More than 80% of the milk stored in the udder, i.e. the alveolar fraction, only available after milk ejection is induced by the release of oxytocin and myoepithelial contraction. Milk ejection is induced by tactile teat stimulation, either manually or by the milking machine. The time from the start of a tactile stimulation until the occurrence of milk ejection spans from 40 s up to more than 2 min and increases with decreasing degree of udder filling. Therefore, cows need a longer pre-stimulation in late stages of lactation or if the milking is performed shortly after the previous milking whereas in full udders, pre-stimulation is less important. Milk ejection can be disturbed under several conditions, for example, during milking in unfamiliar surroundings or for several weeks after parturition in many primiparous cows. We found that a disturbed milk ejection is due to a reduced release or complete absence of oxytocin from the pituitary. During milkings with disturbed milk ejection in unfamiliar surroundings, the concentrations of cortisol and beta-endorphin were elevated. While exogenous glucocorticoids could not induce the disturbance, morphine caused inhibition of oxytocin release during milking. However, spontaneously occurring disturbance of milk ejection could not be abolished by the opioid-antagonist naloxone. On the other hand, severity of disturbed milk ejection and coping capacity towards novel milking environment was inversely related to the cortisol release in response to ACTH, i.e. adrenal cortex activity. Therefore, the susceptibility of individual cows to the inhibition of oxytocin release and milk ejection could be predicted by an ACTH challenge test.  相似文献

The objectives of this study were first to show adrenocortical response to a long‐acting adrenocorticotropic hormone preparation (tetracosactide acetate zinc suspension) (ACTH‐Z) and its effect on adrenocortical function in beef cows ( Experiment 1 ) and second to apply the ACTH‐Z challenge in dairy cows based on cortisol concentrations in milk collected at routine milking ( Experiment 2 ). In Experiment 1 , four beef cows in luteal phase were challenged with ACTH‐Z, and plasma cortisol concentrations were determined for 48 h after the injection at 30‐min to 2‐h intervals. A rapid ACTH test was conducted 3 days before and 2 h after the completion of ACTH‐Z injection for 48 h to investigate the effect on adrenocortical function. Plasma cortisol concentrations increased significantly 30 min after ACTH‐Z injection (p < 0.001), and the high cortisol levels were maintained for approximately 10 h after the injection. In Experiment 2 , eight dairy cows were subjected to ACTH‐Z challenge 1–2 weeks and 4–5 weeks post‐partum. Blood and milk samples were taken at morning and afternoon milking. All the cows showed a significant increase in cortisol concentrations in plasma as well as in skim milk 8 h after ACTH‐Z injection 1–2 weeks and 4–5 weeks post‐partum (p < 0.001). There was a significant correlation between plasma and skim milk cortisol concentrations 8 h after ACTH‐Z challenge (r = 0.74, p < 0.001). The results obtained in this study suggest that elevated levels of plasma cortisol are maintained for approximately 10 h after ACTH‐Z treatment without adverse effect on adrenocortical function and a long‐acting ACTH‐Z challenge based on cortisol concentrations in milk, which were collected at the morning and the afternoon milking, can be a useful tool to monitor adrenocortical function in cows.  相似文献

The acyclic postpartum period in cows is prolonged due to frequent suckling when compared with twice daily milking. In conventional milking systems twice daily milking is common, while in automatic milking systems (AMS) three or more milkings are performed in early lactation. In this study, the hypothesis was tested that an increased milking frequency in AMS causes a delay in resumption of ovarian cyclicity postpartum and increases days open in German Fleckvieh cows (n = 124). Calvings were equally distributed throughout the 1-year experimental period. All cows were housed in one barn. Sixty-three cows were milked in an AMS (AC) and 61 cows were twice daily milked in a conventional milking parlour (PC). Forty-two AC and 36 PC remained in the herd until conception. After parturition, milk samples were analysed for progesterone twice weekly to record the ovarian activity until pregnancy was diagnosed. The first and the second ovulation were determined by the progesterone profile of each individual cow. The mean milking frequency during the first 3 weeks after parturition was higher in AC (3.17 +/- 0.09 milkings/day) than in PC (milked twice daily). The individual mean milking frequency of AC for the first 3 weeks of lactation ranged from 2.0 to 5.9 daily milkings. The time span from parturition until the first and second ovulation did not differ between AC and PC. However, first ovulation was observed earlier in primiparous (23 +/- 1 days) than in multiparous cows (29 +/- 1 days, P < 0.05). Within AC no relationship between milking frequency and duration of the acyclic period was observed. Days open did not significantly differ between AC and PC and between primiparous and multiparous cows. In conclusion, the increased milking frequency in AMS did neither cause a delay in resumption of ovarian cyclicity postpartum nor did it increase days open.  相似文献

