In 2004, the isolation of an influenza virus from racing greyhounds changed the point of reference for discussions about influenza virus in dogs. A virus isolated from greyhounds did not have its origin in a previously described human influenza virus but came from a virus with an equine history. More significantly, evidence emerged to indicate that the virus was capable of transmission from dog to dog. This virus is now referred to as canine influenza virus (CIV) and is the focus of this review. Because the history of CIV is relatively short, the impact of this virus on canine health is yet to be determined.  相似文献

We conducted a cross-sectional convenience sampling study of dogs racing in the 2010 Iditarod to determine the seroprevalence of canine influenza virus (CIV) in the sled dog population. Questionnaires were completed detailing medical and CIV vaccination history, kennel size and location, travel history, and social interactions for each team. A total of 399 dogs were tested for CIV antibodies by hemagglutination inhibition assay. None of these, including 39 samples from dogs reported as CIV vaccinated, were seropositive for CIV antibodies. All of the vaccinated dogs were also negative on virus microneutralization assay. Risk factors for CIV seropositivity could not be determined due to a lack of positive samples. This is the first published study investigating the prevalence of CIV in sled dogs and additional studies are warranted to assess CIV infection among racing sled dogs and to evaluate the ecology of CIV and the vaccine efficacy in this population of dogs.  相似文献

Abstract

AIM: To determine whether canine respiratory coronavirus (CRCoV) and canine influenza virus (CIV) are present in dogs in New Zealand.

METHODS: Serum samples from 251 dogs of varying age, breed and clinical histories were tested for the presence of antibodies to CRCoV and CIV, using indirect fluorescent antibody (IFA) analysis. The population sampled represented a wide geographic area but principally encompassed the central and lower North Island of New Zealand.

RESULTS: Seventy-three of the 251 samples (29%) were seropositive for CRCoV. Dogs <2 years old were less likely to be seropositive for CRCoV than older dogs. None was seropositive for CIV.

CONCLUSIONS: This study revealed the presence of antibodies to CRCoV in dogs in New Zealand. Young dogs are less likely to be seropositive than older dogs, probably due to increased opportunity for exposure to CRCoV over time. Serum antibodies to CIV were not detected in any of the dogs sampled, suggesting that this virus is unlikely to be present in dogs in New Zealand.

CLINICAL RELEVENCE: Canine respiratory coronavirus is present in New Zealand. Although the role of this virus in canine infectious tracheobronchitis has not been fully elucidated, evidence suggests that it may have a causal role in this disease. Veterinarians should consider CRCoV as a differential diagnosis in cases of respiratory disease in dogs in New Zealand. While CIV appears not to be currently present in New Zealand, veterinarians should consider infection with this virus as a differential diagnosis in dogs presenting with respiratory signs.  相似文献

Background: Since 2004, canine influenza virus (CIV) has spread throughout the United States. While studies suggest that CIV is commonly detected in shelter dogs, little is known about its prevalence in household dogs. Objectives: To evaluate the seroprevalence of CIV in pet dogs presented for care in a veterinary hospital in Colorado and to investigate risk factors that might predispose these dogs to CIV infection. Animals: One hundred and forty dogs presenting to the Community Practice service, 110 dogs seen at other clinical services at Colorado State University's Veterinary Teaching Hospital in 2009, and samples from 75 dogs seen before 2004. Methods: In this prospective study, samples were tested with hemagglutination inhibition assays, using 3 CIV isolates. To identify risk factors for CIV infection, 140 owners completed questionnaires at time of sampling. Results: CIV seroprevalence was 2.9% (4/140) for dogs seen by the Community Practice service and 4.5% (5/110) for dogs seen by other hospital services (P= .48). All sera collected before 2004 tested negative for CIV. No differences were seen in antibody titers to the 3 CIV isolates tested. Data from the questionnaires indicated an association between CIV seropositivity and canine daycare visits (P < .001). Conclusion and Clinical Importance: CIV seropositivity in household dogs in Colorado is low, although it has increased since 2004. Antibody titers to the 3 CIV isolates were comparable, suggesting that measurable antigenic drift has not yet occurred. Finally, dogs boarded in kennels or attending daycare might be at an increased risk of CIV infection.  相似文献

Background

Canine influenza virus (CIV) H3N8 infection is thought to be highly contagious and potentially deadly. Low seroprevalence in dogs participating in the sport of flyball in 2009 was reported. It remains unknown if prevalence is changing in dogs at risk.

Objective

To determine seroprevalence of antibodies to CIV H3N8 in a population of pet dogs that participated in an indoor flyball tournament in Pennsylvania in November 2010.

