A study was designed to develop and define a sc tissue chamber as a suitable device for establishing a soft-tissue infection model in cattle and to use this model to study the interaction between Pasteurella haemolytica, sulfadiazine/trimethoprim, and bovine viral diarrhea virus (BVDV). Thermoplastic tissue chambers were implanted in the paralumbar fossae of 20 calves. At 35 days after implantation, calves were allotted to 4 groups of equal size and the calves in 2 groups were inoculated intratracheally with a New York-1 strain of BVDV. At 45 days after implantation, all chambers were inoculated with a 6-hour culture of P haemolytica serotype 1. Starting 36 hours after bacterial inoculation, sulfadiazine/trimethoprim was administered IV once a day to half of the virus-inoculated calves and to half of those calves that had not been exposed to virus. Inoculation of P haemolytica into tissue chambers resulted in the establishment of a localized soft-tissue infection, characteristic of pneumonic pasteurellosis. Despite the maintenance of chamber antimicrobial concentrations that exceeded minimal bactericidal concentrations established in vitro, the infections were not sterilized. This lack of efficacy was associated with decreased pH and increased protein concentrations in chamber fluids after inoculation. Infection with BVDV, which is thought to depress host defenses, had no effect on the response of P haemolytica to sulfadiazine/trimethoprim administration. Observation of responsive antibody titers, bacterial phagocytosis, and high leukocyte viability within P haemolytica-infected chambers documented functional host defenses within tissue chambers.  相似文献

Five of a group of six calves were inoculated with Mycobacterium bovis. Two more uninoculated calves were introduced to the group 84 days later. All the inoculated calves were subsequently shown to be excreting M bovis in nasal mucus. The uninoculated calf in the initial group of six became infected and subsequently excreted M bovis. The two uninoculated calves which were introduced later did not become infected. It was concluded that contact with nasal mucus from the infected cattle resulted in infection of the uninoculated calf and that the density of accommodation of animals excreting M bovis was an important factor in transmission of the disease.  相似文献

Mild clinical disease was produced in conventionally reared calves by the intranasal inoculation of 18-hour cultures of Pasteurella haemolytica simultaneously with Mycoplasma bovis; at necropsy seven days later moderate pneumonic consolidation was observed in two of four calves. Additional intratracheal injection of these organisms did not increase the severity of disease. In contrast, inoculation of six-hour cultures of P haemolytica with M bovis produced more severe disease and more extensive pneumonic consolidation. The most severe disease and greatest degree of pneumonic consolidation was induced by intranasal and intratracheal inoculation of six-hour cultures of P haemolytica one day after the intranasal inoculation of M bovis. Omitting the intranasal injection of P haemolytica reduced the severity and consolidation only slightly. Studies in gnotobiotic calves revealed that more severe disease and more extensive pneumonic consolidation resulted when M bovis was inoculated before P haemolytic rather than vice versa.  相似文献

Pathogenicity evaluations of Moraxella bovis strain EPP 63 grown in the piliated and nonpiliated phase indicated that the organism grown in the piliated phase induced clinical keratoconjunctivitis, whereas the organism grown in the nonpiliated phase did not induce disease under identical challenge conditions. Calves inoculated with culture grown in the piliated phase developed significantly (P less than 0.01) higher lesion scores than did calves inoculated with culture grown in the nonpiliated phase. Vaccination/challenge exposure evaluations indicated that calves immunized with vaccine prepared from piliated culture had significantly lower lesion scores than did control calves (P less than 0.001) or calves immunized with vaccine prepared from nonpiliated culture (P less than 0.05). Similarly, calves immunized with piliated vaccine developed a significantly higher antibody titer against purified pili. Nonvaccinated controls and calves immunized with nonpiliated vaccine did not develop a significant increase in antibody titer. The results indicate that M bovis pili constitute an important factor in the pathogenicity of keratoconjunctivitis and may be used as a vaccine in the control of keratoconjunctivitis in cattle.  相似文献

