A study between August 1995 and December 1997 included 343 dairy cattle on 20 farms in the Dar es Salaam region and 2289 zebu cattle on 39 bomas in the Lugoba area (coast region). The aim was to establish the prevalence of bovine tuberculosis ( Mycobacterium bovis) and bovine brucellosis ( Brucella abortus). In the single intradermal tuberculin test (SIT), 0.9% (3/343) of the animals in Dar es Salaam tested positive and 1.2% (4/343) were doubtful. Positive reactors were found in 10% (2/20) of the farms. In the Lugoba area, 0.6% (14/2206) were positive and 6.8% (149/2206) doubtful, positive cases being found in 21% (8/39) of all bomas. In the slow agglutination test (SAT) for B. abortus, 14.1% (48/341) of the serum samples reacted positively in Dar es Salaam and 2.3% (8/341) were doubtful. Positive SAT reactors were identified on 25% (5/20) of the dairy cattle farms. In the Lugoba area, 12.3% (273/2221) proved to be positive SAT reactors and doubtful reactions were observed in 2.9% (64/2221). SAT-positive animals were detected on 87% (34/39) of all bomas. The prevalence in single herds in Dar es Salaam varied from 4.3% to 5.3% for the SIT and from 2.2% to 50% for the SAT. The prevalence in single herds in Lugoba area was between 1.1% and 2.9% for SIT and from 1.4% up to 62.1% for SAT. The two cattle populations differed significantly ( p<0.001) in the prevalence of both bovine tuberculosis and bovine brucellosis. Two cows that were positive reactors were slaughtered and subjected to post-mortem examination, and organ samples were bacteriologically cultured. The occurrence of Mycobacterium tuberculosis was confirmed by polymerase chain reaction (PCR) in both cows. 相似文献

Smallholder dairy herds around the Dar es Salaam region of Tanzania supply 86% of raw milk consumed by the city dwellers. Previous studies have indicated that clinical mastitis is an important disease affecting smallholder dairy cows and an 18-month questionnaire-based longitudinal field-study was conducted between July 2003 and March 2005 to elucidate risk factors associated with the incidence. A total of 6057 quarter-level observations from 317 lactating cows on 87 randomly selected smallholder dairy herds were analysed at the quarter and cow level using logistic and Poisson regression models, respectively. At the quarter level, the average incidence rate of clinical mastitis was 38.4 cases per 100 quarter-years at risk whereas at the cow level the incidence rate was 43.3 cases per 100 cow-years at risk. The incidence was significantly (P< or =0.001) associated with cow factors (body condition score, parity, stage of lactation, and udder consistency), housing (floor type) conditions and milking (cow and udder preparation) practices. It was concluded that the extrapolation of the classic ten-point mastitis control plan into smallholder dairy herds should be undertaken cautiously. An integrated approach to dairy extension should focus more on the creation of mastitis awareness among smallholder producers and on the improvement of animal nutrition and reproduction indices-factors that may also have a direct impact on milk yield. 相似文献

A cross-sectional study was conducted to determine three parameters of the quality of the raw milk marketed by milk selling
points (MSPs) in Dar es Salaam region. Total bacterial count (TBC) was used as an indicator of the microbial quality of the
milk; antimicrobial residues were determined; and the California mastitis test (CMT) was used to screen for milk somatic cells
as an indication of the mastitis level in the cows that provided the milk. Moreover, a water sample at each MSP was taken
for bacteriological culturing. Finally, a questionnaire survey was conducted with the milk sellers at the MSPs to identify
risk factors for poor milk hygiene. A total of 128 milk samples and corresponding water samples were collected from randomly
selected milk selling points in Dar es Salaam region. The mean TBC was (8.2± 1.9) × 10 6 cfu/ml, and major bacterial isolates from the milk samples were Escherichia coli (6.3%), Bacillus cereus (6.3%), Staphylococcus aureus (6.3%) and Streptococcus agalactiae (6.3%), Enterobacter aerogenes (5.6%) and Enterococcus faecalis (4.7%). In most cases, the organisms identified in milk corresponded to those isolated from the corresponding water samples.
Of milk samples, 79.0% were positive to the CMT and 7.0% were positive for antimicrobial residues. TBC was normalized by log-transformation,
and the possible predictors of TBC were identified by fitting two linear regression models. In a random effect model, water
microbial quality, frequency of cleaning the milk containers, frequency of milk supply, milk storage time and the type of
containers, and mixing of fresh and previous milk were significantly ( p < 0.05) associated with the mean log TBC. In a fixed effect model, in addition to these indicators, water shortage, water
source and the refrigerator condition were significantly ( p ≤ 0.01) associated with log TBC. It was concluded that the milk sold in Dar es Salaam region is of poor quality and is of