Methods

Medical, travel, and activity histories for previous 10.5 months were obtained from consenting owners and serum samples were collected from dogs. Antibodies to CIV H3N8 were measured using hemagglutination inhibition.

Results

Of 251 competing dogs, 103 dogs were tested. Overall, seroprevalence of CIV H3N8 in dogs participating in flyball was 1.9% (95% CI, 0.05–6.8%). Thirty‐five dogs were tested in both 2009 (all titers <8) and 2010 (all titers <8). Ten of 103 dogs had been vaccinated for CIV H3N8. Two dogs had positive titers (>8) and both had been vaccinated twice in the previous month. Titers in remaining 8 vaccinated dogs were undetectable. Seroprevalence in vaccinated dogs was 20% (95% CI, 5.7–50.1%), whereas in unvaccinated dogs it was 0% (95% CI, 0–4.0%). Although 7 dogs had history of respiratory signs, only 1, which was vaccinated, was serologically positive for CIV H3N8.

Conclusions

Consistent with low prevalence of CIV H3N8 exposure previously reported in flyball dogs, unvaccinated dogs had 0% prevalence, suggesting a stable exposure rate. However, low seroprevalence may result in unprotected flyball dogs at risk for developing CIV H3N8.  相似文献

OBJECTIVE: To evaluate canarypox-vectored equine influenza virus (EIV) vaccines expressing hemagglutinins of A/equine/Kentucky/94 (vCP1529) and A2/equine/Ohio /03 (vCP2242) for induction of antibody responses against canine influenza virus (CIV) in dogs. ANIMALS: 35 dogs. PROCEDURES: Dogs were randomly allocated into 4 groups; group 1 (n = 8) and group 2 (9) were inoculated SC on days 0 and 28 with 1.0 mL (approx 10(5.7) TCID(50)) of vCP1529 and vCP2242, respectively. Dogs in group 3 (n = 9) were inoculated twice with 0.25 mL (approx 10(5.7) TCID(50)) of vCP2242 via the transdermal route. The 9 dogs of group 4 were control animals. All dogs were examined for adverse reactions. Sera, collected on days -1, 7, 13, 21, 28, 35, and 42, were tested by hemagglutination inhibition (HI) and virus neutralization (VN) assays for antibodies against CIV antigens A/Canine/FL/43/04-PR and A/Canine/NY/115809/05, respectively. RESULTS: Inoculations were tolerated well. The HI and VN antibodies were detected by 7 days after primary inoculation. Most dogs of groups 1 and 2 and all dogs of group 3 had detectable antibodies by 14 days after initial inoculation. The second inoculation induced an anamnestic response, yielding geometric mean HI titers of 139, 276, and 1,505 and VN titers of 335, 937, and 3,288 by day 42 (14 days after booster inoculation) in groups 1, 2, and 3, respectively. CONCLUSIONS AND CLINICAL RELEVANCE: Canarypox-vectored EIV vaccines induce biologically important antibodies and may substantially impact CIV transmission within a community and be of great value in protecting dogs against CIV-induced disease.  相似文献

In horse populations, influenza A virus subtype H3N8 (equine influenza virus, EIV) is a very important pathogen that leads to acute respiratory disease. Recently, EIV has emerged in dogs, and has become widespread among the canine population in the United States. The interspecies transmission route had thus far remained unclear. Here, we tested whether the interspecies transmission of EIV to dogs could occur as a result of close contact with experimentally EIV-infected horses. Three pairs consisting of an EIV-infected horse and a healthy dog were kept together in individual stalls for 15 consecutive days. A subsequent hemagglutination inhibition test revealed that all three dogs exhibited seroconversion. Moreover, two of the three dogs exhibited virus shedding. However, the dogs exhibited no clinical signs throughout the course of the study. These data suggest that the interspecies transmission of EIV to dogs could occur as a result of close contact with EIV-infected horses without clinical symptoms. Although the interspecies transmission of EIV is unlikely to become an immediate threat to canine hygiene, close contact between EIV-infected horses and dogs should be avoided during an EI epidemic.  相似文献

