1. Embryos of eggs incubated for 42 h after storage for 7 or 14 d were retarded by 5.3 and 12.2 h, respectively, compared with those of non‐stored incubated eggs.

2. This was not due entirely to a delay in the initiation of embryonic development; the rate of development during the first 2 d of incubation was significantly lower in eggs stored for 14 d than in non‐stored eggs.

3. There was no correlation between the developmental age of an embryo after any specified incubation time and either the original weight of the egg or the weight loss during storage.

ABSTRACT

1. Sex-specific variations in early embryonic development rates may pre-empt later variations in embryonic development through to pipping and hatching. Given that erythropoiesis (blood production) can be equated with early embryonic growth rate, it was hypothesised that blood pigment haemoglobin can act as a specific spectral fingerprint for changes in growth rate. Moreover, by measuring longitudinal, rather than lateral, spectral transmission through the egg, a more consistent spectrum with a higher signal-to-noise ratio could be captured.

2. Longitudinal visible transmission (T575/T598 ratio), which is sensitive to haemoglobin, was used to monitor sex-specific early embryonic development rate in white layer chicken eggs from d 0 to 8 of incubation. The sex of these eggs was subsequently confirmed two days after hatching.

3. Embryonic development was detectable from d 3 (72 h) of incubation, 36 h earlier than previously reported lateral spectral measurements, supporting the greater sensitivity of longitudinal measurements.

4. At d 3, the mean T575/T598 ratio for male embryos was significantly lower (P < 0.001) (i.e. higher absorbance of haemoglobin) than for female embryos, which was thought to be due to sex-differences in early embryogenesis. On the other hand, female embryos had a significantly lower (P < 0.05) mean T575/T598 ratio than male embryos at d 7 of incubation, presumably due to the combined effects of oestrogen synthesis receptors and enzymes on erythropoiesis in female embryos at this time.

5. In conclusion, the proposed methodology has the sensitivity to differentiate sex-specific embryonic development rates during early incubation and the potentiality to advance precision incubation management and poultry research.  相似文献

1. Embryos in eggs laid by older birds were developmentally more advanced, both at oviposition and after 24 to 42 h incubation, than those in eggs from younger birds.

2. Storage of eggs caused shrinkage of the blastoderm. The amount of shrinkage tended to decrease with parental age.

3. The developmental age of embryos after 42‐h incubation increased with parental age but was reduced by storage, with embryos in eggs from middle‐aged birds (35 to 40 weeks of age) being least retarded.

4. The rate of development in fresh eggs increased with parental age. Storage reduced the rate of development equally at all flock ages.

1. In two separate experiments, the possibility of detecting embryonic development in chicken eggs was assessed using the same spectrophotometric method used to detect blood in Table eggs, using a combination of two wavelengths (577 and 610 nm) of the transmission spectrum. 2. In the first experiment, during the first 10 d of incubation, transmission spectra of 30 Hisex White eggs and 30 Hybro eggs were measured daily. 3. In the second experiment, 292 Hisex White eggs were incubated. Seven groups were randomly assigned. Six received an injection of sodium azide (NaN3) at different times during incubation in order to stop embryonic development, and during the first 12 d of incubation the transmission spectrum was measured daily. The acoustic resonance analysis method was also used on a group of uninjected eggs. 4. In the first experiment, it was possible to detect embryonic development from 120 h of incubation onwards in fertile eggs. In the second experiment changes in light transmission due to embryonic development were detected from 108 h of incubation. Detection of embryonic development using the acoustic resonance analysis method in the second experiment was possible only from 120 h of incubation. 5. It was concluded that the detection of embryonic development using visible light transmission is not directly linked with the formation of blood, but with the formation of sub-embryonic fluid, which takes place from 72 h of incubation onwards. This fluid makes the yolk sac translucent so that absorption of light at 577 nm can be detected.  相似文献

Two techniques for egg-shell treatment were evaluated for their effects on the quantity and variability of the fluid absorbed by turkey hatching eggs subjected to the temperature-differential method of egg dipping. The use of pin or drill to pierce a uniform hole through the shell and membrane(s) on the large end of each egg caused a significant increase in the quantity of dip solution absorbed. Variability in fluid uptake by the eggs also was reduced significantly. Utilization of dilute acid to remove cuticle completely from the whole egg surface or partially from either end also had similar effects. The techniques described also permit and effective dipping procedure that does not require the preheating step.  相似文献

