Unitary impulse activity from tension receptors in the abomasal antrum in sheep anaesthetized with chloralose was studied with concurrent measurements of isometric tension in the antral wall, intraluminal pressures and electromyography (EMG). Rhythmical background activity had a close correlation with intraluminal pressure changes and usually a close correlation with local increases in EMG activity and isometric tension. Impulse frequencies increased, but were not sustained, following changes from isotonic to isometric conditions. Tension receptors were excited by transmural compression, by antral distension and by local intra-arterial injections of acetylcholine (2.5–25 µg), pentagastrin (10–20 µg), 5-hydroxytryptamine (1–20 µg), adrenaline (10 µg) cholecystokinin-8 (1–5 µg) or bradykinin (0.5–20 µg), and by systemic injections of bethanecol (5–1000 µg). Local intra-arterial perfusions of atropine (50 µg) and hexamethonium (100 µg) did not abolish spontaneous antral movements or affect the enhanced impulse activity evoked by acetylcholine or 5-hydroxytryptamine. Most antral tension receptors were excited by increases in antral tension or passive distension; they therefore functioned in series with the muscularis externa. This experiment investigates the changes in the plasma leptin levels of sheep fed a diet of three energy levels (low, moderate and high). Four mature wethers were used for this experiment. For the first 4 weeks, the sheep were fed diets to provide 1.2 times the maintenance metabolizable energy requirements, low energy levels (LE). During the second 4 weeks, the sheep were fed diets to provide 1.5 times the maintenance metabolizable energy requirements, moderate energy levels (ME). During third 4‐week period, the sheep were fed a diet to provide 1.8 times the maintenance metabolizable energy requirements, high energy levels (HE). Body weight was determined every week. Blood samples were collected prior to the morning meal at 3 days intervals throughout the experiment, and plasma leptin, insulin and nonesterified fatty acid (NEFA) concentrations were assayed. Body weight decreased in week 1 after the start of the experiment, it continued to decrease during the LE feeding, but it gradually increased until the end of HE period. Similarly, plasma leptin concentration decreased during LE feeding, but increased during the HE feeding. Additionally, positive correlation was obtained between leptin and insulin or glucose concentrations, whereas no clear relationship with circulating NEFA was observed. In conclusion, it was suggested that plasma leptin concentrations were affected by the metabolizable energy in feed.  相似文献

The acid secretory capacity of the abomasal mucosa was studied in sheep experimentally infected with Ostertagia leptospicularis. The acidity of the abomasal contents, permanently recorded by a pH probe located inside the abomasum, decreased markedly to mean levels between pH 5 and 6. Subcutaneous administration of histamine or carbachol successfully stimulated acid secretion (pH 3.4). The results indicate that the abomasal mucosa harboured a population of functional parietal cells which were also identified immunohistochemically (H(+)/K(+)-ATPase). Ultrastructural investigation before stimulation revealed that the majority of these cells was in a resting state. Despite high serum gastrin levels, the acid secretion was blocked either at the level of the parietal cell or the enterochromaffin-like cell by an unknown factor, possibly mediated by the parasites. This is the first report of a parietal cell dysfunction associated with a nematode infection in the abomasum. It is suggested that the parasites induce changes in their environment which favour their survival and/or increase their reproduction.  相似文献

