We conducted a cross-sectional study from August 2003 to February 2004 to identify risk factors for bovine tuberculosis (BTB) in the Kafue basin of Zambia. We investigated a total of 106 herds of cattle for presence of BTB using the comparative intradermal tuberculin test (CITT) while an interviewer-administered questionnaire was used to gather epidemiological data on herd structure, management and grazing strategies. BTB prevalence at herd level was estimated and possible risk factors were investigated using the multiple logistic regression model. The true herd level prevalence of BTB was estimated at 49.8% (95% CI: 37.9, 61.7%). The logistic regression model showed that cattle herd BTB status was highly associated with area and husbandry practices. When compared to Kazungula, cattle herds in Blue Lagoon were more likely to test positive for BTB when other factors such as management practices were controlled (OR=10.5). In terms of grazing strategies, transhumant herds (TH) had higher odds (OR=3.0) of being positive compared to sedentary herds (OR=1.0). The results in this study provide preliminary information about potential risk factors that were found to be associated with BTB status in cattle.  相似文献

A retrospective cohort study of Irish cattle herds investigated whether the severity of a herd’s bovine tuberculosis (BTB) breakdown was a predictor of the hazard of a future BTB breakdown in that herd. Data on 10,926 herds not having had BTB in 1995 (the “non-exposed” group) were obtained using a 10% random sample from all herds without BTB in 1995. Data on 6757 herds that had a new BTB breakdown in 1995 (the “exposed” group) were obtained and categorized into five increasing exposure-severity classes based on the total number of standard reactors (to the single intra-dermal comparative cervical tuberculin test) detected during the breakdown. Exposed herds were deemed to be free of BTB after they passed a 6-month check test; non-exposed herds were deemed free as of the date of the first negative herd-test in 1995.

In the 5 years after 1995, 18% of the non-exposed herds had a BTB breakdown, whereas 31% of the exposed herds had a subsequent breakdown. Relative to the hazard for non-exposed herds, the hazard for the first future singleton standard reactor breakdown, was 1.6-times higher for exposed herds with only 1 standard reactor in 1995, and 1.8-times higher for those exposed herds with 4–8 standard reactors during the 1995 episode. When the outcome for future breakdowns was 2 or more standard reactors, the hazard ratios ranged from 1.6 for exposed herds with only 1 standard reactor in 1995 up to 2.9 in exposed herds with 8 or more standard reactors during the 1995 episode. The latter hazard ratio varied over time, decreasing to 1.7 after 3 years of risk. The hazard of a future BTB breakdown increased directly with number of cattle in the herd, a positive history of previous BTB in the herd, and the local herd prevalence of BTB. The presence of confirmed BTB lesions in reactor cattle was not predictive of the future breakdown hazard when the effects of other factors were controlled.  相似文献

Bovine tuberculosis (BTB) is endemic in African buffalo (Syncerus caffer) in the Kruger National Park (KNP). In addition to buffalo, Mycobacterium bovis has been found in at least 14 other mammalian species in South Africa, including kudu (Tragelaphus strepsiceros), Chacma baboon (Papio ursinus) and lion (Panthera leo). This has raised concern about the spillover into other potentially susceptible species like rhinoceros, thus jeopardising breeding and relocation projects aiming at the conservation of biodiversity. Hence, procedures to screen for and diagnose BTB in black rhinoceros (Diceros bicornis) and white rhinoceros (Ceratotherium simum) need to be in place. The Interferon-gamma (IFN-gamma) assay is used as a routine diagnostic tool to determine infection of cattle and recently African buffalo, with M. bovis and other mycobacteria. The aim of the present work was to develop reagents to set up a rhinoceros IFN-gamma (RhIFN-gamma) assay. The white rhinoceros IFN-gamma gene was cloned, sequenced and expressed as a mature protein. Amino acid (aa) sequence analysis revealed that RhIFN-gamma shares a homology of 90% with equine IFN-gamma. Monoclonal antibodies, as well as polyclonal chicken antibodies (Yolk Immunoglobulin-IgY) with specificity for recombinant RhIFN-gamma were produced. Using the monoclonals as capture antibodies and the polyclonal IgY for detection, it was shown that recombinant as well as native white rhinoceros IFN-gamma was recognised. This preliminary IFN-gamma enzyme-linked immunosorbent assay (ELISA), has the potential to be developed into a diagnostic assay for M. bovis infection in rhinoceros.  相似文献

