The virulence ofPhytophthora citrophthora isolated from various host-plants on three peach rootstocks (GF677, PR204, KID I) was examined. There was no significant difference among the rootstocks with respect to their susceptibility to testedP. citrophthora isolates. The most virulent isolate originated from sycamore (Acer pseudoplatanus); isolates from pistachio trees (Pistacia vera) also showed high virulence but were significantly less virulent than the sycamore isolate. Isolates originating from plum (Prunus domestica), almond (Prunus amygdalus) and lemon (Citrus limon) trees were moderately virulent on peach rootstocks; those from cyclamen (Cyclamen persicum) showed the lowest virulence of those tested. There was thus great variation in virulence among the testedP. citrophthora isolates. It is possible that the isolates ofP. citrophthora from sycamore, pistachio, plum, almond and lemon trees are a threat to peach trees, whereas the low virulence of the isolates from cyclamen hosts suggests that these pathogens are not a serious threat to peach trees. http://www.phytoparasitica.org posting Jan. 3, 2002.  相似文献

A severe crown rot of pear trees of cultivar ‘Kondoula’ grafted on quince rootstock was observed in Greece. Isolations from the affected tissues repeatadly yielded aPhytophthora sp. that was determined by morphological and physiological characteristics to beP. citrophthora. The pathogenicity of two of theP. citrophthora isolates was tested by inoculating trunks of 2-year-old pear trees by mycelial agar disks. Thirty-two days after inoculation all inoculated trees were infected. Although the pear isolates could not be differentiated from isolates ofP. palmivora orP. nicotianae based on isozyme profiles of α-esterase or lactate dehydrogenase, RAPD profiles with one selected primer differentiated the pear isolates from the other species and revealed an electrophoretic banding pattern similar to that of aP. citrophthora standard. This is the first report ofP. citrophthora on pear trees in Greece.  相似文献

为明确不同梨和苹果种质对阿太菌果腐病菌Athelia bombacina的抗性以及筛选防治其有效杀菌剂,采用离体菌丝块有伤接种方法对40份梨种质和154份苹果种质进行病斑直径测定,通过聚类分析法和平均病斑直径法对不同种质进行了抗病性分级,并利用菌丝生长速率法测定了13种常用杀菌剂对阿太菌果腐病菌的毒力。结果表明,根据聚类分析法和平均病斑直径法均可将梨和苹果种质划分为高抗、抗、中抗、感和高感5类。与平均病斑直径法相比,梨和苹果种质分别以欧式距离为14和10作为最佳聚类分割点时进行聚类分析能够更加科学地划分类别,从40份梨种质中筛选出金锤子梨1个高抗种质,仅占总数的2.50%,从154份苹果种质中筛选出垂丝海棠、莫斯科透明、新疆苹果、路边石、伏帅等32个高抗种质,占总数的22.73%。药剂试验结果表明,戊唑醇、腈菌唑、咯菌腈和噻呋酰胺对阿太菌果腐病菌菌丝生长的抑制效果较好,抑制中浓度EC_(50)分别为0.027、0.048、0.054和0.095 mg/L。表明梨和苹果种质均可被阿太菌果腐病菌侵染,但不同种质间抗病性差异明显,戊唑醇是采前防治阿太菌果腐病菌菌丝生长的最佳药剂。  相似文献

Root and crown rot of raspberry (Rubus idaeus L.) was observed in a plantation at the experimental station of small fruits in Kostinbrod, Bulgaria. Isolates ofPhytophthora spp. were obtained from diseased plants. Colony morphology, growth rates, features of asexual and sexual structures were studied and as a result twoPhytophthora species were identified:Phytophthora citricola Saw. andPhytophora citrophthora (R.E. Sm. & E.H. Sm.) Leonian. Their pathogenicity was confirmed in artificial inoculation experiments. The isozyme (-esterase) patterns ofP. citrophthora andP. citricola isolates from raspberry and from the collection of the CBS, Baarn the Netherlands were compared, using micro-gel electrophoresis. Both species are reported for the first time as pathogens of raspberry in Bulgaria. This is only the second report in phytopathological literature ofP. citrophthora on raspberry, the first being from Chile [Latorre and Munoz, 1993].  相似文献

