作为内蒙古重要的草地资源,克氏针茅草原在我国畜牧业生产中占有重要的地位。土壤呼吸是大气与陆地生态系统碳循环的主要通量,是土壤碳输出的主要途径。试验区设置在内蒙古锡林浩特市东部毛登牧场内以克氏针茅建群的典型草原;在2011-2013年采用开顶式生长室(OTC)对试验样地进行模拟增温增雨处理,在生长季(5-9月份)使用开路式土壤碳通量测量系统(LI-8100, LI-COR,NE) 对试验样地土壤呼吸强度进行测定;同时监测试验样地气温、土壤温湿度等环境因子,采集土壤样品并室内分析土壤养分特征和微生物生物量,并对土壤呼吸与环境因子间进行相关性分析。克氏针茅草原土壤呼吸受空气温度和土壤含水量影响显著(P<0.05),分别呈现出二项函数和线性相关关系;不同处理下土壤呼吸在相同时间存在显著差异(P<0.05),2012年增温增雨与增温处理下土壤呼吸速率分别为2.60和1.96 μmol/(m2·s), 2013年增温增雨与增温处理下土壤呼吸速率分别为3.38和2.09 μmol/(m2·s);增温增雨处理下,克氏针茅草原空气温度的升高和土壤含水量的降低均对土壤呼吸产生了抑制。土壤呼吸温度敏感度(Q10)在2011-2012年中增温增雨处理明显高于对照处理和控温处理;2011-2013年控温处理都低于对照处理。本研究表明,土壤呼吸主要受水热条件影响,并且空气温度升高和土壤含水量的降低均对土壤呼吸有抑制作用,增温增雨处理下土壤呼吸速率要高于增温处理;土壤呼吸温度敏感系数与土壤温度、土壤含水量、土壤有机碳、全氮和微生物等环境因子之间的关系较为复杂,还有待深入研究。  相似文献

Prescribed burning of aboveground biomass in tallgrass prairie is common and may influence dynamics and magnitudes of carbon (C) movement between the surface and atmosphere. Carbon dioxide (CO2) fluxes were measured for 2 yr using conditional sampling systems on two adjacent watersheds in an ungrazed tallgrass prairie near Manhattan, Kansas. One watershed was burned annually (BA) and the other biennially (BB). Leaf and soil CO2 fluxes were measured in the source area. Net ecosystem exchange (NEE) of CO2 reached a maximum daily gain of 26.4 g CO2·m?2·d?1 (flux toward surface is positive) in July 1998 (year when both sites were burned and precipitation was above normal); gains were similar between sites in 1998. The maximum daily NEE loss of CO2 was ?21.8 g CO2·m?2·d?1 from BA in September 1997 (year when only BA was burned and precipitation was below normal). When data were integrated over the two years, both sites were net sources of atmospheric CO2; NEE was ?389 g C·m?2·2 yr?1 on BA and ?195 g C·m?2·2 yr?1 on BB. Burning increased canopy size and photosynthesis, but the greater photosynthesis was offset by corresponding increases in respiration (from canopy and soil). Carbon losses from fire represented 6–10% of annual CO2 emissions (bulk came from soil and canopy respiration). Data suggest that annual burning promotes C loss compared to less-frequently burned tallgrass prairie where prairie is not grazed by ungulates. Greater precipitation in 1998 caused large increases in biomass and a more positive growing season NEE, indicating that C sequestration appears more likely when precipitation is high. Because C inputs (photosynthesis) and losses (canopy and soil respiration) were large, small measurement or modeling errors could confound attempts to determine if the ecosystems are long-term CO2 sources or sinks.  相似文献

