Using the full-cycle cultured (FC) Pacific bluefin tuna [body weight 16.3±1.9 kg (pre-fasting group, pre-FG), 14.2±0.9 kg (post-fasting group, post-FG)], changes in the physical/chemical properties of the cephalal parts of dorsal (D) and ventral (V) ordinary muscles (OM) by fasting (6 days) during chilled storage (4°C) were investigated. Condition factors were 26.7 (pre-FG) and 20.3 (post-FG, P < 0.05). Fasting changed the liver color to green. Fasting also decreased the amount of protein and lipid contents of the DOM and VOM of FC tuna. The breaking strength and pH of the DOM and VOM of post-FG tuna were higher (P <0.05) than for pre-FG tuna during storage. In contrast, the glycogen contents of DOM and VOM of post-FG tuna were lower than for pre-FG tuna. The color values (L*, a* and b*) of DOM of post-FG tuna were lower than for pre-FG tuna throughout the storage period. In addition, the metmyoglobin (metMb) content of DOM of post-FG tuna was lower (P <0.05) than that of pre-FG tuna, and the metMb content of VOM of post-FG tuna remained low after fasting. These results indicate that fasting suppresses deterioration (especially meat color) of FC tuna muscles during chilled storage.  相似文献

ABSTRACT:   Using the skeletal muscle of full-cycle cultured Pacific bluefin tuna (body weight: 13.1 ± 2.6 kg, cultured for about 21 months), the proximate compositions, breaking strength and histological structure of the front and rear parts of the dorsal ordinary muscles (FD-OM and RD-OM) and the ventral ordinary muscles (FV-OM and RV-OM) were compared. The FV-OM showed low moisture, protein, ash and high fat contents ( P  < 0.05, respectively) for the other three positions. The breaking strength of FD-OM, RD-OM and RV-OM increased up to 15–18 h and decreased later. However, the breaking strength of FV-OM was maintained during chilled storage. The pH of all muscles decreased up to 15 h, and then stayed at pH 5.7–5.8. However, the pH of FV-OM stayed at a higher level (pH 5.9). The histological structure observed by optical microscopy showed a space extension among muscle cells after 24 h in all positions, and these results were also supported by image analysis.  相似文献

The present study investigated the effects of lipid oxidation on quality deterioration in the ordinary and dark muscles of skipjack tuna Katuwonus pelamis during the early stages of ice storage for 72 h. The lipid hydroperoxide content of the dark muscle was significantly higher (P < 0.01) than that of the ordinary muscle throughout 72 h of ice storage. The metmyoglobin content of the ordinary muscle gradually increased, and was accompanied with darkening in the fish meat color. On the other hand, the addition of sodium ascorbate or 6-hydroxy-2,5,7,8-tetramethylchroman-2-carboxylic acid (Trolox®) to the ordinary muscle of skipjack tuna significantly inhibited the formation of lipid hydroperoxide as well as metmyoglobin formation. Thus, a decrease in α-tocopherol content in the ordinary muscle with antioxidant addition was not observed during ice storage period. In conclusion, the rate of lipid oxidation of skipjack tuna ordinary muscle is closely related to metmyoglobin formation, and the addition of antioxidants to fish meat is effective at inhibiting lipid oxidation as well as myoglobin oxidation in post-mortem meat.  相似文献

以菲律宾蛤仔为研究对象,对其活品贮藏特性进行探索。分别采取冷却干藏和常规湿藏两种方式,进行3个处理组贮藏特性的分析比较,即4℃干藏充气组、4℃干藏充氧组以及室温湿藏组;以存活率、磷酸精氨酸、三磷酸腺苷及其关联物、糖原以及pH为指标,跟踪分析各个处理组在10d保藏期间的代谢变化情况。研究结果显示,4℃冷却条件下的干藏保活效果总体好于常规湿藏,其中干藏充氧组的保藏效果最好,10d后存活率仍达到52%,其次为干藏充气组,7d后全部死亡;而湿藏组3d内全部死亡。生化指标分析结果进一步佐证了冷却干藏较常规湿藏有更好的保活效果,冷却干藏条件下肌肉pH、三磷酸腺苷及磷酸精氨酸水平均高于湿藏组,显示出较活跃的生命代谢状态;同时,冷却干藏组的活贝在保藏期间均维持较稳定的糖原水平。  相似文献

