An experiment was performed to evaluate the efficacy of potassium permanganate (KMnO4) as a prophylactic and therapeutic treatment of an experimental subacute infection of Flavobacterium columnare in channel catfish, Ictalurus punctatus . Fish were cutaneously abraded and divided into five treatment groups: (i) challenged by waterborne exposure to F. columnare and not treated with KMnO4 (positive control), (ii) challenged and simultaneously treated with KMnO4, (iii) challenged and treated with KMnO4 at 1, 6 and 9 days post-challenge, (iv) not challenged and treated with KMnO4 at 1, 6 and 9 days post-challenge (first negative control) and (v) not challenged and not treated (second negative control). The dosing of KMnO4 was 2.0 mg L−1 above the potassium permanganate demand for 2 h duration. The survival of the group challenged and simultaneously treated with KMnO4 (99%) was significantly higher than the positive control (78%) and was not significantly different from the negative control groups. The challenged fish treated with KMnO4 post-challenge had 7% higher survival than the positive control (85% compared with 78%), but that difference was not statistically significant. The results demonstrate that KMnO4 has a clear prophylactic value but probably a marginal therapeutic value once the infection has established.  相似文献

Rainbow trout Oncorhynchus mykiss (261.6 × 24.7 g initial weight, mean × SEM) at 13.1 × 0.2 C were exposed for 94 d to one of three CO2 treatments: control (22.1 × 2.8 mg/L), medium (34.5 × 3.8 mg/L), or high (48.7 × 4.4 mg/L). Trout were checked daily for survival, and fish were sampled at 0, 28, 56, and 84 d for physiological responses, growth, and fillet quality assessments. Trout were also challenged to a 15-min crowding stress at 93 d to assess their ability to initiate a stress response during hypercapnia. Chronically exposed trout showed nearly 100% survival through 84 d exposure (1 of 1,500 fish died). Growth and physiological results showed that increasing elevated CO2, concentrations result in corresponding decreased growth rates and CO2specific physiological parameters: The medium and high CO2 treatments had significantly slower growth and subsequently smaller fish by 84 d. Exposed trout also showed significantly ( P < 0.05) decreased plasma chloride for medium and high CO2 treatments compared to the control from 28 through 84 d. Decreased growth and smaller fish in the medium and high CO2 treatments resulted in correspondingly smaller fresh and smoked fillet weights. Chronic CO2 exposure did not result in notable changes in ultimate muscle pH. Exposure to 15-min crowding stress at 93 d resulted in significant changes in hematocrit, plasma cortisoI, glucose, and chloride for all treatment groups. CO2-specific changes were detected in hematocrit, plasma cortisoI, and plasma chloride responses following the 15-min crowding stress.  相似文献

Copper sulfate (CuSO45H2O) is used to reduce the abundance of blue-green algae and combat off-flavor in channel catfish culture. Copper sulfate usually is applied at a concentration of one-one hundredth of the total alkalinity. A study was performed at the Auburn University Fisheries Research Unit to determine the duration of elevated copper (Cu) concentration following copper sulfate applications. Two alkalinity treatments, 20-40 mg/L and 110-130 mg/L (as CaCO3), were examined. Copper sulfate was applied biweekly for 14 wk at 03 mg/L for the low alkalinity treatment and 1.2 mg/L for the high alkalinity treatment. Total copper concentrations in pond waters declined to the background level by 48-h post treatment. In addition, total copper concentrations were determined in waters of 38 catfish production ponds located in west central Alabama. The mean and standard deviation were 0.0092 ± 0.0087 mg Cu/L. Copper quickly precipitates from the water or is absorbed by sediments following copper sulfate treatment. Although concentrations of copper in pond waters increase immediately following copper sulfate treatment, they rapidly decrease and seldom exceed the United States Environmental Protection Agency's National Recommended Water Quality Criteria for Priority Toxic Pollutants of 0.013-mg Cu/L. Findings of this study suggest that copper sulfate treatment will not contaminate effluent from catfish ponds because of the short time that applied copper remains in the water column. Furthermore, the most frequent applications of copper sulfate occur in late summer months when rainfall is minimal and pond overflow is rare.  相似文献