Grasslands and agroecosystems occupy one-third of the terrestrial area, but their contribution to the global carbon cycle remains uncertain. We used a set of 316 site-years of CO2 exchange measurements to quantify gross primary productivity, respiration, and light-response parameters of grasslands, shrublands/savanna, wetlands, and cropland ecosystems worldwide. We analyzed data from 72 global flux-tower sites partitioned into gross photosynthesis and ecosystem respiration with the use of the light-response method (Gilmanov, T. G., D. A. Johnson, and N. Z. Saliendra. 2003. Growing season CO2 fluxes in a sagebrush-steppe ecosystem in Idaho: Bowen ratio/energy balance measurements and modeling. Basic and Applied Ecology 4:167–183) from the RANGEFLUX and WORLDGRASSAGRIFLUX data sets supplemented by 46 sites from the FLUXNET La Thuile data set partitioned with the use of the temperature-response method (Reichstein, M., E. Falge, D. Baldocchi, D. Papale, R. Valentini, M. Aubinet, P. Berbigier, C. Bernhofer, N. Buchmann, M. Falk, T. Gilmanov, A. Granier, T. Grünwald, K. Havránková, D. Janous, A. Knohl, T. Laurela, A. Lohila, D. Loustau, G. Matteucci, T. Meyers, F. Miglietta, J. M. Ourcival, D. Perrin, J. Pumpanen, S. Rambal, E. Rotenberg, M. Sanz, J. Tenhunen, G. Seufert, F. Vaccari, T. Vesala, and D. Yakir. 2005. On the separation of net ecosystem exchange into assimilation and ecosystem respiration: review and improved algorithm. Global Change Biology 11:1424–1439). Maximum values of the quantum yield (α=75 mmol · mol?1), photosynthetic capacity (Amax=3.4 mg CO2 · m?2 · s?1), gross photosynthesis (Pg,max=116 g CO2 · m?2 · d?1), and ecological light-use efficiency (εecol=59 mmol · mol?1) of managed grasslands and high-production croplands exceeded those of most forest ecosystems, indicating the potential of nonforest ecosystems for uptake of atmospheric CO2. Maximum values of gross primary production (8 600 g CO2 · m?2 · yr?1), total ecosystem respiration (7 900 g CO2 · m?2 · yr?1), and net CO2 exchange (2 400 g CO2 · m?2 · yr?1) were observed for intensively managed grasslands and high-yield crops, and are comparable to or higher than those for forest ecosystems, excluding some tropical forests. On average, 80% of the nonforest sites were apparent sinks for atmospheric CO2, with mean net uptake of 700 g CO2 · m?2 · yr?1 for intensive grasslands and 933 g CO2 · m?2 · d?1 for croplands. However, part of these apparent sinks is accumulated in crops and forage, which are carbon pools that are harvested, transported, and decomposed off site. Therefore, although agricultural fields may be predominantly sinks for atmospheric CO2, this does not imply that they are necessarily increasing their carbon stock.  相似文献

A mobile, closed-chamber system (CC) was used to measure carbon and water fluxes on four land-use types common in the Kazakh steppe ecoregion. Land uses represented crop (wheat or barley, WB), abandoned land (AL), crested wheatgrass (CW), and virgin land (VL). Measurements were conducted during the growing season of 2002 in northern Kazakhstan at three locations (blocks) 15–20 km apart. The CC allowed the measurement of the carbon flux components of net ecosystem exchange (NEE), ecosystem respiration (RE) and soil respiration (RS), together with evapotranspiration (ET). Nonlinear regression analyses were used to model gross primary production (GPP) and ET as a function of photosynthetically active radiation (Q); RE and RS were modeled based on air (Tair) and soil (Ts) temperature, respectively. GPP, RE, RS, and ET were estimated for the entire year with the use of continuous 20-min means of Q, Tair, and Ts. Annual NEE indicated that AL gained 536 g CO2 · m?2, WB lost - 191 g CO2 · m?2, CW was near equilibrium (? 14 g CO2 · m?2), and VL exhibited considerable carbon accumulation (153 g CO2 · m?2). The lower GPP values of the land-use types dominated by native species (CW and VL) compared to WB and AL were compensated by positive NEE values that were maintained during a longer growing season. As expected, VL and CW allocated a larger proportion of their carbon assimilates belowground. Non–growing-season RE accounted for about 19% of annual RE in all land-use types. The results of this landscape-level study suggest that carbon lost by cultivation of VLs is partially being restored when fields are left uncultivated, and that VLs are net sinks of carbon. Estimations of carbon balances have important management implications, such as estimation of ecosystem productivity and carbon credit certification.  相似文献

