium of the small intestine and colon, and the activity of alanine aminotransferase tended (P = 0.07) to increase in the epithelium of the small intestine. The activities of glutaminase, glutamate dehydrogenase and isocitrate dehydrogenase remained unaffected by the inclusion of NIV in the diet. In the control pigs the protein content in the epithelium of the small intestine was higher (P < 0.05) than in the stomach and colon, while there were no effects of NIV inclusion in the diet on the protein content. It can be concluded from the present study that the epithelial tissue of the small intestine and colon of pigs exposed to a diet containing NIV will have a reduced enzymatic capacity to utilise alpha-ketoglutarate in the tricarboxylic acid cycle (TCA-cycle), suggesting an impaired energy supply to these organs. 相似文献

The gastrointestinal tract (GIT) of young mammals is submitted to aggressions early in life and GIT stress proteins are up-regulated in pigs following weaning. We hypothesized that transient food deprivation may contribute to these changes. Therefore, the effects of fasting and refeeding on GIT stress proteins in weaned pigs were investigated. A complete block experimental design with three groups of five pigs each was set up with the following treatments: A - food offered, B - fasted for 1.5 days, C - fasted for 1.5 days and then re-fed for 2.5 days. After slaughter, the GIT was removed, weighed and sampled. Intestinal villi and crypts were measured and alkaline phosphatase activity was determined. GIT tissue stress protein concentrations were measured by Western blotting. Fasting led to intestinal mucosa and villous-crypt atrophy (p < 0.01) and reduced mucosal alkaline phosphatase total activity in the proximal small intestine (p < 0.05). Heat shock proteins HSP 27 and HSP 90 (but not HSP 70) and neuronal NO synthase (nNOS) increased (p < 0.01) in the stomach, mid-intestine and proximal colon with fasting. Inducible NOS (iNOS) did so in the stomach (p < 0.001). Refeeding partially or totally restored GIT characteristics and stress protein concentrations, except for gastric HSP 90 and iNOS. Significant correlations (p < 0.05 to p < 0.0001) were found among stress proteins, between nNOS and digesta weight, between HSP 27 or HSP 90 and intestinal mucosa weight, and between intestinal or colonic HSP or nNOS and alkaline phosphatase. In conclusion, fasting and refeeding modulate GIT HSP proteins and nNOS in pigs following weaning. Changes in digesta and intestinal mucosa weights and alkaline phosphatase activity may be involved in the modulation of stress proteins along the GIT. 相似文献

Background
This study aimed to determine the effect of the inclusion of corn gluten feed (CGF) on the apparent and standardized ileal digestibility of protein and amino acids and the apparent ileal and total tract digestibility of energy in growing pigs. The study was performed using 16 barrows (weight, 45.3 ± 4.5 kg) that were fitted with a T cannula at the terminal ileum. There were four treatments: a corn-soybean diet without CGF and three corn-soybean diets containing increasing levels of CGF (65, 130, and 195 g/kg). Data were analyzed according to a randomized complete block design, four blocks with four pigs each (one pig per treatment). The trend of the response (linear or quadratic) was determined using orthogonal contrasts, and when a linear effect was determined, a linear equation was obtained.Results
The results showed that the inclusion up to 195 g/kg of CGF in the corn-soybean diet did not diminish the ileal digestibility (apparent and standardized) of protein and amino acids (P > 0.05), except that of phenylalanine, cystine, and proline. A linear decrease (P < 0.05) per gram of CGF added to the diet in the apparent and standardized ileal digestibility of phenylalanine (0.011 and 0.015 percentage units, respectively), cystine (0.048 and 0.043 percentage units, respectively), and proline (0.045 and 0.047 percentage units, respectively) was noted. Similarly, ileal digestibility of dry matter and energy were adversely affected (reduced by 0.028 and 0.025 percentage units, respectively, per gram of CGF increment in the diet). A significant (P < 0.05) linear reduction in total tract digestibility with increase in CGF amount in the diet was observed for energy (0.027 percentage units), dry matter (0.027 percentage units), crude protein (0.020 percentage units), and neutral detergent fiber (0.041 percentage units) per gram of CGF added to the diet.Conclusion
CGF did not affect the ileal digestibility of protein and most amino acids but reduced the ileal and total tract digestibility of energy. 相似文献15.

