The role of putrescine (PUT) in regulating fruit softening, antioxidative enzymes and biochemical changes in fruit quality was investigated during ripening and cold storage of mango (Mangifera indica cv. Samar Bahisht Chaunsa). Fruit were treated with various PUT concentrations (0.0, 0.1, 1.0 and 2.0 mM) and were allowed to ripen at 32 ± 2 °C for 7 days, or stored at 11 ± 1 °C for up to 28 days. Respiration rate and ethylene production were measured daily during ripening and cold storage. Cell wall degrading enzymes such as exo-polygalacturonase (exo-PG), endo-polygalacturonase (endo-PG), pectin esterase (PE), endo-1,4-β-d-glucanase (EGase), antioxidative enzymes including superoxide dismutase (SOD), peroxidase (POX), and catalase (CAT), fruit firmness as well as biochemical fruit quality characteristics were estimated during ripening and cold storage at 2 and 7 day intervals, respectively. PUT treatments reduced respiration rate, ethylene production and maintained higher fruit firmness during ripening as well as cold storage. PUT-treated fruit exhibited significantly suppressed activities of cell wall enzymes (exo-, endo-PG and EGase), but retained higher PE activity during ripening and cold storage. Total phenolic and antioxidant contents were significantly higher in PUT-treated fruit during ripening as well in the cold storage period than in the controls. Activities of antioxidative enzymes (CAT, POX and SOD) were also significantly higher in PUT-treated fruit during ripening as well as cold storage. SSC and SSC:TA were lower in PUT-treated fruit, while TA and ascorbic acid content showed the reverse trend. In conclusion, pre-storage 2.0 mM PUT treatment inhibited ethylene production and suppressed the activities of cell wall enzymes, while resulting in higher activities of antioxidative enzymes and maintaining better fruit quality during ripening and cold storage.  相似文献

Pear and apple are among the main fruit crops worldwide. These species can be planted in mixed orchards, and they both depend on insect pollination for fruit set. As pollinating insects are attracted by the floral resources, we investigated nectar and pollen production and chemical composition in four pear (‘Concorde’, ‘Conférence’, ‘Doyenné du Comice’, ‘Triomphe de Vienne’) and five apple (‘Braeburn’, ‘Golden Reinders’, ‘Jonagored’, ‘Pinova’, ‘Wellant’) cultivars commonly grown in Belgium. We also investigated whether insect flower visitation rate and pollination efficiency are linked to floral resource quantity and quality. The pear cultivars flowered one week before the apple cultivars in early spring, and their flowers were about six times less visited by insects. The visitors foraged more on the pollen of the pear trees and the nectar of the apple trees. Pear flowers produced higher volumes of nectar than apple flowers (1.3–3.2 μl vs. 0.4-0.6 μl), but with lower sugar concentration (9.6%-10.8% vs. 28.3%-36.4%). Pear flowers also produced fewer pollen grains per anther than apple flowers (2425–4937 vs. 3284–7919), but these had higher polypeptide (346–362 μg/mg vs. 216–303 μg/mg), amino-acid (40–77 μg/mg vs. 12–18 μg/mg) and phytosterol (21–47 μg/mg vs. 15–43 μg/mg) concentrations. The foraging behavior of the insects is thus better explained by nectar and pollen quality rather than quantity. Despite the differences in flower visitation rates, pollination of both species resulted in valuable fruit production.  相似文献

Physiological responses and fruit quality of ‘d’Anjou’ pear fruit from five orchard lots were evaluated after cold storage in air or controlled atmospheres (CA) with the O2 concentration based on assessment of fruit chlorophyll fluorescence (CF) or standard conditions (1.5 kPa O2). The pCO2 for all CA fruit was 0.5 kPa. Softening, acid loss, and peel degreening of all lots were delayed at one or more evaluation dates (2, 4, 6, 8 months) by previous storage at the CF pO2 compared with fruit stored in 1.5 kPa O2 or in air. Superficial scald developed on fruit previously stored in air but not on fruit stored in a CA. Pithy brown core developed on fruit from all lots stored at the CF pO2 and on fruit stored at 1.5 kPa in 3 of the 5 lots. Pithy brown core incidence decreased with advanced harvest maturity. Post-storage ethylene and CO2 production were in most instances lowest for fruit stored at the CF pO2. A significant relationship between fruit ethanol content and pithy brown core incidence was observed. Results indicate low pO2 storage based on CF monitoring slows fruit ripening relative to fruit stored at 1.5 kPa O2, prevents superficial scald development compared with fruit stored in air, however, development of pithy brown core in fruit stored at the CF pO2 was not accompanied by a change in CF.  相似文献

