Clinically healthy calves (aged 3-6 weeks) were exposed to defined ambient temperature for 4 hours (cold: 5 degrees C, 60% humidity, n = 12; warm: 35 degrees C, 60% humidity, n = 11). During the exposure of each animal in a climatic chamber, certain parameters of lung function, respiratory mechanics, blood gas analysis, circulation, metabolism and thermal regulation were registered simultaneously in order to study immediate physiological consequences of different environmental conditions. In comparison to control calves (18-20 degrees C, 60% humidity, n = 13) an insufficient adaptation of these young calves was noticed in both cold and warm conditions. At 5 degrees C, marked changes in lung function were observed, i.e. airway constriction, pulmonary hypertension, and ventilation-perfusion-mismatching leading to hypoxemia and hypercapnia. Due to compensation by the circulatory system, a sufficient O2-consumption of the organism as well as an unchanged body temperature were maintained. At 35 degrees C, the respiratory pattern changed to panting and a higher dead space ventilation. No changes were observed in pulmonary gas exchange and blood arterialisation. Due to hyperventilation, the partial pressure for CO2 decreased in blood. Since the body temperature increased continuously, thermal regulation was insufficient. This situation would have led to animals collapsing after a period of heat stress lasting longer than 4 hours. In conclusion, young calves up to the age of 6 weeks were not able to tolerate acute changes in ambient temperature. This was true for cold conditions (5 degrees C) as well as for hot conditions (35 degrees C). The results of this study should be taken into account in order to optimise transport and farming conditions.  相似文献

A series of studies were carried out to determine the effects of increasing environmental temperature (20 to 28 to 33 °C) at a constant (60%) relative humidity on the physiological responses, energy and nitrogen balance of prepubertal Holstein heifers (n = 4, initial body weight = 198 ± 3 kg). The relatively high environmental temperatures of 28 and 33 °C increased rectal temperature by 0.2 and 1.2 °C and increased respiration by 23 and 58 breaths per min, respectively. At 28 °C, dry matter intake was similar to that at 20 °C, whereas at 33 °C dry matter intake was lower (P < 0.10) by about 9%. However, the dry matter digestibility was higher (P < 0.05) at 33 °C than at 20 °C, hence the thermal treatments did not affect daily weight gain. At 33 °C blood urea nitrogen increased whereas hematocrit and plasma glucose concentrations decreased. At 33 °C energy and nitrogen intake were lower (P < 0.10), while energy and nitrogen digestibility were higher at 33 °C than at 20 °C. Urinary nitrogen was elevated at 33 °C resulting in lower nitrogen retention. Although, energy retention as fat did not differ (P > 0.10) among the treatments, energy retention as protein was lower (P < 0.05) at 33 °C than at 20 °C or 28 °C. These results confirmed that the effects of environmental temperature on prepubertal heifers were severe above 28 °C.  相似文献

试验共选用135头日龄相近的断奶仔猪(40日龄左右),随机将其分到3个日粮处理组中。本试验分两期进行,第1期进行3个重复试验,其中阉公猪2个重复,小母猪1个重复,该期阉公猪和小母猪平均初始体重分别为9.91±1.37kg和10.4±0.86kg;第2期进行2个重复试验,其中阉公猪和小母猪各1个重复,二者平均初始体重分别为9.83±1.00kg和9.57±1.00kg。3个处理日粮分别在基础日粮中添加一定比例的药物添加剂:处理1组为基础日粮+10mg/kg那西肽,处理2组为基础日粮+20mg/kg效美素,处理3组为基础日粮+12mg/kg恩拉霉素。两期试验持续期均为7d,并且都是从断奶后第2d开始进行的。统计结果表明,处理1组仔猪平均日增重显著优于处理3组(p=0.048),而平均日采食量与处理3组相比有提高的趋势(P0.10)。对于其他生产性能指标,各处理组之间差异不显著。从数值上看,处理1组仔猪的采食量和日增重都是最佳的,而饲料效率与处理2组相当,处理3组仔猪在生产性能上的表现最差。不同药物对阉公猪和小母猪增重的影响不尽一致,总体上看小母猪平均日增重比阉公猪大,但这也可能与小母猪初始体重较大有关。从腹泻情况看,各处理组仔猪腹泻频率不存在显著差异。考虑药物的相对价格以及对仔猪生长和健康的影响,在该期饲料中添加10mg/kg那西肽是值得推荐的。  相似文献

