Steroid levels and ovarian follicular morphology were examined in sows on days 19 and 26 (day 5 of next cycle) after injection of adrenocorticotropic hormone (ACTH) or dexamethasone (DXM). Five sows received DXM (30 micrograms/kg bodyweight, intramuscularly) at 12 h intervals from days 9 to 14. Another five sows were given ACTH (2 IU/kg bodyweight, intramuscularly) from day 17 to day 19 or the end of estrus. Five control sows received no treatment. Ovulation occurred only in control sows and progesterone was significantly elevated at day 26. Estradiol values in ovarian vein blood were low but variable on day 19 in DXM- and ACTH-treated animals. Androstenedione values were lower (p less than 0.05) on both days in sows receiving DXM but not in those given ACTH compared to control values on day 19. Morphometric analysis, based on six follicles in each of three sows from each treatment group, indicated that follicular and antral diameters and granulosa cell numbers did not differ for either hormone treatment group on either day compared to those of control sows on day 19. The mitotic index suggested that cell replication continued. However, pyknotic and karyorrhectic nuclei were also seen in the hormone treatment groups. Follicles and oocytes from both DXM- and ACTH-treated sows showed signs of early degenerative changes including disorganization of cumulus cells and large lipid droplets in the cytoplasm of oocytes. Significant differences from control follicles in granulosa cell density and theca interna cell density suggested an association with the altered steroid hormone secretion.  相似文献

Sows housed in freely moving groups have elevated cortisol levels until the rank order is established, which takes place within approximately 48 h. The aim of this investigation was to study the effect of repeated administration of synthetic adrenocorticotropic hormone (ACTH; Synacthen Depot), during the follicular phase (pro-oestrus) on oestrus, ovulation and endocrine parameters. Four multiparous sows were used. Follicular growth and ovulation were recorded by ultrasonography. The first oestrous cycle after weaning was used as control cycle. Onset of oestrus in the sow occurs 3-4 days after the time when plasma progesterone reaches a concentration of 8 nmol/l. The progesterone profile in the control cycle of the individual sow was used for estimation when the ACTH injections should start. In the third pro-oestrus ACTH (2.5 microg/kg) was given via an indwelling catheter every 2 h for 48 h. The sows were euthanased 4-6 days after onset of the third oestrus and the ovaries were examined. Cortisol levels were elevated during the treatment period (p < 0.05). The second cycle, in which the sows were injected with ACTH, was prolonged with 2.5 days compared with the control cycle (p < 0.05). The oestradiol pattern during oestrus was similar in the control and the treatment cycle in ovulating sows. Three sows had ovulated (fresh corpora lutea), but the ovaries contained additionally one or several luteinized follicles/cysts. In conclusion, ACTH administration during pro-oestrus caused a prolongation of the oestrous cycle and a disturbed follicular development.  相似文献

In 7 instances, cystic ovarian follicles resulted when adrenocorticotropin (ACTH) was administered daily during the follicular phase of the estrous cycle in cows. Two cows given daily injections of hydrocortisone (cortisol) during the follicular phase of the estrous cycle did not develop cystic ovaries. Plasma concentrations of estradiol in cows with induced cystic ovarian follicles were similar to the peak values observed at estrus and were between 6 and 12 pg/ml. Progesterone concentrations in plasma of cows with cystic ovaries were low, between 1 and 2 ng/ml. Ovulation occurred when 2 cows were given human chorionic gonadotropin (HCG) during the period of ovarian cyst development with ACTH administration. Several days of administration of ACTH was required to cause cyst development. Ovulation occurred at the expected time in 1 cow when injections began on day 19, that is, late in the follicular period. In another cow, when treatment was stopped on day 3, after the expected time of estrus a delayed ovulation occurred. In 2 cows with induced cystic ovarian follicles, cyst atresia occurred spontaneously about day 13 to 17 of the cycle. In these cows, new follicular growth and ovulation followed (although delayed in 1 cow). The time of atresia of cystic follicles was not influenced by the intrauterine injection of 10 ml of sterile saline solution on days 8, 9, and 10 in 1 cow. When 5 mg of prostaglandin F2alpha in 10 ml of sterile saline solution was given (uterine injection) in 2 cows on days 8, 9, and 10, cyst atresia occurred earlier than the time of spontaneous atresia. Intrauterine administration of 100 mg of indomethacin in 10 ml of sterile saline solution daily for 13 or 14 days to 2 cows, starting on day 12 or 13 of the cycle, resulted in persistence of the induced cystic ovarian follicles. After cessation of indomethacin treatment, atresia of cysts followed and new follicular growth and ovulation occurred.  相似文献

