Yearling steers were treated with ACTH to determine the effect of increased plasma cortisol concentration on bovine lymphocyte and polymorphonuclear leukocyte (PMN) function. The administration of ACTH caused a significant (P less than 0.01) increase in serum cortisol concentration and depression of lymphocyte blastogenesis in response to phytohemagglutinin and concanavalin A. The response to pokeweed mitogen was also depressed, but not significantly. Random migration by PMN was significantly enhanced by ACTH treatment, but there was no effect on ingestion of Staphylococcus aureus, nitroblue tetrazolium reduction, or antibody-dependent cell-mediated cytotoxicity by PMN. The iodination reaction, which evaluates the activity of the myeloperoxidase-hydrogen peroxide-halide antibacterial system of the PMN, was significantly impaired after ACTH treatment. These data indicate that specific parameters of lymphocyte and neutrophil function were impaired directly or indirectly by elevated in vivo concentrations of plasma cortisol.  相似文献

Neonatal Japanese Black (JB) calves show a high incidence of diarrhea. The objective of this study was to analyze the immune cell populations of neonatal JB calves in detail and examine its correlation with the incidence of diarrhea immediately after birth. Understanding the immune cell populations is helpful in clinics in order to determine the condition of the immune system for prevention of diseases. Blood samples were obtained from JB calves on the day of birth. The peripheral leukocyte populations were analyzed separately for calves that had diarrhea within 2 weeks after birth (diarrhea group; n = 26) and for calves without diarrhea (control group; n = 74). The numbers of the peripheral blood CD3(+)TcR1-N12(+) and CD8(+) T cells were significantly lower in the diarrhea group compared with the control group. These findings suggest that the congenital lower peripheral γδ and CD8(+) T cells results in a high risk of diarrhea in neonatal JB calves.  相似文献

To clarify the cellular immune condition in Japanese Black (JB) calves with a weak syndrome, peripheral leukocyte populations were analyzed by flow cytometry. Twenty JB calves were divided into two groups based on clinical observations; one group of calves was weak, because they had experienced an onset of diarrhea within 3 days after birth and needed treatment (Group 1 ;n=10), and the other group of calves was healthy (Group 2; n=10). With regard to leukocyte populations, CD8(+) cells and gamma delta T cells in Group 1 were markedly lower than those in Group 2 during the experimental periods. It is possible that immune-insufficiency might be based on T lymphocyte function in weak syndrome JB calves during the growth process.  相似文献

The effects of cold climatic conditions on energy partitioning were investigated with 49 Hereford-type steers fed an all-concentrate, barley-based diet in a 2 x 3 factorial comparative slaughter trial. Steers (seven per treatment) were kept either indoors or outdoors (n = 2) and fed at 50, 65, or 80 g of DM/kg of BW.75 (n = 3) for up to 106 d. Mean temperatures were 16.9 +/- 2.7 degrees C and -7.6 +/- 6.8 degrees C in indoor and outdoor locations, respectively. Steers housed indoors grew 49% faster (P less than .001) and had 51% better gain: feed ratios (P less than .05) than those kept outdoors. Outdoor steers retained 65% less (P less than .001) energy. Estimated fasting heat production in the outdoor steers was 18% higher (P less than .01) and efficiency of ME use for maintenance 14% lower (P less than .01) than in the indoor steers, which resulted in an estimated increase of 41% in the ME requirements for maintenance in the outdoor steers. The NEg content of the diet was decreased from 1.29 Mcal/kg in the indoor steers to .76 Mcal/kg in the outdoor steers. Outdoor steers deposited 21% of their energy as protein, whereas indoor steers deposited only 14% of their energy as protein, which could explain the low NEg value of the diet in the cold environment. It was concluded that the main factors contributing to reduced energetic efficiency in the cold were an increased maintenance requirement and a greater proportion of the dietary energy retained as protein.  相似文献