A 31-year record-based retrospective study was carried out to determine the aetiology and temporal distribution of bovine clinical mastitis in smallholder dairy herds in the Dar es Salaam region of Tanzania over the period November 1971-December 2002. Laboratory information on 1964 quarter samples from 1365 cows in 281 smallholder dairy herds were retrieved, compiled and studied. Eighty-eight percent of the quarter samples were culture-positive and the predominant mastitis pathogens isolated were Staphylococcus aureus (25.7%), Streptococcus agalactiae (15.4%), Klebsiella pneumoniae (14.3%) and Escherichia coli (14.1%). Other isolates included Pseudomonas aeruginosa (7.5%), Streptococcus dysgalactiae (5.2%) and Streptococcus uberis (4.2%). Contagious mastitis pathogens were isolated from 45.6% of the culture-positive samples, whereas environmental and miscellaneous pathogens were isolated from 48.2% and 5.7%, respectively. Thirty percent of the miscellaneous mastitis pathogens were Candida species. The results demonstrate a steady increase in clinical Candida albicans mastitis. The prevalence of Candida albicans has increased from 1% in 1971 to 17.0% in November 2002. Conversely, despite some fluctuations, the prevalence of Staphylococcus aureus, Streptococcus agalactiae, E. coli and K. pneumoniae remain above 10%. Evaluation trials of the efficacy of buparvaquone (BUTA-kel KELA Laboratoria, N.V. Belgium), as a treatment of field cases of Theileria parva infection (East Coast fever - ECF) were carried out on 63 cattle in the peri-urban of Dar Es Salaam city, Tanzania, during the period November 2004 to August 2005. Thirty-two cattle (56%) received single-dose treatment (2.5 mg buparvaquone per kg body weight), while two and three-dose treatment with interval(s) of 48 h was given to 33% and 11% of total treated cattle, respectively; 38 cattle (60.3%) were treated at an early stage of the disease, while 25 cattle (39.7%) were treated at an advanced stage of the disease. The rectal body temperature of 90.5% of buparvaquone-treated cattle dropped to normal values (37.5-39.5 degrees C) by day 7 of treatment, and by day 15 of treatment 96.8% of treated cattle showed normal values. Pulmonary signs were observed in 8/68 (11.8%) of total ECF diagnosed cattle and were successfully treated, albeit with parvaquone plus frusemide (Fruvexon); were not included in final evaluation of the efficacy of BUTA-kel. The present evaluation trials record a recovery rate of 95.2%. Buparvaquone (BUTA-kel KELA Laboratoria, N.V. Belgium), therefore, records another efficacious and valuable alternative treatment against East Coast fever in Tanzania. 相似文献