We investigated the impact of targeted removal of badgers on the subsequent bovine tuberculosis (BTB) risk in cattle herds in county Laois, Ireland. The study period was 1989-2005. For each of 122 targeted badger-removal licenses (permit to remove badgers in the proximity of cattle herds undergoing a serious BTB episode), the herd number (index herd) for which the license was given was obtained. The herds in the proximity of the index herd were identified from another database. The main "exposure" in our study was the geographical location of herds relative to the area in which targeted badger removal was conducted. We categorized herds into five different exposure groups: herds were classified as non-exposed and denoted as group 0 (reference group) if they were located 500 m or more from the edge of any parcel of land of the index herd; group 1, was the index herds, group 2 the immediate (contiguous) neighbors of the index herd, group 3 herds were not immediate neighbors but within 150 m and group 4 herds were between 150 m and 500 m distance from the edge of any parcel of land of the index herd, respectively. We conducted a survival analysis (allowing multiple failures per herd) to compare the hazard of having a BTB episode in any of the four groups of exposed herds vs. the hazard in herds in the reference group. We controlled for other known risk factors as well taking into account a temporal component. Our analysis showed that the hazard ratio for the index herds (group 1) were non-significantly increased, indicating that there was no difference in the hazard of failing a BTB test (after the targeted badger removal was conducted) between index herds and reference herds. For the rest of the herds farther away from badger removal activities the hazards were lower than herds in areas not under badger removal. A cross-sectional study of bovine tuberculosis (BTB) was conducted in pastoral cattle herds in southern Ethiopia, from February to August 2008 using the comparative intradermal tuberculin test. The prevalence of BTB and the risk factors for having positive reactor herds were assessed in four pastoral associations in two districts of southern Ethiopia, namely Goro-Dola with 242 cattle in 16 herds and Liben with 231 cattle in 15 herds. A herd was considered positive if there was at least one reactor animal in a herd. The test results were interpreted based on the Office Internationale des Epizooties recommended 4-mm and a recently suggested 2-mm cut-off. The apparent individual animal prevalence of tuberculin reactors was 5.5% (95% confidence interval (CI), 4.0–8.0%) and 7.0% (95% CI, 5.0–10.0%), whereas the true prevalence estimate was 4.4% (95% CI, 0.8–8.0%) and 6.1% (95% CI, 2.6–9.5%), when using the 4-mm and the 2-mm cut-offs, respectively. The overall herd apparent prevalence of tuberculin reactor animals was 41.9% (95% CI, 24.9–60.9%) and 48.4% (95% CI, 30.2–66.9%) with the 4-mm and 2-mm cut-offs, respectively. A positive tuberculin test was associated with the age of animals and the main drinking water sources during dry seasons. In order to investigate the public health risks and the epidemiological importance of BTB in the area, we recommend to include other livestock species (camels and goats) as well as humans in future studies.  相似文献

We investigated the prevalence and risk factors to positive herd-level tuberculin reactivity between October 2003 to May 2004 to bovine tuberculosis (BTB) in the four transhumant districts of Uganda: three districts (Karamoja region) of nomadic transhumance cattle rearing (30 superherds and 1522 cattle), and one district (Nakasongola) of fixed-transhumance (7 herds and 342 cattle). We used the comparative intradermal skin-test, sampled 50 animals per superherd/herd, and considered herd positive if there was at least one reactor. Of the 30 superherds under nomadic transhumance, 60% (95% CI 41.4, 79) were tuberculin-test positive; of the 7 fixed herds, 14.3% (95% CI −20.7, 49.2) were tuberculin test positive. The true herd prevalence was estimated at 46.6%. Many risk factors were collinear. The final multivariable logistic-regression model included: recent introductions from market (OR = 3.4; 95% CI 1.1, 10.3), drinking water form mud holes during dry season (OR = 49; 95% CI 9.1, 262), and the presence of monkeys (OR = 0.08; 95% CI 0.0, 0.6) or warthogs (OR = 0.1; 95% CI 0.0, 0.3). No association was found between herd size or number of herd contacts with reactors; it was probably masked by the effect of high between-herd interactions. Provision of water from mud holes in dry river beds and introductions of new animals are risk factors that might be targeted to control BTB in transhumance areas.  相似文献