Studies in infants and foals indicate an age-dependent maturation of peripheral lymphocyte subsets. The age-dependent relationship for maturation of cellular immune responses, such as phagocytosis and lymphocyte responses of the peripheral and pulmonary-derived leukocytes, has not been characterized in foals. Lymphocyte subpopulations, mitogen stimulation response of lymphocytes, lymphokine-activated killing cell activity, phagocytosis and oxidative burst activity, and serum immunoglobulin (Ig) classes G and M concentrations were determined in developing foals. This study illustrates age-dependent changes in immunoglobulin class concentrations, lymphocyte subsets, and EqMHC Class II expression in cells of the peripheral blood and lungs of developing neonatal-to-weanling foals. The increase in peripheral blood and BAL B-lymphocytes and serum immunoglobulins in developing foals suggests expansion of immune cell populations during a time in which environmental pathogen exposure is great. General immune function, mitogenic responses, LAK cell activity, opsonized phagocytosis, and oxidative burst activity of newborns was similar to the adult horse. Total immune-cell numbers, rather than function, seemed to be the limiting factor in the development of the equine neonatal immune system. There was an age-related percent increase in the appearance of pulmonary lymphocytes, but a percent decrease in macrophages. Although development of the respiratory immune system follows changes in the peripheral blood, cellular expansion, activation, and migration may occur at a slower pace, making the respiratory environment susceptible to pathogens prior to optimal immune system maturity.  相似文献

Zymosan-induced and luminol-aided chemiluminescence (CL) of whole blood from beagle dogs was estimated for the function of polymorphonuclear leucocytes (PMNs). Whole blood (0.1 ml) was examined directly and results were obtained within 20 min. A phagocytic function of PMNs can be estimated from the peak CL counts and the number of PMNs in a specimen, and the opsonic activity can also be estimated by the peak time showing peak CL after the addition of non-opsonized zymosan. The optimal temperatures to keep diluted whole blood for the CL measurement was around 13 degrees C. Thus, this method offers information concerning the functions of phagocytic cells in whole blood.  相似文献

Polymorphonuclear leukocyte (PMN) function and serum concentrations of estradiol, progesterone, and cortisol (hydrocortisone) were monitored concurrently in clinically normal cows during the estrous cycle. Five parameters were used to evaluate PMN function: (i) random migration under agarose, (ii) ingestion of 125I-labeled Staphylococcus aureus, (iii) nitroblue tetrazolium (NBT) reduction, (iv) iodination, and (v) antibody-dependent cell-mediated cytotoxicity. Increased serum estradiol concentrations were associated with enhanced random migration, but had no apparent effect on NBT reduction, iodination, or ingestion of S aureus by bovine PMN. Increased serum estradiol was also associated with increased serum cortisol. Increased serum progesterone values were associated with a depression of NBT reduction and iodination by PMN, but with enhanced random migration and antibody-dependent cell-mediated cytotoxicity. These results indicate that physiologic changes in steroid hormone values during the normal estrous cycle of the cow are associated with alterations in PMN function.  相似文献

Japanese black (JB) calves have greater susceptibility to infectious diseases compared to Holstein (Hol) calves. In order to clarify the differences in cellular immune status between JB and Hol calves, the leukocyte population and lymphocyte proliferative ability were analyzed. In total 200 healthy calves, 1 day to 14 weeks of age, were examined: 105 JB and 95 Hol calves. Lower numbers in peripheral blood and percentage in peripheral blood mononuclear cells of CD3+TcR1‐N12+ T cells and major histocompatibility complex class‐II+CD14 B cells were observed in the JB compared to the Hol. The percentage of TcR1‐N12+CD25+ T cell in the JB was significantly lower than that of the Hol at 4–6, and 8–10 weeks. Interleukin (IL)‐2 sensitivity in the JB was lower than that in the Hol, and significant differences were observed in age groups of 6–8 weeks and 10–14 weeks. These findings indicated that the lower numbers of γδ T cells and B cells in the JB compared to the Hol might be associated with the specificity of the immune systems in JB calves.  相似文献

We investigated the effect of supplemental vitamin E on the peripheral blood leukocyte population in Japanese Black calves. Twenty-six calves kept at the same farm were studied. They were divided into two groups; thirteen calves received 300 IU/day of vitamin E orally from 1 to 3 months of age (VE group), and the other thirteen calves did not receive the vitamin E supplement (control group). The VE group showed a higher serum vitamin E concentration at 2 and 3 months of age compared with the control group (P<0.01). The numbers of CD3+ cells and CD4+ cells were higher in the VE group than in the control group, and the difference was statistically significant at 3 months of age (P<0.05). The numbers of CD21+ cells were higher in the VE group than in the control group, and the difference was statistically significant at 2 months of age (P<0.05). The numbers of CD335+ cells tended to be higher in the VE group than in the control group. The numbers of CD8+ cells and CD14+ cells tended to be higher in the VE group than in the control group at 3 and 4 months of age. This study demonstrated that the supplementation of suckling Japanese Black calves with vitamin E might affect the numbers of some immune cell types in the peripheral blood.  相似文献

