Transpiration efficiency (TE) has been recognized as an important source of yield variation under drought stress in groundnut. Here the variation for TE is evaluated in a set of 318 recombinant inbred lines (RILs) of groundnut at F8 generation, derived from a cross between a high TE (ICGV 86031) and a low TE (TAG 24) parent, and the value of specific leaf area (SLA), SPAD chlorophyll meter readings (SCMR) and carbon isotope discrimination (Δ13C) as surrogates of TE are measured. Transpiration efficiency was measured gravimetrically in the 318 RILs and parents under progressive soil drying in a pot culture in two post-rainy seasons. Large and consistent variation for TE existed among the RILs across years. The overall distribution of TE among the RILs indicated that TE was governed by dominant and additive genes. Surrogates SLA and SCMR, were measured prior, during and after completion of the drought period, whereas Δ13C was measured on the dried tissue after harvest. Transpiration efficiency was negatively associated with SLA after the completion of stress treatment (r2 = 0.15) and Δ13C in leaves (r2 = 0.13) and positively associated with SCMR during stress (r2 = 0.17). These associations, all fairly weak, were significant only in 2004. None of these relationships was found in 2005. Although the heritability of SCMR during 2005 was relatively higher than that of TE, and although SCMR has previously been used to identify contrasting germplasm for TE, the stress-dependence of the relationship with TE, and the poor regression coefficients (r2) with that RIL population, do not confer that these surrogates are adequately robust enough in that population. Though more time consuming, a direct gravimetric evaluation for TE appeared to be more reliable.  相似文献

The current study investigates the association between drought tolerance traits and aflatoxin contamination in peanut grown under long-term drought. Two field experiments were conducted at Khon Kaen University, Thailand using a split–split plot design with three drought stress levels as main plots, 11 genotypes as sub-plots, and two soil inoculations of Aspergillus flavus treatments as sub-sub-plots. The effects of temperature, soil moisture and A. flavus population on kernel colonization and aflatoxin contamination, and drought tolerance traits viz. specific leaf area (SLA) and root length density (RLD) were measured. The results demonstrated that elevated soil temperatures and reduced soil moisture, favored aflatoxin production. Drought in combination with higher levels of A. flavus inoculum load in the soil resulted in an increase in the fungal populations in the soil which in turn resulted in increased kernel colonization and subsequent aflatoxin contamination. A combination of SLA and RLD, and kernel colonization had a significant influence on aflatoxin contamination under drought conditions in both seasons (r = 0.73** and 0.76**). The results revealed that drought tolerance traits (SLA and RLD) could be contributing to resistance to aflatoxin contamination suggesting that a combination of SLA, RLD and kernel colonization could be used as selection criteria in selecting parents for aflatoxin resistance.  相似文献

Aflatoxins, produced by the fungi Aspergillus flavus Link ex Fries and Aspergillus parasiticus Speare, are the major toxins affecting the quality of groundnuts (Arachis hypogaea L.) meant for human consumption. Groundnuts can be infected with aflatoxin-producing fungi pre-harvest, at harvest and post-harvest. This survey was conducted in Gujarat province in India in order to assess farmers’ and other stakeholders’ (extension staff and traders) perceptions and knowledge of aflatoxin contamination of groundnuts and to evaluate the agronomic and market practices used to manage it. The survey investigated the effects of the socioeconomic background of the farmers. The results showed that the socioeconomic and psychological characteristics, viz. education, caste, farm size, social participation, extension participation, market orientation, economic motivation, innovativeness and perception had positive and significant associations with farmers’ knowledge. The extension staff and traders had a good understanding of the problem and of the importance of managing aflatoxin contamination but farmers did not. Farmers’ who practised effective crop husbandry in order to increase production were unwittingly managing aflatoxin contamination to some extent. Their marketing practises showed that the problem of aflatoxin contamination was neglected at both the production and marketing stages. We suggest that extension agencies need to train farmers in the use of biological control agents, post-harvest management and identification of aflatoxin contamination. Partnerships need to be forged between research institutions, the departments of agriculture of various states, marketing agencies, NGOs, farmers’ groups, consumer groups, agrochemical manufacturers and other stakeholders in order to develop strategies for addressing the problem of aflatoxin contamination.  相似文献

