Effects of exposure to hot environment on urea metabolism were studied in lactating Holstein cows. Four cows were fed ad libitum a total mixed ration and housed in a temperature‐controlled chamber at constant moderate (18°C) or high (28°C) ambient temperatures in a cross‐over design. Urea nitrogen (N) kinetics was measured by determining urea isotopomer in urine after single injection of [15N2]urea into the jugular vein. Both dry matter intake and milk yield were decreased under high ambient temperature. Intakes of total N and digestible N were decreased under high ambient temperature but urinary urea‐N excretion was increased. The ratio of urea‐N production to digestible N was increased, whereas the proportion of gut urea‐N entry to urea‐N production tended to be decreased under high ambient temperature. Neither return to the ornithine cycle, anabolic use nor fecal excretion of urea‐N recycled to the gut was affected by ambient temperature. Under high ambient temperature, renal clearance of plasma urea was not affected but the gut clearance was decreased. Increase of urea‐N production and reduction of gut urea‐N entry, in relative terms, were associated with increased urinary urea‐N excretion of lactating dairy cows in higher thermal environments.  相似文献

Information on oxidative stress under hot conditions from the levels of cells to organs and the whole body has accumulated in the last decades. Although a hot climate decreased dairy performance, changes of oxidative stress markers under hot conditions have remained obscure. Therefore, the effect of high environmental temperature on ascorbic acid, sulfhydryl (SH) residue and oxidized lipids concentrations in plasma from a total of 128 dairy cows was investigated. The monthly average maximum day temperature varied from 9.2°C in January to 32°C in August of 2004 in this institute. High ambient temperatures increased the rectal temperature of dairy cows up to 39.3°C in August. One of the reducing equivalents in plasma, SH residue concentration, decreased in July compared with December (P < 0.05). Another antiradical molecule, ascorbic acid concentration in plasma, also decreased in July (P < 0.01). The oxidative stress index, thiobarbituric acid reactive substance (TBARS), which was produced from the oxidation of polyunsaturated fatty acids under oxidative conditions, increased in summer (P < 0.05). A significant positive relationship of SH residue and ascorbic acid concentrations in the hot season was observed (P < 0.01). A negative correlation between rectal temperatures and ascorbic acid concentrations in the hot season was obtained (P < 0.01). However, TBARS concentration varied independently of the SH residue and ascorbic acid concentration. These results suggest that the response of oxidative stress markers of SH residue, ascorbic acid and TBARS concentration to oxidative stress under hot conditions were not shown to be the same, and that oxidative stress in dairy cows in the hot season increased.  相似文献

The temperature–humidity index (THI) is widely used to characterize heat stress in dairy cattle. Diet composition is known to induce variation in metabolic‐associated heat production. However, the relationships between THI and diet are poorly characterized with regard to performance and intake behaviour. Therefore, the objectives were to evaluate the impact of THI on water intake (WI), dry matter intake (DMI) and the frequency of drinking and feeding bouts in lactating dairy cows offered four dietary treatments: each contained 20% grass silage and additionally (i) 20% maize silage, 60% concentrate (M‐HC); (ii) 60% maize silage, 20% concentrate (M‐LC); (iii) 20% pressed beet pulp silage, 60% concentrate (BPS‐HC); or (iv) 60% pressed beet pulp silage, 20% concentrate (BPS‐LC) (DM basis). Individual WI and DMI were recorded from April to July 2013. Furthermore, dietary effects on milk production and reticular pH were estimated. Milk yield was lowest for M‐LC, while energy‐corrected milk was similar for all diets. Milk fat percentage was higher and milk protein amount lower for cows offered both LC diets. Reticular pH below 6.3, 6.0 and 5.8 lasted longest for BPS‐LC. WI was higher for HC diets. However, the frequency of drinking bouts was not influenced by the ration. Lower DMI occurred for BPS‐LC compared to M‐LC. Frequency of feeding bouts was significantly higher for LC diets. THI was significantly related to WI, DMI as well as drinking and feeding bouts. Per increasing THI, WI increased slightly more for LC diets and DMI decreased more for HC diets. Frequency of drinking bouts increased slightly higher for BPS rations per rising THI, while the decrease in feeding bouts was highest for M‐HC. In conclusion, TMR composition and moderate heat stress impacted WI and DMI of dairy cows, while both dietary energy density and ruminal filling might intensify the THI impact.  相似文献