This clinical study analysed the anaesthetic sparing effect of a medetomidine constant rate infusion (CRI) during isoflurane anaesthesia in horses. Forty healthy horses undergoing different types of orthopaedic and soft tissue surgeries were studied in a randomized trial. Orthopaedic surgeries were primarily arthroscopies and splint bone extractions. Soft tissue surgeries were principally castrations with one ovariectomy. All horses received 0.03 mg kg?1 acepromazine IM 1 hour prior to sedation. Group A (11 orthopaedic and nine soft tissue surgeries), was sedated with 1.1 mg kg?1 xylazine IV, group B (13 orthopaedic and seven soft tissue surgeries) with 7 µg kg?1 medetomidine IV. Anaesthesia was induced in both groups with 2.2 mg kg?1 ketamine and diazepam 0.02 mg kg?1 IV. Maintenance of anaesthesia was with isoflurane (ISO) in 100% oxygen, depth of anaesthesia was always adjusted by the first author. Group B received an additional CRI of 3.5 µg kg?1 hour?1 medetomidine. Respiratory rate (RR), heart rate (HR), mean arterial blood pressure (MAP), Fe ′ISO and Fe ′CO2 were monitored with a methane insensitive monitor (Cardiocap 5, Ohmeda, Anandic, Diessenhofen) and noted every 5 minutes. Arterial blood was withdrawn for gas analysis (PaO2, PaCO2) 5 minutes after the induction of anaesthesia and every 30 minutes thereafter. Dobutamine (DOB) was given as a CRI to maintain mean arterial blood pressure above 70 mm Hg. Data were averaged over time (sum of measurements/number of measurements) and tested for differences between groups by unpaired t‐tests. There were no significant differences between the groups in terms of body mass (group A, 508 ± 73.7 kg; group B, 529.25 ± 78.4 kg) or duration of anaesthesia (group A, 125.5 ± 36 minutes; group B, 121.5 ± 48.4 minutes). The mean Fe ′ISO required to maintain a surgical plane of anaesthesia was significantly higher in group A (1.33 ± 0.13%) than in group B (1.07 ± 0.19%; p = 2.78 × 10?5). Heart rate was different between the two groups (group A, 42.2 ± 8.3; group B, 32.6 ± 3.5; p = 8.8 × 10?5). Dobutamine requirements were higher in group A (group A, 0.72 ± 0.24 μg kg?1 minute?1; group B, 0.53 ± 0.23 μg kg?1 minute?1; p = 0.023). Respiratory rate, Fe ′CO2, PaO2, PaCO2 were not different between the groups. Adjustment of anaesthetic depth subjectively was easier with the medetomidine infusion and isoflurane (group B) than with isoflurane as a sole agent (group A). In group A 12 horses and in group B five horses showed purposeful movements on 27 (A) and 12 (B) occasions. They were given thiopental (group A, 0.0114 mg kg?1 minute?1; group B, 0.0023 mg kg?1 minute?1). In group A, a further 17 horses were given ketamine to deepen anaesthesia (52 occasions, 0.00426 mg kg?1 minute?1) whereas in group B only nine horses needed ketamine (34 occasions, 0.00179 mg kg?1 minute?1). An infusion of 3.5 µg kg?1 MED during ISO anaesthesia resulted in a significantly reduced ISO requirement.  相似文献

The secretion of acetylcholinesterase (AChE) by female and male Heligmosomoides polygyrus was studied in different in vitro culture media. AChE secretion was increased in the presence of fetal calf serum or bovine serum albumin (BSA). In the absence of crowding effects, specific AChE activity in excretion/secretion products was higher for male (2.41 ± 0.07 µmol min?1 l?1 mg?1) than for female (0.56 ± 0.04 µmol min?1 mg?1) worms but on a per nematode basis both sexes showed comparable rates of secretion. Acetylthiocholine iodide was the favoured substrate of the enzyme. When the nematodes were incubated in vitro with albendazole (ABZ), ricobendazole (RBZ), mebendazole (MBZ), levamisole (LVM), morantel (MRT) or ivermectin (IVM), at concentrations from 1 mM to 10 nM, in RPMI medium for 2 or 6 h and then transferred to a drug‐free medium (RPMI medium supplemented with 0.5 % BSA) for 24 h or continuously exposed to the drugs in supplement‐free medium (24 h), the concentration‐ and time‐dependent inhibitory effects on AChE secretion were observed. The continued exposure to the drugs for all incubation periods (with a single exception for LVM 1 mM) produced the highest levels of inhibition. Under these conditions, the concentrations inhibiting the secretion of AChE by 50 % (IC50) relative to drug‐free controls were estimated. The IC50 values ranged from 0.012 µM (IVM) to 2.96 µM (MRT). The potential of this bioassay for the selective primary evaluation of new compounds with broad‐spectrum anti‐nematodal activity is discussed.  相似文献

The aim of the study was to compare two methods of measuring physiological dead space/tidal volume ratio (Vd /Vt ) and alveolar dead space (Vd ALV). Measurements were obtained by automated single breath CO2 analysis (Ventrak 1550/Capnoguard 1265 (V&C)) and classical calculations were carried out using the Enghoff–Bohr equation in anaesthetized dogs. The V&C consists of a mainstream capnometer, a pneumotachometer, a signal processor, and computer software to determine continuous single‐breath CO2 analysis (SBT‐CO2). Eleven dogs of mixed breed (five female, six male) mean body mass 35 ± 10 kg, aged 9 months to 8 years were studied. Pre‐anaesthetic medication was acepromazine (0.03 mg kg?1) and methadone (0.1 mg kg?1). Anaesthesia was induced with propofol given to effect and maintained with propofol (10 mg kg?1 hour?1) and fentanyl (0.02 mg kg?1 hour?1) by infusion. The dog's trachea were intubated and the carbon dioxide and flow sensor were placed between the tube and the Y‐piece of a circle system (Fi O2 = 1.0). Controlled ventilation was started (tidal volume 10–15 mL kg?1) and settings were not changed throughout the measurement period. Mixed expired PCO2 (P e ?CO2) was measured by analyzing expired gas collected in a mixing box in the expiratory limb of the circle system. The dorsal pedal artery was cannulated for arterial blood sampling and analysis. Measurements were done every 15 minutes for 1 hour. The Vd /Vt was automatically calculated and displayed from the SBT‐CO2 analysis and also obtained using the Enghoff modification of the Bohr equation (Vd /Vt = (PaCO2 ? P e ?CO2)/PaCO2). Alveolar dead space was determined by calculating the physiological dead space (Vd phys = expired volume × (Vd /Vt )) and subtracting the anatomical dead space measured by SBT‐CO2. Values for Vd /Vt and Vd ALV obtained with both methods were compared using Students t‐test. The mean values from the automatic dead space calculation (Vd /Vt : 0.62–0.63; Vd ALV: 56.1–64.3 mL) did not differ significantly from those calculated arithmetically (Vd /Vt : 0.62–0.63; Vd ALV: 54.09–66.31 mL). The mean differences and standard deviation in Vd /Vt was 0.63 ± 0.00 and in Vd ALV 58.98 ± 4.28 mL for the two measurement techniques. Our data indicate that V&C can be used for accurate noninvasive online Vd /Vt and Vd ALV measurements in anaesthetized ventilated dogs.  相似文献

