Occurrence of Heterobasidion annosum s.l. was investigated in 15 coniferous stands in the Giresun and Pontic Mountains in north‐eastern Turkey. Basidiocarps of the fungus were found in 11 stands. Fifty‐two basidiocarp specimens of Heterobasidion were collected from stumps of Abies nordmanniana ssp. nordmanniana and two from stumps of Picea orientalis. Pure cultures were isolated from the basidiocarps and identified with the aid of mating tests. Forty‐five (90%) of the specimens collected from A. nordmanniana were identified as H. abietinum and five (10%) as H. annosum s.s. The former species was also found twice on P. orientalis. This is the first report of H. annosum s.s. for Turkey and the first report of H. abietinum on P. orientalis. Heterobasidion abietinum seems to be mostly a saprotroph on A. nordmanniana.  相似文献

Forty‐five basidiocarp specimens of Heterobasidion were collected from native Abies species in three locations in western Turkey: A. nordmanniana ssp. bornmülleriana in Bolu province, A. nordmanniana ssp. equi‐trojani in Balikesir province and A. cilicica in Antalya province. Pure cultures were isolated from the basidiocarps and identified to the species level with the aid of mating tests. All the specimens proved to belong to the species Heterobasidion abietinum. This root rot fungus is common in the forests investigated and appears to be relatively virulent on Abies in Turkey. This is the first report of H. abietinum outside Europe.  相似文献

Abies pinsapo, a rare fir found in the southernmost mountain region of Spain, is severely affected by Heterobasidion abietinum. It was hypothesized that spread of the disease might be favoured by woody species growing in the same habitat. In order to evaluate this hypothesis, artificial inoculation tests were performed on other potential hosts forming the tree cohort species with A. pinsapo. Inoculation test on seedlings demonstrated that H. abietinum was able to colonize stems of Pinus halepensis, P. pinaster and Quercus ilex subsp. rotundifolia, but failed on Q. faginea. Both Pinus species were colonized to a similar extent as the natural host, but Q. ilex subsp. rotundifolia showed a significantly lower susceptibility than conifers. These results suggest that Pinus spp. growing in mixed stands with A. pinsapo might favour spread of H. abietinum. As a consequence, natural replacement of pure A. pinsapo stands by mixed forest Abies‐Quercus should be facilitated.  相似文献

The efficacy of 30% aqueous urea solution, borax powder and spore suspensions of Phlebiopsis gigantea and Trichoderma harzianum against establishment of artificially inoculated Heterobasidion abietinum on Abies cilicica was tested both in the field and in a simulated stump treatment experiment carried out in a growth chamber. In the field, in each of the four selected stands 20 fresh stumps per control agent were treated and 20 stumps were left as untreated controls. In two of the stands, the treatments were applied in June and in the two others in November. Stumps were sampled twice, at 6‐ and 12‐months after treatment. In the growth chamber, 10 log pieces per treatment and control were used, and sampling performed after 6‐week incubation. Results of the urea and borax treatments were consistent between the experiments; the mean efficacies were 98.8 and 99.4% in the growth chamber, and 80.2–91.3 and 89.4–90.1% in the two samplings of the field experiment, respectively. Despite the high efficacies of the P. gigantea (85.9%) and T. harzianum (97.5%) treatments in the growth chamber, efficacies of these biological control agents in the field were 47.1–49.2 and 61.3–65.5%, respectively.  相似文献

Main branches of Abies nordmanniana Spach. were examined through their first growth season from subapical buds around the leader bud to fully expanded shoots. Plagiotropism was evident in branch orientation, which was almost horizontal, as well as in the orientation of buds developing on the branches. Auxin transport capacity was predominantly basipetal (> 90%) and consistently higher in the middle part of the branch than in the distal and proximal ends. Auxin transport capacity was higher on the dorsal side of the branch during the short initial hyponastic growth phase, but the difference disappeared when the branch became horizontal. No dorsal-ventral differences could be detected in young horizontal branches in concentrations of indole acetic acid, cytokinins, gibberellins or abscisic acid. Branch orientation was unaffected by decapitation of the leader apex or by decapitation and replacement with exogenous auxin. However, decapitation resulted in a less plagiotropic bud arrangement on the branches, and auxin application to the leader bud scar counteracted this effect. Thus, a signal originating in the stem seems to be involved in regulating branch bud positioning, whereas the horizontal branch orientation must rely on a different mechanism, presumably autonomic within the branch.  相似文献