High‐yielding Holstein‐Friesian recipients (n = 43) were used in order to investigate the relation between energy balance status during the early postpartum period and subsequent embryonic mortality after transferring good‐quality frozen embryos. Blood samples were collected during the second, third, fourth, fifth, sixth and seventh weeks postpartum in order to measure energy status indicators. These indicators include β hydroxyl butyric acid (BHBA), non‐esterified fatty acid (NEFA), total cholesterol (T‐chol), glucose and blood urea nitrogen (BUN). Moreover, body condition scores (BCS) were assessed during the same period. Pregnancy diagnosis by ultrasonography at the 28th day postestrus and embryo viability was ascertained until 45 days postestrus in order to detect late embryonic mortality (LEM). The pregnancy rate on day 28 was 44.2% (19/43); however, five cows (11.6%) experienced LEM by day 45. Based on the non‐return rate at day 24, non‐pregnant animals, as diagnosed by ultrasonography, were allocated into animals with longer estrus intervals than 24 days (32.5%; mid‐embryonic mortality (MEM) group) and animals returning to estrus by day 24 postestrus (23.5%; early embryonic mortality (EEM) group). At week 5 postpartum, BCS was significantly (P < 0.05) lower in the LEM group than that of pregnant (PREG), EEM and MEM groups. NEFA was significantly higher in animals that experienced LEM (LEM group) at week 7 postpartum (289.6 ± 47.0 µEq/L; P < 0.01) than that of PREG (196.8 ± 16.0 µEq/L), EEM (157.2 ± 18.6 µEq/L) and MEM groups (191.5 ± 14.4 µEq/L). In conclusion, lower BCS at week 5 postpartum and higher NEFAs at week 7 postpartum may be associated with subsequent LEM in high‐yielding recipient cows.  相似文献

试验旨在研究日粮阴阳离子平衡对泌乳中后期热应激奶牛血液生化指标的影响,选取试验奶牛32头分为4组,每组8头,日粮DCAB分别为对照组83 mEq/kgDM、试验Ⅰ组172 mEq/kgDM、试验Ⅱ组217 mEq/kgDM、试验Ⅲ组357 mEq/kgDM.研究结果表明:DCAB增加可在一定程度上提高奶牛血液的缓冲能力,缓解奶牛热应激,随DCAB增加,血液的pH值及HCO3-浓度升高,CO2分压下降,差异均不显著(P>0.05);高的DCAB能增加血液Na+、K+浓度,K+显著增加,Cl-显著下降(P<0.05);高水平的DCAB日粮,能使泌乳中后期血液ALP含量增加, CK、ALT含量减少,差异不显著(P>0.05).综合DCAB对血液缓冲能力、血液生化指标等方面考虑,在泌乳中后期DCAB为217 mEq/kgDM时饲喂效果较好.  相似文献

Data from six primiparous and nine multiparous Holstein cows were used to clarify the difference of Ca and P mobilization between primiparous and multiparous cows during early lactation. The dry matter intake (DMI) of primiparous cows was lower (P < 0.01) than those of multiparous cows. Milk yield was lower in primiparous cows at 7, 14, 21 (P < 0.01), and 28 days (P < 0.05) after parturition. There was no significant difference in milk Ca and P concentrations between primiparous and multiparous cows. There were no significant differences in plasma Ca and P concentrations between primiparous and multiparous cows. The plasma P level at 7 days postpartum in primiparous cows was lower (P < 0.05) than 28 days postpartum. The concentration of plasma osteocalcin (OC) measured as bone formation marker of primiparous cows was significantly higher than multiparous cows (P < 0.01) at 21 and 28 days postpartum. The urinary deoxypyridinoline (DPD) as bone resorption marker of primiparous cows tended to be higher (P < 0.10) than multiparous cows at 21 days after parturition and decreased to the same level as that of multiparae toward the peak lactation. These results show that Ca and P mobilization of primiparous cows are more active than multiparous cows.  相似文献