Buprenorphine is an effective analgesic when administered epidurally to humans. The purpose of this study was to compare epidural buprenorphine (B; n = 10) with epidural morphine (M; n = 10) for post‐operative analgesia in dogs undergoing cranial cruciate ligament repair. All dogs were premedicated with acepromazine (0.1 mg kg?1 IM), induced with propofol (4–6 mg kg?1 IV) and maintained with halothane in oxygen. Dogs were randomly assigned to receive B (0.004 mg kg?1) or M (0.1 mg kg?1) in the lumbosacral epidural space in a total volume of 0.2 mL kg?1. End‐tidal halothane and CO2 and temperature were recorded every 15 minutes until extubation (t = 0). A numerical rating pain score (SPS) was recorded at t = 0, 1, 2, 4, 6, 10 and 24 hours by a blinded observer. Dogs received rescue morphine (1.0 mg kg?1 IM) if indicated by SPS and the time of rescue analgesic administration was recorded. Observable side‐effects such as urinary retention, sedation or pruritus were recorded. Data were analyzed with repeated measures anova . Mean ± SD body weight (kg) and age (yrs) for B dogs was 34.2 ± 10.8 and 5.5 ± 2.8; for M dogs these values were 36.6 ± 13.5 and 5.9 ± 3.3. Mean ± SD SPS for B dogs at t = 0, 1, 2, 4, 6, 10 and 24 hours were 1.2 ± 0.75, 3.2 ± 2.0, 4.5 ± 4.3, 4.6 ± 3.4, 4.7 ± 3.0, 5.0 ± 4.9 and 5.1 ± 3.5. For M dogs these values were 1.7 ± 0.5, 2.6 ± 2.0, 3.7 ± 0.75, 4.2 ± 2.2, 4.1 ± 3.0, 3.1 ± 2.1 and 3.9 ± 1.9. There were no significant differences between B and M with respect to SPS, times or frequency of rescue morphine administration, end‐tidal halothane and CO2, or esophageal temperature. Fifty per cent of dogs in both groups required rescue morphine. Buprenorphine is as effective as morphine for epidural analgesia in healthy dogs undergoing hindlimb orthopedic surgery.  相似文献

Lidocaine decreases minimum alveolar concentration (MAC) of inhalational anesthetics. This study determined the influence of a low dose, 50 µg kg?1 minute?1 (LDI) and high dose, 200 µg kg?1 minute?1 (HDI) constant rate infusion of lidocaine on the MAC of isoflurane (I) in dogs. Ten mongrel dogs were anesthetized with I in oxygen and mechanically ventilated. End‐tidal anesthetic (Fe ′A) and CO2 (Pe ′CO2) concentrations were monitored at the endotracheal tube adaptor with an infrared gas analyzer calibrated before each experiment with a standardized calibration gas mixture designed for the analyzer. Pe ′CO2 and body temperature were maintained within normal limits. Noxious stimuli included clamping the hindlimb paw (HC) and electrical current (50 V at 50 cycles second?1 for 10 milliseconds pulse?1) applied subcutaneously to the forelimb (FE) at the level of the ulna. After an initial equilibration period of at least 40 minutes at an Fe ′A of 1.7%, the Fe ′A was decreased to a value close to the estimated MAC for dogs. MAC was defined as the Fe ′A mid‐way between the value permitting and preventing purposeful movement. Following baseline MAC, a loading dose of 2 mg kg?1 of lidocaine IV was administered over 3 minutes followed by the LDI, and MAC determinations for the combination started after 30 minutes of infusion. Once determined, the lidocaine infusion was stopped for 30 minutes and the dog maintained at the ETC that prevented movement without the lidocaine. Following this period, a second loading dose of lidocaine was given (2 mg kg?1) over 3 minutes followed by the HDI, and the MAC determination procedure repeated after 30 minutes of infusion. Data were analyzed using an anova for repeated measures. MAC of I was 1.34 ± 0.035% (mean ± SEM) for both the FE and HC stimuli. The LDI significantly decreased MAC to 1.09 ± 0.043% (18.7% reduction) and HDI to 0.76 ± 0.030% (43.3% reduction). In conclusion, lidocaine infusions decreased the MAC of isoflurane in a dose‐dependent manner.  相似文献

