This study was conducted to investigate the effects of feeding sows a bulky diet during gestation on their physiological and metabolic adaptations during the peripartum period, and to determine how these effects may relate to sow and piglet performances. From d 26 of gestation until farrowing, gilts were fed diets that contained 2.8 or 11.0% crude fiber (control and high-fiber diets, respectively, n = 9/group). Daily feed allowance provided the same amount of DE daily (33 MJ of DE/d). Throughout lactation, sows were allowed to consume a standard lactating sow diet ad libitum. Litters were standardized to 12 piglets beyond 48 h after birth. On d 105 of gestation, a jugular catheter was surgically implanted. Preprandial blood samples were collected from d 109 of gestation to the day after farrowing and on d 4, 18, and 26 of lactation. Meal tests and glucose tolerance tests were performed on d 109 of gestation and d 4 and 18 of lactation. During gestation, BW and backfat gain did not differ between treatment groups. During lactation, sows fed the high-fiber diet ate an average of 0.94 kg/d more than control sows (P < 0.02). Piglets born from sows fed the high-fiber diet grew faster than piglets from control sows (P = 0.03). Body weight and backfat losses did not differ between the 2 treatment groups. Sows fed the high-fiber diet during gestation had lesser concentrations of leptin before farrowing than control sows (P < 0.01). Leptin concentrations were negatively correlated with feed intake during lactation (P < 0.05). The prepartal increase in prolactin concentrations tended to be greater in sows fed the high-fiber diet than in control sows (P < 0.1). Preprandial concentrations of glucose, NEFA, lactate, and IGF-I fluctuated over time without significant treatment effect. Glucose half-life was shorter in late gestation than during both stages of lactation, but did not differ between sows in the 2 groups. In late gestation, the postprandial increases in glucose and insulin were delayed, and smaller, after a high-fiber meal than after a control meal. During lactation, glucose and insulin profiles after a standard meal did not differ between sows from treatment groups. In conclusion, the greater appetite of lactating sows fed a high-fiber diet during gestation does not seem related to changes in glucose and insulin metabolism and may be partly due to decreased secretion of leptin. The greater feed consumption was accompanied by a faster growth rate of piglets without sparing effect on maternal body reserves.  相似文献

Insulin-like growth factor-I (IGF-I), growth hormone (GH), and prolactin (PRL) play important roles in milk protein synthesis, and their plasma concentrations were reported to be affected by dietary protein intake. To investigate the relationship between circulating amino acid (AA) and concentrations of these hormones, 18 Wistar rats aged 14 wk were assigned to a low (LP; 9% protein), standard (SP; 21% protein), or high-protein (HP; 35% protein) diet from parturition through day 15 of lactation. Plasma, liver, pituitary gland, skeletal muscle, and mammary gland samples were collected at the end of treatment. Circulating and hepatic IGF-I concentrations increased linearly with elevated dietary protein concentrations (P < 0.0001). Rats receiving the HP diet had higher circulating GH (P < 0.01) and pituitary PRL concentrations (P < 0.0001) but lower pituitary GH concentration (P < 0.0001) relative to those in rats receiving the LP and SP diets. Pearson correlation test performed on composed data across treatments showed that several circulating AAs were correlated with circulating and tissue concentrations of IGF-I, GH, and PRL. Multiple linear regression analyses identified Leu, Gln, Ala, Gly, and Arg as the main AAs associated with hormone responses (R2 = 0.37 ~ 0.80; P < 0.05). Rats fed the LP and HP diets had greater Igf1 and Ghr gene expression in skeletal muscle than those fed the SP diets (P < 0.01). However, LP treatment decreased Prlr mRNA abundance in mammary glands as compared with the SP and HP treatments (P < 0.05). The HP diets increased AA transporter expression (P < 0.01) but decreased mammalian target of rapamycin (P < 0.05) and 70 kDa ribosomal protein S6 kinase 1 (P < 0.01) phosphorylation in mammary glands as compared with the LP and SP diets. The results of the present study suggested that several circulating AAs mediated the effects of dietary protein supply on concentrations of IGF-I, GH, and PRL, which in turn altered the metabolism status in peripheral tissues including the lactating mammary glands.  相似文献

