中国东南沿海青蟹线粒体COI基因部分序列分析   总被引:17,自引:1,他引:17

对我国东南沿海5个地区的72个青蟹个体的线粒体细胞色素氧化酶亚基I (COI)部分序列序列进行了测定和分析。获得的72个细胞色素氧化酶亚基I (COI)序列可分为12个单倍型,与GenBank中已知的Scylla paramamosain COI序列的相似性达到98%以上,与其它3种 青蟹的差异为7.36%~15.54%。这些序列与S. paramamosain的遗传距离仅为0.00783,但是与S. serrataS. olivaceaS. tranquebarica〗的遗传距离却分别达到0.11659、0.17812和0.08423。序列特征、遗传距离和系统进化等分析结果都表明本文研究的青蟹为S. paramamosain。结果提示,在进行青蟹属相关研究应当仔细鉴别采集样本的种类。  相似文献

Dunaliella salina has been selected as the most valuable microalgae for production of carotenoids as a source of natural pigment when grown under defined conditions. Compared with optimal conditions, D. salina contained higher amounts of total carotenoids (TCAR, 12.6%, dw)—including β-carotene (60.4%, of total carotenoids), astaxanthin (17.7%), zeaxanthin (13.4%), lutein (4.6%), and cryptoxanthin (3.9%)—when grown under stress conditions. These TCAR showed greater in vitro antioxidant potential by inhibition of lipid peroxidation of rat liver microsomes induced by Fe++/ascorbate and Fe++/H2O2 model systems than butylated hydroxytoluene (BHT). The toxicological evaluation of D. salina functional ingredients as an antioxidant and natural food colorant was conducted, in which male albino rats were fed ad libitum for 6 weeks with one of the following diets: standard diet (S), S diets containing 0.5 g trans β-carotene/kg diets (S + 0.5 β C), S + 0.5 g D. salina carotenoids/kg diets (S + 0.5 DC), and S + 1.0 g D. salina carotenoids/kg diets (S + 1.0 DC). Generally, no mortality or treatment-related toxicological signs were observed in any of the rats during the 6 weeks of the experiment. The concentrations of glucose and glycogen in liver and glucose, total proteins, bilirubin, and creatinine as well as enzymes activities of aspartate (AST) and alanine aminotransferases (ALT), and alkaline phosphatase (ALP) in plasma were not significantly different from each other in all four diets. In contrast, the levels of total lipids and lipids components showed significant differences in all groups. Groups S + 0.5 βC, S + 0.5 DC, and S + 1.0 DC showed a significant decrease in concentrations of total lipids, triglycerides, total cholesterol, and phospholipids in either plasma or hepatic tissues compared with S group. The results showed that D. salina carotenoids could be potentially rich sources of natural antioxidants and food colorants and have hypolipidemic and hypocholesterolemic properties.  相似文献

Morphological changes in the embryos, egg size and development, incubation period and morphological structures of newly hatched zoea of three mud crab Scylla species were determined. The three species exhibited similar embryonic development composed of 10 stages. The mean egg diameter of Scylla serrata was significantly larger (P<0.05) at the prehatch stage. The mean egg diameters of Scylla tranquebarica and Scylla olivacea were similar (P>0.05). The incubation period was the longest in S. serrata and the shortest in S. olivacea. There was a positive relationship between egg size and larval size, as S. serrata exhibited the largest egg size and first zoea. However, no correlation was detected between egg size at prehatch and lengths of the morphological structures of the newly hatched zoea. The three species exhibited similar lengths of cephalic structures, but S. olivacea had significantly shorter (P<0.05) abdominal structures. The duration of spawning from ablation was the shortest in S. tranquebarica and the longest in S. olivacea. The study is relevant to aquaculture and fisheries management of Scylla species.  相似文献