The objective was to determine the effects of fermentable carbohydrates on phosphorus (P) metabolism, the chemical composition of the faecal mixed bacterial mass (MBM) and the microbial activity in the large intestine (LI) of pigs. Eight barrows (mean BW 35.9 ± 0.9 kg), fitted with simple T‐cannulas at the terminal ileum, were either fed a low‐P corn–soybean meal‐based control diet or 75% of the control diet supplemented with 25% cellulose, starch or pectin according to a 4 × 3 Latin Square design. Both pectin and cellulose caused higher faecal than ileal P recoveries. Ileal volatile fatty acids (VFA) levels were more pronounced for the starch (p < 0.05) rather than the cellulose and pectin treatments, whereas pectin resulted in a higher faecal VFA concentration in comparison to starch and cellulose (p < 0.05). The differences in faecal VFA concentrations corresponded to the pH values obtained in faeces. The N content of MBM was higher (p < 0.05) when cellulose was supplemented. Pectin caused a decrease in the P content of the MBM compared to the control (p < 0.05). As a result, the N:P ratio was significantly higher for the pectin (N:P = 4.33) than for the control treatment (N:P = 2.63), while the Ca:P ratio remained constant for all treatments, suggesting changes in the accumulation of N, P and Ca in MBM, probably due to changes in the species composition and activity of the microflora. 相似文献

The main objective of the present investigation was to study the impact of fermentation of cereals on the degradation of polysaccharides and other macronutrients in the small intestine and total tract of growing pigs. Eight pigs (initial BW, 34.5 ± 0.9 kg) were used in a replicated 4 × 4 Latin square design. Pigs were cannulated and housed individually in metabolism pens during sample collection. The 4 cereal-based diets were nonfermented liquid barley (NFLB), nonfermented liquid wheat (NFLW), fermented liquid barley (FLB), and fermented liquid wheat (FLW). The fermented feeds were prepared by storing the dietary cereals (barley and wheat) and water [1:2.75 (wt/wt)] in a closed tank at 25 °C for 2 d, after which 50% of the volume was removed and replaced with an equal amount of fresh cereals and water after each afternoon meal. At the time of feeding (0730 and 1430 h), the remaining dietary ingredients were added. Water was added to the dry nonfermented feeds [1:1 (wt/wt)] immediately before feeding. The fermentation process reduced the amount of DM in both cereals (P<0.001), whereas the amount of DM was similar (P=0.626) between the fermented cereals. There was an interaction of cereal and treatment for ileal flow of DM (P=0.014), OM (P=0.013), and protein (P=0.006), which were less in pigs fed the NFLB than the FLB diets, but unchanged in pigs fed the NFLW and FLW diets. Conversely, the ileal flow of protein was similar (P=0.605) in pigs fed the barley diets (average, 47.5 ± 1.7 g/kg of DMI) and increased with the FLW diet compared with the NFLW diet (43 vs. 35 g/kg of DMI, respectively). Ileal fat and CH(2)O digestibilities were 7.6 (P=0.002) and 8.9% (P<0.001) greater, respectively, when pigs were fed wheat compared with the barley-based diets, and the ileal digestibility of CH(2)O was greater when pigs were fed the fermented than nonfermented diets (86.5 vs. 84.5%, respectively; P<0.001). Fermentation reduced (P<0.0001) the fecal excretion of DM, OM, and protein in pigs fed the barley diet, but not when fed the wheat-based diet (P=0.305). Fermentation had no effect (P=0.243) on the fecal digestibility of nonstarch polysaccharides in either of the cereals but their digestibility was 10.0% greater (P<0.001) in pigs fed wheat than the barley-based diets. In conclusion, fermentation of cereal before feeding altered the dietary composition and influenced flow and composition of polysaccharides and other macronutrients at the ileum and in feces to a larger extent for barley than wheat. 相似文献