Pre‐slaughter handling involves fasting fish and catching them, which can affect fish welfare and flesh quality, but few studies have considered their combined effects. In this study, adult rainbow trout (320 ± 10 g average weight) were fasted for 7 days (135.6 degree days) and subjected to a long catch duration (20 min), compared with controls (no fasting or short catch duration). Condition factor, organ weight indexes and carcass yield decreased with fasting but not catch duration. Plasma concentrations of cortisol, glucose and lactate increased after a long catch, while plasma triglycerides decreased with fasting. Liver glycogen concentration was lower in fasted fish, and liver luminosity and chroma were higher after fasting with a long catch. Regarding flesh quality, rigor mortis resolved more slowly and final muscle pH at 48 hr post‐mortem was higher for fasted fish with a long catch time. Muscle glycogen concentration was higher in fasted fish, where chroma was also lower. Fasted fish had lower lipid oxidation, but there were no differences in fat content in muscle. Fasted fish with a long catch duration also had less monounsaturated and more saturated fatty acids. In conclusion, a long catch triggered a stress response that had negative effects on flesh quality, independently of fasting.  相似文献

ABSTRACT:   In this paper, the detection of type I collagen degradation during the softening phenomenon of yellowtail muscle, was examined. Acid soluble collagen was isolated from dorsal ordinary muscle at death and after 24-h chilled storage. In the abundant ratio of subunit components, an increase in β12 chain (5.4 points) and a decrease in components with molecular weights larger than γ chain (7.0 points) after 24-h chilled storage, was found. Type I collagen was detected in the alkali-soluble fraction by SDS-PAGE. Its amount calculated from hydroxyproline contents in alkali-soluble fraction was increased from 0.097 mg/g muscle to 0.155 mg/g muscle during 24-h storage. The increased alkali-soluble collagen (0.058 mg/g muscle) was about 1.4% of whole collagen. These results suggest that a slight decomposition of type I collagen of yellowtail muscle may occur and subsequently becomes alkali-soluble corresponding to postmortem softening.  相似文献

This study examines the relationship of the reproductive and reserve storage cycles and spat settlement of the Queen scallop (Aequipecten opercularis) to environmental factors. The possibility of farming this species from spat settled on collectors is considered. The gonad condition index shows maximum values between mid-May and early August. Another peak occurs in mid-November. In spring and summer, the energy needed for gametogenesis is derived from food, although it is possible that direct transfer may occur from the digestive gland to the gonad. The glycogen and lipid content of the reserve organs reaches maximum values between late summer and early autumn. The energy requirements related to gametogenesis, which begins in autumn, involve the consumption of accumulated reserves with the resulting reduction in the condition indices. There is a positive correlation between temperature and the condition indices of the digestive gland, the adductor muscle and their lipid and glycogen content, respectively. Two settlement periods were recorded, one in winter, which could not be associated with the spawning of the experimental animals, and another in summer, which coincided with the spawning that took place in spring. The reduced gonad indices recorded at the end of the summer and in winter did not lead to settlement.  相似文献

Abstract

The effects of freezing and frozen storage on the biochemical properties of scallop adductor muscles stored at 2-4EC were investigated. Glycogen content fell 40% in the first 12 hr of chilled storage and decreased slowly thereafter. ATP content increased for a short time after death and then decreased slowly. Glycogen and ATP degradation was accompanied by a decrease in both pH and 260/250 absorbance ratio of scallop muscle extracts. A significant increase (p < 0.05) in the hypoxanthine (Hx) content was also observed in chilled stored adductor muscles. Adductor muscles showed a significant decrease (p < 0.05) in the 260/250 absorbance ratio of extracts and an increase in the Hx level after freezing. An increase in Hx content was observed during frozen storage of adductor muscles frozen immediately after processing. Expressible juice increased with freezing and frozen storage of the muscles. The results shown in this paper indicate a rapid loss of quality in scallop adductor muscles during storage at 2-4. Freezing and frozen storage enhanced the quality deterioration of the muscles.  相似文献

虾夷扇贝的捕后处置对闭壳肌的硬化产生重大影响,而硬化又是影响闭壳肌生食品质的重要因素。为延长虾夷扇贝死后硬化发生时间,通过宰杀前暂养,缓解捕后胁迫对后期闭壳肌贮藏特性的影响。将采捕后的虾夷扇贝分为3组(对照组、暂养3 d组和暂养7 d组),然后去壳取闭壳肌进行0~7 d冰藏,分析闭壳肌在贮藏过程中的僵直情况、ATP关联物、K值、p H值、糖原及水溶性蛋白等指标的变化。结果显示,不论是否经过暂养,冰藏3 d内K值均低于5%,可以满足生食要求。经过暂养,闭壳肌中ATP的含量略有升高,暂养7 d组暂养3 d组对照组。在贮藏0~2 d内,暂养组的ATP含量基本保持不变,可抑制肌肉收缩并减少汁液流失。暂养组的糖原含量和水溶性蛋白含量低于对照组。研究表明,30 ku蛋白在暂养组中的消失时间比对照组提前了1~2 d。扇贝采捕过程中应尽可能减少胁迫,采捕后应尽快加工,以免影响商品的品质及货架期。  相似文献

