犬瘟热是犬瘟热病毒(Canine distemper virus,CDV)感染犬和其他食肉动物造成的多发性、致死性传染病,本文从分子水平上探讨CDV遗传进化特性、变异情况与流行规律之间的关系.通过收集2002-2010年在中国地区分离的14株CDV野毒株、2006-2007年在全球各地分离的12株CDV野毒株以及从不同宿主分离的12株CDV野毒株和4株疫苗株,将其分为4组,将前3组野毒株分别与国内外正在使用的4株疫苗株的H基因进行遗传变异分析.分析发现,CDV野毒株与疫苗株间H蛋白基因的核苷酸相似性为86.2%~92.1%,其氨基酸相似性为89.1%~91.9%;H基因的584位的天冬酰胺糖基化位点是Asia-Ⅰ型CDV所特有的;H蛋白3555区域的非同义氨基酸替换概率较高.作者推测H蛋白抗原变异可能造成弱毒疫苗免疫效力降低,不能为某些CDV株的感染提供完全有效的保护.  相似文献

Raccoon dogs (Nyctereutes procyonoides) are widespread and common in South Korea. In 2011, we obtained serum samples from 102 wild raccoon dogs to survey their exposure to canine distemper virus (CDV). Forty-five of the 102 animals (44.1%) were seropositive. Field cases of canine distemper in wild raccoon dogs from 2010 to 2011 were investigated. Fourteen cases of CDV infection were identified by a commercially available CDV antigen detection kit. These cases were used for virus isolation and molecular analysis. Sequence analysis of hemagglutinin genes indicated that all viruses isolated belonged to the Asia-2 genotype. H protein residues which are related to the receptor and host specificity (residues 530 and 549) were analyzed. A glutamic acid (E) residue is present at 530 in all isolates. At 549, a histidine (H) residue was found in five isolates and tyrosine (Y) residue was found in 6 isolates. Our study demonstrated that CDV infection was widespread in wild raccoon dogs in South Korea.  相似文献

为了解近年来犬瘟热病毒(canine distemper virus,CDV)在中国毛皮动物主要养殖区的流行情况及遗传变异情况,本试验于2012-2014年从山东、河北、辽宁收集疑似犬瘟热的水貂、狐狸和貉病料,经犬瘟热抗原检测试纸条检测和RT-PCR检测为阳性后,克隆测序了15株CDV F蛋白信号区(Fsp)基因序列.序列分析结果显示,15株CDV野毒株Fsp基因核苷酸序列和氨基酸序列的相似性均为93.6%~100.0%,与疫苗株相似性为80.7%~81.7%.基因系统进化分析结果显示,15株野毒株均属于中国当前流行的Asia-1基因型.氨基酸序列比对分析结果显示,Fsp蛋白在氨基酸3-14、16-37、47-67位等处存在高变异.通过对当前流行CDV野毒株Fsp基因序列分析,结果表明该Fsp基因区具有较高的变异率,可作为CDV基因分型的依据,同时本试验结果为毛皮动物犬瘟热的防控提供了参考依据.  相似文献

Ten wild masked palm civets infected with canine distemper virus (CDV), captured in Japan from 2005 to 2007, were histopathologically and phylogenetically analyzed. Phylogenetic analysis based on the amino acid sequences of the H protein of two CDV isolates from masked palm civets revealed that the two isolates were classified into the clade of recent isolates in Japan. Histopathologically marked lesions of virus encephalitis were present in the brain, whereas gastrointestinal lesions were absent or at a mild degree. The distribution of the lesions resembles that of recent CDV cases in dogs. Therefore, recent CDV infections in masked palm civets could be caused by recently prevalent CDV in dogs. The possibility of the masked palm civet as a spreader of CDV among wildlife is also discussed.  相似文献

