To estimate the importance of leaching of dissolved organic matter (DOM) as a pathway through which organic matter is supplied to stream ecosystems, we examined the amount of leachate over time and chemical properties of DOM leached from leaves in different conditions. The samples used were green leaves, yellow senescent leaves, and leaf litter of Salix gracilistyla Miq., which is the dominant riparian plant species in the middle reaches of rivers in western Japan. We analyzed dissolved organic carbon (DOC), total sugar, and polyphenol in the leachate of leaf samples collected from a fluvial bar in the middle reaches of the Ohtagawa River in Hiroshima Prefecture, Japan. Considerable leaching of DOC from senescent leaves [37.3 mg g−1 dry weight (dw) leaf] and leaf litter (8.1 mg g−1 dw leaf) occurred within 24 h after immersion. In contrast, DOC leached from green leaves was negligible until 1 week after leaf immersion. Carbon loss of leaves by leaching within 24 h after leaf immersion was estimated to be less than 8%, suggesting that leaching of DOC from S. gracilistyla leaves is a minor pathway through which organic matter is supplied to stream ecosystems. DOM leached from the leaves included sugar and polyphenol, which were among the major chemical forms of DOM leached from the leaves (based on the molecular mass). In a laboratory experiment in which the difference in the stability of DOM between the chemical forms was examined, sugar decomposed more rapidly than polyphenol.  相似文献

Aqueous suspensions of Phlebiopsis gigantea oidiospores, used as a biological control agent against Heterobasidion annosum, were either (1) set under pressure, (2) warmed or (3) used in mechanized stump treatment. Survival of P. gigantea following these treatments was monitored by estimation of oidia germination on agar medium and compared with the germination of oidia from control suspensions. Pressure up to 2200 kPa had no effect on the survival of P. gigantea. No effect on the survival was detected following incubation at ambient pressure for between 5 min and 8 h at 20°C. Phlebiopsis gigantea spore germination had an optimum at 30°C. When exposed to temperatures exceeding 30°C, viability of P. gigantea spores decreased with time. During mechanized stump treatment, survival of P. gigantea was not significantly lower, neither after the suspension had passed through the stump treatment device nor after 7 h time, compared with a fresh control suspension. Thus, mechanized application under normal summer conditions in Sweden did not obstruct the spore germination of P. gigantea in this study.  相似文献

Using attached and detached leaves ofAcer palmatum Thunb. andRhaphiolepsis umbellata Makino, pulse-modulated chlorophyll fluorescence and CO2 exchange were measured. Quantum yield of photosynthesis was determined from the fluorescence parameter(Fm′−Fs)/Fm′, where (Fm′−Fs) was defined as the difference between steady state chlorophyll fluorescence (Fs) and maximum fluorescence (Fm′) elicited by a saturating light pulse. The rate of electron transport through photosystem II (total electron flow) was calculated from the product of quantum yield andA (PFD), whereA is the rate of absorbed photons as given by leaf absorptance, and PFD is the photon flux density at the leaf surface. The rate of electron transport dependant on CO2 uptake (assimilative electron flow) was calculated from the gross photosynthetic rate in a leaf. The difference between the rates of total and assimilative electron transport was denoted as the rate of non-assimilative electron transport which depends on photorespiration and oxygen reduction. Available data provided quantitative information on the rate of non-assimilative electron flow in intact leaves. When leaf photosynthesis ofA. palmatum was measured under sunlight, the rates of total and assimilative electron transport were determined to be approximately 900 and 150 μmol equiv. e/mg Chl·h, respectively. The difference (750 μmol equiv. e/mg Chl·h) was attributed to the activity of non-assimilative electron flow. The ratio of total to assimilative electron flow was found to increase gradually with rising in irradiance. The results suggest that non-assimilative electron flow occurred at much higher rate than assimilative electron flow at high irradiance. Implications of the results are briefly discussed in relation to photosynthesis limitation in tree leaves.  相似文献

