The Shaw larval test, in conjunction with adult tick immersion and stall tests, was utilized to confirm that a field strain of B. decoloratus, from Natal, is highly resistant to the ixodicide fenvalerate (Factor of resistance 4,744). The objective of this study was to assess the host status of African buffaloes, Syncerus caffer, for the one-host tick Rhipicephalus (Boophilus) decoloratus. To this end the R. (B.) decoloratus burdens of ten buffaloes examined in three north-eastern KwaZulu-Natal Province (KZN) nature reserves were compared with those of medium-sized to large antelope species in these reserves and in the southern Kruger National Park (KNP), Mpumalanga Province. The R. (B.) decoloratus burdens of the buffaloes were considerably smaller than those of the antelopes in the KNP, but not those in the KZN reserves. The life-stage structure of the R. (B.) decoloratus populations on the buffaloes, in which larvae predominated, was closer to that of this tick on blue wildebeest, Connochaetes taurinus, a tick-resistant animal, than to that on other antelopes. A single buffalo examined in the KNP was not infested with R. (B.) decoloratus, whereas a giraffe, Giraffa camelopardalis, examined at the same locality and time, harboured a small number of ticks. In a nature reserve in Mpumalanga Province adjacent to the KNP, two immobilized buffaloes, from which only adult ticks were collected, were not infested with R. (B.) decoloratus, whereas greater kudus, Tragelaphus strepsiceros, examined during the same time of year in the KNP harboured large numbers of adult ticks of this species. African buffaloes would thus appear to be resistant to infestation with R. (B.) decoloratus, and this resistance is expressed as the prevention of the majority of tick larvae from developing to nymphs.  相似文献

Anaplasma centrale was isolated from a field collection of Rhipicephalus simus. Transstadial transmission of A. centrale with adult ticks was demonstrated, but the infection was not carried transovarially. Ticks from this collection were subsequently reared as a non-infected, laboratory strain. It was proved that the Onderstepoort live blood vaccine strain of A. centrale, isolated by Theiler in 1911, is still tick transmissible after more than 75 years of needle passage through cattle in the laboratory. Attempts to demonstrate transstadial transmission of the vaccine strain with Boophilus decoloratus and Boophilus microplus failed.  相似文献

A field study (February 2000 to August 2001) was conducted on communal and commercial farms in the Eastern Cape and North-West Provinces of South Africa to detect the levels of tick resistance to commonly used acaricides. The larvae obtained from engorged females of the one-host tick Boophilus decoloratus, the two-host tick Rhipicepalus evertsi evertsi and the three-host ticks Amblyomma hebraeum and Rhipicephalus appendiculatus were tested against various concentrations of amitraz, chlorfenvinphos and cypermethrin using the Shaw Larval Immersion Test method. Ticks from the communal farms showed higher levels of resistance to cypermethrin and some resistance to chlorfenvinphos whilst no resistance was detected against amitraz. However, ticks from commercial farms were equally resistant to amitraz, chlorfenvinphos and cypermethrin. The B. decoloratus populations tested were considerably more resistant to all the acaricides tested than the R. evertsi evertsi, A. hebraeum and R. appendiculatus populations. This supports the hypothesis that single-host ticks develop resistance faster than multi-host ticks. This trend was recorded on most of the farms where single- and multi-host ticks co-existed. It was concluded that the use of acaricides at high frequencies and high concentrations was one of the main causes of tick resistance in the study areas. Possible factors which caused the resistance problems are discussed and acaricide management strategies recommended.  相似文献

The susceptibility of the larval offspring of engorged female Boophilus decoloratus, and of the engorged females, collected from cattle on the dairy farms Brycedale, Sunny Grove and Welgevind in the Eastern Cape Province, South Africa, was tested against the acaricides amitraz, chlorfenvinphos and cypermethrin. Resistance was determined by means of the Shaw Larval Immersion Test (SLIT) for larvae and the Reproductive Estimate Test (RET) and Egg Laying Test (ELT) for adults. At Brycedale the tests all indicated resistance to chlorfenvinphos, and RET and ELT indicated resistance to amitraz and emerging resistance to cypermethrin. At Sunny Grove, B. decoloratus was resistant to cypermethrin using SLIT and exhibited emerging resistance to chlorfenvinphos with SLIT and to cypermethrin with both RET and ELT At Welgevind, resistance was recorded against chlorfenvinphos (SLIT) and against cypermethrin (ELT), and emerging resistance against permethrin (RET). The results obtained with RET and ELT were generally comparable, but often differed from those obtained with SLIT. Resistance could be detected within 7 days with ELT compared to 42 days with RET and 60 days with SLIT.  相似文献