Avian-lineage H3N2 canine influenza virus (CIV)-associated respiratory disease, which can be fatal, emerged in South Korean dogs in 2007. We show here that dogs experimentally infected with CIV only developed respiratory tract diseases, as no extrapulmonary lesions and virus antigens were detected. This differs from the multiorgan diseases that avian influenza H5N1 induces in small experimental animals. However, the CIV-infected dogs developed a distinctively severe, long-persistent bronchointerstitial pneumonia, which differs from the acute but short-term bronchopneumonia that human (H1N1 and H3N2) influenza cause in rodents and ferrets. Histopathology and in situ TUNEL assays revealed that the neutrophils infiltrating the lesions were undergoing apoptosis, which probably reflects the attempts by the body to maintain appropriate numbers of neutrophils for defense against secondary bacterial infections. Our observations suggest that neutrophils along with the related chemoattractant cytokines (TNF-α, IL-1 and IL-8, etc.) may play a key role in the pathogenesis of H3N2 CIV in dogs.  相似文献

Background

Since equine influenza A virus (H3N8) was transmitted to dogs in the United States in 2004, the causative virus, which is called canine influenza A virus (CIV), has become widespread in dogs. To date, it has remained unclear whether or not CIV-infected dogs could transmit CIV to horses. To address this, we tested whether or not close contact between horses and dogs experimentally infected with CIV would result in its interspecies transmission.

Methods

Three pairs of animals consisting of a dog inoculated with CIV (108.3 egg infectious dose50/dog) and a healthy horse were kept together in individual stalls for 15 consecutive days. During the study, all the dogs and horses were clinically observed. Virus titres in nasal swab extracts and serological responses were also evaluated. In addition, all the animals were subjected to a gross pathological examination after euthanasia.

Results

All three dogs inoculated with CIV exhibited clinical signs including, pyrexia, cough, nasal discharge, virus shedding and seroconversion. Gross pathology revealed lung consolidations in all the dogs, and Streptococcus equi subsp. zooepidemicus was isolated from the lesions. Meanwhile, none of the paired horses showed any clinical signs, virus shedding or seroconversion. Moreover, gross pathology revealed no lesions in the respiratory tracts including the lungs of the horses.

Conclusions

These findings may indicate that a single dog infected with CIV is not sufficient to constitute a source of CIV infection in horses.  相似文献

Influenza viruses have been isolated from dogs in China; however, the extent of influenza infection among dogs is not yet clear. Here, we examined the seroprevalence of avian-origin canine H3N2, pandemic H1N1/09 and human seasonal H3N2 influenza viruses in pet dogs in China during January 2012 to June 2013. The seropositivity rate of canine H3N2, H1N1/09 and human H3N2 were 3.5%, 1.5%, and 1.2%, respectively. Dogs aged 2–5 years were most commonly seropositive to canine H3N2 virus. It is worth noting that two serum samples were positive against both canine H3N2 and H1N1/09 viruses, suggesting the possibility of coinfection with both viruses. Our findings emphasize the necessity for continued surveillance of influenza viruses in dogs in China.  相似文献

为了解灵缇犬对马流感病毒A/Equine/Huabei/01/2007(H3N8)的易感性,将马流感病毒培养液以1.0 mL(含105.7EID50)经静脉途径接种4条灵缇犬,另以2.0 mL(含2×105.7EID50)经滴鼻、点眼方式接种3条灵缇犬。采用临床症状和病理学观察、免疫酶组织化学染色技术、病毒分离、HI抗体检测和流感病毒受体类型检测技术,对病毒在灵缇犬体内的感染情况进行了系统研究。结果表明,感染的7条犬在整个试验观察期间体温等临床指标均无异常变化,未见明显的肉眼和组织学病变。感染后5 d,4条感染犬的气管、支气管和细支气管上皮细胞内流感病毒M蛋白均为阳性,1条犬的咽拭子病毒分离结果为阳性,1条犬的马流感病毒H3亚型HI抗体阳性。感染后14d,剩余3条感染犬中有2条犬马流感病毒H3亚型HI抗体呈阳性。试验证明,目前流行的A/Equine/Huabei/01/2007(H3N8)病毒可以感染灵缇犬,但不导致灵缇犬出现明显的临床症状。灵缇犬的喉头、气管上皮细胞具有与马流感病毒结合的SAα2,3 Gal受体。  相似文献

为了解广州地区犬流感的流行情况,本研究采集107份犬鼻咽拭子样品和58份血清样品,SPF鸡胚分离病毒并进行抗体检测。结果显示:分离获得3株H3N2亚型犬流感病毒。分离株的EID50为10-2.42/0.1 mL~10-3.5/0.1 mL;MDT为177.6 h~192 h;对乙醚、氯仿、酸和温度均敏感;对血凝素为热不稳定型;能够凝集公鸡、豚鼠、猪和牛的红细胞。  相似文献