本试验旨在比较2种不同剂型酸化剂对哺乳母猪生产性能、初乳成分和肠道菌群结构的影响。试验选择30头体况相近、预产期接近的2~4胎长大二元杂交母猪,随机分成3个组,每组10个重复,每个重复1头母猪。在试验期间各组母猪分别饲喂基础饲粮(对照组)、基础饲粮+0.3%吸附型酸化剂(A组)、基础饲粮+0.1%微囊型酸化剂(B组)。预试期7 d(母猪分娩前7天)、正试期26 d(从母猪分娩开始至泌乳结束)。结果表明:与对照组相比,A组和B组母猪的泌乳期平均日采食量分别提高4.9%(P0.05)和5.3%(P0.05),仔猪断奶均重分别提高2.6%(P0.05)和7.4%(P0.05)。A组和B组的母猪初乳中乳脂、乳蛋白、尿氮素、免疫球蛋白G和免疫球蛋白A含量均高于对照组(P0.05),而乳糖含量则低于对照组(P0.05)。与对照组相比,B组母猪饲粮蛋白质消化率显著提高(P0.05),且粪便中大肠杆菌数量显著降低(P0.05);A组母猪粪便中大肠杆菌数量显著降低(P0.05)。由此可见,微囊型酸化剂在提高仔猪断奶重以及哺乳母猪饲粮蛋白质消化率和改善肠道菌群结构方面有一定功效,而吸附型酸化剂在改善哺乳母猪肠道菌群结构方面有一定功效。  相似文献

Our objective was to study the effects of dietary-induced insulin enhancement during and after lactation on the reproductive performance of primiparous sows. During a 21-d lactation period, 48 sows were allotted to a 2x2 factorial experiment. Treatments were feeding level (high or low; 44 MJ or 33 MJ NE/d) and dietary energy source (fat or starch). After weaning, all sows received the same amount of feed (31 MJ NE/d from weaning to estrus and 17.5 MJ NE/d from breeding until slaughter) of the same energy source as fed during lactation. On d 7, 14, and 21 of lactation and d 22 (weaning), blood samples were taken every 12 min for 12 h and analyzed for plasma glucose, insulin, and LH. Sows were slaughtered on d 35 of the subsequent pregnancy, and ovulation rate was assessed. During lactation, postprandial plasma glucose and insulin concentrations were higher for sows fed the starch diet than for those fed the fat diet (P<.001), whereas feeding level had no effect. Basal and mean LH concentrations were not affected by treatments. The LH pulse frequency on d 7 of lactation was greater for sows fed the starch diet than for those fed the fat diet (.52 vs .17 pulses/12 h; P = .03). The high compared with the low feeding level resulted in a greater LH pulse frequency on d 21 of lactation (.89 vs .47 pulses/12 h; P = .05) and on d 22 (8.63 vs 5.77 pulses/12 h; P = .02), in a higher percentage of sows that exhibited estrus within 10 d after weaning (96 vs. 63%; P = .01), and a tendency for a higher ovulation rate (18.0 vs. 16.2; P = .09). Plasma glucose and insulin concentrations were not related to any of the LH traits. The LH pulse frequency after weaning was related to the weaning-to-estrus interval (WEI) and was best explained by a linear-plateau model. In sows fed the low feeding level, follicle size after weaning was correlated with LH pulse frequency after weaning and with the WEI, whereas in sows fed the high feeding level these correlations were not significant. Our results indicate that an improved dietary-induced insulin status during and after lactation does not overcome the inhibitory effects of lactation on subsequent reproduction at any of the feeding levels.  相似文献