The efficacy of an ophthalmic ointment containing benzathine cloxacillin for treatment of infectious bovine keratoconjunctivitis was determined in 2 experiments. In the first experiment, Holstein calves (n = 6/group) were inoculated with Moraxella bovis and treated on postinoculation days 3 and 6 with either topically applied benzathine cloxacillin (250 mg/eye) or long-acting oxytetracycline formulation (20 mg/kg of body weight, IM). A third group of inoculated calves remained untreated as controls. For the second experiment, 4 groups of calves (n = 6/group) were inoculated and treated on postinoculation days 3 and 6 with 50, 125, 250, or 375 mg of benzathine cloxacillin; a fifth untreated group served as controls. Ocular specimens were obtained for microbiologic culture, and eyes were observed and assigned a clinical score daily. Eyes were photographed on alternate days. Ulcer surface area was measured, using a planimeter. In experiment 1, the week-2 ulcer surface area measurements for both groups of treated calves were smaller than those for controls. There was a greater frequency of M bovis isolation from the ocular secretions of controls than from those of benzathine cloxacillin-treated calves during postinoculation weeks 2 and 3. The number of M bovis isolations from the benzathine cloxacillin- and oxytetracycline-treated calves was not significantly different at any sample collection interval. On week 3, the scores of the benzathine cloxacillin-treated calves were smaller than those of controls. In experiment 2, calves of the 250- and 375-mg groups had smaller ulcer surface area measurements than did controls on week 2. In studies to determine whether vaccination with one strain of Moraxella bovis would protect against challenge with virulent homologous or heterologous strains, calves were intramuscularly inoculated 3 times with formalin-killed M bovis, with 14 days between inoculations. Fourteen days after the 3rd vaccinal dose was given, all calves were exposed to homologous or heterologous virulent cultures of M bovis. The results indicated that vaccination with one strain of M bovis may induce protective immunity against homologous and heterologous challenge exposure; however, because vaccinated cattle resisted infection and disease produced by a homologous strain to a greater extent than they resisted those produced by heterologous strains, polyvalent vaccines or highly immunogenic common antigens may be needed to protect cattle against the numerous strains they might encounter under natural field conditions. There was minimal correlation between the presence of precipitating antibodies against the heterologous strains and the establishment of infection and disease.  相似文献

Excretion of Mycobacterium bovis by experimentally infected cattle   总被引:4,自引:0,他引:4  
Three groups, each of five calves, four to seven months old, were inoculated intranasally with different numbers of Mycobacterium bovis. Infection was established readily in the calves which received an inoculum containing either 10(6) or 10(4) colony forming units (cfu). After every infection there was a lag period during which the organisms could not be isolated from specimens of nasal mucus. All the animals excreted M bovis and the time of commencement, quantity and duration of excretion appeared to be related to the inoculation dose. Excretion continued for many weeks, and for two calves excretion became intermittent over many months. All the calves which were given inocula of 92 cfu failed to develop the disease and no immunological responses were detected; however, M bovis was isolated from nasal secretions from one of these animals 100 days after inoculation.  相似文献

Because of the frequent exposure of cattle to mycobacteria of the avium/intracellulare group, an investigation was carried out into the possible repercussions thereof on the diagnosis of bovine tuberculosis. Three calves from a bovine tuberculosis-free herd, scored avian reactors in the gamma-interferon assay for bovine tuberculosis, were sedated and inoculated endotracheally with a virulent Mycobacterium bovis strain. Then, three other avian reactors were housed with the above donor calves. Mycobacterium bovis was isolated from the nasal swabs of the three endotracheally infected, donor calves. On these samples, TB complex-specific polymerase chain reaction (PCR) tests for IS6110 were also positive, albeit with a different time kinetics. The three contact-infected calves showed clear immunological signs of infection; however, their nasal swabs were always PCR-negative and only Mycobacterium avium was isolated. In the endotracheally infected donor calves there was a rise of the gamma-interferon responses to avian and bovine purified protein derivative (PPD) tuberculins, which reached the same stable plateau levels over the whole experiment. The above effect was also observed in the contact-infected calves, even though the response to avian PPD tuberculin always remained at a higher level. By using conventional bovine and avian PPD tuberculins, the comparative intradermal test was generally positive in endotracheally infected, as opposed to contact-infected calves; a positive intradermal test for M. bovis was obtained in two contact-infected calves by different bovine PPD tuberculins based on M. bovis bacillus Calmette-Guerin (BCG) secreted or somatic antigens. It was concluded that M. bovis infection may be concealed for some time in cattle sensitized by mycobacteria of the avium/intracellulare group and that different diagnostic procedures should be adopted for such animals.  相似文献