The objective was to investigate the spread of S. aureus in seven dairy herds. Milk samples were taken to determine mastitis pathogens and somatic cell count. An approved hygiene programme was established to control the spread of S. aureus from quarter to quarter. S. aureus isolates were differentiated by geno- and phenotyping to trace their spread within a herd.In two herds S. aureus showed a high prevalence, but were eliminated from the herds by the control programme. In these herds and a third herd typing results identified a particular type that was found much more frequently than all other types. The frequent types were repeatedly detectable during the study. In three other herds with a lower S. aureus prevalence and also in one herd with a long history of S. aureus and high prevalence subclinical mastitis was caused by several distinguishable S. aureus types. These types occurred simultaneously in the particular herd and showed little or no tendency to spread from quarter to quarter. They seemed to circumvent the control procedures resulting in a relatively high rate of new infected animals and therefore were hardly eliminated. The typing results and the clinical observations indicated that strains differed in their tendency to spread and their ability to infect udder quarters. In three herds typing methods identified a predominant type with the common epidemiological features of a contagious mastitis pathogen, while in the other herds the S. aureus type patterns were similar to that of environmental pathogens.  相似文献

The prevalence of bovine tuberculosis (BTB) infection in cattle was investigated in extensive and intensive production systems in three districts of northwestern Ethiopia. Single comparative intradermal tuberculin test (SCIDTT) was used in the study. The prevalence of BTB infection as determined by SCIDTT was 9.7% whereas the non-specific infection prevalence was 10.8%. In the extensive system the prevalence was 8.2% and 11.3%; under intensive system the prevalence was 22.1% and 6.3% for BTB and non-specific infections respectively. The prevalence of BTB was significantly higher in the intensive than extensive production systems. Of the 75 herds tested 41 (54.7%) had BTB infections, 68.9% of the BTB positive herds were in the extensive system and 40% of herds in the intensive systems (small dairy farms) had BTB infections.  相似文献

SUMMARY A study of the prevalence, spread and control of caprine arthritis-encephalitis virus (CAEV) in dairy goat herds in New South Wales (NSW) during 1986–1988 found that 56.8% of 1484 goats in 14 dairy herds were infected with CAEV. The prevalence of CAEV infection within most herds not implementing control measures increased during the study. At the end of the study, 59.7% of 1322 goats were infected. The prevalence of CAEV increased with age. Differences between breeds were less apparent. Within seven herds with a high standard of identification of goats, 149 of 812 goats seroconverted in an ELISA. Of these newly infected goats, 142 (95.3%) were > 1 yr of age and 96 (64.4%) were > 2 yr suggesting lateral spread of the virus. Most of the goats > 2 yr of age had been in the milking herd for a minimum of 3 to 6 months. The high seroconversion rate within the milking herd suggested that factors other than the ingestion of infected colostrum and milk before weaning were important for the spread of CAEV. Observations indicated that behaviour of goats, particularly reproductive behaviour among lactating does, and milking herd management practices are important in the spread of CAEV. A high density of livestock, poor livestock control and contamination of feed, water, equipment and personnel were implicated in transmission. Poorly functioning milking machines may also be involved. CAEV was eradicated from 3 herds by the implementation of strict control measures.  相似文献

Bovine herpesvirus type 1 (BHV-1) is an important bovine pathogen, exacerbating poor health and the productivity of cattle. The aims of this study were to detect the efficacy of vaccination programmes in lowering the seroprevalence of BHV-1 gE within the dairy herd and to follow the dynamics of the infection in non-vaccinated herds with uninfected heifers. A two-year longitudinal study was carried out on seven herds that were vaccinated, and in five herds with uninfected heifers without applying a control programme. After the start of the vaccination programme, calves born remained free from the virus. However, in one herd, 7 per cent (95 per cent CI 2 to 18) of these animals showed antibodies to BHV-1 two years after the first vaccination. A decline in BHV-1 antibody prevalence was found in vaccinating herds. Among the five herds not under the control programme, one experienced active virus spread, although one herd experienced self-clearance of the virus. In the herds with high BHV-1 prevalence, vaccinating all cattle from three months of age twice a year with a commercial inactivated marker vaccine efficiently protected offspring from becoming infected, and lowered the prevalence of BHV-1 within the herd. A small proportion of herds may experience self-clearance of the virus.  相似文献