Aflatoxin, a toxin produced by the fungus Aspergillus flavus Link: Fries, occurs naturally in maize (Zea mays L.). Aflatoxin is a potent human carcinogen and is also toxic to livestock, pets, and wildlife. When contaminated with aflatoxin, the value of maize grain is markedly reduced. This investigation was conducted to compare ear rot, aflatoxin accumulation, and fungal biomass in maize single crosses with varying degrees of resistance to aflatoxin accumulation and to determine the relative importance of general combining ability (GCA) and specific combining ability (SCA) in the inheritance of resistance to ear rot, aflatoxin accumulation, and fungal biomass. Eight germplasm lines with different levels of resistance to aflatoxin accumulation were used as parents of a diallel cross. The cross was evaluated for visible ear rot, aflatoxin accumulation, and A. flavus infection in the grain. A. flavus infection was determined by quantitative real-time polymerase chain reaction (qRT-PCR) assays. Both GCA and SCA were significant sources of variation in the inheritance of the three traits although GCA accounted for a greater portion of the variation among single crosses. The interactions of GCA and SCA with years were highly significant for aflatoxin accumulation, but not significant for A. flavus infection. Estimates of GCA effects were highly significant for both reduced A. flavus infection and reduced aflatoxin accumulation for Mp313E, Mp715, and Mp717. Conversely, GCA effects associated with GA209 were significant for reduced levels of A. flavus infection and ear rot, but high levels of aflatoxin accumulation. Mp313E, Mp715, and Mp717 should be useful in breeding programs targeting both reduced levels of fungal infection and aflatoxin accumulation.  相似文献

Drought is a major constraint for rice production and yield stability in rainfed ecosystems, especially when it occurs during the reproductive stage. Combined genetic and physiological analysis of reproductive-growth traits and their effects on yield and yield components under drought stress is important for dissecting the biological bases of drought resistance and for rice yield improvement in water-limited environments. A subset of a doubled haploid (DH) line population of CT9993-5-10-1-M/IR62266-42-6-2 was evaluated for variation in plant water status, phenology, reproductive-growth traits, yield and yield components under reproductive-stage drought stress and irrigated (non-stress) conditions in the field. Since this DH line population was previously used in extensive quantitative trait loci (QTLs) mapping of various drought resistance component traits, we aimed at identifying QTLs for specific reproductive-growth and yield traits and also to validate the consensus QTLs identified earlier in these DH lines using meta-analysis. DH lines showed significant variation for plant water status, reproductive-growth traits, yield and yield components under drought stress. Total dry matter, number of panicles per plant, harvest index, panicle harvest index, panicle fertility, pollen fertility, spikelet fertility and hundred grain weight had significant positive correlations with grain yield under drought stress. A total of 46 QTLs were identified for the various traits under stress and non-stress conditions with phenotypic effect ranging from 9.5 to 35.6% in this study. QTLs for panicle exsertion, peduncle length and pollen fertility, identified for the first time in this study, could be useful in marker-assisted breeding (MAB) for drought resistance in rice. A total of 97 QTLs linked to plant growth, phenology, reproductive-growth traits, yield and its components under non-stress and drought stress, identified in this study as well as from earlier published information, were subjected to meta-analysis. Meta-analysis identified 23 MQTLs linked to plant phenology and production traits under stress conditions. Among them, four MQTLs viz., 1.3 for plant height, 3.1 for days to flowering, 8.1 for days to flowering or delay in flowering and 9.1 for days to flowering are true QTLs. Consensus QTLs for reproductive-growth traits and grain yield under drought stress have been identified on chromosomes 1 and 9 using meta-QTL analysis in these DH lines. These MQTLs associated with reproductive-growth, grain yield and its component traits under drought stress could be useful targets for drought resistance improvement in rice through MAB and/or map-based positional analysis of candidate genes.  相似文献