Changes in ACTH challenge test characteristics in dairy cows changing their physiological status at different lactational stages and different feeding levels were not investigated in terms of repeatability yet. In 23 multiparous Holstein cows (10 cows fed a sole fresh herbage diet without concentrate, 13 cows fed with concentrate), three ACTH challenge tests were performed: once during pregnancy shortly prior to drying off ( T1 ), and in week 3 ( T2 ) and 8 ( T3 ) after parturition. Test characteristics were correlated to performance and metabolic parameters: DMI, BW, energy balance (EB), plasma concentrations of free fatty acids (NEFA) and beta‐hydroxybutyrate (BHB). Basal plasma cortisol concentrations were higher at T1 compared with T2 and T3 (p < .05). The adrenal cortex sensitivity (expressed as total AUC (AUCt) of cortisol response after ACTH application) was lowest at T2 compared with T1 and T3 (p < .05). Ranking of the individual animals’ responses was not repeatable between time points of the ACTH tests. Enhancing the energy deficiency during early lactation by omission of concentrate did not affect baseline cortisol concentrations in plasma, but decreased peak height at T2 (p < .05). Baseline plasma cortisol concentrations were positively correlated with cortisol peak values after ACTH application, previous lactation performance, milk yield and BW (p < .05). The AUCt was positively correlated with baseline cortisol concentrations, EB and DMI. Cortisol release after ACTH injection was lower in animals with high plasma concentrations of NEFA, BHB and with higher contents of fat and free fatty acids in milk (p < .05). Cortisol peak height after ACTH administration was higher in cows with a more positive EB, higher DMI and lower plasma concentrations of NEFA and BHB. In summary, cortisol responses to ACTH challenges in this study were not repeatable in dairy cows changing their physiological status.  相似文献

Recent studies in Japan of the effect of automatic milking systems (AMS) on the behavior of dairy cows have confirmed the labor-saving aspect of AMS, but have not solved some of the problems affecting milking traits. The influence of AMS on milking was examined in 39 Holstein cows at Iwate Agricultural Research Center from June 1998 to December 1999, by measuring and recording the yield per milking, milking finish time, milking frequency, and daily yield of milking. Four milking traits were analyzed separately as the objective variable in a linear model, with parity, days in milking, month after installation of the AMS as independent variables. Primiparous cows milked more frequently in the middle and late stage of lactation, yielded less per milking and less per day in the early stage of lactation than multiparous cows with the AMS. Lower frequencies were observed at noon and midnight for cleaning of the bulk tank. By contrast, higher milking frequency in the forenoon was shown. The yield per milking rose and declined, following a normal lactation curve. However, milking frequency rose slowly, and declined gradually in the late stage of lactation. After installing a one-way gate, the cows milked more frequently, with a lower yield per milking and a greater daily milk yield, than before installation.  相似文献

Administration of 6IU synthetic ACTH1-24 intravenously to six Holstein-Friesian cows resulted in a cortisol peak concentration after 1 hour of 148 +/- 34.2 ng/ml. Basal plasma cortisol concentration (4.84 +/- 0.83 ng/ml) was reached 5 hours after ACTH injection. Until 7 days after ACTH administration no effect on milk yield was recorded. So it is concluded that a dose of 6 IU ACTH1-24 is sufficient for a conspicuous release of cortisol without any alteration in milk production. This dose can be used as a standard test for the evaluation of adrenocortical function in lactating cows when administered intravenously at 9 a.m. and when plasma samples for cortisol assay are collected just prior to administration and at 10 a.m.  相似文献