OBJECTIVE: To investigate the infection of calves with Mycobacterium bovis through oral exposure and transmission of M. bovis from experimentally infected white-tailed deer to uninfected cattle through indirect contact. ANIMALS: 24 11-month-old, white-tailed deer and 28 6-month-old, crossbred calves. PROCEDURE: In the oral exposure experiment, doses of 4.3 x 10(6) CFUs (high dose) or 5 x 10(3) CFUs (low dose) of M. bovis were each administered orally to 4 calves; as positive controls, 2 calves received M. bovis (1.7 x 10(5) CFUs) via tonsillar instillation. Calves were euthanatized and examined 133 days after exposure. Deer-to-cattle transmission was assessed in 2 phases (involving 9 uninfected calves and 12 deer each); deer were inoculated with 4 x 10(5) CFUs (phase I) or 7 x 10(5) CFUs (phase II) of M. Bovis. Calves and deer exchanged pens (phase I; 90 days' duration) or calves received uneaten feed from deer pens (phase II; 140 days' duration) daily. At completion, animals were euthanatized and tissues were collected for bacteriologic culture and histologic examination. RESULTS: In the low- and high-dose groups, 3 of 4 calves and 1 of 4 calves developed tuberculosis, respectively. In phases I and II, 9 of 9 calves and 4 of 9 calves developed tuberculosis, respectively. CONCLUSIONS AND CLINICAL RELEVANCE: Results indicated that experimentally infected deer can transmit M. bovis to cattle through sharing of feed. In areas where tuberculosis is endemic in free-ranging white-tailed deer, management practices to prevent access of wildlife to feed intended for livestock should be implemented.  相似文献

Newborn Holstein male calves were purchased within 3 days after birth and were removed from the local farms to the Dixon Springs Agricultural Research Center. They were hand-fed for 7 weeks and then weaned to a prepared feed. Eight groups, each of 4 calves, were housed in separate pens. In each of 4 pens (pens 2 to 5), 1 calf was inoculated with sporulated oocysts of Eimeria bovis (and was not medicated); 1 calf was inoculated and given feed with added monensin at the dosage level of 10 g/906 kg of feed; and 2 calves were inoculated and given medicated feed with added monensin at the dosage level of 20 g/906 kg or 30 g/906 kg. In the 4 other pens (6 to 9), the calves were inoculated with E zuernii and otherwise were given feed without or with added monensin as in pens 2 through 5. Another group of 5 calves (all kept in 1 pen), served as noninoculated, nonmedicated controls. At 14 days after inoculations with E bovis, the single calves in each of the 4 pens that were given the nonmedicated feed began to show clinical signs of coccidiosis and discharged increasing numbers of oocysts. The other inoculated calves (given monensin) had fewer clinical signs and discharged fewer oocysts in the feces as the level of medication in the feed increased. The calves inoculated with E zuernii developed only moderately severe infections when compared with those inoculated with E bovis. To evaluate the efficacy of a recombinant Moraxella bovis pilin-M. bovis cytotoxin subunit vaccine to prevent naturally occurring infectious bovine keratoconjunctivitis (IBK; pinkeye), a randomized, blinded, controlled field trial was conducted during summer 2005 in a northern California herd of beef cattle. One hundred and one steers were vaccinated with ISCOM matrix (adjuvant control), recombinant M. bovis cytotoxin carboxy terminus+ISCOM matrix (MbxA), or recombinant M. bovis pilin-cytotoxin carboxy terminus+ISCOM matrix (pilin-MbxA); calves received secondary vaccinations 21 days later. Calves were examined once weekly for 18 weeks for the development of corneal ulcers associated with IBK. Overall, the pilin-MbxA vaccinated group had the lowest overall cumulative proportion of ulcerated calves. Calves that received MbxA, whether alone or with pilin had significantly higher M. bovis cytotoxin serum neutralizing titers as compared to control calves. Results of ocular cultures suggested that vaccination with an M. bovis antigen affected organism type isolated from an ulcer: M. bovis was cultured more often from the eyes of control calves than from the eyes of calves vaccinated with MbxA and pilin-MbxA. In addition, vaccination of calves with MbxA and pilin-MbxA resulted in a higher prevalence of Moraxella bovoculi sp. nov. in ocular cultures. While no significant difference was observed between a cytotoxin versus pilin+cytotoxin vaccine against IBK, the reduced cumulative proportion of IBK in the pilin-cytotoxin vaccinated calves suggests it may provide an advantage over a cytotoxin vaccine alone. Mycobacterium bovis was isolated from the respiratory tracts of three cattle which registered negative to tuberculin testing; no tuberculous lesions were found and the culture of lymph nodes and other tissues proved negative. One animal was from a group of five calves which had been inoculated intranasally with M bovis, and the organism was recovered once only from nasal mucus sampled 100 days after inoculation. The second animal had had contact with experimentally infected cattle which were excreting M bovis and the third was from a commercial farm. The results of ELISAS for antimycobacterial antibodies and interferon-gamma, and of lymphocyte transformation assays are presented. The animals' immune responses provided evidence that each of them had been challenged.  相似文献