ObjectiveTo compare cardiac output (CO) measured by Doppler echocardiography and thermodilution techniques in spontaneously breathing dogs during continuous infusion of propofol. To do so, CO was obtained using the thermodilution method (COTD) and Doppler evaluation of pulmonary flow (CODP) and aortic flow (CODA).Study designProspective cohort study.AnimalsEight adult dogs weighing 8.3 ± 2.0 kg.MethodsPropofol was used for induction (7.5 ± 1.9 mg kg?1 IV) followed by a continuous rate infusion at 0.7 mg kg?1 minute?1. The animals were positioned in left lateral recumbency on an echocardiography table that allowed for positioning of the transducer at the 3rd and 5th intercostal spaces of the left hemithorax for Doppler evaluation of pulmonary and aortic valves, respectively. CODP and CODA were calculated from pulmonary and aortic velocity spectra, respectively. A pulmonary artery catheter was inserted via the jugular vein and positioned inside the lumen of the pulmonary artery in order to evaluate COTD. The first measurement of COTD, CODP and CODA was performed 30 minutes after beginning continuous infusion (T0) and then at 15‐minute intervals (T15, T30, T45 and T60). Numeric data were submitted to two‐way anova for repeated measurements, Pearson’s correlation coefficient and Bland &; Altman analysis. Data are presented as mean ± SD.ResultsAt T0, COTD was lower than CODA. CODA was higher than COTD and CODP at T30, T45 and T60. The difference between the COTD and CODP, when all data were included, was ?0.04 ± 0.22 L minute?1 and Pearson’s correlation coefficient (r) was 0.86. The difference between the COTD and CODA was ?0.87 ± 0.54 L minute?1 and r = 0.69. For COTD and CODP, the difference was ?0.82 ± 0.59 L minute?1 and r = 0.61.ConclusionDoppler evaluation of pulmonary flow was a clinically acceptable method for assessing the CO in propofol‐anesthetized dogs.  相似文献

A two-year study was conducted with an overall objective of characterising the structure and function of an arid rangeland in Kenya. A plot measuring 100 × 100m was used for this study. Data on rainfall and temperature were recorded at the plot site whereas data on evaporation rates and relative humidity were obtained from the meteorological office near the study site.

Herbaceous aboveground material was sampled at monthly intervals using a rectangular 0.25m2 quadrat frame. Clipped material was separated by species and classified dead or live by physical examination. A 5cm diameter metal soil corer was used to sample belowground plant material, at monthly intervals. The sampled material was washed with running water over 2mm sieves and classified dead or live using the vital staining technique. All weights and calculations were based on organic weight.

Total aboveground standing crop ranged from 84.6g m?2 to 295.4g m?2, with a mean of 162.3 ± 60.6g m?2. Mean monthly aboveground standing crop for 1992 and 1993 was 142.8 ± 53.8 and 178.5 ± 63.3g m?2 respectively. The two values were significantly different (p<0.10). Aboveground biomass yield ranged from 17.7g m?2 to 242.7g m?2, with a mean of 104.3 ± 58g m?2 and a coefficient of variation of 58%. Mean aboveground standing crop was 59 ± 24g m?2 Monthly values ranged from 28.8g m?2 to 120g m?2, with a 38% coefficient of variation.

The range for total belowground standing crop was from 83.3g m?2 to 232.7g m?2, and a mean of 155.2 ± 46g m?2. The values had a coefficient of variation of 30%. Mean total monthly belowground plant material yield for 1992 and 1993 was 137.6 ± 41g m?2 and 169.9 ± 46g m?2 The coefficients of variation were 59% and 28% respectively. The mean monthly belowground biomass yield was 51.6 ± 33g m?2 with a coefficient of variation of 64%. Mean monthly yield for belowground dead material was 103.7 ± 32g m?2, with a coefficient of variation of 31%. There was no significant difference (p>0.01) in the mean belowground dead material yield between 1992 and 1993.