The aim of this study was to provide baseline data on the population and herd Salmonella seroprevalence in sows and finishers. For the population estimates in 1996 and 1999 and the herd prevalences for sows and gilts, blood samples from swine vesicular disease (SVD) and pseudorabies monitoring programmes were used and tested in an indirect Salmonella enzyme-linked immunosorbent assay (ELISA). The herd prevalence for finishers was determined using blood samples collected at two slaughterhouses.The population prevalence for finishers in 1996 and 1999 was 23.7 and 24.5%, respectively, and for sows 40.5 and 60.4%, respectively. The prevalence in free range (FR) finishers was significantly higher (44.6%) than in intensively housed finishers in 1999, identifying a hazard group for possible extra pork and pork product contamination. Of 406 finishing herds, 9% were completely seronegative for Salmonella (cut-off OD%>10). Of these 406 finishing herds, 69.7% had Salmonella-status I (low prevalence), 21.7% status II (moderate prevalence) and 8.6% status III (high prevalence) (cut-off OD%>40). In 46 multiplying sow herds, 20 breeding sow herds and 20 matching replacement gilt herds, the average herd prevalences were 54, 44.4 and 19.3%, respectively. Two gilt herds were completely seronegative. The prevalence in the gilt herds was never higher than in the matching breeding sow herds. Agreement on methodology and calibration of ELISA tests would make these results comparable between countries and is a prerequisite for a co-ordinated and integrated program to reduce Salmonella in pork in the European Union.  相似文献

A study was conducted to determine the prevalence of Mycobacterium bovis-infection and atypical mycobacterioses in different cattle herd management systems in and around Morogoro, Tanzania. Between April and June 2005, a total of 728 bovines from 49 herds were tested for M. bovis-infection and atypical mycobacterioses. Milk samples were taken from tuberculin positive animals and analysed for the presence of mycobacteria. Total prevalences of 2.5% and 10.1% were found for M. bovis-infection and atypical mycobacterioses respectively, with more M. bovis-infection in cattle in the extensive management system and more atypical mycobacterioses in cattle in the intensive management system. From 8 out of 42 milk samples (19%) atypical mycobacteria were cultured. A higher prevalence of M. bovis-infection in the extensive sector could be due to several factors. In addition, such high prevalence puts herd owners and their families at risk for BTB. Therefore control of BTB, as well as education of cattle owners is important, especially in the extensive sector.  相似文献

Data on the geographic distribution of swine herds tested for pseudorabies virus (PRV) in the state of Illinois (USA) were analyzed to determine whether the prevalence of PRV-infected herds was clustered geographically at the county level. Second-order analysis of spatial dependence indicated there was a spatial clustering of counties of high PRV prevalence rates and that this clustering was greater than the observed clustering of counties with a large number of swine herds. The clustering of county PRV prevalence rates was most apparent within a radius of 120 km (on the average, approximately two couties away). The association of county PRV prevalence rates with average herd size, geographic density of swine herds in the country and regional (within 120 km) density of PRV-infected herds was analyzed using multiple linear regression. The primary factor affecting county PRV prevalence rates was the regional PRV density, which interacted with the other model variables. For counties with a low regional density of PRV infection, county PRV prevalence rates charged little with a change in county herd density or average herd size. In contrast, for counties with a high regional density of PRV infection, PRV prevalence within a county increased with increasing average herd size and increasing geographic density of swine herds in the county. The results of the current and previous studies implicate an important role for the geographic proximity of infected herds in the spread of PRV among swine herds.  相似文献

An epidemiological study was carried out to determine the herd prevalence of Chlamydophila abortus and Coxiella burnetii antibodies in goats covered by a milk recording program in Poland. The survey took place in 2007 and 48 herds located in different parts of the country were involved. A representative sample from each herd was taken by a simple random sampling allowing to detect seropositivity of a herd on a 95% level of confidence. In total 918 goats were tested for specific antibodies against both germs with the use of enzyme-linked immunosorbent assays. In addition, history of reproductive failures was recorded in these herds. The survey revealed that the herd prevalence of C. abortus was 4.2% (2 herds) while no C. burnetii antibodies were found. Abortions were reported to be a problem in 80% of herds while repeating estrus was encountered in 46% of herds. Reproductive failure concerned two seropositive herds as well. Since the germ is present in the population, it has to be taken into consideration in diagnostic process. Nevertheless, the results of the present study indicate that C. abortus infection occurs infrequently in Polish goats. As no antibodies against C. burnetii were detected in the screened sample the risk of goat-to-human transmission of both bacteria in Poland seems to be very low.  相似文献

