Preventive and curative activities of postharvest treatments with selected chemical resistance inducers to control postharvest green (GM) and blue (BM) molds on oranges (cvs. ‘Valencia’ or ‘Lanelate’) artificially inoculated with Penicillium digitatum and Penicillium italicum, respectively, were evaluated. In vivo primary screenings to select the most effective chemicals and concentrations were performed with benzothiadiazole (BTH), β-aminobutyric acid (BABA), 2,6-dichloroisonicotinic acid (INA), sodium silicate (SSi), salicylic acid (SA), acetylsalicylic acid (ASA) and harpin. INA at 0.03 mM, SA at 0.25 mM, BABA at 0.3 mM and BTH at 0.9 mM were selected and tested afterwards as dips at 20 °C for 60 or 150 s with oranges artificially inoculated before or after the treatment and incubated for 7 d at 20 °C. Although it was an effective treatment, SSi at 1000 mM was discarded because of potential phytotoxicity to the fruit rind. Preventive or curative postharvest dips at room temperature had no effect or only reduced the development of GM and BM very slightly. Therefore, these treatments cannot be recommended for inclusion in postharvest decay management programs for citrus packinghouses.  相似文献

The efficacy of three antagonistic yeasts, Metschnikowia pulcherrima strain MACH1, M. pulcherrima strain GS9, and Metschnikowia fructicola strain AL27, against Penicillium expansum and patulin accumulation was evaluated on apples stored at room (22 ± 1 °C for 7 days) and cold temperatures (1 ± 1 °C for 56 days). To increase the potential range of application of the biocontrol agents (BCAs), their efficacy was evaluated on four cultivars of apple, i.e. ‘Golden Delicious’, ‘Granny Smith’, ‘Red Chief’ and ‘Royal Gala’. AL27 was more effective than MACH1 and GS9 in the control of blue mold rot and in the reduction of patulin accumulation. The efficacy of AL27 was in most cases similar to the chemical control used, making the antagonist as competitive as chemical fungicides. In vitro experiments showed that AL27 reduced the conidial germination and germ tube length of P. expansum more than the other strains. The three BCAs were more effective in the control of blue mold rot on ‘Golden Delicious’ apples than on the other tested cultivars.  相似文献

This study aimed to investigate the effect of chitin on the antagonistic activity of Rhodosporidium paludigenum against Penicillium expansum, the cause of blue mold in apple fruit, and the possible mechanisms involved. Our results showed that biocontrol efficacy and population growth of R. paludigenum were greatly enhanced when it was harvested from nutrient yeast dextrose with added chitin (NYCB) medium compared with that harvested from nutrient yeast dextrose (NYDB) medium. The ability of R. paludigenum produced in NYCB to induce resistance to blue mold in apple fruit was significantly enhanced. The enhanced disease control efficacy was correlated with higher levels of polyphenoloxidase (PPO) and superoxide dismutase (SOD) activities in apple fruit treated with R. paludigenum. Moreover, the SOD and catalase (CAT) activities of R. paludigenum were stimulated by cultivating in NYCB, while malondialdehyde (MDA) accumulation in the yeast cells was suppressed. These results indicated that adding chitin to normal media might be an effective method to improve the antagonistic activity of R. paludigenum and the active oxygen metabolism of R. paludigenum might be closely related to the biocontrol activity of the yeast.  相似文献

Penicillium expansum is one of the most important pathogens that cause blue mold in stored apples. Due to the development of resistance to the postharvest fungicide, thiabendazole (TBZ), an increase in blue mold has been observed in apple storages. The sensitivity of three TBZ-sensitive and three TBZ-resistant isolates of P. expansum to diphenylamine (DPA), scald inhibitor, was tested in vitro. Of the 94 isolates, collected later in the storage season, 41% were found resistant to both DPA and TBZ. To manage the fungicide resistance, a reduced-risk fungicide, fludioxonil, was tested against blue mold caused by TBZ-sensitive and -resistant P. expansum on ‘McIntosh’ apples treated with or without 1000 μg ml−1 of diphenylamine. Fruit were assessed for disease and scald incidence during storage. Diphenylamine controlled scald in treated fruit. Higher disease incidence of blue mold was observed in apples treated with diphenylamine and low concentrations of fludioxonil (3, 5, and 75 μg ml−1). DPA neither positively nor negatively affected the control of blue mold when DPA was applied together with 150, 300 and 600 μg ml−1 of fludioxonil during 12 weeks of storage at 4 °C.  相似文献