Pythium spp. that cause damping-off of seedlings also can cause root rot of older plants and lead to yield reductions. This can be especially severe in soilless cultures where the fungus can spread easily with the nutrient solution. 39Pythium isolates obtained from discolored roots were assayed for their ability to cause damping-off on cucumber seedlings in sand-peat and for their pathogenicity in soilless culture of cucumber in rockwool or hydroponic solution. Isolates ofPythium aphanidermatum, P. irregulare, P. sylvaticum andP. ultimum were highly pathogenic in sand-peat, but onlyP. aphanidermatum strains were pathogenic in soilless conditions and led to root decay, plant death in rockwool culture and growth reduction in hydroponic culture. One strain ofP. aphanidermatum significantly reduced the yield of cucumber grown in rockwool under conditions similar to those of commercial cultures.  相似文献

The effect of microjet, drip, and two durations of sprinkler irrigation systems on phytophthora crown and root rot of apple trees was examined under field conditions. This eight year study indicates that crown and root rot caused byPhytophthora cactorum was most severe where young MM. 106 rootstock trees were watered by microjet irrigation for 2.3 h each day. There was no difference in infection byP. cactorum when trees were irrigated either by drip or sprinkler irrigation systems. The MM.106 apple rootstock trees watered by drip irrigation for 2.6 h each day were least affected by phytophthora crown and root rot.  相似文献

The pathogenicity and virulence of ten GreekPseudomonas syringae pv.syringae strains from different hosts (citrus, pear, apple, peach and cherry) were evaluated using three different laboratory methods, which produced results in good agreement. All ten strains were virulent on apple, pear, cherry and peach trees. The extent of tissue colonized varied considerably among strains and cultivars. On excised shoots and twigs of apple and pear, strains BPI 176, BPI 203, PI 2 and PI 14 were the most virulent and strains BPI 689, BPI 992, BPI 4, BPI 20, PI 18 and PI 19 were the least virulent. On excised shoots and twigs of peach and cherry, strains BPI 176, BPI 203, PI 2, PI 14, PI 18 and PI 19 were the most virulent and strains BPI 4 and BPI 20 were the least virulent. Moderate virulence was evinced by strains BPI 689 and BPI 992. These pathogenicity assays are proposed as rapid and reproducible screening systems to evaluate the susceptibility of apple, pear, cherry and peach cultivars to this bacterial pathogen.  相似文献

Potential antagonists ofFusarium solani f. sp.pisi (Fsp) were selected from soil samples with varying degrees of receptivity to this pathogen. They were tested against Fsp isolate 48 (Fs48), in increasingly complex systems. Most species testedin vitro were able to antagonize Fs48. No relation could be establishedin vitro between the receptivity of the soil from which an isolate originated and its antagonism to Fs48. In soils naturally infested with pea root rot pathogens, which were stored humid at 4°C for a period longer than a year, various isolates ofFusarium, Gliocladium andPenicillium spp. were able to reduce root rot. After sterilization of these soils, onlyGliocladium roseum isolates, added at 105 conidia g–1 dry soil, significantly reduced disease severity and prevented root weight losses caused by Fs48 at 104 conidia g–1 dry soil. In soils in which the biota were activated by growing peas before the assays, doses of 106 and 107 ofG. roseum were required to reduce root rot. In these soils, the antagonistic effects of fluorescent pseudomonad strains from soil of low receptivity to Fsp were variable. Some strains of fluorescent pseudomonads, from soil moderately receptive to Fsp and from highly infested soils, were also able to reduce root rot. Disease suppression by pseudomonad strains was more evident in the absence than in the presence ofAphanomyces euteiches in the root rot pathogen complex. The role of receptiveness of the soil with regard to potential antagonists is discussed.  相似文献