The aim of the present study was to determine the effects of natural antioxidant extract isolated from the shells of giant red shrimp (Aristaeomorpha foliacea) on the changes in fatty acid profile of anchovy (Engraulis encrasicolus) during refrigerated storage (2.7°C). Total antioxidant activity of shell extract was determined as 45.84%, total phenolic compound as 17.87 mg/100 g shell, and total carotenoid content in shell as 20.31 mg/100 g shell. Total saturated fatty acid (SFA), monounsaturated fatty acid (MUFA), and polyunsaturated fatty acid (PUFA) contents in the control group and the group with 0.1% shell extract addition were found to be affected by the duration of storage (p < 0.05). A percentage increase was determined in MUFA content, while decreases were observed in PUFA content. No statistically significant change was detected in SFA, MUFA, and PUFA contents of groups with added butylated hydroxytoluene (BHT) and 0.5% of shell extract (p > 0.05). It was concluded that the extract isolated from shrimp shells could be used during the cold storage of fish fillets instead of synthetic antioxidants.  相似文献

With the increasing occurrence of dietary lead (Pb) contamination in aquaculture, a better understanding of the toxic effects of dietary Pb on aquatic animals is needed. Tilapia (Oreochromis niloticus) were exposed to dietary Pb at concentrations of 0, 100, 400 and 800 μg g?1 dry weight for 60 days, and Pb accumulation in tissues and blood, oxidative stress in posterior kidney and DNA damage in peripheral blood cells were investigated. The results showed that dietary Pb exposure resulted in significant Pb accumulation in tissues and blood, which increased with the dietary Pb concentrations. Pb accumulated in sampled tissues in the following order: posterior kidney>bone>liver>gill>spleen>testis>muscle>brain. Dietary Pb caused a significant increase in the malondialdehyde level and a significant decrease in the total antioxidant capacity content when compared with the control group (P<0.05), accompanied by concentration‐dependent decreases in the glutathione content, glutathione peroxidase and superoxide dismutase activities. Pb dose‐dependent DNA damage was observed in peripheral blood cells of tilapia exposed to dietary Pb. The results suggest that dietary Pb exposure can induce significant Pb accumulation in tissues and blood, followed by oxidative stress in posterior kidney and DNA damage in peripheral blood cells of tilapia.  相似文献

This experiment was conducted to assess the effects of antimicrobial peptides at different levels (Diet 1 (0%), Diet 2 (0.1%), Diet 3 (0.2%), Diet 4 (0.4%), Diet 5 (0.6%) and Diet 6 (1%)), on growth, serum biochemical indices and antioxidant effect, feed utilization and disease resistance in Litopeneaus vannamei. There were four replicates in each group in the experiment (mean weight = 0.21 ± 0.00 g) and also fed with their respective diets for 8 weeks. Growth performance compared with the control group (0%) significantly increased at first and then decreased among treatment groups (p < .05) whereas the survival rate ranged from 78% to 96%. No significant differences were observed in terms of moisture, crude protein and ash content, but there was a significant increase in crude lipid (p < .05). In serum, acid phosphatase, alkaline phosphatase, polyphenol oxidase, triglyceride, glucose and total cholesterol changed as compared with 0%. Total antioxidant capacity, glutathione peroxidase, catalase, superoxide dismutase and malondialdehyde were also different from 0%. Disease resistance was increased in shrimp among treatment groups with 0.4% recording the lowest mortality percentage of 37% after the challenge test. The results from the present study suggested that supplementation of AMP at 0.4% in shrimp diet can improve growth performance, antioxidants activities and innate immune response of Pacific whiteleg shrimp.  相似文献

An 8‐week feeding trial was conducted to evaluate the effects of a dairy‐yeast prebiotic (Grobiotic®‐A) on growth performance, body composition, antioxidant capacity and immune functions of juvenile starry flounder, Platichtahys stellatus. Three triplicates of starry flounder (average initial weight of 15.05 ± 0.04 g) were fed one of six diets formulated to supplement dairy‐yeast prebiotic at 0%, 0.4%, 0.8%, 1.2%, 1.6% or 2.0% respectively. Results showed that weight gain, daily feed intake, protein efficiency rate and condition factor increased significantly (P < 0.05) whereas feed conversion ratio decreased significantly with the increasing level of the dairy‐yeast prebiotic supplement. Crude lipid content in dorsal muscle and liver decreased significantly (P < 0.05). No significant differences occurred in moisture and crude protein content of dorsal muscle and liver with the increasing level of the dairy‐yeast prebiotic supplement. Compared with fish feed the basal diet, the activity of catalase and total antioxidant capability in serum and liver, as well as total superoxide dismutase in serum all significantly increased with high level of the prebiotic, while the malondialdehyde content in serum and liver decreased significantly. In serum, the activity of alkaline phosphatase, lysozyme, complement and the content of nitric oxide were significantly increased at higher level of prebiotic supplementation while no significant differences were found in total protein, albumin, globulin, albumin to globulin ratio, haemoglobin, acid phosphatase and myeloperoxidase. Based on weight gain response using the quadratic regression, combine the antioxidant and immune indices, the optimum dietary dairy‐yeast prebiotic level for juvenile starry flounder was estimated to be 1.33% under these experimental conditions.  相似文献