Plants belonging to four “oregano” plant species (Origanum hirtum L., Origanum onites L., Coridothymus capitatus L., and Satureja thymbra L.) were collected during flowering from 33 sites located in the eastern part of the Greek island of Ikaria in the Eastern Aegean during April, May and July 2008. C. capitatus and O. hirtum were mostly observed in higher altitudes, whereas O. onites and S. thymbra in lower ones. The spatial distribution of all species was depicted on a GIS map. All four species exhibited essential oil concentrations higher than those reported in earlier literature, namely O. onites 3-4.3%, S. thymbra 4-6.5%, C. capitatus 3.7-5.6% and O. hirtum 5.5-10.0% (v/w). Carvacrol was the main constituent of the essential oils of all species, followed by γ-terpinene, p-cymene and caryophyllene, while thymol was not detected. All constituents varied remarkably among the four species, with carvacrol exhibiting the lowest variation. Carvacrol content varied between 72.3 and 89.2% in O. onites; 46.5 and 58.0% in S. thymbra; 82.9 and 90.9% in C. capitatus; and 84.4 and 93.8% in O. hirtum. By applying hierarchical cluster analysis on the basis of the essential oil constituents two main groups, divided into four subgroups of the taxa were evident. The first group consisted of O. onites and S. thymbra, while the second one of C. capitatus and O. hirtum. The results are discussed in terms of topography and climatic variation.  相似文献

Rice (Oryza sativa L.) is a semi-aquatic member of the grass family that is poorly adapted to dry environments and has greater sensitivity to water-deficits than other important cereals in this family. To increase productivity in aerobic or water-limited environments rice must overcome its adaptations to flooded environments. Deletion mutants offer an alternative genetic resource for improving drought tolerance. Almost 3500 IR64 deletion mutants were screened under vegetative and reproductive stage drought stress in the field and evaluated for leaf drying and/or grain yield. Seven novel conditional mutants of rice which showed gain of function through continued growth as drought stress developed compared to the wild type were identified. Mutant recovery rate was 0.1%. Further evaluation of putative drought mutants revealed that their average shoot biomass at maturity and grain yield per plant under stress exceeded those of the wild type by two-fold. Studies under controlled conditions confirmed mutants to have continued growth of both roots and shoots as drought developed compared to the wild type, and a tendency for greater water extraction. We propose that deletions in these mutants have affected a regulator of the highly conservative growth response common to irrigated lowland rice cultivars. Our results suggest that screening deletion mutants for performance under managed drought stress in the field could be a highly effective way to identify valuable genetic resources for improved drought response and aerobic adaptation in rice.  相似文献

Phosphorus (P) deficiency is a major constraint for maize production in many low-input agroecosystems. This study was conducted to evaluate genotypic variation in both root (root architecture and morphology, including root hairs) and plant growth traits associated with the adaptation of maize landraces to a P-deficient Andisol in two locations in the Central Mexican highlands. Two hundred and forty-two accessions from the Purhepecha Plateau, Michoacan were grown in Ponzomaran with low (23 kg P2O5 ha−1) and high (97 kg P2O5 ha−1) P fertilization under rain-fed field conditions, and subsequently a subset of 50 contrasting accessions were planted in the succeeding crop cycle in Bonilla. Accessions differed greatly in plant growth, root morphology and P efficiency defined as growth with suboptimal P availability. The accessions were divided into 3 categories of P efficiency using principal component and cluster analyses, and 4 categories according to the retained principal component and their relative weight for each genotype in combination with growth or yield potential. The distribution of accessions among three phosphorus efficiency classes was stable across locations. Phosphorus-efficient accessions had greater biomass, root to shoot ratio, nodal rooting, nodal root laterals, and nodal root hair density and length of nodal root main axis, and first-order laterals under P deficiency. Biomass allocation to roots, as quantified by the allometric partitioning coefficient (K) was not altered by P availability in the efficient accessions, but inefficient accessions had a lower K under low P conditions. Accessions with enhanced nodal rooting and laterals had greater growth under low P. Dense root hairs on nodal root main axes and first-order laterals conferred a marked benefit under low P, as evidenced by increased plant biomass. Late maturity improved growth and yield under low P. These results indicate that landraces of the Central Mexican highlands exhibit variation for several root traits that may be useful for genetic improvement of P efficiency in maize.  相似文献