In 1992, annual NPP was 439.2g m?2, giving a net primary productivity of 1.22g m?2day?l. Monthly NPP ranged from 17.2g m?2 to 90.1g m?2 In 1993, annual NPP was 944.5g m?2, equivalent to a net primary productivity of 2.62g m?2 day?1. Monthly NPP was between 27.4g m?2 and 548.6g m?2. Over the 1992–1993 period, NPP was 1 383.7g m?2, equivalent to a productivity of 1.92g m?2day?1. Trends in monthly NPP closely followed the trend in rainfall. On the whole, herbaceous vegetation production and productivity were episodic in nature and closely linked to rainfall. The high primary productivity puts arid and semi-arid rangelands under sharp focus as CO2 sinks, whose role in the amelioration of greenhouse effect could be more important than is currently appreciated.  相似文献

Stained cytological specimens from 24 dogs with spontaneous soft tissue sarcomas [fibrosarcoma (n = 8), liposarcoma (n = 8) and haemangiopericytoma (n = 8)], and 24 dogs with reactive connective tissue lesions [granulation tissue (n = 12) and dermal fibrosis (n = 12)] were analysed by computer‐assisted nuclear morphometry. The studied morphometric parameters were: mean nuclear area (MNA; µm2), mean nuclear perimeter (MNP; µm), mean nuclear diameter (MND mean; µm), minimum nuclear diameter (Dmin; µm) and maximum nuclear diameter (Dmax; µm). The study aimed to evaluate (1) possibility for quantitative differentiation of soft tissue sarcomas from reactive connective tissue lesions and (2) by using cytomorphometry, to differentiate the various histopathological soft tissue sarcomas subtypes in dogs. The mean values of all nuclear cytomorphometric parameters (except for Dmax) were statistically significantly higher in reactive connective tissue processes than in soft tissue sarcomas. At the same time, however, there were no considerable differences among the different sarcoma subtypes. The results demonstrated that the quantitative differentiation of reactive connective tissue processes from soft tissue sarcomas in dogs is possible, but the same was not true for the different canine soft tissue sarcoma subtypes. Further investigations on this topic are necessary for thorough explication of the role of quantitative morphology in the diagnostics of mesenchymal neoplasms and tumour‐like fibrous lesions in dogs  相似文献

The purpose of this study was to determine the plasma pharmacokinetics (PK) and toxicity of zebularine, an oral cytidine analog with demethylating activity, in dogs. Plasma zebularine concentrations were determined by HPLC‐MS/MS following an oral zebularine dose of 8 or 4 mg kg?1. Plasma zebularine clearance was constant. Mean maximum concentration (Cmax) was 23 ± 4.8 and 8.6 ± 1.4 µM following 8 and 4 mg kg?1, respectively. Mean half‐life was 5.7 ± 0.84 and 7.1 ± 2.1 following 8 and 4 mg kg?1, respectively. A single 8 mg kg?1 dose was well tolerated. Daily 4 mg kg?1 treatment in three laboratory dogs resulted in grade 4 neutropenia (n = 3), grade 1 anorexia (n = 2) and grade 1 or 2 dermatologic changes (n = 2). All adverse events resolved with supportive care. A 4 mg kg?1 dose every 21 days was well tolerated. A follow‐up dose escalation study is in progress with a lower starting dose.  相似文献

Non‐steroidal anti‐inflammatory drugs may potentiate the opioid induced reduction in volatile anaesthetic requirements ( Gomez de Segura et al. 1998 ). This study determined the reduction in the MAC of isoflurane (ISO) produced by ketoprofen (KETO) in dogs anaesthetized with fentanyl (FENT) and ISO. Six healthy female crossbred dogs, weighing 13.5 ± 1.3 (mean ± SD) kg and aged 3.0 ± 0.9 years were studied. Approval of the study was obtained from the institutional ethics committee. Anaesthesia was induced in all dogs via a facemask with 5% ISO in 5 L minute?1 oxygen. The dogs' trachea were intubated and lungs were ventilated to maintain normocapnia (Pe ′CO2 4.7–6 kPa, 35–45 mm Hg). A heating pad was used to maintain body temperature. The animals were anaesthetized four times at one week intervals with the following anaesthetic and analgesic protocols randomly administered. Study 1, MAC (ISO); Isoflurane MAC. Study 2, MAC (ISO + FENT); dogs anaesthetized with ISO received a loading dose of 30 µg kg?1 FENT IV over 20 minutes followed by a maintenance infusion of 0.2 µg kg?1 minute?1 FENT. Study 3, MAC (ISO + FENT + KETO1); as study 2 plus 1 mg kg?1 KETO. Study 4, MAC (ISO + FENT + KETO2); as study 2 plus 2 mg kg?1 KETO. The MAC was determined in duplicate by applying a standard electrical stimulus (50 V, 50 H2 over 60 seconds via two needles placed SC over the tarsus). The stimulus was applied 15 minutes after every step change in anesthetic concentration. The Wilcoxon test was applied to data to determine significant differences among MAC measurements. Fentanyl significantly decreased MAC (ISO) from 1.27% ± 0.02% to 0.73% ± 0.08%, a reduction of 42% (p < 0.05). Ketoprofen 1 mg kg?1 further decreased the MAC value (although not statistically significantly) with a reduction of 47% from MAC (ISO) (0.67% ± 0.13%) and 8% from MAC (ISO + FENT). When KETO 2 mg kg?1 was given, the reduction in MAC was 50% compared to MAC (ISO) (0.63% ± 0.08%; p < 0.05) and 14% compared to MAC (ISO + FENT) p < 0.05. Administration of KETO further reduces MAC (ISO) compared to levels observed with FENT alone. The observed reduction may have clinical advantages.  相似文献