Shot holes on oriental cherry (Prunus serrulata var. spontanea) leaves were investigated to evaluate their role in the elimination of pathogens using analytical electron microscopy. Small brown spots caused by Mycosphaerella cerasella appeared on the leaf surface and developed into shot holes that were approximately 1–5 mm in diameter. Field emission scanning electron microscopy revealed clusters of fungal conidiophores and hyphal growth on the leaf surface around the shot holes. The fungal structures were mostly located within a radius of 500 μm from the shot holes and confined to the epidermal sector separated by veins. X‐ray microanalysis revealed distinct calcium ring patterns encircling the shot holes. The abscised surfaces consisted of suberized plant cells colonized with filamentous fungi and yeasts. Embedded in the mesophyll tissue, druse crystals were co‐localized with calcium and oxygen. Despite the fungal presence around the shot holes, no further expansion of the shot holes was observed on the leaf surface. These results suggest that shot holes may represent an incomplete but neutral abscission of infection loci in the presence of necrotrophic fungal pathogens. This microscopy study provides insight into the formation and roles of calcium oxalate in plants under pathological conditions.  相似文献

A study was conducted on the influence of clonal susceptibility, leaf age and inoculum density on the incubation and presporulation periods, and on the intensity of attack of Melampsora larici-populina races El and E3, under controlled conditions, on artificially inoculated poplar leaf discs. Both races were more virulent on susceptible clones and on 5- to 15-day-old leaves. On susceptible clones the intensity of attack increased progressively with the increase of inoculum density. Where there was complete coverage of the leaf surface, uredia were smaller in size as a result of the increase in density. The incubation period and, above all, the average presporulation period of susceptible clones proved to be longer with 1- to 3-day-old leaves and, generally, with the oldest. The epidemiological consequences of these results are discussed.  相似文献

Colonization of leaf litter by ligninolytic fungi and relationships between mass loss and chemical qualities of surface leaf litter were examined in Acacia mangium plantations and adjacent secondary forests in southern Sumatra Island, Indonesia. Leaves were collected from eight A. mangium plantations of different ages and three secondary forests. Partly decomposed leaves beneath the surface leaf litter were used to measure the bleached area which indicated colonization by ligninolytic fungi. Surface leaf litter was used to measure initial chemical content and subjected to the pure culture decomposition test. The bleached area was greater in secondary forests than in A. mangium plantations. Nitrogen content was higher in all the A. mangium plantations than in the secondary forests, and acid unhydrolyzable residue (AUR) content was generally higher in the A. mangium plantations than in the secondary forests. The bleached area of leaf litter was negatively correlated with nitrogen content of surface leaf litter at all sites, indicating an inhibition of the colonization by ligninolytic fungi of leaves with higher nitrogen content. In a pure culture decomposition test inoculating a ligninolytic fungus to surface leaf litter, mass loss of leaves was negatively correlated with AUR content of surface leaf litter. Mass loss of leaves and AUR was not significantly related to nitrogen content. These results suggested that higher nitrogen content in A. mangium leaf litter had a negative effect by colonization of ligninolytic fungi, but the effect of high N in A. mangium leaf litter on the decomposition of leaf litter and AUR remained unsolved.  相似文献

Leaf epidermal fingerprints of six species of Rhododendron ( Rh. Aureaum, Rh. dauricum, Rh. micranthum, Rh. Mucronulatum, Rh. Redowskianum, Rh. schlippenbachii) were observed by optical microscope with nail polish expression method in Key Laboratory of Forest Plant Ecology of Ministry Education China in Northeast Forestry University in 2004. The leaf morphological features including of stomata types, characters of guard cells, subsidiary cells in lower epidermis were observed. And ordinary cells (in shape and anticlinal walls feature) as well as the trichomes in both sides of the leaves are described in detail. The results showed that there were three types of stoma in six investigated Rhododendron species, from which Pericytic stomata type exists in three species (Rh. dauricum, Rh. micranthum, and Rh. mucronulatum), Anomocytic stomatal type in Rh. Redowskianum, Diacytic stomata type in Rh. aureaum and Rh. schlippenbachii. The subsidiary cells of the Pericytic and Diacytic stomata type are different in shape and surface feature between the species, respectively. The ordinary epidermal cells show a variety from quadrangular to hexagonal, polygonal or irregular in surface view, the anticlinal walls are straight or sinuose. Trichomes (gland scales) are present in the both of the leaf sides in three species (Rh. dauricum, Rh. micranthum, and Rh. mucronulatum). All of these detail leaf features show specific specificity of leave finger print for 6 rhododendrons.  相似文献