To uncover the genetic contributions of agronomic traits to content of total sugar (TS) and find indicator traits for indirect selection on TS in the flue-cured tobacco (Nicotiana tobacum L.), multivariable conditional analysis was conducted based on a genetic model containing additive–dominance and their interactions with environments. Fourteen cultivars (or breeding lines) and derived 41 F1 crosses were grown at four locations in Yunnan province, China. Significant phenotypic contribution to TS was detected for six agronomic traits, plant height (PH), girth of stem (GS), internode length (INL), number of leaves (NL), length of middle leaves (LML) and width of middle leaves (WML). There was large contribution of additive effects due to each of the five agronomic traits (PH, GS, INL, LML and WML). The contribution ratio of dominance effect was high due to PH. By serving as high contributor of additive effects to TS and having high ratios of additive variance to phenotypic variance, INL and PH could be used as indicative agronomic traits for selecting breeding lines with suitable TS. Among the six agronomic traits, PH had the highest contribution to dominance effects of TS for most F1 crosses, and could be used for selecting the crosses with suitable TS.  相似文献

The commercial production of peppermint (Mentha piperita L.) depends on the genetic structure and ecological conditions affecting yield and oil composition. To determine yield and quality characters of two peppermint clones (Clone-3 and Clone-8), field experiments were carried out at four different locations (Ayd?n, Bursa, Izmir and Tokat) in Turkey during 2007 and 2008. Locations with warmer climate gave higher fresh herbage yield (Ayd?n and Izmir 37.0 t/ha and 36.8 t/ha, respectively). Although vigor canopy caused maximum fresh herbage yield, it decreased dried leaf yield due to the defoliation of leaves near to the ground. The main components menthol and menthone showed significant variation with different ecologies. Menthol contents were higher in temperate locations (Bursa and Tokat), while menthone contents were lower. The differences in oil composition of Clone-3 were lower than that of Clone-8 according to different climates, thus Clone-3 can be grown widely in various ecological conditions for oil productions as compared to the Clone-8. It was also concluded that temperate location was more suitable for peppermint oil production with high menthol contents.  相似文献

The susceptibility of three sweet potato cultivars (Ipomoea batatas L.) C4, TIS 3290 and TIS 9162 was evaluated against 156 isolates of Meloidogyne spp. with the aim to include resistant/tolerant sweet potato cultivars in a crop rotation scheme for the management of root-knot nematodes. The nematode isolates corresponded to races 1, 2 and 3 of Meloidogyne arenaria (n = 7), races 1, 2, 3 and 4 of M. incognita (n = 131) and Meloidogyne javanica (n = 18). Also, the isolates of M. incognita were differentiated in virulence groups: Pepper (n = 35), Pepper-Mi (n = 25), Tomato (n = 41) and Tomato-Mi (n = 30), depending on their ability to parasitize resistant pepper and tomato cultivars. The tested isolates of M. javanica parasitized C4 and TIS 3290, but not TIS 9162, whereas M. arenaria parasitized C4 and TIS 9162, but not TIS 3290, and M. incognita was able to parasitize the three sweet potato cultivars tested. C4 was the most susceptible cultivar to all nematode species tested, especially M. incognita, TIS 3290 was the most resistant and TIS 9162 was in between (7.2, 62.9 and 26.9% of resistant plants, respectively). Susceptibility of the sweet potato cultivars showed slight variations depending on the race or virulence group of M. incognita. The results suggest that sweet potato cultivars TIS 3290 and TIS 9162 may be used as rotation crops in fields where root-knot nematodes are present, their selection depending on the Meloidogyne isolates present. The use of resistant sweet potato cultivars would be preferably combined with other management practices to avoid virulence selection in nematode isolates.  相似文献