The corticotrophin (ACTH)-adrenal cortical axis has previously been implicated in the onset of cystic ovaries in the sow. In view of the role of the ACTH-adrenal cortical axis in stress, two sows were subjected to an elevated environmental temperature of 32 degrees C for three hours daily during the follicular phase of the estrous cycle. Plasma concentrations of glucocorticosteroids and progesterone fluctuated markedly in one sow that developed cystic ovaries. Concentrations of these hormones did not vary greatly in the other sow that did not develop cystic follicles. Exposure to an environmental temperature of 32 degrees C for three hours or injection of 1 IU/kg bodyweight of ACTH for each of two ovariectomized sows resulted in an elevation in progesterone values to 5-7 ng/ml plasma from basal levels of 1-2 ng/ml and a rise in total glucocorticosteroids from basal levels of 1 or 2 microgram/100 ml plasma to 4-10 microgram/100 ml. Injection of 2 mg/kg bodyweight of progesterone and 4 mg/kg bodyweight of cortisol into the ovariectomized sows was found to approximate these elevations in plasma steroid values. When either progesterone or cortisol was injected daily during the follicular phase into two intact sows in two successive experiments at these dosage levels, similarly elevated plasma steroid concentrations were seen and cystic ovarian follicles resulted. The results suggest that glucocorticosteroids and progesterone of adrenal origin may be involved in the onset of cystic ovaries in the sow.  相似文献

Three experiments investigated ovarian follicular development in sows whose litters were weaned at 28 to 31 d of lactation. Unilateral ovariectomy near the time of weaning was used to assess early follicular characteristics and to identify those sows that would not return to estrus within 10 d after weaning. This allowed segregation of and exclusion from the study those sows that had a prolonged interval from weaning to first estrus. In Exp. 1, 82 and 72% of the large follicles that were marked at 48 or 72 h after weaning (10 sows per time point) were subsequently identified as corpora lutea. In Exp. 2, sows (seven to nine per time point) were unilaterally ovariectomized at 0, 6, 12, 18, 24, or 48 h after weaning, and follicular fluid was evaluated for changes in steroid concentrations. Progesterone concentrations in fluid from medium-sized (4 to 6 mm) follicles increased by 6 h after weaning and then declined through 24 h concomitant with increases in testosterone and estradiol. For Exp. 3, follicular fluid and granulosa cells from individual follicles were obtained from sows (seven to nine per time point) at 0, 6, and 24 h after weaning. The weaning to estrus and weaning to ovulation intervals in sows are controlled by ovarian follicular growth after weaning. Longer intervals could be caused by smaller diameter follicles at weaning that take more time to reach a preovulatory size. We addressed this hypothesis by decreasing the diameter of follicular populations before weaning and then measuring follicular development and interval to estrus and ovulation after weaning. The posterior vena cava, cranial to the entry of the ovarian vein, was cathetered for blood sampling and infusion in 20 sows at 12 +/- 1 d after farrowing. Sows were assigned randomly to receive either 30 mL of charcoal-treated follicular fluid (FF, n = 9; a treatment known to decrease serum FSH and follicular diameter) or 30 mL of saline (n = 11) by venous infusion thrice daily (0700, 1500, and 2300 h) for 96 h beginning at 14 +/- 1 d after farrowing. Sows were weaned 48 h after the last infusion. Blood samples were collected for FSH analysis thrice daily beginning on the day of catheterization and continuing until ovulation. Follicular diameter was determined once daily by transrectal ultrasonography. A treatment x time interaction was detected for serum FSH (P < 0.001) and follicular diameter (P < 0.001) because serum FSH and the diameter of follicular populations decreased in FF sows during the infusion period. After the infusion period, serum FSH rebounded in FF sows, and follicles resumed growth but grew at the same rate as those of saline-treated sows, thus failing to achieve equivalent diameters relative to saline-treated sows on a given day after weaning. As a result, sows treated with FF had longer (P < 0.05) weaning to estrus (6.1 +/- 0.4 d) and weaning to ovulation (8.6 +/- 0.5 d) intervals compared with saline-treated sows (4.7 +/- 0.4 d and 7.2 +/- 0.4 d, respectively). We conclude that the diameter of the follicular population at weaning is one factor that controls interval to estrus and ovulation in sows. Small follicles at weaning cannot undergo compensatory growth and require additional time to reach a preovulatory size.  相似文献

