This study was conducted to estimate the dietary biotin requirement for maximum growth of zebrafish Danio rerio. Six isonitrogenous and isocaloric purified diets containing 0.031 (biotin‐unsupplemented diet), 0.061, 0.263, 0.514, 1.741 and 2.640 mg biotin kg?1 diet were fed in triplicate tanks for a total of 12 weeks to juvenile zebrafish (initial mean body mass 0.13 ± 0.001 g). From 4 weeks of feeding, fish fed diets with ≤0.061 mg biotin kg?1 showed biotin deficiency signs, such as retarded growth and skeletal deformity, was observed on some dead fish. At the end of the study, gill disorders were observed on some fish and liver glycogen accumulation were also observed in fish fed these diets. Fish fed the biotin‐unsupplemented diet exhibited a lower final body weight, protein efficiency ratio and feed utilization than the fish fed the biotin‐supplemented diets, whereas the highest values were observed with the diet containing 0.51 mg biotin kg?1 diet (P < 0.05). A linear relationship (r= 0.77; P < 0.0001) was observed between whole‐body biotin content and dietary biotin level. A broken‐line analysis indicated that the optimum dietary biotin content for maximal growth expressed as final body weight is 0.51 mg kg?1 diet.  相似文献

Present experiment was conducted to determine the effect of different feeds with varying protein levels on the growth, survival and reproductive performance of zebrafish, Danio rerio. The control diet (T1) was wild‐collected zooplankton from local fish ponds, while test diets with 350 g kg?1 protein (T2), 400 g kg?1 protein (T3) and 450 g kg?1 protein (T4) were formulated and fed to fish for a period of 210 days. The significantly (< 0.05) highest mean weight gain and specific growth rate were observed in T1, which were similar with T3 and T4. The significantly (P < 0.05) highest number of egg production per female and relative fecundity were found in T1, followed by T4 and T3 while T2 produced lowest number of eggs. No significant (> 0.05) differences were observed in brood survival rate, fertilization and hatching rate among the dietary treatments. The highest (< 0.05) fry survival rate was recorded in T1, followed by T3 and T4. Thus, it is suggested that control diet i.e. mixed zooplankton exhibited better growth, reproductive performance and fry survival rate. However, diet containing 400 g kg?1 crude protein also gave comparable results in terms of growth, survival and reproductive performance of zebrafish.  相似文献

Probiotic influence on fish immune response and digestive capacity is extensively discussed in aquaculture. In this experiment, a feeding trial was carried out for 100 days to evaluate the cross‐effects of probiotic supplementation and rearing temperature (17, 20 and 23°C) in juvenile seabass (Dicentrarchus labrax). The experimental diet was supplemented with a commercial probiotic blend (Biomin AquaStar Growout) at 3 g/kg diet (5.23 × 10CFU/kg diet), and tested against a non‐supplemented diet (control). Growth performance and innate immune responses were analysed at 70 and 100 days of feeding, whereas digestive enzyme activities were determined at 100 days of feeding. At the end of the feeding trial, fish were subjected to a handling stress and cumulative mortality was recorded. Digestive enzyme activities were influenced by temperature, with α‐amylase and lipase activities peaking at the higher temperature (23°C) and trypsin at the lower temperature (17°C). Immune parameters showed a significant temperature versus feeding duration effect, with complement system (ACH50) and peroxidase peaking at 70 and 100 days of feeding, respectively. Poststress cumulative mortality was higher at the lowest temperature (17°C), especially in fish fed the control diet. In conclusion, water temperature was the main variable affecting the studied parameters, whereas the dietary probiotic supplementation had influence on the chymotrypsin activity and survival rate in seabass reared at 17°C.  相似文献

Taurine (Tau) has been regarded as a conditional essential nutrient for some fish species. Although its role has been extensively studied in higher vertebrates, limited results are reported with fish especially its role on reproductive performance and the ontogenic changes on Tau levels throughout the life cycle. Therefore, we designed a feeding trial using zebrafish as a model species to test whether Tau supplementation to plant protein diets would have a positive effect on growth and reproductive performance. Zebrafish were fed plant protein diets containing graded levels of Tau (0.2, 4.6, 5.9 and 13.7 g/kg diet) from 10 days post fertilization (dpf) to sexual maturity. An additional commercial diet was used as a positive control for performance. The trial followed a completely randomized design with five treatments (diets) and three replications. After 60 days of feeding, growth, Tau concentration in the body, redox status, lipid body composition, reproductive and offspring performances were analysed. Tau supplementation did not affect growth and/or reproductive performance; however, zebrafish seems to differently modulate Tau concentration according to the growth stage. Tau seemed to induce a hypolipidemic effect in zebrafish by reducing lipid accumulation in their bodies (p < .05). A trend to a more pro‐oxidant effect of Tau supplementation was observed by the decreased reduced glutathione levels. In sum, Tau does not affect growth and reproductive performance of zebrafish but it is important for normal lipid utilization and redox status.  相似文献

