Abstract

Radioactively labeled iron (59Fe) was used to study iron retranslocation from mature leaves of Broad bean (Vicia faba L. var. Scirocco). Our experiments offered the possibility to detect and quantify the translocation of foliar applied iron by imaging technique in combination with tissue analysis. 59Fe labeled solution was placed as a droplet onto the leafs upper surface of intact plants. Distribution of 59Fe was analyzed after 0.5 h up to 2 days. Iron was translocated acropetally (towards the tip of the treated leaf) as well as basipetally. Movement in the apical direction was predominant, amounting to about 65% of 59Fe translocated from the application site. About 35% of 59Fe were transported basipetally, corresponding to absolute amounts of 2.8–53.6 pmol h?1. After 30 min, it was detectable in the petiole, which included a translocation of 20 mm basipetal from the application site. A mean of 15% of the iron retranslocated from a leaflet was detected in non‐treated leaflets of the same leaf. This iron was supposed to have been exchanged from the phloem into the xylem pathway, probably within the petiole. When the loading rate into the phloem was estimated on basis of the sum of retranslocated 59Fe per time and per area of the leaf treated, a range of 0.031–2.21 pmol h?1 mm?2 (mean: 0.62 pmol h?1 mm?2) was obtained. This was not sufficient to meet an estimated demand for iron in the growing terminal bud, but could cover about 25% of it. In conclusion, average iron retranslocation from leaves of Fe‐sufficient plants was not large enough to meet the iron demand of the growing shoot. This was not due to a limitation in iron availability for transport, as an excess amount of iron was supplied which was not biologically bound, but a limitation due to transport facilities, probably in the phloem, seemed to be more likely in this case.  相似文献

An increased root turnover can be a mechanism of improved nutrient‐uptake efficiency. The objectives of this study were to investigate P and K efficiency of faba beans (Vicia faba L.), to determine their root growth and root turnover, and to assess the relevance of root turnover on P and K uptake at limited supply. Faba beans were grown as part of a long‐term fertilization experiment on fertilized plots (control) and plots that had not received any P or K fertilizer for 16 years (P0, K0). Although the unfertilized soils were low and very low in their P‐ and K‐supply level, respectively, no differences in shoot‐dry‐matter production occurred compared to the control. However, relative K concentration in dry matter of the K0 plants (control plants = 100) decreased during the experiment and was only 60% of the control at the final harvest. This indicated a high K‐utilization efficiency of faba bean. Relative phosphorus concentration increased in the P0 treatment and was not different from the control at the last harvest, indicating an improvement in P‐uptake efficiency with time. The size of the standing root system determined by sequential auger sampling (net development) was not influenced by P and K supply. Total root production as measured by the ingrowth‐core method was about 6 times higher than the average size of the standing root system and even increased under low‐K conditions. This indicated a fast root turnover. Modeling soil nutrient transport and uptake revealed that calculated uptake of the control was up to 48% higher when root turnover was taken into account, compared to calculations based on the net development of the root systems. This is due to a better soil exploitation. Under K shortage, root turnover resulted in a 117% higher calculated uptake, which was close to measured K uptake. Root turnover was also of benefit for P uptake, but calculated P uptake was significantly less than measured, indicating that root turnover was of little importance for P uptake of faba beans.  相似文献

植物叶表面的气孔保卫细胞是研究信号转导的模式实验系统,对环境变化反应灵敏而准确,采用蚕豆叶面气孔保卫细胞,研究了铝(AlCl3)对细胞的毒性效应。结果表明,在1~10 mmol.L^-1范围内,AlCl3可使气孔保卫细胞活性降低,部分细胞死亡,且随着浓度的增高细胞死亡率增高;死细胞呈现核固缩、核降解、凋亡小体等典型凋亡特征。凋亡抑制剂Z-Asp-CH2-DCB或TLCK与AlCl3共同作用时,保卫细胞死亡率显著降低;一定浓度的抗坏血酸(AsA)或过氧化氢酶(CAT)以及Ca2+螯合剂乙二醇四乙酸酯(EGTA)或Ca2+通道抑制剂LaCl3与AlCl3共同作用时,细胞死亡率降低。研究结果表明,铝诱导的蚕豆保卫细胞死亡可能是一种细胞凋亡过程,由胁迫诱发的活性氧介导,通过激活质膜钙通道,引起胞内Ca2+水平改变,进而介导细胞凋亡。  相似文献