We sequenced the hemagglutinin (H) genes from four canine distemper virus (CDV) isolates obtained from three dogs and a marten in Korea. These sequences were included in subsequent H gene-focused phylogenetic tree analysis of 89 CDV strains. This analysis revealed eight clades designated as EU1, EU2, EU3, NA1, NA2, Asia 1, Asia 2 and Vaccine. Three of the Korean isolates (97Jindo, 98Marten and 07D111) occurred in the Asia 2 group that also contains many Japanese CDV strains isolated in 1998. The remaining Korean strain (07Q72) fell into the Asia 1 group. The 21 H protein sequences of 25 Asia 1 strains are generally predicted to bear nine potential N-linked glycosylation sites. In contrast, the 9 H protein sequences of 12 Asia 2 strains had eight potential N-linked glycosylation sites. The remaining strains had six (98Marten and 07D111) and seven (97Jindo) potential N-linked glycosylation sites.  相似文献

Canine distemper virus (CDV) is a highly contagious viral pathogen causing lethal disease in dogs and other mammalians. A high degree of genetic variation is found between recent CDV strains and the old CDV isolates used in the vaccines and such genetic variation is regarded as a possible cause of the increasing number of CDV-related diseases in dogs. The H gene shows the greatest extent of genetic variation that allows for distinction of various lineages, according to a geographical pattern of distribution and irrespective of the species of identification. In the present study, hemagglutinin (H) genes obtained from field strains detected from clinical specimens of Italian dogs were analyzed genetically. Phylogenetic analysis revealed that a homogeneous group of CDV strains is widespread in Italian dogs, all which are included into the European lineage. Unexpectedly, strains 179/04 and 48/05 clustered along with CDVs of the Arctic lineage, the highest identity being to strain GR88 (98.0 and 98.4%aa, respectively). The full-length sequence of a red fox CDV strain, 207/00 was also determined and analyzed. The H protein of the fox CDV strain was unrelated to strains within the major European lineage. These results suggest that at least three different CDV lineages are present in Italy.  相似文献

为分析当地非典型犬瘟热病毒(CDV)核衣壳蛋白(N)基因的序列特征及其表达产物的抗原性,根据已发表CDV的N基因序列设计引物,用RT-PCR方法从引起非典型症状的CDV细胞培养物中扩增N基因,进行克隆和序列分析,结果表明:该非典型CDV的N基因与已发表的12个CDV强毒株的核苷酸序列和氨基酸序列同源性分别在96.6%~99.2%和97.9%~99.4%之间,与已发表的4个CDV疫苗弱毒株的同源性分别在93.2%~93.6%和96.4%~97.5%之间;在N基因系统发育进化树上,非典型CDV与12个强毒株处在同一亚群,而且与9个中国分离毒株的亲缘关系近于3个国外毒株。N基因在大肠杆菌中表达的重组N蛋白的分子量为62 ku,主要以包涵体的形式存在;用western blot分析,重组N蛋白可与CDV阳性血清发生特异性反应;以纯化的重组N蛋白为抗原建立的CDV抗体间接ELISA检测方法具有良好的特异性。  相似文献

Two pairs of primers were prepared, both localized within the sequences of the nucleoprotein gene (NP) of canine distemper virus (CDV). A number of experiments were done to optimize the conditions of RT-PCR and nested PCR methods. The nucleic acids of the Onderstepoort, Rockborn, Snyder Hill and Lederle strains of CDV could be detected with these primers. However, they did not react with the sequences of the Edmonston strain of the measles virus. The detection limit for RT-PCR was 10 TCID50 and for nested PCR 0.1 TCID50 of CDV. The RT-PCR was able to demonstrate the nucleic acid of CDV in the blood of all seven puppies vaccinated with a modified live virus. Blood samples of 23 dogs clinically suspected of distemper were examined by RT-PCR combined with nested PCR, and the results were compared with the detection of the CDV antigen in the smears from the mucous membranes by the direct immunofluorescence (IF) test. Of the 23 dogs, 12 were positive in nested PCR, six in the IF assay, and only two in single RT-PCR. It is concluded that nested PCR seems to be the most sensitive method for ante-mortem diagnosis of canine distemper, especially in its subacute or chronic forms.  相似文献