1. A cross-sectional study was conducted from September 2015 to July 2016 in South Tyrol, Northern Italy to examine the prevalence of gastrointestinal helminths in free-range laying hens under mountain farming production conditions. 2. A total of 280 laying hens from 14 free-range mountain farms (4 organic, 10 conventional) were randomly collected at the end of the laying period. Faecal samples were taken to analyse faecal egg counts (FEC) and faecal oocyst counts (FOC). The gastrointestinal tracts were removed post mortem and examined for the presence of helminths. 3. In faeces, FEC values averaged 258 eggs per g of faeces, which were dominated by Ascaridia galli and Heterakis gallinarum. Mean FOC was 80 oocysts/g. In the gastrointestinal tract, at least one nematode species was found in 99.3% of the examined hens. H. gallinarum was the most prevalent nematode (95.7%), followed by Capillaria spp. (66.8%) and A. galli (63.6%). Thirty per cent of the chickens were infected with cestodes (tapeworms). Correlation coefficients between worm counts of H. gallinarum, Capillaria spp. and A. galli ranged from 0.41 to 0.51. 5. The helminth prevalence did not differ between conventional and organic farms, whereas total worm burden was higher in organic compared with conventional farms (318.9 vs. 112.0). Prevalence and infection intensity did not differ between farms that used anthelmintic treatments and those that did not. 6. In conclusion, free-range laying hens under the studied mountain farming conditions are at high risk of nematode infection, especially in organic systems. Screening of subclinical mastitis under field conditions is done using the California mastitis test (CMT). CMT score of ≥1 corresponding to ≥500,000 somatic cells ml −1 is commonly used as threshold of subclinical mastitis in temperate countries. However, given the innately high physiological level of somatic cells in low yielding dairy cows, this threshold may not apply to low yielding dairy cows. The current study was undertaken to investigate the clinical utility of CMT for screening of Staphylococcus aureus subclinical mastitis in low yielding smallholder dairy cows in Tanzania. A total of 1151 of quarter-milk samples were CMT tested, of these 914-originated from cows with a lactation period of 14–305 days. All samples were screened for subclinical mastitis by the CMT as well as microbiological culture of single, duplicate (two consecutive) and triplicate (three consecutive) samples as a gold standard. For the duplicate and triplicate quarter-samples, cows were considered positive for S. aureus subclinical mastitis if results of microbiologic culture for S. aureus were positive for two of two, and for at least two of the first three consecutive quarter-milk samples collected from that cow, respectively. Using a CMT score of ≥1 would classify 78.6% of the 940 quarter-samples as positive. Eighty-two percent of the samples in which S. aureus was isolated had CMT scores ≥2; this would classify 51.6% of the 940 quarter-samples as positive. For the single sample, this cut-off had sensitivity, specificity and likelihood ratio for S. aureus of 0.87, 0.83 and 4.24, respectively. For the duplicate quarter-milk samples this cut-off had sensitivity, specificity, and likelihood ratio of 0.94, 0.86, and 5.19. While, for the triplicate quarter-milk samples this cut-off had sensitivity, specificity and likelihood ratio of 0.97, 0.92 and 7.47, respectively. Based on these results and practical considerations, it is concluded that CMT score of ≥2 corresponding to ≥800,000 somatic cells Ml −1 is the best cut-off to correctly identify S. aureus intramammary infections in low yielding dairy cows in Tanzania. 相似文献

Prevalence of thermophilic Campylobacter infections in humans, chickens and crows was determined in a cross-sectional study that was carried out in urban and rural areas of Morogoro region, Tanzania during the period of January 2003 to December 2004. A total of 632 human stool samples, 536 cloacal swabs from local and broiler chickens and 22 intestinal contents from crows were screened for presence of thermophilic campylobacters using Skirrow's protocol. Representative Campylobacter jejuni isolates from human and chicken samples were also analysed by polymerase chain reaction (PCR) as a definitive identification method. The overall prevalence of thermophilic campylobacters was 9.3% (95% CI: 7.2-11.9), 69.8% (95% CI: 65.7-73.6) and 72.7% (95% CI: 49.8-89.3) in humans, chickens and crows respectively. In humans, 59 thermophilic campylobacters were isolated of which 96.6% were C. jejuni and 3.4%Campylobacter coli. There was a significantly (P<0.001) higher prevalence in young individuals (16%) than in adults (7%). Of 341 isolates from chickens, 91.2% were C. jejuni and 8.8% were C. coli. A significantly (P<0.05) higher infection rate was observed in rural local chicken (76%) than in broilers (60%). In crows, of 16 isolates, 93.8% were C. jejuni and 6.2% were C. coli. Definitive identification of C. jejuni by PCR revealed positive results in 74.1% of 243 analysed isolates. Findings in this study indicate high prevalence of thermophilic campylobacters in humans, chickens and crows in Morogoro, and a higher infection rate of C. jejuni than that of C. coli in different animal species. Age of humans and location of chickens were identified as risk factors for thermophilic Campylobacter infections. Positive isolates to biochemical tests that indicated negative results on PCR indicates the additional value of PCR for definitive diagnosis of C. jejuni. 相似文献