The efficiency of symbiotic dinitrogen (N2) fixation in Vicia faba L. in combination with 3 different Rhizobium leguminosarum strains was studied in a pot experiment during vegetative and reproductive growth. The objective of the experiments was to assess variability among Rhizobium strains inoculated on single legume species and determine possible reasons for observed variations. Dry matter formation, N2 fixation and the carbon (C) costs of N2 fixation were determined in comparison with nodule free plants grown with urea. Nodule number and the capacity of different respiratory chains in the nodules were also measured. The plants inoculated with the Rhizobium strain A 37 formed less dry matter and fixed less N compared to the other two Rhizobium strains (Vic 1 and A 150). This coincided with a lower number of nodules and higher C costs of N2 fixation. The C costs for N2 fixation were in all cases significantly lower during reproductive growth compared to vegetative growth. Neither the latter nor the differences in C expenditure for N2 fixation between the Rhizobium strains could be explained in terms of differences or shifts in the capacity of different respiratory chains in the nodules.  相似文献

Summary A pure line faba bean germplasm collection (BPL) was derived by a process of cyclic single plant progenies starting in 1979 through 1985. In 1985/86, 840 of these pure line accessions from 677 original populations from 41 countries were evaluated for autofertility by comparing the mean of five plants where all flowers were tripped vs. five plants with no tripped flowers for number of seeds produced (seed index). In a replicated trial grown at the same time, seed yield, 100 seed weight, number of pods per plant, seeds per pod, and number of flowers per inflorescence were also evaluated. A high number of BPL accessions failed to set seeds in the screen houses without tripping, the mean seed index measure of autofertility was 0.51 with a C.V. of 74.2%. There were marked differences among countries of origin for autofertility, with Egypt having the highest (SI = 0.94) and the USSR the lowest (SI = 0.26) autofertility, however, most countries expressed a large range, suggesting that breeders should first look in their own material for this trait rather than searching in exotic germplasm. In general, there was a higher level of autofertility in Middle Eastern countries that may be due to natural selection for non-dependency on pollinators for high yield. Contrary to the findings of others, large seeded accessions had a high autofertility (SI = 0.704) compared to small seeded accessions (SI = 0.384), however, both had similar ranges. In general, there was no significant correlations among autofertility and yield and its components. The variability found in this large pure line collection for autofertility is of potential use for developing cultivars not dependent on insect populations for high yield and for developing self-pollinated faba bean.  相似文献

通过盆栽试验研究了崇礼蚕豆出苗及生长过程中不同指标对不同土壤硒浓度的响应。结果表明,低浓度硒(3.2 mg·kg-1)对蚕豆各出苗指标、叶绿素a含量、叶绿素a+b、叶绿素a/b及产量构成因素有一定的促进作用,但高浓度硒(12.4 mg·kg-1)对蚕豆出苗及生长指标有明显的抑制作用。通过各项指标与土壤硒浓度之间的相关分析及10%抑制率的临界指标EC10值表明,不同指标EC10各异,以出苗势或苗期干重作为蚕豆土壤临界值制定的依据更可靠,EC10分别为4.8和5.2 mg·kg-1。  相似文献

Summary Twenty-six faba bean (Vicia faba L.) populations, from five geographic areas and five varietal types have been assayed electrophoretically for variation in seven isozyme systems. Isozyme frequencies in populations, botanical varieties and geographical areas were submitted to the principal component analysis (PCA). Varieties grouped together showed no differences among them except for isolated populations. Populations from Ethiopia and China lay far apart in PCA from those of other origins. It seems that no genetic divergence at the isozyme level took place during the varietal formation and dispersion process, except when geographical isolation of the population occurred. The results support the hypothesis of an isolation of the crop in Ethiopia and in China.  相似文献