Shortened and weakened estrous expressions could be one of the causes of poor heat detection rate. Non-specific acute stresses are assumed to depress expression of estrus by an increase of plasma progesterone which may originate from the adrenal cortex. The objective of the present study was to examine whether the adrenal cortex can secrete significant amounts of progesterone in response to exogenous adrenocorticotropic hormone (ACTH) in lactating cows. Four cows had estrus synchronized and were ovariectomized in the luteal phase. The cows were given 25 IU ACTH through an indwelling catheter 5 h after catheterization. Blood samples were collected at an interval of 30 min. In 3 of the 4 cows, plasma progesterone concentrations increased significantly 0.5-1.5 h after the first ACTH challenge with a mean peak value of 4.2 +/- 0.4 (S.D.) ng/ml. A similar response was also observed after the second ACTH challenge. Peak plasma progesterone concentrations in the 3 cows after first ACTH challenge were comparable with the progesterone values in the luteal phase of each cow. The results suggest that lactating cows have the capability to secrete a significant amount of progesterone from the adrenal cortex.  相似文献

The results of most studies show the beneficial effect of milking automation on production parameters of dairy cows, but its effect on fertility traits is debatable. Therefore, a study was undertaken to predict cow fertility – services per conception (SC) and calving interval (CI) – based on automatic milking system (AMS) data collected in the periparturient period subdivided into the second and first week before calving, 1–4, 5–7, 8–14, 15–21 and 22–28 days of lactation. SC and CI were predicted using daily indicators such as concentrate intake, number of milkings, cow box time, milking time, milking speed, colostrum and milk yield, composition, temperature and electrical conductivity. The study material was derived from the AMS management system and from the SYMLEK milk recording system. The analysis covered data for 16,329 milkings of 398 Polish Holstein-Friesian (PHF) cows, which were used in three AMS herds. The collected numerical data were statistically analysed by correlation analysis in parallel with decision tree technique (SAS statistical package). The present study showed that due to the low, mostly non-significant coefficients of correlation between AMS data collected between 2 weeks before and 4 weeks after calving, it is not possible to predict cow fertility based on single traits. It has been established that the decision tree method may help breeders, already during the postcalving period, to choose the level of factors associated with AMS milking, which will ensure good fertility of cows in a herd. The most favourable number of services per conception is to be expected from cows that were milked <1.6 times per day from 1 to 4 days of lactation and electrical conductivity of their colostrum did not exceed 69 mS during that time. In turn, shortest CI (366 days) will be characteristic of the cows whose average daily colostrum yield did not exceed 20.2 kg and their daily concentrate intake from 8 to 14 days of lactation was at least 5.0 kg.  相似文献

The objective of this study was to clarify the effect of different milking frequencies under an automatic milking system (AMS) on milk yield, plasma metabolite profiles and mammary arterial‐venous (A‐V) differences of milk precursors by mammary tissues in early lactation cows. Twelve Holstein cows were divided into two and four times milking frequency treatments by AMS after calving to 50 days postpartum. Cows were given a partial mixed ration ad libitum and a concentrate diet at every milking. Dry matter intake increased similarly in both treatments with advancing postpartum days. Milk yield was greater (P < 0.001) by 25% with four times milking, but milk composition was not affected by milking frequency. Body weight change was also not affected by milking frequency. Arterial concentrations of glucose and glutamate were lower (P < 0.05) for four times milking frequency. However, arterial concentration of nonesterified fatty acids did not differ between treatments. Although mammary A‐V differences of plasma concentration for most milk precursors did not differ between treatments, estimated plasma flow was higher (P < 0.05) for four times milking frequency. These results indicate that higher milking frequency may increase mammary uptake of milk precursors, whereas may not affect the extent of fat mobilization of early lactating cows from day 20 postpartum onward.  相似文献