This study compared the effectiveness of practical grade chitosan when used in solution with acetic, glutamic, formic and hydrochloric acids, and a water-soluble commercial chitosan formulation, in controlling postharvest diseases of strawberry. The commercial chitosan formulation and other resistance inducers based on benzothiadiazole, oligosaccharides, soybean lecithin, calcium and organic acids, and Abies sibirica and Urtica dioica extracts were also tested. The commercial chitosan formulation was as effective as the practical grade chitosan solutions in the control of gray mold and Rhizopus rot of strawberries immersed in these solutions and kept for 4 days at 20 ± 1 °C. Moreover, the treatment with commercial and experimental resistance inducers reduced gray mold, Rhizopus rot and blue mold of strawberries stored 7 days at 0 ± 1 °C and then exposed to 3 days shelf-life. The highest disease reduction was obtained with the commercial chitosan formulation, followed by benzothiadiazole, calcium and organic acids. The compounds that provided the best results in postharvest applications to control storage decay of strawberries, should be tested in further trials through preharvest treatments, applied at flowering and a few days before harvest.  相似文献

The aim of this study was to investigate the ability of two salts, sodium carbonate and bicarbonate, to activate defence mechanisms in citrus fruit against postharvest green mould caused by Penicillium digitatum. In particular, once there was confirmed salt antifungal activity in the absence of direct contact with the pathogen, changes in enzyme activity and expression levels of chitinase, β-1,3-glucanase, peroxidase and phenylalanine ammonia lyase (PAL), and phytoalexin (scoparone, scopoletin, umbelliferone) and sugar (glucose, fructose, sucrose) contents in treated oranges were analyzed. Overall, sodium carbonate and bicarbonate increases the activity of β-1,3-glucanase, peroxidase, and PAL enzymes in orange tissues. Gene expression analyses confirmed PAL up-regulation particularly 12 h after treatment application. HPLC analyses of peel extracts showed increased amounts of the sugars and phytoalexins, compared to control tissues, with sucrose and scoparone being the most represented. The results suggest that, although salts exert a direct antifungal effect on P. digitatum, they are also able to induce citrus fruit defence mechanisms to postharvest decay. The defence response seems correlated with the up-regulation of the phenylpropanoid pathway, which has a role in the adaptation to various stresses. This response could result in natural reaction to wounding and pathogen attack in citrus, enhancing its protective effect. As a consequence, the fruit might have a better chance of successful defence against the decay.  相似文献

Penicillium digitatum is the most devastating postharvest pathogen of citrus. In addition, Penicillium expansum is the main pathogen of pome fruit, although recent studies have demonstrated its ability to infect oranges under some conditions. In this study, we evaluated wound response in ‘Valencia’ oranges harvested at three different maturity stages and the effect of wound response on the establishment of both pathogens when fruit were stored at two different temperatures (20 and 4 °C). The effect of wounding and pathogen inoculation on lignin content, was also quantified. Lastly, the expression of several phenylpropanoid pathway-related genes was also analyzed by semi-quantitative RT-PCR. Results indicated that, in general, P. digitatum exhibited lower decay incidence and severity as time between wounding and inoculation increased. Decay incidence and severity were higher in fruit from the over-mature harvest than in fruit from immature and commercial harvests. P. expansum was able to infect fruit at 20 °C but lesions were small compared to lesion size of fruit stored at 4 °C. Lignin content in wounded fruit (control) and in samples wounded and inoculated with P. expansum was highest in fruit from the immature harvest at 7 d post-wounding and inoculation. Wounded fruit had higher expression of pal1, comt1 and pox1 genes at 48 h than at 24 h. However, samples inoculated with P. digitatum showed lower expression at 48 h than at 24 h. Our results indicated that maturity and storage temperature play an important role in orange wound response.  相似文献