Knockdown and persistence efficacies of a pour-on containing Amitraz 1% and Cypermethrin 1% and a spray dip containing 12.5% Amitraz were compared. Knock-down and persistence efficacies of the Amitraz spray dip against Rhipicephalus (Boophilus) decoloratus and Amblyomma hebraeum were significantly higher for the duration of the trial than those of the pour-on. In the case of Rhipicephalus appediculatus and Rhipicephalus evertsi evertsi, efficacy was significantly higher on Days 2 and 5. Resistance testing prior to the start of the trial indicated that R. (B.) decoloratus was resistant to both Cypermethrin and Amitraz, yet in the case of the spray dip excellent efficacy results were obtained. It is thought that the cattle's scruffy winter coat may have hindered the spread of the pour-on, but that the thorough wetting and especially the higher concentration of active ingredient applied via the spray dip allowed this formulation to be effective. These results show that under certain conditions a spray dip containing 12.5% Amitraz may be more effective than a pour-on containing 1% Amitraz and 1% Cypermethrin, despite apparent in vitro resistance.  相似文献

The applicability of laboratory bioassays to diagnose ivermectin (IVM) resistance in Rhipicephalus microplus was evaluated. Adult immersion tests (AITs), larval immersion tests (LITs) and larval packet tests (LPTs) were performed to characterise the effects of ivermectin toxicity on adults and larvae of a susceptible reference strain. The AIT was determined to be a reasonable assay but requires a large number of individuals to attain interpretable results. The LIT and LPT were validated with an IVM resistant strain, revealing resistance ratios (RRs) of 6.73 and 1.49, respectively. In a field survey, nine different populations of cattle tick from the states of S?o Paulo and Mato Grosso do Sul, Brazil, were analysed with the LIT. Populations without previous exposure to ivermectin exhibited RRs between 0.87 and 1.01. Populations previously exposed to IVM showed RRs between 1.83 and 4.62. The LIT was more effective at discriminating between resistant and susceptible populations than the LPT. The use of the LIT is recommended for the diagnosis of ivermectin resistance in R. microplus.  相似文献

Delayed-type hypersensitivity skin reactions were used to assess the tick resistance status of Tonga calves in Zambia. The antigen used in the tests was a homogenate of unfed nymphal Rhipicephalus appendiculatus which had been shown to give protective immunity in guinea pigs to adult female R appendiculatus. There was a significant negative correlation between the intensity of the reactions and the total number of ticks (Amblyomma variegatum, R appendiculatus, Hyalomma truncatum, Boophilus decoloratus and Rhipicephalus species) on the animals.  相似文献

The objectives of this study were to determine the diversity, seasonal dynamics and abundance of ticks infesting cattle in urban, small-scale farming communities in and around Botshabelo and Thaba Nchu in the eastern Free State Province, South Africa. A total of ten cattle, ear-tagged for individual identification, were investigated monthly at each of five localities. Adult ticks were removed from the right hand side of each animal and placed in containers filled with 70% ethanol. They were subsequently identified and their numbers quantified. Immature Otobius megnini were counted but not removed. A total of 244,538 adult ticks of ten different species were collected over the 12-month study period. The tick species, in decreasing order of relative abundance, were: Boophilus decoloratus (87.26%), Rhipicephalus evertsi evertsi (6.86%), Hyalomma marginatum rufipes (2.42%), Otobius megnini (1.85%) Rhipicephalus follis (0.76%), Rhipicephalus gertrudae (0.54%), Rhipicephalus sp. (0.21%), Ixodes rubicundus (0.08%), Hyalomma truncatum (0.01%) and Margaropus winthemi (0.004%). The three most abundant species, namely B. decoloratus, R. evertsi evertsi and H. marginatum rufipes, occurred at all localities but with significant differences in abundance. M. winthemi ticks occurred only in the Thaba Nchu area and were not found at any of the there localities in Botshabelo. Significant differences in tick burdens between the six warm months (September to February) and the six cooler months (March to August) were found for most of the species recorded. Boophilus decoloratus occurred in significantly higher numbers in autumn (March to May) and winter (June to August) compared to spring (September to November) and summer (December to February), with 76.8% of the total B. decoloratus burden occurring during the cooler months.  相似文献