The objective of this study was to investigate the effects of substituting 1 kg of a standard lactation diet with 1 kg of a sugar‐rich (15.75 DE MJ/kg) or fat‐rich (23.85 DE MJ/kg) diet during late lactation on blood glucose and insulin changes in primiparous sows. During a 4‐week lactation period, 21 primiparous sows were fed to appetite with a standard lactation diet (14.10 DE MJ/kg). At 9 days before weaning, sows were assigned to a control (C, n = 7), fat (F, n = 6) or sugar (S, n = 8) treatment. During the treatment period (from 8 days before weaning until weaning), 1 kg of the lactation diet was substituted with 1 kg of a sugar‐rich or fat‐rich diet for S and F sows. At 3 days before weaning, serial blood samples were collected for a total of 228 min around feeding to establish pre‐ and postprandial plasma glucose and insulin concentrations. Preprandial plasma glucose and insulin concentrations did not differ between treatments (p > 0.05); however, mean plasma glucose and insulin concentrations were higher for S compared to F (p < 0.05) and intermediate for the C sows. Postprandial plasma concentrations of glucose and insulin were higher for the S sows than for C and F sows (p < 0.05). Sow body weight loss during late lactation did not differ between treatments (p > 0.05). The results from our study suggest that a sugar‐enriched diet during the last week of lactation elevates circulating glucose and insulin concentrations and may potentially improve post‐weaning fertility in primiparous sows.  相似文献

本试验旨在探讨饲粮中添加低聚壳聚糖对母猪繁殖性能、泌乳性能和血浆生化指标的影响。选择24头胎次、体重和预产期相近的健康大白猪母猪,随机分为3个组,每组8个重复,每个重复1头猪。对照组饲喂基础饲粮,试验Ⅰ组和Ⅱ组饲粮在基础饲粮中分别添加50和100 g/t的低聚壳聚糖。试验从母猪妊娠第85天时开始,至分娩后第21天时结束。结果表明:1)与对照组相比,饲粮中添加低聚壳聚糖可显著增加母猪的健仔数和哺乳期平均日采食量(P0.05),显著降低母猪的产程(P0.05),且在一定程度上可改善母猪的便秘和降低母猪的背膘厚损失;与对照组相比,试验Ⅰ组仔猪的初生窝重显著增加(P0.05),试验Ⅱ组母猪的哺乳期泌乳量显著增加(P0.05)。2)与对照组相比,试验Ⅰ组和Ⅱ组母猪初乳中乳脂肪含量显著增加(P0.05),试验Ⅰ组初乳中尿素氮(UN)含量和常乳中精氨酸、总氨基酸、限制性氨基酸含量显著增加(P0.05)。3)试验Ⅰ组和Ⅱ组母猪的血浆谷丙转氨酶(GPT)活性显著低于对照组(P0.05),各组的血浆碱性磷酸酶(ALP)、乳酸脱氢酶(LDH)活性和总蛋白(TP)、白蛋白(ALB)、UN、甘油三酯(TG)、总胆固醇(CHO)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)含量均无显著差异(P0.05)。由此可见,饲粮中添加低聚壳聚糖对母猪的繁殖性能和泌乳性能均有一定的促进作用。综合本研究结果,50 g/t低聚壳聚糖为母猪饲粮中的最佳添加剂量。  相似文献

This study was conducted to evaluate the effects of dietary live yeast (LY) supplementation during late gestation and lactation on reproductive performance, colostrum and milk composition, blood biochemical and immunological parameters of sows. A total of 40 multiparous sows were randomly fed either the control (CON) diet or the CON diet supplemented with LY at 1 g/kg from d 90 of gestation to weaning. Results showed that the number of stillborn piglets and low BW piglets were significantly decreased in the LY-supplemented sows compared with sows in the CON group (P < 0.05). Moreover, the concentrations of protein, lactose and solids-not-fat were increased in the colostrum of LY-supplemented sows (P < 0.05). Interestingly, the plasma activities of aspartate aminotransferase (AST), alkaline phosphatase (ALP), gamma-glutamyl transpeptidase (γ-GGT) at d 1 of lactation and alanine aminotransferase (ALT) at weaning day were decreased by feeding LY diet (P < 0.05). Meanwhile, sows fed LY diet had higher plasma concentration of immunoglobulin G compared with sows fed CON diet at d 1 of lactation (P < 0.05). In conclusion, LY supplementation in maternal diets decreased the number of stillborn piglets and low BW piglets, improved colostrum quality and health status of sows.  相似文献