The effect of oligofructose (FOS) or trans-galactooligosaccharides (TOS) inclusion at 10 g/kg diet for growing pigs, at the expense of cornstarch (CON), on growth performance, nutrient digestibilities, and faecal physicochemical parameters was determined. In addition, concentration of volatile fatty acids (VFA) and bacterial -glucosidase, β-glucosidase, β-galactosidase and β-glucuronidase activities in faeces were determined as markers of intestinal microflora metabolic activity. Twelve 35-day-old castrated pigs were allocated to CON, FOS and TOS dietary treatments for an experimental period of 4 weeks. Dietary treatments did not affect animal growth performance. FOS and TOS pigs tended to have a lower faecal output than CON pigs by approximately 6.0% and 1.7% respectively. FOS and TOS did not affect total tract apparent digestibility of dry matter, organic matter, fat, crude protein, crude fiber, ADF and cellulose. Digestibility of carbohydrate components such as nitrogen free extract and NDF was significantly (P ≤ 0.05) higher in TOS pigs compared to CON and FOS. Digestibility of hemicellulose in TOS pigs did not differ from FOS but was significantly higher from CON. The experimental period had a significant effect on most of the parameters determined. There were no significant (P ≤ 0.05) differences between treatments regarding total VFA concentration and faecal bacterial enzyme activities. Nevertheless, an increased saccharolytic activity in the gut of FOS and TOS pigs could be postulated by the trends for apparently higher VFA concentration and bacterial -glucosidase and β-galactosidase activities. 相似文献

However, road transportation usually causes a stress response in pigs. It is reported that homeopathic remedies supplementation could alleviate the stress response in pigs. This study investigated the effects of road transportation with or without homeopathic remedy (Convermax) supplementation on growth performance, nutrient digestibility, fecal microbiota, and serum cortisol and superoxide dismutase (SOD) concentrations in growing pigs. A total of 180 crossbred 49-d-old growing pigs [(Yorkshire × Landrace) × Duroc] with an initial body weight of 13.17 ± 0.02 kg were randomly allotted to 2 groups based on the initial body weight, containing 18 replicates with 5 pigs (mixed sex) in each. The pigs were fed dietary supplementation of a homeopathic remedy (Convermax) (0 or 200 mg/kg of feed, as-fed) for 35 d. On day 21, 45 pigs (70-d old; 25.25 ± 0.37 kg) were randomly selected from each group and assigned to either 2 hr of road transportation or no road transportation, resulting in a 2 × 2 factorial design. We found that road transportation led to an increase in the fecal coliform bacteria counts (P = 0.023) and serum cortisol concentration (P = 0.039) and a decrease in the serum SOD concentration (P < 0.001). However, supplementing homeopathic remedy (Convermax) to the diet of growing pigs increased gain to feed ratio (P = 0.042), apparent nitrogen digestibility (P = 0.019), and serum SOD concentration (P = 0.007), whereas decreased serum cortisol concentration (P = 0.022). In brief, road transportation induced stress response for growing pigs and increased harmful bacteria counts in their intestines. Dietary supplementation of homeopathic remedy (Convermax) alleviated stress response, improved apparent nitrogen digestibility, and increased gain to feed ratio. However, no significant interactive effects between road transportation with or without dietary homeopathic remedy (Convermax) levels were observed on the detected parameters in growing pigs. 相似文献

本试验研究饲料肽对仔猪后期的生长、氨基酸利用率和肠道消化酶活性的影响.选用体重近似的96头(38±2)日龄的杜长大仔猪,随机分为4个处理组,每个处理4个重复.4个处理分别为:含6%鱼粉的对照组(C)、6%鱼粉和2%血浆蛋白粉的日粮组(T1)以及分别加1%和2%饲料肽等蛋白取代喷雾干燥血浆蛋白粉的试验日粮组(T2和T3),试验21 d结束时进行屠宰试验.结果表明:血浆蛋白粉和饲料肽可显著提高仔猪的平均日增重和日粮必需氨基酸消化率(P<0.05),二者之间的生产性能和日粮必需氨基酸消化率差异不显著(P>0.05).日粮处理对仔猪肠道消化酶活性没有显著影响(P>0.05).这表明饲料肤部分取代喷雾干燥血浆蛋白粉对仔猪后期的生长、氨基酸消化率和消化酶活性无显著影响. 相似文献