ABSTRACT:   Using full-cycle cultured (FC) Pacific bluefin tuna (body length [BL], 42.6–66.4 cm; body weight [BW], 1.66–7.40 kg, n  = 15), the changes in chemical compositions and histological structure of the cephalal parts of the dorsal ordinary muscles (DOM) occurring with growth were investigated. A positive correlation ( r  = 0.9644, P  < 0.05) was observed between BL and BW with growth. The protein, lipid and ash contents of DOM and condition factor did not change with growth. However, the glycogen content of DOM increased ( P  < 0.05) from approximately 55 cm (BL) in this study. Using optical microscopic photographs, the various shapes of muscle fibers were observed and it was noted that the muscle fiber diameter increased ( P  < 0.05) with growth. Using transmission electron microscopic observation, many glycogen granules were observed in muscle fibers (especially, side of connective tissue) of DOM throughout the growth stage in this study. These results indicate that the glycogen content of DOM of FC Pacific bluefin tuna increases before the lipid with growth.  相似文献

为探讨短期禁食提升池塘养殖鱼类食用品质的技术可行性,以3个邻近池塘养殖的商品草鱼为对象,分别转移至另一个池塘的3个网箱中,进行21 d短期禁食。每间隔7 d采集3个池塘来源的草鱼,并将鱼体分割为A、B、C、D共4个身段,采集其肌肉样品,对肌肉水分、粗蛋白质、粗脂肪和糖原含量进行定量分析。结果显示,禁食7 d时,不同身段肌肉营养物质含量即发生改变,14 d时粗蛋白含量和粗脂肪含量变化较大,其中尾部(C、D段)的粗蛋白含量有显著增加的趋势;而10号池塘来源的草鱼A段粗脂肪含量在14和21 d分别比0 d减少了22.60%和17.07%,B、D段分别在7、14和21 d比0 d减少了7.50%、19.41%、12.61%和28.83%、36.68%、13.49%,17号池塘来源的的草鱼A、C和D段在7 d后显著增加,B段在7、14和21 d比0 d分别减少了26.76%、58.41%和62.90%,D段在7和14 d比0 d分别减少了1.52%和22.58%。3个池塘来源的草鱼肌糖原含量(C、D段)有显著增加的趋势。采集草鱼身体B段背部肌肉样品,测定其肌肉系水力、肌肉质构特性和组织切片,发现...  相似文献

The aim of the present study was to evaluate the effects of various sous-vide time–temperature regimes and their interactions on quality parameters of Atlantic mackerel (Scomber scombrus) during chilled storage. The mackerel ?llets were exposed to sous-vide treatment at 60, 75, and 90°C for 10, 15, and 20 min and further stored for 1, 3, and 7 days at 4 ± 1°C before analysis. Changes in pH, water content and cook loss, amount of water- and salt-soluble proteins, texture, and color parameters, as well as accumulation of lipid oxidation products in sous-vide-cooked mackerel were assessed. Sous-vide cooking time and temperature had the lowest contribution to the formation of primary and secondary products of lipid oxidation, as well as increase in yellowness of the fish flesh due to their accumulation; whereas duration of chilled storage led to a significant increase in oxidation and yellowness (p < 0.05). Duration of chilled storage also affected structural and textural properties of the fish muscle, leading to a decreased cook loss. At the same time, sous-vide cooking decreased the firmness of the fish muscle. Duration of chilled storage was found to have the highest significant effect (p < 0.001) on all physicochemical characteristics of sous-vide-cooked mackerel.  相似文献

Juvenile sea bass (Dicentrarchus labrax) were fasted for 22 days and changes in plasma insulin, glucagon and glucose levels, as well as glycogen and protein content in liver and muscle were analyzed. Glucagon increased initially on the 4th day of fasting. The glucagon/insulin ratio (G/I) increased from a value of 0.11±0.02 (24h of fasting) to 0.21±0.05 (4th day of fasting). Thereafter, both glucagon and insulin levels decreased and remained at low concentrations until the 22nd day of fasting. Plasma glucose levels fell at the beginning of fasting, stabilized between the 4th and 8th day, and gradually declined during the rest of the experiment. There was a body weight loss of 15% and a significant decrease in both the hepatosomatic index and mesenteric fat. The decrease in the percentage of muscle proteins was not significant, while liver glycogen content showed a sharp decline.  相似文献