Thirty-four sows (parity=1.8; BW=206 kg) were used to determine the influence of L-carnitine and/or chromium tripicolinate on plasma leptin concentrations of gestating sows fed one meal daily. Treatments were arranged in a 2 x 2 factorial with main effects of carnitine (0 or 50 ppm) and chromium (0 or 200 ppb). Diets were fed for approximately 167 days (through one gestation, the following lactation, the interval from weaning to estrus, and 28 days into the following gestation) prior to blood collection. Leptin concentration was determined in plasma that was collected at feeding, every 15 min for the first 3h after feeding, and at 6, 9, 15, 20, and 24h after feeding. Sows fed diets containing carnitine had greater (P<0.02) overall mean plasma leptin concentrations and greater (P<0.05) leptin concentrations at 2.25, 3, 6, 15, 20, and 24h after feeding compared to sows fed either the control diet or the diet containing chromium. Leptin concentrations of sows fed diets containing carnitine also were greater (P<0.05) than control sows at 2.5 and 2.75 h postprandial and greater than (P<0.05) sows fed diets with both carnitine and chromium at 6h after feeding. Chromium had no effect (P>0.10) on plasma leptin concentration. These results suggest that dietary carnitine, but not chromium, increases circulating leptin in gestating sows fed one meal per day. These results may help to explain the improvements in reproductive function previously observed from feeding sows diets containing carnitine.  相似文献

Effect of nutrient intake on mammary gland growth in lactating sows   总被引:2,自引:0,他引:2  
Sixty-one primiparous sows were used to determine the response of mammary gland growth to different energy and protein intakes during lactation. After birth, litter size was set to 9 or 10 pigs. Sows were slaughtered at selected times up to 30 d of lactation. Individual sows were fed one of four diets that were combinations of different amounts of energy and protein (3.0 Mcal ME and 8.0 g lysine/kg diet; 3.0 Mcal ME and 16.2 g lysine/kg diet; 3.5 Mcal ME and 6.4 g lysine/kg diet; or 3.5 Mcal ME and 13.0 g lysine/kg diet). Mammary glands were collected at slaughter and trimmed of skin and the extraneous fat pad. Each gland was weighed, cut in half to measure cross-sectional area, ground, and stored at -20 degrees C for chemical analysis. Frozen, ground tissue was used to determine dry matter, dry fat-free tissue (DFFT), total tissue protein, ash, and DNA content. Only glands known to have been suckled were included in this data set. Response surface regression was used for statistical analysis. The percentage of protein, fat, ash, and DNA in each suckled mammary gland was affected only by total energy intake (P<.05). The percentage of dry tissue and fat decreased as the total energy consumed during lactation increased, whereas the percentage of protein and DFFT increased as total energy intake increased. There were quadratic effects (P<.05) of both total energy and protein intake on wet weight, dry weight, protein amount, DFFT amount, and DNA amount of each suckled mammary gland during lactation. This study shows that mammary gland growth is affected by nutrient intake during lactation. The weight of suckled mammary glands and the amount of mammary tissue protein, DFFT, and total DNA were maximal on d 27.5 of lactation when sows had consumed an average of 16.9 Mcal of ME and 55 g of lysine per day during lactation. Provision of adequate amounts of nutrients to sows during lactation is important for achieving maximal growth of mammary glands and maximal milk production.  相似文献