为探索不同形态氮素输入对青藏高原高寒灌丛土壤CO2、N2O和CH4排放的影响,采集青藏高原东部金露梅高寒灌丛土壤,设置1个对照(CK)和3个添加不同形态氮素的处理(NH4Cl,NH4NO3,KNO3),在实验室恒温15℃下进行培养,分析了土壤CO2、N2O和CH4的释放量以及土壤NH4+,NO3-和可溶性有机碳(DOC)含量。结果表明:1)所有氮素处理抑制了高寒灌丛土壤CO2的排放,土壤CO2排放量与DOC浓度呈显著正相关关系;2)所有氮素处理显著增加了土壤N2O的排放,而且以添加NO3--N增加的N2O最为显著;3)高寒灌丛土壤N2O的产生过程以反硝化作用为主;4)添加不同形态氮素对高寒灌丛土壤CH4吸收没有显著影响。5)不同形态氮素施入后,高寒灌丛土壤温室气体全球增温潜能(GWP)顺序:KNO3>NH4NO3>NH4Cl>CK。  相似文献

1. The pharmacokinetics of gatifloxacin were investigated following intravenous and oral administration of a single dose at a rate of 10?mg/kg body weight in broiler chicks.

2. Drug concentration in plasma was determined using High Performance Liquid Chromatography with ultraviolet detection on samples collected at frequent intervals after drug administration.

3. Following intravenous administration, the drug was rapidly distributed (t1/2α: 0·33?±?0·008?h) and eliminated (t1/2β: 3·62?±?0·03?h; ClB: 0·48?±?0·002?l/h/kg) from the body.

4. After oral administration, the drug was rapidly absorbed (C max: 1·74?±?0·024?µg/mL; T max: 2?h) and slowly eliminated (t1/2β: 3·81?±?0·07?h) from the body. The apparent volume of distribution (Vd(area)), total body clearance (ClB) and mean residence time (MRT) were 3·61?±?0·04?l/kg, 0·66?±?0·01?l/h/kg and 7·16?±?0·08?h, respectively. The oral bioavailability of gatifloxacin was 72·96?±?1·10 %.

ObjectiveClinical experience suggests that dachshunds are prone to bradycardia during general anaesthesia (GA). The study investigated mean heart rates in anaesthetized dachshunds and other breeds of dog.Study DesignRetrospective clinical study.AnimalsSixty one dachshunds and 62 dogs of other breeds met inclusion criteria.MethodsClinical records of small breed dogs undergoing GA for spinal Magnetic Resonance Imaging between September 2008 and March 2010 were identified and examined. Data collected included drugs administered, baseline heart (HR) and respiratory (fR) rates and rectal temperature. The following information was noted from anaesthetic records: HR, fR, mean non-invasive arterial pressure and end-tidal carbon dioxide (Pe′CO2) and anaesthetic agent (Fe′agent) during the first 60 minutes of anaesthesia; rectal temperature at a time closest to the cessation of anaesthesia, ventilatory mode (spontaneous/mechanical) and fluid infusion rate. Univariate analysis with Student t-test and Fisher's test identified parameters significant in predicting a lowered HR. A multivariate analysis investigated their effect on the mean HR during GA.ResultsNo differences were found between groups regarding: age, baseline HR, baseline temperature, incidence of hypotension, Fe′agent, mean Pe′CO2 and fluid infusion rate. Body mass was smaller for dachshunds (6.7 ± 1.5 kg) compared to other breeds (7.8 ± 1.8 kg) (p = 0.0005). The lowest HR recorded was lower in dachshunds (64 ± 19 beats minute?1) compared to other breeds (72 ± 21 beats minute?1) (p = 0.03). Mean HR was lower in dachshunds (75 ± 21 beats minute?1) compared to other breeds (84 ± 21 beats minute?1) (p = 0.02). Post-procedural temperature (°C) was lower in dachshunds (35.5 ± 1.1) compared to other breeds (36.1 ± 1.2) (p = 0.007) and anticholinergics were also administered more frequently (p = 0.026). Multivariate analysis identified that breed and mean Pe′CO2 affected mean HR during anaesthesia.ConclusionThis study supported our hypothesis that dachshunds have a lower mean HR under GA than other small breed dogs.  相似文献

