Blood samples were taken at weekly intervals from fifty four dairy cows (mainly Friesians and Jerseys) during the periods 8 weeks before and 8 weeks after calving. The cows were divided into two age groups; Group 1 consisting of 26 cows 2 1/2 to 4 years old and Group 2 consisting of 28 cows over 4 years old.The changes in erythrocyte magnesium (EMg) and plasma magnesium (PMg) concentrations before and after calving were expressed as the slope of each blood parameter regressed on weeks. The post-partum EMg concentration slope (increase) was greater (P<0.05) in the older cows than in the young ones. The older cows also had higher (P<0.05) mean EMg concentrations than the young cows. Between cow EMg concentration variations were significant (P<0.05) in both age groups while the weekly EMg concentration variations were only significant (P<0.05) in the older cows after calving. Age did not influence the changes in PMg concentrations before or after calving. Nor did the PMg concentrations vary significantly either between cows or between weeks. 相似文献

In the last few years, the detection of free radical damage and the body's defences against it have become increasingly important in clinical medicine as a complementary tool in the evaluation of metabolic status. The aim of this study was to evaluate, under field conditions, the anti-oxidant status of healthy cows during late pregnancy and lactation onset using two parameters: (1) plasma levels of malondialdehyde (MDA), a degradation product of lipid peroxidation, and (2) total antioxidant status (TAS). Results were compared with those obtained in another group of cows with lesser metabolic demands. We also investigated possible relationships between antioxidant status markers and other relevant blood parameters. Our results confirmed the characteristic metabolic changes associated with late pregnancy and early lactation. MDA and TAS provided an accurate reflection of the internal physiological status of the animal. The data indicated increased lipid peroxidation around parturition, but with wide individual variations that may be attributable not only to the physiological stage but also to unknown factors that will have to be further considered in future studies. 相似文献

The possibility that alterations in liver function may occur during late pregnancy as well as after calving has been investigated in healthy dairy cows and the results compared with those from non-pregnant non-lactating cows. There were significant alterations in plasma total bilirubin, glucose, total ketone body and urea concentrations, in bromsulphthalein clearance and in plasma aspartate aminotransferase and creatine kinase activities in periparturient cows compared with non-pregnant non-lactating cows. Of these, only the alterations in glucose, ketone bodies and urea concentrations and bromsulphthalein half-time and retention were markedly different before calving. The degree of fatty infiltration of the liver was significant two weeks before as well as two weeks after calving which differed from previous reports. It seems likely that the changes in the liver in dairy cows are functional and reversible and related to the metabolic demands of late pregnancy and early lactation. The incidence and severity of fat infiltration in the liver of Finnish Ayrshire cows at 1 and 8 weeks after calving were studied. All multiparous (n = 88) and some primiparous (n = 17) cows that calved in 20 commercial milk recorded herds were investigated. The cows lost weight and a condition score decreased significantly (p < 0.05) during early lactation. Blood samples were taken at 1, 4 and 8 weeks after calving. Ketone body concentrations were highest at 4 weeks after calving. Albumin and total protein concentrations and ornithine carbamyltransferase activity in the blood increased significantly (p = < 0.05) from 1 to 8 weeks after calving. The percentage of liver fat (v/v) was significantly (p < 0.05) greater at 1 (3.9 ± 0.5 %) than at 8 weeks (1.2 ± 0.3 %) after calving. Eighty-nine of 102 cows had less than or equal to 9 % of fat in the liver (mean 2.3 ± 0.2 %), and 13 cows had more than 9% (mean 14.9 ± 1.2%) at 1 week after calving. Only blood acetoacetate and plasma aspartate aminotransferase differed significantly between the groups. In the fatty liver group the incidence rate of treated cases of ketosis was 30.8 ± 13.3 % and of parturient paresis 23.1 ± 12.2 %. In the non-fatty liver group the rates were 10.1 ± 3.2 % and 7.8 ± 2.9 %, respectively. The differences were not significant. 相似文献

The effect of age on erythrocyte Mg, Na and K concentrations and the correlations between these parameters (EMg, ENa and EK) and other blood parameters were studied in female dairy cattle ranging from 1 week to over 8 years of age. EMg concentration was higher than plasma Mg, the concentration being highest in calves 1 week old and lowest in cows 3–4 years old. ENa and EK concentrations were lower and higher respectively in calves 1 week of age than in older animals and did not show any significant age variation in those between 1 and 8 years of age. The EK concentration was more highly correlated with erythrocyte parameters (PCV, RCC, Hb, MCV, MCH, MCHC) than was either EMg or ENa concentrations. 相似文献