蚕豆是重要的生物固氮和植物蛋白源作物,在改良土壤培肥地力、改善温带农业区持续蛋白生产方面尤为重要。然而,蚕豆品种的无限生长习性是导致机械化程度低的主因之一,极大地制约了蚕豆的规模化种植。为了探讨亚有限型在蚕豆育种中的应用及其对合理株型矮化育种研究的推动作用,通过查阅相关文献和多年的选择鉴定,对亚有限生长型新种质的发现过程和主要特征进行了总结归纳。对3个具有不同生长习性的种质与青蚕17号×RF25 F2分离群体主要农艺性状的表现差异进行了分析。结果表明,在F2群体中,亚有限型在开花层数、株高、结荚层数上与有限、无限型均存在显著差异,无限型与亚有限型的单株荚数、单株粒数、单株产量差异不显著,二者均显著高于有限型。说明亚有限生长习性蚕豆在品种改良后,能兼顾产量与机械化收获,在生产中应用优势更明显。  相似文献

通过盆栽试验研究了在不同浓度EDTA、柠檬酸、草酸3种螯合剂作用下不同浓度铀镉复合污染(分别用W1、W2表示)对土壤中蚕豆(ViciafabaL.)幼苗体内叶绿素、丙二醛和细胞质膜透性的影响。结果表明,在所设的两个土壤污染浓度下,较高土壤污染浓度(W2)中植物受损伤程度高于较低土壤污染浓度(W1)中的植物受损伤程度,由此得出较高土壤污染浓度可能更有利于蚕豆对铀镉的吸收;蚕豆的整个生长过程中,对于叶绿素而言,在污染土壤中添加5mg.kg-1的草酸,对其含量影响最大,可能有利于促进蚕豆对土壤中铀和镉的吸收,添加2.5mg.kg-1的柠檬酸次之;对于丙二醛,向污染土壤中添加10mg.kg-1的草酸,对其含量影响最大,可能有利于蚕豆对土壤中铀和镉的吸收,添加2.5mg.kg-1的柠檬酸次之;在细胞质膜透性上,向土壤中添加5mg.kg-1草酸,对其含量影响最大,可能更有利于促进蚕豆吸收土壤中的铀和镉,而添加5mg.kg-1的柠檬酸次之。而EDTA在整个试验中都表现出了对植物较大的生物毒性。  相似文献

 Mixed inoculation of Vicia faba L. with four different Rhizobium/Azospirillum and Rhizobium/Azotobacter combinations led to changes in total content, concentration and/or distribution of the mineral macro- and micronutrients, K, P, Ca, Mg, Fe, B, Mn, Zn and Cu, when compared with plants inoculated with Rhizobium only. The infiltration-centrifugation method is valuable for the estimation of ion concentrations within the leaf apoplast because it opens up the possibility of differentiating between free and bound cations. The K+ concentration in the apoplast of intact leaves of Vicia faba reflects the nutritional status with respect of this ion and can be monitored by the infiltration-centrifugation technique. Transfer into a nutrient solution without K+ led to a significant decrease in apoplastic K+ within two days. Under K+ deficiency, the K+ concentration in the leaf apoplast was maintained at 2 mM. Regardless of K+ supply, young leaves had higher K+ concentrations in the apoplast than older ones. Data on diurnal variations in apoplastic K+ suggest that the leaf apoplast is a transient pool for K+ accumulation within the leaf prior to absorption into the symplast or retranslocation to other growing tissues.  相似文献

This study was initiated to evaluate the effect of locally isolated Rhizobium on nodulation and yield of faba bean at Haramaya, Ethiopia for three consecutive years. Ten treatments comprising of eight effective isolates of rhizobia, uninoculated, and N-fertilized (20 kg N ha?1) were laid out in a randomized complete block design with three replications. The result of the experiment indicated that all inoculation treatments increased nodule number and dry weight over the control check in all cropping seasons. The result, however, showed the non-significant effect of Rhizobium inoculation on shoot length, number of tiller per plant and 100 seed weight in all cropping season. Inoculating Haramaya University Faba Bean Rhizobium (HUFBR)-15 in 2011 and National Soil Faba Bean Rhizobium (NSFBR)-30 in 2012 and 2013 gave the highest grain yields (4330, 5267 and 4608 kg ha?1), respectively. These records were 75%, 48%, and 5% over the uninoculated treatment of respective years. Over the season, NSCBR-30 inoculation resulted in the highest nodulation and grain yield production as compared to the other treatments. In general, isolates from central Ethiopia were better than those isolated from eastern Ethiopia and Tropical Agricultural Legume (TAL)-1035 in enhancing faba bean production at Haramaya site. Therefore, NSFBR-30 is recommended as a candidate isolate for faba bean biofertilizer production in eastern Ethiopia soils.  相似文献