Effects of three dietary lysine (protein) concentrations during lactation on metabolic state, protein metabolism, reproductive hormones, and performance were investigated in 36 primiparous sows. Sows were assigned randomly to one of three diets containing .4% (low lysine, LL), 1.0% (medium lysine, ML), or 1.6% (high lysine, HL) total lysine from intact protein sources. All diets contained 2.1 Mcal NE/kg and exceeded the recommended requirements for all other nutrients. Actual lysine intakes over an 18-d lactation were 16, 36, and 56 g/d for sows fed LL, ML, and HL, respectively. Fractional breakdown rate of muscle was determined on d 4 and 15 of lactation by using a three-compartment kinetic model of 3-methylhistidine metabolism. Increasing lysine intake during lactation did not affect fractional breakdown rate of muscle on d 4 of lactation but decreased it on d 15 (P < .05). Sows fed LL had a reduced number of LH pulses on d 12 and 18 (P < .05) and reduced serum estradiol (E2) concentration on d 18 of lactation compared with sows fed ML and HL treatments. However, LH pulses and E2 concentrations were similar between ML and HL treatments (P > .35). Increasing lysine intake increased serum urea nitrogen (SUN) and postprandial insulin concentrations (P < .05) during lactation but had no effect on plasma glucose concentrations (P > .20). Sows fed HL had greater serum IGF-I on d 6 and 18 than sows fed ML (P < .05). Number of LH peaks was correlated with serum insulin concentration 25 min after feeding on d 6 and 18 (r = .31 to .41; P < .1) and pre- (r = .33 to .46) and postprandial (r = .30 to .58) SUN concentrations (P < .05) during different stages of lactation. Results indicate that, compared with medium lysine intake, low lysine intake increased muscle protein degradation and decreased concentrations of insulin, SUN, and estradiol and LH pulsatility. In contrast, high lysine (protein) intake increased SUN, insulin, and IGF-I, but did not increase secretion of estradiol and LH compared with medium lysine intake. Furthermore, nutritional impacts on reproduction may be mediated in part through associated effects on circulating insulin concentration.  相似文献

Thirty‐two sows were allocated to four treatments to evaluate the effect of dietary star anise (SA) supplementation during gestation and lactation on the lactational performance of sows. At 85 days of gestation, sows were randomly allotted to one of two diets supplemented with 0.5% SA or basal diet. After farrowing, sows were further allotted to one of two lactation diets supplemented with 0.5% SA or basal diet. On a weekly basis, body weight (BW) of sows and piglets was measured. Blood and milk samples were obtained from the sows and piglets. Number of days from weaning to estrus, milk yield and feed intake were also recorded. Weight gain of piglets from sows fed the SA‐supplemented diet during lactation was greater between days 7 and 14, days 14 and 21 and the overall experimental period compared with control groups. Supplementation of SA during lactation improved weaning weight of piglets, milk yield and average daily feed intake (ADFI) of sows. The SA diet increased concentrations of insulin‐like growth factor‐1 (IGF‐1) in ordinary milk and prolactin (PRL) in serum of sows. In conclusion, this study has indicated the beneficial effects of dietary SA addition in improving the lactation performance of sows.  相似文献