Argentine hake (Merluccius hubbsi) was stored chilled for 8 days. The evolution of fluorescence of the aqueous and organic extracts of muscle during storage at 4°C was studied and correlated with common damage indices; pH, total volatile basic nitrogen, free fatty acids, and the fluorescence intensity of the aqueous phase showed a good correlation with storage time. The excitation-emission matrices of the aqueous extracts were also obtained. A fluorescence landscape that could be attributed to tryptophan decreased with storage time, meanwhile a peak (around 340 and 400 nm for excitation and emission wavelengths, respectively) is developed during chilled storage.  相似文献

ABSTRACT:   The post-mortem changes of type I and V collagens in Japanese flounder muscle during chilled storage were examined. The muscle softened significantly within 12 h under chilled storage. Both type I and V collagens isolated by ion-exchange chromatographies showed no remarkable changes in band patterns even after 24 h of storage, suggesting that degraded collagen molecules may be removed from the collagen fraction by a conventional preparation method. In contrast, type I and V collagen molecules were detected by Western blotting with each specific antibody in a saline extract and gradually increased during chilled storage. These results suggest that both type I and V collagens may participate in the post-mortem softening of fish muscle.  相似文献

We evaluated whether body fat content affects the energetic metabolism and growth in pacu submitted to daily feeding, fasting and refeeding. For 15 days, fish were fed different diets to obtain lean and fat conditions, and then subjected, for 20 days to: (1) continuously feeding (control), or (2) fasting for 15 days and refeeding for 5 days. Blood (glucose, triglycerides, cholesterol, non‐esterified fatty acids and total protein) and tissue (liver lipid and glycogen, muscle lipid and mesenteric fat) metabolic indicators, and growth performance parameters (weight gain, specific growth rate, daily feed intake and feed conversion ratio) were measured. Fasting led both lean and fat pacu to make notable use of their energy reserves, through glycogenolysis and lipolysis, reflected in reduced blood glucose and triglycerides, liver glycogen and muscle lipid levels. Lipolysis was confirmed by the high levels of non‐esterified fatty acids, especially in fat pacu. Refeeding led to higher plasma glucose and liver lipid in lean fish. Muscle fat increased in fat fish but was not restored in lean fish, while mesenteric fat index (MFI) remained the same in fat fish and increased in lean fish. Although refeeding occurred only for 5 days, lean fish grew more and were more efficient at utilizing food (higher weight gain and better feed conversion ratio). In conclusion, our results suggest that fat pacu have higher glycogenic and lipogenic abilities, and the higher deposition of lipids in fish does not mean higher availability of energy for growth when compensatory growth is stimulated by refeeding after fasting.  相似文献

Following the successful development of Pacific bluefin tuna (PBT) aquaculture, it is of considerable importance to determine the muscle fiber types and their growth patterns for future development. Muscle fiber profiles of dorsal ordinary, lateral ordinary (LO) and dark muscles and their growth patterns in PBT from 3.0 to 54.3 kg body weight were studied. Muscle fibers were histochemically stained for NADH-diaphorase and myosin adenosine triphosphatase activity (mATPase), and immunohistochemically stained with S-58 slow-muscle myosin antibody. All muscle fibers in dorsal and LO muscles showed low NADH-diaphorase activity, and acid-labile (pH 4.0 or 4.3) and alkali-stable mATPase activity. In LO muscle adjacent to dark muscle, three intensities of mATPase activity were observed after acid pre-incubation at pH 4.5 or 5.0, and the activity was related to the muscle fiber diameter. In dark muscle, all small and some large fibers stained intensely for NADH-diaphorase activity, related to their high aerobic metabolism. The high-active fibers with NADH-diaphorase in dark muscle were positive for S-58 antibody. Some large fibers in dark muscle showed intermediate NADH-diaphorase activity and high mATPase activity after alkali pre-incubations. These are fast-twitch oxido-glycolytic fibers in dark muscle and transformed to red muscle fibers with increasing body weight.  相似文献