We evaluated the lactation performance, liver lipid content and plasma metabolites indicating the energy balance of dairy cows supplemented with conjugated linoleic acid (CLA) pre‐ and post‐partum (PP) vs. only PP. A total of 60 cows were divided into three groups (n = 20). Daily diet of cows was supplemented with 14 g of CLA (7 g cis‐9, trans‐11 and 7 g trans‐10, cis‐12 isomers) from week 3 before the expected date of calving (group CLA1), or from the day of calving (group CLA2) until 77–91 days PP. Control cows were fed an isocaloric, isonitrogenous and isolipidic diet without CLA. Between week 3 and week 6 PP, the milk yield of cows in both CLA‐treated groups was approximately 4.5 kg higher (p < 0.05) than in control. Milk fat concentrations decreased from week 3 and were lower in both CLA groups than in control (p < 0.01). Body condition score loss was lower (p < 0.05) in the CLA1 than in the control group on week 5 PP. By week 11 PP, the body condition of both CLA1 and CLA2 groups exceeded that of control. Plasma non‐esterified fatty acid was lower in CLA1 compared to CLA2 and control during the early PP period (p < 0.05), while this difference faded away by the late PP period. Beta‐hydroxybutyrate (BHBA) increased rapidly in all groups following calving. In CLA1 group, it began to decrease sooner than in CLA2 and control. The prevalence of subclinical ketosis (BHBA > 1.2 mm ) was lower in CLA1 group than in CLA2 and control (p < 0.05). Liver biopsy analyses showed that CLA1 treatment decreased (p < 0.05) the total lipid content of liver compared to control at week 5 after calving. Our results show that CLA supplementation is more efficient in alleviating body mass mobilization and decreasing the incidence of subclinical ketosis when applied as early as 3 weeks before calving than started feeding after calving.  相似文献

We compared the hair cortisol levels of lactating dairy cows in a cold‐ and a warm‐temperate region out of four climatic zones in Japan. We simultaneously investigated the effects of calving number, lactation period and month of hair sampling. Hair of nine Holstein lactating cows chosen from each region (i.e. 18 cows per sampling) was sampled in March, June, September and December. Number of calvings (1, 2, ≥3) and lactation duration (early: <100, middle: 101–200, and late: >201 days) were balanced between regions. Cortisol was extracted from hair by methanol, and its level was determined with a cortisol immunoassay kit. A multi‐way analysis of variance revealed that the effects of month of hair sampling (P < 0.001) and its combination with region (P < 0.05) were significant. In a multiple comparison test, significant differences (P < 0.01) in hair cortisol level (pg/mg of hair) were found between June (13.0 ± 1.0) and the other 3 months, and between September (1.6 ± 0.2) and December (4.5 ± 0.3). The rise in cortisol level from March to June was more intense in the cold‐temperate region. These results demonstrate the necessity of considering seasonal variations in each climatic region when we use hair cortisol level as an indicator of stress.  相似文献

选择40头体质健康的中国荷斯坦奶牛,根据胎次和泌乳天数,按照随机区组试验设计分为Ⅰ组(30kg/d)、Ⅱ组(30~35kg/d)、Ⅲ组(35~40kg/d)和Ⅳ组(40kg/d)。热应激前、热应激前期、热应激中期、热应激后期和热应激后分别于尾静脉采血,用ELISA试剂盒测定热休克蛋白(HSP)27,70,90的表达量。结果显示,Ⅳ组HSP27表达量最高,Ⅱ组表达量最低,Ⅳ组、Ⅲ组和Ⅰ组均显著高于Ⅱ组(P〈0.05)。HSP70表达量各组间没有明显差异,但随产奶量呈线性增加(P〈0.05)。HSP90的表达量,Ⅳ组和Ⅲ组明显高于Ⅱ组(P〈0.05)。HSP27的表达量热应激后差异较大;HSP70的表达量各组整个过程差异较大;HSP90的表达量在热应激前、热应激前期和热应激后差异较大。总之,在热应激过程,高产奶牛血清中热应激蛋白的表达量较高,HSP70表达量随产奶量呈线性增加,而不同热应激蛋白的变化规律差异较大。  相似文献

Temperature gradients in female reproductive tissues seem to influence the success of key processes such as ovulation and fertilization. The objective of this study was to investigate whether pre‐ovulatory follicles are cooler than neighbouring uterine tissue and deep rectal temperatures in lactating dairy cows under heat stress conditions. Temperatures within the pre‐ovulatory follicle, on the uterine adjacent surface and 20 cm deep within rectum, were measured using fine thermistor probes within 45 min after sunrise (dawn). Cows were selected from synchronized groups for fixed‐time insemination during the warm period of the year. Five cows under direct sun radiation and 11 cows in the shade were included in the study. None of the cows in the sun area ovulated within 24 hr, whereas 10 of the 11 cows in the sun area ovulated. Four of the 10 ovulating cows became pregnant. In the ovulating cows, follicular temperatures were 0.74 and 1.54°C significantly cooler than uterine surface and rectal temperatures, respectively, whereas temperatures in the uterine area were 0.80°C significantly cooler than rectal temperatures. No significant differences among temperatures were found in non‐ovulating cows. Follicular size was similar for ovulating and non‐ovulating cows. Environmental temperatures in the shade area were 6.4°C significantly lower than those in the sun area. Results of this study indicate that pre‐ovulatory follicles are cooler than neighbouring uterine tissue and deep rectal temperatures and those temperature gradients were not found in cows suffering ovulation failure.  相似文献