The feasibility of using a lyophilized culture filtrate (LCF) from Lentinula edodes at different concentrations to improve the efficacy of Pichia membranefaciens in controlling postharvest blue mold decay in apple fruit was investigated. Application of LCF alone was effective in inhibiting the blue mold rot in apple fruit wounds in a dose-dependent manner. Moreover, the biocontrol activity of P. membranefaciens against blue mold caused by Penicillium expansum in apple fruit could be enhanced by addition of LCF. The combination of P. membranefaciens and LCF resulted in a more effective control than individual treatment of P. membranefaciens or LCF alone. The combined treatment induced higher phenolic accumulation and the upregulation of superoxide dismutase, catalase, ascorbate peroxidase, chitinase and β-1,3-glucanase activities in fruit than LCF or yeast alone, and resulted in a lower lesion diameter and disease incidence. The use of LCF may be an effective method to improve the biological activity of P. membranefaciens, and induced host defenses appear to contribute to the control mechanism of P. membranefaciens and LCF.  相似文献

Highbush blueberries (Vaccinum spp.) are a major export fruit crop of Chile which is stored at 0 °C and transported to markets in Asia, Europe, and the USA, using more than 15 d of maritime transportation. Under these conditions, gray mold caused by Botrytis cinerea can produce important economic losses. The effectiveness of sulfur dioxide (SO2) concentration × time treatments on gray mold control was determined in the laboratory and validated prior to refrigerating the fruit, using pallet scale SO2 fumigation treatment on the following blueberry cultivars: ‘Brigitta’, ‘Legacy’, ‘Liberty’ and ‘O’Neal’. In inoculated ‘Brigitta’ and ‘Liberty’ blueberries, gray mold prevalence varied from 97.2% to 97.5% in non-treated fruit, and this value was reduced from 7.9% to 6.1% in blueberries that were exposed to a SO2 concentration × time (Ct) product of 400 (μL L−1) h. The relationship between SO2 Ct products and gray mold prevalence under laboratory conditions was best explained by exponential models, which had a determination coefficient (R2) that ranged from 0.88 to 0.96. The estimated EC90 values varied between 245 and 400 (μL L−1) h, and the SO2 Ct between 250 and 350 (μL L−1) h was validated using a pallet scale application treatment to obtain the best control and minimal variation. No visual phytotoxicity symptoms of SO2 were observed with the Ct that was tested in this study. Therefore, SO2 fumigation was demonstrated to be an effective and practical technology for reducing the risk of blueberry gray mold decay during storage, and further effort should be given to register the use of this product for blueberries in the main Chilean export markets.  相似文献

In Israel, black spot caused by Alternaria alternata is the main postharvest factor that impairs the quality and reduces the storability of persimmon fruit (Diospyros kaki cv. Triumph). The fungus infects the fruit in the orchard and remains quiescent until harvest. After harvest, the pathogen slowly colonizes the fruit during storage at 0 °C, which elicits black spot symptom development 2–3 months after storage entry. A commercial postharvest dip treatment in chlorine at 500 mg L?1, released from sodium troclosene tablets, effectively controlled black spot in fruit stored for up to 2 months. However, decay incidence increased as the length of storage was extended beyond 2.5 months. The long incubation period that precedes black spot symptom development after harvest enabled the development of a series of integrative approaches for application at the pre- and postharvest stages, in combination with the commercial chlorine dip treatment, to improve the control of black spot disease. Preharvest treatments included treatment with the cytokinin-like N1-(2-chloro-4-pyridyl)-N3-phenylurea (CPPU) 30 d after fruit set, or a single spray with the curative fungicide polyoxin B 14 d before harvest, and when one of these was applied in combination with the postharvest chlorine dip treatment, the black spot infected area was reduced by 3 and 60%, respectively, compared with the chlorine dip alone. At the postharvest stage, fogging during storage, or post-storage on-line spraying with sodium troclosene, when applied in combination with the postharvest chlorine dip, improved the percentage of marketable fruit by 2 or 10%, respectively, compared with the chlorine dip alone. The results indicate that postharvest pathogens that show a slow colonization pattern might enable the integration of pre- and postharvest disease control methods to improve quality and reduce postharvest disease development.  相似文献

Summary The leaf pH of two year old apple seedlings was found to be sufficiently related to the fruit pH, measured when the seedlings were six to seven years old, to serve as a basis for pre-selection. Discarding all seedlings with an above average leaf pH (40% of the total) eliminated the majority (74%) of seedling with low-acid cum sweet fruits. The method was not effective for reducing the proportion of seedlings with high-acid cum sour fruits.  相似文献