Zebrafish (Danio rerio) is a laboratory model organism used in different areas of biological research including studies of immune response and host–pathogen interactions. Thanks to many biological tools available, zebrafish becomes also an important model in aquaculture research since several fish viral infection models have been developed for zebrafish. Here, we have evaluated the possible use of zebrafish to study infections with fish viruses that have not yet been tested on this model organism. In vitro studies demonstrated that chum salmon reovirus (CSV; aquareovirus A) and two alloherpesviruses cyprinid herpesvirus 1 (CyHV‐1) and cyprinid herpesvirus 3 (CyHV‐3) are able to replicate in zebrafish cell lines ZF4 and SJD.1. Moreover, CSV induced a clear cytopathic effect and up‐regulated the expression of antiviral genes vig‐1 and mxa in both cell lines. In vivo studies demonstrated that both CSV and CyHV‐3 induce up‐regulation of vig‐1 and mxa expression in kidney and spleen of adult zebrafish after infection by i.p. injection but not in larvae after infection by immersion. CyHV‐3 is eliminated quickly from fish; therefore, virus clearing process could be evaluated, and in CSV‐infected fish, a prolonged confrontation of the host with the pathogen could be studied.  相似文献

The present study was conducted to investigate the effects of Bacillus pumilus on the growth performance, innate immunity and digestive enzymes of the giant freshwater prawn, Macrobrachium rosenbergii. Prawns (mean weight: 1.81 ± 0.01 g) were fed different levels of Bpumilus at doses of 0, 1 × 107, 1 × 108 and 1 × 109 CFU/g to produce one control group and three experimental groups (G1, G2 and G3), respectively. After a 60‐day experimental feeding period, the results showed that the final weight, weight gain rate (WGR) and specific growth rate (SGR) were significantly higher in experimental groups than in the control (p < 0.05), whereas these improvements did not differ significantly (p > 0.05) among experimental groups. The improvements in phagocytic activity, respiratory burst (RBs) activity and the activity of catalase (CAT), nitric oxide synthase (NOS) and acid phosphatase (ACP) were significantly higher in G2 and G3 than in the control and G1 (p < 0.05). Further, the activity of CAT, NOS and ACP significantly increased with an increase in the dietary level of B. pumilus (p < 0.05). The activity of phenoloxidase (PO) in G3 was significantly higher than that in the control (p < 0.05). The activity of amylase was significantly higher in the groups G2 and G3 than that in the control group. G3 improved the activity of protease compared with the control (p < 0.05). In conclusion, the results of this study indicated that the potential use of B. pumilusat a dose of 1 × 108 CFU/g feed could improve the growth, immunity and digestive enzymes of M. rosenbergii.  相似文献

This study was conducted to investigate the effects of dietary inulin on growth performance, diet utilization, survival rate, carcass composition and digestive enzymes activities (amylase, lipase and protease) of carp (Cyprinus carpio) fry (0.55 ± 0.02 g). After acclimation, fish were allocated into 9 tanks (40 fish per tank) and triplicate fish groups were fed, control diet (0 g) or diets containing 5 g and 10 g inulin kg?1 for 7 weeks. No significant effect on growth performance and diet utilization of fish fed inulin compared with the control group was observed. However, supplementation of inulin significantly increased survival rate and carcass lipid content, while carcass protein content significantly decreased. Dietary inulin had no significant effects on digestive lipase, protease and amylase activities.  相似文献

A six‐week feeding trial was conducted to determine the effect of butyric acid (BA) inclusion in diet on performance of Barramundi (Lates calcarifer). Fish (12.0 ± 0.2 g) were fed with four experimental diets contained 0.0 (control), 2.5, 5.0 and 10.0 g BA/kg in triplicate. Fish fed on 5 and 10 g BA/kg diets had higher growth and total alkaline protease and lipase activities than the other treatments. Fillet protein content in BA‐supplemented groups was higher than the control. Liver catalase activity was highest in 2.5 g BA/kg group compared with the other groups. The values of liver superoxide dismutase activity and serum total protein concentration in 5 and 10 g BA/kg treatments were higher than those in the control (p < .05). Moreover, the highest and the lowest levels of serum lysozyme activity were noticed in the 10 g BA/kg and the control groups, respectively (p < .05). Also, the highest respiratory burst activity was observed in fish fed 5 g BA/kg feed. Supplementing diet with 2.5 or 5 g BA/kg resulted in higher serum haemolytic activity in fish compared with the other groups. The number of red and white blood cells increased in fish fed BA‐supplemented diets compared with the control (p < .05). In conclusion, the results of this study showed inclusion of BA at 5 g/kg diet in L. calcarifer juveniles improved growth and upgraded the general health condition by enhancing fish antioxidant enzyme activities and haemato‐immunological responses.  相似文献