Wild oat (Avena fatua L.) is the most troublesome weed in cereal crops in Argentina. With the aim of studying the effects of different herbicides, doses, and wild oat growth stage at application on weed control and crop yield, field experiments were conducted in wheat and barley crops during three growing seasons in the south of Buenos Aires Province, Argentina. Treatments were post-emergence applications of new herbicide, pinoxaden + cloquintocet mexyl (5%-1.25%), at doses that ranged from 20 g to 60 g a.i. pinoxaden ha−1, applied at two to three leaves and the beginning of tillering of wild oat. In addition, standard treatments were included and applied at the same wild oat growth stages. Diclofop methyl at 511 g a.i. ha−1 and fenoxaprop-p-ethyl at 55 g a.i. ha−1 were applied in barley. In wheat, diclofop methyl was replaced by clodinafop-propargyl + cloquintocet mexyl (24%-6%) at 36 g a.i. clodinafop-propargyl + 9 g cloquintocet mexyl ha−1 and in 2008/09 wheat experiments, iodosulfuron plus metsulfuron methyl (5%-60%) at 3.75 g a.i. ha−1 + 3 g a.i. ha−1 also was included. In both crops, pinoxaden at 30 g a.i. ha−1 and at higher rates, fenoxaprop-p-ethyl and clodinafop-propargyl gave the best control of wild oat. In 2006/07 wheat crops, treatments applied at tiller initiation provided better control than the early timing averaged across herbicides. However, wheat yield generally was greater with early application. In barley, wild oat control and crop yield were similar regarding time of application. Variations in crop yield were correlated with grain number m−2 both in wheat and barley, but relationships between both grain number and spikes m−2 and with grains per spike were identified only in wheat.  相似文献

Due to adaptation to new ecological and manmade conditions, the large diversity evolved in the European common bean germplasm is of particular interest for plant breeding. The knowledge of the genetic relationships within and among races and gene pools and their performance per se will provide bean breeders with a starting point in designing crosses using contrasting and complementary parents to broaden the genetic base within the different commercial classes. A genetic study of seed size variation and protein markers in progeny derived from 16 intraracial, interracial and inter-gene pools European common bean populations was conducted. General and specific combining ability (GCA and SCA) values were significant for seed weight, indicating that both additive and nonadditive genetic effects were involved in conditioning seed weight. Interracial populations showed transgressive values due to the accumulation of large-seeded alleles. Genetic variation inside Andean germplasm, and Chile and Peru races in particular, exhibited useful genetic progress in these populations, providing lines with a large seed size, and so, an excellent market potential. The distribution of incompatibility between both gene pools (Mesoamerican and Andean) of the common bean was explored. Inter-gene pool populations provided lower means of inbred segregants than the mid-parent value. Therefore, a good option it would be select for large seed size according to a recurrent or congruity inbred-backcrossing selection programs. Analysis of allele markers frequencies in inter-gene populations showed segregation distortion with a higher than expected frequency of alleles from the Mesoamerican gene pool, many of which were fixed in the F7 lines. The presence of a great percentage of markers that showed segregation distortion in these populations (87%) indicated that this phenomenon can be amplified by using distance related common bean genotypes. In addition, a high percentage of heterozygotes for the Phs locus (for the seed storage protein phaseolin) was found, which suggest that the Andean homozygous TT could not be expressed in Mesoamerican genetic background due to the action of some form of female specific mechanisms that affected gene exchange between parental germplasm in inter-gene pool populations. The present work provides useful information in the establishment of large seed size germplasm that could have a great deal of interest among breeders and may offer some possibilities to exploit existing variation within and between common bean races.  相似文献

Conditions to cultivate medicinally important herb Centella asiatica in hydroponic system are reported here for the first time. Growth kinetics of hydroponically grown plants was monitored over a period of 70 days. The maximum growth and dry matter accumulation (156.3% increment over the initial inoculum weight) in the cultured plants occurred around 42nd day. High Performance Liquid Chromatography (HPLC) analysis of the bioactive centellosides in the crude triterpenoids extract of the harvested leaves showed the presence of 11 mg, 1.7 mg, 36.6 mg and 6.3 mg of madecassoside, asiaticoside, madecassic acid and asiatic acid on per gram dry weight basis, respectively. The results of this study suggest that the cultivation of C. asiatica in hydroponic systems can be an effective platform for the production of clean and good quality C. asiatica herb for the pharmaceutical companies.  相似文献