Canine distemper (CD) is a contagious disease, which can damage the immune system, respiratory system, digestive system, and even nervous system, leading to systemic pathological changes, and has a huge threat to pet dogs, fur animals, etc. At present, the commonly used colloidal gold test cannot effectively distinguish vaccine immunity from animal natural infection. To establish an efficient and accurate detection method for identifying wild strains and vaccine strains of canine distemper virus(CDV), the whole genome of six canine distemper wild strains isolated from dogs and three widely used CDV vaccine strains collected from Wuhan area were sequenced. After comparing and analyzing the amino acid and the base sequences, the H gene was determined as the target gene for AS-PCR primer design. By genotyping the H gene, it was found that the prevalent CDVs in Wuhan were all Asia-Ⅰ, while the vaccine strain Y2 was America-I, and the vaccine strains Y1 and Y3 were both America-Ⅱ. Comparing the CDV-H gene sequences of 179 Asia-Ⅰtypes (6 wild-type strain samples + 173 GenBank Asia-Ⅰtype stains) and 3 vaccine strains, using AS-PCR technology (3'mismatch) to design a pair of primers. It can effectively distinguish CDV Asia-I wild strain and vaccine strain. The upstream primer sequence is 5'-TTAATATAATAATGACAGTG-3', and the downstream primer sequence is 5'-CCTCAAGGGGCACA-3'. The results showed that the primer had a strong specificity and the wild strains could amplify an 894 bp fragment, while the vaccine strains could not. There are 9 more regular bases (amino acids) variation sites in H gene of wild strain, and the 277th amino acids of all vaccine strains are Asparagine. These mutations may lead to an increase in N-glycosylation sites, which will have an impact on the virulence of canine distemper virus vaccine strains. The AS-PCR method established in this study can effectively distinguish the canine distemper vaccine and Asia-Ⅰwild strains.  相似文献

The C-terminal part of the nucleocapsid protein gene of 13 canine distemper virus (CDV) isolates from Thailand, were analyzed. The nucleotide sequences were assigned to two clusters; cluster A exhibited a high degree of homology with the vaccine strain Onderstepoort, 99.10 and 97.61%, respectively, in the two isolates examined. Cluster B appeared closely related to virulent strains registered in the GeneBank database and to the virulent reference strain (A75/17); a total of 11 samples were analyzed, with 94.63-99.10% homology at the same position. The deduced amino acid sequences correlated with the two-nucleotide sequence clusters. However, there was no association among the CDV groups with histories of vaccination, sex, ages, clinical findings and evidence of viral antigen in tissues.  相似文献

Sequence and phylogenetic analyses of three isolates of canine distemper virus (CDV) isolated from three dogs with a vaccination history were compared with the same analyses of vaccine virus isolated from a vaccine used for dogs. The three dogs showed clinical signs of a recent major type of CD in Japan, including oculonasal discharge and diarrhea, and pathological findings including non-suppurative encephalitis, pneumonia, mild gastroenteritis and lymphoid depletion. Inclusion bodies were in the stomach without inflammation and encephalitis was without clinical signs. One of the highest titers of CDV in different organs of the three dogs was commonly systemic lymphatic organs, including the spleen, lymph nodes and tonsils. New isolates of CDV joined to the clades of the Asia 1 group that is far from the vaccine group. These results surely indicate that wild strains of CDV from dogs with a vaccination history were not reversed vaccine virus, and that the dogs showed characteristics of recent CD in Japan.  相似文献

犬瘟热是一种接触性传染病,可侵害免疫系统、呼吸系统、消化系统,甚至神经系统,导致全身性的病理变化,对宠物犬、毛皮动物等存在巨大威胁。目前常用的胶体金检测法不能有效区分疫苗免疫与动物自然感染。为建立一种高效准确的鉴别犬瘟热病毒野毒株和疫苗株的检测方法,本试验对从武汉地区收集已确诊犬瘟热的6只犬分离得到的野毒株以及3株广泛使用的CDV疫苗株进行全基因组测序,从氨基酸水平和碱基水平比对分析后,确定H基因为AS-PCR引物设计的靶基因。通过对H基因进行分型,发现武汉地区流行的CDV均为Asia-Ⅰ型,而疫苗株Y2为America-I型,疫苗株Y1和Y3均为America-Ⅱ型。对比179株Asia-Ⅰ型(6株野毒株样品+173株GenBank Asia-Ⅰ型)与3株疫苗株的CDV-H基因序列,采用AS-PCR技术(3'端错配)设计出1对能有效区分犬瘟热Asia-Ⅰ型野毒株和疫苗株的特异性引物,上游引物序列为5'-TTAAATGATAATGACATAGTG-3',下游引物序列为5'-CCTGGCAAGGCAAGA-3'。结果显示该引物有较强的特异性,6株样品野毒株均可扩增出长894 bp的片段,疫苗株不能扩增,且野毒株的H基因上存在9个较为规律的碱基(氨基酸)变异位点,而疫苗株在第277位氨基酸上均为天冬酰胺,这些变异可能导致N-糖基化位点的增加,从而对犬瘟热病毒疫苗株的毒力产生影响。本研究建立的AS-PCR方法能有效区分犬瘟热疫苗和Asia-Ⅰ型野毒株。  相似文献

