A longitudinal study was carried out to investigate the diversity and persistence of Mycoplasma hyopneumoniae (M. hyopneumoniae) strains in four infected pig herds. In each herd, 20 pigs were randomly selected and blood and/or bronchoalveolar lavage (BAL) fluid was collected at 6, 10, 14 and 26 weeks of age. In the BAL fluid, quantitative PCR and MLVA (multiple-locus variable number of tandem repeats (VNTR) analysis) testing were performed for detection and typing of M. hyopneumoniae strains, respectively. At 26 weeks of age, the prevalence and severity of lung lesions were recorded at slaughter (minimum 50 pigs belonging to the same batch as the investigated pigs). The percentage of pigs testing positive on qPCR increased from 35% at 6 weeks to 96% at 26 weeks of age. With MLVA testing, positive pigs were found from 14 weeks onwards. Within each herd, only one distinct strain was detected, although clonal variants were identified in two herds. In three of the herds, the strain remained present until slaughter age. The percentage of pigs with Mycoplasma-like lesions ranged from 38% to 98%, and the average pneumonia score ranged from 1.7 to 11.9, respectively. The present field study documented that within a herd, mainly one distinct M. hyopneumoniae strain was present that persisted in the same animals for at least 12 weeks. This implies that the immune response of the animals following infection is not able to rapidly clear the infection from the respiratory tract.  相似文献

Epidemiological aspects of Mycoplasma hyopneumoniae (Mh), influenza H1N1 and H3N2 viruses, and Aujeszky's disease virus (ADV) were investigated in slaughter pigs from 50 fattening pig herds. Herd factors as potential risk indicators for respiratory disease were obtained by means of a questionnaire. At slaughter, blood samples were collected from each herd, and the proportion of seropositive pigs per herd was assessed for each of these pathogens. The median herd-level seroprevalence of the agents were: Mh 88%, H1N1 100%, H3N2 60% and ADV 90%. The percentage of herds in which all investigated fattening pigs were seronegative for these agents was: Mh 0%, H1N1 0%, H3N2 12% and ADV 18%. The percentage of herds in which all investigated fattening pigs were seropositive for these agents was: Mh 8%, H1N1 71%, H3N2 22% and ADV 40%. A positive association was found between influenza H1N1 and H3N2 viruses, and a negative association between influenza H3N2 virus and ADV. There were no risk indicators for the seroprevalence of Mh. Three risk indicators were associated with the seroprevalence of influenza H1N1 virus: a fully slatted floor, an increasing number of pigs in the municipality and dry feeding. Three risk indicators were found for the seroprevalence of influenza H3N2 virus: purchase of pigs from > or = two herds, an increasing number of pigs in the municipality and natural ventilation. The seroprevalence of ADV was influenced by two risk indicators: an increasing number of pig herds in the municipality and an increasing number of pigs per pen.  相似文献

This study was conducted to compare the effects of a preventive in-feed medication programme using tilmicosin (Pulmotil 200 premix, Elanco Animal Health) at 200 p.p.m. with those of Mycoplasma hyopneumoniae (Mh) vaccination programme (Stellamune Mycoplasma, Pfizer Animal Health). A pig herd with chronic respiratory disease in which infection with Mh played an important role was selected, and a total of 204 piglets were randomly allocated to either the medication (P) or the vaccination (V) group. Pigs in the P group received medicated feed for 3 weeks after weaning (days 34-55), and for 2 weeks late in the nursery period (days 77-98). The piglets in the V group were vaccinated twice intramuscularly, at 4 and 22 days of age. The two groups were compared on the basis of average daily gain (ADG), feed conversion rate (FCR), additional curative medication days (CMD), overall mortality (major variables), a coughing index, pneumonia lesions, and serology against Mh, influenza H1N1 and influenza H3N2 viruses, Actinobacillus pleuropneumoniae (App) and porcine reproductive and respirator, syndrome virus (PRRSV) (minor variables). No significant differences (P > 0.05) were observed for ADG (555 g/day in P group; 567 g/day in V group), FCR (2.64 in P group; 2.41 in V group) and mortality rate (11% in P group; 7% in V group). The average number of additional curative medication days (CMD) per pig was significantly higher (P < 0.01) in the P group (1.5) than in the V group (0.58). At slaughter age, the serological results and the prevalence of macroscopic lung lesions were comparable in the two groups (P > 0.05). With the exception of CMD, the preventive use of tilmicosin at this swine farm was found to confer similar beneficial effects to Mh vaccination.  相似文献