The sperm mobility assay measures the ability of sperm to swim through a dense layer of Accudenz®, and the sperm mobility phenotype has been shown to predict fertility and other sperm performance traits in roosters and turkeys. In this study, we examined turkey sperm morphometry and rates of early embryonic death associated with high‐ and low‐mobility semen. We also assessed whether the hypo‐osmotic stress test, which evaluates the structural integrity of the sperm plasma membrane, may be used as a faster and simpler assay for sperm mobility and viability. We confirmed previous work that found that high‐mobility sperm are faster and swim more linearly than low‐mobility sperm, and that mobility traits were repeatable within males. In contrast to previous studies, we did not find higher rates of fertility, but low‐mobility sperm was associated with higher rates of early embryonic death, though this trend was not significant. High‐mobility sperm had longer sperm heads, explained by longer nuclei, despite shorter acrosomes. Although these sperm were faster, midpiece length and flagellum length did not differ between high‐ and low‐mobility sperm. Finally, mobility was not found to be associated with sperm performance in the hypo‐osmotic stress test.  相似文献

Gamma-tubulin, a member of the tubulin superfamily, is a peri-centriolar component which is considered to be essential for microtubule nucleation. The dynamics of gamma-tubulin during mouse oocyte meiotic maturation, fertilization, and early cleavage as well as the co-localization of gamma-tubulin and alpha-tubulin during the formation of the meiotic I spindle were studied by confocal microscopy. We found that gamma-tubulin was evenly distributed in the germinal vesicle (GV) stage oocyte. After germinal vesicle breakdown (GVBD) gamma-tubulin dots were localized in both the cytoplasm and the vicinity of the condensed chromosomes, and aligned at both poles of the meiotic spindle at prometaphase I and metaphase I. At anaphase I and telophase I, gamma-tubulin was detected between the separating chromosomes, while it was absent in the midbody. At the MII stage, gamma-tubulin was again accumulated at the spindle poles. Alpha-tubulin had a similar distribution pattern as gamma-tubulin in the cytoplasm and radiated from gamma-tubulin foci close to the chromosomes during the meiotic spindle formation. After fertilization, gamma-tubulin was translocated from spindle poles to the area between separating chromatids and distributed around the pronuclei. It aggregated into some dots during the interphase, but was distributed on the mitotic spindle poles in early embryos. Our results suggest that gamma-tubulin is essential for microtubule nucleation and spindle formation during mouse oocyte meiosis, fertilization, and early embryo cleavage.  相似文献

1. In field trials, eggs from two flocks of Large White turkey hens were injected with about 0.2 ml saline solution containing 600 μg of pyridoxine hydrochloride in order to examine its effect on hatchability.

2. Also, in an aseptic laboratory trial, eggs from Large White turkey hens were injected with 0.2 ml of saline solution and 0.2 ml of saline solution containing 600 μg of pyridoxine hydrocholoride.

3. In field trials, hatchability of pyridoxine‐injected eggs was 4.6% higher (P<0.05) than the control (non‐injected) eggs.

4. In the aseptic laboratory trial, hatchability of pyridoxine‐injected eggs was 4.2% higher (P<0.05) than saline‐injected and control (non‐injected) eggs.  相似文献

The objective of this study was to evaluate selected morphological characteristics and the chemical composition of turkey eggs of different weight classes. The percentage of eggs weighing ≥100 g increased to 46.66% in wk 23. Eggs weighing 95 and 110 g had lower yolk content (3.05 and 4.86%) and higher albumen content (by 3.16 and 5.21%) compared with eggs weighing 80 g. The total protein content of the albumen increased (from 9.93 to 10.71%) and the water content of the albumen decreased (from 88.30 to 87.59%) with an increase in egg weight. Albumen weight per gram of yolk increased from 1.87 to between 2.18 and 2.42 g in larger eggs. The energy density of the egg decreased from 5.68 to 4.90 kJ/g with an increase in egg weight, and the energy density of the yolk remained at a similar level in all egg weight classes. The other analyzed yolk and shell parameters were not affected by egg weight. The highest energy and total protein content of the yolk was observed in wk 14 of the laying season. No correlations were found between the progress of the laying cycle and eggshell percentage and the water content of the yolk. The values of the remaining egg parameters varied over the laying season.  相似文献