犬瘟热(CD)是由犬瘟热病毒(CDV)引起的一种急性、高度接触性传染病,流行范围广,发病率、致死率高,临床症状多样。CDV感染宿主广泛,所有日龄的犬都有可能感染。CDV属于副黏病毒科麻疹病毒属,有囊膜包裹的单股负链线性RNA病毒。CDV基因组编码6种蛋白:核衣壳(N)蛋白、磷(P)蛋白、基质膜(M)蛋白、融合(F)蛋白、血凝素(H)蛋白和大(L)蛋白。N、P和L蛋白与病毒复制有关;M蛋白与病毒的装配和出芽有关;F、H蛋白在病毒的侵染过程中起到关键作用。近年来,随着我国宠物业、毛皮经济养殖业的迅速发展,CD在我国的发病率有升高的趋势。论文对CDV分子生物学研究进展进行归纳总结。  相似文献

The nucleotide sequences of the phosphoprotein (P) of canine distemper virus (CDV) strains isolated between 1992 and 1996 in Japan were determined. This is the first report of the complete sequences of the P genes of recently prevalent CDV strains. The deduced amino acid sequences of the P, C and V proteins showed that in the new Japanese isolates, these proteins have approximately 93%, 90-91% and 92% identities with those of the Onderstepoort vaccine strain, respectively. The predicted functional regions were conserved. RNA editing resulting in a shift to the open reading frame (ORF) of the V protein was shown to occur with the same efficiency in both the field isolates and vaccine strain.  相似文献

Canine Distemper is a highly contagious viral systemic disease that affects a wide variety of terrestrial carnivores. Canine Distemper virus (CDV) appears genetically heterogeneous, markedly in the hemagglutinin protein (H), showing geographic patterns of diversification that are useful to monitor CDV molecular epidemiology. In Mexico the activity of canine distemper remains high in dogs, likely because vaccine prophylaxis coverage in canine population is under the levels required to control effectively the disease. By phylogenetic analysis based on the nucleoprotein (N) and on the H genes, Mexican CDV strains collected between 2007 and 2010 were distinguished into several genovariants, all which constituted a unique group, clearly distinct from field and vaccine strains circulating worldwide, but resembling a CDV strain, 19876, identified in Missouri, USA, 2004, that was genetically unrelated to other North-American CDV strains. Gathering information on the genetic heterogeneity of CDV on a global scale appears pivotal in order to investigate the origin and modalities of introduction of unusual/novel CDV strains, as well as to understand if vaccine breakthroughs or disease epidemics may be somewhat related to genetic/antigenic or biological differences between field and vaccine strains.  相似文献