A study of the biochemical oxygen demand (BOD) of waters from ten channel catfish ponds at Auburn, Alabama, revealed that the 5-d BOD (BOD5) seldom exceeded 8 mg/L and that the ultimate BOD (BODu) was usually less than 30 mg/L. Water samples from catfish ponds usually needed to be diluted only 2 or 3 times to permit BOD5 measurements, and nitrification occurred even during a 5-d incubation period. Catfish pond waters were not extremely high in ammonia nitrogen concentration, and ammonia nitrogen introduced in the ammonium chloride-enriched dilution water caused an appreciable increase in BOD of some samples. Plankton respiration is a major component of carbonaceous BOD (CBOD) in catfish pond waters. Thus, the BOD is not expressed as rapidly during 5-d incubations as in typical waste-water. The ultimate BOD (BODu) would be a good measurement of oxygen demand for catfish pond effluents, but it is difficult to measure. Data from this study suggest that BODu can be estimated from BOD5, but the correlation is not strong ( R 2= 0.62). An alternative is to develop a short-term BOD measurement specifically for effluents from channel catfish and other aquaculture ponds. This study suggests that a 10-d BOD conducted without nitrification inhibition or addition of ammonia nitrogen in dilution water might be a better alternative to standard BOD5 or BODu measurements normally used in wastewater evaluation.  相似文献

Three pairs of brood channel catfish Ictalurus punctatus were induced to spawn in aquaria supplied with flowing water from a reservoir filled with ground water (calcium hardness = 110 mg/L as CaCO3= 44 mg/L as calcium). Fertilized egg masses were allowed to incubate in aquaria for 6 h and each mass was then split into five portions. The five portions were then allowed to hatch and the resulting sac fry developed in waters with calcium hardnesses of 0, 1, 5, 10, or 100 mg/L as CaCO3 (0, 0.4, 2, 4, or 40 mg/L as calcium). Test waters were prepared from distilleddeionized water and reagent-grade chemicals; tests were conducted using static-renewal conditions. Survival from hatch to onset of exogenous feeding ("swim-up") averaged 62% in calcium-free water and 98% at all other calcium hardness levels. Wet weight gain, dry weight loss, and resistance to environmental hypoxia were significantly affected ( P < 0.05) by environmental calcium levels: best growth, yolk utilization rate (indicated by changes in dry weight and visual observation), and tolerance to low dissolved oxygen concentrations were found at calcium hardnesses of 10 and 100 mg/L as CaCO3. Based upon these results, a minimum calcium hardness of 10 mg/L as CaCO3 (4 mg/L as calcium) is recommended for channel catfish hatchery water supplies.  相似文献

Abstract. Survival of the early stages (egg, larva and juvenile) of the American lobster, Homarus americanus L., reared under controlled conditions has often been threatened by a variety of fungi and microbial epibionts. A preliminary study was conducted for the purpose of defining toxicity levels of malachite green for 5th stage juveniles during each of the following intermoult (or fused intermoult) states: A-B, C4, D0 and D2-D3 Irrespective of exposure time or concentration, all the lobsters treated during intermoult state A-B died. This contrasts with seemingly good survival of lobsters in intermoult states C4, D0 and D2- D3 treated at concentrations of 900 mg/1 malachite green for exposure periods of 10 min. The findings suggest that an effective treatment combination for lobsters immersed in malachite green may be one which is employed during intermoult state C4, for about 6min and at concentrations of about 700–800 mg/l.  相似文献

The primary objective was to compare thyroid hormones and levels of carp pituitary extract for the artificial production of female Ictalurus punctatus × male I. furcatus hybrid catfish. The effects of different carp pituitary extract dosage rates (5, 6, 9, and 10 mg/kg), carp pituitary extract (6 and 10 mg/kg) supplemented with the thyroid hormones thyroxine (T4) and triiodothyronine (T3), or pregnenolone (DHP) were determined for inducing ovulation of female channel catfish, fertilization of channel catfish eggs with male blue catfish sperm, and hatching rate of these embryos. Hormone treatments thyroxine and tri-iodothyronine with carp pituitary extract, carp pituitary extract alone, and pregnenolone with carp pituitary extract used to artificially produce hybrid catfish were not different in terms of ovulation rates, eggs/kg, fry/kg body weight of female channel catfish, fertilization rates, or hatching rates (P > 0.05). These findings suggest that lower amounts of carp pituitary extract may be used to induce spawn of female channel catfish for production of channel-blue catfish hybrids and the addition of thyroid and steroid hormones is ineffective at the rates used in this study.  相似文献