采用我国北方两个豆科绿肥品种: 箭筈豌豆(Vicia sativa L.)与毛叶苕子(Vicia villosa Roth),在控制条件下通过不同供磷处理的营养液培养,研究了不同绿肥作物适应低磷胁迫根际过程的差异,并揭示其高效利用磷的机理。试验通过分次取样的方法,测定了两种豆科作物在缺磷与供磷条件下的生物量、根系质子释放速率、根系有机酸分泌速率以及根表酸性磷酸酶活性的动态。结果表明,箭筈豌豆与毛叶苕子在生长前期对低磷胁迫的响应存在明显差异。箭筈豌豆主要靠增大质子释放量和提高酸性磷酸酶活性来适应低磷胁迫; 而毛叶苕子主要通过提高根冠比、扩大根系生物量来对外界环境中的缺磷状况做出响应,在缺磷时其根表酸性磷酸酶的活性显著提高。箭筈豌豆与毛叶苕子可通过协调根系形态和生理的适应性变化提高对磷的吸收。  相似文献

We examined the role of jasmonic acid (JA) in faba bean under cadmium (Cd) stress, which reduces the growth, biomass yield, leaf relative water content (LRWC) and pigment systems. Hydrogen peroxide (H2O2) and lipid peroxidation (malondialdehyde [MDA]) levels increased by 2.78 and 2.24-fold, respectively, in plants under Cd stress, resulting in enhanced electrolyte leakage. Following foliar application to Cd-treated plants, JA restored growth, biomass yield, LRWC and pigment systems to appreciable levels and reduced levels of H2O2, MDA and electrolyte leakage. Proline and glycine betaine concentrations increased by 5.73 and 2.61-fold, respectively, in faba bean under Cd stress, with even higher concentrations observed following JA application to Cd-stressed plants. Superoxide dismutase, catalase, ascorbate peroxidase and glutathione reductase levels rose by 87.47%, 130.54%, 132.55% and 37.79%, respectively, with Cd toxicity, with further enhancement of antioxidant activities observed following foliar application of JA. Accumulation of Cd in roots, shoots and leaves was also minimized by external supplementation of JA. In conclusion, JA mitigates the negative impacts of Cd stress in faba bean plants by inhibiting the accumulation of Cd, H2O2 and MDA, and by enhancing osmolyte and antioxidant activities that reduce oxidative stress.  相似文献

The effects of salinity on four faba bean (Vicia faba L) cultivars [Giza 429, Giza 843, Misr 1 (Orobanche-tolerant), and Giza 3 (Orobanche-susceptible)] and soil properties were investigated in a pot experiment with addition of 0, 50, and 100 mM sodium chloride (NaCl) for 9 weeks. Salinity significantly decreased calcium (Ca2+), magnesium (Mg2+), potassium (K+), bicarbonate (HCO3 ?), and sulfate (SO4 2?) while significantly increasing sodium (Na+), chloride (Cl?), pH, and electrical conductivity (EC; dS m?1). Root length density (cm cm?3), root mass density (mg cm?3), total dry weight, and salt-tolerance indexes were significantly reduced as a result of application of salinity. The results presented support evidence on the positive relationship between Orobance tolerance and salt tolerance in the three cultivars (Giza 429, Giza 843, and Misr 1). This adaptation was mainly due to a high degree of accumulation of inorganic nitrogen (N), phosphorus (P), K+, Ca2+, and Mg2+ and lesser quantities of Na+ and Cl?, as well as greater K+/Na+ and Ca2+/Na+ ratios.  相似文献

