Yellow mosaic disease, caused by wheat yellow mosaic virus (WYMV), is one of the most serious diseases of winter wheat (Triticum aestivum L.) in Japan. The three pathotypes of WYMV are distributed in different geographical areas: pathotype I is found mainly in western and central Japan (Kanto), pathotype II in northern Japan (Tohoku and Hokkaido) and pathotype III on the southern island of Japan (Kyushu). A total of 246 doubled‐haploid (DH) lines, derived from a cross between ‘Yumechikara’ (resistant) and ‘Kitahonami’ (susceptible), were evaluated for 2 years for their resistance to WYMV pathotype I. A single major quantitative trait locus, Q.Ymym, mapping to chromosome 2D was associated with resistance to pathotype I in ‘Yumechikara’. This is the first time a QTL responsible for pathotype I resistance has been identified. Fine mapping of Q.Ymym indicated that it was on a tight linkage block originating from ‘Yumechikara’, and the markers associated with this block will accelerate the development of varieties resistant to WYMV pathotype I.  相似文献

Summary Co-isogenic lines of a common wheat, Triticum aestivum, cv. S-615 with each of the following ten major genes were produced by repeated backcrosses; The gene C on 2D chromosome for the compactum character, s on 3D for the sphaerococcum character, Hd on 4B, B1 on 5A, and B2 on 6B for awn suppression, Hg on 1A for glume hairiness, Hp on 4A for peduncle hairiness, Ne1 on 5B and Ne2 on 2B for hybrid necrosis, and v1 on 3B for virescence. Seven of them showed the typical mendelian fashion of inheritance, while three others (C, s and v1) were transmitted at lower frequencies than their corresponding normal alleles.The effects of those major genes on 24 characters of cv. S-615 were investigated, which are summarized as follows: C: Increased node diameter, number of spikelets per ear and spike density, but decreased lengths of all ear rachis, awn, anther, empty and outer glumes, and grain, and grain index. s: Increased culm diameter and thickness, and spike density, reduced lengths of all 1st and 2nd internodes, culm, flag leaf, rachis, awn, anther, empty and outer glumes, and grain, and grain index, and accelerated heading. Hd: Increased number of spikelets per ear and anther length, while decreased length of awn, and empty and outer glumes. B1: Increased 1st internode, rachis, and anther lengths, and grain index, but decreased spike density, awn length and grain thickness. B2: Increased rachis length, number of spikelets per ear, empty and outer glume lengths and grain index, but reduced awn length and grain thickness. Hg: No effects. Hp: Increased number of spikelets per ear. Ne1: Accelerated heading, and increased empty glume length. Ne2: Increased node and culm diameters. v1: No effects.Two species-specific genes, C and s, affected a large number of characters expressed in various developmental stages. These genes were found to have pleiotropic effects, namely, they influenced, at least, two groups of interdependent characters. The wheat progenitors and other wild relatives continue to be important sources of genes for agronomically desirable traits, which can be transferred into durum wheat (Triticum turgidum; 2n = 4x = 28; AABB genomes) cultivars via hybridization. Chromosome pairing in durum × alien species hybrids provides an understanding of genomic relationships, which is useful in planning alien gene introgression strategies. Two durum cultivars, ‘Lloyd’ and ‘Langdon’, were crossed with diploid wheatgrass, Thinopyrum bessarabicum (2n = 2x = 14; JJ), to synthesize F1 hybrids (2n = 3x = 21; ABJ) with Ph1. ‘Langdon’ disomic substitution 5D(5B) was used as a female parent to produce F1 hybrids without Ph1, which resulted in elevation of pairing between durum and grass chromosomes – an important feature from the breeding standpoint. The F1 hybrids were backcrossed to respective parental cultivars and BC1 progenies were raised. ‘Langdon’ 5D(5B) substitution × Th. bessarabicum F1 hybrids were crossed with normal ‘Langdon’ to obtain BC1 progeny. Chromosome pairing relationships were studied in F1 hybrids and BC1 progenies using both conventional staining and fluorescent genomic in situ hybridization (fl‐GISH) techniques. Multicolour fl‐GISH was standardized for characterizing the nature and specificity of chromosome pairing: A–B, A–J and B–J pairing. The A–J and B–J pairing will facilitate gene introgression in durum wheat. Multicolour fl‐GISH will help in characterizing alien chromosome segments captured in the durum complement and in their location in the A and/or B genome, thereby accelerating chromosome engineering research.  相似文献

Summary Azolla accessions (section Azolla) from the germplasm collections of the International Rice Research Institute and Washington State University were fingerprinted and classified by enzyme electrophoresis and leaf trichome morphology. A. filiculoides was enzymatically distinctive and also reliably identified by its prominent one-celled trichomes. Neotropical accessions labelled as A. filiculoides proved to be members of other species. Two groups of isolates were designated A. rubra, but those from Japan were identified as A. filiculoides. The A. rubra of Australia-New Zealand was biochemically unique and possessed less protuberant trichomes than A. filiculoides. A. microphylla, A. mexicana, and A. caroliniana were phenetically similar, but a. microphylla was identifiable from the others in the banding patterns of certain enzymes. A. mexicana and A. caroliniana were closely related enzymatically. The two-celled leaf trichomes of these three species were similar in size and shape.  相似文献
