The conditions of isolating protein from tilapia frame by-products (TFB) by a pH-shift process were investigated along with their physicochemical and gelling properties. The minimum solubility of TFB protein was observed at pH 5.5 and gradually increased at both acid and alkaline sides. The highest solubility was observed at pH 2 and 12. Protein solubility of TFB increased as the ratio of alkaline extraction medium and extraction time increased. The highest protein extraction was found at the ratio of ground sample to extraction medium of 1 to 9 and extraction time of 15 min. The maximum protein recovery was found at pH 12 with 19.19%. The color of all protein isolate samples obtained from either acid or alkaline extraction was dark brown. Whiteness of a TFB protein isolate extracted at pH 2 was the highest with a value of 14.69. An alkaline pH-shift process effectively removed fat as much as 95%. Breaking force and deformation of recovered protein gel from an alkaline pH-shift were higher than those from the acid counterpart.  相似文献

ABSTRACT

Basa fish skin free from fat and noncollagenous proteins was hydrolyzed by different proteinases to produce angiotensin-converting enzyme (ACE) inhibitory peptides. Protamex hydrolysates were found to have the highest ACE inhibitory activity. Response surface methodology (RSM) was applied to optimize the hydrolysis conditions of Protamex—including enzyme-to-substrate ratio, hydrolysis time, and solid (fish skin)-to-liquid (water for hydrolysis)—at a temperature of 50°C and a pH of 7.0. The mathematical model demonstrated a good fit with the experimental data. An enzyme-to-substrate ratio of 6,108 U/g, a hydrolysis time of 1.24 h, and a solid-to-liquid ratio of 1:8.05 g/mL were found to be the optimal conditions to achieve the highest value of ACE inhibitory activity. The IC50 of the hydrolysates for ACE inhibitory activity was 1.417 mg/mL under these conditions.  相似文献

ABSTRACT

Extrusion cooking plays an important role in the production of puffed snacks, which enjoy wide acceptance all over the world and are potential vehicles for increasing the consumption of n-3 fatty acids. Response surface methodology (RSM) with central composite design was adopted to investigate the effects of four independent variables (fish powder, fish oil, moisture, and screw speed) at five levels on expansion ratio, breaking strength index, hardness, color, and thiobarbituric acid reactive substances (TBARS). A second order polynomial model was used to study the main, interactive, and quadratic effects of the independent variables and to explain the relationship between the responses and independent variables. Moisture was found to be the most significant factor (p < 0.001) influencing expansion ratio, breaking strength index, and color. Inclusion of fish powder and fish oil were found to be negatively associated with the expansion characteristics, resulting in decreased lipid oxidation due to less exposed surface area of the extrudates to air. Further, the absence of linear effect of fish powder and fish oil on TBARS demonstrated the possibility to incorporate higher levels of these ingredients to produce oxidatively stable extruded snacks rich in fish proteins.  相似文献

在单因素提取工艺优化的基础上,选取液料比、浸润时间、提取时间和提取温度进行四因素三水平的Box-behnken实验设计,并结合响应面分析法进一步优化丹参中抗无乳链球菌(Streptococcus agalactiae,SA)活性成分提取工艺。优化所得最佳提取工艺为液料比37.8:1.0(ml:g)、浸润时间104 min、提取温度86.7℃、提取时间182 min。响应面模型预测在该提取工艺下抗SA活性成分提取得率极值为25.95%,经4次平行验证实验的实际提取得率为(25.87±0.03)%,为优化前的2.59倍,表明优化方法高效可靠。  相似文献

采用三因素三水平中心组合试验设计方案,研究光照度、盐度、pH及三者交互作用对大竹蛏幼贝滤水率的影响,并进行响应面分析,探寻大竹蛏幼贝最适生长条件组合,构造大竹蛏幼贝滤水率模型。试验期为30d,试验光照梯度为2000lx、1000lx、0,盐度梯度为30、25、20,pH梯度为9.00、8.00、7.00。结果显示,光照度、盐度及pH三者对大竹蛏幼贝滤水率有显著交互作用(P0.05)。通过Design-Expert 8.0软件对数据进行二次多元回归,拟合得到大竹蛏幼贝滤水率y对编码自变量A(光照度)、B(盐度)和C(pH)的二次多元回归方程:y=1.62+0.084A+0.04B-0.14C-0.38A2-0.65B2-0.68C2(r2=0.9821),软件模拟最适大竹蛏幼贝生长的条件组合为光照度1109.17lx,盐度25.12,pH7.89。  相似文献

应用单因素试验、复合保护剂组合试验和响应面分析法,对鳗弧菌载体疫苗MVAV6203A-1冷冻干燥的保护剂配方进行筛选和优化。试验结果表明,众多保护剂中半乳糖、海藻糖和脱脂牛奶的组合使用,能够显著提高细胞对冷冻干燥环境的耐受力。以冻干后的细胞存活率为响应值,响应面法优化的结果为:半乳糖4.6%、海藻糖2.3%、脱脂牛奶10.2%,以优化的最佳配方进行的3次重复冻干试验,细胞平均存活率可达77.3%。优化的保护剂配方适用于鳗弧菌载体疫苗的冻干生产。  相似文献

The main objective of the current work was to evaluate the proximate chemical composition and nutritional quality of amino acids of protein hydrolysates from viscera of Tilapia nilotica fish. The results of amino acid composition showed that all hydrolysates contained sufficient and/or excessive amounts of both essential and nonessential amino acids, with appreciable values of essential amino acid/nonessential amino acid ratio (0.77 to 1.59) and essential amino acid/total amino acid ratio (43.35 to 61.35). All protein hydrolysates had a high content of the flavor enhancer amino acids. The lowest values of essential amino acids were found for phenylalanine and methionine with chemical scores ranging from 0.04 to 0.77 and 0.09 to 1.06, respectively, except for 18 h hydrolysates. The protein efficiency ratio had values greatly exceeding 2.0, which is considered a high quality. On the other hand, the levels of sugar and ash contents ranged from 2.69 to 3.32 g L?1 and 0.48 to 2.1%, respectively, with 9.2 to 14% dry matter. Mineral composition showed that sufficient amounts of calcium, sodium, potassium, magnesium, and iron were present. The results suggest that the studied hydrolysates are appropriate for use in balanced human or animal diets.  相似文献