In July 2007, PRRS was detected for the first time in Sweden. A total of eight positive herds were identified and various measures were taken to eradicate the disease, including restrictions and slaughter of infected herds. Subsequently, both active and passive surveillance activities were undertaken. This study describes stochastic scenario-tree modelling of all the various surveillance system components, to estimate the current probability that Sweden is free from PRRS. The model includes all actions taken after the first positive herd was detected. The surveillance system components included in the model were as follows: investigations undertaken in association with the outbreak, a serological study based on samples collected at slaughter, samples collected in the national PRRS surveillance programme and passive clinical surveillance. The probability of freedom was estimated in time steps of 1 month, from July to December 2007. After each time step, the calculated posterior probability of freedom from the previous month, combined with the probability of introduction, was used as a prior probability for the next month.The result from the model showed a 99.8% probability that Sweden was free from PRRS at the end of December 2007. The estimated total sensitivity of the surveillance system varied between 81.2% and 94.3% and was highest during the first months after the outbreak. For sensitivity analysis purposes, the model was also applied using higher risks of introduction. However, this did not make considerable difference to the final estimates.  相似文献

The milk yields of 1824 cows were used to investigate the effect of lesion-specific causes of lameness, based on farmer treatment and diagnosis of lame cows, on milk yield. A three-level hierarchical model of repeated test day yields within cows within herds was used to investigate the impact of lesion-specific causes of lameness (sole ulcer, white line disease, digital dermatitis and other causes) on milk yield before and after treatment compared with unaffected cows. Cattle which developed sole ulcer (SU) and white line disease (WLD) were higher yielding cattle before they were diagnosed. Their milk production fell to below that of the mean of unaffected cows before diagnosis and remained low after diagnosis. In cattle which developed digital dermatitis (DD) there was no significant difference in milk yield before treatment and a slightly raised milk yield immediately after treatment. The estimated milk loss attributable to SU and WLD was approximately 570 and 370 kg, respectively. These results highlight that specific types of lameness vary by herds and within herds they are associated with higher yielding cattle. Consequently lesion-specific lameness reduction programmes targeting the cow and farm specific causes of lameness might be more effective than generic recommendations. They also highlight the importance of milk loss when estimating the economic impact of SU and WLD on the farms profitability.  相似文献

The program for eradication of pseudorabies virus (PrV) from swine herds in Illinois was evaluated with respect to compliance with Livestock Conservation Institute (LCI) guidelines for selection of intervention strategies and for the effectiveness of these interventions under different herd conditions. The sample consisted of 395 swine operations quarantined between 1988 and 1994. These herds were followed until the end of 1996. The association of herd characteristics (number of sows, sow PrV seroprevalence, type of housing, number of PrV-seropositive farms within 1.5 mi) at the time of quarantine with the producer's selection of an initial intervention strategy (vaccination, offspring segregation, test-and-removal, depopulation-and-repopulation) was analyzed using logistic multiple regression. The interaction of herd characteristics with intervention strategies to affect the duration of quarantine was analyzed using multivariable Cox regression.

Factors favoring selection of vaccination were increased herd size, higher sow PrV seroprevalence, and more PrV-seropositive farms within 1.5 mi. Offspring segregation was preferred when sow PrV seroprevalence was higher, and test-and-removal was preferred when seroprevalence was lower. Depopulation-and-repopulation was more likely in outdoor operations. Except for depopulation-and-repopulation, selection of an intervention strategy was in accordance with LCI guidelines.

Vaccination and offspring segregation were associated with longer times under quarantine, and test-and-removal and depopulation-and-repopulation with shorter times. Test-and-removal was more effective in reducing the duration of quarantine when sow PrV seroprevalence was low. Vaccination increased the duration of quarantine less when sow PrV seroprevalence was high. Vaccination increased the duration of quarantine more when there were one or more PrV-seropositive farms within 1.5 mi than when there were no PrV-seropositive farms within 1.5 mi. It is apparent that herd characteristics affect the duration of quarantine and therefore need to be taken into account in the selection of a PrV-intervention strategy.  相似文献