Rupture of the dorsal ligaments of the tarsus in the greyhound is a rare racing injury. This case report describes dorsal tarsal instability in three greyhounds. A stumble and somersault were observed in two cases and were thought to have caused the injury. Previous reports advocate partial intertarsal arthrodesis as the preferred treatment for dorsal instability of the proximal intertarsal joint, but not all dogs returned to successful racing. In these three cases, stability was restored by the insertion of a wire tension band anchored to two bone screws, allowing the development of periarticular fibrosis. Canine respiratory coronavirus (CRCoV) has recently been detected in dogs; it is a group 2 coronavirus showing similarity to bovine coronavirus (BCoV) but is distinct from canine enteric coronavirus (CECoV). CRCoV may play an important role in canine infectious respiratory disease (CIRD) either by predisposing to further and potentially more serious viral and bacterial infections or possibly as a primary pathogen. The prevalence of serum antibodies to CRCoV, in a population of dogs in the south east of England, has been shown previously to be 30.1% on the first day of entry to a rehoming kennel [Erles, K., Toomey, C., Brooks, H.W., Brownlie, J., 2003. Detection of a group 2 coronavirus in dogs with canine infectious respiratory disease. Virology 310, 216-223]. The purpose of this study was to establish the prevalence of CRCoV in the general canine population within as well as outside the UK. An ELISA, used to test for the presence of antibodies to CRCoV in canine serum samples, identified seropositive dogs in UK, USA, Canada, Republic of Ireland and Greece. The development of an ELISA based on CRCoV antigen and immunofluorescence assay are described here. 54.7% (547/1000) of North American and 36.0% (297/824) of United Kingdom dogs were seropositive for CRCoV. The age and geographical distribution of seropositive dogs was also assessed. The cross-reactivity demonstrated between CRCoV antibodies from different countries and a UK viral isolate suggests immunological similarity. The overall prevalence of this virus in both North America and the UK suggests that CRCoV has international significance and that further epidemiological studies are required.  相似文献

Fifty-one calcaneus fractures associated with (41) or without (10) central tarsal bone (Tc) fractures in racing greyhounds were evaluated and categorized. All calcaneal fractures with no Tc fractures had a plantar proximal intertarsal subluxation. No subluxations were found in dogs with both calcaneal and central tarsal fractures. The calcaneal fractures were treated either with coaptation splints or surgical repair. Surgical techniques included a Steinmann pin with a figure eight tension band device or screw or plate fixation as primary techniques supplemented by Kirschner wires and cerclage wires. In all calcaneal fractures associated with plantar proximal intertarsal subluxation, an arthrodesis of the calcaneoquartal joint was performed. All 22 surgically repaired fractures in dogs available for physical and radiographic reexamination had healed within 1 to 6 months. Eight dogs with fractures of the calcaneus associated with fractures of Tc returned to a racing career. None of the dogs with plantar proximal intertarsal subluxation raced again. Based on the orientation of the fracture lines and on dissection of two tarsi with calcaneal fractures, a hypothesis on the pathogenesis of calcaneal fractures in racing greyhounds was formulated.  相似文献

A sero-epidemiological survey of human and equine H3 influenza A virus infections in dogs and cats using the hemagglutination inhibition (HI) and neuraminidase inhibition (NI) tests was conducted. Serum samples were collected from 582 dogs and 237 cats in Japan during the periods 2002-2008 and 1997-2008, respectively. Although no HI antibodies against equine H3 virus were detected, 9 (3.8%) from cats and 12 (2.1%) from dogs were HI-positive against human H3 virus. Only one serum each from dogs and cats was NI-positive against N2 virus. These findings suggest that although equine H3 influenza virus infections have not been prevalent in companion animals, human H3N2 influenza A virus infections have occurred in dogs and cats in recent years in Japan.  相似文献

The hemagglutination inhibition test is used by many diagnostic and surveillance laboratories for detection of antibodies to influenza viruses. It is well known that the hemagglutination inhibition test is affected by nonspecific inhibitors present in equine serum. Several serum treatments are in use to remove these inhibitors, including treatment with kaolin. Discrepant results were observed in the authors' laboratories when using kaolin treatment before testing equine sera for antibodies against equine influenza virus (EIV) subtype-1 (H7N7). It is demonstrated here that kaolin treatment leads to false positive results when testing for antibodies against EIV subtype-1, as compared to other standard serum treatments (trypsin-periodate, receptor-destroying enzyme). Against EIV subtype-2 (H3N8), however, false positive results were not evident. Trypsinperiodate and receptor-destroying enzyme (RDE) treatments appear to be superior to kaolin for removal of nonspecific inhibitors from equine serum and should be used for serological diagnosis and surveillance of equine influenza virus.  相似文献