Two diets differing in their degrees of ruminal degradation synchronicity between nitrogen (N) and organic matter (OM) were fed to six adult rams at a maintenance level for three months in a two‐way layout design combining a randomized block design. Adaptive changes in microbial N (MBN) supply and N balance were investigated for both diets. The in situ rumen degradation parameters of OM and N were determined for four feeds and subsequently were used to formulate the two diets to have different ruminal degradation synchronicity and to be identical with metabolizable energy (ME), metabolizable protein (MP) and ruminally degradable N concentrations. The animals were divided into two groups and fed asynchronous (ASYNC) or synchronous (SYNC) diet, respectively. On days 30, 60 and 90 of each feeding period, a five‐day metabolism trial was carried out. Animals consumed their diets thoroughly and did not show marked body weight change during the observation. The apparent digestibility of OM and N did not differ significantly between the diets throughout the periods. The MBN supply was significantly greater for ASYNC than that for SYNC (P < 0.01) and was greater on days 60 and 90 than that on day 30 (P < 0.05). Neither diets nor periods had a significant effect on the efficiency of MBN synthesis. There was a significant period effect on N retention (P < 0.05) together with tendency of diet × period interactions (P = 0.1). There was no significant dietary effect. An improvement in N retention with advancing feeding period was observed for both diets. The plasma urea concentration decreased with feeding period (P < 0.01) and there was a concomitant decrease in urinary N excretion. Plasma glucose concentration tended to increase (P = 0.07). The results suggest that lengthening feeding duration from 30 days might have caused ameliorative changes in the utilization of ME and MP, and may have improved microbe synthesis irrespective of the degradation synchronicity.  相似文献

Leptin is an anorectic hormone secreted mainly by peripheral adipocytes but also by other central and peripheral tissues. It acts by means of a receptor called OB‐R, influencing not only appetite and body mass but being also involved in many fields like endocrinology, metabolism and reproduction. Immunohistochemistry and qRT‐PCR techniques were, respectively, used to demonstrate the presence of leptin and its receptor in the gut of adult zebrafish and to evaluate the leptin gene expression response to feeding and fasting. Immunoreactivity for the antibodies utilized was demonstrated in feeding but not in fasting fish, and the gene expression analysis corroborates the data obtained by immunohistochemistry. Therefore, all the obtained results support the hypothesis of the role of this hormone in food regulation in zebrafish.  相似文献

Very few epidemiologic studies have verified the utility of the right paramedian abomasopexy (RPA) technique in cows with right abomasal disorders. This study aimed to investigate the outcomes and risk factors for non-survival in the herd within 30 days of surgery in cows with right abomasal disorders who underwent the RPA technique. Forty-seven Holstein cows with right abomasal disorders (25 with right abomasal displacement [RDA] and 22 with right abomasal volvulus [RAV]) were included. Twenty-two cows with RDA (22/25, 88.0%) and 10 cows with RAV (10/22, 45.5%) survived at 30 days post-surgery. Multivariate logistic regression analysis indicated that hyponatremia, hypokalemia, and the presence of abomasal volvulus were the major risk factors associated with non-survival.  相似文献

将健康的围产期奶牛30头随机分为3组, 每组10头。其中Ⅱ组为对照组, Ⅰ组和Ⅲ组分别为试验组。Ⅰ、Ⅱ、Ⅲ组均于产前28d开始分别饲喂NRC标准减少20%日粮(能量摄入80%组)、NRC标准日粮(能量摄入100%组)、NRC标准增加20%日粮(能量摄入120%组), 产后各组奶牛均饲喂标准日粮, 至产后56d结束。观察妊娠后期奶牛不同能量水平对干物质摄入量、产奶量、乳成分及体重等生产性能和血浆瘦素浓度的影响。试验结果表明: 奶牛产后干物质摄入量、产奶量、乳中蛋白质、乳脂率和非固体物质为Ⅰ组高于Ⅱ组、Ⅱ组又高于Ⅲ组(P>0 05 ); 同时血浆瘦素浓度为Ⅰ组高于Ⅱ组、Ⅱ组又高于Ⅲ组, 经统计分析组间差异极显著(P<0 01) 或差异显著(P<0 05); 但产前28~14d瘦素浓度组间差异均不显著(P>0 05 ), 此后Ⅰ组和Ⅲ组差异显著(P<0 05 )。此外, Ⅰ组体重消耗最小, Ⅲ组体重消耗最大, 各组间差异极显著(P<0 01)。可见, 围产期健康奶牛不同能量水平与干物质摄入量、产奶量、乳成分、体重等生产性能及血浆瘦素浓度之间存在着密切的关系。  相似文献