UV-C inactivation kinetic data of Penicillium expansum on intact and wounded pear disks were determined. P. expansum conidia (0.5 mL, 1.6 × 107 CFU/mL) were spot inoculated onto intact and wounded pear tissue with skin (excised disks), treated with UV-C doses ranging 0.101–3.06 kJ/m2 at 23 °C and surviving conidia were enumerated. Changes in selected physicochemical parameters and sensory quality following UV-C treatment of whole pears were determined immediately after treatment, and 4 and 8 weeks of storage at 4 °C. A greater UV-C intensity was required for similar inactivation levels of P. expansum populations on wounded pear disks (3.1 kJ/m2 for 2.7 log reduction) compared to intact pear disks (1.7 kJ/m2 for 2.8 log reduction). No significant difference in % weight loss, or soluble solids content and texture was observed between UV-C treated and untreated pears. However, browning was observed on UV-C treated pear surfaces after 4 and 8 weeks along with changes in flavor and texture. An increase in consumer preference was noticed for the untreated control pears after 4 weeks storage.  相似文献

为了研究和掌握病害的发生规律及更好的防治病害,本实验从温度、pH值、光照、碳源、氮源对玉竹锐顶镰孢菌(Fusarium acuminatum)菌落生长的影响以及不同种类杀菌剂对病菌的抑菌效果进行了研究。结果表明锐顶镰孢菌菌丝生长最适温度为28℃,最适pH值为7.0,光暗交替有利于菌丝生长,以可溶性淀粉为碳源、以甘氨酸为氮源时利用最优。药剂筛选结果表明有机硫类杀菌剂对玉竹锐顶镰孢菌的抑制效果最优。  相似文献

The effect of commercial degreening with ethylene gas on fruit susceptibility and quality and development of postharvest green (GM) and blue (BM) molds on early season citrus fruit was investigated. Each cultivar was harvested with different peel color indexes (CI). Fruit were exposed for 3 d to 2 μL L−1 ethylene at 21 °C and 95–100% RH before or after artificial inoculation with Penicillium digitatum or Penicillium italicum. Control fruit were kept at the same environmental conditions without ethylene. Fruit were stored at either 20 °C for 7 d or 5 °C for 14 d and disease incidence (%) and severity (lesion diameter) were assessed. No significant effect of commercial degreening was observed on fruit susceptibility to both GM and BM on citrus cultivars inoculated after degreening. Likewise, no significant effect was observed on disease incidence on citrus cultivars inoculated before degreening and stored at either 20 °C for 7 d or 5 °C for 14 d. In contrast, in cultivars like ‘Clemenules’ mandarins and ‘Navelina’ oranges, degreening significantly increased the severity on fruit with higher initial CI (−3.6 and 1.7, respectively). GM and BM severity on degreened and control ‘Clemenules’ mandarins incubated at 20 °C for 7 d was 146 and 118 mm and 56 and 46 mm, respectively. In general, commercial degreening did not significantly affect external and internal quality attributes of citrus cultivars. Commercial degreening after inoculation of less green (more mature) fruit showed a trend to increase mold severity, presumably through an aging effect (acceleration of peel senescence).  相似文献

An antagonistic isolate Bacillus amyloliquefaciens HF-01, sodium bicarbonate (SBC) and hot water treatment (HW) were investigated individually and in combination against green and blue mold and sour rot caused by Penicillium digitatum, P. italicum and Geotrichum citri-aurantii respectively, in mandarin fruit. Populations of antagonists were stable in the presence of 1% or 2% SBC treatment, and spore germination of pathogens in potato dextrose broth was greatly controlled by the hot water treatment of 45 °C for 2 min. Individual application of sodium bicarbonate at low rates and hot water treatment, although reducing disease incidence after 8 weeks or 4 weeks of storage at 6 °C or 25 °C respectively, was not as effective as the fungicide treatment. The treatment comprising B. amyloliquefaciens combined with 2% SBC or/and HW (45 °C for 2 min) was as effective as the fungicide treatment and reduced decay to less than 80% compared to the control. B. amyloliquefaciens HF-01 alone or in combination with 2% SBC or/and HW significantly reduced postharvest decay without impairing fruit quality after storage at 25 °C for 4 weeks or at 6 °C for 8 weeks. These results suggest that the combination of B. amyloliquefaciens HF-01, SBC and HW could be a promising method for the control of postharvest decay on citrus while maintaining fruit quality after harvest.  相似文献