An 8‐week feeding experiment was conducted to determine the dietary magnesium (Mg) requirement and physiological responses of Litopenaeus vannamei in low salinity water of 2 g L?1. Casein–gelatin‐based diets supplemented with seven levels of Mg (0, 0.4, 0.8, 1.6, 3.2, 6.4 and 8.0 g kg?1) were fed to juvenile shrimp. Prior to the experiment, the postlarvae were gradually acclimated to the low salinity media and fed with a basal diet (0.5 g Mg kg?1) for 2 weeks. After 8 weeks of feeding, survival ranged from 80.11% to 85.65% with no significant difference among the treatments. Hepatopancreas Mg2+‐ATPase and Na+/K+‐ATPase activities and muscle content of lipid and protein were not significantly affected by graded levels of Mg. The weight gain and mineral (calcium, potassium, sodium and total phosphorus) content of different tissues were significantly affected by dietary Mg levels, while there were no significant differences in ash and zinc content in tissues. The Mg content in tissues except hepatopancreas was maintained relatively constant regardless of dietary treatments. The dietary Mg requirement for optimal growth was 2.60–3.46 g Mg kg?1 by using the polynomial regression analysis based on growth.  相似文献

Change in environmental salinity level is a major limiting factor for the aquaculture productivity because it imposes severe stress on organisms that in turn retards growth. The orange mud crab (Scylla olivacea) is an important coastal aquaculture species (farming is practised in 10‰–20‰ salinity levels) in Bangladesh. The present study was conducted to investigate the changes in growth, O2 consumption and mRNA expression levels of five selected genes in the orange mud crab (S. olivacea) exposed to three different experimental salinity levels (0‰, 10‰ and 20‰) for three months. Crabs reared at 10‰ and 20‰, showed significantly higher (p < .05) growth performance and expression of growth regulatory genes (Actin and α‐amylase). The highest levels (p < .05) of O2 consumption and expression of ion regulatory genes (Na+‐K+‐ATPase, V‐type H+‐ATPase and Diuretic Hormone) were obtained at 0‰. Moderate levels of growth and expression of selected candidate genes were observed at 10‰ treatment while the highest levels of growth and gene expression were obtained at 20‰ (control salinity). Strong interactions were observed between growth performance and expression of growth genes (R2 = 0.81–0.91), and rate of O2 consumption and expression of ion regulatory genes (R2 = 0.83–0.93), implying that the selected genes are important candidates for growth and ionic balance in S. olivacea. Growth performance was found to be very low at 0‰ initially, after 30 days crabs showed better growth performance at this salinity level. It is thus inferred that orange mud crab individuals might require 3–5 days for acclimation to salinity stress but it can take at least 30 days for acclimation to regular growth. Results indicate that with proper acclimation, the orange mud crab (Scylla olivacea) can be farmed at low salinity conditions and possibly in freshwater condition.  相似文献