Pcnaeus monodon juveniles (35.4 ± 2.2 mm TL) were exposed to seawater (20 ppt) having different concentrations of total ammonia (NH3+ NH,+) and nitrite. Median lethal concentration (LC50) was determined with static-renewal tests. The LC50, of total ammonia-N, NH3-N and niMte-N on shrimps decreased with increase of exposure time. The 24, 48, 72, % and 120 h LC50 were 94.96, 61.09, 47.47 45.58 and 38.00 mg/L total ammonia-N (2.68, 1.73, 135, 1.29 and 1.08 mg/L NH3-N) and 215.85, 185.33, 88.54, 54.76 and 37.97 mg/L nitrite-N, respectively. The 144h LC50 of total ammonia-N and NH3-N was 36.71 mg/L and 1.04 mgL. The "threshold" of ammonia and nitrite was found at 144h and 120h respectively, from the toxicity curve approaching asymptote. A "safe value" was 3.7 mg/L total ammonia-N, 0.1 mg/L NH3-N (20 ppt, pH 7.70, 27C) and 3.8 mg/L nitrite-N for P. monodon juvenile .  相似文献

Application of readily-oxidizable organic substrate to laboratory soil-water systems and fish ponds caused anaerobic conditions in bottom soil and water, and concentrations of soluble reactive phosphorus (SRP) increased. Aeration of ponds increased total phosphorus (TP) concentrations by suspending soil particles in the water, but SRP concentrations declined because of increased oxy- genation of bottom water and soil, Alum [Al2(SO4)3·14H2O] treatment of ponds reduced SRP and TP concentrations in ponds, but the low concentration of alum used, 20 mg/L, had little residual effect on phosphorus concentration. Application of agricultural limestone at 0.2 kg/m2 to ponds with soil pH of 5.5 and Ca2+ concentration of 5 mg/L did not affect SRP and TP concentration. Unless pond soils were anaerobic at their surfaces, a condition not acceptable in thermally-unstratified fish ponds, soils released little phosphorus to the water. Strong adsorption of phosphorus by soils in intensive ponds with feeding is beneficial, because removal of phosphorus by aerobic soils is a control on excessive phytoplankton growth. In fertilized ponds, phosphorus must be applied at frequent intervals to replace phosphorus removed from the water by soils.  相似文献

These studies were conducted to evaluate the efficacy of a live attenuated Edwardsiella ictaluri vaccine against enteric septicemia of catfish. In one study channel catfish fingerlings (72 d of age post hatch) were immersed for 30 min in water containing E. ictaluri RE-33 at dosages of 1 × 106, 1 × 107 and 2 × 107 CFU/ML of water. No mortalities were observed following vaccination. Following exposure to virulent Edwardsiella ictaluri the cumulative mortality of fish vaccinated with dosages of at least 1 × 107 CFU/mL were significantly lower than that of non-vacccinated fish in both laboratory and field challenges. Vaccination with 1 × 106 CFU RE-33mL provided some protection during the laboratory challenge but failed to protect fish under field conditions. In a second study, vaccination of 6 full-sib families of channel catfish at a vaccine dosage of 1 × 107 CFU/mL resulted in a relative percent survival among families ranging from 67.1 to 100%. Significant differences in mortality were found among families and between vaccinated and unvaccinated groups, but there was no family by vaccine interaction. Families with the highest mortality after vaccination were also shown to have the highest mortality without vaccination (r = 0.82; P = 0.04).  相似文献

Uptake and clearance of Edwardsiella ictaluri in the peripheral blood of channel catfish Ictalurus punctatus fingerlings were monitored for 216 h after exposure to E. ictaluri for 4 h and 8 h under static conditions. Most fish exposed to E. ictaluri developed bacteriemia 24 h post-exposure, and by 72 h post-exposure E. ictaluri was recovered from all the blood of all exposed fish. The number of E. ictaluri colony forming units (CFU) in the blood of moribund fish ranged between 1.7 × 103 to 1.6 × 105 CFU/50 μL whole blood. Clearance of bacteria from the blood was observed by 216 h post-exposure and all fish surviving bacterial exposure developed agglutinating antibody against E. ictaluri . The pathogenesis of the infection was accompanied by the shedding of viable E. ictaluri into the water which may serve as a mechanism by which fish to fish transmission occurs.  相似文献

Abstract Two groups of European catfish, Silurus glanis L., fingerlings were infected with channel catfish virus (CCV) by either intraperitoneal injection with 105 TCID50 of CCV, or bathing in water containing 105 TCID50 of CCV per 1·0 ml. The virus was isolated from spleen, intestine and brain of CCV-injected fish at day 1 and the titres ranged from 102·1 to 103·3 TCID50/g. However, the tissue distribution of CCV was irregular and no virus was isolated after day 3 post-exposure. In CCV-bathed fish, the virus was isolated only from the liver of one specimen at day 3 post-exposure. No clinical signs of CCV disease developed in any of the fish. Specimens in each regime from all sampling periods showed some minor histopathological changes, but there were no differences between treatments. Lesions included oedema and focal haemorrhage in the liver and the spleen was congested. Electron micrographs of tissue samples showed the presence of a few virus particles around the nuclei of kidney, spleen and intestinal cells, and in or around a myelinated nerve within the optic lobes of infected fish during the first 4 days of infection.  相似文献