The current experiment was carried out to determine whether exogenous GnRH treatment in primiparous, lactating sows undergoing feed restriction would improve reproductive performance after weaning. Sows were allocated to one of three treatments: AA sows (n = 8) were fed to appetite throughout a 28-d lactation, AR (n = 12) and AR + GnRH (n = 12) sows were fed as AA sows from farrowing to d 21 of lactation, and feed intake was reduced to 50% of the ad libitum intakes from d 22 to 28. The AR + GnRH sows received 800 ng of GnRH i.v. every 6 h from d 22 to 28 of lactation, and AA and AR sows received saline. Sow weight, backfat, and litter weight were recorded weekly. Within 2 d after farrowing, litter size was standardized to 8 to 10. At d 17 of lactation, an indwelling jugular catheter was surgically implanted in each sow. Blood samples were taken for characterization of plasma LH, FSH, insulin, IGF-I, and leptin by RIA at d 21 and before and after weaning on d 28 of lactation. After weaning, all sows were given ad libitum access to feed, checked for onset of standing estrus twice daily with mature vasectomized boars, and inseminated 12 and 24 h after onset of standing estrus with pooled semen from the same fertile boars (3 x 10(9) sperm/AI). After breeding, feed allowance was reduced to NRC (1988) requirements for gestation. At d 28 +/- 3 of gestation, sows were killed and ovulation rate and embryo survival were determined. Restricted sows lost more weight during lactation than AA sows (P < .02). During the period of feed restriction, plasma IGF-I and postprandial insulin and leptin in AR and AR + GnRH sows, and LH pulse frequency in AR sows, were lower than those in AA sows (P < .04). Associations (P < .004) between plasma insulin and leptin and between leptin and mean LH concentrations were established. The LH pulse frequency in AR + GnRH sows did not differ from that in AA sows before weaning. After weaning, maximum, mean, and minimum LH concentrations in the AA and AR sows, and FSH concentrations in AR sows, increased (P < .05) in response to weaning. Paradoxically, GnRH treatment in lactation seemed to suppress the expected LH and FSH responses to weaning. Ovulation rate and embryo survival were not different among the three groups. In conclusion, although exogenous GnRH therapy restored LH secretion in feed-restricted sows, it did not improve overall reproductive performance.  相似文献

The aim of this study was to determine the effect of dietary Forsythia suspensa extract (FSE) supplementation to lactating sows and nursery pigs on post-weaning performance, antioxidant capacity, immunoglobulins, and intestinal health. Based on backfat, body weight (BW), and parity, 24 gestating sows (Landrace × Yorkshire) with average parity of 3.38 ± 0.61 and BW of 234 ± 6.81 kg were allotted into two dietary treatments (control vs. 100 mg/kg FSE) with 12 sows per treatment from day 107 of gestation to day 21 of lactation. After weaning, based on the initial BW and source litter, 192 nursery pigs (Duroc × [Landrace × Yorkshire], average BW of 6.98 ± 0.32 kg, weaned at day 21) were allotted into four dietary treatments with eight replicate pens per treatment, six pigs per pen for a 4-wk study. The treatments included the following: 1) CC (sows and their piglets both fed control diet); 2) CF (sows fed control diet and their piglets fed FSE diet [containing 100 mg/kg FSE]); 3) FC (sows fed FSE diet and their piglets fed control diet); and 4) FF (sows and their piglets both fed FSE diet). The MIXED procedures of SAS for a split-plot arrangement with sow diet as the whole plot and nursery diet as split plot were used to analyze the data. After weaning, piglets from FSE-fed sows had improved (P < 0.05) average daily gain and feed efficiency, and lower (P < 0.05) diarrhea rate in overall (day 1 to 28) compared with those from sows fed control diet. Piglets from FSE-fed sows also had higher (P < 0.05) contents of immunoglobulin G (IgG), growth hormone, superoxide dismutase (SOD), total antioxidant capacity in serum, villus height in ileum, and villus height to crypt depth ratio in jejunum, as well as lower (P < 0.05) content of malondialdehyde (MDA) in serum and crypt depth in ileum compared with those from sows fed control diet. Piglets fed FSE during nursery had increased (P < 0.05) concentrations of IgG, SOD, and catalase, and decreased (P < 0.05) MDA and tumor nuclear factor-α levels in serum compared with those fed control diet during nursery. Piglets from FC group had increased (P < 0.05) protein expression of occludin in jejunal mucosa and relative abundance of Lactobacillus on genus level in colon compared with those from CC group. In conclusion, for the performance and intestinal health, diets supplemented with FSE during lactation phase seemed more efficient to alleviate weaning stress than the nursery phase. In terms of the antioxidant status and immunoglobulins, FSE supplemented in both phases were efficient for nursery pigs.  相似文献