以盐肤木和黄栌为参照树种,用火炬树树叶水浸提液、林下土壤水浸提液浸种24 h,研究了浸提液对侧柏、油松、香椿、臭椿、小麦、小白菜种子萌发及幼苗生长的影响,研究了火炬树的化感作用。结果表明:火炬树完整鲜叶水浸提液处理小白菜种子,其发芽率和发芽势分别为88.0%、82.5%,与对照发芽率(94.0%)和发芽势(91.5%)无显著差异,而破碎鲜叶水浸液则极显著影响小白菜的发芽率和发芽势,分别降低为24.8%和5.0%;火炬树完整鲜叶和破碎鲜叶水浸液都能促进小白菜苗高生长,但都抑制小白菜胚根的伸长。与鲜叶相比,火炬树干叶水浸提液的化感作用明显降低。火炬树破碎鲜叶水浸提液极显著影响小麦发芽率,分别比对照的发芽率和发芽势降低11.0%和83.5%,干叶水浸提液则不影响小麦的发芽率,为95.0%,但发芽势比对照降低23.0%。与盐肤木和黄栌相比,火炬树破碎鲜叶水浸提液对小麦幼茎及胚根生长的抑制作用最强。火炬树破碎鲜叶水浸提液对4个乡土树种的种子萌发产生了不同程度的抑制作用,对香椿及臭椿发芽率的抑制作用极显著。火炬树破碎鲜叶水浸提液对4个树种的苗高以及油松苗胚根的伸长没有显著影响,但却极显著地抑制了侧柏、香椿及臭椿胚根的伸长。火炬树林下表层土壤水浸提液对小麦幼苗的生长没有抑制作用,能明极显著促进香椿的生长;处理55 d时,香椿苗平均高为8.63 cm,极其显著高于对照(5.14 cm)。  相似文献

Ammonium lignosulfonate (ALS) was found to stimulate the germination and growth of Phlebiopsis gigantea. When incubated in 1% and 2.5% solutions germination rates were higher and germ tubes longer at all temperatures tested when compared to water controls. When ALS was used to inoculate P. gigantea into red pine stumps the spores had germinated and hyphae were easily observed after 16 h but no germination had occurred in the stumps treated with oidia in water.  相似文献

A study was conducted to determine the effects of electrostatic field (ESF) treatment on seed germination and seedling growth of Sorbus pohuashanesis. The experiments were arranged by uniform design computed by the Data Processing System (DPS), including three levels of seeds soaking time, four levels of ESF intensity and four levels of ESF treatment time, with 12 treatments. Ten seeds were used in each treatment with three replicates. Seed vigor, seed germinating ability, emergence rate of seedling, survival rate of seedling, and seedling height and diameter, as well as the change in activities of superoxide dismutase (SOD), soluble protein contents, total chlorophyll contents, soluble total sugar contents in leaves of S. pohuashanensis seedlings were measured after ESF treatments. The experiment results show that ESF treatment could improve the water absorption ability of dry seeds of S. pohuashanensis, resulting in fast germination at room temperature under light conditions. Combined treatment of ESF with cold stratification could increase seed germination percentage significantly (to 42.20%), promote seedling height growth, affect leaf SOD activity, and could raise contents of total chlorophyll, soluble protein, and total soluble sugar in leaves. Seed soaking time had a significant effect on seed relative electroconductivity, seed germination under light, SOD activity, soluble protein content and total soluble sugar content of seedling leaves. ESF intensity exerted a moderate effect on these indexes. ESF treatment time only had significant effect on total chlorophyll contents, no evident effect on other indexes.  相似文献