The study deals with evaluation of antifungal and antiaflatoxigenic Caesulia axillaris Roxb. essential oil (EO) against herbal raw materials deteriorating fungi and its free radical scavenging activity. During mycoflora analysis these herbal raw materials were found to be severely contaminated by different fungi and aflatoxins. A total of nine different fungal species were isolated from three herbal raw materials. Aspergillus flavus LHPtc was recorded as the highest aflatoxin B1 producing strain. EOs of some plants were tested for their fungitoxicity against the toxigenic strain A. flavus LHPtc, and C. axillaris EO was found as potent fungitoxicant. C. axillaris EO was chemically characterized through GC-MS analysis which depicted the presence of 18 compounds, dl-limonene and Euasarone being the major components. The EO exhibited broad spectrum of fungitoxicity against fungi causing postharvest deterioration of herbal raw materials. At 1.0 μl ml−1 the oil showed complete inhibition of fungal growth and aflatoxin B1 production was inhibited at 0.8 μl ml−1. Free radical scavenging activity of the oil was also recorded by 2,2-diphenyl-1-picrylhydrazyl assay, and its IC50 value was found 18 μl ml−1. The safety limit of the EO was determined in terms of LD50 on mice, which was 9166.6 μl kg−1, suggesting its non mammalian toxicity. The EO of C. axillaris may be recommended as a plant based preservative in enhancement of shelf life of herbal raw materials by preventing their lipid peroxidation as well as biodeterioration due to fungal and aflatoxin contamination.  相似文献

The contribution of ear photosynthesis to grain filling in wheat (Triticum aestivum L.) is not well known. The main objective of this work was to evaluate this contribution through three different experimental approaches: (1) ear photosynthesis was reduced by removing awns or shading the ears (in combination with a defoliation treatment), (2) grain weight per ear was compared in an ‘all shaded’ crop versus plants where only the vegetative parts were shaded (‘ear emerging’), and (3) ear photosynthesis was reduced with DCMU (3-(3,4-dichlorophenyl)-1,1-dimethylurea), a specific inhibitor of photosystem II.  相似文献

In 2009, a survey was conducted of cereal fields in five prefectures of Greece to establish the frequency and distribution of herbicide-resistant sterile wild oat (Avena sterilis L.). In total, 104 sterile wild oat accessions were collected and screened in a field experiment (conducted in 2009 and repeated in 2010) with several herbicides commonly used to control this weed. Most of the sterile wild oat accessions (89%) were classed as resistant (or developing resistant) to the ACCase-inhibiting herbicide diclofop, while resistance to other ACCase-inhibiting herbicides was markedly lower. The results of the pot experiments showed that some of the sterile wild oat accessions were found to have a very high level of diclofop resistance (resistance index up to 28.6), while cross-resistance with other herbicides was common. The levels of resistance and cross-resistance patterns varied among biotypes with different amount and time of selection pressure, indicating either more than one mechanism of resistance or different resistance mutations in these sterile wild oat biotypes. LA14, which had the highest diclofop resistance level (28.6 resistance index), showed resistance to all APP herbicides applied and non-ACCase inhibitors. Alternative ACCase-inhibiting herbicides, namely tralkoxydim and pinoxaden remain effective on 86 and 92% of the tested sterile wild oat populations, respectively. For the ALS-inhibiting herbicide mesosulfuron + iodosulfuron, nearly all the sterile wild oat accessions were susceptible (97%), with only 3 of them classed as developing resistance. Therefore, there is an opportunity to effectively control sterile wild oat by selecting from a wide range of herbicides and other cultural practices. Early post-emergence herbicide application is strongly suggested, since it could decrease the number of resistant seeds in the field and slow down the dispersal of this major problem.  相似文献