Reptiles are becoming popular pets in many parts of the world. They are also known to harbor numerous gastrointestinal parasites. We used faecal smears to examine 748 stool samples from 14 different agamid lizard species. In addition, we used coproantigen ELISA tests (11 samples) and immunofluorescence assays (IFA) (19 samples) to detect reptile Cryptosporidium infections. In 28 cases, veterinarians requested therapy to treat oxyurid- and/or Isospora amphiboluri-infections and resent fecal samples after proposed therapy and anti-parasitic treatments had been applied. We also performed complete dissections of 24 deceased agamas in order to specify protozoan and helminth parasite infections.Overall, the examined fecal samples contained 6 different taxa. Oxyurids (Pharyngodonidae) were the most prevalent nematodes (41.2%), followed by I. amphiboluri (17.0%), Entamoeba spp. (0.8%), Choleoeimeria spp. (0.5%), Trichomonas spp. (0.3%), Cryptosporidium spp. (0.3%) and Strongyloides-like nematodes (0.1%) . I. amphiboluri infections were significantly more prevalent (Chi-square test: χ2 = 21,5, df = 1, P < 0.001) in juvenile agamid lizards (31.9%) than in adults (14.2%). One of 11 (9.1%) coproantigen ELISA-examined samples was positive for Cryptosporidium. In 10.5% of the samples we found oocysts of Cryptosporidium. Thirteen (54.2%) of necropsied agamid lizards were infected with endoparasites and it is likely that three (12.5%) of them died due to severe parasitic infections. 74.0% of the samples that were submitted after therapy had been applied were negative. The high prevalences and pathological findings of several clinical parasitoses observed in these exotic reptiles calls for more detailed investigations on agamid gastrointestinal parasite fauna. 相似文献

Associations between serum concentrations of haptoglobin, pathological lung lesions indicative of Mycoplasma hyopneumoniae (EP) or Actinobacillus pleuropneumoniae (PL) infection at slaughter and previous rearing environment were investigated in 510 pigs (90-100 kg live weight) from 17 farms in England. Haptoglobin concentrations were significantly higher in pigs showing pathological signs of EP infection compared to those without signs of this disease (EP positive median 0.43 mg ml(-1) vs. EP negative median 0.26 mg ml(-1), p<0.01). However, there were no significant associations between serum haptoglobin concentrations and pathological signs of PL. The presence of solid partitions compared with barred or similar open partitions was associated with a decrease of 0.44 mg ml(-1) farm mean haptoglobin concentration, whilst an increase in pen size of 10 m(2) was associated with a decrease of 0.08 mg ml(-1) farm mean haptoglobin concentration. The findings indicate that pathological signs of EP were associated with increased serum haptoglobin at slaughter, which in turn was influenced by components of the farm environment. 相似文献

To date, the public health relevance of mycobacterial infections in pigs is not well investigated despite high risk of infection.
Recently, there has been a documented increase in opportunistic infections and risk of acquiring opportunistic mycobacterial
infections in HIV/AIDS patients in Mubende district; unfortunately, there has been no published information on the epidemiology
of mycobacterial infections in this area. This study was carried out between September 2008 and February 2009. Investigations
were done to assess the prevalence and associated risk factors of mycobacterial infections in slaughtered pigs in Mubende
district of Uganda. A total of 997 pigs (53.7% male and 46.3% female) from 31 different slaughterhouses were examined for
the presence of lesions compatible with TB and mycobacterial infections. Pathologic tissue specimens were collected for culturing
and isolation of mycobacteria. A cross-sectional technique was used based on convenient visits to slaughterhouses but random
selection of individual slaughtered pigs for a detailed post-mortem inspection on a daily basis. The results reflected a 9.3%
and 3.1% (95% CI) prevalence of Mycobacterium species based on necropsy examinations and culture isolation, respectively. The highest prevalence of mycobacterial infection
was recorded in Buwekula County (the mixed agro-zone) whilst the lowest was in Kassanda County (pastoral zone). A multivariable
logistical regression analysis identified age ( P ≤ 0.001) and sex ( P ≤ 0.05) as risk factors for mycobacterial infections in pigs. Post-estimation statistics of the regression model evaluation
and validation fit it well into the data (HL, χ
2 = 5.9; P = 0.69 for necropsy, HL χ
2 = 2.9; P = 0.94 for culturing). This study documented a high prevalence of mycobacterial infections in slaughter pigs in Mubende district.
The fact that pigs and human often share common housing and environment poses a high risk of zoonotic transmission. This then
warrants further molecular investigation to identify the specific Mycobacterium species and their public health importance in this area. 相似文献