Litopenaeus vannamei is economically important shrimp species in worldwide aquaculture. This study was conducted to assess the effect of different levels of probiotic Pediococcus pentosaceus (PP) on growth performance, feed utilization, digestive enzyme activity, intestinal microflora count and body composition of L. vannamei. Four diets containing different concentrations [0 (PP0), 106 (PPI), 107 (PPII) and 108 (PPIII) CFU/g] of PP were formulated. After 8 weeks feeding trial, the experimental shrimps were challenged with Vibrio anguillarum and noted the surveillance. At the end of the feeding trial, the obtained results revealed a significant increase (< .05) in final body weight, final length, weight gain (WG), survival rate, protease and amylase activities, lactobacillus sp. and Bacillus sp. intestinal count, total haemocyte counts (THC) and lysozyme activity in PPIII group when compared with the other groups. Similarly, WG, amylase activity, Bacillus sp. count, THC and lysozyme activity were significantly enhanced in case of PPII compared to the control group (< .05). Interestingly, FCR and Vibrio sp. counts were significantly decreased in case of PPIII group when compared to the other groups (< .05). Also, significant level of surveillance was noted in the challenging test with V. anguillarum. These results suggested that the P. pentosaceus improved the growth performance, digestive enzyme activity, immunity and tolerance against V. anguillarum of L. vannamei.  相似文献

Lactic acid bacteria (LAB) have a crucial role in inorganic selenium metabolism as well as their known desirable effects on fish. In this study, the synergistic effects of dietary sodium selenite and Pediococcus acidilactici on growth performance, intestinal bacterial counts, selenium bioavailability, hepatic antioxidant enzyme thioredoxin reductase activity and hepatic glycolytic enzyme activity that is hexokinase, phosphofructokinase and pyruvate kinase, and non‐specific immune response such as serum lysozyme and complements C3, C4 and ACH50 activity in rainbow trout (Oncorhynchus mykiss) were investigated. Thus, a 3 × 3 factorial design experiment was conducted with nine purified diets including three levels of sodium selenite (0, 1 and 2 mg/kg) and three levels of P. acidilactici (0, 7 and 9 log CFU/g). After 8 weeks of feeding, weight gain and specific growth rate were increased by increasing dietary sodium selenite and P. acidilactici levels compared to control (p < .05), whereas feed conversion ratio and condition factor was decreased by increasing dietary sodium selenite and P. acidilactici amounts in comparison with control (p < .05). Survival rate was not significantly affected among the experimental treatments (p > .05). Total cultivable bacterial populations after 4 and 8 weeks of the feeding trial were not significantly different among the dietary treatments, while LAB levels were higher in P. acidilactici‐fed groups than in control and selenium‐fed groups (p < .05). Selenocysteine, methylselenocysteine and selenomethionine levels in the intestine of rainbow trout were increased by increasing the sodium selenite and P. acidilactici levels (p < .05), and selenocysteine was found the most selenium species in the trout intestine. The quantity of total selenium in the whole body, intestine, blood, liver and muscle of rainbow trout were increased by increasing the amounts of sodium selenite and P. acidilactici compared to control (p < .05). Hepatic thioredoxin reductase and hexokinase activity were increased by increasing dietary selenium and P. acidilactici levels in comparison to control (p < .05), whereas phosphofructokinase and pyruvate kinase activity in the liver of rainbow trout were not significantly different between the dietary treatments. Serum lysozyme, complements C3, C4 and ACH50 activity were enhanced by increasing dietary selenium and P. acidilactici levels compared to control (p < .05). The most synergistic effects of dietary supplements on growth and metabolism of rainbow trout were obtained at 2 mg/kg sodium selenite and 7 log CFU/g P. acidilactici. The findings revealed the synergistic effect of dietary selenium and P. acidilactici on growth and metabolism in rainbow trout (O. mykiss).  相似文献