为探讨催乳激素对体外培养的山羊乳腺上皮细胞脂肪酸合酶(fatty acid synthase,FASN)基因转录活性的调控作用,试验分别用不同浓度的胰岛素(insulin,INS)、雌激素(estradiol,E2)、催乳素(prolactin,PRL)及不同激素组合(INS+PRL、E2+INS+PRL)处理山羊乳腺上皮细胞24 h,提取细胞总RNA,采用实时荧光定量PCR检测催乳激素对FASN基因mRNA表达水平的影响。细胞转染山羊FASN基因启动子报告基因载体,同样用不同浓度的胰岛素、雌激素、催乳素及激素组合处理24 h,利用双荧光素酶报告基因系统检测催乳激素对FASN基因启动子活性的影响。结果发现,用雌激素、催乳素处理山羊乳腺上皮细胞后,FASN基因启动子活性及mRNA水平极显著或显著上调(P<0.01;P<0.05),雌激素浓度为10、100 μmol/L和催乳素浓度为0.1和1 μg/mL时效果最为明显,而胰岛素对FASN基因的启动子活性及mRNA水平没有显著影响(P>0.05)。用不同激素组合(INS+PRL、E2+INS+PRL)处理细胞均能显著上调FASN基因启动子活性及mRNA表达水平(P<0.05)。结果表明,雌激素和催乳素能够调控FASN基因的转录活性,为进一步研究泌乳过程中FASN基因的分子调控机制提供理论依据。  相似文献

The current studies were designed to investigate relationships among prolactin (PRL) binding, PRL concentrations in plasma and metabolic activity of porcine mammary glands. Preliminary studies revealed specific high-affinity binding of oPRL to porcine mammary gland. Conditions for optimal specific binding were similar to those observed for other species. To address the main objectives of the study, four mammary biopsies and blood samples were obtained from each of four gilts during lactogenesis and lactation (d-11, 4, 21 and 42 of lactation) to measure in vitro rates of metabolic activity, PRL binding to mammary membranes and PRL concentrations in plasma. Metabolic activity, as measured by oxidation of glucose or acetate to CO2 and incorporation into lipid, was low during pregnancy, increased two- to five-fold on d 4, and then paralleled the lactation curve for sows. There were highly significant positive correlations between PRL binding and all measures of mammary metabolism when data from pregnancy and lactation were utilized. Coefficients were positive but generally not statistically significant when lactation data only were utilized. During lactation, significant negative correlations were observed between concentrations of PRL in plasma and PRL binding and between PRL in plasma and mammary metabolic rate. These data provide evidence that binding of PRL to its receptor is an important effector of milk production in sows. Furthermore, oPRL is a suitable ligand to quantify PRL binding to porcine mammary tissue.  相似文献