1. Two experiments are described in which laying pullets maintained at constant temperatures were fed a range of diets with a view to defining optimum combinations of temperature and nutrient intake. 2. In the first experiment, all combinations of 6 temperatures (15 degrees, 18 degrees, 21 degrees, 24 degrees, 27 degrees and 30 degrees C) 9 diets (three protein concentrations and three energy contents) and two stocks were tested for 34 weeks using 4320 pullets. In experiment 2, all combinations of three rearing temperatures, three laying temperatures (18 degrees, 22.5 degrees and 27 degrees C) three diets (protein concentration) and two stocks were tested for 61 weeks using 2160 pullets. 3. As anticipated, higher dietary protein concentrations were needed to maintain egg output at higher temperatures. If diets suplying adequate amino acid intakes were provided, egg output was unaffected by temperatures in the range 15 degrees to 27 degrees C although, at the highest temperature, egg weight was slightly reduced and rate of lay (particularly in the later part of the laying year) was increased. At 30 degrees C, egg output was depressed whichever diet was fed. 4. Dietary energy content had small but significant effects on egg weight and egg output but did not interact with temperature. It was not possible to maintain egg weight or egg output at 30 degrees C by feeding a high energy, high protein diet. 5. Estimated heat output of the birds increased during the course of the experiment at the lower temperatures but decreased with time at 30 degrees C. Feather loss occurred earlier at the lower temperatures and this is interpreted as an effect of temperature on the timing of the annual moult, which also accounts for the better persistency of lay observed at 27 degrees C.  相似文献

以健康荷斯坦黑白花奶牛为试验对象,研究不同环境温度下(5℃、10℃、15℃、20℃、25℃)对奶年乳清的某些生化指标的影响。研究结果表明,环境温度为20℃时,乳清天门冬氨酸氨基转移酶(AST)活性最高,显著高于其它各组;乳清肌酸激酶(CK)活性在5℃和25℃时极显著高于各中间温度组;丙氨酸氨基转移酶(ALT)从5℃至20℃逐渐升高,且20℃组显著高于25℃组;乳清总蛋白(TP)含量5℃组为最低,而10℃、15℃和25℃组为高;乳清中尿素氮(BUN)则随环境温度的升高呈逐渐降低的趋势,且25℃组显著低于其它各温度组。  相似文献

The aim of this study was to investigate the impact of weight gain of calves within the first 3 weeks of life on health status and subsequent performance. Holstein bull calves were reared either intensively (IR; individual hutches and ad libitum milk feeding for the first 3 weeks of life; n = 24), or according to the established protocol [ER; 4 l milk/day in hutches during week 1 and 720 g/day milk replacer (MR) from day 8 to 21 in a group pen; n = 24]. Water, hay and concentrates were freely available to all calves. From week 4, calves of both groups were housed together in a group pen and fed 720 g MR/day; step‐down weaning was performed between week 5 and 10. Key metabolic blood parameters were analysed on day 2, 12, 21 and 70 of life. After weaning, all animals were fed concentrates and corn silage until slaughter at an age of 8 months. Within the first 3 weeks, average daily weight gain was threefold higher in IR calves in relation to ER calves (1.28 vs. 0.38 kg/day, p < 0.001). Neither incidence nor duration of scouring differed significantly between groups. Starter intake (week 4–10) was higher in IR calves in relation to ER calves (49.7 vs. 38.0 kg/calf, p = 0.006). Serum glucose, urea, albumin and insulin were higher at an age of 21 days in IR calves in relation to ER calves; no differences were obvious at an age of 70 days. Plasma GH and IGF‐I concentrations revealed an uncoupling of the somatotropic axis in ER calves within the first 3 weeks of life. At slaughter, body weight of IR calves tended to be higher than that of the ER calves (320 vs. 309 kg, p = 0.07). In conclusion, intensive feeding and individual housing during the first 3 weeks of life had positive long‐term effects on subsequent performance.  相似文献

本研究采用免疫组织化学分析方法,测定了HSP70蛋白在意大利蝗卵子发生期的表达定位及33~42℃高温处理对其相对表达量的影响。结果表明,1)在卵黄发生前期,卵母细胞和滤泡细胞均表达HSP70蛋白,而在卵黄发生期和卵黄发生后期,HSP70蛋白阳性表达位于滤泡细胞;2)在33~42℃温度范围内,随着温度升高,意大利蝗卵子发生期HSP70蛋白相对表达量呈现先上升后下降的变化趋势;其中36℃处理组HSP70蛋白相对表达量最高,显著高于27℃对照组(P0.05);42℃处理组HSP70蛋白相对表达量最低,但与27℃对照组差异不显著(P0.05)。短时高温处理对意大利蝗卵子发生期HSP70蛋白相对表达量有显著影响,推测HSP70蛋白的表达在意大利蝗抵抗短时高温胁迫过程中具有重要作用。  相似文献