在容积为500 L的圆锥形塑料缸中分别用对照(自然海水)、配制的盐度为20、10、5的海水和淡水(地下水)养殖点篮子鱼[(67.76?26.12) g],研究了不同盐度对点篮子鱼存活、生长和抗氧化酶活性的影响。结果显示,淡水组第9天出现死亡,至第27天时死亡率达100%,其余各组未出现异常,死亡率为0%。各盐度组鱼的特定生长率未表现出显著性差异,但盐度10组鱼体重显著高于盐度20和盐度5组,与对照组无显著性差异;盐度5组全长显著低于其余各盐度组。驯化40 d后,对各组鱼鳃、肝脏、肾脏和肌肉中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、抗超氧阴离子自由基、羟自由基活性检测结果显示,除盐度5组外,盐度10、20组点篮子鱼鳃、肝脏、肾脏、肌肉中SOD、CAT、抗超氧阴离子自由基和羟自由基活性在驯化40 d后均恢复到对照组水平,组间无显著性差异;盐度5组中鱼鳃的SOD和抗超氧阴离子自由基活性显著高于盐度10和盐度20组,肝脏、肌肉和肾脏中SOD、CAT、抗超氧阴离子自由基和羟自由基活性均恢复到对照组水平。点篮子鱼不同组织中SOD和CAT酶活力在不同盐度下均以肝脏中最高,肾脏和鳃次之,肌肉中最低;抗超氧阴离子自由基活力以肝脏中最高,极显著高于其余各组织中抗超氧阴离子自由基活力(P<0.01),肌肉次之,肾脏和鳃最低;羟自由基活力以肾脏中最高,显著高于其余各组织中羟自由基活力(P<0.05);鳃次之,肝脏和肌肉中最低,结果表明盐度能影响抗氧化酶活力大小,但并未影响鱼体内酶的分布。  相似文献

为分析马粪海胆和光棘球海胆性腺的脂质组成和抗氧化活性,采用核磁共振和气相色谱—质谱技术对2种海胆性腺油脂的脂质成分和脂肪酸组成进行分析,并通过DPPH自由基清除法、羟基自由基清除法和超氧阴离子自由基清除法对其脂质的抗氧化活性进行研究。结果显示,马粪海胆和光棘球海胆性腺脂质均以甘油三酯和磷脂为主,胆固醇、胆固醇酯和游离脂肪酸含量较低。马粪海胆和光棘球海胆性腺总脂富含C20:4n-6和C20:5n-3,且二者总量分别占脂肪酸含量的35.88%和34.98%;同时2种海胆性腺的中性脂和极性脂的脂肪酸组成存在较大差异,中性脂以C14:0和C16:0等饱和脂肪酸为主,而极性脂以C20:4n-6和C20:5n-3等多不饱和脂肪酸为主。马粪海胆和光棘球海胆性腺脂质对DPPH自由基、羟基自由基和超氧阴离子自由基均具有较好的清除能力,DPPH自由基IC50分别为2.75和1.98 mg/mL,羟基自由基IC50分别为0.33和0.29 mg/mL,超氧阴离子自由基IC50分别为0.33和0.31 mg/mL。研究表明,马粪海胆和光棘球海胆性腺脂质具有较高的营养价值和抗氧化活性,可作为C20:4n-6、C20:5n-3和磷脂等功能性脂质因子的重要膳食来源。  相似文献

Temporal diet changes can be detected by comparing δ13C and δ15N values between tissues with different isotopic turnover times. However, other factors contribute to disparities in δ13C and δ15N signatures between tissues and could confound the interpretation of stable isotope data. We examined the effects of ecological factors on differences in muscle and liver δ13C and δ15N signatures of Arctic charr [Salvelinus alpinus, (L.)] to determine whether their effects were large enough to influence studies of diet change. In addition, we examined whether differences in lipid content accounted for a significant portion of the differences in δ13C values between tissues. Results indicated that life history, diet, reproductive status and gender had significant effects on the differences in δ13C values between tissues, while gender and diet significantly affected differences in δ15N values. Differences in % lipid content between tissues also explained approximately 74% of the observed variation in the difference in δ13C values between tissues. The differences in δ13C values observed between muscle and liver tissues may be partially explained by physiological differences in lipid storage and use between tissues associated with ecological factor effects. These results demonstrate that ecological factors other than a change in diet can influence the differences in δ13C and δ15N signatures between muscle and liver tissues. However, the impact of ecological factors on studies of diet change depends on how the magnitude of the factor effects compared with a biologically meaningful change in diet.  相似文献