OBJECTIVE: To determine whether administration of isoflupredone acetate (ISO) to healthy cows increases the frequency of severe hypokalemia and whether dexamethasone (DEX) has detectable mineralocorticoid properties. ANIMALS: 33 cows at 20 to 25 days of lactation. PROCEDURES: Cows were randomly allocated to 5 treatment groups and received 2 IM injections (on days 0 and 2) of sterile saline (0.9% NaCl) solution (10 mL each), an injection of ISO (20 mg) or DEX (20 mg) followed by 10 mL of saline solution, or 2 injections of ISO or DEX. Milk production was measured, physical examinations were performed, and blood and urine samples were collected daily on days 0 through 7. RESULTS: Physical examination parameters did not differ among groups; however, 1 cow developed atrial fibrillation on day 4. Both corticosteroids significantly increased plasma glucose concentrations, and ISO significantly decreased plasma potassium concentrations and increased total carbon dioxide concentrations with time. One dose of ISO decreased mean plasma potassium concentration by 25% on day 2, compared with day 0, and severe hypokalemia (serum potassium concentration < 2.3 mEq/L) developed in 1 of 6 cows. Mean plasma potassium concentration was 46% lower on day 3 than on day 0 in cows receiving 2 doses of ISO, and 5 of 7 cows became severely hypokalemic. Mean urinary fractional excretion of potassium significantly increased from that on day 0 in cows receiving 2 doses of ISO. CONCLUSIONS AND CLINICAL RELEVANCE: Both corticosteroids had glucocorticoid activity; however, only ISO had mineralocorticoid activity. Three late-lactation Holstein cows were used to determine the effects of environmental temperature on performance and energy partitioning. Each cow was housed in a respiratory chamber for 30 consecutive days and exposed to three different conditions of environmental temperature: (i) 20°C and 20°C (20°C), (ii) 25°C and 20°C (25°C), (iii) 30°C and 25°C (30°C) during the day and night, respectively. The temperature was switched in an interval of 10 days. Humidity in the chamber was maintained at 55–65% through the entire experimental period. The daily mean as well as morning and evening rectal temperatures of Holstein cows increased linearly ( P < 0.05) as chamber temperature increased. There was a significant linear reduction in dry matter (DM) intake ( P < 0.05) and an increase in DM digestibility ( P < 0.05). The response in milk yield, however, was not affected by heat stress. There were no significant differences among treatments for intake energy, heat production, net energy for lactation and net energy for gain. This results of this study disagreed with the assumption that late lactation cows gave priority to increasing body tissue at the expense of milk production under thermal stress. 相似文献

Lactation has been negatively associated with fertility because pregnancy rates in maiden heifers exceed those obtained after first or subsequent calvings. The extent of this difference is less in pasture-fed dairy cows (<10%) than in American Holsteins (>20%) fed grain and conserved forages. The latter cows have pregnancy rates to first insemination and oestrus detection rates of only 40 to 45%. This suggests that the subsequent fertility of inherently fertile Holstein heifers may be severely compromised by high levels of milk production. International comparisons show that pasture-fed dairy cows may experience extended periods of anovulatory anoestrum but have normal fertility (60% pregnancy rate to first insemination) once cycling. The high-producing American Holstein may ovulate within 4 weeks postpartum but is more likely to continue ovulating without being detected in oestrus. Both situations are associated with negative energy balances (NEB) during early lactation. The severity and duration of this NEB may vary with body condition at calving, age or parity, ration formulation, production level and environmental factors. Relative daily milk yield is not an absolute indicator of NEB, because some lower producing cows within a herd have lower feed intakes and more severe energy deficits. NEB is not simple to measure; nonetheless, it is correlated with genetic improvement for milk yield. A positive energy balance, greater weight gain and higher body condition score have all been shown to be positively correlated with plasma progesterone concentrations in early lactation. No studies have investigated the possibility that the rapid increase in metabolic rate at this time may also alter steroid concentrations with consequent effects on oestrous behaviour and fertility. Studies to more precisely define the effects of increasing milk yields in early lactation, especially in Holsteins, may need to be completed in Australia and New Zealand. Oestrus detection rates and pregnancy rates for American Holsteins of less than 50% are accepted widely in the USA. Such low detection rates confound studies on fertility. The objective should be to increase these 2 rates to at least 80% and 60% respectively. This may involve the use of controlled breeding, especially if oestrous behaviour is less overt in high-producing Holstein cows. 相似文献