We measured a bone-formation marker recognizing osteocalcin, and a bone-resorption marker recognizing C-telopeptide (CT(x)) fragments of collagen type I, in a longitudinal study. The levels of these markers in the plasma of dairy cows (n=11) were recorded over a 12 month postpartum period, including a full lactation and a dry period. The plasma concentration of CT(x) was highest in the first week after parturition. It then declined slowly over the next 33 weeks and remained low until the next parturition. Osteocalcin concentration was lowest around parturition, reached a plateau during mid-lactation, then fell again towards term. There were large variations in bone metabolism during a lactation, that were not directly related to milk production. These results may be used to facilitate appropriate adjustments to calcium and phosphorous concentrations in the diet, reflecting the specific needs of each stage of the reproductive cycle.  相似文献

In dairy cows, exogenous α‐amylase is suggested to improve starch utilization and positively affect performance and health traits linked to energy balance and fertility. In a 1‐year feeding experiment, 421 cows were orally supplemented with α‐amylase (treatment: 12.5 g/kg dry matter (DM) addition rate to a concentrated feed) or non‐supplemented (control) on the basis of an ad libitum total mixed ration (TMR). Every cow was allocated to a high‐ (≥32 kg milk/day) or late‐lactation group (<32 kg milk/day), in which the TMR starch content was 220 ± 20.8 g/kg DM and 183 ± 24.8 g/kg DM, respectively. The energetic effect of α‐amylase supplementation seemed to be exclusively related to the high‐lactation stage (5–100 days in milk) in primiparous cows, where the daily milk yield was 32 ± 0.49 versus 31 ± 0.50 kg per cow in the treatment versus control group (< 0.05). The pluriparous cows did not benefit from the supplementation that way. In neither primiparous nor pluriparous cows, was the milk composition, the fat‐to‐protein ratio, the somatic cell score, the backfat thickness, serum total bilirubin, β‐hydroxybutyrate and the fertility found to be systematically affected by α‐amylase supplementation.  相似文献

Dairy cow bone phosphorus (P) mobilization and deposition and their influence on P requirements were studied over the lactation cycle. Thirty Holsteins received a common diet during the dry period and one of the following three dietary treatments that varied in P percentage during the subsequent lactation (44 weeks): (i) 0.36 throughout (constant P, 0.36‐0.36‐0.36), (ii) 0.36 for 30 weeks then 0.29 for 14 weeks (P changed once, 0.36‐0.36‐0.29), and (iii) 0.43 for 10 weeks, 0.36 for 20 weeks, and 0.29 for 14 weeks (P changed twice, 0.43‐0.36‐0.29). Six P balance studies were conducted during the experiment, including one during the dry period and five along lactation, based on P intake, faecal P, urinary P and milk P, when appropriate. Blood samples were taken during balance to analyse bone formation (osteocalcin) and resorption (pyridinoline) marker concentrations and rib biopsies performed to determine bone P content. Phosphorus balance was negative during weeks ?4 to ?1 relative to lactation for all groups and remained negative for cows fed 0.36% P during weeks 1–5, but showed a positive value for cows that received 0.43% P. The balance was close to zero for all groups at weeks 19–23 and showed a clear retention during weeks 38–42; by the end of lactation, cows re‐stored most of the P mobilized earlier. The pattern in P balance was consistent with changes in blood bone metabolism marker concentrations, rib bone P content, and faecal and urinary P concentrations over the experiment, indicating that cows, irrespective of the dietary P treatments received, mobilized P from bone during the late dry period when fed a low‐Ca diet and early lactation, and re‐stored P in late lactation. This dynamic of P metabolism can have important implications for dietary P requirements and ration formulations.  相似文献