Commercial dairy cows milked in a parlor system are packed close together in the holding area before milking. The present study examined the relationships of ‘looking up’ behavior with some other behaviors and the productivity of 1116–1153 cows from five farms. The individual identities of the cows looking up in the holding area were recorded at 5 min intervals during six intermittent afternoon milking sessions. Entrance into the milking parlor and the numbers of steps and kicks by cows while the milking person was attaching the milking cups, were recorded in six milking sessions. Flight responses in the pasture after milking were recorded over four days intermittently. The frequency of ‘looking up’ behavior weakly, but significantly correlated with flight starting distance (r = 0.10, P < 0.05), while the correlation with the number of step‐kicks during milking was not significant. As for productivity, lactation number (r = ?0.18, P < 0.001), milk yield (r = ?0.15, P < 0.001) and fat content (r = ?0.15, P < 0.001) were negatively correlated with the frequency of ‘looking up’ behavior. Age of cows was correlated with the frequency of ‘looking up’ behavior as well as lactation number (r = ?0.21, P < 0.001). Entrance order was positively correlated with the frequency of ‘looking up’ behavior (r = 0.15, P < 0.001). The ‘looking up’ behavior was observed more frequently in cows in their third or less lactation compared with cows which were in their fourth or greater lactation (P < 0.05). The lactation number of cows was correlated with their milk yield (r = 0.36, P < 0.001) and flight starting distance (r = ?0.21, P < 0.001). In conclusion, ‘looking up’ behavior shown by cows in the holding area before milking might be an indicator of low motivation for milking, mainly because of fear of humans, and an aversion to milking caused by insufficient experience in being milked.  相似文献

Three experiments examined the extent of behavioral interactions between housed dairy cows. In Experiment 1, 6 different cows were selected every other day from a dairy herd and allocated to 3 pairs for recording of feeding, standing, lying, and leg-raising behaviors. There was no difference in the between-pair and within-pair coefficients of variation for the behaviors, suggesting that the performance of these behaviors was independent of influences of other cows. Experiment 2 used 9 pairs of cows in a 3-period changeover design. The same behaviors were recorded as in Experiment 1, and data were used only from the last 2 periods, which ensured that cows had been together previously for at least one 48-hour period. There were again no differences in the between- and within-pair coefficients of variation when using behaviors as replicates, either for the first 2 hours, during which time no treatments had been applied, or the entire 4 hours, when some cows had been prevented from lying down and feeding. Experiment 3 investigated the consistency of between-cow associations for repeated observations of feeding, lying, and entering the milking parlor. Although there was evidence of a similar order of entry into the milking parlor between days, the number of feeding associations was similar to that predicted by a model of random behavior, and the pattern of lying associations also suggested a random pattern of associations. Across behaviors, cows were observed with specific partners more often than expected by chance. Thus although there was no evidence of interdependence of feeding, lying, or standing behavior, consistent associations were observed, particularly in parlor entry.  相似文献

本次调研共收集了华北地区99家奶牛场的用水及粪污处理情况数据进行了分析总结。结果表明,①85.9%的牛场以深井水作为水源,其中61.2%的牛场对用水进行消毒处理,43.5%的牛场安装有水表并记录用水情况。②在牛场用水中,牛的饮水量所占比例最高,达60%以上;其次为挤奶厅用水,约占20%。③夏季牛场头均饮水量、挤奶厅用水量、喷淋用水量均显著高于春秋冬季(P<0.05),夏季的头均总用水量比春秋冬季增加了33%(P<0.01)。随着牛场存栏量的增大,奶牛头均总用水量显著减少(P<0.05)。④年产奶量较高的牛场每产1kg牛奶饮水量、挤奶厅用水量显著降低(P<0.01),致使每千克产奶量的总用水量减少52%(P<0.01),用水效率提高。⑤随着养殖规模的增大,牛场雨污分离设施与干湿分离设施配备率分别显著增至62%、72%(P<0.05,P<0.01)。由此可得,存栏量越大、年产奶量越高,牛场每产1kg牛奶用水量越低,用水效率越高,而且配备的雨污分离设施、干湿分离设施比例越高,这些均有利于减少单位产奶量的粪污产生量及其处理压力。  相似文献

The purpose of this paper was to utilize the decision trees technique to determine the factors responsible for high monthly milk yield in Polish Holstein‐Friesian cows from 27 herds equipped with milking robots. The applied statistical method—the decision tree technique—showed that the most important factors responsible for monthly milk yield of dairy cows using robots were, in descending order of importance: milking frequency, lactation number, month of milking, and type of lying stall. At the same time, it has been ascertained that the highest monthly milk yield (47.24 kg) can be expected from multiparous cows kept in barns with a deep bedding that were milked more frequently than three times per day. On the other hand, the lowest milk production (13.56 kg) was observed among dairy cows milked less frequently than two times a day, with an average number of milked quarters lower than 3.97. The application of the decision trees technique allows a breeder to select appropriate levels of environmental factors and parameters that will help to ensure maximized milk production.  相似文献