The relationship between the concentration of plasma progesterone (P4) during embryo attachment or at recognition of pregnancy, and that of pregnancy-associated glycoprotein (PAG) was assessed in dairy cows. The outcome of artificial insemination (AI) was classified as positive (AI+), negative (AI?), or late embryonic mortality (EM) by measuring circulating PAG concentrations and by ultrasonography. Based on P4 concentrations at either day 21 or day 15, AI+ and EM cows were classified into ‘low’ (P4 concentrations < mean) and ‘high’ (P4 concentrations > mean) P4 groups. In both experiments, the threshold of P4 concentration between the ‘low’ and ‘high’ groups was approximately 6 ng/mL. PAG concentrations were lower in the ‘low’ group only when P4 concentrations were below the threshold. The study findings suggest that a possible P4 threshold exists below which PAG secretion may be impaired. 相似文献

Fifty primiparous Finnish Ayrshire cows were used in 2×2 factorial study. The factors were the protein-feeding strategy: constant (cCP) or variable (vCP) protein content in concentrate during lactation and the use of a commercial health product from 2 weeks pre- to 8 weeks postpartum. On treatment cCP, the CP content of the concentrate was 180 g/kg in dry matter (DM) throughout the lactation, and on vCP treatment, the CP content was 210, 180 and 150 g/kg DM during lactation days 0-100, 101-200 and 201-305, respectively. The concentrate blends included barley, oats, rapeseed meal, molassed sugar beet pulp and minerals and vitamins. Grass silage and ensiled wet sugar beet pulp (100 g/kg DM in TMR) were used as forage. The forage-to-concentrate ratio in TMR was 55:45 on both treatments. The experimental period was 305 days of the first lactation.The average daily milk yield (cCP: 26.3 kg/day vs. vCP: 27.5 kg/day) was higher ( P<0.05) when feeding concentrate with decreasing protein content, but the effect was not seen in an energy-corrected milk (cCP: 28.1 kg/day vs. vCP: 29.0 kg/day). Milk composition was not affected by the protein-feeding strategy. Protein-feeding strategy had no effect on DM intake (cCP: 18.5 kg/day vs. vCP: 18.9 kg/day). The efficiency of CP utilization (milk protein/CP intake) was higher (0.327 vs. 0.301; P<0.05) on vCP treatment during the last 100 days of lactation, but during the first 200 days of lactation, no significant difference was found. The average efficiency of CP utilization throughout the lactation was 0.32 with no difference between treatments. The protein-feeding strategy had no effect on the development of body condition scores of the cows. In mid-lactation, the cows on cCP treatment gained more weight than the cows on vCP treatment. The health product had no significant effect on production or feed intake. The results suggest that only small benefits can be obtained from feeding TMR to primiparous cows differing in CP content at various stages of lactation. 相似文献

Two experiments were undertaken to examine the direct and residual responses of late lactation (mean of 232 days calved) autumn calving dairy cows (Experiment 1), and late lactation (mean of 240 days calved) spring calving dairy cows (Experiment 2), to once-daily milking. Experiments 1 and 2 involved 50 and 44 Holstein–Friesian dairy cows respectively. In each of the two experiments, cows were milked either once daily (ODM) or twice daily (TDM), during the late lactation period (mean of 79 and 66 days in Experiments 1 and 2 respectively). Cows in Experiment 1 were managed within a grazing system, and were offered 3.0 kg of concentrate/day, while cows in Experiment 2 were housed, and offered grass silage supplemented with 6.0 kg concentrate/day. Forty-one cows from Experiment 1, and 32 cows from Experiment 2, completed the first eight weeks of the subsequent lactation on a twice-daily milking regime. Food intakes were not measured in Experiment 1, while treatment had no significant effect on dry matter intake in Experiment 2 ( P > 0.05). In each of Experiments 1 and 2, total milk output was increased with twice-daily milking ( P ≤ 0.05), while milk fat ( P ≤ 0.01) and protein ( P < 0.001) concentrations increased with once-daily milking. Somatic cell counts were higher with animals milked once daily in Experiment 1 ( P < 0.001), while not being significantly affected by milking frequency in Experiment 2 ( P > 0.05). Milking frequency had no significant effect on cow live weight or on cow condition score at the point of drying off in either Experiment ( P > 0.05). Milking time per cow during morning milking was unaffected by treatment in either experiment, while total daily milking time per cow (morning and evening combined) was significantly longer with the TDM treatment ( P < 0.001). In Experiment 1, milk flow rates during the morning milking were higher with animals on the ODM treatment, compared to those on TDM treatment ( P < 0.001), while being unaffected by treatment in Experiment 2 ( P > 0.05). Neither daily milk yield, milk fat content, milk protein content, or somatic cell count during the subsequent lactation, were affected by previous lactation milking frequency in either of Experiments 1 or 2 ( P > 0.05). 相似文献