Large multiple cystic ovarian follicles were induced in three sows and small multiple cystic ovarian follicles were induced in three other sows by injections of adrenocorticotrophic hormone (ACTH) during the follicular phase of the oestrous cycle. Plasma progesterone levels in sows with large cysts were relatively high, while those of sows with small cystic follicles were low. Removal of the ovaries with large cysts from one sow resulted in a precipitous drop in progesterone levels indicating that the large cysts were primarily the source of progesterone. The method by which ACTH may induced cystic ovaries was also investigated. Plasma progesterone levels in two ovariectomised sows during ACTH treatment fluctuated markedly during a 24 h period achieving peak values of 4 or 5 ng/ml. The results suggest that progesterone of adrenal cortical origin may be a factor in the development of the cystic ovarian condition.  相似文献

Endocrine differences between European crossbred and Chinese Meishan females have been somewhat modest. Indwelling jugular cannulas were placed in Meishan (n = 7) and White composite (n = 6) multiparous sows before weaning, and blood was sampled from 4 h before to 240 h after removal of litters. Temporal changes in FSH, triiodothyronine (T3), and tetraiodothyronine (T4) after weaning differed between Meishan and White composite sows. Plasma cortisol concentrations were higher in Meishan sows than in White composite sows (P < 0.01), but there were no temporal differences between breeds after weaning. Other hormones monitored (prolactin, GH, IGF-I, and inhibin) were not different between breeds. In the second experiment, Meishan gilts (n = 7) and sows (n = 7) and White composite sows (n = 9) were cannulated during the luteal phase of the estrous cycle and sampled after treatments consisting of GnRH (15 and 150 ng/kg BW), ovariectomy, estradiol cypionate challenge after ovariectomy (10 microg/kg BW), and GnRH antagonist. In response to GnRH challenge, White composite sows had elevated (P < 0.05) concentrations of gonadotropins compared with Meishan. Cortisol concentrations were elevated in Meishan as compared with White composite females (P < 0.01) but unaffected by GnRH treatment. After ovariectomy, LH concentrations increased 3 h sooner in White composite than in Meishan females. After GnRH antagonist, declines in gonadotropins were comparable in both breeds, but LH increased in Meishan females by 20 h but not until after 54 h in White composite females. White composite females demonstrated only a short decline in FSH in response to the GnRH antagonist, but Meishan females had a prolonged decline in FSH concentrations. Consistently elevated cortisol concentrations in Meishan females may positively impact ovarian function; thus, the assumption that high plasma cortisol concentration as an index of stress that impairs reproductive function should be reevaluated in swine. There were few endocrine differences that would relate to or explain increased ovulation rates found in the Meishan breed; thus, other mechanisms must exist to explain the increase in ovarian function in the Meishan breed.  相似文献

The use of urinary cortisol (UC) as an additional tool to evaluate sows welfare was assessed in two experiments. In a preliminary methodological experiment, the kinetics of cortisol excretion in urine was studied during an Adreno Cortico Trophic Hormone (ACTH) challenge test in 10 pregnant sows. In a second experiment, 96 primiparous sows of an experimental unit were assigned to two different housing systems: 48 animals were housed in individual pens (IP) and 48 animals in collective pens (CP) with 6 animals per pen. UC was measured at the beginning and at the end of pregnancy and compared with other welfare indicators such as behaviour or skin damage. In both experiments, UC was measured using a high pressure liquid chromatography assay. In experiment 1, UC was constant on the day before injection of ACTH, with no variations related to circadian rhythm. It began to rise 2 h after the injection, peaked between 2 to 5 h after then returned to the basal concentration on the day after the injection. In experiment 2, UC concentrations were not different between CP- and IP-housed sows but they were higher in sows exhibiting the less stereotypies in comparison with sows exhibiting the most stereotypies. The results of this study suggest that UC is a good indicator of acute stress, more convenient than plasma cortisol measurement since it is a non-invasive method avoiding restraint or catheterisation of sows. They also suggest that UC could also give additional information on the assessment of chronic stress and improve the evaluation of animal welfare if used in conjunction with other welfare indicators.  相似文献