Disease on fruit trees in Bulgaria caused by Phytopthora cactorum and P. citrophthora was found in the period 1998–1999. Leaves of some trees become reddish during July, and later in the season fall off. Infected trees die during the same season, or the next season. Observations on symptom development and spread of Phytophthora root and crown rot of fruit trees was undertaken from 1999 to 2009. Disease incidence is between 2% and 14% in some gardens and nurseries. The disease was registered in the regions of Plovdiv, Kjustendil, Sliven, Yambol, Karnobat, Bourgas and Svishtov. Samples from infected plant tissues were taken and isolations were done on selective PARP media, or by applying a baiting bioassay. Based on morphological and cultural characteristics and temperature requirements the following Phytophthora species have been identified: Phytophthora cactorum, P. citrophthora, P. drechsleri, P. cryptogea, hybrid and Pythium. Pathogenicity of the isolates was tested on green apple fruits or one-year-old apple rootstocks. Laboratory studies of the effect of temperature on mycelia growth showed that most isolates can grow from 5° up to 30°C, with an optimum from 18° to 25°C. Only three strains grew at 35–36°C, two developed slowly, one grew well. The optimal pH for mycelia development was tested. Aiming at control of disease, in vivo pot trials have been carried out for studying resistance of rootstocks to P. cactorum. At the end of the growing season a good level of resistance has been shown in the rootstocks M29C, Gizela 6, and MAXMA 14.  相似文献

A collection of 205 strains ofErwinia amylovora isolated in Israel over a period of 12 years has been established. The strains were isolated from different varieties of pear, apple, loquat and quince grown in Israel, and collected from different locations in the country. They were characterized in respect to degree of virulence on several hosts and serological and molecular characters. Pathogenicity tests carried out on flowering branches of pear and apple, shoots of pears, and on trees of pear and loquat grown in containers outdoors, revealed no significant differences in the severity of blossom blight or shoot blight among the various strains. ELISA and immunofluorescence assays revealed no serotypic groups among the Israeli strains. Genomic diversity was studied by random amplified polymorphic DNA (RAPD) analysis using 24 arbitrary 10-base primers. All the strains examined (45 Israeli and 11 from Egypt, Cyprus and Greece) produced the same RAPD patterns with each of the primers used. Amplification patterns were indistinguishable from those produced by strains isolated from the neighboring countries. Results presented in this study suggest that the population ofE. amylovora in Israel is homogenous.  相似文献

Prospects of durability of resistance in lily to basal rot have been evaluated by testing the virulence and aggressiveness of 31 isolates ofFusarium oxysporum f. sp.lilii towards a number of different resistance sources inLilium spp. Isolates differed strongly in aggressiveness as did species and cultivars ofLilium spp. in resistance. Significant interactions were observed between isolates of the pathogen and genotypes ofLilium spp., but the magnitude was very small compared to the main effects. The interactions were mainly due to a small group of isolates with low aggressiveness. It is argued that the interactions might be based on minor genes. No major break down of the resistance was found. For practical purposes it will be sufficient to use highly aggressive isolates in screening tests.  相似文献

Excised twig assay and excised stem inoculation were used to evaluate nine fungicides (metalaxyl, fosetyl-Al, copper hydroxide, copper sulfate, copper oxychloride, captan, quintozene, propamocarb and chlorothalonil) againstPhytophthora cactorum andP. citrophthora associated with crown rot in peach trees. Segments were soaked in fungicide solutions at different concentrations and then inserted vertically intoP. cactorum orP. citrophthora cultures growing on cornmeal agar plus antibiotics, or inoculated by inserting a mycelium-bearing agar plug directly into the cambium. Following incubation, the bark was scraped off and length of necrosis was measured. Metalaxyl was the only fungicide that inhibited canker development on segments at the manufacturer-recommended concentration. Fosetyl-Al, captan, copper hydroxide and copper sulfate inhibited canker development at 3, 4, 4 and 8 gl-1, respectively. The other fungicides did not affect canker length significantly compared with non-treated twigs, with the exception of propamocarb, which reduced the development ofP. cactorum on excised stems. A boll rot of cotton (Gossypium hirsutum L.) was observed for the first time in Greece in August 1993 in Larissa and Volos counties, and in August and September 1995 in Trikala and Phthiotis counties. Fungi of the genusPhytophthora were isolated from diseased plants. Morphological characteristics of the pathogen were recorded on mounts made directly from the infected tissues or after growth of the isolated fungus on corn meal agar or sterile distilled water. Colony morphology, growth rates, features of asexual and sexual structures and maximum growth temperatures were examined. APhytophthora species new to Europe,Phytophthora boehmeriae Sawada, attacking cotton bolls, was identified. The pathogenicity of the isolates was confirmed by artificial inoculations of detached cotton bolls. Analysis of α-esterase isozymes revealed unique banding patterns for isolates ofP. boehmeriae compared with those ofP. cactorum andP. parasitica, which arePhytophthora species with similar morphology.  相似文献