A 60‐day growth experiment was conducted to investigate the effect of dietary replacement of fishmeal by Spirulina platensis on growth performance, digestion and physiological parameters in juvenile gibel carp (5.0 ± 1.0 g). Four diets were formulated to replace 0 (SP0), 50% (SP50), 75% (SP75) and 100% (SP100) of dietary fishmeal protein by S. platensis respectively. Other two diets based on SP100 were supplemented with 2% dicalcium phosphate (SP100 + P) or 0.28% coated lysine (SP100 + Lys). The results showed that no significant differences of specific growth rate (SGR), feed efficiency (FE), feeding rate (FR) and protein efficiency ratio (PER) were observed between SP0 group and the replacement groups (SP50, SP75 and SP100). FE and PER of fish‐fed SP100 + Lys diet were significantly higher than the fish‐fed SP100 diet (< .05). Apparent digestibility coefficients (ADCs) of dry matter, protein, energy and phosphorus increased significantly with the increasing S. platensis inclusion. Meanwhile, fish from SP100 + P or SP100 + Lys group had higher ADCs of nutrients compared to the SP100 group (< .05). Compared with SP0 diet, fish‐fed S. platensis diets had higher activities of plasma superoxide dismutase, hepatopancreas total antioxidant capacity, plasma alkaline phosphatase and plasma lysozyme, as well as the lower content of plasma malondialdehyde. The present results indicated that dietary fishmeal could be fully replaced by S. platensis without any negative effects on growth performance of gibel carp. Supplementation of lysine in the S. platensis replacement diet could further improve the feed utilization of gibel carp.  相似文献

A‐56 days feeding trial was conducted to evaluate the growth performance, feed utilization, biochemical composition, antioxidant status, lipid metabolism and immunity parameters of Chu's croaker Nibea coibor fed diets supplemented with different levels of conjugated linoleic acid (CLA): 0% (control), 0.5%, 1%, 1.5% and 2%. Each diet was randomly assigned to triplicate groups of 25 fish (initial body weight: 12.8 ± 0.1 g) in 15 floating cage. Weight gain (WG) and specific growth rate (SGR) were not significantly affected. 2% of dietary CLA led to significant lower lipid content in both whole body and liver (< .05). Muscle lipid content was significantly lower in fish fed 1.5% and 2% CLA (< .05). Saturated fatty acid (SFA) increased while monounsaturated fatty acid (MUFA) decreased with increasing dietary CLA in tissues (< .05). Increasing CLA significantly promoted hepatic lipase (HL), lipoprotein lipase (LPL), serum lysozyme (LSZ) and immunoglobulin M (IgM) while decreased serum triglyceride (TG) in both liver and serum (< .05). Liver Superoxide dismutase (SOD), catalase (CAT) and total antioxidant capacity (t‐AOC) increased while malondialdehyde (MDA) decreased (< .05). Our study shown that Chu's croaker can successfully incorporate CLA in tissues up to 2% dietary lipid without growth inhibition, and that CLA increased fish quality due to the increased parameters of lipid metabolism, specific immunity, hepatic antioxidant and CLA accumulation, and the reduction of tissue fat deposition.  相似文献

This study represents a first report on the proximate composition and fatty acid profile of Lagocephalus sceleratus from the Mediterranean Sea as well as seasonal changes and sexual differences. Significant variations in the proximate composition were observed during the months of the study (p < 0.05). The highest moisture, fat, protein, and ash contents were found to be 78.47, 0.33, 21.62, and 1.63%, respectively. Total saturated and monounsaturated fatty acid values were similar throughout the year despite individual variations of 24.22–29.46 and 7.87–13.94%, respectively. However, significant variations were observed in total polyunsaturated fatty acids as well as significant differences between sexes (p < 0.05) with some exceptions. The major fatty acid was found as docosahexaenoic acid (DHA), between 24.17 and 32.65%. Due to the low fat content of this species, the highest eicosapentaenoic acid (EPA) + DHA values were calculated as 80.93 mg/100 g of edible muscle. The results indicated that this species contains high nutritional properties in terms of protein content and fatty acid values.  相似文献