Virgin females of both Boophilus decoloratus and Boophilus microplus, when mated with males of the other species, subsequently produced sterile eggs. Counts of spermiophore capsules in female seminal receptacles showed that the males of both species will mate with the females of both species and that B. microplus males show a slightly greater, but statistically insignificant, mating capacity than B. decoloratus males. South African B. microplus females, when mated with an Australian strain of B. microplus males, produced a 62% yield of viable hybrid progeny while the reciprocal cross produced only a 1,82% hatch of non-viable larvae. The hybrids were sterile when interbred and no hatch resulted when the Fl males were backcrossed with parent females. The reciprocal backcross of hybrid Fl females to parent males resulted in a low percentage hatch of non-viable larvae.  相似文献

Salmonella choleraesuis strain 38 (glycerol-positive fermentation) was repeatedly exposed to porcine neutrophils in an attempt to mimic in vivo conditions of the host immune system. After phagocytosis, viable intracellular S choleraesuis were isolated and the process was repeated at least 5 times. A fifth-passage strain-38 neutrophil-adapted clone, 38PMNa-5X, was isolated, and was compared with the parent wild-type strain 38 for changes. Strain 38PMNa-5X had increased resistance to killing by hydrogen peroxide and phagocyte killing by porcine neutrophils, as measured by 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide reduction. Strain 38PMNa-5X was less invasive than the parent strain on Vero cell monolayers, and had been cured of a 50-kb plasmid. The 50-kb plasmid was marked with bacteriophage mini-Mu (kanamycin resistant) and was reinserted into strain 38PMNa-5X. Strain 38PMNa-5X was avirulent in mice, but the isolates with reinserted plasmids had intermediate resistance to neutrophil and hydrogen peroxide killing and had restored invasiveness and mouse virulence. Differences in complement sensitivity and enzymatic activity were not observed between the strains.  相似文献

In this study the dominant tick species identified were Amblyomma cohaerens 52.2%, Rhipicephaluspravus 19.3% and A. variegatum 14.6%. A. cohaerens occurred throughout the sampling period and showed a peak during March. Their number declined significantly (P < 0.01) from August to December. R. pravus occurred from July to December only and peaking in September (P < 0.01). A. variegatum occurred in low numbers throughout the study period with a marked seasonal variation (P < 0.01) and abundant numbers of ticks were observed from May to July. The other tick species identified were Boophilus decoloratus, R. bergeoni and R. evertsi evertsi. Individual variation in tick infestation level was observed among the animals (P < 0.05). Since all the animals were of the same breed, age group and were managed similarly the selection of animals with low infestation level can be a component of tick control strategy.  相似文献

Free-living stages of ticks on a commercial game farm in the Thabazimbi District, Limpopo Province, South Africa, were collected by drag-sampling with flannel strips during the period September 2003 to August 2004. A total of 5 tick species was collected from 4 sites. Boophilus decoloratus was the most abundant species, followed by Amblyomma hebraeum. Seasonal abundance of the ticks was quantified and an optimum time to implement control measures against the ticks is proposed.  相似文献

While resistance against many other classes of acaricides has been described, products containing benzoylphenyl urea are currently still successfully used against the pesticide-resistant blue tick (Boophilus decoloratus) in South Africa. In order to assess any adverse impact of these tickicides on the important dung beetle (Coleoptera: Scarabaeidae) fauna, a bioassay was undertaken on the ecotoxicological effects of a fluazuron (benzoylphenyl urea) pour-on formulation (Acatak) on the survival and reproduction of the African dung beetles species Onthophagus gazella (Fabricius). The experiment yielded no significant differences in adult or larval survival, egg production, fecundity and fertility between the control and treatment group following three beetle generations over. These results suggested that treatment of cattle with the fluazuron pour-on formulation Acatak was not detrimental to the selected dung beetle species in any notable way.  相似文献