 1993~1995年,对侵染新疆主要农作物的疫霉菌进行了较全面的调查研究,共从9个地区的28种作物上采集表现根腐、根颈腐、果腐症状(包括病土)的病样1531个,结果从其中17种作物上分离到452个疫霉菌株。根据形态特征、生理生化特性、致病性和菌体可溶性蛋白电泳测定,鉴定为7个种:辣椒疫霉(Phytophthora capsici Leon.),恶疫霉[P.cactorum(Leb.et Cohn) Schroeter],掘氏疫霉(P.drechsleri Tucker),菸草疫霉(P.nicotianae van Breda de Haan),苎麻疫霉(P.boehm eriaeSaw.),柑桔褐腐疫霉[P.citrophthora (Sm.et Sm.) Leonian],隐地疫霉(P.cryptogea Pethyb.etLaff.)。这些疫菌是造成新疆茄果类、瓜类、棉花、苹果、梨、枸杞、草霉、红花、白术等幼苗及成株大量死亡及烂果的主要病原,在农业生产中具有十分重要的意义。  相似文献

Forty-two isolates ofColletotrichum gloeosporioidesfrom almond, apple, avocado, mango, pecan, and eight isolates ofC. acutatumfrom apple, peach and pecan were compared by molecular analyses and a pathogenicity assay in order to determine genetic variability and host specificity. Polymerase chain reaction (PCR) amplification of genomic DNA using four different primers andHaeIII digestion patterns of genomic DNA (A+T-rich DNA) grouped theC. acutatumisolates separately from theC. gloeosporioidesisolates. Based on arbitrarily primed PCR (ap-PCR), intraspecies similarity among the isolates ofC. acutatumandC. gloeosporioidesranged from 78 to 93% and from 0 to 38%, respectively. Similarity between the isolates ofC. acutatumandC. gloeosporioidesranged from 0 to 26.5%. A+T-rich DNA grouped theC. acutatumisolates separately from those ofC. gloeosporioides, corresponding to ap-PCR analyses. Artificial inoculations with nine representative isolates on almond, apple, avocado, mango and nectarine fruit showed a variation in levels of infection. TheC. gloeosporioidesisolates from almond grew more slowly, causing significantly smaller lesions on all inoculated fruit than the other isolates. TheC. acutatumisolates from apple and peach caused similar levels of infection on all fruit, but differed significantly from theC. gloeosporioidesisolate from apple. Variation in lesion size was also observed with isolates ofC. gloeosporioidesfrom apple, avocado and mango for most fruit inoculations.  相似文献

The aim of these studies is to develop a method for early detection ofRigidopoms lignosus (Basidiomycete, Polyporaceae), the causative agent of white root disease of rubber tree. Two polyclonal sera were produced against soluble mycelial proteins of twoR. lignosus isolates, one from Africa (FCI2), the other from Asia (FID2). The specificity of the antisera was tested using isoelectric focusing (IEF)/Western-blot and DAS-ELISA. The two sera recognized all 20R. lignosus isolates from various geographical origins. The banding patterns obtained by Western-blot enabled a distinction to be made between isolates from Africa and those from Asia. In DAS-ELISA and Western-blot analyses, strong cross reactions were observed withR. ulmarius. Only slight reactions were observed in Western-blot analysis toR. lineatus andP. noxius, both causative agents of root rot inHevea. These cross reactions were not observed under our DAS-ELISA analysis conditions. Finally, no cross reactions were obtained with 9 otherPolyporaceae orHevea root pathogen species. The sensitivity threshold of the DAS-ELISA method was 5 ng ml–1 ofR. lignosus protein. An initial approach to using the DAS-ELISA test for the detection ofR. lignosus in infected plants was carried out on artificially inoculated root samples. The DAS-ELISA protocol enabled detection ofR. lignosus in the root systems of diseased plants. Moreover, no cross reaction was observed with healthy plant extracts.  相似文献