In the last two decades, stand decline and increased mortality has affected silver fir (Abies alba) forests in the Spanish Pyrenees. Simultaneously severe occurrences of the root rot fungus Heterobasidion annosum s.l. and of the mistletoe Viscum album have been reported. We aimed to improve the understanding of the epidemiology of both pathogens in our region. All H. annosum isolates found on silver fir were typed as H. abietinum. H. abietinum was more frequently observed where cuttings had targeted fir trees rather than other species. H. abietinum fruiting bodies were observed in the most recently cut stumps. V. album was more abundant on more dominant fir trees, and in southern aspect stands. The number of V. album colonies in the stand correlated (R2 = 0.40) with silver fir mortality. Stands with a high level of V. album infection tended to have a smaller percentage of basal area in species other than silver fir, and they tended to be located on more south‐facing slopes. H. abietinum was widespread in silver fir forests of the Pyrenees. Our data suggest that, in the Pyrenees, the observed H. abietinum incidence may represent a combination of both primary and secondary spread of the pathogen. Favouring mixed forests should be tested as a potential control method for V. album. The correlation between silver fir mortality and V. album infection warrants further study, as the observed tree mortality could have occurred due to other factors than V. album, such as drought damage.  相似文献

The monoterpene composition of Picea abies and Abies alba resin was analysed in relation to growth by Heterobasidion spp. Fifteen‐year‐old P. abies and A. alba trees were inoculated on branches with three species of Heterobasidion annosum s.l. After 4 months of incubation, each host was colonized to a significantly greater degree by the pathogen specific to that host (H. parviporum on P. abies, H. abietinum on A. alba) than by the other fungi. Analysis of the enantiomeric monoterpene profiles in the spruce and fir showed that the response in terms of the relative proportions of the monoterpene compounds in the resin differed between tree species. Following challenge with Heterobasidion spp., A. alba trees did not show changes in monoterpene composition in addition to those in the wounding response (increase in (−)‐α‐pinene and (−)‐camphene, and decrease in β‐phellandrene). In P.abies, (−)‐α‐pinene, (+)‐α‐pinene and δ‐3‐carene increased following Heterobasidion attack but not after wounding alone.  相似文献

The tree pathogenic fungi Heterobasidion parviporum and H. abietinum are recently described sibling species that show marked host preference for Picea abies and Abies alba, respectively. H. parviporum shows a higher reproductive character displacement (i.e. higher inter‐sterility) towards H. abietinum in sympatry than in allopatry. We inspected whether this pattern was also present in H. abietinum by studying the inter‐sterility and gene flow amongst sympatric central European populations of H. abietinum and H. parviporum, and two isolated allopatric populations, Pyrenean H. abietinum and Scandinavian H. parviporum. Inter‐ and intra‐specific fixation index values were calculated from DNA sequence data of the GST‐1 locus and four microsatellite loci. Present allopatric and sympatric populations of H. abietinum were equally inter‐sterile towards H. parviporum, not suggestive of relaxed reinforcement. Several hypotheses explaining the observed pattern are presented. Genetic differentiation was observed between H. abietinum in the Pyrenees and in the Alps, suggesting geographical structure of H. abietinum in Europe.  相似文献

Abies nordmanniana and Picea abies seedlings were inoculated with a wild‐type (C 58) or an attenuated strain (rooter or shooter mutants) of Agrobacterium tumefaciens. Large tumours were formed on A. nordmanniana in response to the wild‐type and rooter strains, 60–65 % of the seedlings being susceptible. Smaller tumours were formed on Picea abies in response to the wild‐type strain, at least 12% of the seedlings being susceptible. Tumours from both species induced by the wild‐type strain grew in vitro on medium without added phytohormones.  相似文献

A fungus causing necrosis and needle cast has been discovered on fir trees (Abies alba) in a hight of 1700 m above sea level in Yugoslavia. The intensity of infection was very high, and the needles of all age classes were infected. The consequence of the infection is the death of the tree. The pathogen is described as a new species Tiarosporella durmitorensis Karad?i?. So far, this fungus has been found only in European silver fir stands in the National Park Durmitor and in the National Park Biogradska Gora (Montenegro).  相似文献