This study was conducted to investigate the prebiotic effect of different Dietary Fibre Concentrates (DFC) (Mucilage =MG; Pectin = PN or β‐glucan + mannan = βg + M) on growth and somatic parameters, body composition and digestive enzyme activities of jundiá (Rhamdia quelen). After acclimation, fish (7.16 ± 0.06 g) were allocated into 24 tanks (30 fish per tank) and triplicate groups were fed with Control diet (0 g kg?1 of DFC); diet supplemented with 5 g kg?1 commercial prebiotic (CP) or diets supplemented with 5 or 10 g kg?1 diet of MG; PN or βg + M. At the end of the trial (8 weeks), growth was significantly (P < 0.05) higher in fish fed diets supplemented with DFCs and did not differ from animals supplemented with CP. The animals that were fed Control diet presented a body protein content higher compared to those supplemented with diets containing pectin or β‐glucan + mannan (P < 0.05). However, fish fed diets added with β‐glucan + mannan yielded a higher level of protein deposited in the whole body. The activity of digestive enzymes was lower in the group supplemented with Pectin. Results indicate that supplementation with DFCs in the diet had positive effects on the performance of jundiá and are prebiotic potential candidate.  相似文献

This study was conducted to determine the effects of dietary cellulase addition on improving the nutritive value of Chlorella for juvenile crucian carp Carassius auratus (initial body weight: 2.99 ± 0.02 g, mean ± SEM). Five isonitrogenous and isoenergetic experimental diets were formulated to contain 0.0 (control), 0.5, 1.0, 1.5 and 2.0 g kg?1 cellulase, respectively. Each experimental diet was randomly assigned to triplicate groups with 25 juvenile fish per fibreglass tank for 8 weeks. The results showed that weight gain, specific growth rate, feed intake and the trypsin activity in the anterior intestine increased with increasing dietary cellulase to 1.5 g kg?1 and then declined with further addition. However, the mRNA expression levels of Mrf4 and Myf5, the apparent digestibility coefficients for dry matter, protein, energy and the majority of amino acids, and the activity of lipase in the anterior intestine were highest in fish fed the 1.0 g kg?1 cellulase diet, and then tended to decline with further cellulase supplementation. In conclusion, the optimal dietary cellulase supplementation level was 1.0–1.5 g kg?1, which can improve growth performance, digestive activities and nutrient digestibility in crucian carp.  相似文献

In aquaculture, exposure to stressful conditions and problems related to diseases often occur resulting in serious economic loss. The use of probiotics that control pathogens through different mechanisms, aiding digestion by exoenzyme supply and establishment of beneficial microflora in the digestive tract has got increased attention recently. Two different bacterial species Micrococcus MCCB 104 and Bacillus MCCB 101 were tested as probiotics for two cichlid species Etroplus suratensis and Oreochromis mossambicus. The strains were administered separately and together, at a rate of 103 CFU animal?1 day?1 for 28 days. The effects on digestive enzyme activity and growth were compared with those fed on control diet. FCR of E. suratensis was high in case of Bacillus alone as well as the mixed culture (Micrococcus and Bacillus together) than that of control. However, the SGR of E. suratensis was slightly lower than that of control during 14 days trial. SGR of both the treatments (Bacillus alone and mixed culture) was higher than that of the control on 28th day, and this might be due to the reason that specific duration is needed for the assimilation and conversion of nutrients into proteins. The intestinal digestive hydrolases of E. suratensis were profoundly enhanced mainly by Bacillus and mixed cultures. The hepatopancreatic versions of major digestive enzymes in E. suratensis were influenced variously by different probiotic treatments. The alpha‐amylase activity of hepatopancreas has been elevated significantly by mixed culture during 14 days experiments while on 28 days trials both Bacillus and mixed culture showed an enhancing effect. During 14 days feeding trials, the intestinal lipase activity has been prominently enhanced by all probiotic treatments. It is found that, in O. mossambicus, both 14 days and 28 days feeding experiments, the pepsin as well as intestinal digestive enzymes such as amylase, total alkaline protease, lipase and their hepatopancreatic counter parts showed a significant elevation in their specific activity. Intestinal alpha‐amylase and total alkaline proteases of O. mossambicus showed an increase in specific activities on both 14 days and 28 days feeding experiments with the Bacillus and Micrococcus combination. The hepatopancreatic enzymes were highly influenced by the Bacillus species in O. mossambicus. The results of this study suggest that live probiotic microorganisms such as Micrococcus MCCB 104 and Bacillus MCCB 101 may be incorporated while formulating the cost‐effective, nutritionally balanced diet of E. suratensis and O. mossambicus for better growth performance and nutrient utilization.  相似文献