The objective of this experiment was to study the changes of plasma leptin concentration during puberty and its relationship with testosterone level and testis dimensions in Holstein bull calves. Six Iranian Holstein bull calves with approximately 6 months of age were used. Semen evaluation was conducted at 1‐month interval to determine the puberty state. To detect the plasma leptin and testosterone changes, blood samples were collected from the jugular vein during pre‐puberty (6–7 months of age), puberty (8–9 months of age) and post‐puberty (10–11 months of age). In addition, body weight (BW), body condition score (BCS) and testicular width and length were measured at 3‐week intervals. The effects of time (age) on total sperm number and percentage of progressive motility of sperm, plasma concentration of leptin and testosterone, amplitude and frequencies of testosterone, BW, BCS, testicular dimensions were significant. Sperm number and progressive motility during post‐puberty were higher than those during puberty and pre‐puberty. Plasma concentration of leptin during the pre‐puberty was higher than those during puberty and post‐puberty (p < 0.01). Mean plasma testosterone concentrations during puberty were higher than those during pre‐puberty (p < 0.05). BW, BCS and testicular dimensions consistently increased throughout the trial. Results indicated that in growing bull calves, plasma concentrations of leptin decreased during puberty, while circulating testosterone increased.  相似文献

The objective of the present study was to examine the relationship between plasma leptin levels and fat distribution in crossbred steers. Twenty-four crossbred steers (Japanese Black × Holstein) were used in this study. Six steers belonged to the growing phase (11 months of age), and 18 steers to the finishing phase (from 21 to 28 months of age). Plasma leptin levels correlated positively with body fat weight ( r  = 0.807, P  < 0.01), body fat percentage ( r  = 0.802, P  < 0.01), visceral fat weight ( r  = 0.807, P  < 0.01), visceral fat percentage ( r  = 0.780, P  < 0.01), subcutaneous fat weight ( r  = 0.745, P  < 0.01), subcutaneous fat percentage ( r  = 0.734, P  < 0.01), intermuscular fat weight ( r  = 0.793, P  < 0.01), intermuscular fat percentage (0.775, P  < 0.01), renal fat weight ( r  = 0.694, P  < 0.01), renal fat percentage ( r  = 0.642, P  < 0.01), subcutaneous fat thickness ( r  = 0.755, P  < 0.01), intermuscular fat thickness ( r  = 0.721, P  < 0.01), beef marbling score ( r  = 0.706, P  < 0.01) and the crude fat content of Musculus longissimus thoracis ( r  = 0.592, P  < 0.01). The location of fat deposits did not affect the correlation values. However, when steers were divided into growing ( n  = 6) and finishing ( n  = 18) groups, there was no significant correlation between plasma leptin levels and the carcass characteristics related to fat deposition (fat weight, fat percentage, carcass traits) in either group. These results suggest that plasma leptin levels in crossbred steers positively correlate with body fat content and carcass traits when these parameters vary widely.  相似文献

The relation between plasma leptin and daily ad libitum roughage intake was evaluated during a 14‐day period in eight obese Shetland ponies. When the feeding strategy was changed from maintenance feeding to ad libitum feeding, feed intake increased the first day, decreased the second day and increased again during the following days to reach a constant level after 8 days. Plasma leptin concentration increased during the first 2 days, but remained constant afterwards. Although the same pattern was found in all ponies, the magnitude of the increase in leptin on day 1 and the resulting decrease in feed intake on day 2 differed between ponies. A lower anorectic effect was seen in ponies with higher initial leptin concentration, suggesting the presence of different degrees in leptin sensitivity in obese ponies. High leptin production in a attempt to compensate for the decrease in leptin sensitivity might explain large variations in plasma leptin among obese ponies with similar body condition score. Further research is necessary to clarify whether the reduced leptin sensitivity precedes obesity in equines or vice versa.  相似文献