Twigs of 22 clones of 15‐year‐old Picea abies grafts were inoculated with Heterobasidion parviporum in July and in September 2003. Fungal growth varied between clones and exclusion of the pathogen varied significantly among clones inoculated in July. The results indicate that twigs exhibit the same infection pattern as cuttings when inoculated with H. parviporum and that exclusion of the pathogen in twigs is a clone‐dependent character that occurs during the vegetation period.  相似文献

The occurrence and symptoms of root and butt rot were examined in a 35 × 30 m plot of 68‐year‐old Todo fir plantation in Hokkaido, Japan. Forty‐seven percent of the cut stumps were decayed and 52% of the decayed stumps showed similar decay characteristics with yellowish orange to light brown colouration and expanded pockets in the heartwood. Morphological characteristics of the pure cultures isolated from the decay were similar to the cultures isolated from basidiocarps of Heterobasidion annosum sensu lato, found on fallen logs outside of the research site. Also DNA analysis based on the combined data set of three gene loci (glyceraldehyde 3‐phosphate dehydrogenase, heat shock protein 80–1 and elongation factor 1‐alpha genes) showed that the isolates from the decay are included in the same clade with the Japanese H. annosum s.l. isolates. They form a subclade to H. parviporum (the European S group of H. annosum s.l.). This is the first report of molecular determination of H. annosum s.l. isolated from root and butt rot in a plantation in Japan.  相似文献

A set of quantitative hierarchical real‐time PCR assays was developed for the detection of Heterobasidion irregulare, H. occidentale, H. annosum sensu stricto and of the entire Heterobasidion annosum complex. These assays enable specific and accurate detection and quantification of the target species from DNA extracted on airborne collected spores. Heterobasidion‐specific TaqMan? real‐time PCR detection assays were designed to function under homogeneous conditions so that they may be used in 96‐ or 384‐well plate format arrays for high‐throughput testing of large numbers of samples against multiple targets. Assays were validated for (i) specificity, (ii) sensitivity and (iii) repeatability. All assays were highly specific when evaluated against a panel of pure cultures of target and phylogenetically closely related species. Sensitivity, evaluated by assessing the limit of detection (with a threshold of 95% of positive samples), was found to be between one and a hundred ITS gene region copies or one conidia count equivalent. Precision or repeatability of each assay revealed a mean coefficient of variation of 5.9%. These molecular tools are now available for rapid and reliable monitoring of one of the most significant pathogen species complex of temperate northern coniferous forest around the world.  相似文献

Symptoms and signs associated with root rot caused by Heterobasidion annosum or Armillaria ostoyae in mountain pines (Pinus mugo ssp. uncinata) were investigated in the Swiss Alps. A sample of dying or recently dead mountain pine trees (≥12 cm d.b.h.) and saplings (<1.3 m height) was assessed for root pathogen infection by taking root samples followed by isolations in the laboratory. From a subsample, an additional core was taken from the butt of each tree and evaluated in the same fashion. A total of 157 dying or recently dead mountain pine trees and 184 saplings with roots infected by either of the two pathogens or which lacked infection were analyzed using logistic regression models. The main objectives were to determine the most prominent symptoms induced by the fungi (resinosis), signs of the fungi (mycelia, fruiting bodies and rhizomorphs), and tree characteristics (d.b.h./height and evidence of wounds) that would allow an easy and reliable determination of H. annosum and/or A. ostoyae infection of mountain pines in the field. Heterobasidion annosum caused both root and butt rot on mountain pine, whereas A. ostoyae was mostly restricted to the root systems of the trees sampled. The most discriminating sign for the presence of A. ostoyae infection was the presence of characteristic mycelial fans, and for H. annosum root rot the presence of H. annosum mycelia (sheets of paper‐thin mycelium and mycelial pustules). In addition, resinosis was a powerful predictor for A. ostoyae in trees. Symptoms and signs indicating A. ostoyae or H. annosum infections were more reliable for saplings than for mature trees. Armillaria rhizomorphs were not useful in detecting A. ostoyae infection and, if present, were often formed by saprophytic Armillaria species. Heterobasidion annosum fruiting bodies were rarely observed and poorly reflected the widespread occurrence of this pathogen in the mountain pine forests.  相似文献