Temperate conifer seeds are often considered to be shallowly dormant because a dormancy breakage treatment such as incubation of moist seeds at 4°C (prechilling) for 3–6 weeks improves germination. However, longer prechill durations can induce germination during the cold phase. Since the delicate radicles of prematurely germinated seeds are easily damaged and killed during sowing, potential benefits of extended prechilling are unknown.Described here for the first time is a target moisture content (TMC) prechill. In this dormancy breakage method the seeds are chilled at moisture contents high enough for removal of dormancy but low enough to produce surface dry, free flowing seeds that do not germinate during chilling. The method is a simple, effective and practical way to restrict the amount of water available to seeds from the start of a prechill treatment. The TMC prechill was applied to three conifer species, Douglas fir (Pseudotsuga menziesii (Mirbel) Franco), Lodgepole pine (Pinus contorta Douglas), and Sitka spruce (Picea sitchensis (Bongard) Carrière). All three species benefited from the TMC prechill. Extending the prechill period progressively improved germination by widening the range of temperatures over which germination occurred, raising germination capacity at most temperatures and increasing germination rate at all temperatures. The optimum prechill period combining high germination capacity and low mean germination time was 36 weeks for Douglas fir, from 36 to 76 weeks for Lodgepole pine and 36 or 76 weeks for Sitka spruce depending on incubation temperature. The belowground effects of Phytophthora cinnamomi on 1‐year‐old saplings of two common oak species in mid‐Atlantic US forests, white (Quercus alba) and red oak (Q. rubra), were examined after incubation in pathogen‐infested soilless potting mix. Fine root lengths (0–1.5 mm in diameter) of both oak species were quantified after incubation at successive 30‐day intervals up to 300 days, for a total of 10 incubation periods. In addition, colony‐forming units (CFU) of P. cinnamomi were quantified after white oak saplings were incubated in infested soilless potting mix at different temperature/duration combinations that reflect soil conditions present in the mid‐Atlantic United States. Impact of P. cinnamomi on fine root lengths of red and white oak saplings varied considerably over time. Significant periods of fine root loss occurred primarily during spring, when bud break and leaf flush began for both oak species. Red oaks had 17% fine root loss on average, while white oaks appeared more resistant to P. cinnamomi infection with a 2% decrease in fine roots over the course of the experiment. Phytophthora cinnamomi CFU declined significantly with exposure to all incubation temperatures except 8°C. This was in contrast to in vitro experiments, where the optimum temperature for mycelial growth was determined to be 21°C and above. Significant fine root loss caused by P. cinnamomi depended on plant phenology and the oak species tested. Extreme soil temperatures have a significant adverse impact on temporal changes of P. cinnamomi population.  相似文献

The zoosporic phase of the pathogen Phytophthora ramorum plays a crucial role in the process of plant infection, yet little is known about the fate of zoospores failing to target their hosts. Here, we describe new stages in the life cycle of P. ramorum concerning the in vitro development of monomorphic diplanetism and microcyclic sporulation in free water. Papillate cysts were formed after zoospore suspensions of isolates of the EU1 and NA1 clonal lineages were vortexed. Cysts usually germinated directly forming an emerging tube, or indirectly by releasing a secondary zoospore, which leaves behind an empty cyst with a short evacuation tube. Germinate cysts frequently developed either an appressorium or a microsporangium both terminally. We also observed microcyclic sporulation, i.e. sporangia indirectly germinated by forming a microsporangium, as in microcyclic conidiation of true fungi. Temporal progress of encysted zoospores in solution showed that percentage of germination varied significantly among and within isolates as well as between experiments, suggesting that germination is partly ruled by internal mechanisms. Diplanetism and microcyclic sporulation in P. ramorum may provide a second opportunity for host infection and may increase the chance of long dispersal in moving water.  相似文献

In two different experiments the urediniospores of Melampsoridium betulinum from Betula pendula and Betula pubescens germinated both on mature leaves of greenhouse-grown plants and on sterile in vitro leaves of micropropagated plantlets, which were cloned from the same B. pendula and B. pubescens genotypes. The urediniospores and germ tubes were more easily detached from the leaf surfaces of in vitro leaves. If germination took place on a leaf vein, the growth continued across the veinal ridges; otherwise, no determined growth towards the stomata could be observed with either of the leaf types studied. In both experiments on the in vitro leaves of B. pubescens clone V5944, the germ tubes of the rust isolate from B. pendula mislocated appressoria significantly more often than the germ tubes of the rust isolate from B. pubescens. On the mature leaves of B. pendula clone E4214 and B. pubescens clone V5940 there were also significant differences in appressorial locations between the two rust isolates but the clonal responses were inconsistent. The results of the inoculations suggest that the incompatibility of the rust isolates from B. pendula on the leaves of B. pubescens is not related to the significantly higher ratio of failures in locating appressoria in this host-rust combination.  相似文献