Summary The total steroidal alkaloid compositions of the mature-harvested fruits of the tomato cultivar Allround (Lycopersicon esculentum), of the species L. hirsutum glabratum, resistant to the glasshouse whitefly (Trialeurodes vaporariorum), and of four resistant lines were determined by capillary gas chromatography. The contents of -tomatine in the red-ripe fruits of Allround and in those of the lines were low (5 mg/kg fresh weight). The green-mature fruits of the wild species had a high content (3390 mg/kg fresh weight) of -tomatine. The total alkaloid profiles of Allround and of the lines were similar. Comparison of flame ionisation detector response with thermoionic detector response indicated that in addition to tomatidine, the aglycone of -tomatine, a small amount of one other steroidal alkaloid might be present in Allround and in the lines. The wild species contained five compounds which possibly are steroidal alkaloids.The safety level of -tomatine in tomatoes is discussed. It was concluded that, in respect of the glycoalkaloids of the mature-harvested fruits, the resistant tomato breeding lines are as safe for human cosumption as the standard cultivar.  相似文献

Mature green cherry tomato fruit were harvested and treated with ultraviolet-C (UV-C) irradiation at a predetermined dose of 4.2 kJ m−2, and stored at 18 °C for 35 days. The effects of UV-C treatment on color change, pigment contents, and the expression of major genes involved in carotenoid metabolism, including Psy 1, Pds, Lcy-β, and Lcy-ɛ, encoding phytoene synthase, phytoene desaturase, lycopene β-cyclase and lycopene ɛ-cyclase, respectively, were examined. The UV-C treated fruit developed a pink red color in contrast to the normal orange red color of control fruit. Lycopene accumulation during ripening in UV-C treated fruit was significantly inhibited but its final content was not affected. However, both accumulation and final content of β-carotene were significantly suppressed in UV-C treated fruit. The lower content of β-carotene, leading to a higher lycopene to β-carotene ratio, is probably responsible for the altered color phenotype in UV-C treated fruit. Psy 1, a major gene involved in lycopene synthesis was inhibited by UV-C irradiation. Significantly suppressed expression of Lcy-β gene was also observed in UV-C treated fruit. Thus it is possible that the lower transformation from lycopene to carotenes contributed to the relatively stable content of lycopene.  相似文献

Ají (Capsicum baccatum L.) and rocoto (C. pubescens R. & P.) are two Capsicum pepper species native to the Andean region that have not been subjected to intensive breeding. However, the increase in its demand in European markets has sparked the development of breeding programmes for adaptation to Mediterranean climates, which include breeding for higher levels of bioactive compounds. We have studied the composition of red and yellow carotenoids (C R and C Y, respectively), ascorbic acid (AA), and total phenolics (TP) in 34 accessions of Capsicum, including 23 of C. baccatum, eight of C. pubescens, and 3 controls of C. annuum, which were grown in Spain both under greenhouse (GH) and open field (OF) conditions. The results show that in both growing conditions C. baccatum presents a considerable variation for most of the traits studied, with several accessions having similar or higher levels than C. annuum for the compounds studied. This indicates that C. baccatum is an important source of antioxidant compounds with nutritional value. On the contrary, C. pubescens had a poor performance, with low levels for all the compounds studied, and a poor adaptation to the conditions of Mediterranean climates. When considering the GH or OF growing cycles separately, heritability values were high (>0.75) for C R, C Y and AA, and moderate (0.42 for GH and 0.62 for OF) for TP. The existence of an important genotype × environment interaction resulted in lower levels for heritability when considering both growing cycles together, although the values were still high for C R and AA (>0.6). Positive significant genotypic correlations among all the traits studied were found, except for TP with C R and AA in the OF cycle. This information indicates that there are good prospects for developing C. baccatum varieties with higher levels of bioactive compounds.  相似文献