The relationship between the proportion of the stump surface covered by the biological stump treatment agent PG Suspension, containing Phlebiopsis gigantea and its efficacy against the pathogen Heterobasidion annosum sensu stricto was studied during a first thinning of Corsican pine (Pinus nigra ssp. laricio) in Thetford Forest, UK. PG Suspension was manually applied to 100%, 75%, 50% or 0% of the surface of 150 stumps. Spores of H. annosum were inoculated onto 75 of the stumps, and the remaining stumps exposed to natural airborne spore deposition. The relationship between coverage and efficacy was found to be quantitative. Covering all the stump surface with PG Suspension completely excluded the pathogen, whereas stumps not treated with PG Suspension (the 0% treatment) became infected with H. annosum. Partial (75%) PG Suspension coverage resulted in the pathogen colonizing 40% of stumps following artificial inoculation with H. annosum, and just 7% of stumps exposed to ambient H. annosum spore infection. Decreasing levels of coverage allowed increasing areas of the stump surface to be colonized by H. annosum. Some small gaps in coverage were closed by lateral growth of P. gigantea, but it is recommended that operators aim for full stump coverage to give complete protection against H. annosum.  相似文献

Infections with Heterobasidion parviporum devalue the Norway spruce timber as the decayed wood does not meet the necessary quality requirements for sawing. To evaluate the incorporation of disease resistance in the Norway spruce breeding strategy, an inoculation experiment with H. parviporum on 2-year-old progenies of 466 open-pollinated families was conducted under greenhouse (nursery) conditions. Lesion length in the phloem (LL), fungal growth in sapwood (FG) and growth (D) were measured on an average of 10 seedlings for each family. The genetic variation and genetic correlations between both LL, FG and growth in the nursery trial and wood quality traits measured previously from 21-year old trees in two progeny trials, including solid-wood quality traits (wood density, and modulus of elasticity) and fiber properties traits (radial fiber width, tangential fiber width, fiber wall thickness, fiber coarseness, microfibril angle and fiber length). For both LL and FG, large coefficients of phenotypic variation (>?26%) and genetic variation (>?46%) were detected. Heritabilities of LL and FG were 0.33 and 0.42, respectively. We found no significant correlations between wood quality traits and growth in the field progeny trials with neither LL nor FG in the nursery trial. Our data suggest that the genetic gains may reach 41 and 52% from mass selection by LL and FG, respectively. Early selection for resistance to H. parviporum based on assessments of fungal spread in the sapwood in nursery material, FG, will not adversely affect growth and wood quality traits in late-age performance.  相似文献

To study the bark beetle attacks on Turkish fir stands and to determine the principle site and stand factors influencing beetle attacks, trap log method was used in the Western Black Sea Region of Turkey. The study was carried out in ten different locations prior to flying time in February 2003. On each experimental plot, three healthy fir trees with 30–50 cm diameter and 200 m apart were cut down with a chainsaw. From each sampling tree, needles were sub-sampled for determination of specific leaf-area and macro-nutrient concentrations. In addition, on each experimental plot, crown closure, basal-area, and stand density were measured. To determine the bulk density and nutrient concentrations of the soil, samples were taken around trees on each plot. The relationship between measured stand variables, the number of beetle species and beetle populations were evaluated using correlation and stepwise multiple regression procedure. Eight different species belonging to four different families from Coleoptera were identified in examining the trap logs. Six of these eight species (Pissodes piceae Illig., Pissodes notatus Fabr., Cryphalus piceae Ratz., Pityophthorus pityographus Ratz., Xyloterus lineatus Oliv., Pityokteines curvidens Germ.) were identified as harmful beetles (HB). The other two (Thanasimus formicarius L. and Rhizophagus dispar Payk.) were identified as predator species. The analysis showed that the total number of HB increased as the site’s slope and needle specific leaf area increased. However, as needle N concentrations increased, HB population decreased. Increased Ca concentration in fir needle reduced total insect attack.  相似文献

Growth rates of H. annosum s.s. and H. parviporum were investigated in the functional sapwood of young Pinus sylvestris and Picea abies plants as an indicator of the relative susceptibilities of the hosts to these pathogens. The stems of 520 five‐year‐old P. abies and 321 four‐year‐old P. sylvestris plants were inoculated and the extent of infection determined 16 weeks later. H. annosum s.sgrew further than H. parviporum in P. sylvestris sapwood, while in P. abies, no differences between the two Heterobasidion spp. were found. Both H. annosum s.s. and H. parviporum spread faster in the sapwood of P. abies than in P. sylvestris. There was high within‐host species variation in growth rates for both P. sylvestris and P. abies suggesting it may be possible to identify tree genotypes with lower susceptibility.  相似文献