There is increasing public interest in development of edible natural biodegradable coatings to replace the currently used commercial synthetic waxes for maintaining postharvest quality of citrus fruit. We tested the efficacy of a newly developed polysaccharide-based edible bilayer coating comprising carboxymethyl cellulose (CMC) and chitosan in preserving postharvest quality of various citrus fruit, including ‘Or’ and ‘Mor’ mandarins, ‘Navel’ oranges, and ‘Star Ruby’ grapefruit after simulated storage and marketing. In all citrus species, it was found that the CMC/chitosan bilayer coating was equally effective as the commercial polyethylene wax in enhancing fruit gloss. Furthermore, the CMC/chitosan bilayer coating slightly increased fruit firmness, especially of oranges and grapefruit, but was mostly not effective in preventing post-storage weight loss. Both the CMC/chitosan bilayer coating and the commercial wax had no significant effects on juice total soluble solids and acidity levels, and had similar effects on gas permeability, as indicated by only slight increases in internal CO2 levels and in juice ethanol accumulation after storage. Sensory analyses revealed that neither the CMC/chitosan bilayer coating nor the commercial wax coating had any deleterious effects on flavor preference of ‘Navel’ orange and ‘Star Ruby’ grapefruit. However, application of the commercial wax, and moreover the CMC/chitosan bilayer coating, resulted in a gradual decrease in flavor acceptability of ‘Or’ and ‘Mor’ mandarins because of increased perception of off-flavors. Overall, we showed that the CMC/chitosan bilayer edible coating sufficiently enhanced fruit gloss, but was not effective in preventing postharvest weight loss. The integration of sodium carbonate (SC; dips at 20 °C for 150 s in aqueous 3% SC solutions) treatments and X-ray irradiation (at doses of 510 and 875 Gy) was evaluated on artificially inoculated ‘Clemenules’ clementine mandarins for the control of postharvest green and blue molds, caused by Penicillium digitatum and Penicillium italicum, respectively. Although significant, the reduction of both disease incidence (number of infected fruit) and severity (lesion diameter) on fruit either incubated at 20 °C for 7 days or cold-stored at 5 °C for 21 days was not sufficient for satisfactory disease control under hypothetical commercial conditions. Therefore, the combined treatments could not be a substitute for conventional chemical fungicides. However, pathogen sporulation was greatly inhibited on infected clementines, thus X-irradiation could be of value for management of Penicillium resistant strains and to reduce inoculum levels in citrus packinghouses. X-ray irradiation at 195, 395, 510, and 875 Gy did not influence either decay incidence or the area under the disease progress curve (AUDPC) of lesions of green and blue molds on mandarins inoculated with the pathogens 2, 3, or 6 days after irradiation and incubated for 7 days at 20 °C. Therefore, X-ray treatment did not induce disease resistance in the rind of irradiated fruit. Although X-irradiation at doses up to 875 Gy followed by either 14 days at 20 °C or 60 days at 5 °C caused very slight rind pitting, minor decreases in fruit firmness, and modest increases in juice acetaldehyde and ethanol contents, these changes had no practical impact on fruit quality. Rind color, titratable acidity, soluble solids concentration, maturity index and juice yield were not influenced by irradiation. ‘Clemenules’ can be considered as a clementine cultivar highly tolerant to X-irradiation.  相似文献

Tomato fruit at the mature green stage were treated with ClO2 gas in a sealed container for 12 h, and then stored at 23 °C with 85% relative humidity (RH) for 23 d. Respiration rate, respiration-related enzymes including phosphohexose isomerase (PHI), succinate dehydrogenase (SDH), and glucose-6-phosphate dehydrogenase (G-6-PDH) and 6-phosphogluconate dehydrogenase (6-PGDH), ethylene production, and the expression of LeAOX1a, LeCOX1, LeACS2, LeACS4 and LeACO1 genes were measured. The results showed that application of ClO2 gas was effective in reducing total respiration, cytochrome pathway respiration and the expression of LeCOX1, but no significant reduction in the activities of respiration-related enzymes was observed during storage. Fruit treated with ClO2 resulted in lower ethylene production. Furthermore, the expression of ethylene biosynthesis related genes, including LeACS2, LeACS4 and LeACO1 was reduced by the ClO2 treatment. These results indicate that ClO2 treatment might delay the ripening of tomato fruit, possibly by a mechanism involving suppression of respiration rate and ethylene biosynthesis.  相似文献