In 57 Holstein cows where the dairy farm uses a milking parlor system, the somatic cell count (SCC) increased persistently in the bulk milk (monthly mean 52.3 x 10(4) cells/ml; range 21 to 94 x 10(4) cells/ml). We detected S. aureus in 24 (41.2%) of the 54 lactating cows and in 29 (12.8%) of 227 quarters of the 57 milking cows in the herd. A control program was implemented in an effort to eradicate S. aureus mastitis from this dairy farm. The control plan established improved handling of the lactating cows, improved milking procedures, dry-cow therapy, and culling of infected cows. The program was monitored for 3.5 years by frequent checkups on the rate of S. aureus infection, the SCC, and the changes in milk composition. Eighteen months after the control program was started, the rate of S. aureus infection in the quarter milk decreased dramatically, and no S. aureus isolates were found in the milk of the remaining cows. The SCC in the bulk milk of the herd dropped to a monthly mean of <20 x 10(4) cells/ml. In conclusion, the control program was effective for controlling persistent S. aureus mastitis in this dairy herd.  相似文献

The objective of this study was to describe the responses of the plasma progesterone and cortisol concentrations in ovariectomized lactating cows to low doses of adrenocorticotropic hormone (ACTH). The estrous cycles in 3 lactating cows were synchronized, and the cows were ovariectomized in the luteal phase. ACTH challenge tests were conducted at doses of 3, 6, 12 and 25 IU. Blood samples were collected at 30 min intervals, and the plasma progesterone and cortisol concentrations were analyzed by EIA. A concomitant rise in plasma progesterone and plasma cortisol was observed in cows treated with 12 IU or higher doses of ACTH. Significant increments in the plasma cortisol concentrations were observed at all doses of ACTH. The means (+/- SE) of the peak plasma progesterone concentrations after the 3, 6, 12 and 25 IU ACTH challenge tests were 0.6 +/- 0.1, 1.3 +/- 0.4, 1.5 +/- 0.3 and 2.4 +/- 0.3 ng/ml, respectively. The means of the peak plasma cortisol concentrations in the 3 cows after the ACTH challenge were 14.0 +/- 1.5, 17.0 +/- 2.5, 23.3 +/- 3.0, and 33.3 +/- 7.0 ng/ml, respectively. The effects of the doses, time after treatment, and their interaction on the plasma progesterone concentrations after the ACTH challenge were significant (P<0.01). Likewise, the effects of the doses, time after treatment, and their interaction on the plasma cortisol concentrations after the ACTH challenge were significant (P<0.01). The mean AUC values for the plasma progesterone and cortisol concentrations after the ACTH treatments were also significantly affected by the dose of ACTH (P<0.01 and P<0.05, respectively). A significantly positive correlation was obtained between the peak plasma progesterone and cortisol concentrations after different doses of ACTH (r=0.7, P<0.05). The results suggest that lactating dairy cows are capable of secreting a significant amount of adrenal progesterone, reaching up to the minimal concentration necessary to cause suppression of estrus in response to 12 IU ACTH (P<0.01). The concomitant plasma cortisol concentration was 23.3 ng/ml.  相似文献

Thirteen lactating dairy cows from a herd of 650 died over a 6-week period. Most animals were down in milk production at 1 milking and were found dead at the next milking. Two cows had elevated heart rate and enlarged mandibular lymph nodes. Two others had azotemia, elevated heart rate, hyperglycemia, and weight loss. Necropsy of 10 cows revealed hemorrhages on the intestinal serosa and epicardium, lymphadenopathy, interstitial nephritis, small intestinal hemorrhage, and interstitial pneumonia. Histopathology showed lymphocytic to lymphogranulomatous inflammation in the heart, spleen, kidney, lymph nodes, liver, lung, pancreas, and adrenal gland. Phlebitis was present in 2 livers. The lesions resembled those of hairy vetch toxicosis, but no vetch was being fed. Similar lesions have been reported with the feeding of citrus pulp. Citrus pulp was being fed to the lactating cows and had been added to the diet 6 weeks before the first death. The syndrome resolved with elimination of citrus pulp from the diet.  相似文献