Acute ocular Moraxella bovis infections were induced in the UV-irradiated eyes of 10 calves. Eight calves developed corneal ulcers in at least 1 eye and were used for the treatment experiment. One randomly selected group of 4 calves with corneal ulcers and M bovis infections in 7 eyes was given a long-acting oxytetracycline formulation in 2 IM dosages of 20 mg/kg of body weight each, 72 hours apart. The other 4 calves with corneal ulcers in 6 eyes and M bovis in all 8 eyes served as nontreated controls. Bilateral ocular cultures were obtained and clinical observations were made daily for 20 days after treatment. After administration of the long-acting drug, new ulcers did not develop in the treated calves, whereas 5 new ulcers developed in the control-group calves during this time. The average durations of increased lacrimation/ulcerated eye were 2 and 12 days after treatment in the treatment and control groups, respectively; the average durations of blepharospasm were 3 and 8 days, respectively. Moraxella bovis was not isolated from any of the eyes of the treatment-group calves for the first 6 days after the antibiotic was administered, but was isolated from 1 eye of 1 treated calf on posttreatment day 7 and daily thereafter, for a total of 14 positive cultures of 160 ocular cultures obtained from the treatment-group calves after treatment. The bacterium was isolated from all eyes and from 144 of 160 cultures from the control-group calves during this time.  相似文献

A pure culture of Mycoplasma (M.) bovis was isolated from calves with respiratory disease, exhibiting the picture of lymphohistiocytic proliferative pneumonia with presence of eosinophil plasmatic cells. A mixed infection of M. bovis and Pasteurella (P.) multocida was demonstrated in calves with exudative pneumonia. Both M. bovis and Haemophilus (H.) somnus were recovered from calves with necrotic pneumonia. All 3 organisms--M. bovis, P. multocida, and H. somnus--were present in cases of exudative-necrotic pneumonia. It was also shown that M. bovis played a primary role in the aetiopathogenesis of respiratory diseases caused by mixed infections.  相似文献

Five calves from tuberculosis free herds were each inoculated intranasally with 10(6) viable organisms of a field isolate of Mycobacterium bovis. Four of the calves developed acute tuberculosis. ELISAs employing protein and phosphatide extracts of M bovis as antigens were used to monitor the humoral response of the infected calves. Fourteen days after infection there was a dramatic increase in the level of antibodies demonstrated by the phosphatide antigen. This increase coincided with the first appearance of signs of the disease. The results suggest that the phosphatide antigen may be of potential value in detecting a humoral response, if present, in cattle infected with M bovis. The tests employing the protein antigen demonstrated a humoral response in only one of the infected calves and emphasises the importance of antigen selection to detect antibodies in tuberculous animals.  相似文献