The effects of single IV administered doses of dexamethasone on response to the adrenocorticotropic hormone (ACTH) stimulation test (baseline plasma ACTH, pre-ACTH cortisol, and post-ACTH cortisol concentrations) performed 1, 2, and 3 days (experiment 1) or 3, 7, 10, and 14 days (experiment 2) after dexamethasone treatment were evaluated in healthy Beagles. In experiment 1, ACTH stimulation tests were carried out after administration of 0, 0.01, 0.1, 1, and 5 mg of dexamethasone/kg of body weight. Dosages greater than or equal to 0.1 mg of dexamethasone/kg decreased pre-ACTH plasma cortisol concentration on subsequent days, whereas dosages greater than or equal to 1 mg/kg also decreased plasma ACTH concentration. Treatment with 1 or 5 mg of dexamethasone/kg suppressed (P less than 0.05) post-ACTH plasma cortisol concentration (on day 3 after 1 mg of dexamethasone/kg; on days 1, 2, and 3 after 5 mg of dexamethasone/kg). In experiment 2, IV administration of 1 mg of dexamethasone/kg was associated only with low (P less than 0.05) post-ACTH plasma cortisol concentration in dogs on day 3. In experiment 2, pre-ACTH plasma cortisol and ACTH concentrations in dogs on days 3, 7, 10, and 14 and post-ACTH plasma cortisol concentration on days 7, 10, and 14 were not affected by dexamethasone administration. The results suggest that, in dogs, a single IV administered dosage of greater than or equal to 0.1 mg of dexamethasone/kg can alter the results of the ACTH stimulation test for at least 3 days. This study investigated whether injections of ACTH for 48 h, from the onset of the second standing estrus after weaning, had any impact on time of ovulation and patterns of progesterone, estradiol, luteinizing hormone (LH), and inhibin alpha. The studied sows (n=15) were fitted with jugular vein catheters and randomly divided into a control (C group) and an ACTH group. From the onset of standing estrus, the sows were injected (NaCl or synthetic ACTH, 5 microg/kg) every 4h; blood samples were collected immediately before and 45 min after each injection. Ovulation was monitored using ultrasonography. The ACTH-group sows stopped displaying signs of standing estrus sooner after ovulation in their second estrus, but no impact was found on time of ovulation. There were no significant differences in the intervals between LH peak, estradiol peak, and the onset of standing estrus between the C and ACTH groups. The cortisol and progesterone concentrations were significantly elevated (p<0.001) in samples taken 45 min after ACTH injection. There were minor differences in estradiol and LH concentrations between the groups. Overall inhibin alpha concentrations were significantly higher during the treatment period in the ACTH than in the C group, but there were no significant differences between samples taken either 45 min or 4h after injection. In conclusion, injections of synthetic ACTH during estrus in the sow apparently disturb the duration of signs of standing estrus and the hormonal pattern of progesterone, and possibly of inhibin alpha, estradiol and LH.  相似文献

Full term crossbred sows were selected to study the interaction of the immune system, hypothalamus-pituitary-adrenal axis, and growth in pigs born by Cesarean section (c-section; n=4 sows) or vaginal birth (n=4 sows). Gestation length and birth weight did not differ between vaginal birth and c-section pigs (P=0.34 and 0.62, respectively). Blood and tissue samples were collected from 44 pigs at birth. Forty-five pigs were weaned at 13 d. On d 14, pigs received an i.p. injection of lipopolysaccaride (LPS; 150 microg/kg) or saline at min 0, and blood samples were collected at -20, -10, 0, 5, 10, 20, 40, 60, 90, and 120 min. Vaginal birth pigs had 21% greater average daily gain than c-section pigs on d 14 (P<0.01). Basal serum concentrations of adrenocorticotrophin (ACTH) and cortisol were greater in c-section than vaginal birth pigs at birth (P<0.01) but were not different at 14 d (P=0.99 and 0.80, respectively). LPS increased serum concentrations of ACTH, cortisol, interferon-gamma (IFN-gamma), and tumor necrosis factor-alpha (TNF-alpha; P<0.01) but the response was not different between c-section and vaginal birth (P>0.22). Basal serum concentrations of TNF-alpha tended to be greater in c-section vs vaginal birth pigs at 14 d (P=0.0967); however, basal serum concentrations of IFN-gamma tended to be lower in c-section pigs vs vaginal birth pigs at 14 d (P=0.0787). Expression of interleukin (IL)-6, IL-6 receptor, IL-1beta, and TNF-alpha mRNA did not differ between vaginal birth and c-section pigs but changed in an age and tissue dependent manner. Thus, reduced growth rate of c-section pigs is associated with altered immune system function.  相似文献