Four feeding trials were conducted to determine (1) how the organophosphate ronnel affects thyroid and adrenal circulating hormone levels and blood profile measurements of beef steers and (2) whether the effects of ronnel observed in beef steers could be produced in growing rats. In trial 1, four groups of eight steers each were given either no ronnel or 4 mg ronnel/kg body weight daily in diets fed at either limited (1.8% of body weight) or ad libitum intake. After 7 weeks, intake levels, but not ronnel treatments, were reversed during a 1-week transition period, and feeding was continued for another 7 weeks. In trial 2, four groups of six steers each were given 0, 44, 88 or 176 ppm ronnel premixed in diets fed ad libitum for 18 weeks. Actual ronnel intakes averaged 0, 1.01, 2.12 and 4.73 mg/kg body weight daily in trial 2. In trials 3 and 4 , growth and intake were measured in 64 and 32 growing Sprague-Dawley rats, respectively, fed levels of ronnel ranging from 0 to 100 ppm in the diet. In trial 3, blood plasma was analyzed at various times for triiodothyronine (T3), thyroxine (T4), cholesterol and total lipid content. In trial 1, the concentration of Plasma T4 was 1.31 times higher in steers fed ronnel that in control steers. In trial 2, plasma T4 was 1.30 times higher in steers fed 176 ppm ronnel, the level that improved growth, than in steers fed the lower ronnel levels. Circulating levels of T3, cortisol and aldosterone were similar for both control and ronnel-fed steers. Serum cholesterol concentrations were consistently higher in ronnel-fed steers. The data indicated that the growth-promoting effect of ronnel in steers may be associated with a shift in thyroid function. Effects of ronnel in steers were not observed in rats, demonstrating a species difference between steers and rats.  相似文献

To clarify the effect of nutritive conditions on changes in immune cells in Japanese Black (JB) calves during the growth period, leukocyte populations were analyzed in ten healthy JB calves managed in one herd. The calves were divided into two groups: five calves in Group 1 were given insufficient nutrition, and the other five calves in Group 2 received adequate nutrition. The levels of serum total cholesterol and glucose were significantly lower in Group 1 than in Group 2 at 1 month. The numbers of CD3+, CD4+ and CD8+ cells tended to be lower in Group 1 than in Group 2 at months 1 and 2, and the difference in CD4+ was significant at month 2. The number of MHC class-II(+high) cells was significantly lower in Group 1 than in Group 2 at months 1 and 2. These results suggest that adequate nutrition might stimulate an increase in immune cells in calves during the growth period.  相似文献

Twenty 10‐month‐old Japanese Black steers were used to evaluate the effects of clay on fat necrosis and carcass characteristics. Ten steers (Clay group) were fed the clay (50 g/day) during 10–30 months of age. The other 10 steers (Control group) were not fed it. There was no significant difference in body weight or average daily gain between the two groups (P > 0.05). The occurrence of fat necrotic mass in the Clay group (30%) was lower (P < 0.05) than that in the Control group (90%) at slaughter. The size of necrotic masses in the Clay group was smaller (P < 0.05) than that in the Control group. There was no significant difference in the marbling score, beef color, Longissimus muscle area or subcutaneous fat thickness between the two groups. These results suggest that the clay prevented the occurrence of fat necrosis and did not affect the carcass characteristics in Japanese Black steers.  相似文献

In a study of susceptibilities of sows from 2 herds to experimentally induced Escherichia coli mastitis, a marked difference was seen. The "susceptible" sows were from a conventional herd and "resistant" sows were from a specific-pathogen-free herd. The purpose of the study was to determine whether deficient neutrophil function was associated with increased susceptibility to E coli-induced mastitis. Four in vitro procedures were used to evaluate polymorphonuclear leukocyte (PMN) function: (i) random migration under agarose, (ii) ingestion of 125I-iododeoxyuridine-labeled Staphylococcus aureus, (iii) quantitative nitroblue tetrazolium reduction, and (iv) iodination. After parturition and intramammary inoculation with E coli, sows from the susceptible herd were neutropenic and the neutrophils which were present in the peripheral blood had reduced function. Specifically, there were depressed random migration under agarose, S aureus ingestion, and iodination when compared with PMN function in resistant sows. These data indicate that susceptibility to E coli mastitis was associated with deficiencies in PMN numbers and function. Potential causes of the neutrophil dysfunction are discussed and include possible systemic hormonal aberrations or the presence of an inapparent viral or bacterial infection.  相似文献