Abstract.— Subadult tiger shrimp Penaeus monodon (30.15 ± 2.19 g) were exposed individually in sea water of 25%o to 0 (control), 1.11, 4.76, 10.06 and 19.09 mg/L copper for 24 h. Hemolymph pH, PO2 (partial oxygen pressure), PCO2 (partial carbon dioxide pressure), HCO3, oxyhemocyanin and protein levels were determined. Hemolymph PO2 increased, whereas hemolymph pH, PCO2, HC03-, oxyhemocyanin. protein and the ratio of oxyhemocyanidprotein levels decreased with increasing concentrations of ambient copper in the range of 1.11 to 19.09 ma/L. It is concluded that P. monodon following 24-h exposure to ambient copper as low as 4.76 mg/L shows a reduction of oxyhemocyanin, protein, and the ratio of oxyhemocyanin to protein, and develops metabolic acidosis in the hemolymph.  相似文献

The present experiment was conducted to study growth and tissue responses in Atlantic salmon, Salmo salar L., fed a fish meal based diet supplemented with copper (Cu). The findings of the experiment were used to evaluate the need for dietary Cu supplementation. Atlantic salmon parr, initially weighing ≊ 7.5 g, were randomly distributed among 10 tanks, with 300 fish in each tank. Duplicate groups of fish were fed a fish meal based diet containing 3.5 mg Cu kg−1, or this diet supplemented with 5, 10, 50 or 100 mg Cu kg−1 (as CuSO4*5 H2O) for 12 weeks. Growth was recorded and blood haemoglobin measured. The Cu concentrations in whole body, liver, serum and selected muscle samples were measured, as was liver selenium (Se) concentration.
There were no difference in growth among the dietary treatments. There were, however, significant differences among the dietary groups in liver Cu and Se concentrations. The fish fed the diet supplemented with 5 mg Cu kg−1 had increased liver Cu concentration compared with the other groups. Similar trends were found for serum Cu concentration and whole-body Cu concentration, but these effects were not significant. Liver Cu and Se concentrations were positively correlated and liver Se concentrations were inversely correlated to dietary Cu concentration, confirming an interaction between these two elements in salmon.
There may be a positive effect of a modest Cu supplementation level, and we suggest that a small amount of Cu (5–10 mg kg−1) should be added to fish meal based diets.  相似文献

A generalized nitrogen budget was constructed to evaluate the potential role of mangrove sediments as a sink for dissolved inorganic nitrogen (DIN) in shrimp pond effluents. DIN concentrations were measured in pond effluents from three semi-intensive shrimp ponds along the Caribbean coast of Colombia between 1994–1995. Mean NH4+ concentrations in the discharge water for all farms were significantly higher (67 × 12 μg/L) than in the adjacent estuaries (33 × 8 μg/L). Average NH4+ concentrations in the pond discharge over all growout cycles were similar, representing an approximate doubling in relation to estuarine water concentrations. In contrast, NO2-+ NO3- concentrations were similar in both pond effluent and estuarine waters. Dissolved inorganic nitrogen loading of the ponds was similar. The estimated reduction of DIN in pond effluents by preliminary diversion of outflow to mangrove wetlands rather than directly to estuarine waters would be × 190 mg N/m2 per d. Based on this nitrogen loss and depending upon the enrichment rate, between 0.04 to 0.12 ha of mangrove forest is required to completely remove the DIN load from effluents produced by a 1-ha pond.  相似文献

Abstract.— West Texas saline groundwaters were assessed as potential media for culturing red drum, Sciaenops ocellatus . In 30-d bioassays with juvenile red drum (0.4–3.0 g), highest survival was 85% in a 5-ppt (parts per thousand = g/L) salinity, high-sulfate (1,723 mg/L SO4-2), high-calcium (427 mg/L Ca±2) groundwater from a windmill catchment pond in Pecos County. The lowest survival was 0% in a 35-ppt saline groundwater from a gravel pit, also in Pecos County. In complementary bioassays of low-salinity (<5 ppt) groundwaters from the same region, calcium chloride (CaCl2) addition to a 3-ppt. Reeves County ground-water low in Ca±2 resulted in the greatest increase in survival (0–93). It was unclear whether the beneficial effect of CaCl2 was a result of increasing Ca±2 concentration (36–336 mg/L), increasing Cl- concentration (639–1,296 mg/L). or both. The concentration of total dissolved solids ("salinity") appears to be the single most important consideration in estimating the red drum aquaculture potential of a groundwater, with the optimum salinity being 5–15 ppt. Concentrations of arsenic (As), cadmium (Cd), chromium (Cr), mercury (Hg), lead (Pb), and selenium (Se) tended to be higher in west Texas groundwaters than in typical U. S. surface waters. However, based on current maximum tolerable daily intake recommendations, red drum cultured for 30 d in west Texas groundwaters contained whole-body As, Cd, Cr, Hg, Pb, and Se concentrations that would not pose a substantial health risk to adult human consumers.  相似文献