Safflower (Carthamus tinctorius L.) seeds contain a high proportion of tocopherols (>90 %) in the α-tocopherol form. A mutant with a high concentration of γ-tocopherol (>85 %) was identified in germplasm of wild safflower (Carthamus oxyacanthus M. Bieb.) that showed strong introgression of C. tinctorius, which allowed selection of individuals of both species with high concentrations of either α- or γ-tocopherol. The trait is controlled by a γ-tocopherol methyltransferase (γ-TMT) locus. The objective of this research was to identify γ-TMT sequence mutations associated with the high γ-tocopherol trait. Full length genomic and cDNA sequences of the γ-TMT gene were obtained from plants of C. tinctorius and C. oxyacanthus with both tocopherol profiles. Sequences from high γ-tocopherol plants showed an 11 bp deletion in exon 6 of the γ-TMT gene that disrupted the reading frame and created a premature stop codon, resulting in a predicted protein with a drastically altered amino acid sequence downstream the frameshift site. The data suggested that the frameshift mutation was underlying the γ-TMT loss of function mutant allele that determines the high γ-tocopherol phenotype. The characterized sequence change of 11 bp deletion could be used directly as a functional marker for introgression of the high γ-tocopherol trait into elite safflower cultivars.  相似文献

Summary Twenty locally-adapted but genetically diverse cowpea genotypes were grown in 17 replicated trials in a range of sites and seasons in the drylands of eastern Kenya, and the discrimination against the heavy isotope of carbon (with a mass of 13) () was determined in grain and straw samples. Most genotypes gave similar, low grain yields in the environments which subjected the plants to drought stress, but high yielding and low yielding genotypes were clearly distinct in the less stressful environments. The converse pattern was found for : the genotypes were all very similar in the non-stressful (high-) environments, but in the stressful environments, low- (drought susceptible) and high- (drought avoiding) genotypes were clearly distinct. This pattern was clearest for straw , but was also apparent for grain . The early-maturing genotypes, which escaped terminal drought, were generally those with the most stable and highest , but in the case of straw there were statistically significant deviations from this relationship. It is concluded that provides a reliable measure of the specific response of cowpea genotypes to drought, which may be of value in breeding programs provided that carbon isotope discrimination can be measured reasonably economically.  相似文献

The main objective of this research was the evaluation of the variability present in a segregating wine grape population derived from a cross between Graciano × Tempranillo, two Spanish varieties, in order to select improved genotypes with potential for producing high-quality wines in a climate change scenario. For that purpose, the phenotypic segregation of 16 agronomic traits related to production and phenology and 11 enological traits related to technical and phenolic maturity was studied in the progeny for three consecutive years. All traits presented transgressive segregation and continuous variation. Year effect was significant for all traits except total, extractable and skin anthocyanins content. However, a high level of genotype consistency for enological traits was revealed by repeatabilities and correlations between years. Significant correlations among traits were observed but most associations were weak. Furthermore, the CAPS (Cleaved Amplified Polymorphic Sequence) marker for the VvmybA genotype was tested to determine whether it would be useful in indirect selection for berry anthocyanins content. The results showed that the number of homozygous and heterozygous genotypes for the functional colour allele adjusted to a 1:1 segregation ratio, and that homozygous genotypes had significantly higher anthocyanins content. Principal component analysis found eight variables that contributed up to 80 % of the phenotypic variability present in the population. Seven groups of hybrids were distinguished based on ripening time, cluster weight, berry weight and anthocyanins content by cluster analysis; and fourteen genotypes were pre-selected for further research.  相似文献