Pre-movement testing for bovine tuberculosis (BTB) was compulsory in Ireland until 1996. We determined the proportion of herd restrictions (losing BTB-free status) attributable to the recent introduction of an infected bovid; described events between restoration of BTB-free status (de-restriction) and the next herd-level test for BTB; estimated the proportion of undetected infected cattle present at de-restriction; identified high-risk movements between herds (movements most likely to involve infected cattle); and determined the potential yield of infected cattle discovered (or herds that would not lose their BTB-free status) by pre-movement testing, relative to the numbers of cattle and herds tested. We used national data for all 6252 herds with a new BTB restriction in the 12 months from 1 April 2003 and 3947 herds declared BTB-free in the 12 months from 1 October 2001. We identified higher-risk animals from our logistic generalized estimating-equation models. We attributed 6-7% of current herd restrictions to the recent introduction of an infected animal. There were considerable changes to herd structure between de-restriction and the next full-herd test, and infection was detected in 10% of herds at the first assessment (full-herd test or abattoir surveillance) following de-restriction. Following movement from a de-restricted herd, the odds of an animal being positive at the next test increased with increasing time in the source herd prior to movement, increasing time between de-restriction and the next full-herd test and increasing severity of the source herd restriction. The odds decreased with increasing size of the source herd. We estimated that 15.9 destination-herd restrictions per year could be prevented for every 10,000 cattle tested pre-movement and that 3.3 destination-herd restrictions per year could be prevented for every 100 source herds tested pre-movement. The yield per pre-movement test can be increased by focusing on high-risk movements; however, this would result in a substantial decrease in the total number of potential restrictions identified.  相似文献

A cross-sectional study was carried out to determine the seroprevalence and risk factors associated with bovine respiratory syncytial virus (BRSV) infection in non-vaccinated dairy and dual-purpose cattle herds from Ecuador. A total of 2,367 serum samples from 346 herds were collected from June 2008 to February 2009. A questionnaire, which included variables related to cattle, health, management measures, and the environment, was filled out in each herd. Presence of antibodies against BRSV was analyzed using a commercial indirect ELISA test. A logistic regression model was used to determine risk factors associated with BRSV at herd level. The individual seroprevalence against BRSV in non-vaccinated herds in Ecuador was 80.48% [1,905/2,367; 95% confidence interval (CI)?=?78.9-82.1]. The herd prevalence was 91.3% (316/346; 95% CI?=?88.3-94.3), and the intra-herd prevalence ranged between 25% and 100% (mean, 90.47%). The logistic regression model showed that the existence of bordering cattle farms, the dual-purpose farms, and the altitude of the farm (more than 2,338?m above sea level) were risk factors associated with BRSV infection. This is the first study about BRSV prevalence in Ecuador. It shows the wide spread of the BRSV infection in the country. The risk factors found will help to design effective control strategies.  相似文献

Making valid inferences about herd prevalence from data collected at slaughter is difficult because the observed sample is dependent on the number of animals sampled from each herd, which varies with herd size and culling practices, and the probability of a positive test result, which depends on variable within-herd prevalence levels as well as test sensitivity and specificity. In this study, brucellosis herd prevalence among beef cow-calf operations is estimated from slaughter surveillance data using a method that combines process modeling with Bayesian inference. Inferences are made for two populations; the first population comprises cow-calf beef herds in a typical U.S. state. The second population represents all beef herds in a collection of 46 low-risk states. The Bayesian Monte Carlo method used in this study links process model inputs to observed surveillance results via Bayes Theorem. The surveillance evidence across multiple years is accumulated at a discounted rate based on the probability of introducing new infection into an area. The process model's inputs include herd size, culling rate per herd, within-herd prevalence, serologic test performance, and the probability of successfully investigating positive results. The surveillance results comprise the number of cows and bulls tested at slaughter and the number of affected herds detected each year. The results find at least 95% confidence that brucellosis herd prevalence among beef cow-calf herds is less than 0.014% (3 per 21,500 herds) and 0.00081% (5 per 6,15,770) after 5 years of slaughter surveillance (with no detections of affected herds) in a typical U.S. state and across 46 low-risk U.S. states, respectively. These results were based on conservative modeling assumptions, but sensitivity analysis suggests only slight changes in the results from changing the assumed process model input values. The most influential analytic input was the probability of introducing new infection into a putatively brucellosis-free state or group of states.  相似文献