This study was conducted to evaluate the effect of feeding ensiled alfalfa with waste date on ruminal fermentation characteristics, microbial protein synthesis, protozoa population and blood parameters in sheep. Eight rams were used in a 2 × 2 change over design. Each experimental period consisted of 21 days including 16 days for adaptation and 5 days for sampling. For ensiling, fresh alfalfa (Medicago sativa L.) with different levels of waste date (Phoenix dactylifera L.) were mixed together and ensiled in 100‐l containers for 45 days. Chemical composition of silages such as dry matter (DM), crude protein, NH3‐N, organic matter, NDF, ADF and pH were determined. Then, it was used as 30% (DM basis) in diets. The experimental diets were as follows: (i) control (diet containing alfalfa silage without waste date), (ii) diet containing alfalfa silage with 5 g waste date/95 g DM, (iii) diet containing alfalfa silage with 10 g waste date/90 g DM, and (iv) diet containing alfalfa silage with 15 g waste date/85 g DM. The results of this experiment showed that adding waste date to alfalfa during ensiling, improved silage quality, DM and energy level. Total protozoa population and all of holotrich, cellulolytic and entodinia in rumen fluid were increased linearly by increasing the level of waste date. Nitrogen (N) intake, urinary N excretion and nitrogen retention were affected by dietary treatments. Also, allantoin, uric acid, total purine derivatives, microbial protein synthesis, cholesterol level and blood urea nitrogen were significantly different. In conclusion, direct ensilage of alfalfa can be attained by mixing 15 g waste date/85 g DM and positive associative effects such as increased metabolizable energy (ME) and silage quality occurred.  相似文献

OBJECTIVE: To evaluate the relationship between plasma leptin concentration and body fat content in dogs. ANIMALS: 20 spayed female Beagles that were 10 months old at the start of the experiment. PROCEDURE: Dogs were kept under regulated feeding and exercise conditions for 21 weeks, resulting in a wide range of body weights, body condition scores (BCS), and subcutaneous thicknesses. Plasma leptin concentration was measured by use of a canine leptin-specific ELISA test to evaluate its correlation to body fat content estimated by the deuterium oxide dilution method. Plasma concentrations of glucose, cholesterol, triglycerides (TG), and nonesterified fatty acids (NEFA) were also measured. RESULTS: Body fat content (9 to 60% of body weight) was positively and closely correlated (r = 0.920; n = 20; P < 0.001) to plasma leptin concentration (0.67 to 8.06 ng/ml), compared with other variables (ie, glucose, cholesterol, TG, and NEFA; r = 0.142, 0.412, 0.074, and 0.182, respectively). CONCLUSIONS AND CLINICAL RELEVANCE: The positive relationship between plasma leptin concentration and body fat content in dogs was similar to correlations reported for humans and rodents, suggesting that plasma leptin is a quantitative marker of adiposity in dogs.  相似文献

在内蒙古草原牧区,冬季对绵羊补饲精喂的研究具有重要的意义。为研究产羔前对母畜舍饲精料后,呼伦贝尔短尾羊体重、体尺及经济效益的变化,本研究选取60对待产的呼伦贝尔短尾母绵羊作为试验对象,在母畜产羔前一个月,试验组母畜舍饲精料,对照组仅补饲干草,分析100日龄内家畜体重、体尺及经济效益变化。利用方差及偏最小二乘回归法,分析产前补饲对家畜个体、生产效益的影响及影响家畜体重的主要体尺权重,结果显示,100 d时间段内,母畜体重呈先减少后增加的趋势;仔畜出生后一个月左右的时间段,试验组仔畜的胸围、胸宽、体高、胸深、体长和体斜长显著高于对照组(P<0.05)。影响呼伦贝尔短尾羊体重的主要体尺指标为:胸围、胸宽、体高和胸深,影响权重达到50%以上。补饲使得仔畜的成活率、出售价格显著高于未进行补饲的对照组(P<0.05)。通过补饲母畜不仅可以影响仔畜体重和体尺指标,而且对提高仔畜成活率和经济效益具有重要的作用,这为今后后备母羊的选育、家畜的精准淘汰提供了理论基础。  相似文献