The effects of unilateral ovariectomy on ovarian compensatory hypertrophy (OCH), endocrine profiles and the pituitary response to gonadotropin releasing hormone (GnRH) were studied in 46 multiparous suckled sows. On d 20 of lactation (d 0 of experiment), sows were subjected to sham ovariectomy (Sham; n = 23) or unilateral ovariectomy (ULO; n = 23). On d 1 (n = 16), 2 (n = 15) or 8 (n = 15) following initial surgery the remaining ovaries in both Sham and ULO sows were removed. Immediately following removal of the remaining ovaries, GnRH (10 micrograms) was administered to each sow. Peripheral blood samples were taken every 10 min for 80 min beginning 20 min prior to GnRH administration. No difference in ovarian weight was observed between ULO and Sham sows until d 8, when ovarian weight was greater (P less than .05) for the remaining ovary from ULO sows (3.96 +/- .21 vs 5.91 +/- .39 g). Ovarian follicular fluid weights from ULO sows were greater (P less than .05) than Sham sows on both d 2 and 8. On d 1, plasma concentrations of follicle stimulating hormone (FSH) were greater (P less than .05) in ULO sows than in Sham sows (2.9 +/- .2 vs 2.1 +/- .1 ng/ml). Plasma FSH concentrations, however, did not differ between Sham and ULO sows on either d 2 or 8. Ovarian venous concentrations of estradiol-17 beta were also greater (P less than .05) in ULO sows compared with Sham sows on d 2 but not d 8.  相似文献

Exposing a pregnant sow to stress has been shown to affect the resulting offspring. Our objective was to determine if rough handling of pregnant sows altered the physiology of her offspring and if these alterations were different from an experimentally induced model of prenatal stress. Sow treatments consisted of i.v. injections of ACTH (1 IU/kg of BW), exposure to rough handling for 10 min (Rough), or no treatment (Control) once a week during d 42 to 77 of gestation. To determine the plasma cortisol response to treatments, blood (5 mL) was collected from 30 sows after treatment administration. To conduct the prenatal stress study, a separate group of 56 sows was used in 1 of 4 replicates. At birth, production data were collected for each litter, including birth weight, number born, anogenital distance, and pig viability. At weaning, pigs were blocked by BW and sex, and placed in a nursery pen of 6 pigs, with 2 pigs from each treatment group. To assess the effect of treatments on cortisol, corticosteroid-binding globulin (CBG), and hematological cell profiles, blood was collected every other day for 10 d after weaning. Application of treatments caused plasma cortisol concentrations to be greatest in ACTH sows compared with Control sows (P < 0.001), with Rough sows having intermediate values (P = 0.07). Treatments did not affect the number of pigs born, number of stillborn, or pig viability (P > 0.40). The ratio of cortisol to CBG did not differ between treatments (P = 0.09). Hematological variables did not differ between treatments (P > 0.19). Pigs born to ACTH sows had a smaller anogenital distance compared with controls (P < 0.03), with pigs from Rough sows being intermediate. Our data indicate that swine exposed to prenatal stress (ACTH injection) can have alterations in sexual morphology without effects on growth or the immune cell populations measured in this study.  相似文献