Occurrences of thermophlic Campylobacter in pigs and pig carcasses was investigated in a cross-sectional study that was carried out in three selected slaughter slabs
in Morogoro municipality, Tanzania. Before sampling, the slab hygiene, slaughter, carcass dressing, and meat handling was
assessed. Fecal samples were collected from 66 slaughter pigs at the kill floor. After slaughter, a 100-cm 2 area on medial surface of the thigh muscles of dressed carcasses was sampled using sterile cotton swabs. Thereafter, the
jejunal, cecal, and colon contents were also sampled. The samples were subjected to standard bacteriological examination using
Skirrows protocol. In all slaughter slabs visited, it was found that pig slaughter, dressing, and meat handling was done on
the ground under unhygienic condition. All the slaughter slab environment were dirty and had neither tap water or drainage
systems. Thermophilic Campylobacter prevalence in slaughtered pig was 66.7% while contamination rate of dressed carcasses was 10.6%. Of the Campylobacter-positive carcasses, five (12.2%) were from the animals which were also positive to Campylobacter. The isolation rate of Campylobacter in the cecum was higher (34.8%) compared to the small intestines (28.8%) and colon (16.7%) although the difference was not
statistically significant ( P > 0.05). Campylobacter jejuni was the most prevalent species as it constituted 74% of all isolates, while Campylobacter coli was isolated at 26%. This suggests possible risks of infection to people through consumption of contaminated pork or through
contact with infected pigs. Cecum was found to be the major part of intestine highly colonized by Campylobacter. 相似文献

Epidemiological aspects of Mycoplasma hyopneumoniae (Mh), influenza H1N1 and H3N2 viruses, and Aujeszky's disease virus (ADV) were investigated in slaughter pigs from 50 fattening pig herds. Herd factors as potential risk indicators for respiratory disease were obtained by means of a questionnaire. At slaughter, blood samples were collected from each herd, and the proportion of seropositive pigs per herd was assessed for each of these pathogens. The median herd-level seroprevalence of the agents were: Mh 88%, H1N1 100%, H3N2 60% and ADV 90%. The percentage of herds in which all investigated fattening pigs were seronegative for these agents was: Mh 0%, H1N1 0%, H3N2 12% and ADV 18%. The percentage of herds in which all investigated fattening pigs were seropositive for these agents was: Mh 8%, H1N1 71%, H3N2 22% and ADV 40%. A positive association was found between influenza H1N1 and H3N2 viruses, and a negative association between influenza H3N2 virus and ADV. There were no risk indicators for the seroprevalence of Mh. Three risk indicators were associated with the seroprevalence of influenza H1N1 virus: a fully slatted floor, an increasing number of pigs in the municipality and dry feeding. Three risk indicators were found for the seroprevalence of influenza H3N2 virus: purchase of pigs from > or = two herds, an increasing number of pigs in the municipality and natural ventilation. To investigate the transition in concentration of porcine reproductive and respiratory syndrome virus (PRRSV) and porcine circovirus type 2 (PCV2) and antibody for these viruses in serum, serum samples were collected from 29 pigs on weaning day and at 7, 14, 21, 28, 53, 84, and 120 days after weaning. The concentration of circulated PRRSV and PCV2 in serum was measured by real-time RT-PCR and real-time PCR, respectively. The specific antibody for PRRSV and PCV2 was measured using ELISA. PRRSV was not detected on 0 days post-weaning (dpw). The specific antibody for PRRSV began to increase as the concentration of PRRSV in serum increased, and the level of PRRSV then tended to decrease. PCV2 was detected in 12 of 28 pigs on 0 dpw. The concentration of PCV2 and the specific antibody for PCV2 showed a similar tendency to those of PRRSV. The correlation analysis suggests that a decline in the daily weight gain coincided with an increase in the PRRSV concentration. Pigs with a higher antibody titer against PRRSV or PCV2 on 0 dpw showed the lower level of PRRSV or PCV2, respectively. 相似文献