An 8‐week feeding trial was conducted to determine the effect of dietary conjugated linoleic acid (CLA) on growth performance, muscle fatty acid profile, hepatic intermediary metabolism and antioxidant responses in genetically improved farmed Tilapia (GIFT) strain of Oreochromis niloticus (initial body weight: 42.6 ± 0.4 g, mean ± standard deviation). Three replicated groups of GIFT strain of Nile tilapia were hand‐fed to satiation, twice a day, with the diets in which CLA oil, containing mainly the bioactive cis‐9, trans‐11 and trans‐10, cis‐12 isomers, was included at 0 (control), 0.5%, 1%, 1.5%, 2% and 2.5%, respectively, at the expense of fish oil to maintain the constant lipid and energy levels. Growth performance and feed utilization showed no significant differences among the treatments (P > 0.05). The dietary inclusion of CLA modified total percentages of the main groups of fatty acids. Increasing saturated fatty acid content and reduced mono‐unsaturated fatty acid contents in muscle were observed with increasing dietary CLA inclusion (P < 0.05). Total n‐3 fatty acids and total polyunsaturated fatty acids tended to decline with increasing dietary CLA levels (P < 0.05), but n‐6 fatty acids showed no significant differences among the treatments (P > 0.05). Dietary CLA supplementation resulted in the significant increase in the trans‐10, cis‐12 and cis‐9, trans‐11 CLA isomers in muscle (P < 0.05) and also significantly influenced several hepatic enzymatic activities, such as succinate dehydrogenase, lactate dehydrogenase, malic dehydrogenase, lipoprotein lipase and hepatic lipase activities (P < 0.05). Reduced superoxide dismutase and glutathione peroxidase activities, and the decline in malondialdehyde levels were observed in fish fed the CLA‐supplemented diets (P < 0.05), indicating that dietary CLA supplementation showed a powerful antioxidant effect for this fish species. Our study was the first report involved in the effect of dietary CLA inclusion on hepatic intermediary metabolism and antioxidant responses in fish, which could be used as indicators of nutritional and physiological status of the fish species.  相似文献

The fishery biology of mud crabs Scylla spp. was examined using baited traps and gill nets from September 2001 to August 2005 at Iriomote Island, Japan. To elucidate the growth of the crabs, artificially produced S. serrata juveniles were released and recaptured at the study site. The sizes at which 50% of females and males of S. serrata reached sexual maturity (SM50) were estimated as an external carapace width (ECW) based on the morphology of the abdomen and the chela respectively. Two species, S. serrata and S. olivacea, were identified in the area with S. serrata being the dominant species (>95% of the catch). Changes in the mean ECW and the results of the release and recapture experiments suggested that the recruitment of young crabs to the fishery occurred from December/January to April/May. The SM50 of females and males occurred at 132.4 and 150.7 mm ECW respectively. The body size composition of S. serrata revealed that immature crabs comprised approximately 40 and 65% of the catch for females and males respectively. To maintain a sustainable fishery for S. serrata, a minimum landing size based on the SM50 estimates should be implemented as a fishing regulation.  相似文献

Eight tropical marine macro algae were investigated for in-vitro antioxidant, antibacterial, and biochemical properties. The moisture content [% DW (dry weight)] ranged from 7.21 to 14.72%; ash, 24.92 to 47.04%; lipids, 0.73 to 2.67%; protein, 4.56 to 12.59%; and carbohydrate, 30.1 to 48.51%. The % sum of polyunsaturated fatty acids (PUFAs) ranged from 21.71 to 78.22%, while omega-6/omega-3 (ω6/ω3) ratio was from 0 to 2.08, which remained within the prescribed World Health Organization (WHO) standards (<10). The % sum of essential amino acids (EAAs) ranged from 86.18 to 204.66, and Na/K ratio ranged from 0.37 to 2.85. The extracts exhibited significant (< 0.05) values for total phenol (7.25–16.0 µg PGE mg–1 DW) and total flavonoid (4.06–15.63 µg QE mg–1 DW) at 200 µg/ml. Antibacterial activity was tested against the selected food spoilage and pathogenic bacteria.