Along with the growing number of laboratories that work with zebrafish (Danio rerio), it is necessary to have animals with good sanitary quality. Specific pathogens can interfere with the experimental results and in the life quality of the animals. Pseudoloma neurophilia is a parasite with high potential for interference in behavioural, morphology, toxicological and genetic research, and is very common in zebrafish facilities. With that, we implemented a protocol for the pathogen elimination in a genetically modified lineage (prop 1) using eggs from specific pathogen-free (SPF) wild-type fish (AB line) for in vitro fertilization, along with water recirculation equipment disinfection, appropriate PCR screening and back crossing protocols. This resulted in SPF prop 1 heterozygotes, which allowed us to move forward with subsequent crossings to develop homozygote prop 1 mutants for our research. Hence, this demonstrates a useful strategy for an individual research laboratory to rederive a specific mutant free line that is not available from other SPF laboratories.  相似文献

This experiment aimed to investigate the effects of exogenous multienzyme complex (EC) on growth performance, digestive enzyme activity and non‐specific immunity of the Japanese seabass, Lateolabrax japonicus (initial weight 27.09 ± 0.08 g). EC includes protease, xylanase, glucanase and mannase. Japanese seabass were given six levels of EC (0, 0.5, 1.0, 1.5, 2.0 and 2.5 g/kg) for 28 days. Results show that EC significantly enhanced the weight gain rate and specific growth rate (p < .05), while the feed conversion ratio reduced significantly (p < .05). Activities of lipase and trypsin in liver and intestine significantly increased (p < .05). Alkaline phosphatase, superoxide dismutase and lysozyme activities in serum and liver significantly increased (p < .05), while the content of malondialdehyde in liver significantly declined (p < .05). Regression analysis showed that the optimal supplementation of EC in WGR, SGR, FCR, SOD and LZM activity in serum was 1.66, 1.67, 1.81, 1.71 and 1.53 g/kg, respectively, while the best SOD, LZM activity in liver, trypsin activity in liver and intestine supplement were 1.64, 1.51, 1.81 and 1.97 g/kg. In conclusion, EC supplemented can improve the growth performance, digestive enzyme activity and non‐specific immunity of Japanese seabass, and it is recommended that the optimal supplementation of EC in diets of Japanese seabass is 1.5–2.0 g/kg.  相似文献

This study was performed to determine the effect of starvation and delayed feeding on activities of digestive enzymes and alkaline phosphatase (ALP) of larval red swamp crayfish (Procambarus clarkii), so as to reveal the tolerance to prolonged starvation and the recovery of digestive enzymes after delayed feeding in larval and juvenile P. clarkii. In the control group, activities of trypsin and ALP increased significantly (< .05) with day‐age and then kept constant at 24 days after hatching (DAH) and 10 DAH, respectively, whereas the activities of amylase and pepsin increased firstly then decreased with day‐age, and the activity of lipase increased firstly then decreased and then increased again during the development period of juvenile P. clarkii (1–31 DAH). In the group with continuous starvation (CS), activities of pepsin and lipase both decreased (< .05) after fasting, and the activities of pepsin, lipase and trypsin in the groups with delayed feeding all increased (< .05) and recover to the levels of the control group after food supply. However, the activity of amylase increased (< .05) in the CS group, and it decreased to normal level after food supply. The ALP activity did not significantly (> .05) vary after starvation, whereas it decreased in the groups with delayed feeding after 1 day of food supply, and then increased back to the level similar with the control group. Results from this study could provide information for diet preparation and feeding regime in larval and juvenile red swamp crayfish culture.  相似文献

A feeding trial was carried out to determine effects of dietary supplements of inactivated Pediococcus pentosaceus (PP) on growth, feed utilization and blood characteristics of red sea bream. Five diets containing different concentrations [0 (PP0), 1.6 × 1010 (PPI), 1.6 × 1011 (PPII), 1.6 × 1012 (PPIII) and 3.2 × 1012 (PPIV) cells g?1] of PP were fed to fish for 56 days. At the end of the feeding trial, the obtained results revealed a significant increase (P < 0.05) in weight gain, specific growth rate and skin mucus secretion in all PP‐supplemented groups when compared with control group. Moreover, fish fed PPIII and PPIV diets resulted in significantly higher final body weight, feed efficiency ratio and bactericidal activity than those of fish fed PP0 diet (< 0.05). Further, fish fed the PPIII diet also had a higher feed intake, protein efficiency retention, haematocrit, mucus lysozyme activity (LA), plasma and serum proteins than those of PP0 fed group (< 0.05). Similarly, fish fed PPIV diet resulted in a significant increase in serum LA, peroxidase activity and tolerance against low‐salinity stress test (< 0.05). Considering these promising results, we suggest that the supplementation of PP Prep improves growth and health conditions of Pagrus major.  相似文献