There is a large body of literature describing effects of environmental chemicals (ECs), many of them anthropogenic with endocrine-disrupting properties, on development in rodent laboratory species, some of which lead to impaired reproduction and adverse health. This literature joins extensive human epidemiological data and opportunistic wildlife findings on health effects of ECs. In contrast, the effect of endocrine disruption on foetal development and reproductive performance in domestic species is less extensively documented. This applies both to domestic farm and to companion species even though the former is critical to food production and the latter share our homes and many aspects of the modern developed human lifestyle. In domestic species, the nature of chemicals exposure in utero and their consequences for animal health and production are poorly understood. A complication in our understanding is that the pace of development, ontogeny and efficiency of foetal and maternal hepatic and placental activity differs between domestic species. In many ways, this reflects the difficulties in understanding human exposure and consequences of that exposure for the foetus and subsequent adult from epidemiological and largely rodent-based data. It is important that domestic species are included in research into endocrine disruption because of their (i) wide variety of exposure to such chemicals, (ii) greater similarity of many developmental processes to the human, (iii) economic importance and (iv) close similarities to developed world human lifestyle in companion species.  相似文献

The major purpose of this study was to investigate whether increasing milking frequency for a short period (21 days) increases milk yield and milk components and improves udder health throughout lactation in Turkish Saanen goats. Two groups were formed (control, n = 14, treatment, n = 14); balanced by parity, bodyweight and birth type to make them homogenous. Feeding and management practices were the same for both groups. The treatment group was milked four times a day (4×) for 21 days after weaning whereas the control group was milked twice a day (2×). All the animals were milked twice daily thereafter. The pyronin methyl green stain method was used to estimate the somatic cell count (SCC), which is an indication of udder health. This method provided a count that excluded RNA and background noise while including cells with DNA. Repeated measures analyses indicated that overall differences between the 2× and 4× groups were significant for the morning, evening and total test day milk yields, in that values for the 4× group were increased 14.7, 8.8 and 12.1%, respectively. Differences between the groups for SCC were not significant. The results of this study indicate that increasing milking frequency to 4×, even for a short time (21 days), increases overall milk yield throughout lactation in dairy goats.  相似文献

This study was conducted to determine the effects of low-protein diets supplemented with crystalline amino acids (AA) on performance and intestinal development in barrows (n = 32) over 2 weeks after weaning at 18 ± 1 day. Four maize-soybean meal based diets providing 0.93 g standardized ileal digestible lysine/MJ ME were prepared. The treatments were a control diet containing 23.1% CP (crude protein) and three low-protein diets (21.2, 18.9, and 17.2% CP, respectively), which were supplemented with crystalline AA to achieve an ideal AA pattern. Piglets were raised individually and had free access to feed and water. Average daily gain (ADG) and average daily feed intake (ADFI) were measured weekly. Severity of diarrhea was monitored twice per day. Blood from all piglets was taken for determining serum urea nitrogen on d 0, 7, and 14 and serum free AA concentration on d 14. At the end of the experiment, all piglets were killed to measure morphology of the small intestine and disaccharidase activities. Reducing CP level did not affect ADFI (P > 0.10) but resulted in poorer (linear, P < 0.05; quadratic, P < 0.05) ADG and feed:gain ratio (F:G) over the 2-week study period. However, most of the effect of dietary CP was due to the significant deterioration of performance with the 17.2% CP diet. Faecal consistency was improved linearly (P < 0.01) with dietary CP decrease. Reducing dietary CP from 23.1 to 17.2% resulted in a linear (P < 0.001) and quadratic (P < 0.001) decrease in serum urea nitrogen levels on d 7 and 14. Serum arginine (P < 0.001), glutamine (P < 0.05), and proline (P < 0.05) concentrations were decreased in pigs fed the 17.2% CP diet compared with those fed the control diet on d 14. Villous height was decreased (P < 0.05) in the duodenum and jejunum by the reduction of dietary CP. As dietary CP declined, lactase and sucrase activities were reduced (P < 0.01) in the proximal jejunum. In conclusion, reducing CP concentration from 23.1 to 17.2% led to decreased growth performance associated with morphological changes of the gut and reduced disaccharidase activities in the small intestine. But reducing dietary CP from 23.1 to 18.9% did not affect intestinal morphology and disaccharidase activities.  相似文献