Fifty-five Greek Vicia faba L. populations, collected from diverse areas, were planted at two dry and low fertility sites for evaluation and classification. Yield evaluation, which was carried out by Principal Component Analysis (PCA) on the basis of seven yield traits, showed the number of pods per plant, number of ovules and seeds per pod, and branching from the basal nodes to be the most important traits for population evaluation regarding yield. For population classification, four dissimilarity coefficients (Manhattan, Average Taxonomic Distance, Euclidean distance and squared Euclidean distance) and four multivariate methods (PCA, UPGMA, Neighbor-joining and Principal Coordinate Analysis) were evaluated using fifteen morphological and seven yield traits. Neighbor-joining was chosen as the most suitable multivariate method. This method combined with PCA for the seven yield traits, placed the populations into six groups. As revealed by the application of PCA on all twenty-two traits the grouping was based mainly on pod characteristics, stem thickness, plant height, 1000 seed weight and branching from basal nodes. Based on the results of the present study, a model is proposed for conserving cross-pollinated species, such as faba bean.  相似文献

The effect of prior soil amendment with different N sources at 50 mg N (kg soil)—1 on nodulation and N2 fixation of faba bean (Vicia faba L. cv. Troy) using wheat (Triticum aestivum L. cv. Star) as reference crop was assessed in a pot experiment. Four treatments viz legume manure (LEGM) as clover shoots, cereal manure (CEREM) as barley straw, N fertilizer (FERT‐N) as Ca(NO3)2, and no‐manure control (NOMAN) were investigated consecutively at 45, 70, and 90 days after sowing (DAS). Faba bean nodulated profusely, with an increase on average from 629 nodules per pot at 45 DAS to nearly 2.3‐ and 3.3‐fold at 70 and 90 DAS, respectively. Low nodule numbers and nodule dry matter occurred under FERT‐N and CEREM, whereas high values were found for NOMAN and LEGM. Soil amendment affected percent N2 fixation in relation to N source and plant age. Highest percent N2 fixation (≥ 90 %) was found under the lowest N‐supplying amendments, no‐manure, and cereal manure, respectively. FERT‐N depressed N2 fixation particularly at 45 DAS when N2 fixation was reduced to as low as 23 %. The rise in N2 fixation thereafter suggests that faba bean adjusted after depletion of mineral N in the soil. N2 fixation was also decreased after cereal straw application, even though N concentration in faba bean plants was high. The results indicate that plant residues, both with high and low N concentration, applied to soil to raise its fertility may interfere with N2 fixation of faba bean.  相似文献

V. sativa L. sensu strictu was introduced in Rio Grande do Sul (Southern Brazil) by Italian settlers and is nowadays cultivated for forage in backyards and fields. V. angustifolia L. is a widespread highly polymorphic naturalized ruderal. Both taxa grow sympatrically and morphologically transitional types are often found. We analyzed 37 accessions of V. sativa, V. angustifolia, intermediate types, and V. cordata Wulf. ex. Hoppe from Rio Grande do Sul, for karyotypes, corolla and legume color, seed color pattern, pod constrictions and phenological development. Karyotypes allowed a clear distinction between the taxa: V. sativa (2n=12) had a metacentric marker chromosome, V. angustifolia and intermediate types (2n=12) only acrocentric chromosomes and V. cordata presented 2n=10. Flowering and fruiting periods were shorter for V. sativa than for V. angustifolia, which was separated into early and late flowering groups. Qualitative characters separated the taxa by Jaccard coefficients into three groups: V. sativa at 0.5 similarity level, V. angustifolia and intermediate types at 0.6 and V. cordata joined V. sativa at 0.12. Our results show that karyotypes and a range of qualitative characters are diagnostic for the aggregate represented in Southern Brazil.  相似文献

Phosphorus (P) fertilizers are usually supplied prior to or at planting of potato even though most P is taken up 40 to 80 d after emergence. This may lead to inefficient P use as a result of P leaching or fixation in the soil. This study evaluates the effects of split P application at multiple times during the growth period according to the plant's need for P. Potato (Solanum tuberosum L. cv. Ditta) was grown in pots in climate chambers, and radioactive 32P isotope was used to distinguish between the fertilizer and soil‐derived P sources. Two soils were tested in combination with five application rates of P, and the plants were harvested at four dates. The results show that the recovery of P fertilizer can be significantly enhanced if the P supply is split. The result also showed that the proportion of soil‐derived P, accumulated in the plant, was significantly reduced both when more fertilizer P was applied to the soil and when P supply was split into several applications. The positive effects of multiple P applications on the P recovery were greatest in the soil with low P status and low buffer capacity.  相似文献