Early and late leaf spot (caused by Cercospora arachidicola and Cercosporidium personatum, respectively) are major yield-reducing diseases of peanut (Arachis hypogaea L.) in the southeastern U.S. Effective suppression of both early and late leaf spot, collectively called ‘leaf spot’, can be reached with integrated disease management (IDM) systems utilizing peanut cultivars with moderate levels of resistance to these pathogens. A Bolivian landrace cultivar, Bayo Grande, and a series of progeny breeding lines were evaluated under two IDM systems consisting of various reduced application fungicide regimes and/or tillage treatments in field experiments over two years. In the absence of fungicides or when averaged across fungicide regimes, Bayo Grande and breeding lines had better leaf spot resistance when compared to cultivars Georgia Green, MDR-98 and C-99R in all experiments across years. The addition of three or four fungicide applications to Bayo Grande and several of the breeding lines suppressed leaf spot to levels similar to those genotypes and Georgia Green under the full regime. Conservation tillage reduced the need for fungicides in most genotypes in both years. Yields were higher for Bayo Grande and the breeding lines compared to Georgia Green in three of the four experiments across years. When compared across genotype, yields were greatest for the full season fungicide regime, except in 2002 when yields were similar for all treatments. Leaf spot severity was reduced in conservation tillage compared to conventional tillage in most varieties in the absence of fungicides. However, yields tended to be lower in conservation tillage treatments than in conventional tillage. Under conservation tillage, yields of Bayo Grande and two of the breeding lines were negatively impacted in one year and were overall lower in the other year. Several of these new Bolivian-derived genotypes show promise for use in a reduced fungicide and/or conservation tillage IDM system with the potential to lessen fungicide use compared to standard production practices while maintaining low leaf spot levels and high yields.  相似文献

Very few efforts have been made to improve the nutritional quality of groundnut, as biochemical estimation of quality traits is laborious and uneconomic; hence, it is difficult to improve them through traditional breeding alone. Identification of molecular markers for quality traits will have a great impact in molecular breeding. An attempt was made to identify microsatellite or simple sequence repeat (SSR) markers for important nutritional traits (protein content, oil content and oil quality in terms of oleic acid, linoleic acid and oleic/linoleic acid ratio) in a mapping population consisting of 146 recombinant inbreed lines (RILs) of a cross TG26 × GPBD4. Phenotyping data analysis for quality traits showed significant variation in the population and environment, genotype × environment interaction and high heritability was observed for all the traits. Negative correlation between protein content and oil content, oleic acid and linoleic acid indicated their antagonistic nature. After screening >1000 SSR markers, a partial genetic linkage map comprising of 45 SSR loci on 8 linkage groups with an average inter-marker distance of 14.62 cM was developed. QTL analysis based on single marker analysis (SMA) and composite interval mapping identified some candidate SSR markers associated with major QTLs as well as several minor QTLs for the nutritional traits. Validation of these major QTLs using a wider genetic background may provide the markers for molecular breeding for improving groundnut for nutritional traits.  相似文献

Grain hardness is one of the most important quality characteristics of cultivated bread wheat (Triticum aestivum L.). A large deletion in the puroindoline a (Pina) gene or single nucleotide polymorphisms (SNPs) in the puroindoline b (Pinb) gene results in hard grain texture. So far, nine Pina alleles (Pina-D1aPina-D1b, Pina-D1kPina-D1q) and seventeen Pinb alleles (Pinb-D1aPinb-D1g, Pinb-D1pPinb-D1ab) have been identified in bread wheat. The major Pina and Pinb alleles identified in hard wheat cultivars are Pina-D1b, Pinb-D1b, Pinb-D1c and Pinb-D1d. In this study, a three-primer PCR system was employed to develop nine co-dominant STS markers for genotyping Pina-D1a and Pina-D1b, whereas temperature-switch (TS) PCR was used to develop six co-dominant SNP markers for genotyping the Pinb-D1a, Pinb-D1b, Pinb-D1c and Pinb-D1d alleles. These STS and TS-PCR markers were used to verify the grain hardness genotype of 100 wheat cultivars. The reliability and genotyping accuracy of TS-PCR markers were confirmed through sequencing of PCR products and a comparison with previously published results. Therefore, STS and TS-PCR markers offer a simple, cost-effective and reliable method for high-throughput genotyping Pina and Pinb alleles to select grain hardness in wheat quality breeding programs and for wheat market classification.  相似文献