Immunoglobulins in piglets from sows heat-stressed prepartum   总被引:1,自引:0,他引:1  
Sows were subjected to moderate heat stress in a chamber (32 C) from d 100 of pregnancy until less than 8 h before delivery of first piglet, while control sows were in a thermoneutral chamber (21 C) or farrowing house (22 C). Blood serum and colostrum at parturition of heat-stressed sows and their piglets' serum at birth had elevated cortisol concentrations. Total protein, globulin and immunoglobulin G (IgG) concentrations in sow serum tended to decrease as parturition time was approached; albumin did not change. Total protein and IgG concentrations in colostrum at parturition and in milk 24 and 48 h later tended to be lower in heat-stressed sows. Concentrations of these four protein fractions (total, globulin, IgG and albumin) in piglet serum at birth did not differ among treatment groups, but soon after colostrum ingestion they increased markedly in all groups. Therefore, in all groups total protein remained constant while globulin and IgG decreased. Globulin concentration on d 1 was lowest in piglets from heat-stressed sows, but its rate of decrease after d 1 was not affected by sow treatment. Immunoglobulin G concentration was 11 mg/ml lower, but its rate of decrease through postnatal d 20 was slower in piglets from heat-stressed sows than in those from control sows; a 10-mg/ml difference in IgG concentration on postnatal d 1 has been associated with increased preweaning mortality in piglets. Higher cortisol concentration in serum and lower IgG in colostrum of sows under heat stress was associated in their piglets with higher serum cortisol at birth and lower serum IgG for the first 20 d postnatum.  相似文献

The aim of this study was to monitor changes in cortisol levels in sows around the time of separation from their piglets, in two different intermittent suckling regimes, compared with that in conventionally weaned sows. Sows were either weaned at 21 days of lactation (CONT) or subjected to an intermittent suckling regime (IS) from 14 days of lactation onwards. Sows in the IS regimes were separated from their piglets for 12 h every day, either from 08:00 to 20:00 hours (IS12) or from 08:00 to 14:00 hours and 20:00 to 02:00 hours (IS6). Separation caused a transient increase in cortisol levels on the first (CONT and IS12) and second (IS12) day of separation, compared with a gradual decline from early morning when the sows were still continuously suckling. In IS6 sows, in contrast, the transient rise in cortisol levels after separation at 08:00 hours was observed on the first 3 days and also on day 7 of the IS regime. Cortisol parameters were correlated with peri‐ovulatory characteristics like onset of oestrus, onset of the LH surge and time of ovulation. Onset of the LH surge was delayed in IS6 sows. In conclusion, increase in cortisol levels as a consequence of separation of sows and piglets, is an acute, incidental phenomenon in IS12 and CONT sows, but shows a repeated acute elevation in IS6 sows, possibly placing IS6 sows at a higher risk of influencing peri‐ovulatory processes and developing cystic follicles.  相似文献

The expression and concentration of follistatin and activin change during oestrous cycle suggesting their involvement in the regulation of follicular development. The aim of this study was to determine the level, source and potential role of follistatin in the sheep ovary. Follistatin in ovarian venous blood, measured by radioimmunoassay, remained at its low level from follicular phase (day ?1 and 0) to mid‐luteal phase (days 11–13) phase but were significantly elevated during the late luteal phase (days 14 and 15) when corpora lutea underwent regression. Western blot analyses of follicular fluid at day 15 of the cycle showed two strong bands at 42 and 45 kDa and weakly stained bands at 39 and 31 kDa. At day 0, these bands became weaker and the 39 kDa band became undetectable. However, there were no differences in follistatin concentrations between ovaries with and without functional corpus luteum (CL) during the whole luteal phase. In addition, although the ovaries of Booroola ewes normally contain more corpora lutea than those of normal merino ewes, follistatin concentrations in both jugular and ovarian venous blood were similar in Booroola and normal merino ewes. It is concluded that the secretion of follistatin from the ovary is not related to the formation of CL or high ovulation rate of Booroola ewes. Prenatal stress is known to affect several offspring characteristics, but its effects depend among other factors on the period of gestation in which it is applied. In the present study, oral administration of hydrocortisone-acetate (HCA) was used to elevate cortisol concentrations in pregnant sows to levels also observed after psychological stress. HCA was administered during three different periods of gestation (115 days in pigs): period 1: 21-50 (P1, n = 10), period 2: 51-80 (P2, n = 10) and period 3: 81-110 (P3, n = 10) days after insemination. Control sows (n = 11) received vehicle from 21-110 days after insemination. When P1-, P2- and P3-sows did not receive HCA, they also received vehicle. During gestation, weekly saliva samples were taken from the sows to determine salivary cortisol concentrations. Treatment effects on sow, litter and piglet characteristics were determined. In addition, two female piglets per litter were subjected to an ACTH-challenge test at 6 weeks of age to determine the adrenocortical response to ACTH. Pigs were slaughtered at 6 months of age and slaughter weight, back fat thickness and percentage of lean meat were analysed. During the period of treatment with HCA, salivary cortisol concentrations were increased in P1-, P2- and P3-sows compared to control sows (P < 0.01). The total number of piglets born per litter did not differ among treatment groups (P > 0.30), but pooled HCA-litters had a higher percentage of live born piglets (P < 0.05) and fewer mummies than control litters (P < 0.05). Gestation length did not differ among treatment groups (P = 0.21), but did affect treatment effects on birth weight. Overall, HCA-piglets weighed less at birth, and remained lighter until weaning (P < 0.05). The salivary cortisol concentrations after i.m. injection of ACTH (2.5 IU/kg) were lower in P1- and P3-piglets compared to control piglets. At slaughter, HCA-treatment indirectly decreased lean meat percentage and increased back fat thickness. In conclusion, elevated peripheral cortisol concentrations in pregnant sows affect both litter characteristics and piglet physiology, the latter depending on the period of gestation during which concentrations were elevated. Underlying mechanisms require further investigation.  相似文献