It has been difficult to establish reliable indices of exposure to endocrine disrupting chemicals (EDCs) appropriate for a variety of avian species because of a vast array of reproductive strategies. Data from mammals, reptiles and fish provide insight on likely mechanisms of action for EDCs. However, many of the effects of EDCs are weaker than the actions of the native hormones, making it difficult to assess adverse effects in domestic and wild birds. It is clear that differential sensitivity to EDCs exists across species, due to the timing and mode of exposure, compound toxicity and age of the individual. Our studies on EDCs are conducted in the quail model system, with focus on reproductive endocrine, neuroendocrine and behavioral responses. Studies have included EDC exposure, either by egg injection or via diet. Results from egg injection studies showed the following: (1) estradiol administered by embryonic day 12 demasculinized male sexual behavior, altered hypothalamic neurotransmitters and reduced hen day production and fertility in a dose dependent fashion, (2) methoxychlor (MXC) or vinclozolin impaired male sexual behavior in adult quail and (3) DDE exposure impaired reproductive and immune related end points. Two-generation studies were conducted on Japanese and northern bobwhite quail with dietary methoxychlor (MXC) exposure (0, 5 and 10 ppm) beginning in adults (P1), continuing in their offspring (F1), with F2 offspring raised on control diet. MXC exposure impaired male sexual behavior, hypothalamic catecholamines and plasma steroid hormones. Moreover, MXC exposure had reproductive consequences observable at both the lower and higher doses of MXC in F1 and F2 generations. These data demonstrate that embryonic EDC exposure interferes with sexual differentiation of neural systems that direct reproduction.  相似文献

Methoxychlor (MXC) is a non-steroidal pesticide that is known to possess estrogenic activity, and therefore may be potentially hazardous to the development and/or reproduction. The present study assessed the effects of perinatal exposure to MXC on reproductive function after maturation in both male and female rats. Pregnant rats were fed a phytoestrogen-free diet containing different doses of MXC (24, 240, and 1200 ppm) from day 15 of gestation to day 10 after parturition, and reproductive functions of offspring were examined after maturation. In males, MXC exposure during the perinatal period decreased serum LH and FSH, but not testosterone levels, but it did not affect copulatory behavior. In females, MXC exposure prolonged the days exhibiting cornified vaginal smears during the estrous cycle. In addition, both the lordosis reflex and preovulatory LH surge on the presumptive proestrous evening were suppressed in MXC-exposed females. These results suggest that perinatal exposure to MXC exerts permanent effects on several aspects of the reproductive function in both male and female rats.  相似文献

Background

Studies have reported that heparin may be unsuitable as an anticoagulant in human plasma samples when quantifying cytokines using multiplex bead array assays. For mouse samples, multiplex assays have been validated for serum and EDTA-plasma, but it remains to be elucidated whether heparin influences the quantification of cytokines, and if so – to what extent. Furthermore, laboratory mice are often anesthetized for blood sampling, which causes acute stress that may influence circulating cytokine concentrations and thus bias experimental results. The objectives of the present study were to identify whether specific cytokine concentrations varied between heparin-plasma, serum, and EDTA-plasma, and whether short isoflurane anesthesia would influence the concentrations of these cytokines in the circulation. Twenty-three acute phase and pro-inflammatory cytokines were quantified in matched serum, EDTA-plasma, and heparin-plasma samples from anesthetized and unanesthetized male NMRI mice using a multiplex assay. In addition, samples from unanesthetized mice were spiked with three levels of heparin.

Results

The concentrations of five out of 23 cytokines were significantly different between sample types, but only one cytokine (IL-17A) differed between heparin-plasma and serum. When further spiking the heparin-plasma with increasing concentrations of heparin, there was a significant effect on 11 cytokines, where the cytokine recovery could be correlated to the heparin concentration for ten of these cytokines. Anesthesia resulted in lower concentrations of G-CSF, but had no significant impact on the concentrations of the other 22 cytokines.

Conclusion

In mice, heparin seems like a suitable anticoagulant for obtaining plasma for multiplex assays for the cytokines IL-1α, IL-1β, IL-2, IL-6, IL-9, IL-12p40, IL-12p70, IL-13, G-CSF, GM-CSF, IFN-γ, KC, MCP-1, MIP-1α, MIP-1β, RANTES and TNFα, but an effect of heparin in high concentrations should be considered for the cytokines IL-9, IL-12p40, IL-12p70, KC, MCP-1, MIP-1β and RANTES. Short isoflurane anesthesia had significant impact on G-CSF, but none of the other cytokines.  相似文献

In 5 experiments the weekly rhythm of the heat production of growing young broiler chickens kept in groups between their 5th and 57th days of life was measured according to the method of short-time measuring (30-minute measuring periods) in dependence on environmental temperature (ET) between 40 and 5 degrees C in stages of 5 K by means of gas exchange and indirect calorimetry. The keeping temperature ranged from one experiment to the other from 35, 30, 25, 20 to 15 degrees C. The dependence of heat production on ET is parabolic in the medium and high ranges of temperature. As a rule, heat production increases progressively with very low ET. Thermoneutral temperature depends on both, the age (live weight) of the chickens and environmental temperature. Thermoregulatory heat production decreases with growing age. It changed from 24 kJ/kg LW0.75.d.K at the beginning to 8 kJ/kg LW0.75.d.K at the end of the experiments.  相似文献