Objective To characterize propofol anaesthesia in pregnant ponies. Animals Fourteen pony mares, at 256 ± 49 days gestation, undergoing abdominal surgery to implant fetal and maternal vascular catheters. Materials and methods Pre‐anaesthetic medication with intravenous (IV) acepromazine (20 µg kg?1), butorphanol (20 µg kg?1) and detomidine (10 µg kg?1) was given 30 minutes before induction of anaesthesia with detomidine (10 µg kg?1) and ketamine (2 mg kg?1) IV Maternal arterial blood pressure was recorded (facial artery) throughout anaesthesia. Arterial blood gas values and plasma concentrations of glucose, lactate, cortisol and propofol were measured at 20‐minute intervals. Anaesthesia was maintained with propofol infused initially at 200 µg kg?1 minute?1, and at 130–180 µg kg?1 minute?1 after 60 minutes, ventilation was controlled with oxygen and nitrous oxide to maintain PaCO2 between 5.0 and 6.0 kPa (37.6 and 45.1 mm Hg) and PaO2 between 13.3 and 20.0 kPa (100 and 150.4 mm Hg). During anaesthesia flunixin (1 mg kg?1), procaine penicillin (6 IU) and butorphanol 80 µg kg?1 were given. Lactated Ringer's solution was infused at 10 mL kg?1 hour?1. Simultaneous fetal and maternal blood samples were withdrawn at 85–95 minutes. Recovery from anaesthesia was assisted. Results Arterial blood gas values remained within intended limits. Plasma propofol levels stabilized after 20 minutes (range 3.5–9.1 µg kg?1); disposition estimates were clearance 6.13 ± 1.51 L minute?1 (mean ± SD) and volume of distribution 117.1 ± 38.9 L (mean ± SD). Plasma cortisol increased from 193 ± 43 nmol L?1 before anaesthesia to 421 ± 96 nmol L?1 60 minutes after anaesthesia. Surgical conditions were excellent. Fetal umbilical venous pH, PO2 and PCO2 were 7.35 ± 0.04, 6.5 ± 0.5 kPa (49 ± 4 mm Hg) and 6.9 ± 0.5 kPa (52 ± 4 mm Hg); fetal arterial pH, PO2 and PCO2 were 7.29 ± 0.06, 3.3 ± 0.8 kPa (25 ± 6 mm Hg) and 8.7 ± 0.9 kPa (65 ± 7 mm Hg), respectively. Recovery to standing occurred at 46 ± 17 minutes, and was generally smooth. Ponies regained normal behaviour patterns immediately. Conclusions and clinical relevance Propofol anaesthesia was smooth with satisfactory cardiovascular function in both mare and fetus; we believe this to be a suitable anaesthetic technique for pregnant ponies.  相似文献

Objective To characterize intravenous anaesthesia with detomidine, ketamine and guaiphenesin in pregnant ponies. Animals Twelve pony mares, at 260–320 days gestation undergoing abdominal surgery to implant fetal and maternal vascular catheters. Materials and methods Pre‐anaesthetic medication with intravenous (IV) acepromazine (30 µg kg?1), butorphanol (20 µg kg?1) and detomidine (10 µg kg?1) preceded induction of anaesthesia with detomidine (10 µg kg?1) and ketamine (2 mg kg?1) IV Maternal arterial blood pressure was measured directly throughout anaesthesia and arterial blood samples were taken at 20‐minute intervals for measurement of blood gases and plasma concentrations of cortisol, glucose and lactate. Anaesthesia was maintained with an IV infusion of detomidine (0.04 mg mL?1), ketamine (4 mg mL?1) and guaiphenesin (100 mg mL?1) (DKG) for 140 minutes. Oxygen was supplied by intermittent positive pressure ventilation (IPPV) adjusted to maintain PaCO2 between 5.0 and 6.0 kPa (38 and 45 mm Hg), while PaO2 was kept close to 20.0 kPa (150 mm Hg) by adding nitrous oxide. Simultaneous fetal and maternal blood samples were withdrawn at 90 minutes. Recovery quality was assessed. Results DKG was infused at 0.67 ± 0.17 mL kg?1 hour?1 for 1 hour then reduced, reaching 0.28 ± 0.14 mL kg?1 hour?1 at 140 minutes. Arterial blood gas values and pH remained within intended limits. During anaesthesia there was no change in heart rate, but arterial blood pressure decreased by 10%. Plasma glucose and lactate increased (10‐fold and 2‐fold, respectively) and cortisol decreased by 50% during anaesthesia. Fetal umbilical venous pH, PO2 and PCO2 were 7.34 ± 0.06, 5.8 ± 0.9 kPa (44 ± 7 mm Hg) and 6.7 ± 0.8 kPa (50 ± 6 mm Hg); and fetal arterial pH, PO2 and PCO2 were 7.29 ± 0.06, 4.0 ± 0.7 kPa (30 ± 5 mm Hg) and 7.8 ± 1.7 kPa (59 ± 13 mm Hg), respectively. Surgical conditions were good but four ponies required a single additional dose of ketamine. Ponies took 60 ± 28 minutes to stand and recovery was good. Conclusions and clinical relevance Anaesthesia produced with DKG was smooth while cardiovascular function in mare and fetus was well preserved. This indicates that DKG infusion is suitable for maintenance of anaesthesia in pregnant equidae.  相似文献