In this study, the effects of prolonged, high intensity training on aspects of peripheral blood and bronchoalveolar lavage (BAL)-derived leucocyte function were evaluated in 8 horses. All horses undertook a 7 week endurance training programme, followed by 5 weeks of high intensity training (HIT). Thereafter, horses were divided into control (C) and overtraining (OT) groups. The frequency and intensity of training were increased more substantially for horses in the OT group. Training was terminated in week 32 when horses in the OT group demonstrated a significant performance reduction. Peripheral blood and BAL samples were collected from 4 horses in C and OT groups in training weeks 7, 11, 14, 18, 22, 28 and 32. Flow cytometric techniques were used to assess phagocytosis by peripheral blood neutrophils and pulmonary alveolar macrophages (PAM), and oxidative burst activity of neutrophils, PAM, peripheral blood and BAL-derived lymphocytes. Peripheral blood neutrophil phagocytosis (internalisation) increased during the initial HIT period and decreased from week 16 when the training workload was increased for both groups. The oxidative burst activity of peripheral blood neutrophils and lymphocytes similarly increased and then decreased in response to training. The oxidative burst activity of PAM was reduced towards the end of the overtraining phase of the programme. Pulmonary alveolar macrophage phagocytosis and oxidative burst activity of BAL-derived lymphocytes demonstrated no change throughout the course of the study. There was no difference in results obtained from C or OT group horses, suggesting that protracted HIT, rather than overtraining, was associated with impaired cell function. The detrimental effects observed in peripheral blood neitrophil and PAM function may indicate impaired nonspecific immunity which may adversely affect the health and performance of horses undergoing protracted periods of intense training.  相似文献

OBJECTIVE: To determine the effects of training and sustained submaximal exercise on hematologic values in racing sled dogs. DESIGN: Cohort study. ANIMALS: 39 Alaskan sled dogs bred for endurance racing. Procedures-Blood samples were collected prior to initiation of a 7-month training regimen (n=39), after completion of the training regimen (19), and after completion of an 1,100-mile race (9), and a CBC, differential cell count, and flow cytometry for leukocyte surface antigens were performed. RESULTS: Both training and exercise caused significant decreases in PCV and hemoglobin concentration and significant increases in total WBC count. In contrast, training and exercise were not found to have significant effects on absolute numbers or fractions of CD4+ or CD8+ lymphocytes, other than a significant increase in the fraction of CD8+ lymphocytes associated with training. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggested that training and exercise induced changes in several hematologic values in racing sled dogs. Extracellular fluid volume expansion was the likely explanation for the training-induced decrease in PCV, and acute blood loss secondary to gastrointestinal tract bleeding was likely responsible for the decrease in PCV associated with acute exercise.  相似文献

Two experiments were conducted to evaluate L-carnitine supplementation to cattle fed grain-based diets. In Exp. 1, seven Angus-cross steers (216 kg) were used in a 7 x 4 incomplete Latin square experiment to evaluate the effects of supplemental L-carnitine on N balance and blood metabolites. Steers were fed a corn-based diet (17.5% CP) at 2.5% of BW. Treatments were 0, 0.25, 0.5, 1.0, 1.5, 2.0, and 3.0 g/d of supplemental carnitine. The 18-d periods included 13 d for adaptation and 5 d for collection of feces and urine. Blood was collected before feeding and 3 and 6 h after feeding on d 18 of each period. Dry matter intakes tended to be highest when 1.5 g/d of carnitine was supplied, but N retention was not affected by carnitine and averaged 29.3 g/d. Plasma carnitine concentrations and urinary excretion increased with increasing carnitine supply, indicating that at least some of the carnitine escaped ruminal degradation and was absorbed by the steers. Plasma concentrations of NEFA demonstrated a treatment x time interaction; they decreased linearly in response to carnitine before feeding but increased linearly in response to carnitine at 6 h after feeding. Serum insulin and plasma glucagon, IGF-I, cholesterol, triglyceride, and amino acids were not affected by carnitine. Plasma concentrations of glucose, glycerol, urea, and beta-hydroxybutyrate all were increased by some of the levels of carnitine supplementation, but results for these measurements did not follow easily described patterns and seemed to be related to differences in DMI. In Exp. 2, 95 crossbred steers (357 kg initial BW) were fed finishing diets (14.5% CP) for 129 d. Diets were based on steam-flaked corn and contained 6% alfalfa and 4% tallow. Feed intakes, gains, and feed efficiencies were not affected by supplementation with 2 g/d L-carnitine. However, steers receiving L-carnitine tended to have fatter carcasses, as indicated by tendencies (P < 0.2) for thicker backfat, higher marbling scores, and higher yield grades. In conclusion, carnitine supplementation did not alter lean deposition in growing steers but it did alter plasma NEFA concentrations of growing steers fed a corn-based diet and also seemed to increase fat deposition in finishing cattle.  相似文献