A study of neonatal steatorrhoea in unweaned piglets was carried out in randomly visited herds. Isospora suis and rotavirus were shown to be incriminated in this type of enteric disease and these agents may be considered important enteropathogens for 1-3-week-old piglets in Sweden. More extensive studies of I. suis were undertaken in one large herd. It was found that steatorrhoea may appear in 4-day-old piglets but oocysts were not detected until some days latter. Piglets that were shedding I. suis were not necessarily steatorrhoeic but oocysts were more frequently found in steatorrhoeic faeces than in faeces of normal consistency. The earliest oocyst output was detected at 5 days of age, the average litterage at first appearance being 12.8 +/- 4.0 days. The occurrence of I. suis oocysts in piglets did not correlate with the oocyst output in sows, which almost exclusively included genus Eimeria. Repeated parasitological and virological examination of steatorrhoeic faecal samples from 5-10 litters per herd at an age of 1-3 weeks would be helpful for making a herd diagnosis. 相似文献

In this study, the degree of contractile and metabolic development of myofibers in porcine LM, rectus femoris (RF), and dark and light portions of the semitendinosus (STD and STL, respectively) was determined, and their impact on meat quality was compared at the same age but different BW (trial 1) or at a given BW but different age (trial 2) in 48 Swiss Large White barrows from 12 litters after the growing and finishing period. The barrows had ad libitum (A) or restricted (R, 80% of A) feed access. In trial 1, at 113 and 154 d of age, 6 barrows in treatment A (62.1 and 99.5 kg of BW, respectively) and 6 siblings in treatment R (51.0 and 86.6 kg of BW, respectively) were slaughtered. In trial 2, a similar protocol was used except that the barrows were slaughtered at 61.3 (104 or 119 d of age, respectively) or 101.3 kg of BW (145 or 167 d of age, respectively). Muscle fibers were stained and classified as slow oxidative (SO), fast oxidative-glycolytic (FOG), or fast glycolytic (FG), and fiber area and distribution were determined. At 113 and 154 d of age, R barrows had smaller (P < or = 0.04) SO fibers in the LM, STD, and STL, smaller (P < 0.01) FOG fibers in the STL, smaller (P = 0.03) FG fibers in the LM, and smaller (P < or = 0.04) overall mean area in the LM, STD, and STL. In the STL and RF, R barrows had fewer (P < or = 0.06) FG and more (P < or = 0.08) FOG fibers than A barrows at 113 and 154 d of age. Except for smaller FOG fibers in the STD of R compared with A barrows slaughtered at the same BW, the myofiber size did not differ (P > or = 0.11). However, the LM tended to have fewer (P = 0.06) SO and more (P < 0.01) FG fibers, and the STD had more (P < 0.01) FOG fibers in R barrows. Regardless of whether R barrows were slaughtered at the same age or the same BW as the A barrows, shear force values and cooking losses were greater (P < or = 0.08) in the STD and STL of R barrows. These findings revealed that myofiber hypertrophy was impaired by feed restriction in barrows compared at the same age, but differences in myofiber size vanished at the same BW. By contrast, restricted nutrient supply affected myofiber maturation depending on the age and BW, but the impact differed between muscles. The absence of changes in myofiber type distribution among the younger-lighter and older-heavier barrows indicated that myofiber maturation was already completed in the younger-lighter barrows.