Milk flow was recorded in 21 cows for three days after they were admitted to a large animal hospital. When the spontaneous flow of milk had stopped, a physiological dose (1 iu) of oxytocin was administered intravenously. Five of the cows were, in addition, treated with 0.35 mg of a long-acting analogue of oxytocin (carbetocin) one hour before the first milking after they were admitted. In the 16 cows not treated with carbetocin, only about 30 per cent of the total milk yield was released spontaneously on the first day, and the injection of 1 iu of oxytocin released approximately another 60 per cent of the total milk yield. On the second day, the proportion of the total milk yield released spontaneously increased and the fraction released after the injection of 1 iu oxytocin decreased. In contrast, the five cows treated with carbetocin released on average 94 per cent of the total milk yield spontaneously during the first milking.  相似文献

Adult dairy cows were treated with a range of doses (0.0125mg to 0.4mg) of synthetic ACTH1–24 and the plasma cortisol response was measured. Peak response was independent of dose. Higher doses of ACTH had a more prolonged effect such that the integrated response was dose dependent. Dose response curves were examined by regression analyses. Individual cows had a significant effect on y-axis intercept but not on the slope of the regression lines examined. A dose rate of 0.05mg ACTH iv was identified as a suitable dose rate for use in clinical assessment of adrenal cortex function, with 0, 50 and 120 min following ACTH being critical sampling times to identify the response.  相似文献

Farm managers' decision to cull dairy cows is based on the cows' milk production, history of disorder(s), and reproductive performance, each of which affects dairy cows' lifetime (herd life and productive lifespan). We investigated the relationships among the incidence of clinical mastitis (CM), the reproductive performance, and the culling rate. We also assessed the effects of these relationships on the lifetimes of dairy cows, using the records made before and after the introduction of an automatic milking system (AMS) at Hiroshima University Farm. Milk yield, CM incidence density, and culling rate of dairy cows increased after the AMS introduction. The CM incidence was associated with an elongation of the calving interval in cows with the same parity. CM in the 1st parity might have caused the reductions of the cows' lifetime and their parity at culling. A higher age at first calving (AFC) was associated with an increase in culling rate but did not lead to a significant decrease in lifetime. Investigations of the factors mediating CM in the 1st parity or AFC with CM incidence or culling rate in the later stages might contribute to the control of lifetime of dairy cows.  相似文献

Two directions can be distinguished in the development of high-tech milking equipment: 1) high-capacity milking parlors with a high throughput of cows per person per hour and 2) automatic milking systems in which manual labor is replaced by a milking robot. High-capacity milking parlors are developed in such a way that one operator is able to milk many cows, partly by automation and partly by optimization of available labor. In such parlors, one operator can milk up to 125 cows per hour. This means that there are only a few seconds available for udder preparation. In an automatic milking system, a robot takes over all manual labor during milking. Currently available systems have one robot arm working with one milking stall (one-stall system) or one robot arm working with more milking stalls (multiple-stall systems). Cows have to go to the automatic milking system voluntarily. Therefore, there is a large variation in milking intervals. Moreover, a large variation between milkings and between cows was observed in milk flow rate, machine-on time and udder preparation time. Both developments in high-tech milking have effects on the milk ejection. The small amount of time dedicated to udder preparation in high-capacity milking parlors has negative effects on the milk ejection, among others leading to more bimodal milk flow curves and longer machine-on time. In automatic milking systems, the variation in time between udder preparation and cluster attachment and in milking frequency might have an effect on milk ejection. Lactation physiology can play a role in solving the questions around milk ejection in high-tech milking systems. The introduction of high-tech milking systems makes decision support systems using sensors necessary. These systems should assist in detection of abnormal milk and mastitis. To a lesser extent, diseased cows need to be brought to the attention of the dairy farmer. Some sensors are currently available for this purpose, but they do not fulfill all demands. In the near future other sensors might be developed. It is important that this development is demand driven and not technology driven. Lactation physiology can play an important role in the determination of milk components useful for automatic detection.  相似文献