To achieve proper diagnosis of dogs based on acute clinical symptoms and poorly preserved field samples taken from animals that died due to canine distemper (CD), a new differential diagnostic test has been developed based on polymerase chain reaction (PCR). In this study, more than 150 samples collected from dogs showing respiratory, gastrointestinal and neurological signs suggesting canine distemper virus (CDV) infection were examined. The samples consisted of urine, blood and nasal swabs collected from clinically ill patients, sent to our laboratory by clinicians from various veterinary clinics throughout Hungary. Various organs collected during the necropsy of dogs with pathological changes that suggested CDV infection were also included. Three distinct PCRs were designed. For diagnostic purposes, a primer pair specific to a 409 bases-long segment within the conservative part of the large polymerase region (L) of the CDV genome was designed. Using this test, out of the 150 analyzed samples, 46 (30.66%) proved to be positive for CDV, indicating that CDV still represents a high risk to the canine population in Hungary. For the phylogenetical analysis, a primer pair that completely encompasses the hemagglutinin (H) gene of the CDV genome was designed. The amplicons of this region were sequenced in both directions using the appropriate primers. Our results indicate that several different CDV genotypes are currently present in Hungary. Nine of the analyzed Hungarian strains turned out to belong to the so-called Arctic group of CDVs, and were most closely related to non-European strains from North America, China and Greenland, as well as to the phocine distemper virus 2 (PDV-2) isolated from Baikal seals (Phoca sibirica). One of the Hungarian strains showed high similarity to other European isolates from Denmark, Germany, Italy and Turkey, as well as to other isolates from geographically more distant regions, such as the USA. Three Hungarian strains seem to join a new cluster that is formed by only a couple of strains, one isolated from a mink in Denmark, and another from a dog in North America. Using a third set of primers, a restriction fragment length polymorphism (RFLP) assay has also been designed for the fast and reliable differentiation of the wild-type CDVs from the vaccine strains.  相似文献

A 287 bp fragment from the middle region of the nucleocapsid protein of canine distemper virus (CDV) was amplified from the conjunctival samples of distemper-infected dogs and was cloned into pRSET B vector. The recombinant protein was expressed as a 16-kDa-fusion protein with histidine tag in E. coli. Sera of distemper-infected and vaccinated dogs contained IgG antibodies against the purified recombinant protein as observed by enzyme linked immunosorbent assays (ELISA) and showed a strong correlation (r = 0.882, p < 0.0001 at 95% CI) and good agreement (kappa = 0.718) with the conventional tissue culture viral antigen based ELISA. Further, the results of recombinant protein based ELISA and Western blotting with the sera from the infected and vaccinated dogs correlated well (kappa = 0.8226). These findings recommend the use of the recombinant protein in the serodiagnosis of canine distemper virus infection in dogs.  相似文献

Austrian field cases of canine distemper (14 dogs, one badger [Meles meles] and one stone marten [Martes foina]) from 2002 to 2007 were investigated and the case histories were summarised briefly. Phylogenetic analysis of fusion (F) and haemagglutinin (H) gene sequences revealed different canine distemper virus (CDV) lineages circulating in Austria. The majority of CDV strains detected from 2002 to 2004 were well embedded in the European lineage. One Austrian canine sample detected in 2003, with a high similarity to Hungarian sequences from 2005 to 2006, could be assigned to the Arctic group (phocine distemper virus type 2-like). The two canine sequences from 2007 formed a clearly distinct group flanked by sequences detected previously in China and the USA on an intermediate position between the European wildlife and the Asia-1 cluster. The Austrian wildlife strains (2006 and 2007) could be assigned to the European wildlife group and were most closely related to, yet clearly different from, the 2007 canine samples. To elucidate the epidemiological role of Austrian wildlife in the transmission of the disease to dogs and vice versa, H protein residues related to receptor and host specificity (residues 530 and 549) were analysed. All samples showed the amino acids expected for their host of origin, with the exception of a canine sequence from 2007, which had an intermediate position between wildlife and canine viral strains. In the period investigated, canine strains circulating in Austria could be assigned to four different lineages reflecting both a high diversity and probably different origins of virus introduction to Austria in different years.  相似文献

Canine distemper virus (CDV) causes a multisystemic fatal disease, briefly named as distemper, in domestic and wild animals. Molecular characterization studies serve to identify local strains, accordingly, helps to determine the scope of vaccination in prevention of distemper. We aimed with this study to update the molecular status of CDV in domestic dogs in Turkey.Sequence analysis of the H gene revealed that novel Turkish sequences formed a separated clade in Arctic-like lineage. Italian clade which mainly included strains originated from wild canid or non-canid localized nearly to novel Turkish clade. Codons 530th and 549th determining the affinity of domestic or wild animals to distemper were Asparagine and Tyrosine, respectively.This report presented the presence of CDV strains belonging to Arctic-like lineage for the first time in domestic dogs in Turkey. The findings pave the way for the reassessment of the circulation and geographical shifting of Arctic-like lineages of CDV.  相似文献