卵母细胞胞质中存在着大量的母源性信息,正常受精卵的发育启动和早期卵裂主要由这些母源性信息所控制。为了评估卵胞质含量对早期胚胎发育的影响,以假显微注射兔原核胚为对照,采用显微注射技术将兔原核期胚胎去除或增加一定量的细胞质:试验一,增加5%和20%的细胞质;试验二,移入5%小鼠MⅡ期卵母细胞胞质。胚胎经显微操作后进行R1/R2序贯培养。结果显示,(1)增量5%组胚胎的2细胞胚(72.00%)、8细胞胚(60.00%)、桑椹胚(58.00%)和囊胚(54.00%)的发育率均显著(P〈0.05)高于增量20%组胚胎(分别为47.62%、35.71%、33.33%和30.95%),但与对照组之间无显著差异(P〉0.05);囊胚细胞数在2个增量组和对照组3者之间也无显著差异(P〉0.05)。(2)将5%小鼠MⅡ期卵胞质移入兔原核胚后,各阶段胚胎发育率和囊胚细胞数与增量5%组、对照组相比均无显著差异;早期胚胎经PCR检测,在2细胞胚(5/5)、8细胞胚(5/5)和桑椹胚(5/5)均全部检测到了供体小鼠mtDNA的D—loop区3’端片段,而在囊胚却仅有1枚(1/5)能够检测到。结果表明,增加少量细胞质不会对兔原核胚的体外发育造成显著影响,但胚胎发育率随着细胞质体积的增加有下降趋势,而异种卵胞质对兔原核胚的发育没有明显影响,在囊胚期虽仍能检测到异种卵胞质中mtDNA的存在,但异种mtDNA会随胚胎发育而逐渐减少。  相似文献

The incidence of bacterial contamination in turkey eggs which were rejected at candling or were "dead in shell" was about 4%. This incidence was lower than that previously found in chicken eggs. Analysis of the bacterial flora indicated that the proportion of Enterobacteriaceae was higher and the proportion of Micrococcus spp. lower in turkey eggs.  相似文献

1. A 31P topical magnetic resonance study of whole chicken's eggs in vivo is described.

2. Resonances from adenosine triphosphate, phosphocreatine, inorganic phosphate and phosphorous storage protein were observed. These changed in relative intensity as the embryo developed.

3. No ill effects due to exposure to radiofrequency radiation or magnetic fields were observed.

A precocious line of Eimeria meleagridis was developed by repeated propagation of the parasite in young turkeys and collection of the very first oocysts produced following infection. After 20 generations of selection, the prepatent period was reduced by 4-8 hr. Weight gain of poults given this line, from days 0-3 after infection, was significantly greater than that of poults given the parental strain, indicating that selection resulted in a loss of pathogenicity. Poults immunized with the precocious line produced no oocysts following challenge with the parental strain, indicating that immunogenicity was retained following selection. This is the first published report of the selection of a precocious line of Eimeria in the turkey.  相似文献

Effect of heat stress on early embryonic development in the beef cow   总被引:6,自引:0,他引:6  
Hereford and Hereford X Angus cows (n = 31) were utilized to determine the effects of heat stress on early embryonic development and survival. After acclimation to handling, cows were cannulated via the jugular vein on d 7 and assigned to either a control (C) chamber environment of 22 C, 35% relative humidity (RH) or one of two heat stress treatments. Ambient temperature was maintained at 37 C for 12 h followed by a decrease to 33 C for the remainder of the day in both treatment groups. Relative humidity was maintained at 27% in treatment 1 (TRT 1) and 38% in treatment 2 (TRT 2). On d 8 to 16, daily measurements of respiration rate (RES), rectal temperature (REC) and water intake were taken along with samples of blood, which were analyzed for hematocrit (HEM) and plasma concentration of protein (PP), progesterone (P4), estradiol-17 beta (E2), thyroxine (T4) and glucose (GLU). The uterus was recovered and flushed with saline on d 17 to recover the conceptus and uterine contents. Conceptus (if present) and corpus luteum (CL) wet weight were determined. Cows subjected to TRT 2 had increased RES and REC (P less than .01), while HEM was decreased (P less than .05) compared with C cows. Plasma T4 concentration was decreased (P less than .10) in TRT 2 compared with TRT 1 and cows, while P4 concentration were not significantly different. Corpora lutea wet weights were reduced (P less than .10) in heat-stressed cows vs C cows.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献