Supplementing conjugated linoleic acid ( CLA ) is supposed to spare glucose due to the milk fat‐depressing effect of the trans ‐10, cis ‐12 CLA isomer, and allows repartitioning nutrients despite an energy deficiency in early lactation. However, there is still a lack of knowledge in terms of the dynamic pattern of the glucose turnover in transition dairy cows. We hypothesized that dairy cows supplemented with CLA have an altered rate of glucose turnover and insulin sensitivity during early lactation. We conducted three consecutive hyperglycaemic clamps (HGC ) in weeks ?2, +2 and +4 relative to parturition in Holstein cows supplemented daily either with 70 g of lipid‐encapsulated CLA (6.8 g trans ‐10, cis ‐12 and 6.6 g of the cis ‐9, trans ‐11 CLA isomer; CLA ; n  = 11) or with 56 g of control fat ( CON ; n  = 11). From week ?3 up to week +4 relative to parturition, milk yield and dry matter intake (DMI ) were recorded daily, while body weight (BW ) and milk composition were obtained once weekly. Blood samples were taken once weekly and every 30 min during the HGC . Plasma was analysed for concentrations of glucose, fatty acids (FFA ), beta‐hydroxybutyrate (BHB ), insulin, triglycerides and cholesterol. The CLA supplementation did not affect performance and metabolic parameters except for BHB and cholesterol. Furthermore, insulin concentrations and insulin sensitivity were affected by treatment. During the HGC in early lactation, insulin response was lower and decrease in FFA and BHB greater compared with the HGC in week ?2 although glucose target concentration achieved during the steady‐state period was similar for all three HGC . Our findings in terms of insulin and cholesterol suggest that body reserves are preserved through CLA feeding without restraining animal's performance. Furthermore, CLA effects on cholesterol and triglyceride concentrations indicated beneficial effects on hepatic lipid export contributing to an improved efficiency of prevailing metabolites in circulation.  相似文献

Cull dairy cows contribute almost 10% of national beef production in the United States. However, different factors throughout the life of dairy cows affect their weight and overall body condition as well as carcass traits, and consequently affect their market price. Therefore, the objectives of this study were: (1) to assess relationships between price ratio and carcass merit of cull dairy cows sold through several sites of an auction market and (2) to investigate the effect of animal life history events and live weight on sale barn price (BP) and price ratio (as a measure of relative price), as an indicator of carcass merit. Data from 4 dairy operations included 3,602 cull dairy cow records during the period of 2015 to 2019. Life history events data were collected from each dairy operation through Dairy Comp software; live weight and price were obtained periodically from the auction market, and the carcass data were provided by a local packing plant. Cow price in dollars per unit of live weight ($/cwt) and price ratio were the 2 outcome variables used in the analyses. Price ratio was created aiming to remove seasonality effects from BP (BP divided by the national average price for its respective month and year of sale). The association between price ratio and carcass merit traits was investigated using canonical correlation analysis, and the effect of life history events on both BP and price ratio was inferred using a multiple linear regression technique. More than 70% of the cows were culled in the first 3 lactations, with an average live weight of 701.5 kg, carcass weight of 325 kg, and dressing percentage of 46.3%. On average, cull cows were sold at $57.0/cwt during the period considered. The canonical correlation between price ratio and carcass merit traits was 0.76, indicating that price ratio reflected carcass merit of cull cows. Later lactations led to lower BP compared with cows culled during the first 2 lactations. Injury, and leg and feet problems negatively affected BP. Productive variables demonstrated that the greater milk production might lead to lower cow prices. A large variation between farms was also noted. In conclusion, price ratio was a good indicator of carcass merit of cull cows, and life history events significantly affected sale BP and carcass merit of cull cows sold through auction markets.  相似文献

The objective was to measure the effects of cooling techniques (shade cloth vs. normal roof) on performance and physiology of 16 Friesian crossbred cows (87.5% Holstein Friesian × 12.5% Brahman) located at Sakol Nakhon Livestock Research and Testing Station, Department of Livestock Development, Ministry of Agriculture and Cooperatives (Sakol Nakhon, Thailand). They were divided randomly into two groups of eight. The two groups were used to evaluate the effects of modified roofing (normal roof fitted with woven polypropylene shade cloth) on the subjects' milk yield and reproductive performance under hot humid conditions. Results indicated that the modified roofing offered a more efficient way to minimize heat stress than the normal roof. The difference was sufficient to enable the cows to have a significantly lower mean rectal temperature and respiration rate (38.56°C, 61.97 breaths/min) than that of the cows housed under normal roofing (39.86°C; 85.16 breaths/min). The cows housed under modified roofing produced more milk (P < 0.05) but did not differ significantly in reproductive performance from the cows housed under normal roofing.  相似文献