Leptin gene is expressed in the mammary tissue and the expression of both leptin and its receptor changes significantly during pregnancy and lactation, with high levels during the first half of pregnancy and a decrease at delivery. The aim of this work was to investigate into leptin concentration in plasma and in milk during pregnancy and the first week after parturition in dairy cow and to analyze the correlation between leptin and beta-lactoglobulin (beta-LG) concentrations in plasma and in milk. The trial was conducted on six Holstein dairy cows, reared in the same environmental conditions and evaluated with similar body condition score, during the complete reproductive cycle from insemination to the delivery. Blood from the jugular vein and milk samples were collected at weekly intervals. Plasma leptin concentration showed a lower level (p < 0.05) at the beginning of pregnancy. Milk leptin concentration showed a higher level (p < 0.01) than plasma level from week 23 to week 29 of pregnancy. Plasma beta-LG concentrations were higher (p < 0.01) compared to plasma concentrations during the first part of pregnancy, then milk levels rise and become higher than plasma levels during the last weeks before dry period. A positive correlation (p < 0.01) was observed between leptin and beta-LG both in plasma and in milk profiles.  相似文献

枯草期选甘肃高山细毛羊成年母羊180只,后备母羊38只,羔羊40只,进行为期171 d的放牧与舍饲对比试验。结果表明:试验组(暖棚舍饲)平均每只成年母羊、后备母羊和羔羊体质量分别增加5.1、9.8和6.9 kg,增幅为:11.72%、30.81%和32.86%;对照组(放牧)每只成年母羊、后备母羊和羔羊体质量分别平均下降7.3、5.0和1.0 kg;试验末,试验组成年母羊和后备母羊体质量极显著高于对照组(P0.01),试验组羔羊体质量显著高于对照组(P0.05)。羊毛长度试验组成年母羊、后备母羊和羔羊羊毛每只分别平均增长3.9、3.8和2.7 cm,试验组成年母羊和后备母羊比对照组提高10.4%和16.0%,羔羊比对照组降低11.2%,试验组和对照组差异不显著(P0.05)。试验组各年龄段产羊毛量分别达5.24、4.56和3.79 kg,比对照组分别提高60.24%、73.38%和49.80%,差异极显著(P0.01)。试验组纯收入分别为成年母羊5.35元/只,后备母羊45.65元/只,羔羊59.57元/只,对照组由于掉膘损失严重,成年母羊和后备母羊分别损失28.32和13.05元/只,羔羊收益29.3元/只。  相似文献

Variations in the milk composition of Nigerian cattle, sheep and goats (Bunaji, Yankasa and Red Sokoto breeds, respectively), as well as residual phenotypic correlations between the milk constituents, were investigated. Results indicated that sheep and goats differed significantly (P < 0.05) from cattle in all constituents except protein percentage, which averaged 5.43%, 5.43% and 5.49%, respectively. Caprine milk contained the highest percentages of fat (5.80%), total solids (15.37%) and ash (0.77%), and bovine milk contained the least percentages of fat (0.68%) and lactose (1.84%). Overall, the milk compositions of sheep and goats were very similar as they were not statistically different from each other (P > 0.05). Residual phenotypic correlations between the milk constituents revealed highly significant (P < 0.01) and positive relationships between total solids and solids‐not‐fat (0.97 and 0.98 in cattle and sheep, respectively). All other correlations were positive in cattle (ranging from 0.12 to 0.77), except between protein and total solids (?0.44) and protein and solids‐not‐fat (?0.64). Multiple linear regression equations were fitted to predict the percentages of protein and fat. It was demonstrated that the protein percentage could be predicted from total solids and solids‐not‐fat with the highest accuracy of 94%, 86% and 82% in cattle, sheep and goats, respectively. On the other hand, the accuracy of prediction of fat percentage was very low in all the species (R2 = 0.01, 0.3 and 0.37 in cattle, sheep and goats, respectively).  相似文献