Indica-japonica hybridization is an important approach for developing superior performing hybrids in rice (Oryza sativa L.). In view of the scanty information available on cooking quality characters in indica-japonica crosses, an investigation was undertaken to estimate genetic and genotype × environment variance and covariance components of amylose content, gel consistency and alkali digestion value, and to determine the relative importance of direct genetic effects, maternal genetic effects and cytoplasmic effects in the genetic variations of the three quality characters. Two indica photo-sensitive genic male sterile (PGMS) lines and four japonica varieties were used as parents to make crosses. Genetic model with genotype × environment interactions for triploid endosperm was used for genetic studies of the three cooking quality characters. Variance component analysis revealed that genetic variations of the three characters were mainly attributable to direct additive and maternal additive effects, and the three traits had significant direct and maternal heritabilities. Genotype × environment interactions were mainly dominance × environment (including direct dominance × environment and maternal dominance × environment) and cytoplasm × environment interactions. Environment factors could only affect the expression extent of dominant genes, without changing their directions. Predicted values of genetic effects indicated that the parental lines, ‘VI-70’ and ‘H9304-1’, appeared to be best for amylose content, ‘T 1950’ and ‘Suxuan’ appeared to be best for gel consistency and alkali digestion value. Despite QTL mapping being a routine procedure in plant breeding, approaches that fully exploit data from multi-trait multi-environment (MTME) trials are limited. Mixed models have been proposed both for multi-trait QTL analysis and multi-environment QTL analysis, but these approaches break down when the number of traits and environments increases. We present models for an efficient QTL analysis of MTME data with mixed models by reducing the dimensionality of the genetic variance–covariance matrix by structuring this matrix using direct products of relatively simple matrices representing variation in the trait and environmental dimension. In the context of MTME data, we address how to model QTL by environment interactions and the genetic basis of heterogeneity of variance and correlations between traits and environments. We illustrate our approach with an example including five traits across eight stress trials in CIMMYT maize. We detected 36 QTLs affecting yield, anthesis-silking interval, male flowering, ear number, and plant height in maize. Our approach does not require specialised software as it can be implemented in any statistical package with mixed model facilities.  相似文献

Fourteen genetically diverse inbred lines comprising of four genic male sterile lines and 10 often pollinated lines as testers were crossed following ‘line × tester’ mating design to generate 40 F1s. These F1s, parents and check ‘CH-27’ were evaluated in α-lattice square design, replicated thrice during summer seasons of 2020 and 2021. The male sterile line DPChMS 9-2 was good general combiner for marketable fruit yield and eight other component traits while testers namely, DPCh 10, PBC 535, VVG, Him Palam Mirch-2 and DPCh 40 showed significant GCA for majority of yield related traits. The hybrids DPChMS 9-2 × HPM-2, DPChMS 9-2 × DPCh 40, DPChMS 9-2 × DPCh 101, DPChMS 9-2× VVG and DPChMS 26-1× HPM-1 were the most desirable based on specific combining ability (SCA) and economic heterosis for fruit yield and other important traits. Heterosis was positively correlated with SCA indicating prediction of potential hybrids can be made based on both these parameters simultaneously. Present study provides an opportunity to exploit top hybrids for stability and commercial cultivation.  相似文献

The concentration of amylose, which is synthesised using granule-bound starch synthase, affects the physical properties of food. However, no studies have focused on starch properties and physical characteristics of low-amylose buckwheat. Here, we hypothesised that low-amylose buckwheat would be useful to produce new buckwheat products because low-amylose characteristics change the texture of buckwheat food. In this study, we bred relatively low-amylose buckwheat compared to wild type and investigated the causative genes of the traits, starch properties and physical properties of noodles. In the GBSSa mutant, the amylose concentration was lower than that in the wild type. Compared with the wild type, the mutant exhibited the following traits: Amylose concentration decreased by approximately 2%, setback in the Rapid Visco Analyzer decreased by 30 points and the physical characteristics of noodles in the sensory analysis were soft and sticky. These results suggest that this trait may be useful for changing the texture of foods. In addition, the mutant is promising for producing new foods with physical characteristics that are different from those of the wild type.  相似文献