Grouping of sows is a stressful event until the ranking is established. The purpose of this study was to simulate stress by repeated administration of porcine corticotropin releasing hormone (CRH) and adrenocorticotropic hormone (ACTH)/tetracosactide and to study its influence on endocrine profile and ovulation. Four multiparous sows were used and blood was collected every 2 h from the onset of pro-oestrus until 12 h after ovulation. The first oestrus after weaning was used to check ovulation and acclimate the sows to their environment. The second oestrus after weaning was used as control. At their third oestrus CRH (0.6 microg/kg) and at their fourth oestrus ACTH (5 microg/kg) were given every 4 h from onset of oestrus until ovulation. The total 'area under the curve' of cortisol was twofold larger in two of four sows during the CRH treatment period, and two- to fourfold larger (p < or = 0.05) during the ACTH treatment period, compared with the corresponding control period. In three sows, there was no clear effect of either CRH or ACTH on the levels of oestradiol 17beta, luteinizing hormone (LH) or on the timing of ovulation. One sow was different in all hormonal patterns and also in the timing of ovulation. In all four sows, ACTH treatment lowered the baseline level of prostaglandin F(2 alpha)-metabolite. Therefore, we conclude that stage of the oestrous cycle seems to be of importance when investigating the influence of exogenous administration of CRH/ACTH on hormonal pattern and ovulation time in the sow.  相似文献

Duration and magnitude of hypothalamic-pituitary-adrenal axis suppression caused by daily oral administration of a glucocorticoid was investigated, using an anti-inflammatory dose of prednisone. Twelve healthy adult male dogs were given prednisone orally for 35 days (0.55 mg/kg of body weight, q 12 h), and a control group of 6 dogs was given gelatin capsule vehicle. Plasma cortisol (baseline and 2-hour post-ACTH administration) and plasma ACTH and cortisol (baseline and 30-minutes post corticotropin-releasing hormone [CRH] administration) concentrations were monitored biweekly during and after the 35-day treatment period. Baseline plasma ACTH and cortisol and post-ACTH plasma cortisol concentrations were significantly (P less than 0.05) reduced in treated vs control dogs after 14 days of oral prednisone administration. By day 28, baseline ACTH and cortisol concentrations remained significantly (P less than 0.05) reduced and reserve function was markedly (P less than 0.0001) reduced as evidenced by mean post-CRH ACTH, post-CRH cortisol, and post-ACTH cortisol concentrations in treated vs control dogs. Two weeks after termination of daily prednisone administration, significant difference between group means was not evident in baseline ACTH or cortisol values, post-CRH ACTH or cortisol values, or post-ACTH cortisol values, compared with values in controls. Results indicate complete hypothalamic-pituitary-adrenal axis recovery 2 weeks after oral administration of an anti-inflammatory regimen of prednisone given daily for 5 weeks.  相似文献