We used a null-balance porometer to measure leaf conductance of mature and primary leaves of natural seedlings, saplings, and trees ofPinus roxburghii (chir) during autumn at four sites at 1,320–1,930 m elevation in the central Himalaya of India. Our hypothesis that primary leaves had higher leaf conductance than mature leaves (needles in fascicles), based on measurements in other pines, was rejected. Comparisons on the same saplings and seedlings showed lower leaf conductance for primary leaves than for mature leaves. Primary leaves on seedlings and saplings also did not consistently have higher leaf conductance than mature leaves on near-by trees. Mature leaves on seedlings, however, did often have higher conductance than mature leaves on nearby trees. Mean values for leaf conductance (mmol m−2 s−1, based on total leaf surface area) ranged from 42–82 for mature leaves on trees, 60–121 for mature leaves on seedlings, and 28–67 for primary leaves on seedlings. Compared to published values for other pine species, conductance of mature leaves ofP. roxburghii is relatively low. We used various kinds of wasted tea leaves to develop composite sheets with antibacterial properties. Antibacterial tests showed that the number of viable bacterial cells for the sheet containing wasted green tea leaves was around 10:6–107 CFU/ml after 18 h culture compared to 108 CFU/ml for a tea-free sheet. This indicates that cell growth was signifi cantly inhibited. For sheets containing other types of tea leaves (oolong, black, hojicha, and pu-erh), living cells were not found, indicating that these sheets had superior antibacterial effects. Living cells were also not found in sheets containing wasted black tea leaves or roasted tea leaves at a concentration of 60% by weight after 6 h cultivation. Therefore, roasting treatment of wasted green tea leaves was examined to improve the antibacterial activity of the sheet. In particular, the focus was on structural conversion of catechins by heating.  相似文献

Plant–plant chemical interactions in forests can have a strong impact on the biodiversity and dynamics of these ecosystems, particularly in Mediterranean forests where plants exhibit a high secondary metabolite diversity. Allelopathic interactions in Mediterranean ecosystems have been mostly studied in the first stages of ecosystem dynamics, shrublands and pine forests, but little is known about these interactions in mature oak forests. In this study, the allelopathic effect of three main woody species of downy oak forests (Quercus pubescens, Acer monspessulanum and Cotinus coggygria) on germination and growth of two herbaceous species (Festuca ovina and Linum perenne) was tested through aqueous extracts obtained from different leaf phenological stages (green, senescent and litter). The germination velocity of the two target species was inhibited by the aqueous extracts of senescent leaves from all the woody species. The growth of F. ovina seedlings was affected by aqueous extracts of green leaves of all the woody species, while the growth of L. perenne was only affected by aqueous extracts of green leaves of A. monspessulanum. This shows that (i) allelochemicals released by leaf leachates of the dominant woody species could control the dynamic of the herbaceous species, and then their potential competition with trees and (ii) allelopathic effects of woody species are related to their phenological stage and seem consistent with the development stage of target species.

In glasshouse experiments, Cylindrocladium quinqueseptatum, which was associated with outbreaks of leaf spot and shoot blight in a young plantation of Eucalpytus microcorys in north Queensland during the 1982 and 1983 autumns, was pathogenic on seedlings of E. microcorys and caused varying degrees of leaf spot and shoot blight in ten species of Eucalyptus from major taxonomic groupings of the genus, When the outbreaks occurred, the weather was unusually showery with minimum temperatures above 16°C. In vitro studies showed that free water was essential for conidial germination, which occurred between 14–32°C. The optimum for mycelial growth was 28.5°C. In vivo studies showed that leaf penetration. by germinated conidia was via the stomata. Mancozeb was more effective at controlling the disease than benomyl, captan or copper oxychloride. The potential importance of C. quinqueseptatum and options for control in young Eucalyptus plantations are discussed.  相似文献

Seeds of Strychnos nux-vomica have slow and erratic germination; thus different presowing treatments were applied to enhance the germination of its seeds collected from Tamaraikulam, Tamil Nadu, India. In addition, the effects of desiccation and different storage conditions on the germination of S. nux-vomica seeds were investigated. The results show that soaking in 500 ppm gibberellic acid (GA3) for 24 h, incubation of seeds at 40 °C for 3 days and alternate water soaking (16 h) and drying (8 h) for 14 days significantly increased the percentage germination compared to the control. Desiccation of seeds down to 10% moisture content resulted in better germination. Germination of S. nux-vomica seeds differed significantly between different storage periods, moisture contents of the seed and for first and second order interactions (p<0.001). The highest germination (92%) was achieved when seeds with 10% moisture content were stored at ambient temperature for 30 weeks. Evidence from the present study indicates that S. nux-vomica seeds possess physiological dormancy that can be broken effectively by after-ripening. As seeds of S. nux-vomica are found to be desiccation tolerant, dry seed (10% moisture content) can be hermitically stored at ambient temperature for 30 weeks without losing their viability.  相似文献