The effect of Mannheimia haemolytica infection on the penetration of ceftiofur and desfuroylceftiofur metabolites into tissue chambers was studied in cattle after subcutaneous administration of ceftiofur crystalline free acid sterile suspension (CCFA-SS). Four tissue chambers were implanted subcutaneously in each of 12 calves. Approximately 45 days after implantation, two chambers were inoculated with M. haemolytica (10(6) colony-forming units per chamber) while the remaining two chambers were inoculated with sterile phosphate-buffered saline. Twenty-four hours after inoculation, CCFA-SS was administered subcutaneously in the middle third of the caudal ear pinna of each calf. Chamber fluid and blood samples were collected at predetermined times for 10 days following dosing and analyzed for ceftiofur and desfuroylceftiofur metabolites by high-performance liquid chromatography. Concentrations of ceftiofur and desfuroylceftiofur metabolites in plasma and tissue chamber fluid remained above a threshold of 0.2 microg/mL for at least 8 days. Infected tissue chamber fluid concentrations of ceftiofur and desfuroylceftiofur metabolites were significantly higher than those in non-infected tissue chamber fluid, which correlated with significantly higher total protein concentration in infected tissue chambers. These results indicate that single subcutaneous administration of CCFA-SS at 6.6 mg/kg can be expected to provide effective therapy of susceptible bacterial infections for a period of at least 1 week.  相似文献

Mycoplasma bovis is perceived as an emerging cause of mortality in feedlot beef cattle. This study examined the lesions and infectious agents in naturally occurring M. bovis-associated bronchopneumonia and arthritis and the relationship of this condition with bovine viral diarrhea virus (BVDV) infection. Standardized pathologic, immunohistochemical, and microbiologic investigations were conducted on 99 calves that died or were euthanized within 60 days after arrival in 72 feedlots. Cranioventral bronchopneumonia with multiple foci of caseous necrosis was identified in 54 of 99 calves, including 30 with concurrent fibrinosuppurative bronchopneumonia typical of pneumonic pasteurellosis. Mycoplasma bovis was consistently identified in these lesions by culture and immunohistochemistry, but also commonly in healthy lungs and those with pneumonia of other causes. Focal lesions of coagulation necrosis, typical of pneumonic pasteurellosis, were often infected with both Mannheimia haemolytica and M. bovis. Arthritis was present in 25 of 54 (46%) calves with M. bovis pneumonia, and all calves with arthritis had pneumonia. BVDV infection was more common in calves with lesions of bacterial pneumonia than in those dying of other causes, but BVDV infection was not more common in calves with caseonecrotic bronchopneumonia than those with fibrinosuppurative bronchopneumonia. Retrospective analysis identified cases of M. bovis pneumonia in the early 1980s that had milder lesions than the current cases. The findings suggest that, in at least some calves, M. bovis induces caseonecrotic bronchopneumonia within the lesions of pneumonic pasteurellosis.  相似文献

The efficacy of tulathromycin in the treatment of bovine respiratory disease (BRD) due to Mycoplasma bovis was determined following experimental infection. Two highly pathogenic strains of M. bovis (with minimum inhibitory concentration values for tulathromycin of 1 and >64 microg/ml) were inoculated into 145 calves. Four days after inoculation, calves with clinical BRD were treated subcutaneously with saline or tulathromycin (2.5 mg/kg). Compared with saline, BRD-related withdrawals, peak rectal temperatures, and lung lesion scores were significantly lower for tulathromycin-treated calves (P < .01). Tulathromycin was highly effective in the treatment of BRD due to M. bovis in calves regardless of the minimum inhibitory concentration of the challenge strain (1 or >64 microg/ml).  相似文献

The left eyes of 10 conventional dairy cross calves were inoculated with a pathogenic strain of Moraxella bovis and lesions of infectious bovine keratoconjunctivitis developed in nine of these eyes. M bovis was isolated from all inoculated eyes and lesions developed in five out of 10 eyes which had become naturally infected. The clinical and microbiological findings were similar to those described in field cases.  相似文献