Multicystic renal dysplasia was found in a 6-day-old Japanese black bull. Grossly, both kidneys were markedly small (2.0 x 3.5 cm) with numerous cysts ranging from 1 to 8 mm in diameter. Histopathologically, both kidneys consisted of many irregularly enlarged cysts, immature glomeruli, small ducts and anomalous stromal connective tissues containing focal persistent mesenchyme characterized by a proliferation of stellate cells with myxomatous area. These features are compatible with those of multicystic renal dysplasia in humans and other mammals.  相似文献

提取惠阳胡须鸡脾脏、胸腺、盲肠扁桃体、胸肌、肝脏、肾脏、心脏、未刺激的外周血淋巴细胞(PBL)和ConA刺激后的PBL的总RNA,采用RT-PCR的方法检测IFN-α/β受体α链(IFNAR1)、IFN-α/β受体β链(IFNAR2)和IFN-γ受体β链(IFNGR2)基因的表达情况.在未经刺激的PBL中未检测到IFNAR1、IFNAR2的表达,ConA刺激后的淋巴细胞中IFNAR1、IFNAR2的表达量比较高;在组织中,仅在脾脏、盲肠扁桃体、肝脏和胸腺中检测到了IFNAR1、IFNAR2的表达.在未刺激的PBL、ConA刺激后的PBL、脾脏、盲肠扁桃体、肝脏、胸腺、胸肌中均检测到了IFNGR2的表达,在肾脏和心脏中未检测到IFNGR2的表达.IFNAR1、IFNAR2、IFNGR2在刺激后的PBL、免疫器官和肝脏中分布,表明IFNAR1、IFNAR2、IFNGR2可能参与机体的免疫调节和肝脏的代谢过程.不同的干扰素受体在PBL和各组织中分布情况的差异可能与其各自功能的差异有关.  相似文献

Little is known about the antimicrobial potency and specificity of polymorphonuclear leucocytes which actually appear at the sites of bacterial invasion in tissues. In the present work we have compared inflammatory leucocytes induced by intraperitoneal injection of casein in rabbits with autologous peripheral blood cells in killing Escherichia coli serotype 01 and Staphylococcus aureus 502A. The results indicate that inflammatory leucocytes differ significantly from their virgin blood ancestors. While the blood leucocytes were only able to suppress the growth of the gram-negative bacteria, autologous exudative cells killed more than 95% of the test organisms within 1 h of incubation at 37 degrees C. The enhanced microbicidal activity of the inflammatory cells however, was only specific for the gram-negative bacteria, as evidenced by the failure of leucocytes to kill Staph. aureus to the same extent as the peripheral blood cells. In association with the enhanced gram-negative microbicidal activity the inflammatory cells produced chemiluminescence and released two to three times more O2-anions than the peripheral cells. We interpret these observations to mean that chemotactic factors such as casein activate inflammatory cells to increase their oxidative metabolism. Since microbicidal action of leucocytes is thought to proceed in part through oxygen-dependent reactions, the inflammatory leucocytes would be expected to effectively kill bacteria that are highly susceptible to these lethal oxygen metabolites. It cannot therefore be assumed that assessment of the functional capacity of the virgin peripheral blood PMNs would provide information on the functional characteristics of activated leucocytes which actually migrate to and accumulate at inflammatory sites.  相似文献