Fish, Clarias Lazera were orally administered onion (Allium cepa) or garlic (Allium sativum) juices. Contents of glucose, free amino acids, total lipids, triglycerides, cholesterol, albumin, total protein, urea, aspartate aminotransferase (AST) and alanine aminotransferase (ALT) in serum as well as glycogen, free amino acids, total lipids, triglycerides and cholesterol contents in liver, white and red muscle were determined 5 h and 24 h after a single dose or after repeated doses (1 dose every 24 h for 5 days). Both onion and garlic treatments caused a temporary hypoglycemia as well as liver glycogen muscle depletion accompanied with a significant elevation in white muscle glycogen suggesting that dietary onion or garlic may enhance the process of glycogenesis in white muscle. Onion feeding caused hypolipidemia accompanied with significant decreases in liver content of total lipids in response to the repeated doses for 5 days and a severe rise in the lipid content of red muscle at all periods. Such results suggest that dietary onion may enhance the process of lipogenesis in red muscle of Clarias lazera. Probably, the main precursor of such lipogenesis in the red muscle was the serum fatty acids. Moreover, dietary onion led to hypocholesterolemia accompanied with a significant rise in cholesterol level of both liver and red muscle. On the other hand, dietary garlic significantly decreased serum free amino acids after 5 h and 5 days. Furthermore, significant elevations of liver free amino acids were dedected in the onion-treated fish group (24 h and 5 days) and in the garlic-treated fish group (5 h), probably due to oxidation of thiol components (sulfur attached to the proteins). This result was accompanied with either a marked increase in white muscle free amino acids at all periods of garlic fish group, or a noticeable drop in the onion group (repeated doses). Also, dietary onion and garlic treatment led to an elevation in hepatic ureagenesis in spite of the unchanged serum urea level. Significant decreases of both serum AST and ALT in garlic groups (all periods) and in onion groups (repeated doses) were observed. In liver, AST and ALT showed temporary rises after onion or garlic supplementation. In general, it could be concluded that both dietary onion and garlic have a transitory hypoglycemic action as well as a promoting effect on the process of glycogenesis in white muscle whereas dietary onion promotes lipogenesis in red muscle of fish. Also, dietary garlic may enhance the white muscle uptake of free amino acids which, in turn, may promote protein synthesis.  相似文献

We report the fate of glucose, both as a source of energy and as a temporary store, in the tissues of brown trout (Salmo trutta) in control, fasted and glucose-loaded fish. Tissue glucose utilization (3H-2-deoxyglucose phosphorylation) and storage (conversion of 14C-glucose into glycogen, protein, and lipid) were measured in immature brown trout, and the oxidation rate was calculated as glucose utilization minus storage and 14C-ionic metabolites remaining in the tissue. The glucose utilization rate is tissue-specific, the highest values being found in spleen, kidney, hindgut, brain, and gill. All these tissues also showed a highly active glycolytic pathway. The lowest utilization indices were observed in white and red muscles, skin, stomach and caeca, which also presented the largest proportion of glucose converted into stores (mainly protein and glycogen). Fasting reduced the glucose disappearance rate by 24%, although there were no significant variations in glucose utilization indices or distribution profile. After a glucose load, plasma glucose and insulin levels rose and the rates of glucose utilization, storage, and oxidation also increased in all tissues (from 1.5- to 4-fold). The relative importance of each tissue in glucose disposal was similar to that in normoglycaemia. In liver, only glucose storage was measured reliably; the conversion of glucose to glycogen was higher than in other tissues, and rose markedly (35-fold) in glucose-loaded fish. In most tissues glucose flux into lipids, glycogen and protein increased. The distribution of glucose may not be a merely substrate-mediated process because fasting in glucose-loaded fish caused lower tissue glucose utilization, particularly in gut, red muscle and gills. Conversion of glucose to tissue stores was reduced, lipids being the most affected.  相似文献

The main aim of the present work was to evaluate the effect of dietary butylated-hydroxytoulene (BHT as an antioxidant) and dietary lipid level on flesh lipid quality of Beluga sturgeon during frozen storage. An initial 135-day feeding trial evaluated practical-type diets containing 0 or 250 mg BHT kg?1 with two lipid levels (12 and 24% diet on dry matter basis) in a factorial arrangement. Fillet samples were analyzed fresh or after storage at ?18 ± 1°C for 12 months. Dietary antioxidant supplementation had no significant effect on fat content and fatty acid composition in the studied fish muscle, but increasing the lipid concentration in the feed increased muscle lipid concentration. Lipid oxidation in fish muscle is related to increasing lipid concentration in the feed. In addition, oxidation was reduced for fish fed BHT. Results showed that dietary BHT supplementation can slow down the level of lipid oxidation in Beluga sturgeon muscles during frozen storage, but it had a direct relationship with dietary lipid level.  相似文献