Mammary secretions obtained from four groups of sows at parturition and on days 7, 14 and 21 of lactation were defatted and assayed for total protein and insulin-like growth factor-I (IGF-I). Sows (n = 57) represented two breeds (Landrace and Duroc) and two genetic lines (selected for differences in sow productivity index, SPI) within each breed. Colostrum of Duroc sows was 4-6 fold and 30-60 fold greater in protein (P less than .001) and IGF-I (P less than .001) concentrations, respectively, than the corresponding day 7 milk from these sows. In contrast, the colostrum of Landrace sows was 2-3 fold and 30-50 fold greater in protein (P less than .001) and IGF-I (P less than .001) concentrations, respectively, than the corresponding day 7 milk. The IGF-I content in milk from Duroc sows did not differ among days 7, 14 and 21 of lactation, whereas the IGF-I content of day 7 milk from Landrace sows exceeded those for the corresponding 14 day and 21 day secretion (P less than .05). IGF-I concentration in days 14 and 21 milk was higher in Duroc (P less than .001 respectively) than Landrace sows. Lipid compared with carbohydrate calories in the diet of peripartal sows was examined for specific metabolic effects. Blood samples from second-parity Yorkshire sows fed isocaloric-isonitrogenous diets that differed by either glucose or corn oil supplying 20% of the daily metabolizable energy intake were assayed for growth hormone (GH), insulin and prolactin (PRL) concentrations. On d 97 to 103 of gestation, serum GH levels were reduced following feed intake; the reduction was greater (P less than .05) in glucose-fed sows during the postprandial period. Insulin concentration was increased following consumption of either diet, but to a greater (P less than .05) extent in glucose-fed sows corresponding to the greater (P less than .05) blood glucose response. Basal, fasting concentrations of PRL were not affected by dietary energy source; however, thyrotropin-releasing hormone (TRH) infusion resulted in a greater (P less than .05) PRL response of fat-fed sows. Samples obtained on d 8 to 11 of lactation revealed that TRH stimulated (P less than .05) GH secretion and the response was greater (P less than .05) in fat-fed sows. Plasma insulin concentration was increased (P less than .05) following feeding, and the increase was greater (P less than .05) in glucose-fed sows. Similar to the response measured in gestating sows, TRH-induced increases in PRL was greater in fat-fed, lactating sows. Sham-nursing did not influence maternal PRL concentration compared with the immediate increase noted following natural nursing. No effect of dietary energy source on reproductive performance characteristics was noted. Results of this study have shown that dietary lipid, compared with carbohydrate, selectively increased the sensitivity of the pituitary gland to a GH and PRL secretagogue such as TRH. The beneficial effects of lipid in the diet of peripartal swine may be attributable, in part, to the effect of dietary lipid on maternal pituitary PRL synthesis and(or) release.  相似文献

This experiment was designed to compare the effects of high ambient temperature and of feed restriction on plasma hormones and metabolites in primiparous lactating sows. Females were exposed to a constant thermoneutral (20 degrees C) or hot environment (30 degrees C) during lactation. Sows housed at 30 degrees C were given free access to feed (30AL: n = 12), whereas those housed at 20 degrees C were either pair-fed with those at 30 degrees C (20RF: n = 6) or were fed ad libitum (20AL: n = 6). A jugular vein catheter was surgically inserted in all sows at 100 d of gestation. Absorption of nutrients during the meal induced significant increases in plasma glucose, insulin, and glucagon, and a decrease in nonesterified fatty acids on Day 19 of lactation and Day 1 postweaning (P < 0.05). On Day 19, feed restriction at 20 degrees C was associated with higher plasma glucagon before the meal, lower plasma insulin after the meal and a lower insulin-to-glucagon ratio (I/GA) before and after the meal (P < 0.05). On Day 19, mean plasma concentrations measured in 30AL females were between those measured in 20AL and 20RF sows for nonesterified fatty acids and glucagon before feeding, and for glucose, nonesterified fatty acids, insulin, and glucagon after feeding. None of the differences between the 30AL and the 20RF groups was significant (P < 0.1). On Day 19, the only significant differences between the 30AL and 20AL groups were observed after the meal for plasma insulin and I/GA. Plasma insulin-like growth factor-I increased after farrowing in 20AL and 30AL sows only (P < 0.05). It was higher in 20AL than in 20RF and 30AL sows on Days 4 and 19 of lactation (P < 0.05). Overall, underfeeding at 20 degrees C induced changes in plasma insulin, glucagon, I/GA, and insulin-like growth factor-I, which would favor gluconeogenesis and body-reserve mobilization during lactation. Differences in glucagon and I/GA before the meal between well-fed sows at 20 degrees C and heat-exposed sows were attenuated, which could have detrimental consequences on glucose availability to the mammary gland and hence on milk production at 30 degrees C.  相似文献