The concentrations of carboxyterminal cross-linked telopeptide of type I collagen (CTx), osteocalcin (OCN), parathyroid hormone (PTH) and insulin-like growth factor (IGF-I) were measured in the blood serum of two age groups of mares during late pregnancy (70–50 days before foaling), during early lactation and at the peak of lactation (10–30 and 55–80 days after foaling, respectively). During late pregnancy, the PTH was higher in older (8–19 years old), compared to younger animals (3.5–4 years old) (P < 0.05). The OCN was higher in younger group during late pregnancy (P < 0.05). IGF-I was higher in the younger group during early lactation, in comparison to late pregnancy and the peak of lactation (P < 0.05). IGF-I did not differ between two age groups of mares. The results indicate on the differences in adaptation of bone metabolism to late pregnancy and lactation in older animals, in comparison to younger animals, reflected by elevated PTH secretion. 相似文献

This experiment was designed to evaluate the effects of different concentrate crude protein (CP) concentration on performance, metabolism and efficiency of N utilization (ENU) on early-lactation dairy cows grazing intensively managed tropical grass. Thirty cows were used in a ten replicated 3 × 3 Latin square design. The treatments consisted of three levels of concentrate CP: 7.9%, 15.4%, and 20.5% offered at a rate of 1 kg (as-fed basis)/3 kg of milk. The cows fed low and medium CP had negative balance of rumen degradable protein and metabolizable protein. Increasing CP tended to linearly increase DMI, 3.5% FCM and milk casein, and linearly increased the yields of milk fat and protein. Increasing CP linearly increased the intake of N, the concentration of rumen NH 3–N, and the losses of N in milk, urine, and feces. Increasing dietary CP linearly increased the molar proportion of butyrate but had no effect on the other rumen VFAs and no effect on microbial yield. In conclusion, feeding a concentrate with 20.5% of CP to early-lactation dairy cows grazing tropical grasses, leading to a 17.8% CP diet, tended to increase DMI, increased the yield of 3.5% FCM and the milk N excretion, and decreased ENU by 32%. 相似文献

The bone mineral content in two coccygeal vertebrae of 16 cows of Swedish Red and White Breed was measured with dichromatic photon absorptiometry during the period nine weeks prepartum to one week postpartum. For eight weeks before calving eight cows received 37 g Ca and 50 g P/day and the other eight, 76 g Ca and 50 g P/day. The mean bone mineral changes of the cows on the low Ca ration did not change. The cows on the high Ca ration had a slight increase (4 %) in bone mineral content to some days before calving. The postpartum bone mineral changes were, however, lower than the one prepartum. There was no significant difference between the mean bone mineral changes of the two groups. 相似文献

Data from an observational study, carried out during a 4.5 year period (1986–1990), were used to quantify the effects of health disorders on the risk of culling. The study population consisted of 47 commercial Holstein dairy herds from western France, comprising 4123 cows. Logistic regression was used to assess the relationships between health disorders and early and late culling. Fourteen main health disorders with clinical signs and one subclinical disease were studied: abortion, periparturient accident, calving provided with assistance, digestive disorders, ketosis, locomotor disorders, mastitis, metritis, milk fever, cystic ovaries, respiratory disorders, retained placenta, teat injuries, non-traumatic udder disorders and status with respect to milk somatic cell count. Adjustments were made for year, month of calving, parity, breeding value for milk, best of the two first milk production records and reproductive performance. The possible effects of interactions among variables were also studied. The herd effect was taken into account using random effect models. Non-traumatic udder disorders, teat injuries, milk fever and the occurrence of both ketosis and assistance at calving were significantly associated with an increased risk of being early culled (odds ratios (OR) ranging from 1.6 to 10.3). Early and late abortion, late metritis, poor peproductive performance, retained placenta, non-traumatic udder disorders within 45 days post-partum and mastitis occurring in the first 3 months of the lactation were positively associated with a late culling (OR ranging from 1.2 to 6.6). Cows with lower breeding value for milk and higher parities were high risk groups for culling. A lower level of milk production and occurrence of both reproductive disorders and poor reproductive performance were risk factors for late culling. 相似文献