Fusarium graminearum andF. culmorum are capable of infecting winter cereals at all growth stages. From natural field epidemics of wheat head blight and rye foot rot, three fungal populations were collected with 21, 38 and 54 isolates, respectively; their aggressiveness was analyzed in comparison to collections ofF. graminearum (25 isolates) andF. culmorum (70 isolates) that represent a wide range of geographical locations and host species. All isolates were tested for aggressiveness on young plants of winter rye in the greenhouse and scored for disease severity on a 1–9 scale. Disease ratings of individual isolates ranged from 1.5 to 5.7 indicating quantitative variation of aggressiveness. Genotypic variance was highest in the twoFusarium collections. No substantial difference was found in the amount of genotypic variation betweenF. graminearum andF. culmorum. Individual field populations revealed 57–66% of the total genotypic variation of the collections. This implies a high degree of diversity of aggressiveness within single field populations ofF. graminearum andF. culmorum causing natural epidemics.  相似文献

Thirty barley landraces collected from Morocco in 1985 and 1989, and held in the Polish Gene Bank, IHAR, Radzików, Poland, were screened for resistance to powdery mildew. Fifteen tested landraces (50%) showed powdery mildew resistance reactions and 24 single plant lines were selected. Eighteen lines originating from 13 landraces were tested with 17 isolates of powdery mildew and another six lines originating from six landraces were tested with 23; the isolates were chosen according to their virulence spectra observed on the ‘Pallas’ isolines differential set. Three lines (E 1090-2-2, E 1110-3-2 and E 1077-1-1) showed resistance to all powdery mildew virulence genes prevalent in Europe. In 21 lines, unknown genes alone or in combination with specific ones were detected. Five different resistance alleles(Mlat, Mlal, Mla3, Mlg andMl(CP)) were postulated to be present in the tested lines, alone or in combination:Mlat was postulated to be present in nine (~38%) lines;Mlg andMl(CP) in two lines, andMla1 andMla3 in one tested line each. The use of newly identified sources of resistance in barley breeding as a means of controlling powdery mildew is discussed.  相似文献

Four-hundred-sixty-eightFusarium andFusarium-like isolates were obtained from crowns and subcrown internodes of winter wheat grown in Erzurum, Turkey. Of these isolates, 34.8% wereFusarium acuminatum, 32.3% wereF. equiseti, 16.9% wereF. oxysporum, 15.0% wereMicrodochium nivale (formerlyFusarium nivale), 0.6% wereF. tabacinum and 0.4% wereF. solani. In pathogenicity tests on wheat, the highest disease severity was caused by isolates ofM. nivale, whereas isolates ofF. acuminatum, F. equiseti, F. oxysporum andF. solani were slightly virulent; isolates ofF. tabacinum were nonpathogenic. Stemphylium vesicarium (teleomorph: Pleospora herbarum) is the causal agent of brown spot disease in pear. The species is also able to cause disease in asparagus, onion and other crops. Saprophytic growth of the fungus on plant debris is common. The objective of this study was to investigate whether isolates of S. vesicarium from different hosts can be pathogenic to pear. More than hundred isolates of Stemphylium spp. were obtained from infected pear fruits, dead pear leaves, dead grass leaves present in pear orchard lawns as well as from necrotic leaf parts of asparagus and onion. Only isolates originating from pear orchards, including isolates from dead grass leaves, were pathogenic on pear leaves or fruits in bioassays. Non-pathogenic isolates were also present in pear orchards. Stemphylium vesicarium from asparagus or onion, with one exception, were not pathogenic to pear. Analysis of the genetic variation between isolates using Amplified Fragment Length Polymorphism (AFLP) showed significant concordance with host plants. Isolates from asparagus or onion belonged to clusters separate from the cluster with isolates from pear or grass leaves collected in pear orchards. Multilocus sequencing of a subset of isolates showed that such isolates were similar to S. vesicarium.  相似文献