Recent outbreaks of bovine babesiosis caused by Babesia bovis in Swaziland had indicated the presence of the vector tick Boophilus microplus in the country although it had never before been directly identified. Engorged female Boophilus ticks were collected from cattle at diptanks in the course of a tick resistance survey and used to map the distribution of the two different species of Boophilus. B. decoloratus was found to be widespread throughout the country. B. microplus was identified for the first time in Swaziland and was found to have a patchy distribution. Relative resistance levels of cattle against tick infestations in the communal grazing area of Botshabelo in the south-eastern Free State were determined. The objective was to establish whether differences in resistance can be exploited to contribute to tick control methods used by small-scale farmers in resource-poor environments. Ten cows (Bos taurus crosses) between the ages of 18 months and four years were used and tick counts were conducted once a month over a period of 12 months to compare their total tick burdens. Tick burdens of the various animals were compared mutually as well as with the mean tick burden of the group as a whole. Tick numbers varied throughout the year on all individuals but some animals consistently tended to have either higher or lower numbers than the mean of the group. Tick burdens on cattle classified as having a relatively low resistance to tick infestations increased eleven-fold from January to June 1996 compared to a six-fold increase on cattle categorized as belonging to the high resistance group. Twenty-eight percent of the cattle in the total study group carried 50% of the ticks collected (60,079). It is recommended that farmers in the region visually assess B. decoloratus burdens, the most abundant tick species, and sell or cull the most susceptible animals first in their normal program of utilization of the animals. This should eventually result in the direct improvement of the overall tick resistance of their cattle herds.  相似文献

Adult females of the tick Boophilus decoloratus were removed from 6 breeds of cattle on 2 farms in the Northern Transvaal. Highest numbers of female ticks were collected from Simmentaler, followed by Santa Gertrudis, Bonsmara, Afrikaner, Brahman and Nguni. Resistance levels of each breed to B. decoloratus was positively correlated with the amount of Bos indicus genes in the breed, with the exception of Nguni, which is a sanga type but not pure B. indicus.  相似文献

A study was undertaken to determine the seasonal abundance of the ticks infesting cattle owned by resource-limited farmers in the north-eastern Free State Province of South Africa. Infestations of Boophilus decoloratus Koch, 1844, Rhipicephalus evertsi evertsi Neumann, 1897, Rhipicephalus follis Donitz, 1910, Rhipicephalus gertrudae Feldman-Muhsam, 1960 and Rhipicephalus warburtoni Walker & Horak, 2000, were monitored on cattle of mixed breeds at monthly intervals from May 1998 to April 1999. High tick intensity on the cattle was observed between March and June, with a peak in May to June 1998 for B. decoloratus and R. evertsi evertsi. Few ticks from the other three species were recovered from the cattle. Small peaks in November to December 1998 for R. warburtoni, and in December 1998 for R. follis and R. gertrudae were observed. Following the winter (June to August), numbers of B. decoloratus declined markedly, while those of R. evertsi evertsi and other rhipicephalids increased. These results represent the first published information on ticks of veterinary importance infesting cattle in the north-eastern part of the Free State Province.  相似文献

A simulation model for the African tick Boophilus decoloratus is presented. This model is based on the use of a dynamic life table that is driven by abiotic variables (temperature and vegetation status) remotely sensed (AVHRR sensor of the NOAA series of satellites) over time. The model incorporates temperature-dependent rates of egg production and development, climate-driven density-independent mortality rates, and density-dependent regulation of on-host stages. The model successfully describes both the seasonality and annual variation in the numbers of questing larvae and engorging females observed in eight sites throughout sub-Saharan Africa. Climate data from 1983 to 1999 in 10-day intervals are used as the basic input for modelling the dynamic patterns of activity at four different sites in Africa and to understand how abiotic factors can modulate the long-term life cycle of B. decoloratus.  相似文献

The effect of tick infestation of Hereford, Bonsmara and Nguni cows on the weaning masses of their calves in a dipping versus non-dipping situation was investigated. Breed had a major effect on the level of tick infestation, Herefords being most susceptible and Ngunis least. The productivity of Nguni cows, as measured by the weaning masses of their calves was also least affected. The effect of infestation on the productivity of Hereford cows was the greatest and that on the Bonsmara cows was intermediate. It was estimated that each engorged female tick (predominantly Boophilus decoloratus) caused a reduction of 8.9 g, 8.0 g and 8.6 g in the weaning masses of calves from Hereford, Bonsmara and Nguni cows respectively. The effect of infestation on the productivity of the Nguni cows was nevertheless small because a limited number of ticks fed to maturity on this breed due to its natural resistance.  相似文献