Abstract. Rainbow trout exposed to environmental free C02 concentrations ranging from 12–60 mg/1 for a minimum of 275 days developed nephrocalcinosis. Gross, histopathological and chemical examinations demonstrated that prevalence and severity of nephrocalcinosis increased with increasing CO2 concentrations. The condition was characterized by calcification and granuloma formation in kidney and stomach. Diet, but not water, PO4 -P concentration appeared to modify the prevalence and severity at each CO2 concentration tested.  相似文献

Abstract. Ultrastructural changes occurring in juvenile channel catfish, Ictalurus punctatus Ratinesque, hepatocytes were examined following exposure to carbon tetrachloride (CCl4). Juvenile catfish were intraperitoneally injected with a sublethal dose of undiluted CCl4and controls, used for comparisons, received physiological saline. The rough endoplasmic rcticulum (RER) was slightly dilated by 1 h and was significantly dilated by 4 and 24 h post-CCl4-exposure. At 4 h, the RER became degranulated, mitochondrial eristae indistinct with electron dense flocculant matrieal deposits, and nuclear membranes indented. Autophagic vacuoles were present in some hepatocytes. Within 48 h post-exposure, the hepatocytes were indistinguishable from control hepatocytes. CCl4 can induce reversible or irreversible cell damage which appears dose dependent.  相似文献

Juvenile red drum Sciaenops ocellatus were tank-reared in two, separate 30-d trials to assess effects of total water hardness and chloride concentratlon on growth, survival, and feed conversion. Two levels of total hardness and chloride were used in 2 × 2 factorial designs to obtain total hardness and chloride levels in trial one of 100 and 200mg/L CaCO3 and 125 and 250mg/L Cl, and hardness and chloride levels of 100 and 400mg/L CaCO3 and 150 and 500mg/L Cl in trial two. In trial one, average daily growth and feed conversion were significantly ( P ≤ 0.05) improved at 100mg/L hardness levels compared to 200mg/L at both chloride levels. Average daily growth was 0.11 ± 0.02g/d at 100mg/L hardness and 125mg/L Cl compared to 0.06 ± 0.01g/d at 200 mg/L hardness and 125mg/L chlorides. No significant ( P ≥ 0.05) difference in survival could be attributed to levels of total hardness or chloride. In trial two, 500 mg/L Cl treatments produced significantly ( P ≤ 0.01) better growth, survival and feed conversion than 150mg/L Cl at either level of total hardness (100 and 400mg/L). Survival at 150mg/L chloride and 100mg/L hardness was 48.8 ± 18.7%, while at 500mg/L chloride and 400mg/L hardness survival increased to 96.6 ± 2.1%. Growth also increased from 0.14 ± 0.03 to 0.27 ± 0.01g/d and feed conversion improved from 5.77 ± 1.56 to 1.87 ± 0.06 at the same levels. No interactions between levels of hardness or chlorides were shown to affect red drum growth, survival, or feed conversion in either trial.  相似文献

Abstract. An enzootic, Australian, atypical strain of Aeromonas salmonicida isolated from diseased goldfish, Carassius auratus (L.), was inoculated into Atlantic salmon, Salmo salar L., brown trout, S. trutta L., rainbow trout, S. gairdneri Richardson, and brook trout, Salvelinus fontinalis (Mitchill), fingerlings by intraperitoneal injection (i.p.) and by bath challenge, the latter with and without prior abrasion of skin. The 10-day LD50 (i.p.) was estimated to be 7·4 × 10-3 colony forming units (cfu) for Atlantic salmon, 3·0 × 10-2 cfu for brown trout, 3·7 × 102 cfu for brook trout and 6·4 × 103 cfu for rainbow trout. Brown, rainbow and brook trout succumbed to bath challenges with between 105–106 cfu/ml, developing ulcers of the skin and septicaemia. The organism was trasmitted from inoculated fish to five of 195 within-tank control fish via water and established a carrier state in one of 14 Atlantic salmon. It was concluded that the organism poses a significant threat to the salmonid farming industry and wild salmonid fisheries in Australia.  相似文献