为探究围产期亚临床酮病与泌乳奶牛繁殖性能及卵泡发育之间的关系,本试验选择了黑龙江省某大型集约化牛场开展试验,根据产后血酮水平确定亚临床酮病组和健康组奶牛共80头,该牛场亚临床酮病发生率为71%;两组奶牛跟踪到产后50 d进行直肠检查和B超检查了解卵泡发育状况,并记录繁殖性能数据。结果显示:与健康组奶牛相比,亚临床酮病组奶牛产后首次发情天数延长10 d,产后60 d内发情次数少0.58次;产后50 d子宫复旧不全高15%,与亚临床酮病呈显著正相关(R^2=8.74);产后60 d卵泡直径小3 mm(P<0.05)。结论:该牛场奶牛亚临床酮病发病率较高,是引起奶牛产后繁殖障碍的风险因素,也是造成卵泡发育受阻的不良因素,会导致奶牛繁殖力下降。 相似文献

Concentrations of penicillin-G in serum were determined after single intravenous doses of potassium penicillin-G (10 mg/kg body wt) to ewes and cows in late pregnancy and in early lactation. Penicillin-G in serum was analysed by a microbiological method and pharmacokinetic parameters were calculated by model-independent methods. Serum concentrations were lower in early lactation than in late pregnancy in both ewes and cows. The differences were statistically significant at most sampling times in ewes but only during the first hour in cows. Weight corrected values of clearance and volume of distribution were significantly higher in early lactation than in late pregnancy in both ewes and cows. Mean residence time and elimination-half life were shorter in lactating than in pregnant ewes, however for the latter parameter the difference was not statistically significant. Neither of these parameters changed in cows. It is concluded that in both ewes and cows pharmacokinetic parameters of penicillin-G are altered from late pregnancy to early lactation but that these changes have little practical impact and do not call for a revised dosage regime of the studied drug. 相似文献

As a result of research conducted in the US, recommendations for dry cow vitamin E intakes have increased seven fold there, however there has been no change to recommendations in the UK. As part of a larger study comparing the impact of existing UK and new US recommended vitamin E intakes on the health and fertility of commercial dairy cows in the UK, a study was set up to investigate the effect of route of supplementation and stage of lactation, over a 21 day period, on the response to mega-supplementation of cattle receiving supposedly adequate vitamin E. The study assessed the response of dry, peak lactation and mid lactation cows to in-feed or parenteral vitamin E supplementation (7 animals per treatment/lactation stage group) by measuring plasma and milk vitamin E concentrations, blood glutathione peroxidase (GSH-Px) activity and milk yields over a 21 day period. Plasma vitamin E concentrations were significantly influenced by a time, stage and treatment interaction ( P = 0.046). Both dry and lactating animals had significantly higher plasma vitamin E concentrations at some time points in the parenteral supplemented cows compared to the in-feed supplementated animals ( P ≤ 0.011 and P < 0.01, respectively). Milk vitamin E concentrations did not significantly differ between lactation stages but treatment had a significant effect on concentrations ( P < 0.008) when lactation stage was removed from the model. There was no significant difference in milk yield between treatment groups. A significant relationship between plasma and milk vitamin E concentrations was only found in the parenterally supplemented cows ( r = 0.435, P < 0.001). In cattle with intakes greater than the ARC recommendations, measurement of plasma vitamin E concentration may be of limited value in determining whether there has been a response to supplementation. The relationship between plasma and milk vitamin E concentrations is too poor for milk vitamin E concentrations to be used as a proxy for plasma vitamin E. 相似文献

This study was carried out to examine the changes in plasma concentrations of the Ca‐binding antimicrobial proteins S100A7 and S100A8 during pregnancy in dairy cows. Holstein Friesian cows ( n = 19) were inseminated with Holstein Friesian semen. Blood was collected at days 30, 60, 90, 120, 150, 180, 210, 240 and 270 after insemination. Plasma was used for measuring the concentrations of S100A7 and S100A8. Both S100A7 and S100A8 concentrations showed similar patterns during gestation; they increased during the midgestation, between days 90 and 180, and then declined before calving. The findings indicated that plasma concentrations of S100A7 and S100A8 did not change significantly during pregnancy in cows. Further studies are required to determine the roles of S100A7 and S100A8 in physiological function during pregnancy in dairy cows. 相似文献
